Description
本発明は、動物細胞培養用の培地添加剤、動物細胞培養用の培地添加剤の製造方法、および細胞の培養方法に関する。
培養細胞は、本来生体内で起こる生命現象を生体外で再現することができるため、生命現象に関するメカニズムの解明、機能分子の産生、および薬剤の評価等に有用であり、研究、産業、製薬、および臨床を含む様々な分野において、多様な生物種で利用されている。
細胞を培養するためには、培養対象の細胞に適した培地を使用する必要がある。細胞の種類および実験系に応じて様々な培地が使用されており、培地には、アミノ酸、ビタミン、無機塩、糖、成長因子およびホルモン等の様々な成分が含まれる。
特に、ヒト等の哺乳類の細胞、ならびにショウジョウバエおよびカイコ等の昆虫の細胞を含む動物細胞を培養する場合には、アミノ酸、ビタミン、無機塩および糖等の基本培地に血清を添加することが必須である。血清は、多数の細胞増殖因子およびホルモン等の供給源であることが知られているが、血清中のすべての有効成分が同定されているわけではない。
動物細胞の培養用培地に添加する血清としては、ウシ胎児血清(Fetal Bovine Serum、FBS)、ウシ血清、ウマ血清、新生仔ウシ血清、ヤギ血清、ウサギ血清、ブタ血清、およびニワトリ血清等が使用されている。培養する細胞と同種の動物由来の血清を使用する場合もあるが、必ずしも入手が容易ではなく、また汎用性の観点からも、FBSが最も一般的に使用されている。多くの種類の細胞、例えば、ヒト等の哺乳類の細胞だけでなく、ショウジョウバエおよびカイコ等の昆虫細胞の培養においても、FBSは広く使用されている。乳牛および肉牛の生産において多くのオス牛が堕胎されており、FBSはその副産物として製造されているため、比較的安価に入手できるという利点がある。
しかし、FBSを利用する際に最も大きな問題となるのが、人獣共通感染症のウイルス等の病原体の混入である。FBSの原料となるのは、FBSの生産用に管理された牛ではなく、前述のように一般の畜産農家で堕胎されたオス胎児である。FBSに混入するリスクのある病原体は10種類ほど存在しているが、必ずしもそのすべての病原体の混入が検査されているわけではなく、病原体が混入した製品が流通するおそれがある。もし培地に病原体が混入すれば、その培地で培養した試料のすべてが病原体で汚染されてしまう。
FBSの原料となる牛の飼育地域で病原体に起因する牛の感染症が流行した場合、FBSの供給が絶たれるという問題がある。細胞培養用血清は海外輸入に大きく依存しており、感染症の流行等によりサプライチェーンが崩壊し、安定的に供給できない事態が起こりうる。また、牛は遺伝的に均質な系統が確立されているわけではないため、個体差が大きく、FBSの成分等の品質もロットによる差が大きいという問題もある。
したがって、均質性が高く安定的に供給できる血清が求められている。現在、FBSの代替品として、合成血清が販売されている。合成血清は、数十種類の細胞増殖因子を遺伝子組み換え大腸菌で合成し、精製して混合したものである。しかし、前述のように血清の有効成分がすべて同定されているわけではないため、合成血清を使用した場合の再現度は不十分である。また、合成血清は非常に高価である。
FBSの代替として、様々な生物由来の成分を含有する添加剤が提案されている。例えば、特許文献1および特許文献2では、魚類由来の成分を含有する動物細胞用の培地添加剤が提案されている。特許文献1には、添加剤を添加した培地を用いた臍帯由来ヒト血管内皮細胞の培養では、FBSと同等の増殖活性がみられたことが記載されている。また、非特許文献1には、カイコ由来の血清成分を添加した培地を用いてカイコの細胞を培養することが記載されている。

本発明は上述の事情に鑑みてなされたものであり、種々の細胞の培養に使用可能で、病原体混入のリスクがなく安定的に供給できる動物細胞培養用の培地添加剤、および動物細胞培養用の培地添加剤の製造方法を提供することを目的とする。また、種々の細胞の培養に使用可能で、病原体混入のリスクがない細胞の培養方法を提供することを目的とする。

