Abstract
CHAPS를 포함하는 생체 조직 투명화제를 사용한 생체 조직의 투명화 방법은, 조직의 변성 없이 빠른 투명화가 가능하다.

생체 조직의 투명화 방법에 관한 것이다.
조직 투명화 기술은 생체 조직 내부 구조 및 단백질 분포를 확인할 수 있고 기존 기술의 관찰 한계를 뛰어넘어 더 깊은 곳까지 조직 구조를 관찰하고 다양한 기관계(system)로부터 통합적인 구조와 분자 정보의 접근을 가능하게 하므로 최근 다양한 방법으로 조직을 투명화하는 기술이 개발되고 있다.
종래의 조직 투명화 기술로는 유기 용매를 이용한 조직투명화 과정인 Spatleholz, BABB, Scale S, iDISCO 법과, 폴리머 주입법인 ACT(active CLARITY technology)법이 있다. ACT를 제외한 방법의 경우, 형광과 항원의 보존성이 감소하는 문제를 갖는다. ACT의 경우 90% 이상의 항원 보존성이 있으며, 이는 CLARITY와 같이 고정된 단백질에 추가로 하이드로젤 폴리머와의 결합을 필요로 하는 방법에 비하면 보다 높은 보존성을 보인다. 그러나 강한 조직 고정 과정은 항원성의 손실을 유발하며, 사용할 수 있는 항체가 감소하는 등의 문제점이 발생하므로, 기술의 개선이 필요하다.
또한, 최근에 개발된 조직 투명화 방법인 'CLARITY' 기반 기술은 조직 내 하이드로겔(Hydrogel)을 추가하여 DNA나 단백질 등 진단에 중요한 중요 물질들을 붙잡아주는 일종의 조직 내 그물망 지지체를 만든 뒤 지질만을 선택적으로 제거하는 방법을 이용한다. (국제특허공개공보 WO 2016/108359)
그러나, 상기 'CLARITY' 기반 기술은 하이드로젤 지지체를 조직에 침투시켜야 하는데, 하이드로젤의 농도가 높아지면 단백질과의 결합도가 많아지며 보다 촘촘한 그물형 구조가 만들어지기 때문에 조직이 더 단단해진다. 이렇게 조직이 단단해지면 계면활성제에 의해서 지질이 빠져나가기 어려워지기 때문에 투명화에 걸리는 시간이 길어진다. 또한, 상기 방법은 조직 표면에 공기 및 검은 입자 침착을 야기하거나, 조직을 노랗게 변색시키는 문제점이 있다.
또한, 상기 'CLARITY' 기반 기술은 과정이 너무나 복잡하고 부가적인 장비가 많이 필요하다. 예를 들면 뇌 하나를 투명화하기 위해서는 적어도 50,000,000 만원 이상이 필요하다. 뿐만 아니라, 한 번에 하나의 뇌만 투명화하는 것이 가능하므로 경제적, 시간적으로 많은 손실이 유발된다. 보다 큰 문제는 항체을 이용한 염색이 그물망 구조를 이루는 폴리아크릴아마이드(polyacrylamide)의 사이를 투과하기에 어려움이 있다.
따라서 폴리아크릴아마이드(polyacrylamide)의 사용 없이 조직의 투명화 과정을 유도하고, 조직의 구조와 단백질의 손상 없이 생체 조직을 투명화할 수 있는 기술이 요구되고 있다.
본 발명의 일 측면에서의 목적은 조직의 변성 없이 빠른 투명화가 가능한 생체 조직 투명화용 조성물을 제공하는 것이다.

CHAPS를 포함하는 생체 조직 투명화제를 사용한 생체 조직의 투명화 방법은, 조직의 변성 없이 빠른 투명화가 가능할 뿐만 아니라, 투명화제 처리 전에 당류를 함유하는 용액을 전처리한 후 투명화를 유도하면, 보다 우수한 조직 투명화가 가능한 효과가 발생한다.
도 1은 PBS를 단독으로 처리할 경우 마우스 뇌의 투명화가 진행되지 않음을 확인한 사진이다.

본 발명의 일 측면에서, 하기 화학식 1로 표시되는 화합물 또는 이의 수화물을 포함하는, 생체 조직 투명화용 조성물이 제공된다:

상기 생체 조직 투명화용 조성물은 빛과 다른 분자의 투과를 막는 지질 성분을 생체 조직으로부터 제거하고, 단백질의 구조적 변성을 일으키지 않으며, 조직을 단단하게 하는 역할을 한다.
상기 생체 조직 투명화용 조성물에 있어서, 상기 화학식 1로 표시되는 화합물의 농도는 2 내지 55 w/v%(중량/부피%)의 농도로 포함될 수 있으며, 4 내지 50 w/v% 농도가 바람직하다. 이때, 상기 농도를 나타내기 위한 용액으로는 통상의 분야에 사용하는 유사 생체 용액(simulated body fluid)일 수 있으며, 보다 구체적으로는 물(증류수), PBS(phosphate buffer saline), TBS(tris buffer solution) 등을 사용할 수 있으나 이에 한정되는 것은 아니다.
