Abstract
本发明的目的在于提供即使用于口腔用组合物和饮食品也没有安全性的问题,而且有效地促进脱钙了的牙釉质的再钙化的同时,藉此积极地抑制龋蚀的再钙化促进剂和含有该促进剂的口腔用组合物及饮食品。本发明由将含有半乳糖酐的红藻类和/或其提取物作为有效成分的牙釉质的再钙化促进剂和含有该促进剂的口腔用组合物及饮食品构成。
Description
技术领域
本发明涉及牙釉质的再钙化促进剂和含有该促进剂的口腔用组合物及饮食品。
背景技术
通常,龋蚀开始于变形链球菌(Streptococcus mutans)和远缘链球菌(Streptococcus sobrinus)等口腔内链球菌(龋齿菌)附着于牙齿表面,由于这些细菌所具有的葡萄糖苷转移酶的作用而产生葡聚糖,形成蚀斑(牙垢)。该蚀斑中,由于上述细菌代谢食物残渣中的砂糖和淀粉等而生成的酸使牙釉质脱钙,即形成所谓的初期的龋蚀状态。
另一方面,唾液中存在钙和磷酸盐,它们通过将上述脱钙部分修复,即再钙化,从而起到使牙齿恢复原状的作用。即,在牙齿表面,一直发生脱钙和再钙化这两种相反的现象,通常保持所需的平衡。但是,如果该平衡中的蚀斑增大,则向脱钙的一边倾斜,发生龋蚀。
构成牙齿表面的釉质的结晶由作为六方晶系的羟磷石灰Ca10(PO4)6OH2的磷酸钙构成。初期的龋蚀中所确认的脱钙是牙釉质无机成分的溶解,再钙化可以实现溶解残留的已有的磷酸钙结晶的修复和再生长。
以往,为了防止龋蚀,开发有针对龋齿菌的牙齿附着抑制剂、抗菌剂或抑制龋齿菌的葡聚糖形成的葡萄糖苷转移酶抑制剂等。但是,例如抗菌剂并非仅对龋齿菌表现出抗菌作用的特异性材料,安全性存在问题,葡萄糖苷转移酶抑制剂存在容易受到唾液产生的影响的问题。
此外,已知掺入具有与牙齿的无机成分类似的结晶结构的羟磷石灰和氟化物,将牙齿脱钙表层部再钙化的龋齿预防组合物(例如参照专利文献1),但将氟化钠、氟磷酸钠或氟化亚锡等氟化物掺入口腔用组合物和饮食品中的方法从安全性的角度来看存在问题。
另外,已知通过组合羟磷石灰的微粒和木糖醇使用来将脱钙的牙釉质再钙化的技术方案(例如参照专利文献2),但工业化制造的羟磷石灰是化学上稳定的化合物,因此缺乏反应性,在严格意义上其结晶结构与人体中的构成牙齿的羟磷石灰不同,所以再钙化的效果不足。
另外,还已知包含液状化磷酸钙化合物的口腔用组合物(例如参照专利文献3),但是仅依靠磷酸钙,再钙化的效果不足。
此外,还已知包含海藻类、特别是红藻类的隐丝藻目的海萝的牙釉质的再钙化促进剂(例如参照专利文献4、5),但牙釉质的再钙化促进效果的稳定性根据海藻的种类存在差异,再钙化的效果不足。即,红藻类中含有半乳糖酐的红藻类和/或其提取物,例如选自κ-角叉菜聚糖(carrageenan)、ι-角叉菜聚糖和帚叉藻聚糖(furcellaran)的至少1种多糖类特别具有牙釉质的再钙化促进效果,对于这一点过去完全是未知的。

发明的揭示
鉴于上述情况,本发明的目的在于提供即使用于口腔用组合物和饮食品也没有安全性的问题,而且有效地促进脱钙了的牙釉质的再钙化的同时,藉此积极地抑制龋蚀的再钙化促进剂和含有该促进剂的口腔用组合物及饮食品。
