Abstract
本発明の目的は、ヒドロキシクエン酸を含む植物エキスを含有する医薬又は食品組成物において、ヒドロキシクエン酸の減量を抑制する製剤処方を提供することである。ヒドロキシクエン酸を含む植物エキスを含有する医薬又は食品組成物において、前記植物エキスとともに、コンドロイチン硫酸及び/又はその塩を配合すると、ヒドロキシクエン酸の減量を抑制できる。
Description
本発明は、医薬又は食品組成物に関する。より具体的には、本発明は、ヒドロキシクエン酸を含む植物エキスを含みながら、ヒドロキシクエン酸の減量が抑制された医薬又は食品組成物に関する。
ヒドロキシクエン酸は、脂肪代謝への作用(非特許文献1)、運動負荷ヒト骨格筋におけるグリコーゲン合成促進作用(非特許文献2)等の生理活性が報告されている有用成分である。
ヒドロキシクエン酸は、ガルシニアやハイビスカス等の植物に含まれている。このような植物から抽出したエキスは、ヒドロキシクエン酸による生理活性に基づく効果を期待して、体重減少剤及びサプリメント等の医薬又は食品組成物に配合して用いられている。また、ガルシニア抽出エキスをカフェインと組み合わせることで、脂肪代謝をより促進できる脂肪代謝促進組成物も報告されている(特許文献1)。
一方、ヒドロキシクエン酸を含む組成物は、ヒドロキシクエン酸の可溶性を向上させる観点から、医薬的に許容できる塩の形態で用いられることがある。ヒドロキシクエン酸の塩の中には吸湿性のものがあるため、カルシウム塩の形態で組成物に配合することで吸湿性を抑制することが報告されている(特許文献2)。

ヒドロキシクエン酸を含む製剤の安定性に関しては、上述のように、ヒドロキシクエン酸を所定の塩の形態に変更することで吸湿性を抑制するアプローチは知られている。しかしながら、製剤の安定性に影響する要素は吸湿性の抑制に限られるものではなく、有効成分の生物学的利用能を高めるために、製剤中での有効成分の減量を抑制することが望まれる。そして、ヒドロキシクエン酸を含む製剤について、ヒドロキシクエン酸の減量を抑制する技術については十分に検討されてこなかった。
本発明者は、ヒドロキシクエン酸を含む植物エキスを含有する製剤の安定性を検討したところ、製剤調製後にヒドロキシクエン酸が減量するという不安定性の課題に直面した。一方で、ヒドロキシクエン酸を単独化合物として含有する製剤ではそのような不安定性は認められなったことから、当該不安定性は、植物エキスを含有する製剤に特有の課題であることを見出した。
そこで本発明の目的は、ヒドロキシクエン酸を含む植物エキスを含有する医薬又は食品組成物において、ヒドロキシクエン酸の減量を抑制する製剤処方を提供することにある。
本発明者らは、鋭意検討の結果、ヒドロキシクエン酸を含む植物エキスを含有する医薬又は食品組成物において、コンドロイチン硫酸及び/又はその塩を配合することで、ヒドロキシクエン酸の減量が抑制できることを見出した。さらに、ヒドロキシクエン酸を含む植物エキスを含有する医薬又は食品組成物を錠剤として製剤した場合には、コンドロイチン硫酸及び/又はその塩を配合することで、ヒドロキシクエン酸の減量抑制だけでなく、水分がある条件に晒された場合に生じるひび割れ及び/又は面荒れを抑制できることも見出した。本発明は、これらの知見に基づいて更に検討を重ねることにより完成したものである。
即ち、本発明は、下記に掲げる態様の発明を提供する。
項1. ヒドロキシクエン酸を含む植物エキスと、コンドロイチン硫酸及び/又はその塩とを含有する、医薬又は食品組成物。
項2. 前記植物エキスがガルシニアエキス及び/又はハイビスカスエキスである、項1に記載の医薬又は食品組成物。
項3. 前記植物エキスの乾燥重量換算量100重量部中の前記ヒドロキシクエン酸の量が、10~80重量部である、項1又は2に記載の医薬又は食品組成物。
項4. 前記コンドロイチン硫酸及び/又はその塩の平均分子量が477~10万である、項1~3のいずれかに記載の医薬又は食品組成物。
項5. 前記植物エキスの乾燥重量換算量1重量部に対する前記コンドロイチン硫酸及びその塩の総量が、0.0006~3.6重量部である、項1~4のいずれかに記載の医薬又は食品組成物。
項6. 錠剤である、項1~5のいずれかに記載の医薬又は食品組成物。
項7. 前記植物エキスの含有量が60重量%以上である、項1~6のいずれかに記載の医薬又は食品組成物。