상기 화학식 1로 표시되는 화합물의 농도가 2 w/v% 미만으로 포함되는 경우에는 생체 조직의 투명화 속도가 현저히 느려질 수 있고, 55 w/v% 초과로 포함되는 경우에는 상기 화학식 1로 표시되는 CHAPS가 완전히 용해되지 않을 수 있다.
상기 생체 조직 투명화용 조성물은 삼투압을 조절하여 생체 조직의 투명화를 빠르게 촉진시키는 역할을 하는 물질을 더 포함할 수 있다. 이때, 상기 생체 조직의 투명화를 빠르게 촉진시키는 물질은 우레아(urea), CHAPSO(3-([3-Cholamidopropyl]dimethylammoni o)-2-hydroxy-1-propanesulfonate), 수크로오스(sucrose), 프룩토오스(fructose), 글리세롤, 다이아트라이조산(Diatrizoic acid), Triton X-100, Tween-20, 2,2'-싸이오디에탄올, 이오헥솔(Iohexol), 클로랄 수화물(Chloral hydrate), 또는 이의 조합을 사용할 수 있으나, 이에 제한되는 것은 아니다.
이때, 상기 생체 조직의 투명화를 빠르게 촉진시키는 물질은 5 내지 80 w/v%의 농도로 포함될 수 있으며, 5 내지 75 w/v%의 농도로 포함될 수 있으며, 10 내지 70 w/v%의 농도로 포함될 수 있으며, 5 내지 50 w/v% 농도로 포함될 수 있으며, 35 내지 60 w/v% 농도로 포함될 수 있다. 이때, 상기 농도가 5 w/v% 미만인 경우 조직의 투명화 속도가 느려지며 80 w/v% 초과인 경우 결정이 유발되거나 용액에 녹지 않을 수 있다. 하나의 구체적인 일례로, 만약 상기 생체 조직의 투명화를 빠르게 촉진시키는 물질로 우레아(urea)를 사용할 경우, 상기 우레아는 10 내지 70 w/v%의 농도가 포함될 수 있으며, 20 내지 60 w/v% 농도가 보다 바람직하다. 또한, 상기 생체 조직의 투명화를 빠르게 촉진시키는 물질의 농도는 상기 화학식 1로 표시되는 화합물의 바람직한 농도 범위와 적절하게 조절할 수 있다.
종래의 투명화 방법을 사용하기 위해서는 조직과 용액의 굴절률을 맞추기 위해 mounting 용액을 따로 추가로 구입 및 제작이 필요하였으나, 본 발명에 따른 생체 조직 투명화용 조성물은 굴절률을 맞추는 용액이 필요 없으며, 이는 비용절감의 효과를 나타낸다. 더 나아가 기존의 투명화 방법은 조직이 부풀어지는 현상을 유발하는데 본 발명은 조직의 크기가 전혀 변하지 않는 효과가 있다.
상술한 바와 같이, 본 발명에 따른 생체 조직의 투명화용 조성물은 고가의 전기 영동 장치와 고가의 용액을 필요로 하지 않고, 뇌, 간, 폐, 신장, 장, 심장, 근육, 혈관 등의 다양한 생체 조직에 손상 없이 적용 가능할 뿐만 아니라, 조직의 부풀어짐, 버블 형성, 변색 및 검은 퇴적물이 나타나지 않고 생체 조직의 투명성이 향상되므로 생체 조직의 투명화용 조성물로 유용하게 사용할 수 있다.
본 발명의 다른 일 측면에서, 고정화된 생체 조직을, 상기 생체 조직 투명화용 조성물과 접촉시켜 생체 조직을 투명화시키는 단계를 포함하는, 생체 조직의 투명화 방법이 제공된다.
이하, 상기 생체 조직의 투명화 방법을 구체적으로 설명한다.
상기 생체 조직의 투명화 방법은 고정화된 생체 조직을 상기 조성물과 접촉시켜 투명화시키는 단계를 포함한다.
구체적으로, 본 발명에 따른 생체 조직의 투명화 방법은 고정화된 생체 조직을 상기 화학식 1로 표시되는 화합물을 포함하는 조성물과 접촉시킴으로써, 생체 조직의 물리화학적 특성을 변형하여 투명하게 함으로써 보다 깊이 빛이 침투할 수 있도록 만들어 투명하게 하는 것이다.
본 발명에 따른 생체 조직의 투명화 방법은 버블 형성, 변색 및 검은 퇴적물이 나타나지 않고 생체 조직의 투명성을 향상시키며 단백질 변성 등에 의하여 원하는 조직 내 정보가 손실 또는 왜곡되지 않아, 특히 GFP단백질 등 다양한 형광원(Fluorophore)을 조직 내 정보 검출 유용하게 사용될 수 있다.
