すなわち、本発明は以下の通りである。
[1]水酸基、チオール基、アミノ基、カルボキシル基、チオカルボキシル基、及び酸アミド基からなる群より選ばれる、塩素と反応し得る官能基を少なくとも1つ有する反応基質と塩素含有化合物とを、炭素数9~40のトリアルキルアミンと有機溶媒と共にフロー式リアクターに供給し、前記反応基質と塩素含有化合物とを反応させる有機化合物の製造方法。
[2]前記フロー式リアクターが2以上の原料供給口と、供給された原料を混合する混合部と、混合液を流通するリアクター部とを有しており、

[9]前記反応基質が(S)-1-フェニル-1,2,3,4-テトラヒドロイソキノリンであり、前記塩素含有化合物がホスゲン(5)又はその合成等価体であり、これらの反応から得られる前記有機化合物が塩化(S)-1-フェニル-1,2,3,4-テトラヒドロイソキノリンカルボニルである、[6]に記載の製造方法。

ここでR4は、反応基質のうち前記塩素反応性基以外の部分を表す。n、mは、それぞれ独立して1~3の整数を表す。n及びmは、好ましくは1~2であり、より好ましくは1である。
NR5のR5となるアルケニル基としては、エテニル基、プロペニル基、ブテニル基、ペンテニル基、ヘキセニル基等の炭素数2~10の基が好ましく、炭素数2~4の基がより好ましい。
NR5のR5となるシクロアルキル基としては、シクロプロピル基、シクロブチル基、シクロペンチル基、シクロヘキシル基等の炭素数3~10の基が好ましく、炭素数5~6の基がより好ましい。
NR5のR5となるアラルキル基としては、ベンジル基、フェネチル基、フェニルプロピル基等の炭素数7~15の基が好ましく、炭素数7~10の基がより好ましい。
NR5のR5となるアリール基としては、フェニル基、トルイル基、ナフチル基等の炭素数6~10の基が好ましく、炭素数6~8の基がより好ましい。
NR5のR5となるヘテロアリール基としては、ピリジニル基、ピラジニル基、ピリミジニル基、ピリダジニル基等が挙げられる。
NR5のR5となるアルコキシ基としては、メトキシ基、エトキシ基、プロポキシ基、ブトキシ基等の炭素数1~10の基が好ましく、炭素数1~4の基がより好ましい。
NR5のR5となるアラルキルオキシ基としては、ベンジルオキシ基、フェネチルオキシ基等の炭素数7~15の基が好ましく、炭素数7~10の基がより好ましい。
NR5のR5となるアリールオキシ基としては、フェニルオキシ基、ナフチルオキシ基等の炭素数6~10の基が好ましく、炭素数6~8の基がより好ましい。
前記R5の基が有していてもよい置換基としては、例えば、フッ素原子;メトキシ基、エトキシ基、フェノキシ基、ベンジルオキシ基等のアルコキシ基;エポキシ基等の環状エーテル基;メチルチオ基等のアルキルチオ基;トリフルオロメチル基;アセチル基;ベンゾイル基;シアノ基;ニトロ基;メトキシカルボニル基、エトキシカルボニル基等のアルコキシカルボニル基;ジメチルアミノ基、ジエチルアミノ基、ピロリジル基等のジアルキルアミノ基;ベンジルオキシカルボニルアミノ基、tert-ブチルカルボニルアミノ基、アセチルアミノ基、ベンゾイルアミノ基等の保護アミノ基等が挙げられる。また置換基は2価の基であってもよく、例えば、-CH2-O-などが含まれる。-CH2-O-の2つの結合手が同一の炭素原子に結合する場合、オキシラン環が形成される。置換基の数に制限はない。
(式中、R6、R7、及びR8は、反応基質のうち前記塩素反応性官能基以外の部分を表す。Xは、前記と同じである。Xが複数ある場合、それらは同一でもよく、異なっていてもよい。)
R6~R8も、R4と同様、反応基質に応じて種々の複雑な構造をとることが可能であるが、単純な構造であってもよい。反応基質が単純構造の基質の場合、R6としては、例えば、置換基を有していてもよい炭素数1~20のアルキル基、置換基を有していてもよい炭素数2~20のアルケニル基、置換基を有していてもよい炭素数3~20のシクロアルキル基、置換基を有していてもよい炭素数7~20のアラルキル基、置換基を有していてもよい炭素数6~20のアリール基、又は置換基を有していてもよい炭素数3~20のヘテロアリール基が挙げられる。R6のアルキル基、アルケニル基、シクロアルキル基、アラルキル基、アリール基、ヘテロアリール基としては、R5と同様の基が挙げられる。またR6の置換基もR5と同様の基が挙げられる。
