Abstract

本発明の目的は、発酵乳の生産効率を低下させずに、十分な硬さを有し、かつ食感が滑らかな発酵乳を製造することができる、発酵乳の製造方法を提供することであり、本発明は、原料乳の平均粒径を小さくするように、原料乳を均質化する工程と、原料乳を均質化する工程の前または後に、原料乳を115℃~150℃で殺菌する工程と、原料乳を均質化する工程および原料乳を殺菌する工程の後に、原料乳を発酵させる工程とを含む、発酵乳の製造方法である。

Description

発酵乳およびその製造方法
 本発明は、流通時の振動に耐えられる硬度(強度)を有し、かつ食感の滑らかさに優れた発酵乳を得るための製造方法、およびその製造方法によって得られる発酵乳に関する。
 発酵乳は、乳または乳と同程度の無脂乳固形分を含む乳等を乳酸菌または酵母で発酵させて、糊状、液状、固形状にしたもの、もしくは、これらを凍結したものであり、二つのタイプに大別できる。一つは前発酵タイプ、もう一つは後発酵タイプである。前者(前発酵タイプ)は、原料乳に所定量のスターター(乳酸菌等)を添加し、流通用の個食容器に詰める前のタンク等を用いて、この原料乳を所定の乳酸酸度や所定のpH等に到達するまで発酵させてから冷却した後に、この得られた発酵乳を破砕等して、必要に応じて、果肉や甘味料(糖液等)等を混合してから、流通用の個食容器(紙容器、プラスチック容器、ガラス容器等)に充填したものである。後者(後発酵タイプ)は、原料乳に所定量のスターターを添加し、この原料乳を流通用の個食容器に充填してから、発酵室等を用いて、この原料乳を所定の乳酸酸度や所定のpH等に到達する時間まで発酵させて、プリン状に固化させた後に冷却したものである。前発酵は、果肉入りのソフトタイプのヨーグルトや甘味料入りのドリンクタイプのヨーグルト等の製造に多く用いられる。一方、後発酵は、果肉や甘味料等を含まないハードタイプ(セットタイプ)のヨーグルト等、いわゆるプレーンタイプのヨーグルト等の製造に多く用いられる。
 発酵乳は、殺菌した原料乳にスターターを添加して発酵させることにより製造される。原料乳を超高温殺菌処理(UHT処理、たとえば120℃以上の加熱処理)して発酵させると、食感が極めて滑らかになり、嗜好性が有意に向上するものの、カードの硬度が小さい(脆い)発酵乳が得られることとなる。このため、セットタイプのヨーグルト等において、カードの硬度が大きい(頑丈な)発酵乳を得ようとする場合、一般には、原料乳を高温短時間殺菌処理(HTST処理、たとえば85~95℃の加熱処理)して発酵させている。しかし、原料乳を高温短時間殺菌処理して発酵させると、食感が十分に滑らかな発酵乳を得ることが困難である。
 高温短時間殺菌処理を用いる発酵乳の製造方法として、特許文献1には、原料乳の溶存酸素濃度を低減(5ppm以下)させてから、原料乳を高温短時間殺菌処理した後に、低温(30~37℃)で発酵させる、滑らかな食感に優れ、流通時に組織を維持できる、硬度が十分な発酵乳の製造方法が記載されている。また、特許文献2には、原料乳の溶存酸素濃度を低減(5 ppm以下)させてから、原料乳を高温短時間殺菌処理した後に、低温(30~40℃)で発酵させる、滑らかな食感に優れ、流通段階で組織を維持できる、硬度が十分な発酵乳の製造方法が記載されている。

 特許文献1および2の発酵乳の製造方法では、原料乳の溶存酸素濃度を低減させるため、発酵乳の一般的な製造設備の他に、不活性ガス(窒素(N2)等)を注入する設備、減圧する設備、脱泡する設備等の付帯の製造設備が必要となる。また、原料乳を高温短時間殺菌処理するため、原料乳を超高温殺菌処理する場合に比べて、食感が十分に滑らかな発酵乳が得られるとは言いきれない。つまり、従来よりも食感を改善して、発呼乳の食感の滑らかさを向上させることが求められる。原料乳を高温短時間殺菌処理する場合、一般的に殺菌時間が長くなってしまうため、発酵乳の生産効率が良いとは言い切れない。つまり、従来よりも殺菌時間を短くして、発酵乳の生産効率を向上させることが求められる。
 また、牛乳や乳飲料等を冷蔵で流通する場合、耐熱性の芽胞菌を殺菌して、細菌的な品質を担保するために、牛乳や乳飲料等の製造工程において、生乳や原料乳等を超高温殺菌処理することが求められる。
 つまり、発酵乳の製造工程において、生乳や原料乳等を高温短時間殺菌処理する場合、牛乳や乳飲料等の製造工程における殺菌条件と異なるため、発酵乳の製造工程と、牛乳や乳飲料等の製造工程において同じ殺菌設備を利用することになると、それぞれの製造工程において殺菌条件を切り替えながら変更しなければならない。そのため、乳業工場の全体において、各種の製品の生産効率を低下させていた。また、発酵乳の製造工程と、牛乳や乳飲料等の製造工程において異なる殺菌設備を利用することになると、それぞれの製造工程において殺菌設備を設置しなければならない。そのため、乳業工場の全体において、各種の製品の製造設備の点数を増加させていた。
 上述したように、従来、流通時に組織が維持される十分な硬度を有するとともに、食感が滑らかな発酵乳を製造するためには、原料乳の溶存酸素濃度を低減させたり、原料乳を牛乳や乳飲料の殺菌温度よりも低温で殺菌したり、原料乳にホエイタンパク質(α-ラクトアルブミン、β-ラクトグロブリン等)を添加したりしなければならなかった。そのため、発酵乳の製造費が掛かるとともに、発酵乳の生産効率の低下が避けられなかった。
 そこで、本発明の目的は、発酵乳の生産効率を低下させずに、十分な硬度を有し、かつ食感が滑らかな発酵乳を製造することができる、発酵乳の製造方法を提供することにある。
 本発明者らは、上記課題を解決するために鋭意検討を行った結果、原料乳を均質化等して、原料乳の平均粒径を小さくすることにより、原料乳を超高温殺菌処理した場合でも、十分な硬度を有する発酵乳を製造することができることを見出し、本発明を完成させた。
 本発明は、原料乳の平均粒径を小さくするように、原料乳を均質化する工程と、原料乳を均質化する工程の前または後に、原料乳を115℃~150℃で殺菌する工程と、原料乳を均質化する工程および原料乳を殺菌する工程の後に、原料乳(原料乳に乳酸菌のスターター等を添加した後の状態である、発酵乳基材の意味も含む)を発酵させる工程とを含む、発酵乳の製造方法を提供する。
 また、本発明は、上記均質化する工程において、原料乳の平均粒径が0.8μm以下になるように、原料乳を均質化する、発酵乳の製造方法を提供する。
 また、本発明は、上記製造方法において、原料乳を発酵させる工程の前に、原料乳の溶存酸素濃度を低減する工程をさらに含む、発酵乳の製造方法を提供する。
 また、本発明は、発酵乳がセットタイプのヨーグルトである、発酵乳の製造方法を提供する。
 また、本発明は、原料乳を均質化する工程と、原料乳を均質化する工程の前または後に、原料乳を115℃~150℃で殺菌する工程と、原料乳を均質化する工程および原料乳を殺菌する工程の後に、原料乳を発酵させる工程とを含む、発酵乳の硬度の向上方法を提供する。
 また、本発明は、原料乳を均質化する工程と、原料乳を均質化する工程の前または後に、原料乳を115℃~150℃で殺菌する工程と、原料乳を均質化する工程および原料乳を殺菌する工程の後に、原料乳を発酵させる工程とを含む、発酵乳の食感の滑らかさの向上方法を提供する。
 また、本発明は、原料乳を均質化する工程と、原料乳を均質化する工程の前または後に、原料乳を115℃~150℃で殺菌する工程と、原料乳を均質化する工程および原料乳を殺菌する工程の後に、原料乳を発酵させる工程とを含む、発酵乳の硬度および食感の滑らかさの向上方法を提供する。
 また、本発明は、硬度が26g以上であり、ヨーグルトナイフの侵入角度が60度未満であり、かつ撹拌後の平均粒径が43μm以下である、発酵乳を提供する。
 また、本発明は、上記発酵乳において、付着力が0.0008J/m2以上である、発酵乳を提供する。
 また、本発明は、上記発酵乳において、上記発酵乳の製造方法によって製造された、発酵乳を提供する。
 また、本発明は、上記発酵乳において、容器詰めされた発酵乳を提供する。