項8. ヒドロキシクエン酸を含む植物エキスを含有する医薬又は食品組成物においてヒドロキシクエン酸の減量を抑制する方法であって、前記ヒドロキシクエン酸とともに、コンドロイチン硫酸及び/又はその塩を配合する、減量抑制方法。

本発明によれば、ヒドロキシクエン酸を含む植物エキスを含有する医薬又は食品組成物において、ヒドロキシクエン酸の減量を抑制する製剤処方が提供される。さらに、ヒドロキシクエン酸を含む植物エキスを含有する医薬又は食品組成物を錠剤として製剤する場合には、ヒドロキシクエン酸の減量抑制だけでなく、水分がある条件に晒された場合に生じるひび割れ及び/又は面荒れを抑制する製剤処方も提供される。
定義
本明細書においてヒドロキシクエン酸の「減量」とは、医薬又は食品組成物の保存時にヒドロキシクエン酸の含有量が経時的に低減することをいう。「ヒドロキシクエン酸の減量」の度合いは、例えば、調製直後における医薬又は食品組成物に含有されるヒドロキシクエン酸の重量を100%とした場合の、50℃、RH60%の条件下で3週間保存した後における医薬又は食品組成物に含有されるヒドロキシクエン酸量の相対重量(すなわちヒドロキシクエン酸残存量)(%)で示される。
本明細書においてヒドロキシクエン酸の「減量」とは、医薬又は食品組成物の保存時にヒドロキシクエン酸の含有量が経時的に低減することをいう。「ヒドロキシクエン酸の減量」の度合いは、例えば、調製直後における医薬又は食品組成物に含有されるヒドロキシクエン酸の重量を100%とした場合の、50℃、RH60%の条件下で3週間保存した後における医薬又は食品組成物に含有されるヒドロキシクエン酸量の相対重量(すなわちヒドロキシクエン酸残存量)(%)で示される。
本明細書においてヒドロキシクエン酸に対する「減量抑制」とは、上記の「減量」を抑制することをいう。
本明細書においてヒドロキシクエン酸に対する「減量抑制方法」とは、上記の「減量」を抑制する方法をいう。
本明細書においてヒドロキシクエン酸に対する「減量抑制剤」とは、上記の「減量」を抑制することを目的として用いられる剤をいう。
1.医薬又は食品組成物
本発明の医薬又は食品組成物は、ヒドロキシクエン酸を含む植物エキスと、コンドロイチン硫酸及び/又はその塩とを含有することを特徴とする。

植物エキス
本発明の医薬又は食品組成物は、ヒドロキシクエン酸を含む植物エキスを含有する。このような植物エキスが医薬又は食品組成物中に配合されると、ヒドロキシクエン酸が減量するが、本発明の医薬又は食品組成物は、ヒドロキシクエン酸の減量が抑制されている。
本発明の医薬又は食品組成物は、ヒドロキシクエン酸を含む植物エキスを含有する。このような植物エキスが医薬又は食品組成物中に配合されると、ヒドロキシクエン酸が減量するが、本発明の医薬又は食品組成物は、ヒドロキシクエン酸の減量が抑制されている。
ヒドロキシクエン酸は、2つの不斉中心を有するα-ヒドロキシ三塩基酸(1,2-ジヒドロキシプロパン-1,2,3-トリカルボン酸)であり、2対のジアステレオ異性体又は4つの異なる異性体を形成する。具体的には、ヒドロキシクエン酸としては、(-)ヒドロキシクエン酸、(+)ヒドロキシクエン酸、(-)アロ-ヒドロキシクエン酸、及び(+)アロ-ヒドロキシクエン酸が挙げられる。これらの異性体は、1種単独で使用してもよく、また2種以上を組み合わせて使用してもよい。これらの異性体の中でも、好ましくは(-)ヒドロキシクエン酸が挙げられる。
本発明において用いられる植物エキスの由来となる植物については、ヒドロキシクエン酸を含む限りにおいて特に限定されず、例えば、オトギリソウ科フクギ属のガルシニア種(具体例として、ガルシニア・カンボジア(Garcinia cambogia)、ガルシニア・インディカ(Garcinia indica)、ガルシニア・アトロビリディス(Garcinia atroviridis)、ガルシニア・マンゴスターナ(Garcinia mangostana)、ガルシニア・スベリプティカ(Garcinia subelliptica)等が挙げられる。)、アオイ科フヨウ属のハイビスカス(Hibiscus L.)等の植物が挙げられ、好ましくはガルシニア属が挙げられ、より好ましくはガルシニア・カンボジア(Garcinia cambogia)が挙げられる。植物は、栽培により生産されたものであっても天然より採取されたものであってもよい。
