Abstract
Se proporciona un método donde una proteína poco hidrosoluble o una proteína en un estado poco extraíble se detecta en una inmunoreacción posterior, mientras se mantiene una alta eficiencia de extracción de la muestra. Se revela un inmunoensayo altamente sensible y sencillo, caracterizado porque la proteína poco hidrosoluble de la muestra o la proteína en estado poco extraíble, se extrae/solubiliza con un solvente acuoso que contiene un tensioactivo iónico en un grado de concentración relativamente elevado, y luego la proteína del extracto se detecta directamente por medio de un anticuerpo generado contra la proteína previamente desnaturalizada con el tensioactivo iónico. Reivindicación 1: Un inmunoensayo para detectar la presencia de una proteína poco soluble/poco extraíble en una muestra, que comprende los siguientes pasos: (1) extraer o solubilizar una proteína poco hidrosoluble/poco extraíble en una muestra con un solvente acuoso que contiene un tensioactivo iónico, (2) agregar un anticuerpo obtenido mediante el uso de la proteína poco hidrosoluble/poco extraíble como inmunógeno previamente desnaturalizada con el tensioactivo iónico utilizado en el paso (1)a: a) la solución de proteína obtenida en el paso (1) sin diluir sustancialmente la solución, o b) una dilución en la cual la solución de proteína obtenida en el paso (1) precedente se diluye en una escala tal que la concentración del tensioactivo iónico no se reduce al 0,03% (p/v) o menos, con lo cual se forma un complejo antígeno-anticuerpo entre la proteína poco hidrosoluble/poco extraíble y el anticuerpo y (3) detectar el complejo de antígeno-anticuerpo formado. Reivindicación 12: Un anticuerpo apropiado para detectar la presencia de una proteína en un solvente acuoso que contiene un tensioactivo iónico, donde la proteína se selecciona del grupo que consta de ovoalbúmina, ovomucoide, caseína, beta-lactoglobulina, proteína de trigo sarraceno, proteína de trigo y proteína de maní y dicho anticuerpo es generado contra dicha proteína desnaturalizada con dicho tensioactivo iónico.
