<B>Procédé pour isoler un</B> cétostéroïde <B>à</B> partir <B>d'un mélange le contenant</B> La présente invention est relative à un pro cédé pour isoler un cétostéroïde à partir d'un mélange le contenant. Ce procédé s'applique en particulier à la purification et l'isolement de substances, telles que l'hormone femelle ou es- trone ou une hormone corticosurrénale comme la cortisone. On sait que les hormones stéroïdes sont fa briquées actuellement, soit par extraction de produits naturels complexes, soit par des syn thèses très laborieuses.
Aussi sait-on que, le plus souvent, un cétostéroïde recherché pour ses usages en médecine ou en art vétérinaire peut être souillé de substances présentant des caractères de solubilité presque identiques à ceux du produit principal, de sorte qu'on n'aboutit pas par cristallisations fractionnées à des résultats satisfaisants.
Un premier progrès a été obtenu en sépa rant les cétostéroïdes des substances non céto niques qui les accompagnent, par exemple par isolement, sous la forme de sa semi-carbazone, de la trans-déhydroandrostérone parmi les pro duits neutres d'oxydation du cholestérol, et par isolement de l'estrone des urines de jument, à l'aide des réactifs T et P de Girard et Sandu- lesco, qui permettent d'éngager les cétones en combinaisons hydrosolubles, sous forme d'hy- drazones à fonction ammonium quaternai?e. Cependant, les semi-carbazones,
pratiquement toutes insolubles, ne permettent, d'une façon générale, aucun fractionnement net des cétosté roïdes entre eux, alors que les réactifs de Girard et Sandulesco exigent des conditions fines de travail permettant une application res treinte seulement.
D'une manière générale d'ailleurs, les ré actifs cétoniques utilisés jusqu'à présent présen tent l'inconvénient de ne pas posséder une action sélective suffisante sur les divers cétos téroïdes ou bien encore ils donnent avec les cétostéroïdes des dérivés à partir desquels il est difficile de régénérer les cétostéroïdes. L'inven tion se propose de remédier à ces inconvé nients.
La titulaire a en effet découvert que l'hy- drazide correspondant à l'acide diphénylglyco- lique ou acide benzilique donne avec certains cétostéroïdes des hydrazones peu solubles qu'on peut séparer sélectivement de mélanges contenant ces cétostéroïdes et à partir desquel les il est aisé de régénérer ces cétostéroïdes.
L'hydrazide benzilique, qu'on désignera également dans la suite sous le nom de ré actif B , réagit avec les cétostéroïdes selon l'équation suivante

<tb> (hydrazone <SEP> <SEP> B <SEP> ) Le procédé selon l'invention consiste donc à faire réagir 1.'hydrazide benzilique sur ledit mélange contenant un cétostéroïde à isoler,
à séparer l'hydrazone benzilique dudit cétosté- roïde et à régénérer, à partir de cette hydra- zone, ledit cétostéroïde. La mise en réaction avec l'hydrazide peut avoir lieu, soit à chaud, soit à froid. Après séparation de l'hydrazone benzilique, il peut être avantageux de la puri fier par cristallisation ou solubilisation.
L'hydrazide benzilique est un produit bien connu et est obtenu selon la méthode de Cur- tius (Journ. prakt. Chem., 1917, [2] 95, p. 196).
Les hydrazones des cétostéroïdes sont pro duites, par exemple, en faisant réagir une solu tion dudit mélange contenant un cétostéroïde, dans un solvant à réaction neutre tel que le méthanol, l'isopropanol, le dioxane ou le ben zène, avec ledit hydrazide benzilique. Dans cer tains cas, il est à conseiller d'ajouter au mé lange une petite quantité d'un acide organique à faible poids moléculaire tel que l'acide acéti que, quia un effet catalytique sur la réaction qu'il accélère.
On peut aussi faire réagir l'hy- drazide et ledit mélange contenant un cétosté- roïde dans l'acide acétique pur. La réaction se produit à froid, après un contact plus ou moins prolongé des produits de réaction, ou en faisant chauffer le mélange réactionnel au reflux.