 また、本発明は、上記発酵乳において、セットタイプのヨーグルトである発酵乳を提供する。
 また、本発明は、原料乳の平均粒径を小さくするように、原料乳を均質化する工程と、原料乳を均質化する工程の前または後に、原料乳を115℃~150℃で殺菌する工程と、原料乳を容器に充填する工程と、原料乳を均質化する工程、原料乳を殺菌する工程および原料乳を充填する工程の後に、原料乳を発酵させる工程とを含む、発酵乳の製造方法によって製造された、セットタイプのヨーグルトを提供する。
 また、本発明は、原料乳の平均粒径を小さくするように、原料乳を均質化する工程と、原料乳を均質化する工程の前または後に、原料乳を115℃~150℃で殺菌する工程と、原料乳を発酵させて発酵乳を得る工程と、発酵乳を攪拌する(発酵乳のカードを破砕する)工程と、必要に応じて、果肉、野菜、プレパレーション、ソースおよび糖液等を混合する工程と、発酵乳を容器に充填する工程とを含む、発酵乳の製造方法によって製造された、ソフトタイプのヨーグルトを提供する。
 また、本発明は、原料乳の平均粒径を小さくするように、原料乳を均質化する工程と、原料乳を均質化する工程の前または後に、原料乳を115℃~150℃で殺菌する工程と、原料乳を発酵させて発酵乳を得る工程と、発酵乳を攪拌する(発酵乳のカードを破砕する)工程と、発酵乳を均質化する(発酵乳のカードを微細化する)工程と、必要に応じて、果肉、野菜、プレパレーション、ソースおよび糖液等を混合する工程と、発酵乳を容器に充填する工程とを含む、発酵乳の製造方法によって製造された、ドリンクタイプのヨーグルトを提供する。
 本発明の発酵乳の製造方法は、原料乳の平均粒径を小さくするように、原料乳を均質化し、かつ原料乳を115℃~150℃で殺菌することが主要な要件となっているので、発酵乳の生産効率を低下させずに、十分な硬度を有し、かつ食感が滑らかな発酵乳を製造することができる。

 本発明は、発酵乳の製造方法を提供する。
 本明細書において「発酵乳」とは、乳等を乳酸菌または酵母等で発酵させることにより得られる乳製品および加工品であり、乳等省令で定義される「発酵乳」、「乳製品乳酸菌飲料」および「乳酸菌飲料」等を含む。本発明によって製造される発酵乳は、たとえば、ヨーグルトであってもよい。本発明によって製造される発酵乳は、原料乳をタンク内等で発酵させてから容器に充填する前発酵タイプのヨーグルト、および原料乳を容器に充填してから発酵させる後発酵タイプのヨーグルトのうち、いずれであってもよい。本発明によって製造される発酵乳は、たとえば、プレーンヨーグルト、セットタイプヨーグルト(固形状発酵乳)、ソフトヨーグルト(糊状発酵乳)およびドリンクヨーグルト(液状発酵乳)等であってもよい。本発明の製造方法は、後述するように、十分な硬さを有する発酵乳を製造することができるため、セットタイプヨーグルトに好適に利用することができる。
 本発明の製造方法は、原料乳の平均粒径を小さくするように、原料乳を均質化する工程を含む。
 本明細書において「原料乳」とは、乳、乳成分または乳成分を含む組成物である。乳成分は、たとえば、生乳、牛乳、脱脂乳、全脂粉乳、脱脂粉乳、全脂濃縮乳、脱脂濃縮乳、加糖練乳、加糖脱脂練乳、無糖練乳、無糖脱脂練乳、乳清(ホエイ)、ホエイパウダー、脱塩ホエイ、脱塩ホエイパウダー、ホエイタンパク質濃縮物(WPC)、ホエイタンパク質分離物(WPI)、α-ラクトアルブミン、β-ラクトグロブリン、乳タンパク質濃縮物(MPC)、カゼイン、ナトリウムカゼイネート、カルシウムカゼイネート、クリーム、発酵クリーム、コンパウンドクリーム、クリームパウダー、バター、発酵バター、バターミルク、バターミルクパウダーおよびバターオイル等を含む。原料乳は、乳成分を2種以上で含んでもよい。原料乳は、さらに、乳成分の他に、たとえば、水、脂質、タンパク質、糖類、香味成分、香料、色素、ミネラル(塩類)、ビタミンおよびその他の食品用添加物等を含んでもよい。原料乳は、また、予め加温して溶解したゼラチン液等を含んでもよい。原料乳は、たとえば、公知のヨーグルトミックスでもよい。
 本明細書において「原料乳の平均粒径」とは、原料乳に含まれるタンパク質および/または脂質(脂肪)によって構成される粒子の粒径の平均値である。タンパク質および/または脂肪によって構成される粒子は、たとえば、カゼインミセルおよび/または脂肪球等である。原料乳の平均粒径は、レーザー回折式の粒度分布測定装置(たとえば、SALD-2200、島津製作所)によって評価されてもよい。
 本明細書において「原料乳を均質化する」とは、原料乳に含まれるタンパク質および/または脂質によって構成される粒子を細かく粉砕(微細化)することをいう。本発明の原料乳を均質化する方法には、たとえば、原料乳を加圧して押し出しながら、狭い間隙を通過させる方法や、原料乳を減圧して吸引しながら、狭い間隙を通過させる方法を用いることができる。原料乳を均質化する圧力および流量(流速)は、原料乳の平均粒径を0.8μm以下、好ましくは0.77μm以下、より好ましくは0.75μm以下、さらに好ましくは0.73μm以下、最も好しくは0.7μm以下に調整することができる圧力および流量であればよく、適宜設定することができる。このとき、原料乳を均質化する圧力は、好ましくは180kg/cm2以上であり、より好ましくは200kg/cm2以上であり、さらに好ましくは220kg/cm2以上であり、さらに好ましくは240kg/cm2以上であり、さらに好ましくは260kg/cm2以上であり、さらに好ましくは280kg/cm2以上であり、さらに好ましくは300kg/cm2以上である。そして、原料乳を均質化する圧力は、好ましくは800kg/cm2以下であり、より好ましくは700kg/cm2以下であり、さらに好ましくは600kg/cm2以下であり、さらに好ましくは550kg/cm2以下である。また、原料乳を均質化する流量は、好ましくは100~30000kg/hであり、より好ましくは150~25000kg/hであり、さらに好ましくは200~20000kg/hであり、さらに好ましくは250~15000kg/hである。なお、原料乳を均質化する方法および設備には、上述した方法に限らず、任意の公知の方法および設備を用いることができる。
 原料乳を均質化する工程は、原料乳を均質化する工程の直後から、少なくとも原料乳を発酵させる工程の直前までに、原料乳の平均粒径が0.8μm以下を達成するように行われてもよい。すなわち、原料乳を均質化する工程は、たとえば、原料乳を均質化する工程の直後、原料乳を殺菌する工程の前、原料乳を殺菌する工程の後、または原料乳を発酵させる工程の直前に、原料乳の平均粒径が0.8μm以下を達成するように行われてもよい。ここで、原料乳を均質化する工程は、原料乳を殺菌する工程の前、原料乳を殺菌する工程の後、または原料乳を発酵させる工程の直前に行われてもよいが、本発明の製造方法を簡略化する(原料乳を衛生的に取扱いやすい等)等の観点から、原料乳を均質化する工程は、好ましくは、原料乳を殺菌する工程の前(直前)に行われる。原料乳を均質化する工程は、1回で行われてもよいし、2回以上で行われてもよいが、本発明の製造方法を簡略化する等の観点から、原料乳を均質化する工程は、好ましくは1回で行われる。本発明の製造方法では、原料乳を所定条件で均質化等して、原料乳の平均粒径を小さく調整することで、原料乳を高温の115℃~150℃で処理した場合でも、流通時の振動に耐えられる十分な硬度の発酵乳を得ることができる。このとき、本発明の原料乳の平均粒径は、好ましくは0.8μm以下であり、より好ましくは0.77μm以下であり、さらに好ましくは0.75μm以下であり、さらに好ましくは0.73μm以下であり、さらに好ましくは0.7μm以下であり、さらに好ましくは0.67μm以下であり、さらに好ましくは0.65μm以下である。そして、本発明の原料乳の平均粒径は、好ましくは0.2μm以上であり、より好ましくは0.25μm以上であり、さらに好ましくは0.3μm以上であり、さらに好ましくは0.35μm以上であり、さらに好ましくは0.4μm以上である。また、本発明の原料乳の粒径の標準偏差は、好ましくは0.16μm以下であり、より好ましくは0.15μm以下であり、さらに好ましくは0.14μm以下であり、さらに好ましくは0.13μm以下である。そして、本発明の原料乳の粒径の標準偏差は、好ましくは0.01μm以上であり、より好ましくは0.05μm以上であり、さらに好ましくは0.08μm以上であり、さらに好ましくは0.1μm以上である。