植物エキスは、上記植物のヒドロキシクエン酸を含む部位(例えば、ガルシニア種の果皮、ハイビスカスの花弁等)の抽出物であればよい。植物エキスを得る方法は特に限定されないが、例えば次のようにして得ることができる。植物エキスは、例えば、上記植物のヒドロキシクエン酸を含む部位をそのままの生の状態で又は乾燥物の状態で用意し、そのままの大きさで、または更に必要に応じて裁断または粉砕した後、溶媒抽出、超臨界抽出等の慣用の抽出方法に従って調製することができる。抽出溶媒としては、水(温水及び熱水を含む)、有機溶媒(メタノール、エタノール、n-プロパノール、イソプロパノール、n-ブタノール等の炭素数1~4の低級アルコール;プロピレングリコール、1,3-ブチレングリコール等の多価アルコール;アセトン等のケトン類;ジエチルエーテル、ジオキサン、アセトニトリル、酢酸エチルエステル等のエステル類;キシレン、ベンゼン、クロロホルム等)、これらの混合物が挙げられ、好ましくは、水、低級アルコール、これらの混合物が挙げられ、より好ましくは、温水、熱水などの加熱水が挙げられ、更に好ましくは熱水が挙げられる。これらの溶媒は1種単独で用いてもよく、2種以上を組み合わせて用いてもよい。
得られた植物エキスは、濃縮されない液状の非濃縮エキスの態様であってもよいし、濃縮された液状の軟エキスの態様であってもよいし、非濃縮エキスまたは軟エキスをさらに乾燥処理に供して得たエキス末の態様であってもよい。乾燥処理としては、スプレードライ処理及び凍結乾燥処理が挙げられる。これらの態様の中でも、好ましくはエキス末が挙げられる。
植物エキスの乾燥重量換算量100重量部中のヒドロキシクエン酸の量としては、例えば10~80重量部、好ましくは30~75重量部、より好ましくは40~70重量部、さらに好ましくは50~65重量部、一層好ましくは55~62重量部が挙げられる。
本発明の医薬又は食品組成物における植物エキスの配合量は、ヒドロキシクエン酸の配合量が、例えば20重量%以上、好ましくは30重量%以上、より好ましくは34重量%以上となる量が挙げられる。当該配合量の上限については特に制限されないが、ヒドロキシクエン酸の配合量が、例えば75重量%以下、55重量%以下、45重量%以下、又は35重量%以下となる量が挙げられる。
本発明の医薬又は食品組成物における植物エキスの具体的な配合量については、乾燥重量換算量で、例えば40重量%以上、好ましくは45重量%以上、より好ましくは50重量%以上、更に好ましくは60重量%以上が挙げられる。
本発明の医薬又は食品組成物は、錠剤として製剤される場合、水分がある条件に晒された場合のひび割れ及び/又は面荒れに対する耐性を備えている。通常、ヒドロキシクエン酸を含む植物エキスを含む医薬又は食品組成物が錠剤として製剤される場合、水分がある条件に晒された場合に錠剤のひび割れ及び/又は面荒れが生じやすい。このような水分に対するひび割れ及び/又は面荒れの発生は、ヒドロキシクエン酸を含む植物エキスを比較的多く、具体的には60重量%以上含む医薬又は食品組成物の錠剤で認められやすくなり、特に、ヒドロキシクエン酸を含む植物エキスを68重量%以上含む医薬又は食品組成物の錠剤では顕著になる。錠剤として製剤される場合の本発明の医薬又は食品組成物は、ヒドロキシクエン酸を含む植物エキスを比較的多く含んでいても、水分がある条件に晒された場合のひび割れ及び/又は面荒れを抑制することができる。このような観点から、錠剤として製剤される場合の本発明の医薬又は食品組成物に含まれるヒドロキシクエン酸を含む植物エキスの好適な量としては、好ましくは60重量%以上、特に好ましくは68重量%以上が挙げられる。
当該植物エキスの配合量の上限については特に制限されないが、例えば好ましくは99重量%以下、好ましくは95重量%以下、更に好ましくは92重量%以下、90重量%以下、85重量%以下、80重量%以下、又は75重量%以下が挙げられる。
コンドロイチン硫酸及び/又はその塩
本発明の医薬又は食品組成物は、コンドロイチン硫酸及び/又はその塩を含む。コンドロイチン硫酸及びその塩は、ヒドロキシクエン酸を含む植物エキスを含有する医薬又は食品組成物において、ヒドロキシクエン酸の減量を抑制する。