On a trouvé que ledit réactif B ne réagis sait pas du tout avec un certain nombre de stéroïdes usuels. Avec certains autres, il pro duit des hydrazones de faible solubilité dans le solvant choisi. En employant, par exemple, des mélanges de cétostéroïdes, il arrive très souvent qu'un seul cétostéroïde soit précipité sous la forme d'un hydrazone B, tandis que les autres ne réagissent pas, ou bien leurs produits de ré action sont solubles dans le solvant.
Après séparation par filtrage ou essorage, puis éventuellement recristallisation, l'hydra- zone B est clivée de telle sorte que le cétosté- roïde qui a réagi avec ledit réactif B est régé néré et récupéré. Selon l'hydrazone B spécifi que obtenue, cette régénération du cétostéroïde est obtenue par l'hydrolyse avec des acides mi néraux ou des acides organiques forts ou par double décomposition et échange avec un al déhyde ou une cétone possédant une plus grande affinité pour le réactif B que le céto- stéroïde combiné.
C'est pourquoi ledit aldé hyde ou la cétone est de préférence employé en excès, par rapport à la quantité nécessaire, afin de provoquer une rupture d'équilibre. Le benz- aldéhyde ou l'acide pyruvique, par exemple, conviennent particulièrement bien pour de telles réactions d'échange, le dernier composé étant employé avantageusement en solution dans l'acide acétique dilué.
Le réactif B fournit, par exemple, des hy- drazones cristallisées et peu solubles, avec les cétostéroïdes suivants : l'estrone, c'est-à-dire le 3-hydroxy-17-céto-A1,3.5(10)-estratriène ; la cis- testostérone, c'est-à-dire le 17a-hydroxy-3- céto-A4-androstène ; la nor-cholesténolone, c'est-à-dire le 3(p-hydroxy-25-céto-A5-27-nor- cholestène ;
les esters de désoxycorticostérone, spécialement le 21-acétoxy-3,20-dicéto-A4-pré- gnène ; les esters de la cortisone, spécialement le 17 a-hydroxy-21-acétoxy-3,11,20-tricéto-A4- prégnène. Dans des conditions de réaction tout à fait analogues, on n'observe, par exemple, au cune prépicipation avec les composés suivants la trans-déhydroandrostérone, c'est-à-dire le 3(3-hydroxy-17-céto-A5-androstène ; la A4-an- drostènedione-3,17 ;
la testostérone (trans), c'est-à-dire le 17(3-hydroxy-3-céto-A4-andro- stène ; la prégnénolone, c'est-à-dire le 3 (3- hydroxy-20-céto-A5-prégnène ; la progestérone, c'est-à-dire le 3,20-dicéto-A4-prégnène ; l'équi- line, c'est-à-dire le 3-hydroxy-17-céto-A1.3,5c10r7- estratétraène pour ne citer ici que les représen tants stéroïdes les mieux connus.
Il est très surprenant de constater qu'il existe d'aussi grandes différences de solubilité entre les hydrazones benziliques des cétosté roïdes, d'autant plus que les hydrazones déri vées d'acides très voisins comme l'acide mandé- lique et l'acide diphénylacétique ne présentent pas de telles différences de solubilité. Les hy- drazones mandéliques sont presque toutes solu bles dans les conditions de réaction habituelles lorsque l'on emploie le méthanol ou l'éthanol comme solvants.
Les hydrazones diphénylacétiques, d'autre part, ne présentent pas de différences de solu bilité ; presque toutes ces hydrazones sont in solubles de sorte qu'on ne peut faire aucune précipitation sélective, comme on le voit sur le tableau suivant

<SEP> -f- <SEP> -I- <SEP> - <SEP> - En conséquence, l'hydrazide benzilique per met d'effectuer la séparation du cétostéroïde envisagé, dilué dans une grande quantité d'au tres représentants sans valeur et/ou de purifier le stéroïde en question lorsqu'il est souillé de composés voisins avec lesquels il a tendance à syncristalliser.