 本発明の製造方法は、原料乳を115℃~150℃で殺菌する工程を含む。
 原料乳を殺菌する工程は、原料乳を均質化する工程の前に行われてもよいし、原料乳を均質化する工程の後に行われてもよい。原料乳を殺菌する方法では、いわゆる「高温殺菌処理(HTST)」または「超高温殺菌処理(UHT)」を用いることができる。原料乳を殺菌する温度および時間は、十分な硬度を有し、かつ食感が滑らかな発酵乳を製造することができる温度および時間であればよく、適宜設定することができる。このとき、原料乳を殺菌する温度は、好ましくは115℃以上であり、より好ましくは116℃以上であり、さらに好ましくは120℃以上であり、さらに好ましくは125℃以上であり、さらに好ましくは130℃以上である。そして、原料乳を殺菌する温度は、好ましくは150℃以下であり、より好ましくは145℃以下であり、さらに好ましくは、140℃以下である。また、原料乳を殺菌する時間は、好ましくは1~300秒間であり、より好ましくは1~120秒間であり、さらに好ましくは1~60秒間であり、さらに好ましくは1~30秒間であり、さらに好ましくは1~10秒間であり、最も好ましくは1~5秒間である。なお、原料乳を殺菌する方法および設備には、上述した方法に限らず、任意の公知の方法および設備を用いることができる。なお、原料乳を高温の115℃~150℃で処理することで、食感の滑らかさを向上させた発酵乳を得ることができる。
 本発明の製造方法は、原料乳(原料乳に乳酸菌のスターター等を添加した後の状態である、発酵乳基材の意味も含む)を発酵させる工程を含む。
 原料乳を発酵させる工程は、原料乳を均質化する工程および原料乳を殺菌する工程の後に行われる。原料乳を発酵させる工程では、原料乳を均質化してから、原料乳を殺菌した後に、あるいは、原料乳を殺菌してから、原料乳を均質化した後に、原料乳に乳酸菌や酵母のスターター等を添加(接種)して、原料乳を発酵させることができる。また、原料乳を発酵させる工程では、原料乳を殺菌した後に、原料乳に乳酸菌や酵母のスターター等を添加してから、原料乳を均質化して、原料乳を発酵させることができる。すなわち、原料乳にスターターを添加する工程は、原料乳を殺菌する工程より後であって、原料乳を発酵させる工程より前であれば、原料乳を均質化する工程の前および原料乳を均質化する工程の後のうち、いずれの段階で行われてもよいが、本発明の製造方法を簡略化する(原料乳を衛生的に取扱いやすい等)等の観点から、原料乳にスターターを添加する工程は、好ましくは、原料乳を発酵させる工程の前(直前)に行われる。原料乳にスターターを添加する工程は、1回で行われてもよいし、2回以上で行われてもよいが、本発明の製造方法を簡略化する等の観点から、原料乳にスターターを添加する工程は、好ましくは1回で行われる。
 原料乳を発酵させる工程、あるいは原料乳にスターターを添加する工程では、任意の乳酸菌、ビフィズス菌および酵母等のスターター等を用いることができる。スターターには、たとえば、ラクトバチルス・ブルガリカス(ブルガリア菌、L.bulgaricus)、ストレプトコッカス・サーモフィルス(サーモフィラス菌、S.thermophilus)、ラクトバチルス・ラクティス(ラクティス菌、L.lactis)、ラクトバチルス・ガッセリ(ガセリ菌、L.gasseri)、ラクトバチルス・プランタラム(プランタラム菌、L.plantarum)、ラクトバチルス・カゼイ(カゼイ菌、L.casei)、ラクトバチルス・アシドフィラス(L.acidophilus)およびビフィドバクテリウム(Bifidobacterium)等のように、発酵乳の製造において一般的に用いられる乳酸菌や酵母から選択した1種を単独で用いることもできるし、または2種以上を組合せて用いることもできる。なお、スターターには、コーデックス規格において、ヨーグルトスターターとして規格化されている等の観点から、好ましくは、ブルガリア菌とサーモフィラス菌の混合スターターをベースとするスターターである。また、実際に得ようとする発酵乳に応じて、ヨーグルトスターターをベースとしながら、ガセリ菌、ビフィズス菌および酵母等を添加してもよい。このとき、スターターの添加量は、発酵乳の製造において一般的に用いられる数量であればよく、原料乳に対して、たとえば0.1~10重量%、好ましくは0.2~5重量%、より好ましくは0.5~4重量%、さらに好ましくは1~3重量%である。また、スターターの添加方法は、発酵乳の製造において一般に用いられる方法であればよく、たとえば、原料乳がタンク内等に溜められた状態で、スターターが無菌的に添加する方法や、原料乳が配管内を流れている状態で、スターターがインラインで添加される方法である。なお、原料乳を発酵させる方法および設備には、上述した方法に限らず、任意の公知の方法および設備を用いることができる。
 原料乳を発酵させる方法は、原料乳にスターターを添加した後に、原料乳を容器に充填してから、発酵室内で保持する方法であってもよいし、原料乳にスターターを添加した後に、原料乳をタンク等に充填してから、タンク内等で保持する方法であってもよい。そして、原料乳をタンク等に充填してから、タンク内等で保持する方法では、発酵乳がタンク内等で調製されてから、この発酵乳を撹拌した(発酵乳のカードを破砕した)後に、必要に応じて、果肉、野菜、プレパレーション、ソースおよび糖液等を混合し、この発酵乳を容器に充填する方法を追加して、ソフトタイプのヨーグルトを製造してもよい。また、原料乳をタンク等に充填してから、タンク内等で保持する方法では、発酵乳がタンク内等で調製されてから、この発酵乳を撹拌し(発酵乳のカードを破砕し)、発酵乳を均質化した(発酵乳のカードを微細化した)後に、必要に応じて、果肉、野菜、プレパレーション、ソースおよび糖液等を混合し、この発酵乳を容器に充填する方法を追加して、ドリンクタイプのヨーグルトを製造してもよい。
 原料乳を発酵させる条件は、原料乳に添加される乳酸菌等の種類および添加量や、実際に得ようとする発酵乳の風味、食感および物性等を考慮して調整されればよい。原料乳を発酵させる温度および時間は、十分な硬度を有し、かつ食感が滑らかな発酵乳を製造することができる温度および時間であればよく、適宜設定することができる。このとき、原料乳を発酵させる温度(発酵温度)は、好ましくは30℃以上であり、より好ましくは33℃以上であり、さらに好ましくは35℃以上であり、最も好ましくは37℃以上である。そして、原料乳を発酵させる温度は、好ましくは50℃以下であり、より好ましくは45℃以下であり、さらに好ましくは43℃以下である。また、原料乳を発酵させる時間(発酵時間)は、好ましくは1~24時間であり、より好ましくは1~12時間であり、さらに好ましくは2~8時間であり、さらに好ましくは2~6時間であり、さらに好ましくは2~4時間である。また、原料乳を発酵させる際に、乳酸酸度(酸度)は、組成によって異なるが、無脂乳固形分が8重量%程度であれば、好ましくは0.5%に到達し、より好ましくは0.6%に到達する。また、乳酸酸度(酸度)は、無脂乳固形分が10重量%程度であれば、好ましくは0.6%に到達し、より好ましくは0.7%に到達する。また、乳酸酸度(酸度)は、無脂乳固形分が12重量%程度であれば、好ましくは0.7%に到達し、より好ましくは0.8%に到達する。また、乳酸酸度(酸度)は、無脂乳固形分が14重量%程度であれば、好ましくは0.8%に到達し、より好ましくは1.0%に到達する。また、乳酸酸度(酸度)は、無脂乳固形分が16重量%程度であれば、好ましくは1.0%に到達し、より好ましくは1.1%に到達する。また、乳酸酸度(酸度)は、無脂乳固形分が18重量%程度であれば、好ましくは1.1%に到達し、より好ましくは1.3%に到達する。また、乳酸酸度(酸度)は、無脂乳固形分が20重量%程度であれば、好ましくは1.2%に到達し、より好ましくは1.4%に到達する。なお、原料乳を発酵させる方法および設備には、上述した方法に限らず、任意の公知の方法および設備を用いることができる。なお、原料乳を低温の35℃~40℃で発酵処理することで、食感の滑らかさをより向上させた発酵乳を得ることができる。
