Abstract

Description
本発明は、経口組成物に関する。より具体的には、本発明は、ヒドロキシクエン酸及び/又はその塩を含みながら、呈味が向上した経口組成物に関する。
ヒドロキシクエン酸は、ガルシニアやハイビスカスに含まれる有用成分であり、柑橘類に含まれるクエン酸と非常に類似した構造を有する。一方、ヒドロキシクエン酸はクエン酸より水酸基を1個多く有することから、体内酵素と結合しやすく、その生理活性が注目されている。
ヒドロキシクエン酸の生理活性作用に関しては、脂肪代謝への作用(非特許文献1)、運動負荷ヒト骨格筋におけるグリコーゲン合成促進作用(非特許文献2)等が報告されている。このような生理活性作用に基づく効果を期待して、ヒドロキシクエン酸及び/又はその塩は、体重減少剤及びサプリメントに配合して用いられている。

ヒドロキシクエン酸及び/又はその塩が配合された体重減少剤及びサプリメントは、一般的に、カプセル又はコーティング錠の形態で提供されている。一方、ヒドロキシクエン酸及びその塩には特有の異味があり、呈味を直接感じるような製剤形態では後味が悪い。また、ヒドロキシクエン酸及びその塩には特有のざらざらした舌感があり、舌ざわりを直接感じるような製剤形態では口どけが悪い。
そこで本発明の目的は、ヒドロキシクエン酸及び/又はその塩を含みながら、呈味及び/又は口どけが向上した経口組成物を提供することにある。
本発明者らは、鋭意検討の結果、コンドロイチン硫酸、プロテオグリカン、及びグルコサミン等の所定の糖を、ヒドロキシクエン酸及び/又はその塩とともに配合することで、ヒドロキシクエン酸及びその塩に特有の異味がマスキングされ後味が良くなること、及び特有のざらざらした舌感が抑制され口どけが良くなることを見出した。本発明は、かかる知見に基づいて更に検討を重ねることにより完成したものである。
即ち、本発明は、下記に掲げる態様の発明を提供する。

本発明においては、ヒドロキシクエン酸及びその塩の中でも上記式(I)で表される化合物を「非ラクトン体」又は「フリー体」とも記載し、式(II)で表される化合物(つまり、フリー体の脱水縮合環化物)を「ラクトン体」とも記載する。
上記式(I)及び(II)に示す化合物がヒドロキシクエン酸の場合、上記式(I)及び(II)中、M1、M2及びM3はいずれも水素原子である。
上記式(I)及び(II)に示す化合物がヒドロキシクエン酸の塩の場合、上記式(I)におけるM1、M2及びM3、並びに上記式(II)におけるM1及びM2が同時に水素原子になることはない。また、ヒドロキシクエン酸の塩としては、薬学的又は香粧学的に許容されるものである限り特に制限されない。
具体的には、上記式(I)及び(II)に示す化合物がヒドロキシクエン酸の塩である場合の上記式(I)におけるM1、M2及びM3、並びに上記式(II)におけるM1及びM2は、それぞれ独立に、アルカリ金属又はアルカリ土類金属、若しくは有機塩基を表す。アルカリ金属としては、カリウム、ナトリウムが挙げられる。アルカリ土類金属としては、カルシウムが挙げられる。有機塩基としては、モノエタノールアミン基、ジエタノールアミン基、トリエタノールアミン基、アミノメチルプロパノール基、アミノメチルプロパンジオール基が挙げられる。これらの塩は、1種単独で使用してもよく、また2種以上を組み合わせて使用してもよい。
本発明の経口組成物において、ヒドロキシクエン酸類として、フリー体及びラクトン体のうちいずれか一方を使用してもよいし、両方を組み合わせて用いてもよく、また、上記のフリー体及びラクトン体の中から、1種を選択して使用してもよいし、2種以上を組み合わせて使用してもよい。これらのヒドロキシクエン酸類のフリー体及びラクトン体の中でも、好ましくはフリー体が挙げられる。
また、本発明の経口組成物において、ヒドロキシクエン酸類として、ヒドロキシクエン酸及びヒドロキシクエン酸の塩のうち、いずれか一方を使用してもよいし、両方を組み合わせて用いてもよく、また、上記のヒドロキシクエン酸及びヒドロキシクエン酸の塩の中から、1種を選択して使用してもよいし、2種以上を組み合わせて使用してもよい。これらのヒドロキシクエン酸及びその塩の中でも、好ましくはヒドロキシクエン酸の塩が挙げられ、より好ましくはアルカリ金属塩及びアルカリ土類金属塩が挙げられ、さらに好ましくはアルカリ土類金属塩が挙げられ、特に好ましくはカルシウム塩が挙げられる。
