Description
本発明は、医薬の分野で有用な化合物である、ビスイミノビオチン化合物の薬物送達用の用途に関する。
ビオチン(下記式1で表される)は生体内に存在する分子の一つであり、ストレプトアビジンと強固に結合する(Kd = 10-15 M)ことが知られ、生物学的研究に広く用いられている。
これらの強力な相互作用を利用し、結合力が調整された改変型ビスビオチンとストレプトアビジンを利用した医薬への利用が知られている。
イミノビオチン(下記式2で表される)は改変型ビオチンの一つであり、ビオチンの環状ウレア構造が環状グアニジンとなった化合物である。イミノビオチンはストレプトアビジンとの結合力を調整できる構造として用いられ、さらに一般式3で表されるビスイミノビオチン化合物が、プレターゲティング医薬に利用可能な構造として知られている(特許文献1)。
現在までに、一般式3で表されるビスイミノビオチン化合物のスペーサーVに結合する側鎖Wにはポリエチレングリコールやアミド結合を有するものがあり、その末端には蛍光化合物や薬剤の有効成分としての化合物を結合するためのアミンやカルボン酸を有するものが報告されている。しかしこれらの末端に蛍光化合物などを付ける際に、反応性の高い官能基を保護し、後から脱保護する必要があった。
これらの強力な相互作用を利用し、結合力が調整された改変型ビスビオチンとストレプトアビジンを利用した医薬への利用が知られている。
イミノビオチン(下記式2で表される)は改変型ビオチンの一つであり、ビオチンの環状ウレア構造が環状グアニジンとなった化合物である。イミノビオチンはストレプトアビジンとの結合力を調整できる構造として用いられ、さらに一般式3で表されるビスイミノビオチン化合物が、プレターゲティング医薬に利用可能な構造として知られている(特許文献1)。
現在までに、一般式3で表されるビスイミノビオチン化合物のスペーサーVに結合する側鎖Wにはポリエチレングリコールやアミド結合を有するものがあり、その末端には蛍光化合物や薬剤の有効成分としての化合物を結合するためのアミンやカルボン酸を有するものが報告されている。しかしこれらの末端に蛍光化合物などを付ける際に、反応性の高い官能基を保護し、後から脱保護する必要があった。
例えば、特許文献1記載の以下の式4、式5で表されるビスイミノビオチン化合物は分子内に蛍光団や薬剤を有しており、プレターゲッティング法に基づくドラッグデリバリーや診断技術に有用であると報告されている。しかしそれらの分子の合成には多段階を要し、合成終盤での官能基選択的な修飾や保護基の着脱が問題となる場合がある。
そのため、ビスイミノビオチン部分は共通で末端のみを簡便に変換可能な化合物の開発が必要であった。そこで、ビスイミノビオチン化合物の合成が多段階となり、工業的生産の観点からは改良の余地があり、ビスイミノビオチン部分と薬剤や蛍光化合物をより簡便に結合する手法が求められていた。
しかしながら、ビスイミノビオチン部分と薬剤や蛍光化合物を簡便に結合するための手法は、従来技術には開示されていないのが現状である。
本発明の技術課題は、ビスイミノビオチン部分と薬剤や蛍光化合物を簡便に結合するための構造を有する、ストレプトアビジンで標識化された物質への薬物送達用として有用なビスイミノビオチン化合物を提供することにある。

本発明の課題を解決するために、本発明者らはK. B. Sharplessにより定義されるクリック反応を利用した分子デザインを考案した。
クリック反応の中でアルキンとアジド基を反応させトリアゾールを形成するヒュスゲン環化は最も用いられる反応の一つである。ビスイミノビオチン末端にアルキンやアジド基を有した化合物を合成することで、煩雑な保護基の着脱なしで側鎖の末端を簡便に修飾することが可能になると考えた。
本発明者は、以上の点に基づいて本発明にかかるビスイミノビオチン化合物の分子構造を得て本発明を完成した。
本発明にかかるビスイミノビオチン化合物は、下記一般式9で表されることを特徴とする。
クリック反応の中でアルキンとアジド基を反応させトリアゾールを形成するヒュスゲン環化は最も用いられる反応の一つである。ビスイミノビオチン末端にアルキンやアジド基を有した化合物を合成することで、煩雑な保護基の着脱なしで側鎖の末端を簡便に修飾することが可能になると考えた。
本発明者は、以上の点に基づいて本発明にかかるビスイミノビオチン化合物の分子構造を得て本発明を完成した。
本発明にかかるビスイミノビオチン化合物は、下記一般式9で表されることを特徴とする。