본 발명에 따른 생체 조직의 투명화 방법에 있어서, 생체 조직은 투명화하기 전에 통상적으로 항원성의 손실을 유발하지 않으면서 생체 조직을 고정화하는 방법이라면 특별한 제한없이 적용하여 고정화할 수 있다.
보다 구체적으로 상기 생체 조직의 고정화는 파라포름알데하이드(paraformaldehyde), 에텔렌글리콜 디글리시딜에테르(ethylene glycol diglycidyl ether), 디프로필렌 글리콜 디글리시딜에테르(dipropylene glycol diglycidyl ether), 1,4-부탄디올 디글리시딜에테르(1,4-butanediol diglycidyl ether), 글리세롤 폴리글리시딜 에테르(glycerol polyglycidyl ether), 글루타르알데하이드(glutaraldehyde), 폴리아크릴아마이드 또는 이들의 조합을 사용하여 통상적인 방법을 통해 수행할 수 있으나, 이에 한정되는 것은 아니다.
본 발명에 따른 생체 조직의 투명화 방법에 있어서, 구체적인 실시예로서 CHAPS와 Urea 혼합물을 처리하면 단백질의 구조적 결합력이 증가되고, 변성을 일으키지 않으며, 조직을 더 단단하게 할 뿐만 아니라, 조직 투명화 과정에서 나타나는 조직의 부풀어 오름을 방지하고 항체 처리과정, 세척(washing) 과정 등에서 일어나는 조직의 갈라짐을 방지할 수 있다. 상기 농도를 나타내기 위한 용액으로는 통상의 분야에 사용하는 유사 생체 용액(simulated body fluid)일 수 있으나, 증류수, PBS(phosphate buffer saline), TBS(tris buffer solution), 증류수 바람직하나 이에 한정되는 것은 아니다. 또한, 담지의 온도는 10℃ 내지 50℃ 온도 범위에서 수행할 수 있고, 12 내지 48℃ 온도 범위에서 수행할 수 있고, 14 내지 46℃ 범위에서 수행할 수 있고, 16 내지 44℃ 범위에서 수행할 수 있고, 18 내지 42℃ 범위에서 수행할 수 있고, 20 내지 40℃ 범위에서 수행할 수 있고, 24 내지 39℃ 범위에서 수행할 수 있고, 28℃ 내지 38℃에서 수행할 수 있고, 30℃ 내지 37℃에서 수행할 수 있고, 33 내지 34℃에서 수행할 수 있다.
본 발명에 따른 생체 조직의 투명화 방법은 다양한 척추 동물의 조직(tissue)에 적용할 수 있으며,
특히, 뇌(brain), 혈관(blood vessel), 간(liver), 폐(lung), 신장(kidney), 췌장(pancreas), 장(intestine) 등에 적용하는 것이 바람직하고, 상기 생체 조직의 전체 부분을 한번에 투명화할 수 있다.
본 발명은 상기 투명화된 생체 조직 내 중요 정보, 즉, DNA, RNA, 단백질, 형광신호 등의 검출 방법을 제공한다.
본 발명에 따른 투명화된 생체 조직은 GFP 형광, 면역 염색을 통하여 단백질 또는 mRNA를 검출할 수 있다. 단백질의 경우 고정과정 동안 아미노산에 존재하는 아미노기끼리의 공유결합을 이루면서 서로 네트워크를 이루기 때문에, 매우 안정적인 반면, RNA나 DNA와 같은 핵산의 경우 아미노기가 존재하지 않기 때문에, 고정된 조직에서도 상대적으로 불안정하다. 특히, 전기영동 과정이 포함되는 경우, 핵산이 가진 전기적 성질에 의해 조직 내 위치가 바뀔 우려가 있다. 반면, 본 발명에 따른 투명화된 생체 조직은 GFP(Green Fluorescent Protein) 세포 및 cholinergic neuron 마커 항체인 Choline Acetyltransferase 등에 대한 형광 염색이 우수하게 수행된다.
본 발명에 따른 생체 조직 투명화 방법은 손상되지 않은 생체 조직의 세포와 분자의 3차원 분포를 이미지화하여 관찰할 수 있으므로, 복잡한 구조를 가지는 다양한 생체 조직에 대하여 하나의 완전한 구조로 수백 마이크로미터 이상의 크기로 관찰 연구를 수행할 수 있으므로 조직 내의 유용한 정보를 취득하여 뇌질환 등의 다양한 질환의 원인을 규명하는데 효과적으로 사용할 수 있다.
본 발명의 다른 일 측면에서, 고정화된 생체 조직에, 당류를 함유하는 용액을 처리하는 전처리 단계;
상기 전처리된 생체 조직을, 상기 생체 조직 투명화용 조성물과 접촉시켜 생체 조직을 투명화시키는 단계를 포함하는, 생체 조직의 투명화 방법이 제공된다.