なおXがNR5である時、R5とR6は互いに結合してもよい。R5とR6が結合した場合、窒素原子を構成要素として含む環が形成されるが、該環としては、R5とR4が結合して形成される環と同様の環が例示される。すなわち、R5とR6が結合して形成される環としては、構造式中に「X-R4」を有する場合と同様の環が挙げられる。
なお反応基質と塩素含有化合物とは適当に組み合わせることが可能であり、特に以下の組み合わせが好ましく、その時に得られる生成物は以下の通りである。
また反応基質としてのベンジルアルコール(式(a1)の化合物の一つ)が塩化ベンゾイル(式(2)の化合物の一つ)と反応することで、安息香酸ベンジル(式(10)の化合物の一つ)が生成物として得られる。
反応基質としての(R)-1-フェニルエチルアミン(式(a1)の化合物の一つ)が塩化ベンゾイル(式(2)の化合物の一つ)と反応する場合は、(R)-N-ベンゾイル-α-フェニルエチルアミン(式(10)の化合物の一つ)が生成物として得られる。
反応基質としての(S)-1-フェニル-1,2,3,4-テトラヒドロイソキノリン(式(a1)の化合物の一つ)がホスゲン(5)又はその合成等価体と反応する場合は、塩化(S)-1-フェニル-1,2,3,4-テトラヒドロイソキノリンカルボニル(式(13)の化合物の一つ)が生成物として得られる。
式(a1)~(a3)で表される反応基質から得られる生成物としては、好ましくは安息香酸ベンジル、(R)-N-ベンゾイル-α-フェニルエチルアミン、塩化(S)-1-フェニル-1,2,3,4-テトラヒドロイソキノリンカルボニル、(2R,3R)-3-(2,4-ジフルオロフェニル)-3,4-エポキシ-2-ブチルメタンスルホネートであり、特に好ましくは(2R,3R)-3-(2,4-ジフルオロフェニル)-3,4-エポキシ-2-ブチルメタンスルホネートである。
例えば、好ましい反応基質であるN-[1-(S)-エトキシカルボニル-3-フェニルプロピル]-L-アラニン(式(a6)の化合物の一つ)、4-[({(2S,5R)-5-[(ベンジロキシ)アミノ]ピペリジン-2-イル}カルボニル)アミノ]ピペリジン-1-カルボン酸ベンジル(式(a5)の化合物の一つ)、4-[({(2S,5R)-5-[(ベンジロキシ)アミノ]ピペリジン-2-イル}カルボニル)アミノ]ピペリジン-1-カルボン酸tert-ブチル(式(a5)の化合物の一つ)、(2S,5R)-5-ベンジロキシアミノ-ピペリジン-2-カルボン酸ベンジル(式(a5)の化合物の一つ)、(2S,5R)-5-ベンジロキシアミノ-ピペリジン-2-カルボン酸メチル(式(a5)の化合物の一つ)、(2S,5R)-5-ベンジロキシアミノ-ピペリジン-2-カルボン酸アミド(式(a5)の化合物の一つ)がホスゲン又はトリホスゲンと反応することで、N-[1-(S)-エトキシカルボニル-3-フェニルプロピル]-L-アラニル-N-カルボン酸無水物(式(37)の化合物の一つ)、4-({[(2S,5R)-6-(ベンジロキシ)-7-オキソ-1,6-ジアザビシクロ[3.2.1.]オクト-2-イル]カルバモイル}アミノ)ピペリジン-1-カルボン酸ベンジル(式(36)の化合物の一つ)、4-({[(2S,5R)-6-(ベンジロキシ)-7-オキソ-1,6-ジアザビシクロ[3.2.1.]オクト-2-イル]カルバモイル}アミノ)ピペリジン-1-カルボン酸tert-ブチル(式(36)の化合物の一つ)、(4R,6S)-3-(ベンジルオキシ)-2-オキソ-1,3-ジアザビシクロ[2,2,1]ヘプタン-6-カルボン酸ベンジル(式(36)の化合物の一つ)、(4R,6S)-3-(ベンジルオキシ)-2-オキソ-1,3-ジアザビシクロ[2,2,1]ヘプタン-6-カルボン酸メチル(式(36)の化合物の一つ)、(4R,6S)-3-(ベンジルオキシ)-2-オキソ-1,3-ジアザビシクロ[2,2,1]ヘプタン-6-カルボン酸アミド(式(36)の化合物の一つ)が生成物として得られる。
前記フロー式リアクターとしては、マイクロリアクター、サイクロン型反応器、積層型マイクロ流体チップ等の公知の装置を適宜利用できる。
下記表1に示す種々のアミン1gを下記表1に示す各溶媒10mLに加え、濃度10%(重量/容量)溶液を調製した。この溶液に、塩酸のn-プロパノール溶液(塩酸濃度34重量%)を1.1当量(=塩化水素のモル量/アミンのモル量)となるまで添加し、室温で1時間攪拌した。攪拌後、固体の析出の有無を目視で確認した。表中、○は固体が析出していなかった事を意味し、×は固体が析出していた事を意味する。