本发明人反复认真研究后结果发现,红藻类中含有半乳糖酐的红藻类和/或其提取物可实现上述目的,从而完成了本发明。
即,本发明的牙釉质的再钙化促进剂将含有半乳糖酐的红藻类和/或其提取物作为有效成分,或者将含有半乳糖酐的红藻类和/或其提取物、木糖醇以及磷酸氢钙作为有效成分。作为上述半乳糖酐,较好是选自κ-角叉菜聚糖、ι-角叉菜聚糖和帚叉藻聚糖的至少1种多糖类,特别是κ-角叉菜聚糖可发挥显著的效果。
即,本发明涉及将上述的含有半乳糖酐的红藻类和/或其提取物、根据需要采用的木糖醇和/或磷酸氢钙作为有效成分的再钙化促进剂和含有该促进剂的口腔用组合物及饮食品,包括附带增强牙齿的再钙化的内容的表示的口腔用组合物及饮食品。
本发明的牙釉质的再钙化促进剂将含有半乳糖酐的红藻类和/或其提取物作为有效成分,或者将含有半乳糖酐的红藻类和/或其提取物、木糖醇以及磷酸氢钙作为有效成分,即使用于口腔用组合物和饮食品也没有安全性的问题,而且有效地促进脱钙了的牙釉质的再钙化的同时,可以藉此积极地抑制龋蚀。
特别是并用上述含有半乳糖酐的红藻类的提取物、木糖醇以及磷酸氢钙时的效果非常显著,所述提取物为选自κ-角叉菜聚糖、ι-角叉菜聚糖和帚叉藻聚糖的至少1种多糖类,特别是κ-角叉菜聚糖。即,基于含有半乳糖酐的红藻类的提取物(选自κ-角叉菜聚糖、ι-角叉菜聚糖和帚叉藻聚糖的至少1种多糖类,特别是κ-角叉菜聚糖)的自表层的强再钙化和基于木糖醇的自深层的再钙化相叠加,与不含半乳糖酐的红藻类相比,牙釉质的再钙化促进效果得到非常显著的促进。
实施发明的最佳方式
作为本发明的含半乳糖酐的红藻类,可以从红毛菜目、紫球藻目、角毛藻目、弯枝藻目、海索面目、石花菜目、隐丝藻目、杉藻目、掌藻目或仙菜目等适当选择使用,特别是含有选自κ-角叉菜聚糖、ι-角叉菜聚糖和帚叉藻聚糖的至少1种多糖类的杉藻科的角叉菜、线形软刺藻和红翎菜科的麒麟菜发挥显著的效果,是理想的。
此外,作为本发明的含半乳糖酐的红藻类,当然可以单独使用进行所需的干燥处理后切细或粉末化的制品、获得提取物后将其粒状化或粉末化的制品等,也可以将它们并用。另外,虽然并不被预期具有含半乳糖酐的红藻类的程度的效果,但也可以使用其它海藻类。
作为本发明的红藻类的提取物,帚叉藻聚糖、κ-角叉菜聚糖、ι-角叉菜聚糖是有效的,对于获得红藻类的提取物的方法没有特别限定,例如使用水或有机溶剂,特别是具有与水的相溶性的有机溶剂进行提取。其提取物可以进一步通过有机溶剂、色谱法等进行分离纯化后进行使用。
另外,上述红藻类或其提取物也可以溶解或分散于适当的液体载体,或者与适当的粉末载体混合或吸附于其上,根据情况添加乳化剂、分散剂、悬浮剂、展开剂、浸透剂、湿润剂或稳定剂等,制剂化为乳剂、水合剂、粉剂或片剂等后进行使用。
上述红藻类和/或其提取物即使仅将其用于口腔用组合物或饮食品,也可以充分促进牙釉质的再钙化,通过再并用木糖醇和/或磷酸氢钙,可以显著地促进该再钙化。
本发明的牙釉质的再钙化促进剂中,红藻类和/或其提取物、木糖醇和磷酸氢钙的配比较好是0.02~5.0∶50∶0.02~2.0,更好是0.05~0.2∶50∶0.1~0.4。
