본 출원은 마이크로파 가열 전처리를 포함하는 레토르트 살균방법을 이용하여 종래의 살균방법에 비하여 야채, 육, 해물 원물 식감과 고추, 마늘, 생강 등의 향신야채 맛이 많이 남아 있으면서 곰팡이, 효모, 병원성균 및 포자형 내열성 미생물을 효과적으로 사멸시키는 살균방법에 관한 것이다.
최근 현대인의 생활 순환에 따라 전자레인지 조리식 등과 같은 편의형 가공 식품에 대한 수요가 증대하고 있다. 편의식품의 대표적인 예로는 레토르트 살균한 상온 유통 파우치, 트레이 제품이 있고 이들의 대량 생산을 위해서는 원료 전처리, 조미, 조리, 살균 및 포장 등 다양한 가공공정의 적용이 필요하며, 이러한 공정들에 대한 최적화 조건의 확립은 우수한 품질과 미생물 안전성 등의 확보에 있어서 무엇보다 중요한 과정이다.
이러한 편의식품의 주요 원료 중 하나가 마늘, 양파 등의 향신야채와 감자, 당근 등의 근채류로서 이 원료에는 토양에서 기인하는 바실러스 계통의 영양세포와 포자가 많이 생육하고 있어 미생물 안전성이 크게 저하되는 문제가 발생하게 된다.
특히 바실러스 계통의 내열성 포자형 미생물에 의한 오염은 신맛 발생에 의하여 관능품질 저하를 가져오게 되며 심한 경우 가스발생, 팽창 등의 원인이 되기 때문에 상품화를 위해서는 우선적으로 적절한 살균공정의 설계와 적용이 검토되어야 한다.
일반적으로 내열성 포자류를 사멸시키기 위해 상업적으로 밀봉 후 121의 고온, 고압에서 수 십분 가열하는 레토르트 살균방법을 적용하는데 내열성 포자는 사멸될 수 있으나 고온에 의해 원물의 맛, 외관, 조직감 등의 관능품질의 손상이 심하고 영양성분도 많이 파괴되어 고품질의 다양한 가공 식품 상품화에 있어서 제약점이 많다.
한편, 마이크로파 살균 기작은 주로 높은 주파수에 의한 유전체 역할을 하는 식품의 급속한 열발생에 의한 미생물 사멸로 밝혀져 있으며, 미생물에 대한 전자장의 비가열적 살균효과는 아직 명확하게 규명되어 있지 않다. 마이크로파를 이용한 가공 식품에 대한 살균 연구는 대부분 냉장 제품에 적용 가능한 저온살균(pasteurization)으로 50 ~ 82℃에서 주로 병원성 및 부패 미생물, 효소 불활성화 등에 관한 것으로 멸균(sterilization)에 있어 중요한 영향 인자인 에너지 사용량, 처리식품의 초기온도, 식품 및 포장재의 물리적 특성, 통과속도에 대한 세부 연구는 많이 수행되지 않았다.
따라서, 현재 상용화된 가공 및 살균 공정으로는 원물의 관능품질과 영양성분을 보존하면서 다양한 야채 원료 내 내열성 미생물을 효과적으로 제어할 수 있는 방법이 없는 문제점이 있었고, 품질을 보존하면서 미생물을 효과적으로 제어할 수 있는 새로운 살균방법이 요구되어 왔다.
본 출원은 상기의 원물의 관능품질과 영양성분을 보존하면서 다양한 야채 원료 내 내열성 미생물을 효과적으로 제어할 수 있는 방법이 없는 문제점을 해결하기 위한 것으로 원물의 관능품질과 영양성분을 보존하면서 미생물을 효과적으로 제어할 수 있는 살균방법을 제공하는 것을 목적으로 한다.

상기 마이크로파 가열처리에 의한 감균은 높은 주파수에 의한 유전체 역할을 하는 식품의 급속한 열발생에 의한 미생물 사멸에 의한 것으로 밝혀져 있다. 마이크로파에 의한 발열은 주파수 300MHz ~ 300GHz, 파장 1mm ~ 1m의 전자파 (electromagnetic wave)에 의한 쌍극자 (dipole) 및 분극 (polarization) 현상으로 발생하는 유전손실 (dielectric loss)에 의해 일어난다. 식품의 경우에는 마이크로파에 의한 물분자의 진동에너지로 급속한 발열이 일어나 빠른 건조 속도, 수분 균일화, 표면 경화 및 균열방지, 표백효과, 살균효과가 발생하여 현재, 살균, 해동, 조리, 팽화 등에 이용되고 있다. 특히 최근에는 미생물, 해충의 살균, 유통기한 연장, 살균시간 단축으로 품질향상 및 포장 후 살균처리 가능 등의 여러 장점으로 새로운 살균 방법으로 많이 활용되고 있다.