Tel est, en premier lieu, le cas de l'isole ment de l'estrone dans des résidus de fabrica tion à hautes teneurs en équilénine et équiline. La solubilité de l'hydrazone B de l'estrone, dont la formule est la suivante

Ainsi,
lorsqu'on isole l'estrone de l'urine de jument gravide par la méthode connue qui con siste à extraire l'urine hydrolysée par un sol vant des lipides, à saponifier cet extrait et. à séparer de l'insaponifiable brut, par traitement aux réactifs T ou P de Girard, la fraction céto nique, celle-ci donne, après fractionnement par la soude pour séparer les cétophénols, dont fait partie l'estrone, un mélange de cristaux constitués d'estrone, d'équipne et d'équilénine. Après une ou deux recristallisations,
le mélange cède la majeure partie de l'estrone sous forme pure, mais une partie de l'hormone demeure dans les- eaux-mères sous forme de mélange quasi inextricable avec les deux satellites, équi- line et équilénine. On peut séparer de ce mé lange la majeure partie de l'équilénine à l'état de picrate G. Sandulesco, Wang, A. Girard, C. R. Acad. Sciences,-1933, <I>136,</I> 137, mais ce traitement néglige la récupération de l'estrone qui est cependant le constituant le plus pré cieux.
Or, même dans ce cas, on obtient encore une précipitation sélective de l'hormone femelle sous la forme de son hydrazone B. Les satelli tes de l'estrone, c'est-à-dire principalement l'équiline, forment des hydrazones qui, dans les mêmes conditions, ne précipitent que peu ou même ne précipitent absolument pas. On régé nère ensuite l'estrone par hydrolyse avec un rendement pratiquement quantitatif (Exemple 1).
Le procédé suivant l'invention trouve en core une autre application industrielle dans la préparation de la cortisone. Au cours de la syn thèse de cette hormone, on peut être conduit à oxyder le 11,20-dicéto-3u,17a-dihydroxy-21- acétoxy-prégnane en 3,11,20-tricéto-17a- hydroxy-21-acétoxy-prégnane, et il se trouve alors que de faibles quantités de produit non oxydé gênent considérablement les réactions suivantes. Là encore, la purification par re cristallisation est 'peu satisfaisante pour élimi ner l'impureté. Le passage par l'hydrazone B permet de résoudre facilemnt le problème.
Le 11,20 - dicéto - 3 a,17a - dihydroxy- 21- acétoxy - prégnane ne forme pas d'hydrazone B insolu ble en méthanol, alors que le dérivé du produit oxydé est remarquablement peu soluble (Exem ple 4).
D'autre part, dans la série de la cortisone comme dans la série estrogène, une seule liaison éthylénique dans la structure stéroïde apparaît suffisante pour modifier considérablement le caractère de solubilité des hydrazones benzili- ques. On a vu plus haut que l'hydrazone B de l'équiline était 50 fois plus soluble en méthanol que celle de l'estrone. De même, l'hydrazone B de l'acétate de cortisone de formule

D'un point de vue pratique, on peut égale ment choisir un solvant dans lequel les hy- drazones B formées seront toutes solubles, puis en précipiter l'une par addition d'un autre sol vant, ou encore distiller le solvant à sec, puis reprendre les hydrazones B restantes par un solvant dans lequel telle ou telle hydrazone B sera soluble ou insoluble.
Les exemples suivants illustrent l'inven tion. Comme solvants neutres, on peut utiliser, en lieu et place de ceux mentionnés dans les exemples, d'autres solvants équivalents tels que l'isopropanol, le dioxane, le benzène et, au lieu de l'acide acétique utilisé comme catalyseur, employer d'autres acides organiques tels que les acides formique ou tartrique, seuls ou en mé lange.
