PATENTANSPRÜCHE 1. Verfahren zur Herstellung von Aralkylaminen der Formel
R1—CH2—N—R2 R3
(I)
und deren Säureadditionssalze ein- oder mehrbasischer physiologisch verträglicher Säuren durch Umsetzung von A Alkyl-oder Arylsulfonyloxy- bzw. -halogenverbindungen und B Aminen, wobei a) Komponente A der Formel R1-CH2-X (II) und Komponente B der Formel Hrjf-R2 (III) oder

b) Komponente A der Formel X-R2 (IV) und Komponente B der Formel R1-CH2—NH (V) oder
R3
c) Komponente A der Formel X—R3 (VI) und Komponente B der Formel R'-CHa-N-R2 (VII)

entspricht, wobei in den Formeln
R1 ein substituierter Phenylrest ist, der als Substituenten 2 bis 4 gleiche oder verschiedene Alkylreste mit je 1 bis 4, zusammen jedoch nicht mehr als 6 Kohlenstoffatomen, wobei etwa in p-Stellung stehende Alkyl-Substituenten von wenigstens einem anderen Alkylrest durch ein Ring-C-Atom getrennt sind, oder 1 oder 2 Halogenatome oder Nitrogruppen enthält, wobei etwa vorhandene Fluor-Substituenten in p-Stel-lung angeordnet sind und, wenn 2 Halogen-Substituenten vorhanden sind, diese nur in o-Stellung angeordnet sind, wobei die p- und o-Stellung jeweils auf die substituierte Aminome-thylengruppe bezogen sind,
R2 ein verzweigter oder unverzweigter Alkylrest mit 4 bis 10 Kohlenstoffatomen, jedoch mindestens 4 in einer Kette, ist, wobei die Kette in Form einer ununterbrochenen oder einer durch ein Äthersauerstoffatom unterbrochenen und/oder einer durch eine Hydroxylgruppe substituierten Alkylkette vorliegt, und wobei a) etwaige Verzweigungen in /3-Stellung zum N-Atom nur zusammen mit mindestens einer weiteren Verzweigung vorliegen,
b) die Alkylkette höchstens drei Verzweigungen enthält,
c) ein alleiniger Alkylrest in a-Stellung zum N-Atom stets Methyl ist, und
R3 ein Wasserstoffatom oder ein Alkylrest mit 1 bis 4 Kohlenstoffatomen ist, der mit einer Dialkylphosphinylgruppe der Formel
Phenylrest ist, der durch drei Alkylreste in 2,4,6-Stellung mit zusammen nicht mehr als 6 Kohlenstoffatomen substituiert ist.
3. Verfahren nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass solche Verbindungen umgesetzt werden, in
5 denen R2 ein unsubstituierter Alkylrest mit 5 bis 8, vorzugsweise bis 7 C-Atomen ist, der frei von Sauerstoffunterbrechungen ist.
4. Verfahren nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass solche Verbindungen umgesetzt werden, in io denen R2 in Nachbarstellung zum N-Atom eine Methylgruppe aufweist.
5. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass bei der Umsetzung a) eine Verbindung der Formel II, worin X eine Alkyl- oder Arylsulfonyloxygruppe bedeutet, in
15 Gegenwart eines inerten Lösungsmittels umgesetzt wird.
6. Verfahren nach Ansprüchen 1 und 5, dadurch gekennzeichnet, dass die Umsetzung a) in Gegenwart eines Säureakzeptors der Gruppe Alkalicarbonat, tert. Amine oder einem mindestens äquimolaren Überschuss des Amins der Formel III
20 erfolgt.
7. Anwendung des Verfahrens gemäss Anspruch 1 auf Verbindungen der Formel

8. Anwendung gemäss Anspruch 7, dadurch gekennzeich-40 net, dass R1 ein Phenylrest ist, der durch drei Alkylreste in
2,4,6-Stellung mit zusammen nicht mehr als 6 Kohlenstoffatomen substituiert ist.
9. Anwendung gemäss Anspruch 7 oder 8, dadurch gekennzeichnet, dass R2 ein unsubstituierter Alkylrest mit 5 bis 8, vorzugsweise bis 7 C-Atomen ist, der frei von Sauerstoffunterbrechungen ist.
10. Anwendung gemäss Anspruch 7 oder 8, dadurch gekennzeichnet, dass R2 in Nachbarstellung zum N-Atom eine Methylgruppe aufweist.