상기 마이크로파는 공지의 마이크로파 발생장치에 의해 발생될 수 있고, 바람직하게는 연속식 또는 배치식 마이크로파 가열장치를 이용할 수 있으나, 이에 한정되지 않는다. 상기 마이크로파는 700 ~ 1,200W 출력의 마이크로파 일 수 있다. 상기 마이크로파 출력이 700W 미만일 경우에는 감균효과가 미미하고 1,200W 초과일 경우에는 과도한 가열로 품질이 저하되는 문제점이 있다.
상기 1) 단계 감균효과를 향상시키기 위해 상기 마이크로파 가열처리는 가압상태에서 이루어질 수도 있다. 구체적으로 1,000 ~ 1,700mmHg 조건에서 이루어질 수 있다. 가압조건이 1,000mmHg 미만일 경우 감균효과의 향상을 기대하기 어렵고 1,700mmHg 초과할 경우 공정에 비용이 많이 들어가고 원물의 품질이 나빠질 수 있다.
상기 1) 단계는 1 ~ 30분간 이루어질 수 있다. 바람직하게는 2 ~ 10분간 이루어질 수 있다. 마이크로파 가열 처리가 1분 미만일 경우 전처리 감균의 효과를 보기 어렵고, 30분을 초과할 경우 원물이 과도하게 가열되어 전체 살균을 거친 뒤 품질이 저하되는 문제점이 있다.
본 출원의 상기 2) 단계는 전처리된 원물을 포장하는 단계이다. 전처리된 원물을 포장하는 포장재는 공지의 포장재를 사용할 수 있고, 바람직하게는 트레이 또는 파우치가 사용될 수 있으나, 이에 한정되지 않는다. 포장 장치는 공지의 장치를 사용할 수 있고, 바람직하게는 자동 로터리 팩커 또는 트레이 자동포장기를 사용할 수 있으나, 이에 한정되지 않는다. 상기 2) 단계의 일 예시로 전처리된 원물을 파우치 또는 트레이에 투입하고 소스 또는 육수를 투입한 뒤 실링하여 포장할 수 있다.
본 출원의 상기 3) 단계는 포장된 원물을 레토르트 살균하는 단계이다. 종래의 레토르트 살균은 밀봉 후 121℃의 고온, 고압에서 15 ~ 20분 내외 가열하므로 내열성 포자는 사멸될 수 있으나 장시간 고온에 의해 원물의 품질이 저해되는 문제가 있었다. 그러나 본 출원은 전술한 마이크로파 가열처리하는 전처리 감균단계를 포함하고 또는 추가적으로 천연항균제 용액 처리된 원물을 사용함으로써 종래의 레토르트 살균에 비하여 마일드한 레토르트 살균을 하더라도 종래의 살균방법과 동일한 감균효과가 있다. 상기 3) 단계는 공지의 장치에 의해 수행될 수 있고, 바람직하게는 열수식 또는 스프레이식 레토르트 설비로 살균할 수 있으나, 이에 한정되지 않는다. 또한, 상기 3) 단계는 종래의 레토르트 살균에 비하여 마일드한 조건에서 수행할 수 있다. 바람직하게는 110 ~ 115℃에서는 20 ~ 40분 가열하는 저온 레토르트 살균, 또는 121 ~ 125℃에서 5 ~ 15분, 바람직하게는 7 ~ 12분 가열하는 단기 레토르트 살균에 의해 가열살균 할 수 있다. 상기 3) 단계 이후 가공편의성을 위해 제품의 표면을 건조하는 단계를 더 포함할 수 있다.
본 출원의 살균방법은 종래의 레토르트 살균방법에 비하여 야채, 육, 해물을 원료로 사용하는 대용식, 반찬, 요리 등 다양한 편의형 가공 식품에서 열에 의한 고유의 맛, 향과 식감과 같은 품질의 손상을 최소화하여 야채, 육, 해물 원물 식감과 고추, 마늘, 생강 등의 향신야채 맛이 많이 남아 있도록 하고, 원료 내의 위생 및 품질저하에 관련된 곰팡이, 효모, 병원성균 및 포자형 내열성 미생물을 효과적으로 살균할 수 있는 효과가 있다.