コンドロイチン硫酸は、N-アセチルガラクトサミン(N-アセチル-D-ガラクトサミンを指す。以下において同様。)及びグルクロン酸(D-グルクロン酸を指す。以下において同様。)又はイズロン酸(L-イズロン酸を指す。以下において同様。)の2糖を反復構造単位とする糖鎖に硫酸が結合した酸性ムコ多糖として公知の化合物である。
コンドロイチン硫酸としては、コンドロイチン硫酸A(構成2糖は、グルクロン酸及びアセチルガラクトサミン4硫酸)、コンドロイチン硫酸B(構成2糖は、イズロン酸2硫酸及びアセチルガラクトサミン4硫酸;デルマタン硫酸とも呼ばれる)、コンドロイチン硫酸C(構成2糖は、グルクロン酸及びアセチルガラクトサミン6硫酸)、コンドロイチン硫酸D(構成2糖は、グルクロン酸2硫酸及びアセチルガラクトサミン6硫酸)、コンドロイチン硫酸E(構成2糖は、グルクロン酸及びアセチルガラクトサミン4,6二硫酸)が挙げられる。これらのコンドロイチン硫酸は、1種単独で使用してもよいし、2種以上を組み合わせて使用してもよい。本発明で用いられるコンドロイチン硫酸は、少なくともコンドロイチン硫酸Aを含むことが好ましい。
コンドロイチン硫酸の塩としては特に限定されないが、例えば、ナトリウム塩、カリウム塩等のアルカリ金属塩;カルシウム塩、マグネシウム塩等のアルカリ土類金属塩;アンモニウム塩;アルギニン、リジン、ヒスチジン、オルニチン等の塩基性アミノ酸塩;モノエタノールアミン塩、ジエタノールアミン塩等のアミン塩等が挙げられる。これらの塩は、1種単独で使用してもよいし、2種以上を組み合わせて使用してもよい。
また、本発明において、コンドロイチン硫酸及びコンドロイチン硫酸の塩のうちいずれか一方を使用してもよいし、コンドロイチン硫酸及びコンドロイチン硫酸の塩の両方を組み合わせて使用してもよい。これらの中でも、ヒドロキシクエン酸の減量抑制効果を高める観点から、好ましくはコンドロイチン硫酸が挙げられる。
コンドロイチン硫酸及びその塩の平均分子量としては、例えば477~10万が挙げられ、好ましくは477~10万、より好ましくは477~1万、さらに好ましくは480~5000、一層好ましくは485~1000、より一層好ましくは490~800、特に好ましくは490~550が挙げられる。なお、本発明において、コンドロイチン硫酸及びその塩の平均分子量は、光散乱法を用いたゲルパーミエーションクロマトグラフ(コンドロイチン硫酸標準品(生化学工業株式会社製、サメ軟骨由来又はクジラ軟骨由来のコンドロイチン硫酸標準品)を標準物質とする。)により測定される重量平均分子量である。
コンドロイチン硫酸及びその塩としては、化学合成したものを用いてもよいし、天然物由来の材料から抽出又は精製等したものであってもよい。また、コンドロイチン硫酸又はその塩としては、試薬として市販されているコンドロイチン硫酸又はその塩を使用してもよい。
コンドロイチン硫酸又はその塩を天然物由来の材料から抽出又は精製等して製造する方法としては、特に限定されず、天然物由来の材料から公知の方法で抽出、精製等する方法が挙げられる。コンドロイチン硫酸を抽出するための天然物由来の材料としては、例えば、豚、牛等の哺乳動物の軟骨;鶏等の鳥類の軟骨;サケ、エイ、サメ等の魚類の軟骨等が挙げられ、好ましくは哺乳動物の軟骨が挙げられ、より好ましくは豚軟骨が挙げられる。
本発明の医薬又は食品組成物において、コンドロイチン硫酸又はその塩の含有量としては特に限定されないが、例えば、植物エキス1重量部(乾燥重量換算量)に対するコンドロイチン硫酸及びその塩の総量として、0.0006~3.6重量部、好ましくは0.0009~2重量部、より好ましくは0.01~1重量部、さらに好ましくは0.1~1重量部、一層好ましくは0.3~1重量部、より一層好ましくは0.7~1重量部又は0.8~1重量部が挙げられる。
本発明の医薬又は食品組成物におけるコンドロイチン硫酸又はその塩の具体的な配合量については、特に制限されず、好ましくは、植物エキスの配合量との比率が上記の比率となるように設定すればよい。具体的なコンドロイチン硫酸又はその塩の配合量の例としては、総量で、0.01~30重量%、好ましくは0.1~20重量%、より好ましくは0.3~15重量%、更に好ましくは1~10重量%が挙げられる。
その他の成分
本発明の医薬又は食品組成物は、上記成分以外に、本発明の効果を損なわない範囲で、適用形態に応じて他の成分を含有していてもよいし、含有していなくてもよい。このような他の成分としては、例えば、生理活性物質、及び添加物等が挙げられる。