2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass solche Verbindungen umgesetzt werden, in denen Ri ein
55 Substituierte Hexylamine, wie das 2-Isopropylamino-6-me-thylheptan und das N-2-Cyclohexyläthyl-hexylamin, sind schon als Arzneimittel mit antihistaminischer bzw. vasodila-tierender Wirkung in Form ihrer Hydrochloride eingesetzt worden.
60 Die reduktive Alkylierung von Aminen, z. B. mit einem Überschuss von Natriumcyanoborhydrid und Formaldehyd in Acetonitril, mit Formaldehyd und Ameisensäure oder mit Formaldehyd, Wasserstoff und Palladiumkohle in Gegenwart von Wasser, ist bekannt, ebenso die Umsetzung von Amin mit 65 Natrium in Pyridin und die Äthylierung der Amingruppe mit Äthylen.
Es wurde nun ein Verfahren zur Herstellung von Aralkyi-aminen der Formel

R2 ein verzweigter oder unverzweigter Alkylrest mit 4 bis 10 Kohlenstoffatomen, jedoch mindestens 4 in einer Kette, ist, wobei die Kette in Form einer ununterbrochenen oder einer durch ein Äthersauerstoffatom unterbrochenen und/oder einer durch eine Hydroxylgruppe substituierten Alkylkette vorliegt und wobei a) etwaige Verzweigungen in ß -Stellung zum N-Atom nur zusammen mit mindestens einer weiteren Verzweigung vorliegen,
b) die Alkylkette höchstens drei Verzweigungen enthält,
c) ein alleiniger Alkylrest in a-Stellung zum N-Atom stets Methyl ist, und
R3 ein Wasserstoffatom oder ein Alkylrest mit 1 bis 4 Kohlenstoffatomen ist, der mit einer Dialkylphosphinylgruppe der Formel