以下、本発明の実施の形態について詳細に説明する。なお、本発明は下記の実施の形態及び図面によって限定されるものではない。なお、下記の実施の形態において、“有する”、“含む”または“含有する”といった表現は、“からなる”または“から構成される”という意味も包含する。
本実施の形態に係るカイコ細胞を除く動物細胞培養用の培地添加剤は、カイコの血清に由来する成分を含む。カイコ(Bombyx mori)は、カイコガ科に属する昆虫である。「血清に由来する成分」とは、単離した血清に含まれる1種以上の成分を意味し、「血清に由来する成分」には、単離した血清から一部の成分を除去したものが包含される。
カイコ等の昆虫は牛等の哺乳類に比べ非常に小型であり、また哺乳類とは進化的に非常に離れているため、昆虫血清をヒト等の哺乳類の細胞の培養に利用しようという試みはなされてこなかった。しかし、カイコは、ヒト等の哺乳類と共通の病原体を持たないため、ヒト等の哺乳類の細胞を培養する際に、添加剤に由来する病原体の混入リスクを回避することができる。さらに、遺伝的に均質な系統のカイコを用いることで高い均質性を確保できる。
本実施の形態に係る培地添加剤の製造方法の一例について説明する。培地添加剤の製造方法は、抽出工程と、添加工程と、分離工程と、第1の加熱工程と、除去工程と、を含む。
抽出工程では、カイコ幼虫から血液を抽出する。血液はカイコ幼虫を遠心分離することで抽出できる。カイコ幼虫は、特に限定されないが、例えば、5齢幼虫である。
カイコの血液が体外に抽出された後、酸化酵素の一種であるフェノールオキシダーゼの作用によりメラニンが生成される。メラニンの生成過程においてキノン類が生成されることが知られており、これらは反応性が高く、細胞毒性を示す。そのため、カイコの血清をそのまま細胞培養用の培地添加剤として用いることは好ましくない。このため、添加工程では、カイコから抽出した血液に還元剤を添加する。
添加工程において使用する還元剤としては、フェニルチオウレア(phenylthiourea、PTU)、チオ硫酸ナトリウム、L-グルタチオン、L-システイン、L-アスコルビン酸、およびクエン酸等が例示される。PTUおよびチオ硫酸ナトリウムは、フェノールオキシダーゼと銅イオンとの結合を阻害することによりフェノールオキシダーゼの活性を阻害する。そのため、フェノールオキシダーゼだけでなく、細胞における金属イオン依存性の酵素反応にも影響するおそれがある。したがって、好ましくは、還元剤は、フェノールオキシダーゼの活性を直接阻害する物質ではなく、メラニン生合成にのみ関係する代謝物であるドーパキノンを阻害する物質である。例えば、還元剤はL-グルタチオンまたはL-システインである。最も好ましくは、還元剤はL-システインである。
還元剤は、メラニンの生成を抑制し、かつ細胞の増殖を阻害しない濃度であることが好ましい。還元剤がL-グルタチオンである場合、培地添加後のL-グルタチオンの終濃度は、好ましくは2.0~5.0mMである。L-グルタチオンの終濃度は、より好ましくは2.5mM~3.0mMである。還元剤がL-システインである場合、培地添加後のL-システインの終濃度は、好ましくは0.1mM~12.5mM、より好ましくは0.5mM~2.0mM、さらに好ましくは1.2mM~1.6mMである。
分離工程では、血液から血清を遠心分離等により分離する。遠心分離の条件は適宜調整されるが、例えば、1210×g、4℃で15分間である。
第1の加熱工程では、血液から分離した血清成分を加熱する。血清成分を加熱することで、フェノールオキシダーゼ等のメラニン化関連酵素を失活させることができる。第1の加熱工程における加熱温度は50~90℃程度である。好ましくは、加熱温度は53~80℃であり、より好ましくは55~57℃である。第1の加熱工程における加熱時間は10分間以上である。好ましくは、加熱時間は25~60分間であり、より好ましくは30分間である。第1の加熱工程では、例えば、血清成分は56℃で30分間加熱される。