生理活性物質としては、例えば、コラーゲン、II型コラーゲン、非変性活性2型コラーゲン、コラーゲンペプチド、メチルスルホニルメタン(MSM)、S-アデノシルメチオニン、クレアチン、テアニン、ピぺリン、マスリン酸、5-アミノレブリン酸リン酸塩、キャッツクロー、ブラックジンジャー、ボスウェリアセラータ、アーティチョーク、アミノ酸、ビタミンA、ビタミンB1、ビタミンB2、ビタミンB6、ビタミンB12、ビタミンC、ビタミンD2、ビタミンD3、ビタミンE,ビタミンK、葉酸、ヒアルロン酸、カツオ由来エラスチンペプチド、イミダゾールジペプチド、ケルセチン配糖体、クリルオイル由来EPA・DHA、モリンガ種子由来グルコモリンギン、イタドリ、デビルズクロー、鶏足由来ヒアルロン酸産生促進剤(HAS-II)、大豆イソフラボン、β-クリプトキサンチン、ボーンペップ、濃縮乳清活性たんぱく質(CBP)、3-ヒドロキシ-3-メチルブチレート(HMB)、カルシウムビス-3-ヒドロキシ-3-メチルブチレートモノハイドレート(HMBカルシウム)、マルトビオン酸カルシウム、カルシウム、マグネシウム、亜鉛、鉄、セレン、カリウム、エストロゲン、カルシトニン、アスピリン、ステロイド性消炎剤、非ステロイド性消炎剤等が挙げられる。これらの生理活性物質は、1種を選択して使用してもよく、2種以上を組み合わせて使用してもよい。
添加物としては、薬学的又は食品学的に許容される賦形剤、崩壊剤、希釈剤、滑沢剤、着香剤、着色剤、甘味剤、矯味剤、懸濁剤、湿潤剤、乳化剤、分散剤、補助剤、防腐剤、緩衝剤、結合剤、安定剤、増量剤、増粘剤、pH調整剤、界面活性剤、コーティング剤、栄養成分等が挙げられる。これらの添加物は、1種を選択して使用してもよく、2種以上を組み合わせて使用してもよい。
本発明の医薬又は食品組成物が他の成分を含む場合、減量抑制効果を向上させる観点並びに錠剤として製剤された場合にあっては水分がある条件に晒された場合のひび割れ及び/又は面荒れを抑制する効果を向上させる観点から、他の成分としては、好ましくは滑沢剤及び/又は賦形剤が挙げられる。滑沢剤としては、好ましくはステアリン酸、ステアリン酸カルシウム、ステアリン酸マグネシウムが挙げられ、より好ましくはステアリン酸カルシウム(日本薬局方)が挙げられる。賦形剤としては、乳糖、マンニトール、エリスリトール、結晶セルロース、でんぷん等が挙げられ、好ましくは結晶セルロース及びでんぷんが挙げられる。
本発明の医薬又は食品組成物が他の成分を含む場合、その含有量の下限としては、当該他の成分の配合目的が達成される程度であればよいが、減量抑制効果を向上させる観点並びに錠剤として製剤された場合にあっては水分がある条件に晒された場合のひび割れ及び/又は面荒れを抑制する効果を向上させる観点から、総量で、好ましくは10重量%以上、より好ましくは18重量%以上、さらに好ましくは25重量%以上、一層好ましくは28重量%以上が挙げられる。また、その他の成分の含有量の上限としては、減量抑制効果を向上させる観点並びに錠剤として製剤された場合にあっては水分がある条件に晒された場合のひび割れ及び/又は面荒れを抑制する効果を向上させる観点から、総量で、好ましくは40重量%以下、好ましくは30重量%以下、20重量%以下、好ましくは17重量%以下、又は15重量%以下が挙げられる。また、他の成分として賦形剤を含む場合、賦形剤の含有量としては、例えば1~39重量%、好ましくは10~30重量%、より好ましくは15~20重量%が挙げられる(結晶セルロースを含む場合、結晶セルロースの含有量としては、例えば1~15重量%、好ましくは1.5~10重量%、より好ましくは2~5重量%が挙げられ、でんぷんを含む場合、でんぷんの含有量としては、例えば1~38重量%、好ましくは10~31重量%、より好ましくは15~28重量%が挙げられる)。他の成分として滑沢剤を含む場合、滑沢剤の含有量としては、例えば0.5~25重量%、好ましくは1~10重量%、より好ましくは1~5重量%が挙げられる。
製剤形態等
本発明の医薬又は食品組成物は、上記植物エキスとコンドロイチン硫酸及び/又はその塩とを含み、且つ医薬又は食品組成物として調製される限り、その形態及び性状は特に限定されない。

