Abstract

Description
本发明涉及液体配制剂,其生产方法及其作为动物饲料和人类食品的添加剂的用途。
类胡萝卜素由于其微溶于水和化学不稳定性而特别难配制。因此已做出很多努力以提供一方面稳定且另一方面显示出良好的生物利用率并在使用时具有所需给色量的类胡萝卜素配制剂。液体类胡萝卜素配制剂特别重要,因为获得粉末形式类胡萝卜素及使用者制备液体配制剂变得不必要。
具有纳米级粒度的类胡萝卜素初级颗粒的生产对于获得足够的生物利用率和给色量是关键的。为了生产这类颗粒,现有技术中使用研磨法,例如如WO 91/06292和WO 94/19411中所述。使用胶体磨在水性或油性介质中研磨类胡萝卜素,获得纳米级粒度。
EP 1 460 060 A描述了通过在油中研磨而生产叶黄素分散体。还描述了通过将叶黄素溶解于水混溶性有机溶剂中并在乳化剂的存在下在改性淀粉中包封而生产的可冷水分散的叶黄素配制剂。
US 2005/0084462描述了通过在作为载体的阿拉伯树胶的存在下在水中研磨色料并随后加热至30-60℃而生产用于食品、药品和化妆品中的色料混合物。
US 5,827,539描述了在油中研磨类胡萝卜素分散体,与淀粉、糖和抗氧化剂的混合物形成乳液并且随后喷雾干燥。
WO 9613178描述了通过在其中番茄红素基本上不溶的液体介质中研磨番茄红素而生产稳定的番茄红素提浓物。所用液体介质为甘油、丙二醇或乙醇。然而,不使用亲水胶体。
WO 96/23429描述了通过研磨虾青素,在研磨过程中或之后添加油而生产粒度<2μm的油性虾青素悬浮液。保护胶体和乳化剂的使用未公开。它指出颗粒易于团聚。因此,考虑通过在固化温度以下贮存该悬浮液而进一步稳定。
WO 93/04598描述了在油、水分散性基体成型剂如糖的分散体和稳定剂如明胶或酪蛋白的分散体、乳化剂及合适的话非油性溶剂如甘油中生产包含类胡萝卜素的类胡萝卜素组合物。
本发明的目的为提供能够容易生产并且使用者无需额外努力便能够使用的液体类胡萝卜素配制剂。
目前已惊人地发现该目的通过除类胡萝卜素和亲水性保护胶体之外还包含至少一种水混溶性醇的液体配制剂实现。
本发明因此涉及一种包含至少一种类胡萝卜素、至少一种亲水性保护胶体和至少一种水混溶性醇的液体配制剂。
可以使用的类胡萝卜素为可由天然源或通过合成获得的已知代表。此外,可以使用可由天然源或通过合成获得的已知代表。其实例单独或作为混合物为β-胡萝卜素、番茄红素、叶黄素、虾青素、玉米黄质、隐黄质、柠黄质(citranaxanthin)、斑蝥黄、海胆酮、红木素、β-阿朴-4-胡萝卜素醛、β-阿朴-8-胡萝卜素醛、β-阿朴-4-胡萝卜素酯。优选虾青素、β-胡萝卜素、β-阿朴-8-胡萝卜素醛、β-阿朴-8′-胡萝卜素酸乙酯、斑蝥黄、柠黄质、海胆酮、叶黄素、番茄红素和玉米黄质。特别优选虾青素和斑蝥黄,尤其是虾青素。
类胡萝卜素的粒度通常为50nm-10μm,优选为100nm-5μm,特别优选为100nm-3μm、150nm-2μm,尤其是200nm-1μm。
水溶性或水溶胀性胶体适合作为亲水性保护胶体。保护胶体优选为蛋白质基保护胶体、改性淀粉衍生物和改性纤维素衍生物。蛋白质基保护胶体特别是酪蛋白、酪蛋白酸盐、牛明胶、猪明胶或鱼明胶,尤其是勃鲁姆数(Bloom number)为0-250的酸降解或碱降解明胶及其混合物。改性淀粉衍生物和改性纤维素衍生物特别是酯如淀粉辛烯基琥珀酸酯。相应的产品由National Starch以名称Purity Gum 2000或者由Roquette以Clear GumCO 01、由National Starch以Hi Cap 100或Capsul市售。
优选将改性淀粉用作亲水性保护胶体。
合适的水混溶性醇为一元醇和多元醇。一元醇为链烷醇如甲醇、乙醇、异丙醇等。然而,优选使用多元醇。这些特别是二元醇或三元醇。其实例为乙二醇、丙二醇、丁二醇、聚乙二醇、聚丙二醇、甘油、甘油与C1-C5单羧酸的单酯或甘油的单醚。本发明配制剂特别优选包含甘油。
类胡萝卜素通常以基于配制剂总重为0.1-50重量%,优选0.2-30重量%,尤其是1.0-15重量%的量存在于本发明配制剂中。