以下の実施例1~4及び比較例1、2では、(2R,3R)-3-(2,4-ジフルオロフェニル)-3,4-エポキシ-2-ブチルアルコールとメタンスルホニルクロリドから(2R,3R)-3-(2,4-ジフルオロフェニル)-3,4-エポキシ-2-ブチルメタンスルホネートを製造した。HPLC法によって生成物を定量し、収率を算出した。HPLC条件は以下の通りである。

(2R,3R)-3-(2,4-ジフルオロフェニル)-3,4-エポキシ-2-ブチルアルコール3.00gにトルエン5.54gとトリブチルアミン4.44gを入れて均一溶液とし、A液とした。また、メタンスルホニルクロリド2.57gにトルエン10.68gを入れて均一溶液とし、B液とした。T字ミキサー(内径:2mm、材質:ポリテトラフルオロエチレン(PTFE))及び滞留ライン中(チューブ内径:2mm、材質:ポリテトラフルオロエチレン(PTFE))を50℃の恒温バスに入れた後、A液及びB液をそれぞれ2ml/分の速度にてダイアフラムポンプ(KNF社製)で送液・T字ミキサー内で混合し、滞留ライン内で12分間通流して反応させた(アミン/溶媒=27.5/100(重量比))。反応液はフラスコに入った水6.00g中に撹拌下、連続的にクエンチし、分液後、(2R,3R)-3-(2,4-ジフルオロフェニル)-3,4-エポキシ-2-ブチルメタンスルホネートを4.08g含有する有機層を47.08g取得した(収率98%)。尚、反応中に結晶は析出せず、反応液はクリアーな溶液であった。
(2R,3R)-3-(2,4-ジフルオロフェニル)-3,4-エポキシ-2-ブチルアルコール3.00gにトルエン5.54gとトリブチルアミン4.44gを入れて均一溶液とし、A液とした。また、メタンスルホニルクロリド2.57gにトルエン10.68gを入れて均一溶液とし、B液とした。T字ミキサー(内径:2mm、材質:ポリテトラフルオロエチレン(PTFE))及び滞留ライン中(チューブ内径:2mm、材質:ポリテトラフルオロエチレン(PTFE))を0℃の恒温バスに入れた後、A液及びB液をそれぞれ2ml/分の速度にてダイアフラムポンプ(KNF社製)で送液・T字ミキサー内で混合し、滞留ライン内で3分間通流して反応させた(アミン/溶媒=27.5/100(重量比))。反応液はフラスコに入った水6.00g中に撹拌下、連続的にクエンチし、分液後、(2R,3R)-3-(2,4-ジフルオロフェニル)-3,4-エポキシ-2-ブチルメタンスルホネートを4.13g含有する有機層を41.34g取得した(収率99%)。尚、反応中に結晶は析出せず、反応液はクリアーな溶液であった。
(2R,3R)-3-(2,4-ジフルオロフェニル)-3,4-エポキシ-2-ブチルアルコール3.00gにトルエン5.54gとトリプロピルアミン3.44gを入れて均一溶液とし、A液とした。また、メタンスルホニルクロリド2.57gにトルエン9.68gを入れて均一溶液とし、B液とした。T字ミキサー(内径:2mm、材質:ポリテトラフルオロエチレン(PTFE))及び滞留ライン中(チューブ内径:2mm、材質:ポリテトラフルオロエチレン(PTFE))を0℃の恒温バスに入れた後、A液及びB液をそれぞれ2ml/分の速度にてダイアフラムポンプ(KNF社製)で送液・T字ミキサー内で混合し、滞留ライン内で12分間通流して反応させた(アミン/溶媒=22.7/100(重量比))。反応液はフラスコに入った水6.00g中に撹拌下、連続的にクエンチし、分液後、(2R,3R)-3-(2,4-ジフルオロフェニル)-3,4-エポキシ-2-ブチルメタンスルホネートを4.17g含有する有機層を46.90g取得した(収率100%)。尚、反応中に結晶は析出せず、反応液はクリアーな溶液であった。
(2R,3R)-3-(2,4-ジフルオロフェニル)-3,4-エポキシ-2-ブチルアルコール3.00gにトルエン5.54gとトリオクチルアミン8.48gを入れて均一溶液とし、A液とした。また、メタンスルホニルクロリド2.57gにトルエン14.68gを入れて均一溶液とし、B液とした。T字ミキサー(内径:2mm、材質:ポリテトラフルオロエチレン(PTFE))及び滞留ライン中(チューブ内径:2mm、材質:ポリテトラフルオロエチレン(PTFE))を0℃の恒温バスに入れた後、A液及びB液をそれぞれ2ml/分の速度にてダイアフラムポンプ(KNF社製)で送液・T字ミキサー内で混合し、滞留ライン内で12分間通流して反応させた(アミン/溶媒=42.1/100(重量比))。反応液はフラスコに入った水6.00g中に撹拌下、連続的にクエンチし、分液後、(2R,3R)-3-(2,4-ジフルオロフェニル)-3,4-エポキシ-2-ブチルメタンスルホネートを3.96g含有する有機層を62.38g取得した(収率95%)。尚、反応中に結晶は析出せず、反応液はクリアーな溶液であった。