また、本発明の医薬又は食品組成物の性状は、液状であってもよいし、固形状であってもよい。液状の例としては、液剤、飲料剤、乳剤、懸濁剤、酒精剤、シロップ剤、エリキシル剤、軟エキス剤等を含む)等が挙げられ、固形状の例としては、錠剤(好ましくは素錠剤)、丸剤、散剤、細粒剤、顆粒剤、トローチ剤、チュアブル剤、カプセル剤(ハードカプセル及びソフトカプセルを含む)等が挙げられる。これらの中でも、減量抑制効果を向上させる観点から、好ましくは固形状が挙げられる。固形状の中でも、減量抑制効果を向上させる観点から、好ましくは、錠剤、丸剤、散剤、細粒剤、顆粒剤、トローチ剤、カプセル剤が挙げられ、より好ましくは錠剤、散剤、細粒剤、顆粒剤、カプセル剤が挙げられ、更に好ましくは錠剤、散剤、カプセル剤が挙げられ、一層好ましくは錠剤、散剤が挙げられる。また、本発明の医薬又は食品組成物が錠剤として製剤される場合、ヒドロキシクエン酸を含む植物エキスを比較的多く含んでいても、水分がある条件に晒された場合のひび割れ及び/又は面荒れを抑制することができることから、特に好適な製剤形態として、錠剤が挙げられる。
本発明の医薬又は食品組成物が錠剤である場合、錠剤の包装形態としては、PPT包材等を用いた個包装であってもよいし、パウチ容器又は広口容器等の、開封のたびに外気の湿気に触れる容器内に所定量がまとめて収容される形態(バラ包装)であってもよい。本発明の医薬又は食品組成物が錠剤である場合、水分がある条件に晒された場合のひび割れ及び/又は面荒れ耐性に優れているため、外気の湿気に繰り返し触れる条件に晒されても、効果的にひび割れ及び/又は面荒れを抑制することができる。このような観点から、本発明の医薬又は食品組成物が錠剤である場合、錠剤の好ましい包装形態として、バラ包装が挙げられる。
本発明の医薬又は食品組成物は、一般飲食品、保健機能食品(特定保健用食品、栄養機能食品、機能性表示食品、サプリメント等を含む)、病者用食品、医薬品、医薬部外品として使用することができ、また、食品、医薬品、医薬部外品等への添加剤等として使用することもできる。特に、日常的及び/又は継続的に気軽に摂取させる観点から、サプリメントとして使用することが好ましい。
製造方法
本発明の医薬又は食品組成物の製造方法は、植物エキスとコンドロイチン硫酸及び/又はその塩と、必要に応じて配合されるその他の成分とを用いて、各種形態及び性状、並びに使用目的に応じ、従来公知の通常の製剤手順に従えばよい。

2.減量抑制方法及び減量抑制剤
上述のとおり、コンドロイチン硫酸及びその塩は、上記植物エキスを含む医薬又は食品組成物におけるヒドロキシクエン酸の減量を抑制できる。従って、本発明は、ヒドロキシクエン酸を含む植物エキスを含有する医薬又は食品組成物においてヒドロキシクエン酸の減量を抑制する方法であって、前記ヒドロキシクエン酸とともに、コンドロイチン硫酸及び/又はその塩を配合する、減量抑制方法;並びに、ヒドロキシクエン酸を含む植物エキスを含有する医薬又は食品組成物中の前記ヒドロキシクエン酸に対する減量抑制剤であって、コンドロイチン硫酸及び/又はその塩を含む、減量抑制剤も提供する。
上述のとおり、コンドロイチン硫酸及びその塩は、上記植物エキスを含む医薬又は食品組成物におけるヒドロキシクエン酸の減量を抑制できる。従って、本発明は、ヒドロキシクエン酸を含む植物エキスを含有する医薬又は食品組成物においてヒドロキシクエン酸の減量を抑制する方法であって、前記ヒドロキシクエン酸とともに、コンドロイチン硫酸及び/又はその塩を配合する、減量抑制方法;並びに、ヒドロキシクエン酸を含む植物エキスを含有する医薬又は食品組成物中の前記ヒドロキシクエン酸に対する減量抑制剤であって、コンドロイチン硫酸及び/又はその塩を含む、減量抑制剤も提供する。
減量抑制方法及び減量抑制剤において、植物エキス及びコンドロイチン硫酸及び/又はその塩の詳細、含有の有無を問わないその他の成分、製剤形態、製造方法等の詳細については、上記「1.医薬又は食品組成物」と同様である。
以下に実施例を示して本発明をより具体的に説明するが、本発明はこれらに限定されるものではない。
試験例1
表1及び表2に示す組成の医薬又は食品組成物(以下において、製剤と記載する。)を、それぞれ、散剤及び錠剤(素錠)の形態で調製した。各成分の配合のために用いた材料の詳細については次の通りである。
・ガルシニアエキス