 本発明の製造方法では、後述した実施例の通り、具体的には、カードの組織の緻密さ、後味のまろやかさ、風味のクリーミー感、濃厚感、後味のミルク感、滑らかさおよび食べごたえに優れており、風味が良好であり、かつ食感の滑らかさに優れた発酵乳を得ることができる。
 本発明の製造方法は、原料乳(原料乳に乳酸菌のスターター等を添加した後の状態である、発酵乳基材の意味も含む)を発酵させる工程より前に、原料乳の溶存酸素濃度を低減する工程をさらに含んでもよい。
 原料乳の溶存酸素濃度を低減する工程は、原料乳を発酵させる工程より前に行えばよく、たとえば、原料乳を均質化する工程の前、原料乳を均質化する工程の後、原料乳を殺菌する工程の前、原料乳を殺菌する工程の後、原料乳にスターターを添加する工程の前、および原料乳にスターターを添加する工程の後のうち、いずれに行ってもよい。原料乳の溶存酸素濃度を低減する工程は、1回で行われてもよいし、2回以上で行われてもよいが、本発明の製造方法を簡略化する等の観点から、原料乳の溶存酸素濃度を低減する工程は、好ましくは、原料乳を殺菌する工程の前、および原料乳を発酵させる工程の前のうち、いずれかの1回または2回で行われる。このとき、原料乳の溶存酸素濃度を低減する工程は、発酵開始時における原料乳の溶存酸素濃度が通常よりも低くなるように、たとえば5ppm以下、好ましくは4ppm以下、より好ましくは3ppm以下、さらに好ましくは2ppm以下、さらに好ましくは1ppm以下となるように処理すればよい。発酵開始時における原料乳の溶存酸素濃度を低減することで、乳酸酸度が所定の数値に早く到達するため、発酵時間を短縮することができ、生産効率を向上させることができる。
 原料乳の溶存酸素濃度を低減する方法は、原料乳に不活性ガスを注入して、原料乳の酸素と不活性ガスを置換する方法であってもよいし、原料乳を低圧または真空の状態に保持して、原料乳を減圧して脱気し、原料乳の酸素を除去する方法であってもよい。原料乳の溶存酸素濃度を低減する方法および設備には、上述した方法に限らず、公知の方法および設備を用いることができる。なお、不活性ガスには、たとえば、N2を用いてもよいし、ヘリウム、ネオン、アルゴンおよびキセノン等の希ガスを用いてもよい。このとき、原料乳の溶存酸素濃度を低減する方法は、原料乳を加熱(殺菌)する方法において、加熱温度における保持時間を所定値に設定する(幾らか長くする)方法であってもよく、たとえば、原料乳の加熱温度が115℃~150℃の場合、この保持時間を好ましくは5~60秒間、より好ましくは5~30秒間、さらに好ましくは5~10秒間に設定すればよい。このとき、原料乳の溶存酸素濃度を低減する工程は、原料乳を殺菌する工程と同時に行うことができる。
 本発明の製造方法は、原料乳(原料乳に乳酸菌等のスターター等を添加した後の状態である、発酵乳基材の意味を含む)を発酵させる工程より前に、原料乳を容器に充填する工程をさらに含んでもよい。
 原料乳を容器に充填する工程は、原料乳を発酵させる工程より前に行えばよく、たとえば、原料乳を均質化する工程の後、原料乳を殺菌する工程の後、原料乳にスターターを添加する工程の前、および原料乳にスターターを添加する工程の後のうち、いずれに行ってもよい。原料乳を容器に充填する工程は、たとえば、原料乳を均質化および殺菌してから、原料乳にスターターを添加した後に、原料乳を容器に充填してもよい。なお、容器には、発酵乳(乳製品)の製造において一般的に用いられる容器であればよく、たとえば、プラスチック製、ガラス製および紙製等の容器であればよい。
 本発明の製造方法では、原料乳を均質化する工程において、原料乳の平均粒径を小さくし、かつ原料乳を殺菌する工程において、高温の115℃以上で処理することで、流通時の振動に耐えられる十分な硬さを有するだけでなく、食感の滑らかさにも優れた、発酵乳を提供することができる。また、本発明の製造方法では、原料乳を殺菌する工程において、高温の115℃以上で処理することで、牛乳や乳飲料等の発酵乳以外の製品と同じ殺菌条件を設定することができる。したがって、本発明の製造方法では、原料乳を殺菌する工程において、牛乳や乳飲料等の発酵乳以外の製品と同じ方法および設備を用いることができ、乳業工場の全体において、各種の製品の生産効率を低下させることがなく、また、各種の製品毎に設備を新たに設置する必要もない。
 また、本発明は、十分な硬度を有し、かつ食感の滑らかさに優れた、発酵乳を提供する。本発明の発酵乳は、硬度が26g以上であってもよい。また、本発明の発酵乳は、ヨーグルトナイフの侵入角度(滑らかさを示す指標の一つ)が60度未満であってもよい。また、本発明の発酵乳は、撹拌後の平均粒径(滑らかさを示す指標の一つ)が43μm以下であってもよい。また、本発明の発酵乳は、上述した発酵乳の製造方法によって製造されることができる。また、本発明の発酵乳は、上述した発酵乳の製造方法によって製造されることで、十分な硬さを有し、かつ食感の滑らかさに優れた物性に調整することができる。
 本明細書において、「十分な硬さを有する」とは、流通時の振動に耐えられ、輸送中の衝撃等で破砕されない硬度(強度)を有することをいう。本明細書において「硬度」は、強度やカードテンションと言い換えることができる。本発明の発酵乳における「硬度」は、カードメーター(たとえば、MAX ME-500、アイテクノエンジニアリング社)において、ヨーグルトナイフの破断点の測定値として評価されてもよく、具体的には、測定温度を5~10℃とし、荷重を100gとして、カードメーターの破断点によって測定して評価されてもよい。このとき、本発明の発酵乳では、硬度が26g以上であればよく、流通時に組織を安定的に維持しながら、輸送中の衝撃等で破砕されることを効果的に抑制している。つまり、本発明の発酵乳の硬度は、好ましくは26g以上であり、より好ましくは28g以上であり、さらに好ましくは30g以上であり、さらに好ましくは32g以上である。そして、本発明の発酵乳の硬度は、好ましくは100g以下であり、より好ましくは90g以下であり、さらに好ましくは80g以下であり、さらに好ましくは70g以下である。
 本明細書において「食感の滑らかさに優れた」とは、カードの組織が緻密であり、口腔内に入れたときに、舌触りがザラザラしていないことをいう。本発明の発酵乳における「食感の滑らかさ」は、カードメーター(たとえば、MAX ME-500、アイテクノエンジニアリング社)において、ヨーグルトナイフの侵入角度の測定値として評価されてもよく、具体的には、測定温度を5~10℃とし、荷重を100gとして、カードメーターの測定曲線において、原点を通る破断点に向けた接線と、破断点の後の時間-荷重曲線との角度によって測定して評価されてもよい。この角度が大きいと、ザラザラした粗い食感を有する発酵乳と評価され、この角度が小さいと、滑らかな食感を有する発酵乳と評価される。このとき、本発明の発酵乳では、ヨーグルトナイフの侵入角度が60度未満であってもよく、流通時に組織を安定的に維持しながら、舌触りの滑らかさを効果的に向上させている。つまり、本発明の発酵乳のヨーグルトナイフの侵入角度は、好ましくは60度未満であり、より好ましくは56度未満であり、さらに好ましくは52度未満であり、さらに好ましくは48度未満である。そして、本発明の発酵乳のヨーグルトナイフの侵入角度は、好ましくは10度以上であり、より好ましくは15度以上であり、さらに好ましくは20度以上であり、さらに好ましくは25度以上である。