本発明の経口組成物において、ヒドロキシクエン酸類は、天然物から取得されたものであってもよいし、化学合成されたものであってもよい。天然物としては、オトギリソウ科フクギ属のガルシニア種(具体例として、ガルシニア・カンボジア(Garcinia cambogia)、ガルシニア・インディカ(Garcinia indica)、ガルシニア・アトロビリディス(Garcinia atroviridis)、ガルシニア・マンゴスターナ(Garcinia mangostana)、ガルシニア・スベリプティカ(Garcinia subelliptica)等が挙げられる。)、アオイ科フヨウ属のハイビスカス(Hibiscus L.)等の植物が挙げられ、好ましくはガルシニア属が挙げられ、より好ましくはガルシニア・カンボジア(Garcinia cambogia)が挙げられる。植物は、栽培により生産されたものであっても天然より採取されたものであってもよい。植物からヒドロキシクエン酸類を取得する方法は従来公知である。本発明の経口組成物は、天然物からのヒドロキシクエン酸類の単離精製物を含んでいてもよいし、天然物からのヒドロキシクエン酸類の粗精製物を含んでいてもよい。
ヒドロキシクエン酸類の粗精製物の例としては、上記植物の加工処理物が挙げられ、好ましくは果皮の加工処理物が挙げられる。上記植物の加工処理物の具体的な態様としては、植物乾燥物、植物粉砕物(生及び乾燥物を含む)、植物エキスが挙げられる。植物の加工処理物としては、単一の植物由来のものを用いてもよく、異なる植物由来のものを2種以上組み合わせて用いてもよい。これらの植物の加工処理物の中でも、好ましくは植物エキスが挙げられる。
上述の植物の加工処理物の粗精製物のうち、好ましくは植物エキスが挙げられ、より好ましくはガルシニア属から得られるエキス(ガルシニアエキス)(特に好ましくは、ガルシニア・カンボジア(Garcinia cambogia)から得られるエキス)が挙げられる。植物エキスは、搾汁、溶媒抽出物、溶媒抽出物のヒドロキシクエン酸類を含む分画物であればよい。植物エキスを得る方法は特に限定されないが、例えば次のようにして得ることができる。植物エキスは、例えば、ガルシニア種等の植物の果皮をそのままの生の状態で、またはガルシニア種等の植物の果皮の乾燥物の状態で用意し、そのままの大きさで、または更に必要に応じて裁断または粉砕した後、溶媒抽出、超臨界抽出等の慣用の抽出方法に従って調製することができる。抽出溶媒としては、水(温水及び熱水を含む)、有機溶媒(メタノール、エタノール、n-プロパノール、イソプロパノール、n-ブタノール等の炭素数1~4の低級アルコール;プロピレングリコール、1,3-ブチレングリコール等の多価アルコール;アセトン等のケトン類;ジエチルエーテル、ジオキサン、アセトニトリル、酢酸エチルエステル等のエステル類;キシレン、ベンゼン、クロロホルム等)、これらの混合物が挙げられ、好ましくは、水、低級アルコール、これらの混合物が挙げられ、より好ましくは、温水、熱水などの加熱水が挙げられ、更に好ましくは熱水が挙げられる。これらの溶媒は1種単独で用いてもよく、2種以上を組み合わせて用いてもよい。
得られた植物エキス(搾汁、溶媒抽出物、溶媒抽出物のヒドロキシクエン酸類を含む分画物等)を経口組成物に含ませる場合、当該植物エキスは、そのままの濃縮されない非濃縮エキスの態様であってもよいし、濃縮された液状の軟エキスの態様であってもよいし、非濃縮エキスまたは軟エキスをさらに乾燥処理に供して得たエキス末の態様であってもよい。乾燥処理としては、スプレードライ処理及び凍結乾燥処理が挙げられる。
上記のヒドロキシクエン酸類の粗精製物(好ましくは植物の加工処理物、より好ましくは植物エキス)の乾燥重量換算量100重量%中のヒドロキシクエン酸類の量としては、ヒドロキシクエン酸換算量で、例えば10~80重量%が挙げられる。