ガルシニア・カンボジア(Garcinia cambogia)の果皮を乾燥させ、10倍量の水を加えて熱水抽出した。得られた抽出液をろ過した後、ロータリーエバポレーターにて減圧濃縮し、得られた濃縮液を更に噴霧乾燥させた。乾燥させた抽出物を粉砕して、粉末状のガルシニアエキスを得た。なお、このガルシニアエキス中のヒドロキシクエン酸((-)ヒドロキシクエン酸)含有量は、60重量%である。
・コンドロイチン硫酸素材(ブタ軟骨抽出物、コンドロイチン硫酸(主たるコンドロイチン硫酸は、コンドロイチン硫酸A)含有量のUSP-HPLC法による測定値84重量%、重量平均分子量499.38)
・その他の成分
結晶セルロース(旭化成ケミカルズ社製)
ステアリン酸カルシウム(日本薬局方ステアリン酸カルシウム)
表1及び表2に示す組成の医薬又は食品組成物(以下において、製剤と記載する。)を、それぞれ、散剤及び錠剤(素錠)の形態で調製した。各成分の配合のために用いた材料の詳細については次の通りである。
・ガルシニアエキス
ガルシニア・カンボジア(Garcinia cambogia)の果皮を乾燥させ、10倍量の水を加えて熱水抽出した。得られた抽出液をろ過した後、ロータリーエバポレーターにて減圧濃縮し、得られた濃縮液を更に噴霧乾燥させた。乾燥させた抽出物を粉砕して、粉末状のガルシニアエキスを得た。なお、このガルシニアエキス中のヒドロキシクエン酸((-)ヒドロキシクエン酸)含有量は、60重量%である。
・コンドロイチン硫酸素材(ブタ軟骨抽出物、コンドロイチン硫酸(主たるコンドロイチン硫酸は、コンドロイチン硫酸A)含有量のUSP-HPLC法による測定値84重量%、重量平均分子量499.38)
・その他の成分
結晶セルロース(旭化成ケミカルズ社製)
ステアリン酸カルシウム(日本薬局方ステアリン酸カルシウム)
調製直後の製剤と、50℃、RH60%の条件下で当該製剤を3週間保存した後の製剤とについて、ヒドロキシクエン酸量をHPLC分析により定量した。調製直後におけるヒドロキシクエン酸の重量を100%とした場合の、保存後におけるヒドロキシクエン酸量の相対重量(%)をヒドロキシクエン酸残存量として導出した。結果を表1及び表2に示す。
比較例1と実施例1、及び比較例2と実施例2との対比から明らかな通り、ガルシニアエキスにコンドロイチン硫酸を組み合わせることで、ヒドロキシクエン酸の減量が抑制されたことが示された。なお、ガルシニアエキスの代わりに(-)ヒドロキシクエン酸(単独化合物)を用いた場合には、比較例1及び2のようなヒドロキシクエン酸の減量は認められなかったことから、製剤後におけるヒドロキシクエン酸の減量の課題が生じること及びコンドロイチン硫酸によるヒドロキシクエン酸の減量抑制の効果が奏されることは、植物エキスを配合した製剤に特有の特性であることが分かった。
試験例2
表3に示す組成の医薬又は食品組成物を、それぞれ、錠剤(素錠)の形態で調製した。表3に示す他の成分は、結晶セルロース、ステアリン酸カルシウム、及びデンプンの組み合わせである。製剤条件は、各成分を混合し、篩過(メッシュサイズ:500μm)して造粒物を得た後に、各造粒物をテーブルプレス機(商品名:TB-20H、エヌピーエーシステム株式会社製)により圧縮成型(打錠圧力:直径10mmに対し1t)し、錠剤を得た。得られた錠剤の1錠当たりの重量は180mg、直径10mm、厚み5mmであった。なお、各成分の配合のために用いた材料の詳細については、試験例1と同じである。
表3に示す組成の医薬又は食品組成物を、それぞれ、錠剤(素錠)の形態で調製した。表3に示す他の成分は、結晶セルロース、ステアリン酸カルシウム、及びデンプンの組み合わせである。製剤条件は、各成分を混合し、篩過(メッシュサイズ:500μm)して造粒物を得た後に、各造粒物をテーブルプレス機(商品名:TB-20H、エヌピーエーシステム株式会社製)により圧縮成型(打錠圧力:直径10mmに対し1t)し、錠剤を得た。得られた錠剤の1錠当たりの重量は180mg、直径10mm、厚み5mmであった。なお、各成分の配合のために用いた材料の詳細については、試験例1と同じである。
得られた錠剤に水を1滴(25μL)垂らして表面を濡らし、50℃の恒温槽で全開放条件下で1週間保管した。保管後、錠剤の外観(ひび割れの程度及び面荒れの程度)を、以下の基準に基づいて評価した。結果を表3に示す。
<ひび割れ>
× 錠剤表面を完全に横切るほどの大きな亀裂が認められる。