상기 당류는 단당류, 이당류, 다당류 등일 수 있고, 보다 구체적으로 상기 단당류는 과당(fructose), 갈락토스(galactose), 글루코스(glucose) 또는 만노스(mannose)이고; 상기 이당류는 수크로오스(sucrose), 락토스(lactose), 말토스(maltose), 트레할로스(trehalose), 투라노스(turanose) 또는 셀로비오스(cellobiose)이고; 상기 다당류는 덱스트란(dextran), 디에틸아미노에틸-덱스트란(diethylamino ethyl-dextran), 덱스트린(dextrin), 셀룰로오스(cellulose) 또는 베타-글루칸(β-glucans)를 사용할 수 있다. 바람직하게는 상기 당류로서 수크로오스를 사용할 수 있다. 또한, 상기 당류를 함유하는 용액은 당류를 함유하는 수용액인 것이 바람직하다.
상기 당류를 함유하는 용액의 당류 농도는 10 내지 70 w/v% 범위일 수 있고, 20 내지 60 w/v% 범위일 수 있고, 25 내지 50 w/v% 범위일 수 있고, 30 내지 40 w/v% 범위일 수 있다.
생체 조직에 상기 당류 용액을 처리하면 빛과 다른 분자의 투과를 막는 지질 성분이 생체 조직으로부터 미리 제거되고, 탈수가 유도되어 결과적으로 조직을 고정화하는 시약과 생체조직 단백질과의 구조적 결합력이 증가하여 변성을 일으키지 않고, 조직을 더 단단하게 할 수 있으며, 조직 투명화 과정에서 나타나는 조직의 부풀어오름을 방지하고, 항체 처리과정, 세척 과정 등에서 일어나는 조직의 갈라짐을 방지할 수 있는 현저한 효과가 발생한다. 다만, 상기 당류를 함유하는 용액의 당류 농도가 10 w/v% 미만일 경우 상기 효과가 발생하지 않는 문제가 있고, 상기 당류를 함유하는 용액의 당류 농도가 70 w/v% 초과일 경우 경제적이지 못한 문제가 있다.
상기 생체 조직의 투명화 방법은 4℃ 내지 50℃ 온도 범위에서 수행할 수 있고, 10℃ 내지 50℃ 온도 범위에서 수행할 수 있고, 12 내지 48℃ 온도 범위에서 수행할 수 있고, 14 내지 46℃ 범위에서 수행할 수 있고, 16 내지 44℃ 범위에서 수행할 수 있고, 18 내지 42℃ 범위에서 수행할 수 있고, 20 내지 40℃ 범위에서 수행할 수 있고, 24 내지 39℃ 범위에서 수행할 수 있고, 28℃ 내지 38℃에서 수행할 수 있고, 30℃ 내지 37℃에서 수행할 수 있고, 33 내지 34℃에서 수행할 수 있다.
본 발명의 다른 일 측면에서, 당류 용액을 포함하는 생체 조직 투명화 전처리용 조성물; 및 상기 생체 조직 투명화용 조성물을 포함하는, 생체 조직 투명화용 키트가 제공된다.
이하, 본 발명을 실시예 및 실험예에 의해 상세히 설명한다.
단, 하기의 실시예 및 실험예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기 실시예 및 실험예에 의해 한정되는 것은 아니다.

실험예
1> CHAPS에 의한 생체조직 투명화 평가
CHAPS를 단독으로 포함하는 생체 조직 투명화제를, 생체 조직에 접촉시킬 경우, 생체조직 투명화가 용이하게 유도되는지 평가하기 위하여 하기와 같은 실험을 수행하였다. 단, 본 명세서 내에서 모든 동물 실험 과정은 안전성평가연구소 동물 자원 대학위원회의 지침서(Approval No. RS17003)에 따라 수행하였다.
먼저, 흡입 마취제인 이소푸루란(Isoflurane) (1cc/min) 을 사용하여 성인 생쥐(8주령)를 마취시켰다. 쥐의 혈관을 염색하기 위해 꼬리 정맥(tail vein)으로렉틴-488(Lectin-488, Cat# DL1174)을 주입하였다. 렉틴을 주입하고 5분 경과 후, 경심 관류로 얼음처럼 차가운 1X 인산완충용액 (phosphate-buffered saline) 50 mL을 관류시킨 후, 얼음처럼 차가운 4% PFA(Paraformaldehyde)가 포함된 PBS를 관류시켰다. 그 다음 기관을 적출하고 4%의 PFA(Paraformaldehyde)용액에 담근 후, 4℃에서 12 h 동안 인큐베이션 하였다. 이때, 상기 얼음처럼 차가운 온도 범위는 특별히 제한되는 것은 아니지만 -20℃ 내지 40℃ 범위일 수 있다.