作为上述再钙化促进剂、即红藻类和/或其提取物或者再含有木糖醇和/或磷酸氢钙的口腔用组合物,可以例举牙膏、牙粉或洁牙液等洁牙剂类,漱口剂,牙龈按摩软膏,漱口用片剂或锭剂等。此外,作为饮食品,可以例举口香糖,硬糖、压片糖、软糖、巧克力、饼干或点心等零食,冰淇淋、雪糕或冰点心等冷冻食品,饮料,面包,烤饼,乳制品,火腿、香肠等畜肉制品类,鱼糕、鱼卷等鱼肉制品,副食品,布丁,汤或果酱等。
通过在如上所述的日常使用的口腔用组合物和日常食用的饮食品中掺入本发明的牙釉质的再钙化促进剂,可以日常食用再钙化促进剂,能够容易且轻松地进行龋齿的预防。此外,通过在口腔用组合物和饮食品中掺入再钙化促进剂,再钙化促进剂在口中的停留时间变长,带来再钙化促进剂在口腔中大范围地扩散等进一步的效果,因此是特别理想的。具体来说,通过在锭剂、口香糖、硬糖、压片糖、软糖、巧克力、冰淇淋、雪糕、冰点心、牙膏、牙粉、牙龈按摩软膏中掺入再钙化促进剂,再钙化促进剂在口中的停留时间变长,是非常理想的。此外,在其它食品中掺入再钙化促进剂时,再钙化促进剂含于残存在齿间的食品残渣中,是非常理想的。此外,通过在饮料、汤、洁牙液、漱口剂中掺入再钙化促进剂,再钙化促进剂遍布整个口腔中(牙齿的间隙等),因此是非常理想。
作为红藻类和/或其提取物向口腔用组合物或饮食品的添加量,较好是0.01~10.0重量%。此外,与红藻类和/或其提取物并用的木糖醇、磷酸氢钙的添加量根据采用的口腔用组合物或饮食品的种类和形态等而不同,难以一概而论,但分别较好是1~95重量%和0.01%~5.0%。
本发明中,作为将红藻类和/或其提取物或者木糖醇、磷酸氢钙添加到口腔用组合物或饮食品中的方法,可以在该制品的制造过程的任意阶段添加,或者与剩余的原料混合。此外,并用红藻类和/或其提取物以及木糖醇、磷酸氢钙的情况下,可以将它们预先混合后添加到该口腔用组合物或饮食品中,也可以分别进行添加。
实施例
以下,对于本发明的实施例和试验例进行说明,但本发明的范围并不受到这些例子的限定。
试验例1(本发明的牙釉质的再钙化促进剂的效果)
确认本发明的牙釉质的再钙化促进剂的效果的试验以牙科学报Vol.89,No.9,1441~1455(1989).中记载的基于使用人拔除牙的再钙化促进效果确认试验的方法作为参考如下进行。另外,作为含有半乳糖酐的红藻多糖类的提取物,使用旭东化学产业公司制的κ-角叉菜聚糖、ι-角叉菜聚糖、帚叉藻聚糖以及λ-角叉菜聚糖。
对人拔除牙的釉质块的表面整体以粘蜡被覆并留下3×4mm的窗口,将其在加温至50℃的0.01M乙酸·乙酸钠缓冲液(pH4.0)中浸渍2天,形成脱钙层(参照图1)。然后,将窗口的一半以蜡被覆,制成试验用牙釉质块。
再钙化处理中,使用含有1mM CaCl2、0.6mM KH2PO4、100mM NaCl的以50mM KOH溶液调制至pH7.3的再钙化溶液制备以下的A)~J)这10种溶液,将其调至37℃,并且分别浸渍各1个试验用牙釉质块2周。其中,各溶液每隔2天更换新的溶液。

再钙化处理后,除去各试验用牙釉质块的蜡,用聚酯树脂(Regolac树脂)包埋,制成100μm的研磨切片,拍摄接触式显微放射照片(CMR)。摄影条件为10kV、3mA,照射时间为30分钟,同时以铝箔阶式光楔作为基准进行摄影。显影以通常的方法进行。