ヒドロキシクエン酸類の単離精製物を得る方法としては特に限定されないが、上述の抽出物のヒドロキシクエン酸類含む分画物をさらに精製処理する方法が挙げられる。精製処理としては、ヒドロキシクエン酸類を単離して精製度をさらに高める方法であればよく、常法に従って行うことができる。例えば、クロマトグラフィー等の分離処理、再結晶処理等が挙げられる。
本発明の経口組成物における(A)成分の配合量については特に制限されず、期待すべき効果に応じて適宜決定することができる。また、本発明の経口組成物は呈味及び/又は口どけの向上効果に優れるため、(A)成分の配合量が多くても、効果的に好ましい呈味及び/又は口どけを得ることができる。本発明の経口組成物における(A)成分の具体的な配合量としては、ヒドロキシクエン酸換算量で、例えば10重量%以上、好ましくは15重量%以上、より好ましくは25重量%以上、さらに好ましくは30重量%以上、より一層好ましくは40重量%以上、特に好ましくは45重量%以上が挙げられる。当該配合量の上限については特に制限されないが、呈味及び/又は口どけの向上効果を高める観点から、好ましくは90重量%以下、より好ましくは80重量%以下、より好ましくは70重量%以下、さらに好ましくは60重量%以下、一層好ましくは50重量%が挙げられる。
(B)所定の糖
本発明の経口組成物は、(B)成分として、(B1)コンドロイチン硫酸及びその塩、(以下において、「(B1)成分」とも記載する。)、(B2)プロテオグリカン、(以下において、「(B2)成分」とも記載する。)、並びに(B3)N-アセチル化されていてもよいヘキソサミンからなる群より選択される所定の糖を含む。これら(B)成分は、(A)成分に特有の異味をマスキングし、及び/又は(A)成分に特有のざらざらした舌感を抑制できるため、経口組成物の呈味及び/又は口どけを向上させる。
本発明の経口組成物は、(B)成分として、(B1)コンドロイチン硫酸及びその塩、(以下において、「(B1)成分」とも記載する。)、(B2)プロテオグリカン、(以下において、「(B2)成分」とも記載する。)、並びに(B3)N-アセチル化されていてもよいヘキソサミンからなる群より選択される所定の糖を含む。これら(B)成分は、(A)成分に特有の異味をマスキングし、及び/又は(A)成分に特有のざらざらした舌感を抑制できるため、経口組成物の呈味及び/又は口どけを向上させる。
(B1)成分であるコンドロイチン硫酸は、N-アセチルガラクトサミン(N-アセチル-D-ガラクトサミンを指す。以下において同様。)及びグルクロン酸(D-グルクロン酸を指す。以下において同様。)又はイズロン酸(L-イズロン酸を指す。以下において同様。)の2糖を反復構造単位とする糖鎖に硫酸が結合した酸性ムコ多糖として公知の化合物である。
コンドロイチン硫酸としては、コンドロイチン硫酸A(構成2糖は、グルクロン酸及びアセチルガラクトサミン4硫酸)、コンドロイチン硫酸B(構成2糖は、イズロン酸2硫酸及びアセチルガラクトサミン4硫酸;デルマタン硫酸とも呼ばれる)、コンドロイチン硫酸C(構成2糖は、グルクロン酸及びアセチルガラクトサミン6硫酸)、コンドロイチン硫酸D(構成2糖は、グルクロン酸2硫酸及びアセチルガラクトサミン6硫酸)、コンドロイチン硫酸E(構成2糖は、グルクロン酸及びアセチルガラクトサミン4,6二硫酸)が挙げられる。これらのコンドロイチン硫酸は、1種単独で使用してもよいし、2種以上を組み合わせて使用してもよい。本発明で用いられるコンドロイチン硫酸は、少なくともコンドロイチン硫酸Aを含むことが好ましい。
コンドロイチン硫酸の塩としては特に限定されないが、例えば、ナトリウム塩、カリウム塩等のアルカリ金属塩;カルシウム塩、マグネシウム塩等のアルカリ土類金属塩;アンモニウム塩;アルギニン、リジン、ヒスチジン、オルニチン等の塩基性アミノ酸塩;モノエタノールアミン塩、ジエタノールアミン塩等のアミン塩等が挙げられる。これらの塩は、1種単独で使用してもよいし、2種以上を組み合わせて使用してもよい。
また、(B1)成分として、コンドロイチン硫酸及びコンドロイチン硫酸の塩のうちいずれか一方を使用してもよいし、コンドロイチン硫酸及びコンドロイチン硫酸の塩の両方を組み合わせて使用してもよい。これらの中でも、呈味及び/又は口どけの向上効果を高める観点から、好ましくはコンドロイチン硫酸が挙げられる。