フラスコに(2R,3R)-3-(2,4-ジフルオロフェニル)-3,4-エポキシ-2-ブチルアルコール1.00g、トルエン11.00g、THF10.20g及びトリエチルアミン0.66gを入れて撹拌下、内温を0℃に温調した。次に、メタンスルホニルクロリド0.69gを内温を維持する速度でゆっくりと添加し、添加後、0℃にて1時間撹拌した(アミン/溶媒=3.1/100(重量比))。続いて反応液に水を2g入れてクエンチし、分液後、(2R,3R)-3-(2,4-ジフルオロフェニル)-3,4-エポキシ-2-ブチルメタンスルホネートを1.38g含有する有機層を22.86g取得した(収率99%)。尚、アミン濃度が参考例より低くても、反応中に結晶が析出し、反応液はスラリーであった。
比較例2(バッチ式)
フラスコに(2R,3R)-3-(2,4-ジフルオロフェニル)-3,4-エポキシ-2-ブチルアルコール1.00g、トルエン9.42g及びトリエチルアミン0.66gを入れて撹拌下、内温を0℃に温調した。次に、メタンスルホニルクロリド0.69gを内温を維持する速度でゆっくりと添加し、添加後、0℃にて1時間撹拌した(アミン/溶媒=7.0/100(重量比))。続いて反応液に水を2g入れてクエンチし、分液後、(2R,3R)-3-(2,4-ジフルオロフェニル)-3,4-エポキシ-2-ブチルメタンスルホネートを1.31g含有する有機層を10.84g取得した(収率94%)。尚、アミン濃度が参考例より低くても、反応中に結晶が析出し、反応液はスラリーであった。
以下の実施例5~6及び比較例3では、ベンジルアルコールと塩化ベンゾイルから安息香酸ベンジルを製造した。HPLC法によって生成物を定量し、収率を算出した。HPLC条件は以下の通りである。

フラスコにベンジルアルコール1.00g、トルエン7.60g及びトリエチルアミン1.22gを入れて撹拌下、内温を0℃に温調した。次に、塩化ベンゾイル1.56gを内温を維持する速度でゆっくりと添加し、添加後、0℃にて1時間撹拌した(アミン/溶媒=16.1/100(重量比))。続いて反応液に水を3.33g入れてクエンチし、分液後、安息香酸ベンジルを1.92g含有する有機層を9.83g取得した(収率98%)。尚、反応中に結晶が析出し、反応液はスラリーであった。
以下の実施例7~8及び比較例4では、(R)-1-フェニルエチルアミンと塩化ベンゾイルから(R)-N-ベンゾイル-α-フェニルエチルアミンを製造した。HPLC法によって生成物を定量し、収率を算出した。HPLC条件は以下の通りである。

(R)-1-フェニルエチルアミン3.00gにテトラヒドロフラン5.11gとトリブチルアミン5.97gを入れて均一溶液とし、A液とした。また、塩化ベンゾイル4.18gにテトラヒドロフラン11.53gを入れて均一溶液とし、B液とした。T字ミキサー(内径:2mm、材質:ポリテトラフルオロエチレン(PTFE))及び滞留ライン中(チューブ内径:2mm、材質:ポリテトラフルオロエチレン(PTFE))を20℃の恒温バスに入れた後、A液及びB液をそれぞれ2ml/分の速度にてダイアフラムポンプ(KNF社製)で送液・T字ミキサー内で混合し、滞留ライン内で2分間通流して反応させた(アミン/溶媒=35.9/100(重量比))。反応液はフラスコに入った水30.00g中に撹拌下、連続的にクエンチすることで、(R)-N-ベンゾイル-α-フェニルエチルアミン5.58gを取得した(収率100%)。尚、反応中に結晶は析出せず、反応液はクリアーな溶液であった。
フラスコに(R)-1-フェニルエチルアミン0.50g、テトラヒドロフラン2.50g及びトリエチルアミン0.54gを入れて撹拌下、内温を0℃に温調した。次に、塩化ベンゾイル0.70gを内温が維持可能な速度でゆっくりと添加し、添加後、0℃にて1時間撹拌した(アミン/溶媒=21.6/100(重量比))。続いて反応液に水を5.00g入れてクエンチし、分液後、(R)-N-ベンゾイル-α-フェニルエチルアミンを0.93g含有する有機層を2.43g取得した(収率100%)。尚、反応中に結晶が析出し、反応液はスラリーであった。