한편, 선택적으로 본 출원의 살균방법은 상기 1) 단계에서 원물은 천연항균제 용액에 침지 처리된 원물일 수 있다. 전처리 감균전에 천연항균제 용액에 침지 처리함으로써 원물의 초기 미생물 균수를 줄여 이후의 전처리 감균 단계 및 마일드 레토르트 공정에서의 감균, 살균효과가 상승될 수 있다. 상기 천연항균제 용액은 유기산, 계면활성제, 박테리오신, 칼슘제제 중 선택되는 어느 하나 이상의 천연항균제를 포함할 수 있다. 상기 유기산은 젖산, 구연산, 피틴산, 사과산, 초산, 숙신산 중 하나 이상일 수 있다.
상기 천연항균제 용액은 천연항균제를 0.01 ~ 3.0중량% 포함하고 원물을 10 ~ 120분간 침지 처리하는 것일 수 있다. 천연항균제 중량이 0.01중량% 미만이거나 침지시간이 10분 미만인 경우 원물의 초기 미생물 균수를 감균 효과를 얻기 어렵고 천연항균제 중량이 3.0중량% 초과이거나 침지시간이 120분을 초과할 경우 원물의 관능에 영향을 미쳐 품질이 저하될 수 있다.
상기 천연항균제 용액 침지 처리의 일 예시로 천연항균제로서 박테리오신 계통인 니신(Nisin) 또는 니신을 함유한 발효 추출물, 그리고 폴리라이신, 비타민B1라우릴황산염 등의 계면활성제를 0.01 ~ 1.0중량% 포함한 용액에 원물을 침지하여 처리할 수 있다.
상기 천연항균제 용액 침지는 상기 마이크로파 가열처리하는 전처리 감균단계와의 조합을 통해 원물내의 초기균수를 감균시킴으로서 레토르트 조건을 마일드하게 하더라도 더욱 효과적으로 미생물을 살균하면서도 원물의 관능품질과 영양성분을 보존할 수 있는 효과가 있다.
또한, 선택적으로 본 출원의 살균방법은 상기 2) 단계에서 원물을 소스 또는 육수와 함께 포장할 수 있다. 상기 소스 또는 육수는 별도의 소스 또는 육수 제조단계를 통해 제조될 수 있다. 구체적으로는 목적하는 제품에 따라 결정된 소스 또는 육수의 재료 및 함량을 85 ~ 95℃에서 10 ~ 30분간 가열 교반하면서 제조될 수 있으나, 이에 한정되지 않는다.
상기 소스 또는 육수는 마이크로파 가열 처리하여 전처리 될 수 있다. 상기 마이크로파 가열 처리는 전술한 원물의 감균 처리와 동일할 수 있으나, 소스 또는 육수의 특성에 따라 세부적인 전처리 조건을 변경할 수 도 있다.
본 발명의 상기 가공 식품은 상온 유통용일 수 있다. 상기의 가공 식품의 살균방법에 의해 살균 된 가공 식품은 종래의 살균방법에 비하여 가공 식품의 원료가 열에 의해 발생하는 고유의 맛, 향과 식감과 같은 품질의 손상을 최소화하여 야채, 육, 해물 원물 식감과 고추, 마늘, 생강 등의 향신야채 맛이 많이 남아 있도록 하고, 원료 내의 위생 및 품질저하에 관련된 곰팡이, 효모, 병원성균 및 포자형 내열성 미생물을 효과적으로 살균할 수 있는 효과가 있어 상기 가공 식품은 상온 유통 가능하다.
이하에서는 실시예를 통해 본 출원을 더욱 구체적으로 설명한다. 그러나 실시예의 기재된 내용은 본 출원의 일 예시일 뿐 본 출원의 범위가 실시예의 범위로 한정되는 것은 아니다.
[실시예]
제조예 1: 마이크로파 가열 전처리 감균 및 마일드 레토르트 적용 야채, 육, 해물 및 소스(육수) 베이스 가공 식품 제조
본 출원의 마이크로파 가열 전처리 감균 및 마일드 레토르트 융합 살균 공법을 적용한 야채, 육, 해물 및 소스 또는 육수 베이스 가공 식품의 대표적인 제조 공정은 다음과 같으며, 그 공정도는 도 1에 나타내었다.
(1) 원물 원료 선별, 세척 및 절단 공정
야채, 육, 해물 표면에 붙어 있는 먼지, 이물 등 오염물질을 제거하기 위해 먼저 깨끗한 물로 3회 정도 세척한 다음, 메뉴 특성에 맞는 적당한 크기로 절단하여 원료로 준비한다.