此外,通过观察以以下的5级评价作为基于显微放射照相法(MR)的结果的再钙化度。
再钙化度0)釉质脱钙层中未确认到再钙化。
再钙化度1)釉质脱钙表层中确认少量再钙化。
再钙化度2)釉质脱钙表层中确认较强的再钙化。或者釉质脱钙表层和深层中确认再钙化。
再钙化度3)从釉质脱钙表层至深层都确认再钙化。
再钙化度4)从釉质脱钙表层至深层都确认强的再钙化。
刚使试验用牙釉质块形成脱钙层后的显微放射照相法(MR)的结果和将同一块再钙化处理后的显微放射照相法(MR)的结果示于图1~5。
图1为刚脱钙处理后的MR,未确认到再钙化(再钙化度0)。图2为采用A)的再钙化溶液处理后的MR,脱钙面整体确认缓和的再钙化(再钙化度1)。图3为采用B)的再钙化溶液处理后的MR,不仅釉质脱钙表层,深层也确认再钙化(再钙化度2)。图4为采用C)的再钙化溶液处理后的MR,釉质脱钙表层确认强的再钙化(再钙化度2)。图5为采用D)的再钙化溶液处理后的MR,釉质脱钙表层和深层都确认强的再钙化(再钙化度4)。图6为采用E)的再钙化溶液处理后的MR,脱钙面整体确认缓和的再钙化(再钙化度1)。图7为采用F)的再钙化溶液处理后的MR,不仅釉质脱钙表层,深层也确认再钙化(再钙化度2)。图8为采用G)的再钙化溶液处理后的MR,釉质脱钙表层确认强的再钙化(再钙化度2)。图9为采用H)的再钙化溶液处理后的MR,釉质脱钙表层和深层都确认强的再钙化(再钙化度4)。图10为采用I)的再钙化溶液处理后的MR,釉质脱钙表层确认强的再钙化(再钙化度2)。图11为采用J)的再钙化溶液处理后的MR,釉质脱钙表层和深层都确认强的再钙化(再钙化度4)。
如上所示,确认与使用红藻类提取物中不含半乳糖酐的多糖的情况相比,含有半乳糖酐的多糖具有显著的再钙化促进效果,而且通过在其中并用木糖醇、磷酸氢钙,其再钙化促进效果显著提升。
试验例2(本发明的饮食品(口香糖)的牙釉质的再钙化促进效果)
分别作为含有半乳糖酐的红藻多糖类的提取物使用κ-角叉菜聚糖、ι-角叉菜聚糖、帚叉藻聚糖,并且使用木糖醇和磷酸氢钙,制成基于表1的配比的实施例1~3(分别添加0.1%κ-角叉菜聚糖、ι-角叉菜聚糖、帚叉藻聚糖)和比较例1(未添加红藻提取物)、比较例2(添加0.1%λ-角叉菜聚糖)的口香糖。

实施例1~3和比较例1、2的口香糖的再钙化效果通过以下的方法进行评价。口香糖的有效成分的提取操作以“用于综合评价食品和代用糖的龋蚀诱发性的基础研究(课题编号04304045)平成5年度科学研究费辅助金研究成果报告书(研究代表人山田正);p86~89”作为参考进行。
对于上述实施例1~3和比较例1、2的各口香糖,将其切成细片,称量10g。分别在其中加入50ml与试验例1的情况相同组成的再钙化溶液(60℃),用玻棒充分打碎,溶出含有成分,再向其中加入50ml上述再钙化溶液(60℃),再次进行溶出操作后,通过离心分离去除细碎的口香糖基质,得到对应于实施例1~3和比较例1、2的各口香糖的5种口香糖提取液。