上記一般式9で表されるビスイミノビオチン化合物を用いる、ストレプトアビジンで標識化された物質への薬物送達方法は、薬物送達用基材に薬物を結合させること、該薬物が結合した薬物送達用基材を前記ストレプトアビジンで標識化された物質に結合させて、該薬物送達用基材を介して前記ストレプトアビジンで標識化された物質に該薬物を結合すること、を含み、
前記薬物送達用基材が、上記一般式9で表されるビスイミノビオチン化合物である、ことを特徴とする。
上記一般式9で表されるビスイミノビオチン化合物は、ストレプトアビジンで標識化された物質への薬物送達用の薬物送達剤の製造方法における、薬物送達用基材として使用することができる。
上記一般式9で表されるビスイミノビオチン化合物を用いる、ストレプトアビジンで標識化された物質への薬物送達用の薬物送達剤の製造方法は、一般式9で表されるビスイミノビオチン化合物に官能基Jを利用して薬物を結合させること、を含むことを特徴とする。
薬物としては、生物活性物質又は蛍光化合物を用いることができる。
上記一般式9で表されるビスイミノビオチン化合物に薬物を結合させて得られる、薬物結合ビスイミノビオチン化合物は、ストレプトアビジンで標識化された物質への薬物送達剤としての使用することができる。

前記薬物送達用基材が、上記一般式9で表されるビスイミノビオチン化合物である、ことを特徴とする。
上記一般式9で表されるビスイミノビオチン化合物は、ストレプトアビジンで標識化された物質への薬物送達用の薬物送達剤の製造方法における、薬物送達用基材として使用することができる。
上記一般式9で表されるビスイミノビオチン化合物を用いる、ストレプトアビジンで標識化された物質への薬物送達用の薬物送達剤の製造方法は、一般式9で表されるビスイミノビオチン化合物に官能基Jを利用して薬物を結合させること、を含むことを特徴とする。
薬物としては、生物活性物質又は蛍光化合物を用いることができる。
本発明によれば、ビスイミノビオチン部分と薬剤を簡便に結合するための構造を有し、ストレプトアビジンで標識化された物質への薬物送達用として有用なビスイミノビオチン化合物を提供することができる。
本発明にかかるビスイミノビオチン化合物の一形態によれば、以下の反応が可能となる。
上記の反応において、式6で表されるビスイミノビオチン化合物のアルキン構造を有する末端と、式7で表される化合物のアジド基からなる末端を、銅触媒条件下に付すことで反応させることにより、トリアゾール環を有する末端を有する式8で表されるビスイミノビオチン化合物が得られる。
式7、式8のYとして、生物活性物質や蛍光化合物などの薬物を用いることができる。式8の化合物はその分子構造から、前述の式4や式5と同様に、プレターゲッティングや標的探索に利用可能である。
クリック反応で頻繁に利用されるヒュスゲン環化反応は官能基選択的に起こることから、上記の反応における式6や式7の他の部分に保護基などの修飾を必要としない。そのため合成の最後に二つの分子を結合させることが可能となる。さらにビスイミノビオチン部分を共構造とし、アルキン構造とアジド基を入れ替えた化合物でも同様に反応できることから、化合物に合わせた柔軟な合成が可能となる。
よって、式8の薬物送達剤の有効成分として有用なビスイミノビオチン化合物を合成するためには、薬物送達用基材としての式6のような化合物が必要となる。
以下、本発明について詳細に説明する。
薬物送達用基材として有用なビスイミノビオチン化合物は、以下の一般式9で表される。
式7、式8のYとして、生物活性物質や蛍光化合物などの薬物を用いることができる。式8の化合物はその分子構造から、前述の式4や式5と同様に、プレターゲッティングや標的探索に利用可能である。
クリック反応で頻繁に利用されるヒュスゲン環化反応は官能基選択的に起こることから、上記の反応における式6や式7の他の部分に保護基などの修飾を必要としない。そのため合成の最後に二つの分子を結合させることが可能となる。さらにビスイミノビオチン部分を共構造とし、アルキン構造とアジド基を入れ替えた化合物でも同様に反応できることから、化合物に合わせた柔軟な合成が可能となる。
よって、式8の薬物送達剤の有効成分として有用なビスイミノビオチン化合物を合成するためには、薬物送達用基材としての式6のような化合物が必要となる。
以下、本発明について詳細に説明する。
薬物送達用基材として有用なビスイミノビオチン化合物は、以下の一般式9で表される。
(式中、A、D、Eは2つのビシクロ環を結合するスペーサーであり、Eは分岐をとることのできる構造を表し、それぞれ置換基を有してもよく、環構造を形成してもよい。Jはクリック反応を行うための官能基を表し、GはEとJを結合するスペーサーを表し、Rは、水素、アセチル基、ベンジル基、トリフルオロアセチル基またはBoc基を表す。)
但し、以下の化合物10-24及び化合物10-25は、上記一般式9で示されるビスイミノビオチン化合物から除かれる。