substituiert sein kann, worin R4 und R5 je einen Alkylrest mit zusammen 2 bis 6 Kohlenstoffatomen bedeuten, R3 jedoch bei Methode c) kein Wasserstoffatom ist, und
X ein Halogen oder eine Alkylsulfonyloxy- oder Arylsul-fonyloxygruppe bedeutet, und dass die erhaltenen Verbindungen als solche isoliert werden und dass man die erhaltenen Verbindungen gegebenenfalls mit physiologisch verträglichen Säuren zu Säureadditionsverbindungen umsetzt.
Durch das erfindungsgemässe Verfahren und die so hergestellten therapeutisch wirksamen Amine lässt sich die Basis an substituierten Aminen für therapeutische Zwecke erweitern.
Zweckmässig ist in den erfindungsgemäss hergestellten Verbindungen R1 ein Phenylrest, der durch 3 Alkylreste in ortho-, para-Stellüng substituiert ist.
Der Rest R2 ist vorteilhaft ein substituierter Alkylrest mit 5 bis 8, vorzugsweise bis 7 C-Atomen, der frei von Sauerstoffunterbrechungen ist. Dieser Alkylrest R2 kann auch in Nachbarstellung zum Stickstoffatom eine Methylgruppe aufweisen, s Vielfach sind solche Verbindungen besonders geeignet, in denen R2 ein reiner, jedoch verzweigter Alkylrest mit höchstens drei Verzweigungen ist, in dem die Alkylsubstituenten benachbart oder durch 1, 3, 4 oder 5 weitere C-Atome der Hauptkette voneinander getrennt sind. Sehr gut eignen sich io auch Verbindungen, in denen R2 eine unverzweigte Alkylkette mit 4 bis 6 oder 8 bis 10 C-Atomen ist, die durch ein Äthersauerstoffatom unterbrochen ist. Auch Verbindungen, in denen R2 ein verzweigter Alkylrest ist, der durch ein Äthersauerstoffatom unterbrochen ist, haben gute Eigenschaften. 15 Bei den erfindungsgemäss hergestellten Aralkylaminen handelt es sich um bisher nicht beschriebene Verbindungen, die als solche oder zweckmässig in Form von Additionsverbindungen physiologisch verträglicher Säuren für therapeutische Zwecke eingesetzt werden können. Erfindungsgemäss herge-20 stellte Verbindungen zeigen gefässtonisierende, kapillarabdichtende und/oder antiarhythmische Wirkungen. Physiologisch verträgliche Säuren sind z. B. Mineralsäuren, wie Salzsäure, Salpetersäure, Phosphorsäure, Schwefelsäure, Sulfonsäuren, wie Cyclohexylsulfaminsäure, Carbonsäuren, wie Gluconsäu-25 re, Zitronensäure, Maleinsäure oder Adipinsäure.
Geeignete Verbindungen der Formel II sind 3,4,5-Trime-thylchlorid, 2,4,6-Trimethylbenzylchlorid, 2,3,5,6-Tetrame-thylbenzylbromid, 4-tert.-Butyl-2,6-dimethyl-benzyljodid,
3-tert.-Butyl-6-methyl-benzylchlorid, 4-Chlorbenzylchlorid, 30 3-Chlor-benzylchlorid, 2-Chlor-benzylchlorid, 2,6-Dichlor-
benzylchlorid, 3-Brombenzylchlorid, 2-Brombenzylchlorid,
4-Nitrobenzylchlorid, 2,4,6-Trimethylbenzyltoluolsulfonsäu-reester oder 4-Fluorbenzylchlorid.
Geeignete Verbindungen der Formel IV sind z. B. 1-, 2-35 oder 3-Chlor-, -Brom- oder -Jod-pentan, -hexan, -heptan, -octan, -nonan, -decan, l-Chlor-4-methyl-pentan, l-Chlor-2-äthyl-hexan, 2-Chlor-3-methyl-pentan, 2-Jod-6-methyl-hep-tan, 2-Chlor-5-hydroxy-5-methyl-heptan, 2-Brom-2,4,4-tri-methylpentan, 3-Chlor-propyl-äthyläther, 3-Chlor-propyl-bu-40 tyläther, 3-Chlor-propyl-hexyläther, 3-Chlor-propyl-isopropyl-äther und Heptyl(2)-toluolsulfonsäureester.
Als Verbindungen der Formel VI können z. B. Methyljo-did, Propylbromid, n- oder iso- Butyl-chlorid und Dimethyl-phosphinylmethylchlorid eingesetzt werden.
45 Geeignete Amine der Formel III sind z. B. die verschiedenen 1-, 2- oder 3-Aminopentane, -Aminohexane, -Aminohep-tane, -Aminooctane, -Aminononane, -Aminodecane, wie die normalen Verbindungen oder die folgenden verzweigten Verbindungen: 1-, 2-oder 5-Amino-4-methyl-pentane, 1-Ami-50 no-2-äthyl-hexane, 2-Amino-3-methyl-pentane, 2-Amino-5-methyl-hexane, 2-Amino-4-methyl-hexane, 2-Amino-6-me-thyl-heptane, 2-Amino-5-hydroxy-5-methyl-heptane, 2-Ami-no-2,4,4-trimethyl-pentane, 3-Aminopropyl-äthyl-äther, 3-Aminopropyl-hexyl-äther, 3-Aminopropyl-isopropyl-äther, 55 2-Methyl-amino-heptan, 2-Propylamino-heptan, 2-Dimethyl-phosphinylmethyl-aminoheptan, 2-Amino-n-nonan, 2-Ami-no-n-decan, 3-Amino-n-octan, 2-Amino-7-methyl-octan, 2-Amino-3,6-dimethyl-heptan.
Als Amine der Formel V werden beispielsweise genannt: so 3,4,5-Trimethyl-benzyl-amin, 2,4,6-Trimethyl-benzylamin, 2,3,5,6-Tetramethyl-benzylamin, 4-tert.-Butyl-2,6-dimethyl-benzylamin, 3-tert.-Butyl-6-methylbenzylamin, 4-Fluorbenzyl-amin, 4-Chlorbenzylamin, 3-Chlorbenzylamin, 2-Chlorbenzyl-amin, 2,6-Dichlorbenzylamin, 3-Brombenzylamin, 2-Brom-65 benzylamin, 4-Nitrobenzylamin, aber auch die entsprechenden sekundären Amine, die am Stickstoffatom z. B. mit einem Methyl-, Äthyl-, Propyl-, Dimethylphosphinylmethyl- oder Dimethylphosphinyl-propylrest substituiert sind.