コンドロイチン硫酸及びその塩の平均分子量としては、例えば477~10万が挙げられ、好ましくは477~10万、より好ましくは477~1万、さらに好ましくは480~5000、一層好ましくは485~1000、より一層好ましくは490~800、特に好ましくは490~550が挙げられる。なお、本発明において、コンドロイチン硫酸及びその塩の平均分子量は、光散乱法を用いたゲルパーミエーションクロマトグラフ(コンドロイチン硫酸標準品(生化学工業株式会社製、サメ軟骨由来又はクジラ軟骨由来のコンドロイチン硫酸標準品)を標準物質とする。)により測定される重量平均分子量である。
コンドロイチン硫酸及びその塩としては、化学合成したものを用いてもよいし、天然物由来の材料から抽出又は精製等したものであってもよい。また、コンドロイチン硫酸又はその塩としては、試薬として市販されているコンドロイチン硫酸又はその塩を使用してもよい。
コンドロイチン硫酸又はその塩を天然物由来の材料から抽出又は精製等して製造する方法としては、特に限定されず、天然物由来の材料から公知の方法で抽出、精製等する方法が挙げられる。コンドロイチン硫酸を抽出するための天然物由来の材料としては、例えば、豚、牛等の哺乳動物の軟骨;鶏等の鳥類の軟骨;サケ、エイ、サメ等の魚類の軟骨等が挙げられ、好ましくは哺乳動物の軟骨が挙げられ、より好ましくは豚軟骨が挙げられる。
(B2)成分であるプロテオグリカンは、ムコ多糖とタンパク質との複合体であり、タンパク質のポリペプチド鎖をコアとして、これに複数本のムコ多糖鎖が結合して構成されている。プロテオグリカンの構成ムコ多糖は、N-アセチル化されていてもよいヘキソサミンとグルクロン酸又はイズロン酸との繰返し構造からなる多糖であればよい。N-アセチル化されていてもよいヘキソサミンとしては、グルコサミン、N-アセチルグルコサミン、N-アセチルグルコサミン硫酸及びその塩、ガラクトサミン、N-アセチルガラクトサミン、N-アセチルガラクトサミン硫酸及びその塩が挙げられる。当該ムコ多糖の例としては、コンドロイチン硫酸(上記(B1)成分として記載した酸性ムコ多糖)、ヘパラン硫酸(グルコサミン及びグルクロン酸またはイズロン酸を反復構造単位とする糖鎖に硫酸が結合した酸性ムコ多糖)、ヘパリン(グルコサミン及びグルクロン酸またはイズロン酸を反復構造単位とする糖鎖に硫酸が結合した酸性ムコ多糖)、及びケラタン硫酸(N-アセチルグルコサミン及びガラクトースを反復構造単位とする糖鎖に硫酸が結合した酸性ムコ多糖)が挙げられる。
プロテオグリカンは、1種単独で使用してもよいし、2種以上を組み合わせて使用してもよい。
プロテオグリカンとしては、化学合成したものを用いてもよいし、天然物由来の材料から抽出又は精製等したものであってもよい。また、プロテオグリカンとしては、試薬として市販されているプロテオグリカンを使用してもよい。
プロテオグリカンを天然物由来の材料から抽出又は精製等して製造する方法としては、特に限定されず、天然物由来の材料から公知の方法で抽出、精製等する方法が挙げられる。プロテオグリカンを抽出するための天然物由来の材料としては、例えば、豚、牛等の哺乳動物の軟骨;鶏等の鳥類の軟骨;サケ、エイ、サメ等の魚類の軟骨等が挙げられ、呈味及び/又は口どけの向上効果を高める観点から、好ましくは魚類の軟骨が挙げられ、より好ましくはサケの軟骨が挙げられる。
(B3)成分であるN-アセチル化されていてもよいヘキソサミンは、N-アセチル化されていないヘキソサミン又はN-アセチル化ヘキソサミンであればよい。本発明においては、(B3)成分として、N-アセチル化されていないヘキソサミン及びN-アセチル化ヘキソサミンのうちいずれか一方のみを用いてもよいし、両方組み合わせて用いてもよい。
N-アセチル化されていないヘキソサミンとしては、グルコサミン、ガラクトサミン、マンノサミン及びフルクトサミンが挙げられ、N-アセチル化ヘキソサミンヘキソサミンとしては、N-アセチルグルコサミン、N-アセチルガラクトサミン、N-アセチルマンノサミン及びN-アセチルフルクトサミンが挙げられる。本発明においては、(B3)成分として、これらの化合物から1種を単独で用いてもよいし、複数種を組み合わせて用いてもよい。