EのA、D及びGと結合する部分としては、窒素原子、炭素原子、アミド基、ベンゼン環またはヘテロ環(例えばフラン環、ピリミジン環、ピロール環、ピリジン環など)を好ましく用いることができる。
A、D及びEが有する環構造や置換基は、本発明の目的効果が得られる範囲において選択することができる。
A、D及びEからなる部分としては、Aがa1-a2-a3-a4からなり、Dがd1-d2-d3-d4からなり、A-E-Dがa1-a2-a3-a4-E-d1-d2-d3-d4で表され、a1、a2、a3、a4、E、d1、d2、d3及びd4がそれぞれ独立して以下の表1の各欄から選択される部分が好ましい。
EのA、D及びGと結合する部分としては、窒素原子、炭素原子、アミド基、ベンゼン環またはヘテロ環(例えばフラン環、ピリミジン環、ピロール環、ピリジン環など)を好ましく用いることができる。

本発明において表1~表4に記載される「結合」は、隣接する基を直接結合する直接結合を意味する。

Gとして好ましい連結基として、以下に示す一般式(3-1)~(3-22)で表される連結基を挙げることができる。

アルキン構造を有する基としては、アルキニル基、アルキニルオキシ基、アルキニルアミノ基等を挙げることができ、これらの基に含まれるアルキニル基としてはエチニル基、プロパギル基等の炭素数2~3のアルキニル基を挙げることができる。
好ましいアジド基及びアルキン構造を有する基としては、以下の式(4-1)~(4-10)で表される各基を挙げることができる。なお、下記各式においてGが表示されている位置はGとの結合位置を示す。

以下に一般式9の製造に利用し得る縮合反応の条件について記述する。
縮合反応で使用される溶剤としては、N,N-ジメチルホルムアミド、アセトニトリル、テトラヒドロフラン、テトラヒドロピラン、ジクロロメタン、1,4-ジオキサン、クロロホルム、トルエン、ベンゼンの何れか一つまたはこれらの2種以上を併用して用いることができる。
溶媒として好ましくは、N,N-ジメチルホルムアミド、アセトニトリル、テトラヒドロフラン、ジクロロメタン、1,4-ジオキサンの何れか一つまたはこれらの2種以上を併用して用いる。
溶媒としてより好ましくは、N,N-ジメチルホルムアミド、アセトニトリル、テトラヒドロフラン、ジクロロメタンまたは1,4-ジオキサンを単一溶媒として用いる。