다음으로, 상기 샘플(3mm 두께)을 50ml의 PBS로 2번 세척하였다. 상술한 과정에 의해 고정화된 샘플을, 40% CHAPS 수용액(40% CHAPS in DW)에서, 37℃ 온도 하에 120 rpm으로 5일 동안 인큐베이션하였다. 대조군으로는 상기 고정화된 샘플을 PBS에서, 37℃ 온도 하에 120 rpm으로 5일 동안 인큐베이션하였다. 그 결과를 도 1, 2에 나타내었다.
도 1은 PBS를 단독으로 처리할 경우 마우스 뇌의 투명화가 진행되지 않음을 확인한 사진이다.
도 2는 CHAPS를 단독으로 처리할 경우 마우스 뇌의 투명화가 진행되었음을 확인한 사진이다.
도 1, 도 2에 나타난 바와 같이,
CHAPS가 단독으로 마우스 뇌에 처리될 경우 마우스의 뇌 투명화가 현저히 유도됨을 알 수 있다. 뿐만 아니라, 조직의 크기에 변화가 없는 점으로부터 CHAPS는 조직의 변성과 손상 없이 투명화를 유도할 수 있음을 알 수 있다.

실험예
2> CHAPS 및
우레아에
의한 생체조직 투명화 평가
CHAPS 및 우레아를 포함하는 생체 조직 투명화제를, 생체 조직에 접촉시킬 경우, 생체조직 투명화가 용이하게 유도되는지 평가하기 위하여 하기와 같은 실험을 수행하였다. 단, 본 명세서 내에서 모든 동물 실험 과정은 안전성평가연구소 동물 자원 대학위원회의 지침서(Approval No. RS17003)에 따라 수행하였다.

다음으로, 상기 샘플을 50ml의 PBS로 2번 세척하였다. 상술한 과정에 의해 고정화된 샘플을, 20% CHAPS w/v% 및 60 w/v% 우레아의 PBS 혼합용액에서, 37℃ 온도 하에 220 rpm으로 3일 동안 인큐베이션하였다. 그 결과를 도 3에 나타내었다.
도 3은 본 발명에 따른 조직 투명화 방법을 통해 생쥐 뇌 투명화를 진행하기 전과, 투명화를 진행한 후를 관찰한 사진이며, 도 3의 좌측 사진이 투명화를 진행하기 전 생쥐 뇌를 나타내며, 도 3의 우측 사진이 투명화를 진행한 후 생쥐 뇌를 나타낸다.
이후, 20% CHAPS w/v% 및 60 w/v% 우레아의 PBS 혼합용액으로부터 생쥐 뇌를 3차 증류수로 옮기고, 50ml의 3차 증류수로 12 h 동안 3번 세척한 후, 마운팅(mounting) 용액인 CHAPS와 Urea의 혼합용액에 뇌를 옮긴 후, 뇌의 투명화를 관찰하였였다. 그 결과를 도 4에 나타내었다.
도 4는 본 발명에 따른 조직 투명화 방법을 통해 투명화가 완료된 생쥐의 뇌와, 형광 밝기를 확인한 사진이며, 도 4의 좌측이 투명화를 완료한 생쥐의 뇌이고, 도 4의 우측이 Ultraviolet (UV)를 통해 투명화가 완료된 뇌에 있는 GAD 67-GFP(Glutamic Acid Decarboxylase67-GFP) 형광의 밝기를 확인한 사진이다.
도 3 및 도 4에 나타난 바와 같이, CHAPS 및 우레아를 포함하는 생체 조직 투명화제는 생체 조직을 조직의 변성 없이 빠른 시간 내 투명화시킬 수 있음을 알 수 있다.

실험예
3> CHAPS 및
우레아에
의해 투명화된 생체조직의 형광 이미지 평가
본 발명에 따른 투명화 방법으로 투명화된 뇌의 형광을 확인하기 위해 광학 현미경(Leica)에서 1X 대물렌즈를 이용하여 생쥐 뇌의 면역 염색 형광과 Green fluorescent protein(GFP) 신호를 확인하였다. 그 결과를 도 5에 나타내었다.
도 5는 상기 <실험예 2>에서 투명화가 완료된 생쥐 뇌에 콜린 아세틸트랜스페라제(Choline Acetyltransferase) 항체를 처리한 후, GAD67-GFP와 Alexa fluor-647 형광을 관찰한 이미지이며, 도 5의 좌측이 GAD67-GFP 형광을 관찰한 이미지이고, 도 5의 우측이 Alexa fluor-647 형광을 관찰한 이미지이다.
도 6, 7, 그리고 8은 각각 Microscopy Lightsheet Z.1에서 5X, 20X 대물렌즈를 이용하여, 상기 <실험예 2>에서 투명화가 완료된 생쥐 뇌의 면역 염색이미지와 GFP 신호를 확인한 결과를 나타내는 이미지이다.
도 9는 상기 <실험예 2>에서 투명화가 완료된 생쥐 뇌에 타이로신 수산화 효소(Tyrosine Hydroxylase) 항체를 처리한 후, GAD67-GFP와 Alexa fluor-647 형광을 병합하여 관찰한 이미지이다.