Les points de fusion figurant dans les exem ples sont des points de fusion instantanés déter minés au bloc de Maquenne. <I>Exemple 1</I> <I>Séparation</I> estrone-équiline 100 g d'un mélange de cétones contenant de l'estrone sont dissous, à chaud, dans 2 litres de méthanol et 80 cm3 d'acide acétique cristal- lisable. On ajoute 125 g de réactif B et fait bouillir à reflux pendant deux heures.
S'il s'agit d'un produit riche en estrone, l'hydrazone com mence à se séparer pendant la durée de l'ébul lition. S'il s'agit d'un produit pauvre, on amorce la cristallisation. On essore et lave au méthanol.
L'hydrazone obtenue est reprise par 2 litres de méthanol bouillant. Il s'agit ici d'une diges tion alcoolique et non d'une recristallisation. Après repos jusqu'au lendemain, on essore et on lave à plusieurs reprises avec du méthanol.
Le produit séché et contrôlé présente les cons tantes suivantes F = 169-170 C,<B>MD</B> = -I- 751, (c = 1 0/0, dioxane). L'hydrolyse de l'hydrazone fournit l'es- trone, F = 262- C, [a]D = -f- 163 (c = 1 o/o, dioxane), avec un rendement quantitatif.
<I>Exemple 2</I> <I>Séparation</I> cis-testostérone <I>et</I> trans-testosté- rone On traite, comme décrit à l'exemple 1, un mélange de trans- et cis-testostérone provenant des recristallisations de testostérone technique pesant 58 g.
On obtient 20 g de combinaison B de cis-testostérone. Les eaux-mères sont éva porées à sec et le résidu hydrolysé comme pré cédemment fournit 44 g de produit brut qui, par recristallisation, donnent 30,8 g de trans- testostérone. <I>Exemple 3</I> <I>Purification de l'acétate de cortisone</I> On traite 40 g d'acétate de cortisone impur, obtenus selon un quelconque des procédés classiques de synthèse de cette hormone, F = 2300 C,
par 30 g de réactif B dans 1000 cm3 d'alcool méthylique à 1 % d'acide acétique. Par refroidissement, on obtient l'hy- drazone de l'acétate de cortisone, F = 1960 C, [a]D = -I- 235 à 2400 (c = 1 o/o, chloroforme).
L'hydrolyse de l'hydrazone est effectuée de la façon suivante On traite 50 g d'hydrazone de l'acétate de cortisone par un mélange de 200 cm3 d'acide acétique glacial, 20 cms d'acide pyruvique et 10 cm3 d'eau. Les cristaux formés sont filtrés et la masse lavée avec une solution de bicarbo nate de sodium puis à l'eau. Après séchage, on obtient l'acétate de cortisone, F = 2470 C, [a]D = -f- 177o (c = 1 a/o, acétone).
<I>Exemple 4</I> <I>Purification du</I> 3,11,20-tricéto-17a-hydroxy-21- acétoxy-prégnane On traite par le réactif B, au reflux, du méthanol contenant 1 % d'acide acétique, 20 g de 3,11,
20-tricéto-17a-hydroxy-21-acétoxy- prégnane contenant comme impureté 20 % de 11,20- dicéto - 3 a,17at- dihydroxy-21- acétoxy - prégnane. Par refroidissement, on recueille 23 g de cristaux F = 210o C,
représentant en hydrazone 95 % du dérivé 3-carbonylé mis en jeu. La décomposition de l'hydrazone benzili- que effectuée selon l'exemple 3 fournit le<B>3,11,</B> 20-tricéto-17at-hydroxy-21-acétoxy-prégnane, F =<B>2310</B> C,<B>[ ID</B> = -I- 820 (c = 1 0/0, acétone).
<I>Exemple 5</I> <I>Séparation du</I> 3,11,20-tricéto-17a,21-dihydroxy- prégnane <I>et de l'acétate de cortisone</I> 43 g de résidu provenant de l'évaporation d'eaux-mères de cortisone qui renferment du 3,11,20-tricéto-17a,21-dihydroxy-prégnane sont acétylés par traitement à l'anhydride acétique et à la pyridine. Le produit réactionnel, sous forme de gomme jaune,
est dissous à l'ébulli- tion dans 300 cm3 de méthanol contenant 1 % d'acide acétique.