Ein Amin der Formel VII ist z. B. 2-(2,4,6-Trimethyl-benzyl-amino)-heptan.
Sofern die genannten Amine mindestens ein asymmetrisches Kohlenstoffatom aufweisen, lassen sie sich auch in Form ihrer Razemate oder ihrer optisch aktiven Isomeren einsetzen.
Die Umsetzung gemäss Arbeitsweisen a) bis c) können zweckmässig in Lösungsmitteln erfolgen. Nach Arbeitsweise a) sind gegenüber den Reaktionsteilnehmern inerte Lösungsmittel, wie Alkohole mit 1 bis 4 Kohlenstoffatomen, aromatische Kohlenwasserstoffe, wie Benzol, Toluol, Xylo], Mesitylen oder Dimethylformamid, zweckmässig. Dies ist insbesondere bei der Umsetzung in Gegenwart von Alkylestern von Mineralsäuren oder organischen Sulfonsäuren der Fall. Mit Vorteil setzt man hierbei in Gegenwart eines Säureakzeptors, z. B. einer mindestens äquimolaren Menge mindestens eines Alkalicarbonats oder eines tertiären Amins bzw. in Gegenwart eines mindestens äquimolaren Überschusses der Amine der Formeln III, V oder VII um. Im allgemeinen arbeitet man, insbesondere bei den Arbeitsweisen a) bis c), bei Temperaturen zwischen 50 und 160° C, vorzugsweise bei 75 bis 135° C. Benutzt man z. B. Dimethylformamid als Lösungsmittel, ist zwar kein Säureakzeptor, aber meist eine Temperatur von 70 bis 160° C nötig.
Als Alkalicarbonat-Säureakzeptoren kommen insbesondere z. B. Natriumhydrogencarbonat, Natriumcarbonat oder Kaliumcarbonat in Frage. Aus der Reihe der tertiären Amine haben sich Diisopropylamin, Tributylamin, Triäthylamin, Pico-lin und Pyridin vor allem bewährt. Im Falle der Umsetzung eines Aralkylchlorids kann die Reaktion durch einen geringen Zusatz von Natriumjodid beschleunigt werden.
Wenn Alkyl-, insbesondere Methyl- und/oder Äthylamine der Formel I nach Ausführungsform c) hergestellt werden sollen, können die üblichen Synthesewege benutzt werden.
Verbindungen der Formel VII werden durch Hydrierung einer Azomethinverbindung der Formel oder der Formel