在这5种口香糖提取液中分别于37℃浸渍各3个试验用牙釉质块2周。其间,每隔2天更换新的口香糖提取液。接着,对于各试验用牙釉质块的基于显微放射照相法(MR)的结果,与再钙化促进效果试验1同样地评价通过观察以5级评价作为的再钙化度,基于上述实施例1~3和比较例1、2的再钙化度以各3个试验用牙釉质块的平均值算出,图表化示于图12。
比较例1的口香糖(未添加红藻提取物)的再钙化度为1.67,而实施例1的口香糖(添加0.1%κ-角叉菜聚糖)的再钙化度为2.67,实施例2的口香糖(添加0.1%ι-角叉菜聚糖)的再钙化度为2.33,实施例3的口香糖(添加0.1%帚叉藻聚糖)的再钙化度为2.00。此外,比较例2的口香糖(添加0.1%λ-角叉菜聚糖)的再钙化度为1.67。由该结果可知,与使用不具有半乳糖酐的红藻多糖类的情况下相比,掺入了具有半乳糖酐的红藻多糖类的口香糖显示出再钙化促进效果。
试验例3(本发明的饮食品(压片糖)的牙釉质的再钙化促进效果)
分别作为含有半乳糖酐的红藻多糖类的提取物使用κ-角叉菜聚糖、ι-角叉菜聚糖、帚叉藻聚糖,并且使用木糖醇和磷酸氢钙,制成基于表2的配比的实施例4~6(分别添加0.1%κ-角叉菜聚糖、ι-角叉菜聚糖、帚叉藻聚糖)和比较例3(未添加红藻提取物)、比较例4(添加0.1%λ-角叉菜聚糖)的压片糖。

实施例4~6和比较例3、4的压片糖的再钙化效果通过与试验例2同样的方法进行评价。接着,对于各试验用牙釉质块的基于显微放射照相法(MR)的结果,与再钙化促进效果试验1同样地评价通过观察以5级评价再钙化度,基于上述实施例4~6和比较例3、4的再钙化度以各3个试验用牙釉质块的平均值算出,图表化示于图13。
比较例3的压片糖(未添加红藻提取物)的再钙化度为1.67,而实施例4的压片糖(添加0.1%κ-角叉菜聚糖)的再钙化度为2.67,实施例5的压片糖(添加0.1%ι-角叉菜聚糖)的再钙化度为2.00,实施例6的压片糖(添加0.1%帚叉藻聚糖)的再钙化度为2.33。此外,比较例4的压片糖(添加0.1%λ-角叉菜聚糖)的再钙化度为1.67。由该结果可知,与使用不具有半乳糖酐的红藻多糖类的情况下相比,掺入了具有半乳糖酐的红藻多糖类的压片糖显示出再钙化促进效果。
以下,作为本发明的口腔用组合物或饮食品的实施例,示例实施例7~20的配比(数字为重量%)。另外,作为红藻多糖类,制成分别使用κ-角叉菜聚糖、ι-角叉菜聚糖和帚叉藻聚糖这三种的制品。

Claims (7)
1.牙釉质的再钙化促进剂,其特征在于,将含有半乳糖酐的红藻类和/或其提取物作为有效成分。
2.牙釉质的再钙化促进剂,其特征在于,将含有半乳糖酐的红藻类和/或其提取物以及木糖醇和/或磷酸氢钙作为有效成分。
3.如权利要求1或2所述的牙釉质的再钙化促进剂,其特征在于,含有半乳糖酐的红藻类的提取物为选自κ-角叉菜聚糖、ι-角叉菜聚糖和帚叉藻聚糖的至少1种多糖类。
4.口腔用组合物,其特征在于,包含权利要求1~3中的任一项所述的牙釉质的再钙化促进剂。
5.如权利要求4所述的口腔用组合物,其特征在于,附带增强牙齿的再钙化的内容的表示。
6.饮食品,其特征在于,包含权利要求1~3中的任一项所述的牙釉质的再钙化促进剂。
7.如权利要求6所述的饮食品,其特征在于,附带增强牙齿的再钙化的内容的表示。