On ajoute ensuite une solution bouillante de 26 g de réactif B (excès de 10 0/0) dans 200 cm3 de méthanol à 1 % d'acide acé- tique et porte au reflux au bain-marie. La com binaison B cristallise rapidement. On aban donne au repos une nuit. On essore la masse cristallisée, lave au méthanol puis à l'acide chlorhydrique normal pour éliminer l'excès de réactif B, puis à l'eau, et sèche à 1200 C.
On obtient 45,8 g (68,5 %) de combinaison B fon- dant vers 200-2100 C.
On reprend ce mélange de combinaisons B (45,8 g) par 200 cm3 de dichloréthane. On tié dit au bain-marie pendant quelques minutes, puis refroidit à température ambiante et essore l'insoluble. On répète deux fois l'opération avec -200 cm3 puis 50 cm3 de dichloréthane. Après séchage, on obtient 26 g de combinaison B du 3,11,20-tricéto-17a-hydroxy-21-acétoxy- prégnane, soit 56,5 % du mélange,
F = 210n C.
Les dichloréthanes d'extraction sont réunis et distillés à sec sous vide. On obtient 18,5 g (40 0/0) de combinaison B de l'acétate de cor tisone cristallisant par addition d'acide acétique aqueux.
Par clivage des deux combinaisons B sui vant les exemples précédents, on récupère, d'une part, l'acétate de cortisone et, d'autre part, le 3,11,20-tricéto-17a-hydroxy-21- acétoxy-prégnane. De manière analogue, on peut se servir des hydrazones benziliques suivantes pour isoler les cétostéroïdes correspondants : hydrazones ben- ziliques de l'acétate de 11-désoxycorticosté- rone : plaquettes, F = 1430 C ; de la norchol- esténolone :
paillettes, F = 2551, C ; de l'an- drostane-17a-benzoyloxy-3-one : microcristaux, F = 1550 C ; de l'équiline, F = 182a C (solu ble dans le méthanol) ; de l'équilénine, F = 1700 C (soluble dans le méthanol).
<B> Method for isolating a </B> ketosteroid <B> from </B> from <B> a mixture containing it </B> The present invention relates to a process for isolating a ketosteroid from 'a mixture containing it. This process applies in particular to the purification and isolation of substances, such as the female hormone or estrone or an adrenal cortex hormone such as cortisone. We know that steroid hormones are currently manufactured, either by extracting complex natural products, or by very laborious synthesis.
Also we know that, most often, a ketosteroid sought for its uses in medicine or in veterinary art can be contaminated with substances presenting characteristics of solubility almost identical to those of the main product, so that one does not end up by fractional crystallizations with satisfactory results.
A first progress has been obtained by separating ketosteroids from the non-keto nic substances which accompany them, for example by isolation, in the form of its semi-carbazone, of trans-dehydroandrosterone from the neutral oxidation products of cholesterol, and by isolation of estrone from mare's urine, using T and P reagents from Girard and Sandu- lesco, which enable the ketones to be released in water-soluble combinations, in the form of hydrazones with a quaternary ammonium function ? e. However, semi-carbazons,
practically all insoluble, generally do not allow any net fractionation of the cetostraids between them, whereas the reagents of Girard and Sandulesco require fine working conditions allowing only a limited application.
In general, moreover, the ketone reagents used up to now have the drawback of not having sufficient selective action on the various keto teroids or else they give, with the ketosteroids, derivatives from which it is is difficult to regenerate from ketosteroids. The invention proposes to remedy these drawbacks.
The holder has in fact discovered that the hydrazide corresponding to diphenylglycolic acid or benzilic acid, together with certain ketosteroids, gives poorly soluble hydrazones which can be selectively separated from mixtures containing these ketosteroids and from which it is easy. to regenerate these ketosteroids.