도 10은 상기 <실험예 2>에서 투명화가 완료된 생쥐 뇌에 혈관염색 Lectin-647과 GAD67-GFP 형광 이미지이다.
도 11은 상기 <실험예 2>에서 투명화가 완료된 생쥐 뇌에 타이로신 수산화 효소(Tyrosine Hydroxylase) 항체를 처리한 후, Donkey Anti-rabbit IgG Alexa Fluor-647를 염색한 후 관찰한 형광 이미지이다.
상기 결과로부터, 본 발명에 따른 투명화 방법으로 투명화된 뇌는 형광 이미지 확인이 가능함을 알 수 있다.

실험예
4> CHAPS 및
우레아의
농도 변화에 따른 생체조직 투명화 평가
CHAPS 및 우레아를 포함하는 PBS 혼합용액에서, CHAPS와 우레아의 농도 변화에 따라 생체조직 투명화 정도에 변화가 있는지 평가하기 위하여 하기와 같은 실험을 수행하였다.
<실험예 2>에서는 20% CHAPS w/v% 및 60 w/v% 우레아의 PBS 혼합용액(20C + 60U%)을 사용하였으며; 본 실험에서는 추가로 10% CHAPS w/v% 및 65 w/v% 우레아의 PBS 혼합용액(10C + 65U%); 40% CHAPS w/v% 및 45 w/v% 우레아의 PBS 혼합용액(40C + 45U%)을 사용하여 <실험예 2>와 동일한 방법으로 생쥐 뇌의 투명화를 유도하였다. 그 결과를 도 12에 나타내었다.
도 12는 CHAPS 및 우레아를 포함하는 PBS 혼합용액에서, CHAPS와 우레아의 농도 변화에 따라 생체조직 투명화 정도에 변화가 있는지 평가한 결과를 나타내는 사진이다.
도 12에 나타난 바와 같이, CHAPS 및 우레아를 포함하는 PBS 혼합용액에서, CHAPS와 우레아의 농도를 변화시켜도, 생쥐 뇌의 투명화를 조직의 변성 없이 유도할 수 있음을 알 수 있다.

실험예
5> CHAPS 및 urea를 포함하는 조성물의 투명화 우수성 확인
상기 <실험예 2>와 같이 실험을 수행하여 생체 조직의 투명화를 유도하되, 우레아(urea)를 사용하는 대신 수크로오스(sucrose) 또는 글리세롤(glycerol)을 CHAPS와 함께 사용하여 비교군으로서 생체 조직의 투명화를 유도하였다. 또한, 우레아를 단독으로 사용하여 생체 조직의 투명화를 유도하였다. 나아가, 어떤 처리도 하지 않은 무처리군 생체조직도 관찰하였다. 그 결과를 도 13 내지 17에 나타내었다.

도 14는 20 w/v% CHAPS와 50 w/v% 수크로오스를 사용하여 생체 조직 투명화를 유도한 결과를 나타내는 이미지이다.

도 13 내지 17에 나타난 바와 같이,
CHAPS와 우레아를 함께 포함하는 생체 조직 투명화제로 생체 조직의 투명화를 유도할 경우,
수크로오스 또는 글리세롤을 더 포함하는 생체 조직 투명화용 조성물, 또는 우레아를 단독으로 포함하는 생체 조직 투명화제에 비해 단백질의 구조 변성 없이 조직의 빠른 투명화가 가능한 것을 알 수 있다.
특히, CHAPS와 우레아를 함께 포함하는 생체 조직 투명화제로 생체 조직의 투명화를 유도할 경우, 조직의 변성 없이, 5일 이내에, 투명화가 완료되는 반면(도 16 참조),
우레아를 단독으로 포함하는 생체 조직 투명화제로 생체 조직의 투명화를 유도할 경우, 뇌 조직 구조에 손상이 발생하고, 20일이 경과하여도 투명화 정도가 상대적으로 떨어짐을 알 수 있다(도 17 참조).
한편, CHAPS를 단독으로 처리한 도 2와, 우레아를 단독으로 처리한 도 17을 비교하면, CHAPS를 단독으로 처리한 경우는 조직이 투명화될 뿐만 아니라 조직의 크기가 유지되므로, 뇌 조직의 구조에 손상이 발생하지 않음을 확인할 수 있으나, 우레아를 단독으로 처리한 경우는 조직의 크기가 커지는 것으로부터 뇌 조직의 구조에 손상이 발생함을 알 수 있다.

실험예
6> 당류 전처리에 따른 조직의 및 단백질구조를 보존한 상태에서 생체 조직의 투명화 확인
당류 전처리 과정의 존재 여부에 따라 조직의 투명화 정도에 어떤 변화가 있는지 평가하기 위하여 하기와 같은 실험을 수행하였다. 단, 본 명세서 내에서 모든 동물 실험 과정은 안전성평가연구소 동물 자원 대학위원회의 지침서(Approval No. RS17003)에 따라 수행하였다.
