Composition pharmaceutique contenant de la N-acétyl-cystéine.
La N-acétyl-cystéine est une substance pharmaceutiquement active qui est bien connue pour ses propriétés favorables dans la fluidification des mucus.
Cette substance fluidifie avantageusement les sécrétions en cas notamment d'affections des voies respiratoires et de mucoviscidose. Elle fait partie du groupe des substances appelées mucolytiques ou expectorantes.
Pour son utilisation pratique comme mucolytique, la N-acétyl-cystéine peut être prise par voie buccale sous forme d'une solution aqueuse obtenue par dissolution d'un granulé ou d'un comprimé effervescent, ou sous forme d'un comprimé ou d'une capsule à ingérer avec de l'eau.
Les propriétés organoleptiques de cette substance active sont toutefois très désagréables. Pour la réalisation de ces formulations à administrer par voie buccale il a fallu procéder à de nombreuses recherches pour pouvoir masquer ce goût repoussant de la N- acetyl-cystéine.
Les formulations à absorber par voie orale, dont on vient de parler ci-dessus, sont soit à prendre avec de l'eau et la dissolution se fait à l'intérieur du corps, soit à absorber directement après la dissolution.
La présente invention a pour but de mettre au point une composition pharmaceutique contenant de la Nacétyl-cystéine qui puisse être conservée à l'état fluide. Avantageusement, pendant une durée prolongée, cette composition sera stable à l'entreposage dans des

conditions de température normale et elle conservera un goût agréable où les propriétés organoleptiques désagréables de la substance active seront masquées.
Pour résoudre ces problèmes, on a prévu suivant l'invention une composition pharmaceutique présentant les particularités des revendications 1 à 11 données ci-après.
La formulation prévue suivant l'invention est extemporanée, c'est-à-dire qu'elle concerne une poudre, à dissoudre le moment venu dans un solvant pharmaceutiquement compatible, de préférence de l'eau, pour préparer le sirop en soi.
Le sirop résultant a un bon goût, qui masque celui de la N-acétyl-cystéine, et il conserve ce bon goût pendant la durée de son utilisation. Il est stable dans des conditions normales d'entreposage pendant un temps suffisamment long pour son utilisation et il n'a pas besoin d'être entreposé dans des conditions de réfrigération.
La poudre présente la composition suivante :
N-acétyl-cystéine 7-10 % en poids
Citrate trisodique 7-10 % en poids
Saccharose 80-86 % en poids, et éventuellement des quantités mineures d'additifs usuels habituellement utilisés en pharmacie.
Au moment de l'utilisation, cette poudre est dissoute sous agitation et le sirop est prêt pour l'usage.
Le sirop peut se présenter sous la forme d'ampoules à dose posologique unitaire contenant une quantité de N-acétyl-cystéine comprise entre 100 et 600 mg, de préférence entre 100 et 200 mg, avantageusement de 200 mg.
Il se présente de manière tout à fait avantageuse sous la forme d'un volume unique de sirop égal à

plusieurs doses posologiques unitaires. Il est alors contenu dans une bouteille par exemple de 100,150 ou 250 ml de sirop après addition du solvant. Dans ces bouteilles, la concentration en N-acétyl-cystéine est telle que chaque administration (par exemple une mesurette appropriée ou une cuillère à café) contienne une quantité de substance active d'environ 100 à environ 600 mg, de préférence de 100 à 200 mg.

On complète le tout à 150 ml en ajoutant de l'eau.
On a ajouté un excès de 5 % de N-acétylcystéine en vue d'équilibrer la perte durant la vie en pot de la solution reconstituée.

On mélange ensuite les poids appropriés d'édétate de sodium, de méthylparahydroxybenzoate de sodium, d'agent sapide et 10 % de saccharose pendant 20 minutes à 70 tours/minute.
On ajoute les autres ingrédients : citrate trisodique, N-acétyl-cystéine et 90 % de saccharose, et on continue à mélanger pendant 30 minutes, à 65 tours/minute.
On transvase 74 g du mélange poudreux dans chaque bouteille.
Pendant la fabrication le taux d'humidité ne peut pas dépasser 30 %.
Après reconstitution de la solution, par addition d'eau pour obtenir 150 ml, le pH de la solution reconstituée est de 4,3 + 0,5.
Il doit être entendu que la présente invention n'est en aucune façon limitée aux formes de réalisation décrites ci-dessus et que bien des modifications peuvent y être apportées sans sortir du cadre du présent brevet.

Pharmaceutical composition containing N-acetyl-cysteine.
N-acetyl-cysteine is a pharmaceutically active substance which is well known for its favorable properties in the thinning of mucus.
This substance advantageously fluidifies the secretions in particular in the case of respiratory tract diseases and cystic fibrosis. It is part of the group of substances called mucolytics or expectorants.
For its practical use as a mucolytic, N-acetyl-cysteine can be taken orally in the form of an aqueous solution obtained by dissolving a granule or an effervescent tablet, or in the form of a tablet or a capsule to ingest with water.
The organoleptic properties of this active substance are however very unpleasant. For the production of these formulations to be administered by the oral route, a great deal of research had to be done in order to mask this repulsive taste of N-acetyl-cysteine.
The formulations to be absorbed orally, which have just been mentioned above, are either to be taken with water and the dissolution takes place inside the body, or to be absorbed directly after the dissolution.
The present invention aims to develop a pharmaceutical composition containing Nacetyl-cysteine which can be stored in the fluid state. Advantageously, for a prolonged period, this composition will be stable on storage in

normal temperature conditions and it will retain a pleasant taste where the unpleasant organoleptic properties of the active substance will be masked.
To solve these problems, a pharmaceutical composition is provided according to the invention having the features of claims 1 to 11 given below.
The formulation provided according to the invention is extemporaneous, that is to say that it relates to a powder, to be dissolved when the time comes in a pharmaceutically compatible solvent, preferably water, to prepare the syrup per se.
The resulting syrup has a good taste, which masks that of N-acetyl-cysteine, and it retains this good taste for the duration of its use. It is stable under normal storage conditions for a time long enough for its use and does not need to be stored under refrigeration conditions.
The powder has the following composition:
N-acetyl-cysteine 7-10% by weight
Trisodium citrate 7-10% by weight
Sucrose 80-86% by weight, and possibly minor amounts of usual additives usually used in pharmacy.
At the time of use, this powder is dissolved with stirring and the syrup is ready for use.
The syrup can be in the form of ampoules with a unit dosage dose containing an amount of N-acetyl-cysteine of between 100 and 600 mg, preferably between 100 and 200 mg, advantageously 200 mg.

The appropriate weights of sodium edetate, sodium methyl parahydroxybenzoate, sapid agent and 10% sucrose are then mixed for 20 minutes at 70 rpm.
The other ingredients are added: trisodium citrate, N-acetyl-cysteine and 90% sucrose, and the mixing is continued for 30 minutes, at 65 revolutions / minute.
74 g of the powdery mixture are transferred to each bottle.
During production, the humidity level cannot exceed 30%.