Benzilic hydrazide, which will also be referred to below under the name of reagent B, reacts with ketosteroids according to the following equation

separating benzilic hydrazone from said ketosteroid and regenerating said ketosteroid from this hydra-zone. The reaction with the hydrazide can take place either hot or cold. After separation of the benzilic hydrazone, it may be advantageous to purify it by crystallization or solubilization.
Benzilic hydrazide is a well-known product and is obtained according to the method of Curtius (Journ. Prakt. Chem., 1917, [2] 95, p. 196).
The hydrazones of ketosteroids are produced, for example, by reacting a solution of said mixture containing a ketosteroid, in a neutral reacting solvent, such as methanol, isopropanol, dioxane or ben zene, with said benzilic hydrazide. In some cases it is advisable to add to the mixture a small amount of a low molecular weight organic acid such as acetic acid which has a catalytic effect on the reaction which it accelerates.
It is also possible to react the hydrazide and said mixture containing a ketosteroid in pure acetic acid. The reaction takes place in the cold, after more or less prolonged contact of the reaction products, or by heating the reaction mixture to reflux.
It has been found that said Reagent B does not react at all with a number of common steroids. With some others, it produces hydrazones of low solubility in the chosen solvent. Using, for example, mixtures of ketosteroids, it very often happens that only one ketosteroid is precipitated as hydrazone B, while the others do not react, or their reaction products are soluble in water. solvent.
After separation by filtering or draining, then optionally recrystallization, the hydra-zone B is cleaved in such a way that the ketosteroid which has reacted with said reagent B is regenerated and recovered. Depending on the specific hydrazone B obtained, this regeneration of the ketosteroid is obtained by hydrolysis with mineral acids or strong organic acids or by double decomposition and exchange with an aldehyde or a ketone having a greater affinity for the reagent. B than the combined keto-steroid.
This is why said aldehyde or ketone is preferably used in excess, relative to the amount necessary, in order to cause a disruption of equilibrium. Benzaldehyde or pyruvic acid, for example, are particularly suitable for such exchange reactions, the latter compound being advantageously employed in solution in dilute acetic acid.
Reagent B provides, for example, crystalline and sparingly soluble hy- drazones with the following ketosteroids: estrone, i.e. 3-hydroxy-17-keto-A1,3.5 (10) -estratriene ; cis-testosterone, i.e. 17α-hydroxy-3-keto-A4-androstene; nor-cholestenolone, i.e. 3 (p-hydroxy-25-keto-A5-27-nor-cholestene;
deoxycorticosterone esters, especially 21-acetoxy-3,20-diketo-A4-pregnene; esters of cortisone, especially 17α-hydroxy-21-acetoxy-3,11,20-triketo-A4-pregnene. Under completely analogous reaction conditions, for example, no prepicipation with the following compounds is observed at all trans-dehydroandrosterone, that is to say 3 (3-hydroxy-17-keto-A5- androstene, A4-an-drostenedione-3,17;
testosterone (trans), i.e. 17 (3-hydroxy-3-keto-A4-androstene; pregnenolone, i.e. 3 (3-hydroxy-20-keto -A5-Pregnene; progesterone, i.e. 3,20-diketo-A4-pregnene; equiline, i.e. 3-hydroxy-17-keto-A1. 3,5c10r7- estratetraene to name here only the best known steroid representatives.
It is very surprising to note that there are such large differences in solubility between the benzilic hydrazones of cetostraids, especially since the hydrazones derived from closely related acids such as mandelic acid and acid diphenylacetic do not exhibit such differences in solubility. Almost all of the mandelic hydrazones are soluble under the usual reaction conditions when methanol or ethanol are employed as solvents.
