Abstract

Se describe una Plataforma de Integración de Transgenes Modificados por Ingeniería Genética (ETIP) que puede insertarse en forma aleatoria o en lugares determinados en genomas de plantas para facilitar la rápida selección y detección de un GOI (gen de interés) que es perfectamente reconocido (tanto los extremos 5 como 3) en la ubicación genómica de ETIP. Un elemento en la presente descripción es la introducción de roturas de doble cadena específicas dentro de la ETIP. En algunas realizaciones, una ETIP se describe utilizando sitios de unión de nucleasa de dedo de zinc, pero puede utilizar otras tecnologías de reconocimiento tal como meganucleasas, CRISPRs, TALs, o cierres de leucina. También se describen composiciones de plantas transgénicas y métodos para producir las mismas en donde el ADN donante o de carga expresa uno o más productos de una secuencia de ácido nucleico exógeno (por ejemplo, proteína o ARN) que ha sido establemente integrado en una ETIP en una célula de planta. En realizaciones, la ETIP facilita la prueba de candidatos de genes y vectores de expresión de plantas a partir de ideación por fases de desarrollo.