本実施の形態に係る培地添加剤の製造方法には、第1の加熱工程において変性したタンパク質を除去する除去工程をさらに含むことができる。タンパク質を除去する方法は、当分野で知られる任意のタンパク質除去方法を使用することができ、濾過、透析、塩析、遠心分離、吸着、抽出、およびクロマトグラフィー等が例示される。好ましくは、タンパク質を除去する方法は濾過であり、例えば孔径0.22μmのステリトップ(Merck Millipore社製)を用いることができる。
なお、除去工程で得られた上清を透析して、L-システインの濃度を低くしてもよい。また、得られた血清成分に対して滅菌処理を行ってもよい。
本実施の形態に係る培地添加剤の製造方法は、除去工程の後に、血清成分をさらに加熱する、第2の加熱工程をさらに含むことができる。第2の加熱工程における加熱温度は、56~90℃、好ましくは65~80℃である。第2の加熱工程における加熱時間は10分間以上である。好ましくは、加熱時間は25~60分間であり、より好ましくは30分間である。第2の加熱工程では、例えば、血清成分は65℃で30分間加熱される。第2の加熱工程の後に、上記除去工程と同様に、変性したタンパク質を除去してもよい。
本実施の形態に係る培地添加剤は、上述の抽出、添加、分離、加熱、および除去工程を経て得られるカイコの血清に由来する成分そのままでもよいし、当分野で知られる手法によりカイコの血清に由来する成分を濃縮または希釈したものであってもよい。培地添加剤の形態は、液体または固体であってもよく、当分野で知られる手法を用いて、乾燥粉末、散剤、または錠剤等、任意の形態としてもよい。
本実施の形態に係る培地添加剤は、カイコの血清に由来する成分および還元剤以外に、任意の他の成分をさらに含んでいてもよい。培地添加剤に含み得る他の成分としては、限定されないが、塩類、アミノ酸、ペプチド、タンパク質、動物または植物由来の抽出物、緩衝剤、ビタミン、ホルモン、脂質、糖、浸透圧調節剤、増殖補助因子、および核酸等が例示される。
カイコの血清に由来する成分および還元剤以外の他の成分としては、具体的には、L-アラニン、L-アルギニン、L-アスパラギン、L-アスパラギン酸、L-システイン、L-シスチン、L-グルタミン、L-グルタミン酸、グリシン、L-ヒスチジン、L-イソロイシン、L-ロイシン、L-リジン、L-メチオニン、L-オルニチン、L-フェニルアラニン、L-プロリン、L-セリン、L-スレオニン、L-トリプトファン、L-チロシン、L-バリン等、好ましくはL-アラニン、L-アルギニン、L-アスパラギン、L-アスパラギン酸、L-グルタミン、L-グルタミン酸、グリシン、L-ヒスチジン、L-イソロイシン、L-ロイシン、L-リジン、L-メチオニン、L-フェニルアラニン、L-プロリン、L-セリン、L-スレオニン、L-トリプトファン、L-チロシン、およびL-バリン等のアミノ酸;i-イノシトール、ビオチン、葉酸、リポ酸、ニコチンアミド、ニコチン酸、p-アミノ安息香酸、パントテン酸カルシウム、塩酸ピリドキサール、塩酸ピリドキシン、リボフラビン、塩酸チアミン、ビタミンB12、およびアスコルビン酸等のビタミン;塩化コリン、酒石酸コリン、リノール酸、オレイン酸、リノレン酸、およびコレステロール等の脂質;グルコース、ガラクトース、マンノース、およびフルクトース等の炭素源;塩化ナトリウム、塩化カリウム、および硝酸カリウム等、好ましくは塩化ナトリウム等の浸透圧調節剤;EDTA鉄、クエン酸鉄、塩化第一鉄、塩化第二鉄、硫酸第一鉄、硫酸第二鉄、および硝酸第二鉄等の鉄源;炭酸水素ナトリウム、塩化カルシウム、リン酸二水素ナトリウム、HEPES、およびMOPS等の緩衝剤などが例示される。