Diphenylacetic hydrazones, on the other hand, do not show differences in solubility; almost all of these hydrazones are insoluble so that no selective precipitation can be made, as seen in the following table

when estrone is isolated from the urine of a pregnant mare by the known method which consists in extracting the urine hydrolyzed by a lipid-containing sol, in saponifying this extract and. in separating from the crude unsaponifiable, by treatment with T or P reagents from Girard, the keto fraction, this gives, after fractionation with sodium hydroxide to separate the ketophenols, of which estrone is part, a mixture of crystals made up of estrone, equipne and equilenin. After one or two recrystallizations,
the mixture gives up most of the estrone in pure form, but some of the hormone remains in the mother liquors as an almost inextricable mixture with the two satellites, equine and equilenine. The major part of the equilenin in the picrate state can be separated from this mixture by G. Sandulesco, Wang, A. Girard, C. R. Acad. Sciences, -1933, <I> 136, </I> 137, but this treatment neglects the recovery of estrone which is however the most valuable constituent.
However, even in this case, we still obtain a selective precipitation of the female hormone in the form of its hydrazone B. The estrone satellites, that is to say mainly equilin, form hydrazones which , under the same conditions, precipitate little or even do not precipitate at all. The estrone is then regenerated by hydrolysis with a practically quantitative yield (Example 1).
The process according to the invention also finds another industrial application in the preparation of cortisone. During the synthesis of this hormone, it may be necessary to oxidize 11,20-diketo-3u, 17a-dihydroxy-21-acetoxy-pregnane to 3,11,20-triketo-17a-hydroxy-21-acetoxy -Pregnane, and it is then that small amounts of non-oxidized product considerably interfere with the following reactions. Again, purification by re-crystallization is unsatisfactory in removing the impurity. The passage through hydrazone B easily solves the problem.
11,20 - diketo - 3a, 17a - dihydroxy - 21 - acetoxy - pregnane does not form hydrazone B which is insoluble in methanol, whereas the derivative of the oxidized product is remarkably poorly soluble (Example 4).
On the other hand, in the cortisone series as in the estrogen series, a single ethylenic bond in the steroid structure appears sufficient to considerably modify the solubility character of the benzyl hydrazones. We have seen above that the hydrazone B of equiline was 50 times more soluble in methanol than that of estrone. Likewise, hydrazone B of cortisone acetate of formula

From a practical point of view, it is also possible to choose a solvent in which the hy- drazones B formed will all be soluble, then to precipitate one of them by adding another solvent, or to distill the solvent to dryness, then take up the remaining hydrazones B in a solvent in which such or such hydrazone B will be soluble or insoluble.
The following examples illustrate the invention. As neutral solvents, it is possible to use, instead of those mentioned in the examples, other equivalent solvents such as isopropanol, dioxane, benzene and, instead of acetic acid used as catalyst, to employ d other organic acids such as formic or tartaric acids, alone or in mixture.
The melting points shown in the examples are instantaneous melting points determined at the Maquenne block. <I> Example 1 </I> <I> Separation </I> estrone-equiline 100 g of a mixture of ketones containing estrone are dissolved, hot, in 2 liters of methanol and 80 cm3 of acid crystallizable acetic. 125 g of reagent B are added and the mixture is boiled under reflux for two hours.
If it is a product rich in estrone, the hydrazone begins to separate during the time of the boil. If it is a poor product, crystallization is initiated. It is filtered off and washed with methanol.
The hydrazone obtained is taken up in 2 liters of boiling methanol. This is an alcoholic digestion and not a recrystallization. After standing overnight, filtered and washed several times with methanol.
The dried and checked product exhibits the following constants F = 169-170 C, <B> MD </B> = -I- 751, (c = 1 0/0, dioxane). Hydrolysis of the hydrazone affords estrone, F = 262-C, [a] D = -f-163 (c = 1 o / o, dioxane), in quantitative yield.
<I> Example 2 </I> <I> Separation </I> cis-testosterone <I> and </I> trans-testosterone As described in example 1, a mixture of trans- and cis-testosterone from recrystallizations of technical testosterone weighing 58 g.