(2) 천연항균제 용액 침지 전처리 감균 공정
선별, 세척 및 절단이 끝난 야채, 육, 해물 원료는 각각 물 중량 대비 0.01 ~ 3.0 (w/w, %)의 천연항균제 용액에서 30 ~ 120분간 침지한 후 세척하여 사용한다. 이때 사용하는 천연항균제는 유기산, 계면활성제, 박테리오신, 칼슘제 등으로 원물의 관능특성 및 미생물 현황 등을 감안하여 선택적으로 단독 또는 혼합하여 사용할 수 있다. 원료 내 내열성균 등의 자생균이 적은 경우 천연항균제 용액 침지 공정은 생략하고 바로 마이크로파 사전 가열 감균 공정을 적용할 수 있다.
(3) 마이크로파 가열 전처리 감균 공정
천연항균제 용액 침지를 끝낸 야채, 육, 해물 원물 원료는 연속식 또는 배치식 마이크로파 가열 장치를 통과시켜 고온에서 단시간 가열 살균을 실시한다. 마이크로파 가열 시간은 상압에서 2 ~ 10분 사이에서 원물 관능특성과 미생물 특성을 감안하여 선택적으로 적용할 수 있으며, 살균효과를 향상시키기 위해 가압 상태에서 가열살균을 실시할 수 있다. 또한, 마이크로파 가열 전처리 공정은 살균 효과외에도 물 또는 스팀 블렌칭 효과를 낼 수 있는 장점도 있다.
또한, 야채, 육, 해물 원물 외에 소스 또는 육수도 마이크로 가열 설비를 이용하여 연속적으로 살균할 수 있다.
(4) 소스 또는 육수 제조 공정
소스 또는 육수 원료를 계량, 준비하여 혼합 후 85 ~ 95℃에서 10 ~ 30분간 가열하여 제조한다. 향신야채 등의 원료를 많이 사용하는 소스 또는 육수의 경우 원료 내 내열성균이 많을 자생하고 있으므로 선택적으로 마이크로파 가열 살균 설비를 통과시켜 전처리 감균할 수 있다.
(5) 원물 과 소스 또는 육수 포장 공정
마이크로파 가열 전처리 감균한 야채, 육, 해물 원물과 소스 또는 육수를 메뉴 특성 감안하여 파우치 또는 트레이에 정량 투입하여 실링, 포장한다. 파우치 경우는 자동 로터리 팩커 설비를, 트레이 경우는 트레이 자동 포장기를 이용하여 중량 및 이물 확인 후 포장한다.
(6) 마일드 레토르트 본살균 공정
파우치 또는 트레이에 포장한 완제품은 열수식 또는 스프레이식 레토르트 설비를 이용하여 110 ~ 121℃에서 10 ~ 40분간 가열 살균한다. 이때 살균 조건은 천연항균제 침지 및 마이크로파 가열를 이용한 전처리 감균을 사전 적용 하였으므로 기존 레토르트 조건 보다 50% 내외 저감화된 가열 살균 조건을 적용할 수 있다. 구체적으로 110 ~ 115℃ 에서는 20 ~ 40분 가열하는 저온 레토르트 살균 내지 121 ~ 125℃에서 5 ~ 15분, 바람직하게는 7 ~ 12분 가열하는 단기 레토르트 살균에 의해 가열살균 할 수 있다.
실험예 1: 원물내 미생물에 대한 마이크로파 가열 전처리 감균 및 마일드 레 토르트 조합 살균 효과 측정
야채, 육, 해물의 마이크로파 가열 전처리 감균 처리는 대상시료를 2-10분간 각각 처리 후 총균, 바실러스 계통의 내열성균, 효모 및 곰팡이와 같은 진균류에 대한 살균 효과를 각각 고찰 하였다.
(1) 야채에 마이크로파 가열 적용시 살균 효과 측정
상온 유통 제품에서 주요 살균 목표균인 바실러스 계통 내열성균에 대한 마이크로파 가열살균 효과를 살펴 본 결과, 도 2 에서와 같이 바실러스 아밀로리쿠에파시엔스 (Bacillus
amyloliquenfaciens)를 대상으로 했을 경우 3분 이상 가열시 102 (CFU/g) 내외의 감균 효과를 나타내었다. 통상적으로 가공 식품 내 포자형성 미생물수는 102 ~ 103 (CFU/g)이므로 102 (CFU/g) 내외의 감균 효과를 나타내는 마이크로파 가열을 상업적인 전처리 살균 방법으로 적용할 수 있다.