erhalten, worin
R8 in Formel IX ein Alkylidenrest mit einem Wasserstoffatom weniger als der ununterbrochene und unsubstituierte Alkylrest R2, aber sonst mit derselben Bedeutung wie R2 ist.
Hierfür kommt vorzugsweise die an sich bekannte Methode der reduktiven Alkylierung in Frage (siehe «Organic Reactions, Vol. IV, Seiten 174 ff). Bei dieser katalytischen Wasserstoffanlagerung werden als Katalysatoren z. B. Platin, Palladium, Nickel und Kobalt bevorzugt. Auch Reduktionen mit Natrium oder Natriumamalgam in Alkohol, Zink in Essigsäure und elektrolytisch an der Blei- oder Kupferkathode können durchgeführt werden.
Die Stabilität der erfindungsgemäss erhaltenen Verbindungen, die gewöhnlich kristallin sind, erlaubt die Herstellung von Arzneimittelzubereitungen für die Human- und Veterinärmedizin für orale, parenterale, rectale und äusserliche Verabreichung. Die Herstellung dieser Zubereitungen kann nach der üblichen Praxis unter Zumischen passender und verträglicher, fester oder flüssiger Hilfe- oder Trägerstoffe, wie Stärke, Milchzucker, Cellulosederivate, Stearinsäure oder ihrer Salze, Lösungsmittel, Lösungsvermittler, Zäpfchenmasse, Chloriden, Phosphaten und Carbonaten, z. B. Natriumcarbonat, erfolgen, und zwar in an sich bekannter Weise zu Pulvern, Tabletten Dragees, Kapseln, Lösungen, Pasten oder Suspensionen. Oral, z. B. als Tabletten, Dragees oder Pulver, können die erfin-
dungsgemässen Verbindungen beispielsweise in einer Dosis von 25 bis 50 mg jeweils 2 bis 3 Mal täglich verabreicht werden. Als Injektion oder bevorzugt als Dauerinfusion kann man sie z. B. in einer Dosis von 10 bis 20 mg pro Tag anwenden, s Wie die pharmakologischen Versuche zeigten, handelt es sich bei den beanspruchten Verbindungen um Gefässtonus regulierende Substanzen. Diese Verbindungen erhöhen den Venentonus, ohne einen Spasmus auszulösen, erweitern die Blutgefässe, setzen die erhöhte Permeabilität der Gefässe io herab und entfalten antiödematöse und analgetische Wirkungen. Das genannte Wirkungsspektrum sowie die gute Verträglichkeit macht die erfindungsgemässen Substanzen als gefäss-aktive Mittel besonders interessant und den bisher bekannten Standardpräparaten überlegen.

Beispiele
1. 2-(2,3,5,6-Tetramethyl-benzyl)-aminoheptan-hydrochlorid (siehe Formelverzeichnis Nr. 1)
36,5 g (0,2 Mol) 2,3,5,6-Tetramethyl-benzylchlorid lässt 20 man 2 Stunden mit 46,0 g (0,4 Mol) 2-Aminoheptan in 100 ml siedendem Benzol reagieren. Darauf wird das Lösungsmittel im Vakuum entfernt. Zur Freisetzung der Base gibt man darauf zum Rückstand eine Lösung von 8 g (0,2 Mol) Natriumhydroxyd in Methanol. Das Reaktionsprodukt lässt sich vom 25 ausgefallenen Natriumchlorid mittels einer Filternutsche und von 2-Aminoheptan mittels Vakuumdestillation befreien. Zur Überführung in das gewünschte Hydrochlorid behandelt man das verbleibende 2-(2,3,5,6-Tetramethyl-benzyl)-aminoheptan mit ätherischer Salzsäure und kristallisiert aus Methanol/Essig-30 ester um. Der Schmelzpunkt des Hydrochlorids liegt bei 155° C. Ausbeute: 55,4 g (93% der Theorie).
2. 2-(2,4,6-Trimethyl-benzyl)-aminoheptan-hydrochlorid
(siehe Formelverzeichnis Nr. 2)
Die Lösung von 168,7 g (1 Mol) 2,4,5-Trimethylbenzyl-chlorid in 260 ml Toluol wird mit einer Lösung von 230,4 g (2 Mol) 2-Aminoheptan in 250 ml Toluol versetzt und das Reaktionsgemisch 2 Stunden am Rückfluss gehalten. Nach dem Abdampfen des Toluols im Vakuum wird der Eindampfrückstand mit einer Lösung von 50 g Natriumhydroxyd in 500 ml Wasser gut vermischt und die organische Phase mit Methylenchlorid ausgeschüttelt. Die Methylenchlorid-Lösung wird über wasserfreiem Natriumsulfat getrocknet und im Vakuum eingeengt. Der Eindampfrückstand ergibt bei der Vakuum-45 fraktionierung neben 115 g 2-Aminoheptan 236 g (95,5% der Theorie) 2-(2,4,6-Trimethyl-benzyl)-aminoheptan vom Siedepunkt 175-178° C (bei 12 Torr). Durch Behandlung der Base mit ätherischer Salzsäure erhält man das gewünschte 2-(2,4,6-Trimethyl-benzyI)-aminoheptan-hydrochIorid, dessen 50 Kristalle nach der Umkristallisation aus Methylenchlorid/Äther (1:1) einen Schmelzpunkt von 167° C zeigen. Bei der Umsetzung kann 1 Mol 2-Aminoheptan durch 1 Mol einer inerten tertiären Base, wie Triäthylamin, Tributylamin oder Diisopropylamin, ersetzt werden.