20 g of cis-testosterone combination B are obtained. The mother liquors are evaporated to dryness and the residue hydrolyzed as above gives 44 g of crude product which, on recrystallization, gives 30.8 g of trans-testosterone. <I> Example 3 </I> <I> Purification of cortisone acetate </I> 40 g of impure cortisone acetate, obtained by any of the conventional methods for the synthesis of this hormone, are treated, F = 2300 VS,
per 30 g of reagent B in 1000 cm3 of methyl alcohol at 1% acetic acid. On cooling, the hydrazone of cortisone acetate, F = 1960 C, [a] D = -I-235 to 2400 (c = 1 o / o, chloroform) is obtained.
The hydrolysis of the hydrazone is carried out as follows. 50 g of hydrazone of cortisone acetate are treated with a mixture of 200 cm3 of glacial acetic acid, 20 cm3 of pyruvic acid and 10 cm3 of water . The crystals formed are filtered off and the mass washed with sodium bicarbonate solution and then with water. After drying, cortisone acetate is obtained, F = 2470 C, [a] D = -f- 177o (c = 1 a / o, acetone).
<I> Example 4 </I> <I> Purification of </I> 3,11,20-triketo-17a-hydroxy-21-acetoxy-pregnane The reagent B is treated at reflux with methanol containing 1% acetic acid, 20 g of 3.11,
20-triketo-17a-hydroxy-21-acetoxy-pregnane containing as impurity 20% of 11,20-diketo-3a, 17at-dihydroxy-21-acetoxy-pregnane. By cooling, 23 g of crystals F = 210o C are collected,
representing 95% hydrazone of the 3-carbonyl derivative involved. The decomposition of the benzyl hydrazone carried out according to Example 3 provides <B> 3,11, </B> 20-tricéto-17at-hydroxy- 21-acetoxy-pregnan, F = <B> 2310 </B> C, <B> [ID </B> = -I- 820 (c = 1 0/0, acetone).
<I> Example 5 </I> <I> Separation of </I> 3,11,20-triketo-17a, 21-dihydroxy-pregnan <I> and of cortisone acetate </I> 43 g of residue resulting from the evaporation of cortisone mother liquors which contain 3,11,20-triketo-17a, 21-dihydroxy-pregnane are acetylated by treatment with acetic anhydride and pyridine. The reaction product, in the form of a yellow gum,
is dissolved at the boil in 300 cm3 of methanol containing 1% acetic acid.
A boiling solution of 26 g of reagent B (excess of 10%) in 200 cm3 of 1% methanol of acetic acid is then added and brought to reflux on a water bath. Combination B crystallizes quickly. We give up a night's rest. The crystallized mass is filtered off, washed with methanol and then with normal hydrochloric acid to remove the excess of reagent B, then with water, and dried at 1200 C.
45.8 g (68.5%) of combination B melting at about 200-2100 C. are obtained.
This mixture of combinations B (45.8 g) is taken up in 200 cm3 of dichloroethane. Tié said in a water bath for a few minutes, then cooled to room temperature and filtered off the insoluble material. The operation is repeated twice with -200 cm3 and then 50 cm3 of dichloroethane. After drying, 26 g of combination B of 3,11,20-triketo-17a-hydroxy-21-acetoxy-pregnane are obtained, ie 56.5% of the mixture,
F = 210n C.
The extraction dichloroethanes are combined and vacuum distilled to dryness. 18.5 g (40 0/0) of combination B of the crystallizing cor tisone acetate are obtained by addition of aqueous acetic acid.
By cleavage of the two combinations B following the preceding examples, one recovers, on the one hand, the acetate of cortisone and, on the other hand, the 3,11,20-triketo-17a-hydroxy-21-acetoxy-prégnane . Analogously, the following benzilic hydrazones can be used to isolate the corresponding ketosteroids: benzilic hydrazones of 11-deoxycorticosterone acetate: platelets, Mp = 1430 C; norchol-estenolone:
sequins, F = 2551, C; an-drostan-17a-benzoyloxy-3-one: microcrystals, Mp = 1550 C; equilin, Mp = 182a C (soluble in methanol); equilenin, Mp = 1700 C (soluble in methanol).