 本発明の発酵乳における「食感の滑らかさ」は、発酵乳(最終製品、中間製品)の撹拌後の平均粒径の測定値として評価されてもよく、発酵乳の撹拌後の平均粒径は、レーザー回折式の粒度分布測定装置(たとえば、SALD-2200、島津製作所)によって測定して評価されてもよい。この撹拌後の粒子径が大きいと、ザラザラした粗い食感を有する発酵乳と評価され、この撹拌後の粒子径が小さいと、滑らかな食感を有する発酵乳と評価される。このとき、本発明の発酵乳では、撹拌後の平均粒径が43μm以下であってもよく、流通組織を安定的に維持しながら、舌触りの滑らかさを効果的に向上させている。つまり、本発明の発酵乳の攪拌後の平均粒径は、好ましくは43μm以下であり、より好ましくは42μm以下であり、さらに好ましくは41μm以下であり、最も好ましくは40μm以下である。
 本発明の発酵乳は、ヨーグルトであってもよく、前発酵タイプのヨーグルトおよび後発酵タイプのヨーグルトのうち、いずれであってもよいが、好ましくは、後発酵タイプのヨーグルトである。また、本発明の発酵乳は、プレーンタイプのヨーグルトであってもよく、セットタイプのヨーグルト(固形状発酵乳)、ソフトタイプのヨーグルト(糊状発酵乳)およびドリンクタイプのヨーグルト(液状発酵乳)のうち、いずれであってもよいが、好ましくはプレーンタイプのヨーグルトであって、セットタイプのヨーグルトである。
 本発明の発酵乳は、容器詰めされていてもよい。本明細書において「容器詰めされた」とは、容器(内)に充填され密封されたことをいう。容器は、発酵乳(乳製品)の製造において一般的に用いられる容器であればよく、たとえば、プラスチック製、ガラス製および紙製等の容器であればよい。本発明の発酵乳は、原料乳(原料乳に乳酸菌のスターター等を添加した後の状態である、発酵乳基材の意味も含む)を容器に充填した後に、原料乳を容器内で発酵させたもの(後発酵タイプ)であってもよいし、発酵乳を容器に充填する前に、原料乳をタンク内等で発酵させたもの(原料乳をタンク内等で発酵させた後に、発酵乳を容器に充填させたもの)(前発酵タイプ)でもよい。
 本発明では、原料乳および/または発酵乳(最終製品および/または中間製品)の無脂乳固形分が好ましくは8重量%以上であり、より好ましくは8.2重量%以上であり、さらに好ましくは8.5重量%以上であり、さらに好ましくは8.7重量%以上であり、さらに好ましくは9重量%以上である。そして、原料乳および/または発酵乳の無脂乳固形分が好ましくは30重量%以下であり、より好ましくは25重量%以下であり、さらに好ましくは20重量%以下であり、さらに好ましくは18重量%以下であり、さらに好ましくは15重量%以下であり、さらに好ましくは13重量%以下であり、さらに好ましくは10重量%以下であり、さらに好ましくは10重量%未満である。また、本発明では、原料乳および/または発酵乳の脂肪(脂質)が好ましくは0.3重量%以上であり、より好ましくは0.5重量%以上であり、さらに好ましくは1重量%以上であり、さらに好ましくは1.5重量%以上であり、さらに好ましくは2重量%以上であり、さらに好ましくは2.5重量%以上であり、さらに好ましくは3重量%以上であり、さらに好ましくは3重量%超である。そして、原料乳および/または発酵乳の脂肪(脂質)が好ましくは8重量%以下であり、より好ましくは7重量%以下であり、さらに好ましくは6重量%以下であり、さらに好ましくは5重量%以下であり、さらに好ましくは4.5重量%以下であり、さらに好ましくは4重量%以下であり、さらに好ましくは3.5重量%以下である。また、本発明では、原料乳および/または発酵乳の乳脂肪が好ましくは0.3重量%以上であり、より好ましくは0.5重量%以上であり、さらに好ましくは1重量%以上であり、さらに好ましくは1.5重量%以上であり、さらに好ましくは2重量%以上であり、さらに好ましくは2.5重量%以上であり、さらに好ましくは3重量%以上であり、さらに好ましくは3重量%超である。そして、原料乳および/または発酵乳の乳脂肪が好ましくは8重量%以下であり、より好ましくは7重量%以下であり、さらに好ましくは6重量%以下であり、さらに好ましくは5重量%以下であり、さらに好ましくは4.5重量%以下であり、さらに好ましくは4重量%以下であり、さらに好ましくは3.5重量%以下である。また、本発明では、原料乳および/または発酵乳の炭水化物が好ましくは3重量%以上であり、より好ましくは4重量%以上であり、さらに好ましくは5重量%以上であり、さらに好ましくは5重量%超である。そして、原料乳および/または発酵乳の炭水化物が好ましくは15重量%以下であり、より好ましくは13重量%以下であり、さらに好ましくは10重量%以下であり、さらに好ましくは8重量%以下であり、さらに好ましくは6重量%以下である。また、本発明では、原料乳および/または発酵乳の乳糖が好ましくは3重量%以上であり、より好ましくは4重量%以上であり、さらに好ましくは5重量%以上であり、さらに好ましくは5重量%超である。そして、原料乳および/または発酵乳の乳糖が好ましくは15重量%以下であり、より好ましくは13重量%以下であり、さらに好ましくは10重量%以下であり、さらに好ましくは8重量%以下であり、さらに好ましくは6重量%以下である。
 また、本発明の発酵乳の付着力は、0.0008J/m2以上であってもよく、好ましくは0.0009J/m2以上である。また、本発明の発酵乳の付着力は、0.0050J/m2以下であってもよく、好ましくは0.0030J/m2以下であってもよい。本明細書において「付着力」とは、生体表面を模倣した傾斜板上を流下または滑落する際の、傾斜面の単位面積あたりの消費エネルギーを表す。付着力は、後述する実施例の試験6に示したような計測装置を用いて計測することができる。本発明の発酵乳は、後述する実施例において示したように、従来の発酵乳と比較して付着力が高いため、コクのある発酵乳とすることができる。
 本発明の発酵乳は、無脂乳固形分が8~30重量%であり、かつ硬度が26g以上である発酵乳であってもよい。また、本発明の発酵乳は、無脂乳固形分が8~10重量%であり、かつ硬度が26g以上である発酵乳であってもよい。また、本発明の発酵乳は、脂肪(脂質)が0.3~8重量%であり、かつ硬度が26g以上である発酵乳であってもよい。また、脂肪(脂質)が0.1~4重量%であり、かつ硬度が26g以上である発酵乳であってもよい。また、本発明の発酵乳は、乳脂肪が0.3~8重量%であり、かつ付着力が0.0008J/m2以上0.0050J/m2以下である発酵乳であってもよい。また、本発明の発酵乳は、乳脂肪が0.5~3.8重量%であり、かつ付着力が0.0008J/m2以上0.0040J/m2以下である発酵乳であってもよい。また、本発明の発酵乳は、無脂乳固形分が8~30重量%であり、かつ付着力が0.0008J/m2以上0.0040J/m2以下である発酵乳であってもよい。また、本発明の発酵乳は、無脂乳固形分が8~10重量%であり、かつ付着力が0.0008J/m2以上0.0050J/m2以下である発酵乳であってもよい。また、本発明の発酵乳は、脂肪(脂質)が0.3~8重量%であり、かつ付着力が0.0008J/m2以上である発酵乳であってもよい。また、本発明の発酵乳は、脂肪(脂質)が0.3~3.8重量%であり、かつ付着力が0.0008J/m2以上0.0050J/m2以下である発酵乳であってもよい。また、本発明の発酵乳は、乳脂肪が0.5~3.8重量%であり、かつ付着力が0.0008J/m2以上0.0050J/m2以下である発酵乳であってもよい。また、本発明の発酵乳は、乳糖が3~15重量%であり、かつ付着力が0.0008J/m2以上0.0050J/m2以下である発酵乳であってもよい。
 また、本発明の発酵乳の原材料表示は、乳、生乳、乳製品、乳たんぱく質のいずれか1つ以上および/または糖類(砂糖、液糖、オリゴ糖)、甘味料を組合せてもよい。プレーンタイプとして、本発明の発酵乳の原材料表示は、好ましくは、乳(生乳)および乳製品または乳タンパク質のみであり、より好ましくは、乳(生乳)および乳製品のみであり、さらに好ましくは乳(生乳)のみである。すなわち、本発明の発酵乳では、原材料表示が乳(生乳)のみであっても、従来通りの輸送に耐えうる硬度(強度)を有しており、具体的には、硬度が26g以上である。
