Abstract
Se describen agentes de unión específica que interactúan con el factor de crecimiento de hepatocitos (HGF). Se describen métodos de tratamiento del cáncer mediante la administración de una cantidad efectiva desde el punto de vista farmacéutico de un agente de unión específica a HGF. Se describen métodos de detección de la cantidad de HGF en una muestra que utilizan un agente de unión específica a HGF. Reivindicación 1: Un polipéptido aislado que comprende por lo menos una región determinante de complementariedad (CDR) seleccionada entre CDR1a, CDR2a y CDR3a; en la cual CDR1a comprende la secuencia de aminoácidos a b c d e f g h i j k l m n o p q, en el cual el aminoácido a se selecciona entre lisina, arginina o glutamina; el aminoácido b se selecciona entre serina o alanina; el aminoácido c es serina; el aminoácido d es glutamina; el aminoácido e se selecciona entre serina, glicina o ácido aspártico; el aminoácido f se selecciona entre valina o isoleucina o no está presente; el aminoácido g se selecciona entre leucina o fenilalanina o no está presente; el aminoácido h se selecciona entre fenilalanina o tirosina o no está presente; el aminoácido i es serina o no está presente; el aminoácido j es serina o no está presente; el aminoácido k se selecciona entre asparagina, treonina o no está presente; el aminoácido l se selecciona entre asparagina, isoleucina o valina; el aminoácido m se selecciona entre lisina, arginina, asparagina o ácido aspártico; el aminoácido n se selecciona entre asparagina o serina; el aminoácido o se selecciona entre tirosina, ácido aspártico, triptofano o asparagina; el aminoácido p es leucina; y el aminoácido q se selecciona entre alanina, glicina o asparagina; en la cual CDR2a comprende la secuencia de aminoácidos r s t u v w x, en la cual el aminoácido r se selecciona entre triptofano, alanina, valina, ácido glutámico o glicina; el aminoácido s es alanina; el aminoácido t es serina; el aminoácido u se selecciona entre treonina, serina o ácido aspártico; el aminoácido v se selecciona entre arginina o leucina; el aminoácido w se selecciona entre ácido glutámico, glutamina o alanina; y el aminoácido x se selecciona entre serina, asparagina o treonina; en la cual CDR3a comprende la secuencia de aminoácidos y z a' b' c' d' e' f' g' h', en la cual el aminoácido y se selecciona entre glutamina o leucina; el aminoácido z se selecciona entre glutamina, asparagina o arginina; el aminoácido a' se selecciona entre tirosina, histidina, alanina o serina; el aminoácido b' se selecciona entre fenilalanina, tirosina, ácido aspártico, asparagina o isoleucina; el aminoácido c' se selecciona entre serina, glicina o asparagina; el aminoácido d' se selecciona entre prolina, tirosina, treonina, fenilalanina, ácido aspártico, leucina o triptofano; el aminoácido e' es prolina; el aminoácido f' es prolina o no está presente; el aminoácido g' es triptofano, leucina, prolina, tirosina o isoleucina; y el aminoácido h' es treonina o asparagina; y en la cual el polipéptido, en asociación con una cadena pesada de un anticuerpo, es capaz de unirse al factor de crecimiento de hepatocitos (HGF). Reivindicación 8: Un polipéptido aislado que comprende por lo menos una región determinante de complementariedad (CDR) seleccionada entre CDR1b, CDR2b y CDR3b; en la cual CDR1b comprende la secuencia de aminoácidos a b c d e f g, en el cual el aminoácido a es serina o no está presente; el aminoácido b se selecciona entre ácido aspártico o glicina o no está presente; el aminoácido c se selecciona entre ácido aspártico, glicina, serina, valina, treonina o isoleucina; el aminoácido d es tirosina; el aminoácido e se selecciona entre tirosina o glicina; el aminoácido f se selecciona entre isoleucina, metionina o triptofano; y el aminoácido g se selecciona entre histidina, asparagina o serina; en la cual CDR2b comprende la secuencia de aminoácidos h i j k l m n o