縮合反応におけるカルボン酸の活性化には、酸ハロゲン化物(例えば、酸塩化物や酸フッ化物、酸臭化物など)、活性エステル(例えば、N-ヒドロキシコハク酸イミドエステル、N-ヒドロキシスルホコハク酸イミドエステル、1-アシルオキシ-7-アザベンゾトリアゾール、1-アシルオキシベンゾトリアゾールなど)の少なくとも1種を用いることができる。
カルボン酸の活性化に、好ましくは、酸ハロゲン化物、コハク酸エステル、活性エステルの少なくとも1種を用いる。
カルボン酸の活性化に、より好ましくは、酸塩化物、N-ヒドロキシコハク酸イミドエステル、N-ヒドロキシスルホコハク酸イミドエステルの少なくとも1種を用いる。

一般式14のGとKの間の結合は以下の反応により形成することができる。
例えば、一般式9’の構造中のJがアルキンの場合には、一般式7で表されるアジドを反応させることで、一般式18で表される構造を形成する。または一般式9’’の構造中のJがアジドの場合には、一般式19で表されるアジドを反応させることで一般式20で表される構造を形成する。(但し、Y、Zはアルキンまたはアジドと結合する生物活性物質や蛍光化合物の構造を表す)
以下に一般式14の製造に利用し得る環化反応の条件について記述する。
環化反応で使用される溶剤としては、N,N-ジメチルホルムアミド、アセトニトリル、テトラヒドロフラン、テトラヒドロピラン、ジクロロメタン、1,4-ジオキサン、ジメチルスルホキシド、クロロホルム、トルエン、ベンゼン、水、酢酸、メタノール、エタノール、イソプロパノール、ノルマル-ブチルアルコール、ターシャリー-ブチルアルコールの何れか一つまたはこれらの2種以上を併用して用いることができる。
溶剤として好ましくは、N,N-ジメチルホルムアミド、アセトニトリル、テトラヒドロフラン、1,4-ジオキサン、クロロホルム、トルエン、ベンゼン、水、酢酸、メタノール、エタノール、イソプロパノール、ノルマル-ブチルアルコール、ターシャリー-ブチルアルコールの何れか一つまたはこれらの2種以上を併用して用いる。

環化反応で使用される触媒としては、一価の銅塩(例えば、塩化銅(I)、臭化銅(I)、ヨウ化銅(I)、酢酸銅(I)など)、二価の銅塩(例えば、塩化銅(II)、臭化銅(II)、ヨウ化銅(II)、酢酸銅(II)、硫酸銅(II)など)、銀塩(塩化銀、臭化銀、ヨウ化銀、酢酸銀など)、ルテニウム塩の何れか一つまたはこれらの2種以上を併用して用いることができる。
あるいは触媒を添加せずに環化反応を行ってもよい。
触媒として好ましくは、一価の銅塩、二価の銅塩、銀塩の何れか一つまたはこれらの2種以上を併用して用いる。
触媒としてさらに好ましくは、塩化銅(I)、臭化銅(I)、ヨウ化銅(I)、酢酸銅(I)、塩化銅(II)、臭化銅(II)、ヨウ化銅(II)、酢酸銅(II)、硫酸銅(II) の何れか一つを用いる。
環化反応の添加剤としては、アスコルビン酸、アスコルビン酸ナトリウム、アスコルビン酸カリウム、アスコルビン酸カルシウムの何れか一つまたはこれらの2種以上を併用して用いることができる。あるいは、添加剤を添加せずに環化反応を行ってもよい。

環化反応の精製には、結晶化、ろ過洗浄、シリカゲルカラムクロマトグラフィー、ゲル濾過クロマトグラフィーを用いることができる。
好ましい精製方法として、結晶化、ろ過洗浄、シリカゲルカラムクロマトグラフィーの少なくとも1種を用いる。

あるいは触媒を添加せずに環化反応を行ってもよい。
触媒として好ましくは、一価の銅塩、二価の銅塩、銀塩の何れか一つまたはこれらの2種以上を併用して用いる。

HPLC分析は、以下の2条件で分析を行った。

・流速:1 mL/min.
・カラム温度:40 ℃
・検出波長:254 nm
・移動相:0.1%トリフルオロ酢酸水溶液/アセトニトリル
(分析条件B)