本実施の形態に係る培地添加剤は、サイトカイン、アルブミン、および宿主細胞由来タンパク質等のタンパク質;硫酸銅、硫酸マンガン、硫酸亜鉛、硫酸マグネシウム、塩化ニッケル、塩化スズ、塩化マグネシウム、および亜ケイ酸ナトリウム等の微量金属元素;Tween(登録商標)80、ポリソルベート、およびプルロニック(登録商標)F68等の界面活性剤;Cl-、Br-、NO2-、およびSO42-等の無機アニオン、Li+、Na+、K+、およびMg2+等の無機カチオン;コハク酸、およびピルビン酸等の有機酸、低級アミン類を含む無機物;インスリン、トランスフェリン、インスリン様成長因子(insulin-like growth factor、IGF)、表皮成長因子(epidermal growth factor、EGF)、繊維芽細胞成長因子(fibroblast growth factor、FGF)、血小板由来成長因子(platelet-derived growth factor、PDGF)、トランスフォーミング成長因子(transforming growth factor、TGF)-α、塩酸エタノールアミン、亜セレン酸ナトリウム、レチノイン酸、および塩酸プトレッシン等の増殖等の増殖因子;デオキシアデノシン、デオキシシチジン、デオキシグアノシン、アデノシン、シチジン、グアノシン、およびウリジン等のヌクレオシド;ストレプトマイシン、ペニシリンGカリウム、およびゲンタマイシン等の抗生物質;フェノールレッド等のpH指示薬;2-メルカプトエタノール、およびシメチコン等の添加物等をさらに含んでもよい。
本実施の形態に係る培地添加剤は、当分野で知られる任意の動物細胞培養用の培地に添加して使用することができる。好ましくは、当該培地添加剤は、無血清培地に添加される。動物細胞培養用の培地は、培養する細胞に応じて適した培地を選択することができる。培地の成分は、塩類、アミノ酸、ペプチド、タンパク質、動物または植物由来の抽出物、緩衝剤、ビタミン、ホルモン、脂質、糖、浸透圧調節剤、増殖補助因子、および核酸等の、培地の成分として知られる任意の物質を含有していてもよい。当分野で知られる動物細胞培養用の培地としては、BME(Basal Medium Eagle)培地、MEM(Minimum Essential Medium)培地、DMEM(Dulbecco’s Modified Eagle Medium)培地、199培地、RPMI(Roswell Park Memorial Institute)1640培地、ハムF-12培地、シュナイダー昆虫培地、およびグレース昆虫培地等が例示される。通常、これらの動物細胞用の培地には、FBS等の血清を添加して使用されるが、FBS等の血清に替えて、本実施の形態に係る培地添加剤を使用することができる。
本実施の形態に係る培地添加剤は、カイコの血清に由来する成分の終濃度が20%(v/v)以下となるように培地に添加される。好ましくは、培地におけるカイコの血清に由来する成分の終濃度は1~15%(v/v)または2~12%(v/v)であって、好ましくは10%(v/v)である。
動物細胞培養用の培地は、通常、哺乳類の細胞ではpH7.2~7.4、昆虫の細胞ではpH6.0であることが好ましい。本実施の形態に係る培地添加剤の添加後に、HClまたはNaOH等を加えて、培養する細胞の至適pHとなるように調整してもよい。
本実施の形態に係る培地添加剤を添加した動物細胞培養用の培地を用いて培養される細胞は、カイコ細胞以外の動物細胞であれば特に限定されない。細胞は、例えば、ヒト、サル、牛、ヒツジ、ブタ、ウマ、ウサギ、マウス、ラット、ハムスター、イヌ、およびネコ等の哺乳類、鶏、およびアヒル等の鳥類、爬虫類、両生類、魚類、およびショウジョウバエ等のカイコを除く昆虫の細胞である。細胞の種類は、あらゆる発生段階のあらゆる組織由来の細胞であってもよく、具体的には、ハムスター腎臓由来BHK細胞、イヌマクロファージ由来DH82細胞、アフリカミドリザル腎臓由来Vero細胞、ヒト乳腺由来MCF7細胞等の動物組織由来の細胞、ならびにヒト由来初代培養細胞、ラット由来初代培養細胞、およびマウス由来初代培養細胞等の初代培養細胞等が例示される。