p q r s t u v w x, en la cual el aminoácido h se selecciona entre triptofano, tirosina, valina, asparagina o ácido glutámico; el aminoácido i se selecciona entre isoleucina, fenilalanina o valina; el aminoácido j se selecciona entre asparagina, serina, triptofano o tirosina; el aminoácido k se selecciona entre prolina, serina, tirosina o histidina; el aminoácido l se selecciona entre asparagina, serina o ácido aspártico; el aminoácido m se selecciona entre serina o glicina; el aminoácido n se selecciona entre glicina o serina o no está presente; el aminoácido o se selecciona entre glicina, treonina, ácido aspártico, serina, isoleucina o asparagina; el aminoácido p se selecciona entre treonina, isoleucina o lisina; el aminoácido q se selecciona entre asparagina o tirosina; el aminoácido r se selecciona entre tirosina o histidina; el aminoácido s se selecciona entre alanina o asparagina; el aminoácido t se selecciona entre glutamina, ácido aspártico o prolina; el aminoácido u se selecciona entre lisina o serina; el aminoácido v se selecciona entre fenilalanina, valina o leucina; el aminoácido w se selecciona entre glutamina o lisina; y el aminoácido x se selecciona entre glicina o serina; en la cual CDR3b comprende la secuencia de aminoácidos y z a' b' c' d' e' f' g' h' i' j' k' l' m' n' o' p' q' r', en la cual el aminoácido y se selecciona entre ácido glutámico, ácido aspártico, serina o glicina o no está presente; el aminoácido z se selecciona entre leucina, ácido glutámico, ácido aspártico, histidina, prolina o glicina o no está presente; el aminoácido a' se selecciona entre ácido glutámico, tirosina leucina o no está presente; el aminoácido b' se selecciona entre leucina, asparagina, glicina, histidina, tirosina o triptofano o no está presente; el aminoácido c' se selecciona entre arginina, serina, ácido glutámico, tirosina, glicina o fenilalanina o no está presente; el aminoácido d' es glicina o no está presente; el aminoácido e' se selecciona entre triptofano o tirosina o no está presente; el aminoácido f' es ácido aspártico o no está presente; el aminoácido g' se selecciona entre serina o arginina o no está presente; el aminoácido h' es serina o no está presente; el aminoácido i' se selecciona entre glicina o tirosina o no está presente; el aminoácido j' se selecciona entre tirosina, ácido glutámico o ácido aspártico o no está presente; el aminoácido k' se selecciona entre tirosina, fenilalanina o ácido aspártico o no está presente; el aminoácido l' se selecciona entre tirosina, ácido aspártico, histidina o triptofano o no está presente; el aminoácido m' se selecciona entre tirosina, glicina, ácido aspártico, prolina o serina o no está presente; el aminoácido n' se selecciona entre glicina, valina, tirosina o ácido aspártico o no está presente; el aminoácido o' se selecciona entre leucina, alanina, glicina o tirosina o no está presente; el aminoácido p' se selecciona entre metionina, fenilalanina o tirosina; el aminoácido q' es ácido aspártico; y el aminoácido r' se selecciona entre valina, tirosina, isoleucina o prolina; y en la cual el polipéptido, en asociación con una cadena liviana de un anticuerpo, es capaz de unirse al HGF. Reivindicación 51: Una molécula de ácido nucleico aislada que comprende por lo menos una secuencia de nucleótidos seleccionada entre las SEQ ID NO: 1, 3, 5, 7, 9, 11, 13, 15, 17 y 19. Reivindicación 57: Una célula huésped que comprende la molécula de ácido nucleico de la reivindicación 53. Reivindicación 59: Una línea celular aislada que produce un agente de unión específica de acuerdo con las reivindicaciones 34 ó 36. Reivindicación 92: Un método para obtener un anticuerpo capaz de unirse al factor de crecimiento de hepatocitos (HGF) que comprende la administración de por lo menos un polipéptido seleccionado entre las SEQ ID NO: 164 y 165 a un animal y la obtención de un anticuerpo capaz de unirse al HGF del animal.