3. (2-Dimethylphosphinylmethyl)-2,4,6-trimethylbenzyl)-amino-heptan-hydrochlorid (siehe Formelverzeichnis Nr. 30)
a) 12,4 g (0,05 Mol) 2-(2,4,6-Trimethyl-benzyl)-amino-heptan (Beispiel 2) werden mit 6,4 g (0,05 Mol) Chlorme-thyl-dimethyl-phosphinoxyd in einer Mischung von 100 ml Toluol und 50 ml Dimethylformamid in Gegenwart von 7,5 g Triäthylamin bei einer Badtemperatur von 135° C bis zum chromatographisch kontrollierten Ende der Reaktion umgesetzt. Nach Abtrennung von gebildetem Triäthylamin-hy-drochlorid, restlichem Triäthylamin und Lösungsmittel wird das Reaktionsprodukt in Äthylacetat aufgenommen. Durch Zugabe von einem geringen Überschuss einer ätherischen Salzsäurelösung wird (2-Dimethylphosphinylmethyl)-(2,4,6-trimethylbenzyl)-amino-heptan-hydrochlorid in kristalliner Form erhalten.
Schmelzpunkt: 123° C, Ausbeute: 17,2 g (92% der Theorie).
b) 10,3 g (0,05 Mol) 2-(Dimethylphosphinylmethyl)-ami-noheptan werden analog a) mit 8,5 g (0,05 Mol) 2,4,6-Trime-thylbenzylchlorid und c) 12,0 g (0,05 Mol) Dimethylphosphinylmethyl-(2,4,6-trimethylbenzyl)-amin werden mit 9,0 g (0,05 Mol) 2-Brom-heptan analog a) umgesetzt.
Die nach a), b) und c) erhaltenen Hydrochloride sind identisch.
Pharmakologische Prüfung Die therapeutische Wirksamkeit der erfindungsgemäss erhaltenen Substanzen ergibt sich aus folgenden Versuchen.
1. Isolierter Venen- und Aortenstreifen Die Versuche wurden an isolierten Venen- und Aortenstreifen von Kaninchen nach der abgewandelten Methode von Furchgott et al. [J. Pharmacol. exp. Ther. 108 (1953), Seite 129] vorgenommen. Die Ergebnisse sind aus Tabellen 1 und 2 ersichtlich. Dabei wurde die Kontraktion des Venenstreifens in Adrenalin und die des Aortenstreifens in Noradrenalin mit einer Menge von jeweils 10"® g/ml mit einem Wert von 100 zugrundegelegt.
2. Isolierte Rattenpfortader Die Versuche an der Rattenpfortader, einem Organ mit Eigenrhythmik, wurden unter Berücksichtigung der Methode von Berti et al. [Arch. int. Pharmacodyn. 184 (1970), Seite 328} durchgeführt. In Tabelle 3 ist die Tonussteigerung in Milligramm in Abhängigkeit von der Adrenalinkonzentration angegeben. Zum Vergleich sind die Werte für die bekannte Substanz Heptaminol (= 6-Amino-2-methylheptan-2-ol) mit angegeben.
3. Durchflussänderung am isolierten Kaninchenohr Die Versuche wurden nach Krakow-Pissemski [Pflügers Archiv 151 (1913), Seite 583 und 156 (1914), Seite 426] durchgeführt. In Tabelle 4a sind die ermittelten Werte für die gefässverengende Wirkung in Ringer-Lösung und in Tabelle 4b für die gefässerweiternde Wirkung in Ringer-Norfenefrin-Lösung in Abhängigkeit von der Konzentration der jeweiligen Lösung zusammengestellt. Zum Vergleich wurden die mit dem bekannten Präparat Äescin-Natrium ermittelten Werte angegeben.
4. Relative Coronardurchflussänderung am isolierten Meerschweinchenherz nach Langendorff Nach diesem bekannten Verfahren [Pflügers Archiv 61 (1895), Seite 219] wurden die Werte im Vergleich zu den
Ergebnissen mit dem bekannten Heptaminol in Tabelle 5 zusammengesstellt.
5 5. Histamin-Quaddel-Test
Der Test nach Halpern [Presse médicale 65 (1949), Seite 949] wurde modifiziert und dabei die kapillarpermeabilitäts-hemmende Wirkung der erfindungsgemässen Verbindungen geprüft. Die Blaufärbung an 2 intracutanen Histamin-Injek-io tionsstellen pro Tier, ausgelöst durch i.v. verabreichtes Trypan-blau (2 Minuten vor Histamin), wird nach der folgenden Wertskala beurteilt:

20 Die vorstehende Bewertung ergibt sich aus der Summe der beiden Injektionsstellen.
Die Prüfsubstanzen werden 30 Minuten vor der Histamin-lnjektion i.p. verabreicht. Die Ergebnisse im Vergleich zu
25 Äscin-Natrium zeigt Tabelle 6. Die Vergleichssubstanz Äs-cin-Natrium wurde wegen ihrer optimalen Wirksamkeit 16 Stunden vor der Injektion verabreicht, um vergleichbare Ergebnisse zu erhalten.

6. Toxizität an der Maus
Die Toxizität wurde nach Litchfield und Wilcoxon [J. Pharmacol. exp. Ther. 97 (1949), Seite 399] an der Maus bestimmt. Im Vergleich dazu wurden die Werte mit dem bekann-
35 ten Äscin ermittelt. Die Ergebnisse sind aus Tabelle 7 ersichtlich. Die vergleichende Prüfung mit jeweils 0,1 %iger Lösung auf Venen- und Augenverträglichkeit am Kaninchen führte zu gleichsinnigen Ergebnissen.

7. Hemmung des Carrageenin-Pfotenödems
Nach diesem Test wurden entzündungshemmende und antiödematöse Effekte am Carrageenin- und Serotonin-Pfoten-ödem an der Ratte nach Siegmund et al. [Oroc. Soc. exp. Biol.
45 Med. 95 (1957), Seite 729] nachgewiesen. Die Werte sind in Prozent, bezogen auf die Werte der scheinbehandelten Kontrollversuche, angegeben. Dabei wurde die antiphlogistische Wirkung der erfindungsgemässen Substanzen geprüft. Die Ergebnisse zeigt Tabelle 8.