また、細胞は胚性幹細胞(Embryonic Stem Cell、ES細胞)または人工多能性幹細胞(induced pluripotent stem cell、iPS細胞)等の人工的に作製した幹細胞であってもよい。また、細胞は、当分野で知られる方法により外来のDNAを導入したトランスジェニック動物由来の細胞であってもよいし、当分野で知られる方法により外来のDNAまたはRNAを一過的または安定的に導入した細胞であってもよい。
本実施の形態に係る培地添加剤を添加した動物細胞培養用の培地を用いて細胞を培養する方法は、接着細胞または浮遊細胞等の細胞の性質、および実験に応じて、静置培養であってもよいし、振とう培養であってもよい。培養温度、培養時間、および気体組成等の培養条件は、培養する細胞および実験に応じて適宜選択してもよい。
本実施の形態に係る培地添加剤によれば、細胞培養用の培地に添加して、ヒト等の哺乳類の細胞、およびショウジョウバエ等の昆虫の細胞等のカイコを除く動物の細胞を培養することができる。当該培地添加剤は、ヒト等の哺乳類または他の脊椎動物と共通する病原体を有しないカイコの血清に由来する成分を含むため、哺乳類または他の脊椎動物等の細胞を培養する場合には、培地添加剤に含まれ得る病原体による汚染を回避することができる。カイコは、牛等と異なり、既に遺伝的に均質な系統が確立されており、また、新たに遺伝的に均質な系統を確立することも比較的容易であるため、ロットの違いによる成分の相違等が少なく、均質な培地添加剤を安価かつ安定的に供給することができる。
本実施の形態に係る培地添加剤の製造方法によれば、カイコの血清に由来する成分から生成されるメラニンの生成を抑制し、高品質な培地添加剤を製造することができる。当該製造方法は、さらに血清成分を超音波処理する工程を含んでもよい。
なお、別の実施の形態では、細胞の培養方法が提供される。細胞の培養法方法は、上述の培地添加剤を含む培地において、カイコ細胞を除く動物細胞を培養する方法である。細胞は初代培養細胞であってもよい。
以下の実施例により、本発明をさらに具体的に説明するが、本発明は実施例によって限定されるものではない。
(実施例1:動物細胞培養用の培地添加剤の調製)
カイコ(Bombyx mori)は、N17系統の遺伝的に均質なものを使用した。カイコの幼虫は、25℃で桑の葉の上で飼育した。5齢幼虫の5日目のカイコから簡易遠心分離器を用いて血液を採取した。採取した血液に、培地添加後の終濃度が1.4mMとなるようにL-システイン(富士フイルム和光純薬社製)を添加し、1,210×g、4℃で15分間遠心分離して血球成分を除去した。上清を回収し、56℃で30分間加熱してメラニン化関連酵素等を失活させた。さらに4℃で15分間、9,000×gで遠心分離して変性タンパク質を除去することで血清上清を得た。この血清上清をリン酸緩衝液(1×濃度等)に対して透析し、透析後の溶液を孔径0.22μmのステリトップ(Merck Millipore社製)で滅菌濾過することで、培地添加剤としてのカイコ血清を得た。

(試験例1:FBSとカイコ血清由来培地添加剤の比較)
10%(v/v)FBSを含むMEM培地、および実施例1で調製したカイコ血清を10%(v/v)の濃度で含むMEM培地を使用して、ハムスター腎臓由来BHK細胞の増殖を比較した。12穴プレートのウェルに、1×105細胞/mLのBHK細胞を加えた。PBSで細胞を2回洗浄した後、各ウェルにMEM培地を1mLずつ加えた。FBSまたは実施例1で調製したカイコ血清を10%(v/v)となるように各ウェルに加えた。陰性対照として、同量のPBSを加えた。37℃、5%CO2で3日間培養し、細胞の状態を顕微鏡で観察した。