以下の実施例9~11では、N-[1-(S)-エトキシカルボニル-3-フェニルプロピル]-L-アラニンとトリホスゲンからN-[1-(S)-エトキシカルボニル-3-フェニルプロピル]-L-アラニル-N-カルボン酸無水物を製造した。HPLC法によって生成物を定量し、収率を算出した。HPLC条件は以下の通りである。

N-[1-(S)-エトキシカルボニル-3-フェニルプロピル]-L-アラニン1.50gにテトラヒドロフラン13.50gとトリブチルアミン1.09gを入れて均一溶液とし、A液とした。また、トリホスゲン0.64gにトルエン15.50gを入れて均一溶液とし、B液とした。T字ミキサー(内径:2mm、材質:ポリテトラフルオロエチレン(PTFE))及び滞留ライン中(チューブ内径:2mm、材質:ポリテトラフルオロエチレン(PTFE))を60℃の恒温バスに入れた後、A液及びB液をそれぞれ2ml/分の速度にてダイアフラムポンプ(KNF社製)で送液・T字ミキサー内で混合し、滞留ライン内で3分間通流して反応させた(アミン/溶媒=3.8/100(重量比))。反応液はフラスコに入った18%リン酸水75.00g中に撹拌下、連続的にクエンチし、分液後、N-[1-(S)-エトキシカルボニル-3-フェニルプロピル]-L-アラニル-N-カルボン酸無水物を1.25g含有する有機層を40.21g取得した(収率76%)。尚、反応中に結晶は析出せず、反応液はクリアーな溶液であった。
N-[1-(S)-エトキシカルボニル-3-フェニルプロピル]-L-アラニン1.50gにテトラヒドロフラン13.50gとトリブチルアミン1.09gを入れて均一溶液とし、A液とした。また、トリホスゲン0.64gにトルエン15.50gを入れて均一溶液とし、B液とした。T字ミキサー(内径:2mm、材質:ポリテトラフルオロエチレン(PTFE))及び滞留ライン中(チューブ内径:2mm、材質:ポリテトラフルオロエチレン(PTFE))を60℃の恒温バスに入れた後、A液及びB液をそれぞれ2ml/分の速度にてダイアフラムポンプ(KNF社製)で送液・T字ミキサー内で混合し、滞留ライン内で4分間通流して反応させた(アミン/溶媒=3.8/100(重量比))。反応液はフラスコに入った18%リン酸水75.00g中に撹拌下、連続的にクエンチし、分液後、N-[1-(S)-エトキシカルボニル-3-フェニルプロピル]-L-アラニル-N-カルボン酸無水物を1.25g含有する有機層を46.59g取得した(収率76%)。尚、反応中に結晶は析出せず、反応液はクリアーな溶液であった。
N-[1-(S)-エトキシカルボニル-3-フェニルプロピル]-L-アラニン1.50gにテトラヒドロフラン13.50gとトリブチルアミン1.09gを入れて均一溶液とし、A液とした。また、トリホスゲン0.64gにトルエン15.50gを入れて均一溶液とし、B液とした。T字ミキサー(内径:2mm、材質:ポリテトラフルオロエチレン(PTFE))及び滞留ライン中(チューブ内径:2mm、材質:ポリテトラフルオロエチレン(PTFE))を35℃の恒温バスに入れた後、A液及びB液をそれぞれ2ml/分の速度にてダイアフラムポンプ(KNF社製)で送液・T字ミキサー内で混合し、滞留ライン内で4分間通流して反応させた(アミン/溶媒=3.8/100(重量比))。反応液はフラスコに入った18%リン酸水75.00g中に撹拌下、連続的にクエンチし、分液後、N-[1-(S)-エトキシカルボニル-3-フェニルプロピル]-L-アラニル-N-カルボン酸無水物を1.13g含有する有機層を45.00g取得した(収率69%)。尚、反応中に結晶は析出せず、反応液はクリアーな溶液であった。
以下の実施例12~14では、4-[({(2S,5R)-5-[(ベンジロキシ)アミノ]ピペリジン-2-イル}カルボニル)アミノ]ピペリジン-1-カルボン酸ベンジルとトリホスゲンから4-({[(2S,5R)-6-(ベンジロキシ)-7-オキソ-1,6-ジアザビシクロ[3.2.1.]オクト-2-イル]カルバモイル}アミノ)ピペリジン-1-カルボン酸ベンジルを製造した。HPLC法によって生成物を定量し、収率を算出した。HPLC条件は以下の通りである。