本発明においては、呈味及び/又は口どけの向上効果を高める観点から、(B3)成分として、好ましくはN-アセチル化されていないヘキソサミンが挙げられ、より好ましくはグルコサミン、ガラクトサミンが挙げられ、さらに好ましくはグルコサミンが挙げられる。
本発明において、(B)成分としては、上記(B1)成分、(B2)成分及び(B3)成分の3タイプの成分のうち、単独タイプのみを使用してもよいし、複数タイプを組み合わせて用いてもよい。
本発明の経口組成物において、(B)成分の含有量としては特に限定されないが、例えば、(A)成分のヒドロキシクエン酸換算総量1重量部に対する(B)成分の総量として、0.001~6重量部が挙げられ、本発明の効果をより一層高める観点から、好ましくは0.0015~4重量部、より好ましくは以下に挙げる含有量が挙げられる。また、(B)成分を、(A)成分を含む植物エキスとともに用いる場合、植物エキス1重量部に対する(B)成分の量として、0.0006~3.6重量部が挙げられ、本発明の効果をより一層高める観点から、好ましくは0.0009~2.4重量部、より好ましくは以下に挙げる含有量が挙げられる。
(B1)成分を用いる場合:
(A)成分のヒドロキシクエン酸換算総量1重量部に対する(B1)成分の量として、好ましくは0.008~1重量部、より好ましくは0.03~0.4重量部、さらに好ましくは0.035~0.2重量部、0.035~0.1重量部、0.035~0.05重量部、0.04~0.2重量部、0.04~0.1重量部、又は0.04~0.05重量部が挙げられる。
(A)成分のヒドロキシクエン酸換算総量1重量部に対する(B1)成分の量として、好ましくは0.008~1重量部、より好ましくは0.03~0.4重量部、さらに好ましくは0.035~0.2重量部、0.035~0.1重量部、0.035~0.05重量部、0.04~0.2重量部、0.04~0.1重量部、又は0.04~0.05重量部が挙げられる。
(B2)成分を用いる場合:
(A)成分のヒドロキシクエン酸換算総量1重量部に対する(B3)成分の量として、好ましくは0.0015~0.35重量部、より好ましくは0.005~0.3重量部、さらに好ましくは0.04~0.25重量部、一層好ましくは0.06~0.2重量部、より一層好ましくは0.08~0.15重量部が挙げられる。
(A)成分のヒドロキシクエン酸換算総量1重量部に対する(B3)成分の量として、好ましくは0.0015~0.35重量部、より好ましくは0.005~0.3重量部、さらに好ましくは0.04~0.25重量部、一層好ましくは0.06~0.2重量部、より一層好ましくは0.08~0.15重量部が挙げられる。
(B3)成分を用いる場合:
(A)成分のヒドロキシクエン酸換算総量1重量部に対する(B3)成分の量として、総量で、好ましくは0.0015~3重量部、より好ましくは0.009~3重量部、さらに好ましくは0.05~3重量部、一層好ましくは0.3~3重量部、より一層好ましくは0.6~3重量部、特に好ましくは0.8~3重量部又は0.8~2重量部が挙げられる。
上記の(B3)成分の量は、(B3)成分として用いる成分の種類に関わらず適用できるが、特に好ましくは、(B3)成分としてグルコサミンを用いる場合に適用される。
(A)成分のヒドロキシクエン酸換算総量1重量部に対する(B3)成分の量として、総量で、好ましくは0.0015~3重量部、より好ましくは0.009~3重量部、さらに好ましくは0.05~3重量部、一層好ましくは0.3~3重量部、より一層好ましくは0.6~3重量部、特に好ましくは0.8~3重量部又は0.8~2重量部が挙げられる。
上記の(B3)成分の量は、(B3)成分として用いる成分の種類に関わらず適用できるが、特に好ましくは、(B3)成分としてグルコサミンを用いる場合に適用される。
本発明の経口組成物における(B)成分の具体的な配合量については、特に制限されず、好ましくは、(A)成分の配合量との比率が上記の比率となるように設定すればよい。具体的な(B)成分の配合量の例としては、0.1~50重量%、好ましくは0.5~40重量%、より好ましくは0.8~30重量%、更に好ましくは1~30重量%が挙げられる。
その他の成分