10%(v/v)FBSを含むMEM培地、および実施例1で調製したカイコ血清を10%(v/v)の濃度で含むMEM培地を使用して、ハムスター腎臓由来BHK細胞の増殖を比較した。12穴プレートのウェルに、1×105細胞/mLのBHK細胞を加えた。PBSで細胞を2回洗浄した後、各ウェルにMEM培地を1mLずつ加えた。FBSまたは実施例1で調製したカイコ血清を10%(v/v)となるように各ウェルに加えた。陰性対照として、同量のPBSを加えた。37℃、5%CO2で3日間培養し、細胞の状態を顕微鏡で観察した。
(結果)
図1は、FBS、カイコ血清またはPBSを添加したMEM培地で培養したBHK細胞の顕微鏡写真を示す。陰性対照であるPBS添加区では細胞の増殖はほとんど認められなかった。FBS添加区およびカイコ血清添加区では、細胞の増殖が認められた。FBS添加区では、細胞はコンフルエントまで増殖していた。カイコ血清添加区では、細胞はセミコンフルエントまで増殖していたが,FBS添加区に比べると細胞数は少なかった。このことから、カイコ血清は、BHK細胞に対し一定の増殖活性を有することが示された。
図1は、FBS、カイコ血清またはPBSを添加したMEM培地で培養したBHK細胞の顕微鏡写真を示す。陰性対照であるPBS添加区では細胞の増殖はほとんど認められなかった。FBS添加区およびカイコ血清添加区では、細胞の増殖が認められた。FBS添加区では、細胞はコンフルエントまで増殖していた。カイコ血清添加区では、細胞はセミコンフルエントまで増殖していたが,FBS添加区に比べると細胞数は少なかった。このことから、カイコ血清は、BHK細胞に対し一定の増殖活性を有することが示された。
(試験例2:カイコ血清を添加した培地での様々な細胞の培養)
実施例1で調製したカイコ血清を10%(v/v)の濃度で含むMEM培地を使用して、ハムスター腎臓由来BHK細胞、イヌマクロファージ由来DH82細胞、アフリカミドリザル腎臓由来Vero細胞、およびヒト乳腺由来MCF7細胞を培養した。24穴プレートのウェルに、それぞれ2×104細胞/mLのBHK細胞、DH82細胞、Vero細胞、またはMCF7細胞を加えた。PBSで細胞を2回洗浄した後、各ウェルに10%(v/v)のカイコ血清を含有するMEM培地を1mLずつ加えた。37℃、5%CO2で4日間培養し、細胞の状態を顕微鏡で観察した。
実施例1で調製したカイコ血清を10%(v/v)の濃度で含むMEM培地を使用して、ハムスター腎臓由来BHK細胞、イヌマクロファージ由来DH82細胞、アフリカミドリザル腎臓由来Vero細胞、およびヒト乳腺由来MCF7細胞を培養した。24穴プレートのウェルに、それぞれ2×104細胞/mLのBHK細胞、DH82細胞、Vero細胞、またはMCF7細胞を加えた。PBSで細胞を2回洗浄した後、各ウェルに10%(v/v)のカイコ血清を含有するMEM培地を1mLずつ加えた。37℃、5%CO2で4日間培養し、細胞の状態を顕微鏡で観察した。
(結果)
図2は、カイコ血清を添加したMEM培地で培養したBHK細胞、DH82細胞、Vero細胞、およびMCF7細胞の顕微鏡写真である。上段は細胞培養開始時、下段は4日目の細胞の状態を示す写真である。BHK細胞、DH82細胞、Vero細胞、およびMCF7細胞のいずれの細胞においても細胞の増殖を示した。これにより、実施例1で調製したカイコ血清は、様々な種類の細胞の培養に使用できることが示された。
図2は、カイコ血清を添加したMEM培地で培養したBHK細胞、DH82細胞、Vero細胞、およびMCF7細胞の顕微鏡写真である。上段は細胞培養開始時、下段は4日目の細胞の状態を示す写真である。BHK細胞、DH82細胞、Vero細胞、およびMCF7細胞のいずれの細胞においても細胞の増殖を示した。これにより、実施例1で調製したカイコ血清は、様々な種類の細胞の培養に使用できることが示された。
(試験例3:カイコ血清を添加した培地でのヒト由来初代培養細胞の培養)