 また、本発明は、発酵乳の硬度の向上方法および発酵乳の食感の滑らかさの向上方法を提供する。本発明の方法(発酵乳の硬度の向上方法および発酵乳の食感の滑らかさの向上方法)は、原料乳を均質化する工程と、原料乳を115℃~150℃で殺菌する工程と、原料乳を発酵させる工程とを含む。本発明の方法では、本発明の製造方法について上述した各工程と同様にして、原料乳を均質化する工程、原料乳を115℃~150℃で殺菌する工程および原料乳を発酵させる工程を行うことができる。
 本発明の方法において、原料乳を均質化する工程は、原料乳を均質化する工程の直後から、少なくとも原料乳を発酵させる工程の直前までに、原料乳の平均粒径が0.8μm以下を達成するように行われてもよい。このとき、発酵乳の硬度をさらに向上させる観点や発酵乳の食感の滑らかさをさらに向上させる観点から、本発明の原料乳の平均粒径は、好ましくは0.77μm以下であり、より好ましくは0.75μm以下であり、さらに好ましくは0.73μm以下であり、最も好ましくは0.7μm以下である。
 本発明の方法は、原料乳の溶存酸素濃度を低減する工程をさらに含んでもよい。本発明の方法では、本発明の製造方法について上述した原料乳の溶存酸素濃度を低減する工程と同様にして、原料乳の溶存酸素濃度を低減する工程を行うことができる。
 本発明の方法では、原料乳を所定条件で均質化等して、原料乳の平均粒径を小さく調整することで、原料乳を高温の115℃~150℃で処理した場合でも、流通時の振動に耐えられる十分な硬度の発酵乳を得ることができる。また、原料乳を高温の115℃~150℃で処理することで、食感の滑らかさを向上させた発酵乳を得ることができる。つまり、本発明の方法では、原料乳を均質化する工程において、原料乳の平均粒径を小さくし、かつ原料乳を殺菌する工程において、高温の115℃以で処理することで、流通時の振動に耐えられる十分な硬度を有するだけでなく、食感の滑らかにも優れた、発酵乳を提供することができる。
 本発明の方法では、後述した実施例の通り、具体的には、カードの組織の緻密さ、後味のまろやかさ、風味のクリーミー感、濃厚感、後味のミルク感、なめらかさおよび食べごたえに優れており、風味が良好であり、かつ食感の滑らかさに優れた発酵乳を得ることができる。
 以下に実施例を示し、本発明の実施の形態について、さらに詳しく説明するが、本発明は、以下の実施例に限定されるものではない。
 〔平均粒径の測定方法〕
 本実施例において、レーザー回折式の粒度分布測定装置SALD-2200(島津製作所)を用いて、原料乳の平均粒径および標準偏差を測定した。具体的には、原料乳をイオン交換水で希釈し、この回折・散乱の光強度の分布の最大値が35~75%(絶対値:700~1500)になるように調整した。そして、粒度分布測定装置用のソフトウェアWingSALD IIを用いて、この光強度の分布を解析し、平均粒径±標準偏差を求めた。
 〔発酵乳の硬度の測定方法〕
 本実施例において、カードメーターMAX ME-500(アイテクノエンジニアリング社)を用いて、発酵乳の硬度(強度またはカードテンション)を測定した。具体的には、100gの重りを付けたヨーグルトナイフを発酵乳の天面に静置し、発酵乳を継続的に上昇させて、2g/秒程度で加重しながら、この加重の経過時間に合わせて、この加重の測定値を曲線で表現した。このとき、この加重の経過時間(秒)を縦軸、この加重の測定値を横軸とし、縦軸の10gと横軸の4秒を同じ距離として表現した。そして、発酵乳が破断に至った場合、発酵乳の天面からヨーグルトナイフが侵入することで、この時間-荷重曲線に変曲点(破断点)が生じ、この破断に至るまでの加重を硬度(g)の指標とした。
 〔発酵乳の滑らかさの評価方法〕
 本実施例において、ヨーグルトナイフの侵入角度および撹拌後の平均粒径を測定して、発酵乳の食感の滑らかさを評価した。ここで、上述した発酵乳の硬度の測定方法において、カードメーターの測定曲線における、原点を通り破断点に向けた接線と、破断点の後の時間-荷重曲線との角度を測定して、ヨーグルトナイフの侵入角度を評価した。また、プロペラ型の撹拌翼または薬匙を用いて、発酵乳(試料)を100回転程で撹拌し、カードを崩してから、レーザー回折式の粒度分布測定装置SALD-2200(島津製作所)を用いて、発酵乳の攪拌後の平均粒径を測定した。
 〔試験1〕
 (実施例1)
 生乳、脱脂粉乳および水を混合して、原料乳(ヨーグルトミックス;脂肪:3.1重量%、無脂乳固形分:9.70重量%)を調製し、原料乳を加温(80℃程度)して均質化(400kg/cm2)してから、超高温殺菌(UHT;130℃、2秒間)した後に冷却(約10℃)した。
 このとき、原料乳の平均粒径±標準偏差は、0.52±0.13μmであった。
 前記の原料乳を加温(43℃程度)してから、N2を注入し、原料乳の溶存酸素濃度(DO)を5ppmに低減した後に、乳酸菌のスターター(明治ブルガリアヨーグルトLB81(株式会社明治)から分離した)を3重量%で添加(接種)した。
 カップ容器(容量:100g、プラスチック製)へ充填してから、発酵室(43℃)で保管して、乳酸酸度が0.70%に到達するまで静置(約3時間)した後に、冷蔵室(10℃以下)で保管して、セットタイプのヨーグルト(実施例1)を製造した。
 (比較例1)
 生乳、脱脂粉乳および水を混合して、原料乳(ヨーグルトミックス;脂肪:3.1重量%、無脂乳固形分:9.7重量%)を調製し、原料乳を加温(80℃程度)して均質化(150kg/cm2)してから、超高温殺菌(UHT;130℃、2秒間)した後に冷却(約10℃)した。
 このとき、原料乳の平均粒径±標準偏差は、1.38±0.13μmであった。
 前記の原料乳を加温(43℃程度)してから、N2を注入し、原料乳の溶存酸素濃度(DO)を5ppmに低減した後に、乳酸菌スターター(明治ブルガリアヨーグルトLB81(株式会社明治)から分離した)を3重量%で添加(接種)した。
 カップ容器(容量:100g、プラスチック製)へ充填してから、発酵室(43℃)で保管して、乳酸酸度が0.7 % に到達するまで静置(約3時間)した後に、冷蔵室(10℃以下)で保管して、セットタイプのヨーグルト(比較例1)を製造した。
 (発酵乳の硬度)
 実施例1と比較例1のヨーグルトの硬度(カードテンション)を測定した。図1は、実施例1と比較例1のヨーグルトの硬度を示すグラフである。ここで、硬度が26g以上であれば、流通時に組織を維持できる発酵乳であると言える。
 図1に示すように、比較例1(原料乳の平均粒径:1.38±0.13μm)のヨーグルトでは、硬度が25gと、26g未満であり、硬度が不十分であって、流通時に組織を維持できないことが示された。これに対して、実施例1(原料乳の平均粒径:0.52±0.13μm)のヨーグルトでは、硬度が51gと、26g以上であり、硬度が十分であって、流通時に組織を維持できることが示された。以上の結果から、原料乳の平均粒径を十分に小さくせずに、原料乳を超高温殺菌処理した場合には、流通時の振動に耐えられる十分な硬度の発酵乳を得られないことを確認できた。これに対して、原料乳の平均粒径を小さくすると、原料乳を超高温殺菌処理した場合でも、流通時の振動に耐えられる十分な硬度の発酵乳を得られることを確認できた。
 〔試験2〕
 (実施例2)
 生乳、脱脂粉乳および水を混合して、原料乳(ヨーグルトミックス;脂肪:3.1重量%、無脂乳固形分:9.7重量%)を調製し、原料乳を加温(80℃程度)して均質化(400kg/cm2)してから、超高温殺菌(UHT;130℃、2秒間)した後に冷却(約10℃)した。
 このとき、原料乳の平均粒径±標準偏差は、0.52±0.13μmであった。
 前記の原料乳を加温(39℃程度)してから、窒素(N2)を注入し、原料乳の溶存酸素濃度(DO)を5ppmに低減した後に、乳酸菌のスターター(明治ブルガリアヨーグルトLB81(株式会社明治)から分離した)を3重量%で添加(接種)した。
 カップ容器(容量:100g、プラスチック製)へ充填してから、発酵室(43℃)で保管して、乳酸酸度が0.7%に到達するまで静置(約3時間)してから、冷蔵室(10℃以下)で保管して、セットタイプのヨーグルト(実施例2)を製造した。
