La présente invention est relative à des perfectionnements à la préparation de nouveaux composés stéroïdes, qui peuvent être représentés par la formule générale suivante :

Ces nouveaux composés de formate I constituent des intermédiaires précieux dans la synthèse de la cortisone et de substances Y apparentées à partir de l'hécogénine, à cause de la présence du groupe céto dans la pe- sition 11 et du fait que les noyaux-oxygénés à 5 et 6 éléments peuvent être aisément transformés en la chaine latérale en C17 de la cortisone et d'autres stérol:des d'importance physiologique.
L'invention concerne, dès lors, à titre de nouveaux composés, les composés de formule générale I donnée plus haut, dans laquelle R et X ont les significations indiquées.
On préfère le composé, dans lequel R est un groupe acétoxy et X du brome. L'invention concerne donc, à titre de nouveau composé stéroï-

de spécifique préféré, le 3 fi -acétoxf-23 bromo-12 ( -chloro 11-00-5 c( : 22a-spirostane.
Pour permettre une identification plus aisée de ce nouveau composé stéroïde préféré, on en donne ci-après certaines caractéristiques physiques :

Il est évident que les caractéristiques données ci-avant ont été obtenues pour la matière la plus pure que l'on a été à même de préparer jusqu'à présent et peuvent être sujettes à des variations, selon la pureté d'un échantillon particulier du composé.
On a constaté que les nouveaux composés suivant l'invention peuvent être préparés par oxydation du groupe 11-hydroxy d'un composé de formule générale :

L'invention concerne, dès lors, également un procédé pour la préparation des nouveaux composés de formule générale I, dans lequel un composé de formule générale II est traité en solution à l'aide d'un agent oxydant.
L'agent oxydant préféré pour exécuter le traitement est l'oxyde chromique,.: La réaction peut avantageusement être exécutée à une températu- re comprise entre 0 et 130 C.
La réaction peut être exécutée soit dans un système à une phase liquide, soit dans un système à deux phases liquides. Si la réaction doit avoir lieu dans un système à une seule phase liquide, on peut employer de l'acide chromique dans de l'acide acétique ou de la pyridine. Lorsqu'on utilise de l'acide acétique, on préfère exécuter la réaction en présence d'un tampon approprié, tel que l'acétate de sodium, à des températures élevées, par exemple à la température de l'acide acétique-bouillant (118 C).
Lorsqu'on utilise un système à deux phases liquides, on préfère utiliser une solution de chlorhydrine de formule II dans du benzène et une solution d'acide chromique dans de l'acide acétique aqueux.
Les matières de départ du présent procédé, à savoir les composés de formule générale II, peuvent être préparées de la manière décrite dans la demande de-brevet belge n 417.092 déposée le 27 août 1954 au nom de la demanderesse.
Le produit obtenu peut être séparéet purifié de toute manière convenable. Il peut, par exemple, être extrait à l'aide de benzène, évaporé jusqu'à si(cité,trituré avec du méthanol et séparé par filtration. On a, toutefois, constaté que ce produit peut aussi être converti, si on le désire, sans isolement préalable, en 11-oxo-tigogénine et ses 3-esters, ces com- posés constituant, comme on le sait déjà, des intermédiaires de valeur dans la synthèse de la cortisone et des substances-y apparentées. La ll-oxo-ti- gogénine et ses 3-esters peuvent être représentés par la formule de structure suivante :

L'invention concerne, dès lors, également un procédé pour la préparation de 11-oxo-tigogénine et ses 3-esters, dans lequel procédé un composé de formule générale I est déshalogéné, de préférence par chauffage avec du zinc et un acide aliphatique liquide, tel que l'acide acétique.
La 11-oxo-tigogénine ou son 3-ester ainsi préparé peut être iso- lé et purifié de toute manière convenable. Le zinc peut, par exemple, être séparé par filtration et le filtrat peut ensuite être dilue à l'eau, de ma- niére à obtenir le produit cristallin, qui est à son tour séparé par fil- tration et séché.
Pour que l'invention puisse être bien comprise, les exemples suivants sont donnés à seul titre illustratif.

On met du 3 fi -acétoxy-23-bromo-12 cé -chloro-11 /Î -hydroxy- 5 [alpha]: 22a-spirostane (2 gr) en suspension dans un mélange de benzène (20 ce), d'acide acétique-aqueux à 50 % (20 ce) et d'acide chromique 5N dans de 1' acide acétique (4 ce).
Le mélange est agité mécaniquement pendant 18 heures dans un flacon bouché. Pendant ce temps, la sapogénine passe en solution. La couche benzénique est séparée, lavée à l'eau et au bicarbonate de sodium aqueux, séchée sur sulfate de sodium et distillée jusqu'à siccité. On ajoute du 3 ss -acétoxy-23-bromo-12 [alpha] -chloro-11 ss -hydroxy- 5 [alpha] : 22a-spirostane (1 gr) dissous dans de la pyridine sèche (20 ce) à une suspension-de complexe CrO3/pyridine, obtenu en ajoutant du CrO3 (1 gr) à de la pyridine-sèche (20 ce). Le mélange est agité mécaniquement pendant 3 heures et est alors dilué avec de l'eau et extrait avec du benzène. L'exi- trait benzénique est lavé à l'eau, à l'acide chlorhydrique 2N et à l'eau, séché sur sulfate de sodium et distillé jusqu'à siccité. Le résidu est tri- turé avec-du méthanol (10 ce) et le produit cristallin est séparé par fil- tration et séché. 0,9 gr; P.F. 234-235 (décomposition); [[alpha]] D = -68 (CHCl3).
Exemple 3.
3 ss -acétoxy-11-oxo-5 [alpha] : 22a-spirostane On chauffe du 3 ss -acétoxy-23-bromo-12 [alpha] -chloro-11-oxo-5 [alpha] : 22a-spirostane (1 gr) au reflux dans de l'acide acétique glacial (15 ce) avec de la poudre de zinc (4 gr) et de l'acétate de¯sodium fondu (0,5 gr) pendant-1 1/2 heures. Le zinc est séparé par filtration et le filtrat est dilué à l'eau, en sorte qu'on obtient un produit cristallinincolore exempt d'halogène.¯ 0,70 gr.; P.F. : 217-220 ; [[alpha]] D = -40 (chloroforme).
Par chromatographie sur de l'alumine,-on obtient dans les éluats benzéni- ques une matière fondant à 224-225 , [[alpha]] D = -40 (CHCl3).
Exemple 4.
3 -acétoxy-23-bromo-12 [alpha] -chloro-11-oxo-5 [alpha]: 22a-spirostane.
On met du 3 ss -acétoxy-23-bromo-12 [alpha] -chloro-11 ss -hydroxy- 5 [alpha] : 22a-spirostane (10 gr) en suspension, en agitant, dans de l'acide acé- tique glacial (40 cc) contenant de l'acétate de sodium-fondu (5 gr). La sus- pension est chauffée à l'ébullition sous reflux, tandis qu'une solution d'oxy- -de chromique (4 gr) dans de l'acide acétique aqueux (4 ce d'eau, 3 ce d'aci- de acétique) est ajoutée en l'espace d'1/4 d'heure. La réaction est achevée par chauffage à l'ébullition et agitation pendant encore 3/4 d'heure. On -ajoute de l'eau (50 ce) au mélange réactionnel chaud et après refroidisse- ment à température ambiante, le produit est isolé par filtration.
Rendement : 9,3 gr (93 %); [[alpha]]D20 : -67,9 .