本発明の経口組成物は、上記(A)成分及び上記(B)成分以外に、本発明の効果を損なわない範囲で、適用形態に応じて他の成分を含有していてもよいし、含有していなくてもよい。このような他の成分としては、例えば、生理活性物質、及び添加物等が挙げられる。

生理活性物質としては、例えば、コラーゲン、II型コラーゲン、非変性活性2型コラーゲン、コラーゲンペプチド、メチルスルホニルメタン(MSM)、S-アデノシルメチオニン、クレアチン、テアニン、ピぺリン、マスリン酸、5-アミノレブリン酸リン酸塩、キャッツクロー、ブラックジンジャー、ボスウェリアセラータ、アーティチョーク、アミノ酸、ビタミンA、ビタミンB1、ビタミンB2、ビタミンB6、ビタミンB12、ビタミンC、ビタミンD2、ビタミンD3、ビタミンE,ビタミンK、葉酸、ヒアルロン酸、カツオ由来エラスチンペプチド、イミダゾールジペプチド、ケルセチン配糖体、クリルオイル由来EPA・DHA、モリンガ種子由来グルコモリンギン、イタドリ、デビルズクロー、鶏足由来ヒアルロン酸産生促進剤(HAS-II)、大豆イソフラボン、β-クリプトキサンチン、ボーンペップ、濃縮乳清活性たんぱく質(CBP)、3-ヒドロキシ-3-メチルブチレート(HMB)、カルシウムビス-3-ヒドロキシ-3-メチルブチレートモノハイドレート(HMBカルシウム)、マルトビオン酸カルシウム、カルシウム、マグネシウム、亜鉛、鉄、セレン、カリウム、エストロゲン、カルシトニン、アスピリン、ステロイド性消炎剤、非ステロイド性消炎剤等が挙げられる。これらの生理活性物質は、1種を選択して使用してもよく、2種以上を組み合わせて使用してもよい。
添加物としては、薬学的又は食品学的に許容される賦形剤、崩壊剤、希釈剤、滑沢剤、着香剤、着色剤、甘味剤、矯味剤、懸濁剤、湿潤剤、乳化剤、分散剤、補助剤、防腐剤、緩衝剤、結合剤、安定剤、増量剤、増粘剤、pH調整剤、界面活性剤、コーティング剤、栄養成分等が挙げられる。これらの添加物は、1種を選択して使用してもよく、2種以上を組み合わせて使用してもよい。これらの中でも好ましくは滑沢剤が挙げられ、呈味及び/又は口どけの向上効果を高める観点から、好ましくはステアリン酸、ステアリン酸カルシウム、ステアリン酸マグネシウムが挙げられ、より好ましくはステアリン酸カルシウム(日本薬局方)が挙げられる。
製剤形態
本発明の経口組成物は、上記(A)成分及び上記(B)成分を含み、且つ経口組成物として調製される限り、その形態及び性状は特に限定されない。

また、本発明の経口組成物の性状は、液状であってもよいし、固形状であってもよい。液状の例としては、液剤、飲料剤、乳剤、懸濁剤、酒精剤、シロップ剤、エリキシル剤、軟エキス剤等を含む)等が挙げられ、固形状の例としては、錠剤(好ましくは素錠剤)、丸剤、散剤、細粒剤、顆粒剤、トローチ剤、チュアブル剤等が挙げられる。本発明の経口組成物は、(A)成分を含んでいながらも呈味及び/又は口どけが向上しているため、呈味を直接感じるような製剤形態、及び/又は舌ざわりを直接感じるような製剤形態である場合に特に有用である。このような観点から、本発明の経口組成物は好ましくは固形状であることが好ましく、より好ましくは、素錠剤、丸剤、散剤、細粒剤、顆粒剤、トローチ剤、チュアブル剤が挙げられ、さらに好ましくは、散剤、細粒剤、顆粒剤が挙げられる。
本発明の経口組成物は、一般飲食品、保健機能食品(特定保健用食品、栄養機能食品、機能性表示食品、サプリメント等を含む)、病者用食品、医薬品、医薬部外品として使用することができ、また、食品、医薬品、医薬部外品等への添加剤等として使用することもできる。特に、日常的及び/又は継続的に気軽に摂取させる観点から、サプリメントとして使用することが好ましい。
製造方法
本発明の経口組成物の製造方法は、上記(A)成分及び上記(B)成分と、必要に応じて配合されるその他の成分とを用いて、各種形態及び性状、並びに使用目的に応じ、従来公知の通常の製剤手順に従えばよい。

本発明の経口組成物は、ヒドロキシクエン酸及び/又はその塩による生理活性作用を利用する任意の用途に用いることができる。
2.呈味及び/又は口どけの向上方法、並びに、呈味及び/又は口どけ向上剤