 実施例2(原料乳の平均粒径:0.52±0.13μm)のヨーグルトでは、硬度が49gと、26g以上であり、流通時に組織を維持できることが示された。そして、ヨーグルトナイフの侵入角度は47度であり、攪拌後の平均粒径は27μmであり、食感の滑らかさに優れていることが示された。以上の結果から、原料乳の平均粒径を小さくして、原料乳を超高温殺菌処理した場合には、流通時の振動に耐えられる十分な硬度を有し、食感の滑らかさに優れた発酵乳を得られることを確認できた。
 (比較例2)
 生乳、脱脂粉乳および水を混合して、原料乳(ヨーグルトミックス;脂肪:3.1重量%、無脂乳固形分:9.70重量%)を調製し、原料乳を加温(80℃程度)して均質化(150kg/cm2)してから、高温短時間殺菌(HTST;95℃、達温殺菌)した後に冷却(約10℃)した。
 このとき、原料乳の平均粒径±標準偏差は、1.40±0.15μmであった。
 前記の原料乳を加温(39℃程度)してから、N2を注入し、原料乳の溶存酸素濃度(DO)を5ppmに低減した後に、乳酸菌のスターター(明治ブルガリアヨーグルトLB81(株式会社明治)から分離した)を3重量%で添加(接種)した。
 カップ容器(容量:100g、プラスチック製)へ充填してから、発酵室(43℃)で保管して、乳酸酸度が0.7%に到達するまで静置(約3時間)してから、冷蔵室(10℃以下)で保管して、セットタイプのヨーグルト(比較例2)を製造した。
 比較例2(原料乳の平均粒径:1.40±0.15μm)のヨーグルトでは、硬度が55gと、26g以上であり、流通時に組織を維持できることが示された。ただし、ヨーグルトナイフの侵入角度は91度であり、攪拌後の平均粒径は45μmであり、食感の滑らかさに劣っていることが示された。
 (官能評価)
 実施例2と比較例2のヨーグルトの官能評価を実施した。官能評価では、ヨーグルトの製造から6日後に、7段階の尺度(-3~+3)を基づいて、専門パネルの17名による二点比較法を実施した。危険率1%で有意差がある(「**」で表す)と評価した。図2は、実施例2と比較例2のヨーグルトの官能評価の結果を示す図である。
 実施例2のヨーグルト(原料乳の平均粒径:0.52±0.13μm、殺菌:UHT)では、比較例2のヨーグルト(原料乳の平均粒径:1.40±0.15μm、殺菌:HTST)に比べて、(カードの)組織の緻密さ、後味のまろやかさ、風味のクリーミー感、濃厚感、後味のミルク感、滑らかさ、食べごたえ、総合評価の各項目が有意に優れていた。つまり、これらの各項目において、実施例2と比較例2の間に統計的な有意差があることを確認できた。したがって、原料乳を均質化して、原料乳の平均粒径を小さく調整し、かつ超高温殺菌処理した場合には、組織が緻密であり、まろやかさ、風味のクリーミー感、濃厚感、後味のミルク感、滑らさおよび食べごたえに優れた発酵乳を得られることを確認できた。すなわち、原料乳を均質化して、原料乳の平均粒径を小さく調整し、かつ超高温殺菌処理することで、風味が良好であり、食感の滑らかさに優れた発酵乳を得られることを確認できた。
 〔試験3〕
 (実施例3)
 生乳、脱脂粉乳および水を混合して、原料乳(ヨーグルトミックス;脂肪:3.1重量%、無脂乳固形分:9.70重量%)を調製し、原料乳を加温(80℃程度)して均質化(400kg/cm2)してから、超高温殺菌(130℃、2秒間)した後に冷却(約10℃)した。
 このとき、原料乳の平均粒径±標準偏差は、0.52±0.13μmであった。
 前記の原料乳を加温(43℃程度)してから、N2を注入し、原料乳の溶存酸素濃度(DO)を5ppmに低減した後に、乳酸菌のスターター(明治ブルガリアヨーグルトLB81(株式会社明治)から分離した)を3重量%で添加(接種)した。
 カップ容器(容量:100g、プラスチック製)へ充填してから、発酵室(43℃)で保管して、乳酸酸度が0.7%に到達するまで静置したところ、2時間50分(約3時間)が経過し、その後、冷蔵室(10℃以下)で保管して、セットタイプのヨーグルト(実施例3)を製造した。
 (実施例4)
 生乳、脱脂粉乳および水を混合して、原料乳(ヨーグルトミックス;脂肪:3.1重量%、無脂乳固形分:9.7重量%)を調製し、原料乳を加温(80℃程度)して均質化(400kg/cm2)してから、超高温殺菌(130℃、2秒間)した後に冷却(約10℃)した。
 このとき、原料乳の平均粒径±標準偏差は、0.52±0.13μmであった。
 前記の原料乳を加温(43℃程度)してから、原料乳の溶存酸素濃度(DO)を調整せず、乳酸菌のスターター(明治ブルガリアヨーグルトLB81(株式会社明治)から分離した)を3重量%で添加(接種)した。
 カップ容器(容量:100g、プラスチック製)へ充填してから、発酵室(43℃)で保管して、乳酸酸度が0.7%に到達するまで静置したところ、3時間50分(約4時間)が経過し、その後に、冷蔵室(10℃以下)で保管して、セットタイプのヨーグルト(実施例4)を製造した。
 (発酵時間)
 実施例3と4のヨーグルトの発酵時間(乳酸酸度が0.7%に到達するまでの時間)を測定して、表1に示した。表1に示すように、実施例3(発酵前の脱酸素処理:有)のヨーグルトでは、実施例4(発酵前の脱酸素処理:無)のヨーグルトに比べて、発酵時間が大きく短縮されることが示された。以上の結果から、発酵前に脱酸素処理し、原料乳の溶存酸素濃度を低減した場合には、発酵時間が短縮され、発酵乳を製造するための所要時間を短縮できるため、発酵乳の生産効率を向上できることを確認できた。

 〔試験4〕
 (実施例5~8および比較例3)
 生乳、脱脂粉乳および水を混合して、原料乳(ヨーグルトミックス;脂肪:3.1重量%、無脂乳固形分:9.7重量%)を調製し、原料乳を加温(80℃程度)して均質化(後述した各種の圧力)してから、超高温殺菌(130℃、2秒間)した後に冷却(約10℃)した。
 このとき、原料乳を均質化する条件(圧力)を調整することで、平均粒径を様々に変更して、原料乳を調製した。そして、実施例5~8と比較例3のヨーグルトにおいて、原料乳の均質化後の平均粒径を表2に示した。
 前記の原料乳を加温(43℃程度)してから、N2を注入し、原料乳の溶存酸素濃度(DO)を5ppmに低減した後に、乳酸菌のスターター(明治ブルガリアヨーグルトLB81(株式会社明治)から分離した)を3重量%で添加(接種)した。
 カップ容器(容量:100g、プラスチック製)へ充填してから、発酵室(43℃)で保管して、乳酸酸度が0.7%に到達するまで静置(約3時間)してから、冷蔵室(10℃以下)で保管して、セットタイプのヨーグルト(実施例5~8と比較例3)を製造した。
 (発酵乳の硬度)
 実施例5~8と比較例3のヨーグルトの硬度を測定して、表2に示した。表2に示すように、比較例3のヨーグルト(原料乳の平均粒径:1.30μm)では、硬度が20gと、26g未満であり、硬度が不十分であって、流通時に組織を維持できないことが示された。これに対して、実施例5~8のヨーグルト(原料乳の平均粒径:0.40~0.73μm、0.73μm以下、原料乳の均質化圧力:250~400kg/cm2)では、硬度が34~59gと26g以上であり、硬度が十分であって、流通時に組織を維持できることが示された。以上の結果から、原料乳を均質化する圧力を150kg/cm2(150kg/cm2以下)に設定して、原料乳の平均粒径を1μm以下まで小さくせずに、原料乳を超高温殺菌処理した場合には、流通時の振動に耐えられる十分な硬度の発酵乳を得られないことを確認できた。これに対して、原料乳を均質化する圧力を250~400kg/cm2(180kg/cm2以上)に設定して、原料乳の平均粒径を1μm以下(0.8μm以下)まで小さくすると、原料乳を超高温殺菌処理した場合でも、流通時の振動に耐えられる十分な硬度の発酵乳を得られることを確認された。

 〔試験5〕
 (実施例9~12)
 生乳を加温(80℃程度)して均質化(400kg/cm2)してから、超高温殺菌(保持時間:2秒間)した後に冷却(約10℃)した。