먼저, 흡입 마취제인 이소푸루란(Isoflurane) (1cc/min) 을 사용하여 성인 생쥐(8주령)를 마취시키고, 경심 관류로 차가운 1X PBS에 50 mL을 관류시킨 후, 차가운 4% PFA (Paraformaldehyde)를 관류시켰다. 그 다음 기관을 적출하고 4%의 PFA용액(수용액)에 담근 후 4℃에서 24 h 동안 인큐베이션 하였다. 이때, 상기 차가운 온도 범위는 특별히 제한되는 것은 아니지만 0℃~10℃ 범위일 수 있다. (고정화 단계)
다음으로, 상기 샘플(기관)을 40% 수크로오스 용액에 0℃~10℃ 범위에서 24h 동안 인큐베이션 하였다. 이때, 상기 40% 수크로오스의 농도는 w/v%(중량/부피%)이며 수용액을 사용하였다. (전처리 단계)
대조군으로는 상기 전치리 단계를 수행하지 않고 상기 고정화 단계만 수행한 것으로 설정하였다.
도 18에 상기 고정화 단계와 전처리 단계를 모두 수행한 생쥐의 뇌 이미지와, 상기 고정화 단계만 수행한 생쥐의 뇌 이미지를 나타내었다. 도 18의 아래 형광 이미지는 Ultraviolet(UV)를 통해 뇌에 있는 GAD 67-GFP(Glutamic Acid Decarboxylase67-GFP) 형광의 밝기의 변화를 나타내는 이미지이다.
상기 샘플 조직을 투명화하기 위하여, 20% CHAPS w/v%, 50 w/v% Urea, 50 mM 소듐 아자이드(Sodium azide)의 혼합 수용액을 35℃에서 100 rpm으로 5h, 48h 동안 인큐베이션 하였다. (투명화 단계)
이때, 상기 CHAPS란 본 명세서 내 화학식 1로 표시되는 화합물 또는 이의 수화물이다.
도 19에 상기 고정화 단계, 전처리 단계, 5h 동안의 투명화 단계를 모두 수행한 생쥐의 뇌와, 전처리 단계만 수행하지 않은 상기 대조군 뇌 이미지를 나타내었다. 도 19의 아래 형광 이미지는 Ultraviolet(UV)를 통해 뇌에 있는 GAD 67-GFP(Glutamic Acid Decarboxylase67-GFP) 형광의 밝기의 변화를 나타내는 이미지이다.
도 20에 상기 고정화 단계, 전처리 단계, 48h 동안의 투명화 단계를 모두 수행한 생쥐의 뇌와, 전처리 단계만 수행하지 않은 상기 대조군 뇌 이미지를 나타내었다. 도 20의 아래 형광 이미지는 Ultraviolet(UV)를 통해 뇌에 있는 GAD 67-GFP(Glutamic Acid Decarboxylase67-GFP) 형광의 밝기의 변화를 나타내는 이미지이다.
도 19 및 도 20에 나타난 바와 같이, 수크로오스로 전처리한 생쥐 뇌는, 수크로오스로 전처리하지 않은 생쥐 뇌보다 CHAPS 및 Urea를 사용한 조직 투명화시 형광의 밝기를 유지함을 알 수 있다. 반면, 수크로오스로 전처리하지 않은 생쥐의 뇌는 GFP의 형광이 감소하는 것을 알 수 있다. GFP 형광의 감소는 단백질 구조의 손상을 의미한다. 수크로오스 전 처리는 단백질 구조를 유지를 위해서 중요한 단계임을 의미한다.
이로부터, 본 발명에 따른 당류를 전처리하는 단계를 포함하는 생체 조직 투명화 방법은, 당류를 전처리하는 단계를 포함하지 않는 생체 조직 투명화 방법에 비해 단백질 구조를 유지하는데 현저한 효과가 있음을 알 수 있다.

실험예
7> 세척과정 후 생체 조직의 투명화 확인
상기 <실험예 6>에서 투명화한 조직을 세척한 후 다시 투명화시킬 경우, 당류 전처리 과정의 존재 여부에 따라 조직의 투명화 정도에 어떤 변화가 있는지 평가하기 위하여 하기와 같은 실험을 수행하였다.
상기 <실험예 6>에서 투명화가 진행된 조직을 50ml의 멸균 증류수로 0℃~10℃ 범위에서 20 rpm으로 24 h 동안 3회 교체하며 인큐베이션 하였다. (도 21 참조)
그 후, 다시 20% CHAPS w/v%, 50 w/v% Urea, 50 mM 소듐 아자이드(Sodium azide)의 혼합 수용액을 35℃에서 100 rpm으로 24h 동안 인큐베이션 하였다. (도 22 참조)
도 22에 나타난 바와 같이, 세척 과정 후 조직을 다시 투명화 용액에 다시 넣었을 때 조직의 투명도가 현저히 좋아지는 것을 알 수 있다. 세척 과정 또한 조직의 투명화에 중요한 역할을 하는 것으로 나타났다. 더 나아가 수크로오스로 전처리한 생쥐 뇌는, 세척 과정을 거친 후, 다시 조직 투명화를 하여도 형광의 밝기를 유지함을 알 수 있다. 반면, 수크로오스로 전처리하지 않은 생쥐의 뇌는 GFP의 형광이 여전히 감소되어 있다. GFP 형광의 감소는 단백질 구조의 손상을 의미한다. 수크로오스 전 처리는 단백질 구조를 유지를 위해서 중요한 단계임을 의미한다.