上述のとおり、(B)成分は、(A)成分を含む経口組成物の呈味及び/又は口どけを向上できる。従って、本発明は、(A)ヒドロキシクエン酸及び/又はその塩を含む経口組成物の呈味及び/又は口どけの向上方法であって、前記(A)成分とともに、(B)コンドロイチン硫酸及びその塩、プロテオグリカン、並びにN-アセチル化されていてもよいヘキソサミンからなる群より選択される糖を配合する、呈味及び/又は口どけの向上方法;並びに、コンドロイチン硫酸及びその塩、プロテオグリカン、並びにN-アセチル化されていてもよいヘキソサミンからなる群より選択される糖を含む、ヒドロキシクエン酸及び/又はその塩を含む経口組成物に対する呈味及び/又は口どけ向上剤も提供する。
上述のとおり、(B)成分は、(A)成分を含む経口組成物の呈味及び/又は口どけを向上できる。従って、本発明は、(A)ヒドロキシクエン酸及び/又はその塩を含む経口組成物の呈味及び/又は口どけの向上方法であって、前記(A)成分とともに、(B)コンドロイチン硫酸及びその塩、プロテオグリカン、並びにN-アセチル化されていてもよいヘキソサミンからなる群より選択される糖を配合する、呈味及び/又は口どけの向上方法;並びに、コンドロイチン硫酸及びその塩、プロテオグリカン、並びにN-アセチル化されていてもよいヘキソサミンからなる群より選択される糖を含む、ヒドロキシクエン酸及び/又はその塩を含む経口組成物に対する呈味及び/又は口どけ向上剤も提供する。
呈味及び/又は口どけの向上方法、並びに、呈味及び/又は口どけ向上剤において、(A)成分及び(B)成分の詳細、含有の有無を問わないその他の成分、製剤形態、及び製造方法等の詳細については、上記「1.経口組成物」と同様である。
以下に実施例を示して本発明をより具体的に説明するが、本発明はこれらに限定されるものではない。
試験例
表1A、表1B及び表2に示す組成の経口組成物を散剤の形態で調製した。各成分の詳細については次の通りである。
・(A)成分
(-)ヒドロキシクエン酸-3カルシウム(シグマ・アルドリッチ製)
・(B)成分
(B)成分の配合のために以下の材料を用いた。
(B1)コンドロイチン硫酸素材(豚軟骨エキス;豚軟骨由来コンドロイチン硫酸(主たるコンドロイチン硫酸は、コンドロイチン硫酸A)含量84重量%(USP-HPLC法による測定値);重量平均分子量499.38)
(B2)プロテオグリカン素材(鮭(鼻軟骨)エキス;鮭(鼻軟骨)由来プロテオグリカン含量22.4重量%)
(B3)グルコサミン(エビ・カニ由来)
・その他の成分
クエン酸3ナトリウム
ステアリン酸カルシウム(日本薬局方ステアリン酸カルシウム)
表1A、表1B及び表2に示す組成の経口組成物を散剤の形態で調製した。各成分の詳細については次の通りである。
・(A)成分
(-)ヒドロキシクエン酸-3カルシウム(シグマ・アルドリッチ製)
・(B)成分
(B)成分の配合のために以下の材料を用いた。
(B1)コンドロイチン硫酸素材(豚軟骨エキス;豚軟骨由来コンドロイチン硫酸(主たるコンドロイチン硫酸は、コンドロイチン硫酸A)含量84重量%(USP-HPLC法による測定値);重量平均分子量499.38)
(B2)プロテオグリカン素材(鮭(鼻軟骨)エキス;鮭(鼻軟骨)由来プロテオグリカン含量22.4重量%)
(B3)グルコサミン(エビ・カニ由来)
・その他の成分
クエン酸3ナトリウム
ステアリン酸カルシウム(日本薬局方ステアリン酸カルシウム)
得られた経口組成物を、100mgずつ、訓練された味覚パネル4名で以下の基準に基づいて判定し、評点の平均値を導出した。評点の平均値が高いほど、効果が低いことを示す。さらに、それぞれの表現平均値について、実施例にあっては、比較例1の評点平均値を1とした場合の相対評点を導出し、参考例2~4にあっては、参考例1の評点平均値を1とした場合の相対評点を導出した。相対評点が1を下回る場合に、所定の効果が得られていることを示す。結果を表1及び表2に示す。
<呈味の向上効果>
(A)成分特有の異味を感じる程度に基づく以下の基準により採点した。評点が小さいほど、呈味の向上効果が高い。
5:異味を強く感じ、後味が悪い
4:異味をやや強く感じ、後味がやや悪い
3:異味は強いとも弱いともいえない
2:弱い異味しか感じず、後味がやや良い
1:異味を感じず、後味が良い