 このとき、原料乳を殺菌する条件(温度)を調整することで、熱履歴を様々に変更して、原料乳を調製した。そして、実施例9~12のヨーグルトにおいて、原料乳の均質化後の平均粒径を表3に示した。
 上記の原料乳を加温(43℃程度)してから、窒素(N2)を注入し、原料乳の溶存酸素濃度(DO)を5ppmに低減した後に、乳酸菌のスターター(明治ブルガリアヨーグルトLB81(株式会社明治)から分離した)を3重量%で添加(接種)した。
 カップ容器(容量:100g、プラスチック製)へ充填してから、発酵室(43℃)で保管して、乳酸酸度が0.7%に到達するまで、静置(約3時間)してから、冷蔵室(10℃以下)で保管して、セットタイプのヨーグルト(実施例9~12)を製造した。
 (発酵乳の硬度)
 実施例9~12のヨーグルトの硬度を測定して、表3に示した。表3に示すように、実施例9~12のヨーグルト(原料乳の平均粒径:0.45~0.57μm(0.57μm以下)、原料乳の殺菌温度:130~144℃)では、硬度が50~60gと、26g以上であり、硬度が十分であって、流通時に組織を維持できることが示された。以上の結果から、原料乳の平均粒径を1μm以下(0.8μm以下)まで小さくすると、原料乳を殺菌する温度を130~144℃程度(115℃以上)に設定して、原料乳を超高温殺菌処理した場合でも、流通時の振動に耐えられる十分な硬度の発酵乳を得られることを確認できた。

 〔試験6〕
 (実施例13)
 生乳、脱脂粉乳および水を混合して、原料乳(ヨーグルトミックス;脂肪:3.1重量%、無脂乳固形分:9.7重量%)を調製し、原料乳を加温(80℃程度)して均質化(250kg/cm2+50kg/cm2)してから、超高温殺菌(130℃、2秒間)した後に冷却(約10℃)した。
 このとき、原料乳の平均粒径±標準偏差は、0.5±0.13μmであった。
 前記の冷却した原料乳を43℃程度に加温してから、窒素(N2)を注入し、原料乳の溶存酸素濃度(DO)を5ppmに低減した後に、乳酸菌スターター(明治ブルガリアヨーグルトLB81(株式会社明治)から分離した)を3重量%で添加(接種)した。
 カップ容器(容量:100g、プラスチック製)へ充填してから、発酵室(43℃)で保管して、乳酸酸度が0.7%に到達するまで約3時間静置してから、冷蔵室(10℃以下)で保管して、セットタイプのヨーグルト(実施例13)を製造した。
 (付着力の評価)
 喫食および嚥下の特性の計測装置(以下、喫食および嚥下計測装置)を用いて、実施例13の発酵乳と、従来のセットタイプヨーグルトである明治ブルガリアヨーグルトLB81(株式会社明治;比較例4)との風味特性(主に喫食および嚥下時の食感)の差異を評価した。
 使用した喫食および嚥下計測装置の構成を図3に示す。喫食および嚥下計測装置は、生体表面を模倣した素材(水、親水性ポリビニルアルコールおよびジメチルスルホスキドを混合して作製した)を貼付した傾斜板を備えている。この傾斜板に、所定量の試料(液体、固体、半固体およびゼリーなど)を供給すると、試料が傾斜板の上を流下または滑落することにより、喫食および嚥下の現象を模倣することができる。喫食および嚥下計測装置は、試料が傾斜板の上を流下または滑落する挙動を、複数のセンサと複数のカメラで多面的に計測して数値解析することにより、喫食および嚥下の現象を模擬的に定量化する計測装置である。
 具体的には、喫食および嚥下計測装置は、図3に示すように、傾斜面を有する傾斜板と、傾斜面上に試料を提供する供給部(高精度定量供給ピストンポンプ)と、供給部から傾斜面上へ供給された試料を検出する供給センサ(吐出確認センサ)と、傾斜面上の所定の地点を流下する試料を検出する到達センサ(上部到達確認センサおよび下部到達確認センサ)と、各センサの出力を記録するデータロガーと、傾斜面上を流下する試料を傾斜面の上方から撮像して上面画像を生成する上面カメラと、傾斜面上を流下する試料を傾斜面の側方から撮像して側面画像を生成する側面カメラと、データロガーの出力、側面画像および上面画像の少なくとも一つを使用し、傾斜面上を流下する試料の状態パラメータを演算する演算部とを備えている。
 喫食および嚥下計測装置は、傾斜板上を流下または滑落する食塊の挙動を疑似的な嚥下現象と定義し、流下または滑落時の速度、加速度、圧力、力、せん断速度、壁面せん断応力、壁面せん断力、壁面で消費されるエネルギー、動的接触角、流下面積、滑落面積、流下軌跡、滑落軌跡、試料流下時の中心の厚さ、滑落時の中心の厚さおよび付着力を求めることが可能である。ここで付着力とは、傾斜面の単位面積あたりの、傾斜面で消費されるエネルギーをいう。喫食および嚥下計測装置を用いることで、異なる物性や特性を有する食塊サンプルについて、喫食および嚥下時に発生する物理量の差異を評価することが可能となり、それによって食塊の物性が嚥下時の挙動に与える影響について、評価することが可能となる。
 各物理量は、以下に示す式により算出することができる。ここで、せん断速度γ、厚さδ、粘度μ、せん断応力τ、力F、面積S、長さL(センサ間の距離)、仕事量Wおよび付着力fs[J/m2]として表す。なお、粘度は、たとえば、レオメータ(動的粘弾性測定装置)で測定する。

 上述した喫食および嚥下計測装置を用いて、明治ブルガリアヨーグルトLB81(株式会社明治;比較例4)と実施例13の発酵乳の流下または滑落時の各物理量を各4回ずつ計測した。測定した各物理量の結果を表4に示す。また、図4は、比較例4と実施例13の付着力を示すグラフである。
 表4および図4に示すように、従来の発酵乳(明治ブルガリアヨーグルトLB81)と比較して、実施例13の発酵乳は、付着力が高いことが示された。実施例13の発酵乳の付着力は、0.0008[J/m2]以上であり、従来の発酵乳と比較して約1.3倍の付着力を有していた。また、本発明の発酵乳は、表示上「乳」のみで調製されたものであるが、従来のセットタイプヨーグルト(明治ブルガリアヨーグルトLB81など)と同等の硬度を維持することができることが示された。

 本発明は、ヨーグルト等の発酵乳の製造に利用することができ、特にセットタイプのヨーグルトの製造に好適に利用することができる。

Claims (11)

  1.  原料乳の平均粒径を小さくするように、前記原料乳を均質化する工程と、
     前記均質化する工程の前または後に、前記原料乳を115℃~150℃で殺菌する工程と、
     前記均質化する工程および前記殺菌する工程の後に、前記原料乳を発酵させる工程と
    を含む、発酵乳の製造方法。
  2.  前記均質化する工程において、前記原料乳の平均粒径が0.8μm以下になるように、前記原料乳を均質化する、請求項1に記載の発酵乳の製造方法。
  3.  前記発酵させる工程の前に、前記原料乳の溶存酸素濃度を低減する工程をさらに含む、請求項1または2に記載の発酵乳の製造方法。
  4.  発酵乳がセットタイプのヨーグルトである、請求項1~3のいずれか1項に記載の発酵乳の製造方法。
  5.  原料乳を均質化する工程と、
     前記均質化する工程の前または後に、前記原料乳を115℃~150℃で殺菌する工程と、
     前記均質化する工程および前記殺菌する工程の後に、前記原料乳を発酵させる工程と
    を含む、発酵乳の硬度の向上方法。
  6.  原料乳を均質化する工程と、
     前記均質化する工程の前または後に、前記原料乳を115℃~150℃で殺菌する工程と、
     前記均質化する工程および前記殺菌する工程の後に、前記原料乳を発酵させる工程と
    を含む、発酵乳の食感の滑らかさの向上方法。
  7.  硬度が26g以上であり、ヨーグルトナイフの侵入角度が60度未満であり、かつ撹拌後の平均粒径が43μm以下である、発酵乳。
  8.  付着力が0.0008J/m2以上である、請求項7に記載の発酵乳。
  9.  請求項1~4のいずれか1項に記載の製造方法によって製造された、請求項7または8に記載の発酵乳。
  10.  容器詰めされた、請求項7~9のいずれか1項に記載の発酵乳。
  11.  発酵乳がセットタイプのヨーグルトである、請求項7~10のいずれか1項に記載の発酵乳。