(2) 야채 내 총균 , 내열성균 및 진균에 대한 마이크로파 가열 및 마일드 레 토르트 조합 살균 효과 측정
제조예 1에서 처리한 야채 원물에 대한 마이크로파 단독 가열 및 마일드 레토르트 조합 후 감균 및 살균 효과를 관찰하였고 종래 가열 레토르트 살균 효과와 비교하여 그 결과를 표 1에 나타내었다.

표 1에서와 같이 야채의 대표 시료인 감자, 고구마, 단호박 모두 마이크로파 가열 처리만 할 때 총균은 104 (CFU/g) 내외, 내열성균은 101 (CFU/g) 내외 감균 되었으며, 진균류는 모두 사멸되었다. 마이크로파 가열 전처리 감균 후 마일드 레토르트를 조합 적용 시 모든 균이 사멸되었다. 또한, 본 출원의 마이크로파 전처리 및 마일드 레토르트 처리 시 종래의 레토르트 처리와 비교하여 전반 맛의 점수가 높았다. 따라서 본 출원의 살균방법을 이용하면 기존 레토르트 방식보다 고온 가열에 의한 관능품질 손상을 줄일 수 있어 상온 제품의 품질향상을 도모할 수 있다는 것을 확인할 수 있었고 이는 본 출원의 차별화 요소라 할 수 있다.
(3) 육 및 해물 내 총균 , 내열성균 및 진균에 대한 마이크로파 가열 및 마일 드 레토르트 조합 살균 효과 측정
제조예 1에서 처리한 육 및 해물 원물에 대한 마이크로파 단독 가열 및 마일드 레토르트 조합 후 감균 및 살균 효과를 관찰하였고 종래 가열 레토르트 살균 효과와 비교하여 그 결과를 표 2 에 나타내었다.

표 2와 같이 육 및 해물의 대표 시료인 닭고기, 쇠고기, 새우 모두 마이크로파 가열 처리만 할 때 총균은 103 (CFU/g) 내외, 내열성균은 101 (CFU/g) 내외 감균 되었으며, 진균류는 모두 사멸되었다. 본 출원의 마이크로파 가열 전처리 감균 후 마일드 레토르트를 조합 적용 시 모든 균이 사멸되었다. 또한, 본 출원의 마이크로파 전처리 및 마일드 레토르트 처리 시 종래의 레토르트 처리와 비교하여 전반 맛의 점수가 높았다. 따라서 본 출원의 살균방법을 이용하면 기존 레토르트 방식보다 고온 가열에 의한 관능품질 손상을 줄일 수 있어 상온 제품의 품질향상을 도모할 수 있다는 것을 확인할 수 있었고 이는 본 출원의 차별화 요소라 할 수 있다.
이상 결과에서 야채, 육, 해물 등의 식품 원료 내 다양한 자생균과 오염균을 마이크로파 가열과 마일드 레토르트를 적절히 조합시 품질을 향상시키면서 멸균에 도달시킬 수 있다는 결론을 도출하여 본 출원의 마이크로파 가열 살균법을 전처리 살균 기술로서 유용하게 활용할 수 있었다.

표 3과 같이 1중량% 유기산 베이스 천연항균제에 60분간 침지 시 총균수 및 내열성균이 101-102 (CFU/g) 내외 감균되어 마이크로파 가열 및 마일드 레토르트 조건의 저감화가 가능하여 품질 향상을 더 한층 가져올 수 있을 것으로 기대되었다. 또한, 본 출원의 천연 항균제 처리한 원물을 마이크로파 전처리 및 마일드 레토르트 처리 시 종래의 레토르트 처리와 비교하더라도 전반 맛의 점수가 높았다. 따라서 천연항균제에 원물을 침지하는 단계를 추가하더라도 종래의 레토르트 처리보다 상온 제품의 품질향상을 도모할 수 있다는 것을 확인할 수 있다.
이상의 연구 결과에서, 천연항균제 침지 및 마이크로파 가열 전처리 감균과 마일드 열살균 공법을 융합, 적용하였을 때 야채, 육, 해물 원물 베이스의 가공 식품내 영양세포 및 내열성 포자류, 진균류 등의 미생물이 멸균 수준에 도달되어 기존 고온, 고압 살균 방식인 레토르트 제품 대비 동일한 살균 효과를 거두면서도 레토르트 가열살균 강도를 50% 내외 저감화 할 수 있어, 본 출원의 가공 식품 살균 기술은 향후 차별화 상온 제품 개발에 활용할 수 있는 유용한 기술인 것을 알 수 있다.