8. Writhing-Test
Nach dieser Prüfung [siehe Winter et al. Proc. Soc. exp. Biol. Med. 111 (1962), Seite 544] wurde die Hemmung der
55 Schleifbewegungen an der Maus beobachtet, um etwaige analgetische Wirkungen der erfindungsgemässen Verbindungen festzustellen. Die Ergebnisse sind in Tabelle 9 zusammengestellt.

9. Hot-plate-Test
Dieser Test wurde nach Chen und Beckman [siehe Science 113 (1951), Seite 631] an der Maus durchgeführt.
50 mg/kg der Verbindung Nr. 2 (vergleiche Formelver-
65 zeichnis) per os verabreicht, verlängern die Reaktionszeit der Tiere um 70%. Im Vergleich dazu wurde für die Vergleichssubstanz Aminophenazon für die effektive Dosis von 59%> (ED 50) eine erfolderliche Menge von 84 mg/kg ermittelt.

Diskussion der Ergebnisse Nach Tabellen 1 und 2 zeigen die erfindungsgemäss erhaltenen Verbindungen, insbesondere Verbindung 26 in Tabelle 1 und Verbindung 27 in Tabelle 2, stärkere vasotonisierende Effekte als das Vergleichspräparat Ascin-Natrium.
Die Ergebnisse nach Tabelle 3 stehen in Ubereinstimmung mit den Befunden in den vorhergehenden Tabellen. An der isolierten Pfortader entfalten die genannten Substanzen — insbesondere Verbindungen 2 und 22 — eine deutlich stärkere Tonussteigerung als die Vergleichssubstanz Heptaminol.
Besonders günstige Ergebnisse im Vergleich zum Äscin-Natrium sind die Effekte am perfundierten Kaninchenohr, vgl. Tabelle 4. So zeigen die erfindungsgemäss hergestellten Verbindungen nach Tabelle 4a keine Gefässspasmen. Äscin-Na-trium löst dagegen bereits mit 10-5 g/ml Gefässstenosen bis nahe zum Verschluss aus (siehe Tabelle 4a).
An Organen mit tonisierenden Gefässen erweitern die genannten Verbindungen konzentrationsabhängig die Gefässe, und zwar sowohl am mit Ringer-Norfenefrin-Lösung perfundierten Kaninchenohr (siehe Tabelle 4b) als auch am perfundierten Meerschweinchenherz (siehe Tabelle 5). Im Gegensatz dazu verstärkt die Vergleichssubstanz Heptaminol am Kaninchenohr die Ringer-Norfenefrin-Verengung (siehe Tabelle 4b)
und zeigt am Meerschweinchenherz keine Wirkung (siehe Tabelle 5). Die Vergleichssubstanz Äscin-Natrium zeigt am tonisierten Kaninchenohr (siehe Tabelle 4b) keine Wirkung und zeigt am Meerschweinchenherz Coronargefäss- veren-50 gende Effekte.
Wie der Histamin-Quaddel-Test nach Tabelle 6 zeigt, bewirken die hier zusammengestellten Verbindungen eine deutlich bessere Hemmung der durch Histamin bedingten Kapillarpermeabilitätssteigerung als Äscin-Natrium. 55 Die Toxizität der erfindungsgemäss erhaltenen Substanz an der Maus (siehe Tabelle 7) ist deutlich günstiger als die von Äscin-Natrium. Analoge Ergebnisse zeigen auch Versuche mit Kaninchen.
Etwaige entzündungshemmende und antiödematöse Ef-60 fekte der genannten Substanzen ergeben sich aus den Versuchen am Carrageenin- und Serotonin-Pfotenödem. Die hierbei, insbesondere anhand von Tabelle 8, nachgewiesenen Effekte sowie die ermittelten analgetischen Wirkungen der Substanz bei Writhing-Test (siehe Tabelle 9) und im Hot-pla-65 te-Test machen das Wirkungsspektrum der Substanzen, insbesondere von solchen mit gefässerweiternder Wirkung, besonders interessant.