상기 결과로부터, 본 발명에 따른 40% 수크로오스(Sucrose)의 전처리 과정은 생체 조직 투명화 조성물로 유발될 수 있는 단백질 및 조직의 손상으로부터 보호 효과가 우수하여 투명화된 조직에서 형광을 유지시킬 수 있는 현저한 효과가 있음을 알 수 있다.
CHAPS를 포함하는 생체 조직 투명화제를 사용한 생체 조직의 투명화 방법은 생체 조직 투명화에 유용하다.
Claims (12)
- 제1항에 있어서,상기 생체 조직 투명화용 조성물은,우레아(urea), CHAPSO(3-([3-Cholamidopropyl]dimethylammoni o)-2-hydroxy-1-propanesulfonate), 수크로오스(sucrose), 프룩토오스(fructose), 글리세롤, 다이아트라이조산(Diatrizoic acid), Triton X-100, Tween-20, 2,2'-싸이오디에탄올, 이오헥솔(Iohexol) 및 클로랄 수화물(Chloral hydrate)로 이루어지는 군으로부터 선택되는 1종 이상을 더 포함하는 것을 특징으로 하는,생체 조직 투명화용 조성물.
- 고정화된 생체 조직을,제1항의 생체 조직 투명화용 조성물과 접촉시켜 생체 조직을 투명화시키는 단계를 포함하는,생체 조직의 투명화 방법.
- 제3항에 있어서,상기 생체 조직은 뇌(brain), 혈관(blood vessel), 간(liver), 폐(lung), 신장(kidney), 췌장(pancreas) 또는 장(intestine)인 것을 특징으로 하는,생체 조직의 투명화 방법.
- 제3항에 있어서,상기 고정화된 생체 조직은 파라포름알데하이드(paraformaldehyde), 에텔렌글리콜 디글리시딜에테르(ethylene glycol diglycidyl ether), 디프로필렌 글리콜 디글리시딜에테르(dipropylene glycol diglycidyl ether), 1,4-부탄디올 디글리시딜에테르(1,4-butanediol diglycidyl ether), 글리세롤 폴리글리시딜 에테르(glycerol polyglycidyl ether), 글루타르알데하이드(glutaraldehyde) 및 폴리아크릴아마이드로 이루어진 군으로부터 선택되는 1종 이상으로부터 고정화된 생체 조직인 것을 특징으로 하는,생체 조직의 투명화 방법.
- 제3항에 있어서,상기 생체 조직의 투명화 방법은 4℃ 내지 50℃ 온도 범위에서 수행하는 것을 특징으로 하는,생체 조직의 투명화 방법.
- 고정화된 생체 조직에, 당류를 함유하는 용액을 처리하는 전처리 단계;상기 전처리된 생체 조직을, 제1항의 생체 조직 투명화용 조성물과 접촉시켜 생체 조직을 투명화시키는 단계를 포함하는,생체 조직의 투명화 방법.
- 제7항에 있어서,상기 당류는 단당류, 이당류 및 다당류로 이루어지는 군으로부터 선택되는 1종 이상인 것을 특징으로 하는,생체 조직의 투명화 방법.
- 제8항에 있어서,상기 단당류는 과당(fructose), 갈락토스(galactose), 글루코스(glucose) 또는 만노스(mannose)이고;상기 이당류는 수크로오스(sucrose), 락토스(lactose), 말토스(maltose), 트레할로스(trehalose), 투라노스(turanose) 또는 셀로비오스(cellobiose)이고; 및상기 다당류는 덱스트란(dextran), 디에틸아미노에틸-덱스트란(diethylamino ethyl-dextran), 덱스트린(dextrin), 셀룰로오스(cellulose) 또는 베타-글루칸(β-glucans)인 것을 특징으로 하는,생체 조직의 투명화 방법.
- 제7항에 있어서,상기 당류를 함유하는 용액은 당류를 함유하는 수용액인 것을 특징으로 하는,생체 조직의 투명화 방법.
- 제7항에 있어서,상기 당류를 함유하는 용액의 당류 농도는 10 내지 70 w/v% 인 것을 특징으로 하는,생체 조직의 투명화 방법.
- 당류 용액을 포함하는 생체 조직 투명화 전처리용 조성물; 및제1항의 생체 조직 투명화용 조성물을 포함하는,생체 조직 투명화용 키트.