△ 少しひびが入っているのが認められる。
○ 保管前と変わらない。
<面荒れ>
× 表面に浮き出たぶつぶつが認められる。
△ 表面のツヤが失われている。
○ 保管前と変わらず表面にツヤがある。
× 錠剤表面を完全に横切るほどの大きな亀裂が認められる。
△ 少しひびが入っているのが認められる。
○ 保管前と変わらない。
<面荒れ>
× 表面に浮き出たぶつぶつが認められる。

表3に示す通り、ガルシニアエキスを含む錠剤は、水分がある条件に晒されるとひび割れ及び面荒れを生じる(比較例3~5)が、ガルシニアエキスを含む錠剤にコンドロイチン硫酸を配合することで、当該ひび割れ及び面荒れを抑制できた(実施例3~9)。特に、当該ひび割れ及び面荒れが、比較例4、5の錠剤で格段顕著に生じることに鑑みると、実施例3~9の中でも、実施例8、9の錠剤におけるひび割れ及び面荒れ抑制効果が各段顕著であることが判明した。
なお、実験例3~9の錠剤は、比較例3~5の錠剤と比較して、上記試験例1での保存条件に供した場合に、コンドロイチン硫酸によるヒドロキシクエン酸の減量抑制効果も認められた。
処方例
表4、表5及び表6に示す処方の散剤又は素錠を調製した。使用した成分のうち、ガルシニアエキス、コンドロイチン硫酸、ステアリン酸Ca、及び結晶セルロースの配合に用いられた材料は、上記試験例1と同じである。また、ハイビスカスエキスは、次の方法により調製したものを用いた。
表4、表5及び表6に示す処方の散剤又は素錠を調製した。使用した成分のうち、ガルシニアエキス、コンドロイチン硫酸、ステアリン酸Ca、及び結晶セルロースの配合に用いられた材料は、上記試験例1と同じである。また、ハイビスカスエキスは、次の方法により調製したものを用いた。
(ハイビスカスエキスの調製)
ハイビスカス(Hibiscus sabdariffa L. var ruber (Malvaceae ))の花弁を乾燥させ、10倍量の含水エタノールを加えて抽出した。得られた抽出液をろ過した後、ロータリーエバポレーターにて減圧濃縮し、得られた濃縮液を更に噴霧乾燥させた。乾燥させた抽出物を粉砕して、粉末状のハイビスカスエキスを得た。ハイビスカスエキス中の(+)HCA・フリー体(すなわち(+)アロ-ヒドロキシクエン酸フリー体)の含有量は60重量%未満である。
ハイビスカス(Hibiscus sabdariffa L. var ruber (Malvaceae ))の花弁を乾燥させ、10倍量の含水エタノールを加えて抽出した。得られた抽出液をろ過した後、ロータリーエバポレーターにて減圧濃縮し、得られた濃縮液を更に噴霧乾燥させた。乾燥させた抽出物を粉砕して、粉末状のハイビスカスエキスを得た。ハイビスカスエキス中の(+)HCA・フリー体(すなわち(+)アロ-ヒドロキシクエン酸フリー体)の含有量は60重量%未満である。
いずれの散剤又は素錠についても、製剤後に上記試験例1での保存条件に供してもコンドロイチン硫酸によりヒドロキシクエン酸の減量が抑制されていた。また、素錠については、上記試験例2での条件に供しても、コンドロイチン硫酸によりヒドロキシクエン酸のひび割れ及び面荒れが認められなかった。
Claims (9)
- ヒドロキシクエン酸を含む植物エキスと、コンドロイチン硫酸及び/又はその塩とを含有する、医薬又は食品組成物。
- 前記植物エキスがガルシニアエキス及び/又はハイビスカスエキスである、請求項1に記載の医薬又は食品組成物。
- 前記植物エキスの乾燥重量換算量100重量部中の前記ヒドロキシクエン酸の量が、10~80重量部である、請求項1に記載の医薬又は食品組成物。
- 前記コンドロイチン硫酸及び/又はその塩の平均分子量が477~10万である、請求項1に記載の医薬又は食品組成物。
- 前記植物エキスの乾燥重量換算量1重量部に対する前記コンドロイチン硫酸及びその塩の総量が、0.0006~3.6重量部である、請求項1に記載の医薬又は食品組成物。
- 錠剤である、請求項1に記載の医薬又は食品組成物。
- 前記植物エキスの含有量が60重量%以上である、請求項1に記載の医薬又は食品組成物。
- ヒドロキシクエン酸を含む植物エキスを含有する医薬又は食品組成物においてヒドロキシクエン酸の減量を抑制する方法であって、前記ヒドロキシクエン酸とともに、コンドロイチン硫酸及び/又はその塩を配合する、減量抑制方法。
