<Desc/Clms Page number 4>

<Desc/Clms Page number 6>
<Desc/Clms Page number 7>

The main patent discloses the preparation of novel 4,5-epoxy-3-ketosteroids serving as a starting point for the synthesis of novel steroids substituted in position 4, which have very interesting pharmacological activity and which are useful as intermediates for the synthesis of 'other steroids substituted in position 4.
In order to prepare the 4,5-epoxy-3-ketositoids used as a starting point in the main patent, a 3-keto 4-steroid dissolved in an organic solvent, preferably miscible with water, is reacted. water, such as ethanol, methanol, etc ... with hydrogen peroxide in an alkaline medium. By this reaction, 4,5-epoxy epimeric bata and alpha are obtained, which are easily separated and purified by crystallization and chromatography.
In the preparation of 4,5-epoxy-3-keto-corticosteroids, that is to say compounds having an alpha-ketolic or dihydroxyacetone group at position 17 of the steroid nucleus, as shown by the formula:
EMI3.1
where R 'represents (H) COCH2OH (OH) COCH2OH; R "represented H2; (H) OH; = 0; the action of alkaline hydrogen peroxide not only causes the formation of 4,5-epoxide, but also the oxidation of the side chain, which results in a remarkable lowering of roundness.
<Desc / Clms Page number 4>
In the case of a keto group in position 17, together with the desired 4,5-epoxide, the epoxide of carboxylic acid-17 is also obtained, while in the case of a dihydroxyacetone group in position 17, the epoxides of ketone-17 and 17alpha-hydroxy-17-carboxylic acid are obtained, as shown by the following partial formulas:
EMI4.1
No useful effect was obtained using the normal esters of these steroids, such as acetate, propionate, benzoate, because these esters saponify quickly in alkaline solution.
It has now been found, which is the object of the present invention, that one can reliably protect the side chain of corticosteroids against alkalis and alkaline oxidizing agents by blocking the hydroxyl group at position 21 by virtue of etherification by chloride

<Desc / Clms Page number 6>
Due to their ease of formation or decomposition, triphenylmethyl ethers of corticosteroids are useful for preparing derivatives which can be more easily purified, and they are more stable than non-etherified compounds.
Another very important use of the triphenylmethyl ethers of corticosteroids is based on the fact that only the primary alcohols readily react with triphenylmethyl chloride, in the absence of excess chlorine.
In this way, one can protect a primary hydroxyl group by etherification and then the desired reactions can be carried out on the secondary alcohol.
For example, one can etherify 3beta-21-dihydroxy = ¯4-pregnene at position 21, and then oxidize it to obtain a 3-keto-¯4 derivative according to Oppenhauer:
EMI6.1
From this 3-keto-¯4 derivative, deoxy-corticosterono is obtained by separating the triphenylmethyl radical
Another example of the utility of these ethers is the following: from the compound S, 17alpha, 21-dihydorxy pre-genene-3,20-dione, the compound epi-F is prepared, oxidizing with the aid of Rhizopus nigricans 6227b, according to US Patent No. 2,602,769 applied for February 23, 1952.

After acidification, steam distillation and ordinary extraction, 1.6 g of crude 21-triphenylmethoxy-deoxycorticosterone are obtained. 1.6 of this product is refluxed with 30 cm3 of 80% acetic acid. The solution is poured into water and extracted with ethyl acetate; the ethyl acetate solution is treated with water, dried and distilled. The residue is acetylated with pyridine and acetic anhydride at room temperature for 16 hours. The solution is diluted with water and extracted with ethyl acetate.
The extract is washed with dilute hydrochloric acid and then with dilute alkali, and with water, it is dried and the solvents are removed. 1.6 g of crude product are obtained which, recrystallized by 'ether, gives 0.8 g of deoxycorticosterone acetate, melting point 150-155 c' the melting point of a mixture with a pure sample is not lowered.

S (l7alphaf 21-dih, ydroxy 4 pregnene.-3, 20-diono aida from Rhizopus nigricans 6227b according to US Pat. No. 2,602,769 already cited above, and etherified with an equimolar amount of chloride of triphenylmethyl in pyridine at room temperature for 7 days.
By proceeding as in Example 8, 0.8 g of 21-triphenylmethyl ether of the epi-F compound is obtained. This product is oxidized by treating it with a suspension of 0.4 g of CrO3 in 10 cm3 of pyridine, at room temperature, for 16 hours.
The reaction mixture is diluted with methylene chloride, washed with dilute alkali and with water, and finally dried. The solvent is evaporated off and the residue is fractionated by chromatography on Florisil. The fractions eluted with methylene chloride are recrystallized, with methanol distilled over sodium, and 0.5 g of 21 triphenylmethoxy-cortisone is obtained, point of melting point 223-226 C, which does not lower the melting point when mixed with a sample of the product prepared as in Example 2.
By decomposing the triphenylmethoxy-cortisone according to Example 3 and by acetylating with acetic anhydride in the usual way, one obtains, with a yield close to 100% cortisone acetate, melting point 240 c identical to a sample known.