10%(v/v)FBSを含むDMEM培地、および実施例1で調製したカイコ血清を10%(v/v)の濃度で含むDMEM培地を使用して、ヒト由来初代膵星細胞の増殖を比較した。12穴プレートのウェルに、1×105細胞/mLの膵星細胞を加えた。PBSで細胞を2回洗浄した後、各ウェルに培地を1mLずつ加えた。FBSまたは実施例1で調製したカイコ血清を10%(v/v)となるように加えた。37℃、5%CO2で培養し、細胞の状態を顕微鏡で観察した。同じ培養試験を独立して2回行った。
10%(v/v)FBSを含むDMEM培地、および実施例1で調製したカイコ血清を10%(v/v)の濃度で含むDMEM培地を使用して、ヒト由来初代膵星細胞の増殖を比較した。12穴プレートのウェルに、1×105細胞/mLの膵星細胞を加えた。PBSで細胞を2回洗浄した後、各ウェルに培地を1mLずつ加えた。FBSまたは実施例1で調製したカイコ血清を10%(v/v)となるように加えた。37℃、5%CO2で培養し、細胞の状態を顕微鏡で観察した。同じ培養試験を独立して2回行った。
(結果)
図3は、FBSを含むMEM培地またはカイコ血清を含むMEM培地で培養した膵星細胞の4日目の顕微鏡写真を示す。FBS添加区およびカイコ血清添加区では、細胞の増殖が認められた。カイコ血清添加区では、FBS添加区に比べると細胞数は少なかった。このことから、実施例1で調製したカイコ血清の添加によって、膵星細胞は一定の増殖活性を有することが示された。よって、カイコ血清は初代培養細胞の培養においても有用であることが示された。
図3は、FBSを含むMEM培地またはカイコ血清を含むMEM培地で培養した膵星細胞の4日目の顕微鏡写真を示す。FBS添加区およびカイコ血清添加区では、細胞の増殖が認められた。カイコ血清添加区では、FBS添加区に比べると細胞数は少なかった。このことから、実施例1で調製したカイコ血清の添加によって、膵星細胞は一定の増殖活性を有することが示された。よって、カイコ血清は初代培養細胞の培養においても有用であることが示された。
(試験例4:Vero細胞に対するカイコ血清の効果)
アフリカミドリザル腎臓上皮細胞由来Vero細胞の増殖に対するカイコ血清の効果を、無血清またはFBSと比較した。96穴プレートに、1×104細胞/ウェルでVero細胞を播種し、2%(v/v)FBSを含むDMEM培地で一晩培養した(前培養)。血清を含まないEMEM培地(2%(v/v)PBSを添加)、10%(v/v)FBSを含むDMEM培地、2%(v/v)FBSを含むDMEM培地、および実施例1で調製したカイコ血清を2%(v/v)の濃度で含むDMEM培地のいずれかを各ウェルに加え、37℃、5%CO2の条件下で3~6日間培養した。3日目および6日目に、以下の方法でMTTアッセイを行った。1.5mg/mL MTT solution in PBSを各ウェルに10μL添加し、37℃、5%CO2で3.5時間培養した。培養上清を除去し、MTT wash solution(イソプロパノール中の4mM HCl、0.1%NP40)を150μL添加し、30分間、室温で振とうした。650nmをリファレンスとして、プレートリーダーで570nmの吸光度を測定した。
アフリカミドリザル腎臓上皮細胞由来Vero細胞の増殖に対するカイコ血清の効果を、無血清またはFBSと比較した。96穴プレートに、1×104細胞/ウェルでVero細胞を播種し、2%(v/v)FBSを含むDMEM培地で一晩培養した(前培養)。血清を含まないEMEM培地(2%(v/v)PBSを添加)、10%(v/v)FBSを含むDMEM培地、2%(v/v)FBSを含むDMEM培地、および実施例1で調製したカイコ血清を2%(v/v)の濃度で含むDMEM培地のいずれかを各ウェルに加え、37℃、5%CO2の条件下で3~6日間培養した。3日目および6日目に、以下の方法でMTTアッセイを行った。1.5mg/mL MTT solution in PBSを各ウェルに10μL添加し、37℃、5%CO2で3.5時間培養した。培養上清を除去し、MTT wash solution(イソプロパノール中の4mM HCl、0.1%NP40)を150μL添加し、30分間、室温で振とうした。650nmをリファレンスとして、プレートリーダーで570nmの吸光度を測定した。
