- 权 利 要 求1. 一种检测 HCV抗体的试剂盒, 该试剂盒以 "支持物-第一抗原-待 测 HCV抗体-笫二抗原-标记物-可识别信号" 形式完成检测, 其中第二抗 原与标记物之间进行一步或多步的结合反应, 其特征在于所述笫二抗原为 HCV蛋白与标签的结合物, 所述标签为肽类标签或非肽类标签。2. 权利要求 1 的试剂盒, 其中所述非肽类标签为化合物、 半抗原、 维 生素、 类固醇、 染料、 激素、 抗生素、 核酸或者为它们与肽或蛋白的组合 物。3.权利要求 2 的试剂盒, 其中所述化合物为二硝基苯酚或溴脱氧尿 苷; 维生素为生物素或其衍生物; 类固醇为地高辛; 染料为吖啶酯、 罗丹 明、 丹磺酰氯或荧光素。4. 权利要求 1的试剂盒, 其中所述肽类标签为含有 His Tag、 T7 Tag,S Tag、 Flag Tag, HA Tag或 HCV肽片段的肽或蛋白。5.权利要求 4的试剂盒, 其中所述肽类标签为外源蛋白时, 其分子量 小于 30KD。6.权利要求 5的试剂盒, 其中所述夕卜源蛋白分子量小于 20KD。7. 权利要求 6的试剂盒, 其中所述夕卜源蛋白分子量小于 15KD。8.权利要求 7的试剂盒, 其中所述外源蛋白分子量小于 10KD。9.权利要求 8的试剂盒, 其中所述外源蛋白分子量小于 5KD。10. 权利要求 5的试剂盒, 其中所述外源蛋白位于 HCV蛋白的 N端 时, 其分子量小于 5KD; 位于 HCV蛋白的 C端时, 其分子量小于 20KD。11. 权利要求 1-10任一项的试剂盒, 其中笫一抗原和笫二抗原包括 HCV NS3及 CORE蛋白区段, 所述 NS3蛋白区段为从 a.a.l201±5处起始 至 a.a.l465±8 处终止的任一区段, 该蛋白区段可以存在突变而保持其抗原 性不变; 所述 CORE蛋白区段为含有 a.a.10-17全序列的区段, 进一步为含 有 a.a.7-21全序列的区段, 更进一步为含有 a.a.6-23全序列的区段的区段, 而且不包括 a.a.29-90全序列的区段, 进一步为不包括 a.a.29-70全序列的区 段, 再进一步为不包括 a.a.29-59全序列的区段, 更进一步为不包括 a.a.32- 48 全序列的区段, 该蛋白区段可以存在突变而保持其抗原性和结合能力不 变。12. 权利要求 1-10任一项的试剂盒, 其中笫一抗原的 CORE蛋白区 段为 a.a.2-59; 笫二抗原的 CORE蛋白区段为 a.a.1-28; 笫一抗原和笫二抗 原的 NS3蛋白区段均为 a.a.l201-1465。13. 权利要求 1-12任一项的试剂盒, 其中所述标签为生物素或其衍 生物、 生物素或其衍生物与肽或蛋白的结合物时, 其结合配偶体为亲和素、 链霉亲和素、 中性链亲和素、 它们的类似物、 抗生物素或其衍生物的抗体; 所述标签为肽或蛋白时, 其结合配偶体为抗该肽或蛋白的单克隆抗体或多 克隆抗体。14. 一种制备权利要求 1试剂盒的方法, 其中包括 HCV蛋白与标签 结合的步骤, 所述结合方法包括:1) HCV蛋白的生物素化;2) HCV蛋白与肽的嵌合表达。15. 权利要求 14的方法, 其中 HCV蛋白生物素化的方法包括:1)体内酶促生物素化;2)体外生物素化;3) 间接生物素化;4)氨基酸的生物素化。16. 权利要求 15的方法, 其中体内酶促生物素化的方法为:使编码 HCV蛋白与生物素接受蛋白的嵌合基因 X同编码生物素 -乙酰 辅酶 A羧化酶合成酶的基因 Y共同在宿主体内表达, 在表达宿主体内利用 生物素-乙酰辅酶 A羧化酶合成酶通过体内酶促反应使嵌合基因. X编码蛋 白的特定赖氨酸结合上生物素, 具体方法包括:1) 多顺反子表达;2) 共转化表达;3) 多启动子表达; 4)单顺反子表达;5)与宿主的生物素-乙酰辅酶 A羧化酶合成酶基因共表达。17. 权利要求 15 的方法, 其中体外生物素化为通过偶联活化生物素 或体外酶促反应来完成对 HCV蛋白的生物素化。18. 权利要求 16 的方法, 其中所述生物素接受蛋白为生物素羧基载 体蛋白家族或其功能区段、 SEQ ID N0.3多肽或其功能类似物。19. 权利要求 16 的方法, 其中所述生物素-乙酰辅酶 A羧化晦合成 酶是分类号为 EC6.3.4.15的酶或其功能区段、 功能类似物。