4-[({(2S,5R)-5-[(ベンジロキシ)アミノ]ピペリジン-2-イル}カルボニル)アミノ]ピペリジン-1-カルボン酸ベンジル250mgにテトラヒドロフラン2.53gとトリブチルアミン317.9mgを入れて均一溶液とし、A液とした。また、トリホスゲン127.2mgにテトラヒドロフラン2.97gを入れて均一溶液とし、B液とした。T字ミキサー(内径:2mm、材質:ポリテトラフルオロエチレン(PTFE))及び滞留ライン中(チューブ内径:2mm、材質:ポリテトラフルオロエチレン(PTFE))を0℃の恒温バスに入れた後、A液及びB液をそれぞれ2ml/分の速度にてシリンジポンプ(YMC社製)で送液・T字ミキサー内で混合し、滞留ライン内で2分間通流して反応させた(アミン/溶媒=5.8/100(重量比))。反応液はフラスコに入った13%リン酸水1.56gとトルエン4.00gの混液中に撹拌下、連続的にクエンチすることで、4-({[(2S,5R)-6-(ベンジロキシ)-7-オキソ-1,6-ジアザビシクロ[3.2.1.]オクト-2-イル]カルバモイル}アミノ)ピペリジン-1-カルボン酸ベンジル224mgを取得した(収率85%)。尚、反応中に結晶は析出せず、反応液はクリアーな溶液であった。
4-[({(2S,5R)-5-[(ベンジロキシ)アミノ]ピペリジン-2-イル}カルボニル)アミノ]ピペリジン-1-カルボン酸ベンジル250mgにテトラヒドロフラン2.53gとトリブチルアミン317.9mgを入れて均一溶液とし、A液とした。また、トリホスゲン127.2mgにテトラヒドロフラン2.97gを入れて均一溶液とし、B液とした。T字ミキサー(内径:2mm、材質:ポリテトラフルオロエチレン(PTFE))及び滞留ライン中(チューブ内径:2mm、材質:ポリテトラフルオロエチレン(PTFE))を25℃の恒温バスに入れた後、A液及びB液をそれぞれ2ml/分の速度にてシリンジポンプ(YMC社製)で送液・T字ミキサー内で混合し、滞留ライン内で8分間通流して反応させた(アミン/溶媒=5.8/100(重量比))。反応液はフラスコに入った13%リン酸水1.56gとトルエン4.00gの混液中に撹拌下、連続的にクエンチすることで、4-({[(2S,5R)-6-(ベンジロキシ)-7-オキソ-1,6-ジアザビシクロ[3.2.1.]オクト-2-イル]カルバモイル}アミノ)ピペリジン-1-カルボン酸ベンジル190mgを取得した(収率72%)。尚、反応中に結晶は析出せず、反応液はクリアーな溶液であった。
以下の実施例15では、(S)-1-フェニル-1,2,3,4-テトラヒドロイソキノリンとトリホスゲンから塩化(S)-1-フェニル-1,2,3,4-テトラヒドロイソキノリンカルボニルを製造した。HPLC法によって生成物を定量し、収率を算出した。HPLC条件は以下の通りである。
移動相:ヘキサン/イソプロピルアルコール=98/2 流速:0.7ml/分

(S)-1-フェニル-1,2,3,4-テトラヒドロイソキノリン2.00gにトルエン10.25gとトリブチルアミン5.31gを入れて均一溶液とし、A液とした。また、トリホスゲン2.27gにトルエン16.55gを入れて均一溶液とし、B液とした。T字ミキサー(内径:2mm、材質:ポリテトラフルオロエチレン(PTFE))及び滞留ライン中(チューブ内径:2mm、材質:ポリテトラフルオロエチレン(PTFE))は10℃の恒温バス内に入れた後、A液及びB液をそれぞれ2.0ml/分の速度にてシリンジポンプ(YMC社製)で送液・ミキサー内で混合し、滞留ライン内で2分間通流して反応させた(アミン/溶媒=19.8/100(重量比))。反応液はフラスコに入った2N塩酸水40g中に撹拌下、連続的にクエンチすることで、分液後、塩化(S)-1-フェニル-1,2,3,4-テトラヒドロイソキノリンカルボニルを1.64g含有する有機層を38.04g取得した(収率63%)。尚、反応中に結晶は析出せず、反応液はクリアーな溶液であった。
Claims (12)
- 水酸基、チオール基、アミノ基、カルボキシル基、チオカルボキシル基、及び酸アミド基からなる群より選ばれる、塩素と反応し得る官能基を少なくとも1つ有する反応基質と塩素含有化合物とを、炭素数9~40のトリアルキルアミンと有機溶媒と共にフロー式リアクターに供給し、前記反応基質と塩素含有化合物とを反応させる有機化合物の製造方法。
- 前記フロー式リアクターが2以上の原料供給口と、供給された原料を混合する混合部と、混合液を流通するリアクター部とを有しており、
