"Sirop contenant de la N-acétyl-cystéine".
La présente invention est relative à une composition sous la forme d'un sirop contenant de la Nacétyl-cystéine comme ingrédient actif.
La N-acétyl-cystéine (en abrégé appelée ciaprès NAC) est un composé doué de plusieurs propriétés pharmacologiques intéressantes, faisant de celui-ci un médicament largement utilisé.
Dans le traitement des rhumes ou refroidissements, la NAC s'est révélée intéressante grâce à ses propriétés mucolytiques.
L'une des formes pharmaceutiques largement utilisée dans la thérapie des rhumes ou refroidissements est le sirop, qui se révèle également particulièrement approprié dans les applications pédiatriques.
La NAC est disponible sous plusieurs formes pharmaceutiques ; toutefois la réactivité élevée de la molécule, l'instabilité relative et l'odeur et le goût sulfureux caractéristiques rendent extrêmement incommode la réalisation de formes liquides pour une utilisation orale qui soient stables dans le temps et avec une bonne sapidité.
La préparation d'un sirop prévoit, pratiquement dans tous les cas, l'utilisation d'un disaccharide (sucrose) ou d'un simple sucre comme agent édulcorant et épaississant de la solution aqueuse du médicament. Toutefois, l'interaction de la NAC avec les sucres donne à la solution une coloration brune inacceptable et le titre en NAC diminue.

C'est ainsi qu'il n'est pas possible de laisser la NAC en solution pendant une longue période de temps en présence de sucrose.
D'une façon similaire, les réactions donnant une coloration brune et provoquant un mauvais goût se produisent également entre la NAC et les monosaccharides dont l'utilisation est décrite dans la demande de brevet français nO 2.631. 831 au nom de Calco Anstalt.
Dans la mesure des connaissances, la seule formulation de sirop contenant de la NAC sur le marché consiste en une composition à préparer au moment de l'utilisation par dissolution dans l'eau d'un granulat.
Après la préparation, le sirop extemporané doit être utilisé dans les 3 semaines.
On a constaté à présent d'une manière surprenante qu'il est possible de préparer un sirop de NAC stable dans le temps ayant un goût plaisant sans l'utilisation de sucres.
Par conséquent, le but de la présente invention est une composition sous la forme d'un sirop contenant (chaque composant par rapport à 100 ml) :

Il s'ensuit que la solution susmentionnée est stable dans une bouteille fermée sous du gaz inerte pendant au moins deux ans sous des conditions ambiantes.

Lorsque l'on ouvre la bouteille, la solution montre un goût plaisant et est exempte de mauvaises odeurs ; la NAC maintient le titre initial et le sirop ouvert se montre stable pendant au moins 5 semaines.
Par agent édulcorant, on veut dire une substance pouvant donner un goût sucré à la solution mais qui ne contient pas de sucres. Des exemples spécifiques d'agents édulcorants sont la saccharine, la saccharine de sodium et les cyclamates ou leurs mélanges.
La quantité d'agent édulcorant sera la quantité minimale suffisante pour donner un goût plaisant à la solution.
L'agent épaississant est choisi parmi la carboxyméthylcellulose de sodium, l'hydroxypropylméthyl- cellulose et leurs mélanges. D'une façon surprenante, ces substances se sont révélées compatibles avec la NAC et peuvent donner à la solution une épaisseur convenant pour la sapidité usuelle d'un sirop.
A ces trois composants de base (NAC, agent édulcorant et agent épaississant), on peut ajouter de l'eau jusqu'au volume choisi, en rectifiant éventuellement le pH résultant de telle sorte qu'il soit maintenu dans l'intervalle de 5 à 8. Avantageusement, le pH sera de 6,5 à 7.
Le sirop pourrait également, mais non nécessairement, contenir un agent aromatisant pour améliorer la sapidité, en particulier dans le cadre d'une utilisation pédiatrique.
Puisque le sirop est d'une manière générale fabriqué en l'absence d'asepsie, la pharmacopée requiert la présence obligatoire d'un agent de conservation. L'agent de conservation est en outre nécessaire pour assurer la qualité microbiologique pendant la durée d'utilisation par le patient.

Dans ce cas, on utilisera avantageusement des agents de conservation choisis parmi le benzoate de sodium, le 4-hydroxybenzoate de méthyle, le 4-hydroxybenzoate de propyle et leurs mélanges.
Avec la double fonction d'agent de conservation et d'agent de chélation métallique, on pourrait également utiliser de l'EDTA de sodium en plus ou partiellement à la place des agents de conservation susmentionnés.
La préparation du sirop suivant la présente invention est réalisée sous un gaz inerte par la simple dissolution des substances dans la quantité d'eau prédéterminée.
La solution résultante est secouée dans des bouteilles à dose multiple ou dans des récipients à dose unique.
Le conditionnement de bouteilles à dose multiple pourrait constituer l'apport simultané d'une mesure graduée pour le dosage correct des simples doses pour des individus à l'âge adulte (par exemple 10 ml) ou pour des enfants (par exemple 5 ml).
Afin de mieux illustrer la présente invention, on donne à présent les exemples suivants.

Le pH de la solution résultante a été rectifié à la valeur correcte de 6,5 par l'addition d'hydroxyde de sodium.
La solution a été répartie à raison de 150 ml/bouteille.
Les bouteilles ont été conservées pendant 2 ans à la température ambiante.
Après cette période, le titre de NAC s'est révélé supérieur à 95 % du titre initial.
Ce titre s'est révélé supérieur à 90 % du titre initial également 5 semaines après l'ouverture de la bouteille.
Le sirop, également après 2 ans de conservation, a montré une odeur et un goût plaisants et une bonne sapidité générale.

N-acetyl-cysteine (abbreviated as NAC below) is a compound with several interesting pharmacological properties, making it a widely used drug.

By sweetening agent, we mean a substance which can give a sweet taste to the solution but which does not contain sugars. Specific examples of sweetening agents are saccharin, sodium saccharin and cyclamates or their mixtures.
The amount of sweetening agent will be the minimum amount sufficient to give a pleasant taste to the solution.
The thickening agent is chosen from sodium carboxymethylcellulose, hydroxypropylmethylcellulose and their mixtures. Surprisingly, these substances have been found to be compatible with NAC and can give the solution a thickness suitable for the usual palatability of a syrup.