(結果)
図4に570nmの吸光度を示す。陰性対照である血清を含まないDMEMでは細胞の増殖が見られないのに対し、カイコ血清を含むDMEMでは穏やかな細胞の増殖を示した。細胞の増殖速度は、FBSを含むDMEMに比べると遅かった。しかし、長期間の培養では、FBSを含むDMEMでは細胞の過密による負の影響により細胞数が著しく減少したのに対し、カイコ血清を含むDMEMでは細胞数の著しい減少が見られなかった。結果として、6日目において、2%(v/v)カイコ血清を含むDMEMでは、2%(v/v)FBSを含むDMEM培地と同等の細胞数となった。
図4に570nmの吸光度を示す。陰性対照である血清を含まないDMEMでは細胞の増殖が見られないのに対し、カイコ血清を含むDMEMでは穏やかな細胞の増殖を示した。細胞の増殖速度は、FBSを含むDMEMに比べると遅かった。しかし、長期間の培養では、FBSを含むDMEMでは細胞の過密による負の影響により細胞数が著しく減少したのに対し、カイコ血清を含むDMEMでは細胞数の著しい減少が見られなかった。結果として、6日目において、2%(v/v)カイコ血清を含むDMEMでは、2%(v/v)FBSを含むDMEM培地と同等の細胞数となった。
(試験例5:カイコ血清の熱処理による効果)
実施例1で調製したカイコ血清を、65℃、70.7℃、74.4℃、81.9℃、85.6℃、89.3℃、または95℃で30分間処理した。その後、5000rpmで15分間遠心分離し、上清を回収して熱処理カイコ血清とした。96穴プレートに、1×104細胞/ウェルでVero細胞を播種し、2%(v/v)FBSを含むDMEM培地で一晩培養し(前培養)、56℃で熱処理したサンプルとして実施例1で調製したカイコ血清、および各温度で熱処理した熱処理カイコ血清を、2%(v/v)または10%(v/v)となるように添加した。添加後2日目に、試験例4と同様の方法でMTTアッセイを行い、570nmの吸光度を測定した。
実施例1で調製したカイコ血清を、65℃、70.7℃、74.4℃、81.9℃、85.6℃、89.3℃、または95℃で30分間処理した。その後、5000rpmで15分間遠心分離し、上清を回収して熱処理カイコ血清とした。96穴プレートに、1×104細胞/ウェルでVero細胞を播種し、2%(v/v)FBSを含むDMEM培地で一晩培養し(前培養)、56℃で熱処理したサンプルとして実施例1で調製したカイコ血清、および各温度で熱処理した熱処理カイコ血清を、2%(v/v)または10%(v/v)となるように添加した。添加後2日目に、試験例4と同様の方法でMTTアッセイを行い、570nmの吸光度を測定した。
(結果)
図5に570nmの吸光度を示す。56℃の熱処理カイコ血清を高濃度(10%(v/v))で添加した場合は毒性が生じ、血清を含まない対照と比較して細胞数が少なかった。一方、65~80℃の熱処理カイコ血清では、高濃度で添加した場合であっても毒性はなく、対照および熱処理カイコ血清を低濃度(2%(v/v))で添加した場合と比較して、顕著な細胞増殖が見られた。
図5に570nmの吸光度を示す。56℃の熱処理カイコ血清を高濃度(10%(v/v))で添加した場合は毒性が生じ、血清を含まない対照と比較して細胞数が少なかった。一方、65~80℃の熱処理カイコ血清では、高濃度で添加した場合であっても毒性はなく、対照および熱処理カイコ血清を低濃度(2%(v/v))で添加した場合と比較して、顕著な細胞増殖が見られた。
上述した実施の形態は、本発明を説明するためのものであり、本発明の範囲を限定するものではない。すなわち、本発明の範囲は、実施の形態ではなく、特許請求の範囲によって示される。そして、特許請求の範囲内及びそれと同等の発明の意義の範囲内で施される様々な変形が、本発明の範囲内とみなされる。

カイコ細胞を除く動物細胞培養用の培地添加剤。 - カイコから抽出した血液に還元剤を添加する添加工程と、