前記反応基質と前記トリアルキルアミンとを溶解した有機溶媒溶液と、前記塩素含有化合物を溶解した有機溶媒溶液とを別々に前記原料供給口から前記リアクター部に供給する請求項1に記載の製造方法。 - 前記有機溶媒が、脂肪族炭化水素系溶媒、芳香族炭化水素系溶媒、エーテル系溶媒、エステル系溶媒、ケトン系溶媒、ニトリル系溶媒、及びアミド系溶媒から選ばれる少なくとも一種である、請求項1又は2に記載の製造方法。
- 前記トリアルキルアミンの量が、溶媒100重量部に対して、3重量部以上である請求項1~3のいずれかに記載の製造方法。
- 前記トリアルキルアミンがトリプロピルアミン、トリブチルアミン、トリヘキシルアミン、又はトリオクチルアミンである、請求項1~4のいずれかに記載の製造方法。
- 前記反応基質が下記式(a1)~(a3);
(式中、R4は、反応基質のうち前記官能基以外の部分を表す。Xは、O、S、又はNR5であり、ここで前記R5は、水素原子、置換基を有していてもよい炭素数1~20のアルキル基、置換基を有していてもよい炭素数2~20のアルケニル基、置換基を有していてもよい炭素数3~20のシクロアルキル基、置換基を有していてもよい炭素数7~20のアラルキル基、置換基を有していてもよい炭素数6~20のアリール基、置換基を有していてもよい炭素数3~20のヘテロアリール基、置換基を有していてもよい炭素数1~20のアルコキシ基、置換基を有していてもよい炭素数7~20のアラルキルオキシ基、又は置換基を有していてもよい炭素数6~20のアリールオキシ基を表し、前記R4とR5は互いに結合していてもよい。Xが複数ある場合、それらは同一でもよく、異なっていてもよい。n、mは、それぞれ独立して1~3の整数を表す。)で表される化合物から選ばれる少なくとも1種であり、
前記塩素含有化合物が下記式(1)~(8);
(式中、R1、R2、R3は、同一又は異なって、置換基を有していてもよい炭素数1~20のアルキル基、置換基を有していてもよい炭素数2~20のアルケニル基、置換基を有していてもよい炭素数3~20のシクロアルキル基、置換基を有していてもよい炭素数7~20のアラルキル基、置換基を有していてもよい炭素数6~20のアリール基、又は置換基を有していてもよい炭素数3~20のヘテロアリール基を表す。尚、R1とR2、R1とR3及びR2とR3は結合していてもよい。)で表される化合物及びそれらの合成等価体から選ばれる少なくとも1種であり、これらを反応させて得られる前記有機化合物が下記式(9)~(34);
- 前記R4が、置換基を有していてもよい炭素数1~20のアルキル基、又はこのアルキル基から水素原子が1~2個除かれた2~3価の基、置換基を有していてもよい炭素数2~20のアルケニル基、又はこのアルケニル基から水素原子が1~2個除かれた2~3価の基、置換基を有していてもよい炭素数3~20のシクロアルキル基、又はこのシクロアルキル基から水素原子が1~2個除かれた2~3価の基、置換基を有していてもよい炭素数7~20のアラルキル基、又はこのアラルキル基から水素原子が1~2個除かれた2~3価の基、置換基を有していてもよい炭素数6~20のアリール基、又はこのアリール基から水素原子が1~2個除かれた2~3価の基、或いは置換基を有していてもよい炭素数3~20のヘテロアリール基、又はこのヘテロアリール基から水素原子が1~2個除かれた2~3価の基を表し、前記Xが、O、S、又はNR5であり、前記R5が、水素原子、置換基を有していてもよい炭素数1~20のアルキル基、置換基を有していてもよい炭素数2~20のアルケニル基、置換基を有していてもよい炭素数3~20のシクロアルキル基、置換基を有していてもよい炭素数7~20のアラルキル基、置換基を有していてもよい炭素数6~20のアリール基、置換基を有していてもよい炭素数3~20のヘテロアリール基、置換基を有していてもよい炭素数1~20のアルコキシ基、置換基を有していてもよい炭素数7~20のアラルキルオキシ基、又は置換基を有していてもよい炭素数6~20のアリールオキシ基を表し、前記R4とR5は互いに結合していてもよい基である、請求項6に記載の製造方法。
- 前記反応基質が(2R,3R)-3-(2,4-ジフルオロフェニル)-3,4-エポキシ-2-ブチルアルコールであり、前記塩素含有化合物がスルホニルクロリド化合物であり、これらの反応から得られる前記有機化合物が(2R,3R)-3-(2,4-ジフルオロフェニル)-3,4-エポキシ-2-ブチルスルホネートである、請求項6に記載の製造方法。