グラジュエント条件は、例えば0.1%トリフルオロ酢酸水溶液/アセトニトリル = 85/15(12分)35/65と記載するが、0.1%トリフルオロ酢酸水溶液を85%から35%に12分かけて減少させ、そのあと85%に戻す条件を表す。
HPLC分析は、以下の2条件で分析を行った。

135 mg (0.13 mmol) のビス(Boc-イミノビオチン)22に1 mLの脱水ジメチルホルムアミドを加え、さらに22 mg(1.05当量)のカルボニルジイミダゾールを加えた。40 ℃で1時間撹拌し、0.7 mLの脱水クロロホルムに溶解させた36 mg (1当量)のアミン 23(Shigma-Aldrich、CAS No; 1255942-06-3) を加え、室温で5時間撹拌した。反応終了後、減圧濃縮し10 mLのクロロホルムを加え、0.1 N塩酸でpH 5に調整した。沈殿物が生じ、水とクロロホルムを除去した。沈殿物をメタノールに溶解し、硫酸マグネシウムで乾燥し、濃縮して残渣を得た。シリカゲルカラム (クロロホルム/メタノール = 20/1から5/1)で精製し、128 mgのアミド24 を得た。(収率75%)

(ビスイミノビオチン-フルオロセイン化合物28の合成)
実施例1-2で合成した13 mg(12 μmol)のビスイミノビオチン26のトリフルオロ酢酸塩を1mLの脱水ジメチルホルムアミドに溶解し、0.72 mLのクロロホルム溶液を加えた。さらに、 11 mg(2当量)の6-カルボキシフルオロセインアジド27のクロロホルム溶液を加えた。室温で1夜撹拌した後、反応液を60 ℃で減圧濃縮した。残渣を2 mLの 1 規定塩酸水溶液で二回洗浄した後、減圧乾燥してアモルファス状の25 mgのビスイミノビオチン-フルオロセイン28を得た。

(ビスイミノビオチン-アジド化合物37の合成)
実施例1-2において、15 mg (11.6 μmol) のアミド 25を使用する代わりに、実施例4-1で合成した16 mg(13 μmol) のビス(Boc-イミノビオチン)-アジド36を用い、実施例1-2記載の合成法に従って反応を行い、14.5 mgのビスイミノビオチン-アジド37 のトリフルオロ酢酸塩を得た。(収率100%)

Claims (13)
- 請求項1乃至5のいずれか1項に記載のビスイミノビオチン化合物の、ストレプトアビジンで標識化された物質への薬物送達用基材としての使用。
- 請求項6に記載の薬物結合ビスイミノビオチン化合物の、ストレプトアビジンで標識化された物質への薬物送達用の薬物送達剤としての使用。
- ストレプトアビジンで標識化された物質への薬物送達方法であって、薬物送達用基材に薬物を結合させること、該薬物が結合した薬物送達用基材を前記ストレプトアビジンで標識化された物質に結合させて、該薬物送達用基材を介して前記ストレプトアビジンで標識化された物質に該薬物を結合すること、を含み、
前記薬物送達用基材が、請求項1乃至5のいずれか1項に記載のビスイミノビオチン化合物である、ことを特徴とする薬物送達方法。 - ストレプトアビジンで標識化された物質への薬物送達用の薬物送達剤の製造方法であって、請求項1乃至5のいずれか1項に記載のビスイミノビオチン化合物に官能基Jを利用して薬物を結合させること、を含むことを特徴とする、薬物送達剤の製造方法。
- 前記薬物が、生物活性物質又は蛍光化合物である、請求項10に記載の薬物送達用の化合物の製造方法。
- ストレプトアビジンで標識化された物質への薬物送達用の薬物送達剤の製造方法における、請求項1乃至5のいずれか1項に記載のビスイミノビオチン化合物の使用。
- 前記薬物が、生物活性物質又は蛍光化合物である、請求項12に記載の使用。