亲水性保护胶体的量通常为1-40重量%,优选为2-30重量%,特别是3-25重量%。
类胡萝卜素与亲水性保护胶体的重量比通常为5∶1-1∶5,优选3∶1-1∶3。水混溶性醇的量基于配制剂总重通常为30-95重量%,特别是30-90重量%,特别优选35-90重量%。
本发明配制剂可以包含基于配制剂总重为至多40重量%,优选至多35重量%的水。
本发明配制剂优选不包含油。
为了增加活性成分相对于氧化降解的稳定性,有利的是添加稳定剂如α-生育酚、叔丁基羟基甲苯、叔丁基羟基茴香醚、抗坏血酸或乙氧喹。
本发明配制剂可以额外包含辅助物质如增稠剂,硬脂,螯合剂如柠檬酸、肌醇六磷酸或膦酸的碱金属或碱土金属盐和/或乳化剂。乳化剂的实例为棕榈酸抗坏血酸酯,聚甘油脂肪酸酯如聚甘油3多聚蓖酸酯(PGPR 90),脱水山梨醇脂肪酸酯如脱水山梨醇单硬脂酸酯(span 60)、PEG(20)脱水山梨醇单油酸酯,丙二醇脂肪酸酯或磷脂如卵磷脂。
通过研磨类胡萝卜素和亲水性保护胶体在所用水混溶性醇中的分散体而制备本发明配制剂。然而,还可以在研磨过程中添加部分醇。继续研磨直到类胡萝卜素晶体和亲水性保护胶体的平均粒度达到<5μm,优选<1μm。可以使用本领域技术人员已知的常规装置,例如胶体磨、球磨机如KDL或Multilab类型的Dynomill作为研磨装置。
可以将本发明配制剂直接加入含水或不含水制剂中。然而,它们也可以在加入前进一步用所用醇稀释。
所述配制剂尤其适合作为动物饲料添加剂和人类食品制剂添加剂或配合饲料添加剂,适合作为生产药品和化妆品制剂的组合物并适合生产人类和动物领域的膳食补充剂产品。悬浮液例如可优选通过在挤出时混入饲料颗粒或通过施加或喷雾至饲料颗粒上而用作家畜营养品中的饲料添加剂。特别通过例如如所谓的后造粒施加那样直接喷雾本发明配制剂而用作饲料添加剂。饲料颗粒优选在降低的压力下加载配制剂。
在人类食品领域的一般应用范围例如为饮料,乳制品如酸乳酪、牛奶饮料或牛奶冰和牛奶冻粉,蛋制品,烘烤混合料和糖果的维生素化和着色。在化妆品领域,可以将油性悬浮液用于例如装饰性个人护理组合物,例如以面霜、乳液的形式,或作为唇膏或彩妆。
本发明还涉及包含本发明配制剂的膳食补充剂、动物饲料、人类食品和药品及化妆品制剂。优选加载有配制剂的动物饲料,尤其是饲料颗粒。
膳食补充剂产品和药品制剂尤其指包含本发明配制剂的片剂、包覆片剂及优选硬和软明胶胶囊。
以下实施例用于解释本发明而不限制本发明。
实施例1
生产10.5%浓度的β-胡萝卜素分散体
将784.16g甘油引入2L玻璃烧杯中并搅拌加入110.84g(水含量5.27%)Purity Gum 2000(辛烯基琥珀酸酯-淀粉)。随后,搅拌加入105.0g晶体β-胡萝卜素直到完全润湿(30-40分钟)。然后将由此获得的分散体用来自Siegmund Lindner的Dynomill Multilab使用钇稳定且直径为0.4-0.6mm的SiLi珠Zr珠研磨。研磨容器装有400ml这些研磨珠。在54-70℃的温度、0.10mm的研磨狭缝和2986rpm的磨机转速下进行研磨。
实施例2
制备7.0%浓度的β-胡萝卜素分散体
通过实施例1描述的方法研磨210.0g晶体β-胡萝卜素、221.68g PurityGum 2000(水含量5.27%)和2568.32g甘油的分散体。在52-68℃的温度下进行研磨。1.5小时后,获得其中β-胡萝卜素粒度为438nm的低粘度分散体。

8.根据权利要求7的配制剂,其中所述改性淀粉衍生物为淀粉酯,特别是淀粉辛烯基琥珀酸酯。
9.根据上述权利要求中任一项的配制剂,其包含0-40重量%的水。
10.根据上述权利要求中任一项的配制剂,其中所述类胡萝卜素为虾青素。
11.根据权利要求10的配制剂,其中所述多元醇为甘油。
12.根据权利要求1-11中任一项的配制剂作为动物饲料、人类食品和膳食补充剂及药品和化妆品组合物的添加剂的用途。
13.包含根据权利要求1-11中任一项的配制剂的动物饲料、人类食品或膳食补充剂。