- 前記反応基質が(S)-1-フェニル-1,2,3,4-テトラヒドロイソキノリンであり、前記塩素含有化合物がホスゲン(5)又はその合成等価体であり、これらの反応から得られる前記有機化合物が塩化(S)-1-フェニル-1,2,3,4-テトラヒドロイソキノリンカルボニルである、請求項6に記載の製造方法。
- 前記反応基質が下記式(a4)~(a6);
(式中、R6、R7、及びR8は、反応基質のうち前記官能基以外の部分を表す。Xは、O、S、又はNR5であり、ここで前記R5が、水素原子、置換基を有していてもよい炭素数1~20のアルキル基、置換基を有していてもよい炭素数2~20のアルケニル基、置換基を有していてもよい炭素数3~20のシクロアルキル基、置換基を有していてもよい炭素数7~20のアラルキル基、置換基を有していてもよい炭素数6~20のアリール基、置換基を有していてもよい炭素数3~20のヘテロアリール基、置換基を有していてもよい炭素数1~20のアルコキシ基、置換基を有していてもよい炭素数7~20のアラルキルオキシ基、又は置換基を有していてもよい炭素数6~20のアリールオキシ基を表す。Xが複数ある場合、それらは同一でもよく、異なっていてもよい)で表される化合物から選ばれる少なくとも1種であり、前記塩素含有化合物がホスゲン又はその合成等価体であり、これらを反応させて得られる前記有機化合物が下記式(35)~(37);
(式中、R6~R8及びXは前記に同じ)で表される化合物から選ばれる少なくとも1種である請求項1~5のいずれかに記載の製造方法。 - 前記R6が、置換基を有していてもよい炭素数1~20のアルキル基、置換基を有していてもよい炭素数2~20のアルケニル基、置換基を有していてもよい炭素数3~20のシクロアルキル基、置換基を有していてもよい炭素数7~20のアラルキル基、置換基を有していてもよい炭素数6~20のアリール基、又は置換基を有していてもよい炭素数3~20のヘテロアリール基を表し、前記R7及びR8は、置換基を有してもよい炭素数1~10のアルキレン基を表し、XがO、S、又はNR5であり、ここで前記R5が、水素原子、置換基を有していてもよい炭素数1~20のアルキル基、置換基を有していてもよい炭素数2~20のアルケニル基、置換基を有していてもよい炭素数3~20のシクロアルキル基、置換基を有していてもよい炭素数7~20のアラルキル基、置換基を有していてもよい炭素数6~20のアリール基、置換基を有していてもよい炭素数3~20のヘテロアリール基、置換基を有していてもよい炭素数1~20のアルコキシ基、置換基を有していてもよい炭素数7~20のアラルキルオキシ基、又は置換基を有していてもよい炭素数6~20のアリールオキシ基を表し、前記R5とR6は互いに結合していてもよい基である、請求項10に記載の製造方法。
- 前記反応基質がN-[1-(S)-エトキシカルボニル-3-フェニルプロピル]-L-アラニン、4-[({(2S,5R)-5-[(ベンジロキシ)アミノ]ピペリジン-2-イル}カルボニル)アミノ]ピペリジン-1-カルボン酸ベンジル、4-[({(2S,5R)-5-[(ベンジロキシ)アミノ]ピペリジン-2-イル}カルボニル)アミノ]ピペリジン-1-カルボン酸tert-ブチル、(2S,5R)-5-ベンジロキシアミノ-ピペリジン-2-カルボン酸ベンジル、(2S,5R)-5-ベンジロキシアミノ-ピペリジン-2-カルボン酸メチル、又は(2S,5R)-5-ベンジロキシアミノ-ピペリジン-2-カルボン酸アミドであり、前記塩素含有化合物がホスゲン又はトリホスゲンであり、これらの反応から得られる前記有機化合物がN-[1-(S)-エトキシカルボニル-3-フェニルプロピル]-L-アラニル-N-カルボン酸無水物、4-({[(2S,5R)-6-(ベンジロキシ)-7-オキソ-1,6-ジアザビシクロ[3.2.1.]オクト-2-イル]カルバモイル}アミノ)ピペリジン-1-カルボン酸ベンジル、4-({[(2S,5R)-6-(ベンジロキシ)-7-オキソ-1,6-ジアザビシクロ[3.2.1.]オクト-2-イル]カルバモイル}アミノ)ピペリジン-1-カルボン酸tert-ブチル、(4R,6S)-3-(ベンジルオキシ)-2-オキソ-1,3-ジアザビシクロ[2,2,1]ヘプタン-6-カルボン酸ベンジル、(4R,6S)-3-(ベンジルオキシ)-2-オキソ-1,3-ジアザビシクロ[2,2,1]ヘプタン-6-カルボン酸メチル、(4R,6S)-3-(ベンジルオキシ)-2-オキソ-1,3-ジアザビシクロ[2,2,1]ヘプタン-6-カルボン酸アミドである、請求項10に記載の製造方法。