(A)成分特有の異味を感じる程度に基づく以下の基準により採点した。評点が小さいほど、呈味の向上効果が高い。
5:異味を強く感じ、後味が悪い
4:異味をやや強く感じ、後味がやや悪い
3:異味は強いとも弱いともいえない
2:弱い異味しか感じず、後味がやや良い
1:異味を感じず、後味が良い
<口どけの向上効果>
舌感のざらつきを感じる程度に基づく以下の基準により採点した。評点が小さいほど、口どけの向上効果が高い。
5:舌感のざらつきを強く感じ、口どけが悪い
4:舌感のざらつきをやや強く感じ、口どけがやや悪い
3:舌感のざらつきは強いとも弱いともいえない
2:弱いざらつきしか感じず、口どけがやや良い
1:舌感のざらつきを感じず、口どけが良い
舌感のざらつきを感じる程度に基づく以下の基準により採点した。評点が小さいほど、口どけの向上効果が高い。
5:舌感のざらつきを強く感じ、口どけが悪い
4:舌感のざらつきをやや強く感じ、口どけがやや悪い
3:舌感のざらつきは強いとも弱いともいえない
2:弱いざらつきしか感じず、口どけがやや良い
1:舌感のざらつきを感じず、口どけが良い
表1Bの比較例1と実施例1~10との対比から明らかなとおり、(B)成分は(A)成分の異味を低減することで呈味を向上することが認められた。一方、表1Aの参考例1と参考例2~4との対比から明らかなとおり、(B)成分は(A)成分と非常に構造が似ているクエン酸3ナトリウムに対しては異味を抑制できなかった。すなわち、実施例1~10で認められた呈味の向上効果は、(A)成分を有する経口組成物に特有の効果であることが分かった。また、表2に示す通り、比較例1と対比した実施例1~10の口どけの評点も、表1Bにおける呈味の評点と同様であった。つまり、表2の比較例1と実施例1~10との対比から明らかなとおり、(B)成分は(A)成分のざらざらした舌感を低減し口どけを向上できることも認められた。
以上より、本発明は、(B)成分の配合により、(A)成分を含有する経口組成物における(A)成分の呈味及び口どけを向上できる。このため、本発明は(A)成分を含む植物エキスを含有する経口組成物にも適用できる。なお、当該植物エキスを含有する経口組成物にコンドロイチン硫酸を配合すると、調製直後での(A)成分の量に対する、50℃、RH60%の条件下で3週間保存した後での(A)成分の減量が、コンドロイチン硫酸を配合しない場合に比べて抑制されており、安定性が向上することも確認できた。
Claims (8)
- (A)ヒドロキシクエン酸及び/又はその塩と、(B)コンドロイチン硫酸及びその塩、プロテオグリカン、並びにN-アセチル化されていてもよいヘキソサミンからなる群より選択される糖とを含む、経口組成物。
- 前記N-アセチル化されていてもよいヘキソサミンが、グルコサミン及び/又はガラクトサミンである、請求項1に記載の経口組成物。
- 前記(A)成分のヒドロキシクエン酸換算総量1重量部に対する前記(B)成分の総量の含有量が、0.001~6重量部である、請求項1に記載の経口組成物。
- 前記(A)成分のヒドロキシクエン酸換算総量1重量部に対する前記コンドロイチン硫酸及びその塩の総量の含有量が、0.01~1重量部である、請求項1に記載の経口組成物。
- 前記(A)成分のヒドロキシクエン酸換算総量1重量部に対する前記プロテオグリカンの含有量が、0.0015~0.35重量部である、請求項1に記載の経口組成物。
- 前記(A)成分のヒドロキシクエン酸換算総量1重量部に対する前記N-アセチル化されていてもよいヘキソサミンの含有量が、0.0015~3重量部である、請求項1に記載の経口組成物。
- (A)ヒドロキシクエン酸及び/又はその塩を含む経口組成物の呈味及び/又は口どけの向上方法であって、前記(A)成分とともに、(B)コンドロイチン硫酸及びその塩、プロテオグリカン、並びにN-アセチル化されていてもよいヘキソサミンからなる群より選択される糖を配合する、呈味及び/又は口どけの向上方法。
- コンドロイチン硫酸及びその塩、プロテオグリカン、並びにN-アセチル化されていてもよいヘキソサミンからなる群より選択される糖を含む、ヒドロキシクエン酸及び/又はその塩を含む経口組成物に対する呈味及び/又は口どけ向上剤。
