本発明は、ラミニン、または、ヘテロ3量体を形成しているラミニンフラグメントのα鎖のN末端、β鎖のN末端およびγ鎖のN末端の少なくとも1か所にコラーゲン結合性分子が結合している改変ラミニンを提供する。
ラミニンは、α鎖、β鎖およびγ鎖の3本のサブユニット鎖からなるヘテロ3量体分子である。α鎖はα1~α5の5種類、β鎖はβ1~β3の3種類、γ鎖はγ1~γ3の3種類が知られており、それらの組み合わせで少なくとも12種類以上のアイソフォームが存在する(表1参照)。本発明の改変ラミニンを構成するラミニンは、いずれのアイソフォームであってもよい。すなわち、本発明の改変ラミニンを構成するラミニンまたはヘテロ3量体を形成しているラミニンフラグメントは、α1~α5から選択される1種のα鎖もしくはそのフラグメント、β1~β3から選択される1種のβ鎖もしくはそのフラグメント、およびγ1~γ3から選択される1種のγ鎖もしくはそのフラグメントからなるものであればよい。具体的には、表1に記載の12種類、およびこれら以外のすべてのアイソフォームおよびそのフラグメントを好適に用いることができる。好ましくはラミニンα5β1γ1もしくはそのフラグメント、ラミニンα3β3γ2もしくはそのフラグメント、ラミニンα1β1γ1もしくはそのフラグメント、ラミニンα1β2γ1もしくはそのフラグメント、ラミニンα2β1γ1もしくはそのフラグメント、ラミニンα2β2γ1もしくはそのフラグメント、ラミニンα3β1γ1もしくはそのフラグメント、ラミニンα3β2γ1もしくはそのフラグメント、ラミニンα4β1γ1もしくはそのフラグメント、ラミニンα4β2γ1もしくはそのフラグメント、ラミニンα5β2γ1もしくはそのフラグメントであり、より好ましくはラミニンα3β3γ2もしくはそのフラグメント、ラミニンα5β1γ1もしくはそのフラグメントである。

本発明の改変ラミニンは、コラーゲン結合性分子が結合していない鎖のN末端およびα鎖のC末端にコラーゲン結合性分子以外の分子が結合していてもよい。コラーゲン結合性分子以外の分子としては、例えば、細胞接着分子や増殖因子結合分子等の細胞増殖制御分子が挙げられる(特許文献2参照)。
本発明は、上記本発明の改変ラミニン、ならびにコラーゲンおよび/またはゼラチンを含む細胞外マトリックス材料を提供する。本発明の細胞外マトリックス材料は、改変ラミニンおよびコラーゲンからなるものでもよく、改変ラミニンおよびゼラチンからなるものでもよく、改変ラミニン、コラーゲンおよびゼラチンからなるものでもよく、これら以外の成分を含むものでもよい。これら以外の成分としては、細胞培養に使用できる成分であれば特に限定されないが、例えば、コラーゲンおよびゼラチン以外の細胞外マトリックス成分が好ましい。コラーゲンおよびゼラチン以外の細胞外マトリックス成分としては、例えば、フィブロネクチン、マトリゲル、プロテオグリカン、ヒアルロン酸、テネイシン、エラスチン、ラミニン、フィブリノーゲン(フィブリン)等が挙げられる。
本発明は、上記本発明の改変ラミニン、ならびにコラーゲンおよび/またはゼラチンがコーティングされている培養基材を提供する。本発明の培養基材を用いて培養する細胞は特に限定されず、培養可能な細胞であればどのような細胞でもよい。好ましくは哺乳動物細胞であり、より好ましくは哺乳動物の幹細胞である。幹細胞には体性幹細胞、多能性幹細胞などが含まれる。体性幹細胞としては、神経幹細胞、間葉系幹細胞、造血幹細胞、心臓幹細胞、肝臓幹細胞、小腸幹細胞などが挙げられる。多能性幹細胞としては、ES細胞(胚性幹細胞)、iPS細胞(人工多能性幹細胞)、mGS細胞(多能性生殖幹細胞)、ES細胞と体細胞との融合細胞などが挙げられる。哺乳動物としては、ヒト、マウス、ラット、ウシ、ブタ等が挙げられる。なかでもヒトが好ましい。本発明の培養基材は、従来フィーダー細胞を用いて培養している細胞を、フィーダー細胞を用いることなく培養する場合にも有用である。
本発明は、幹細胞を分化誘導して三次元組織構造体を構築するためのスキャフォールドを提供する。本発明のスキャフォールドは、上記本発明の改変ラミニン、ならびにコラーゲンおよび/またはゼラチンを含むものであればよい。本発明のスキャフォールドを構成する材料は、細胞の足場としての役割を果たすものであればどのような材料でもよく、特に限定されない。例えば、コラーゲン、ゼラチン、フィブリン、ヒアルロン酸、アルギン酸、デンプン、キチン、ペクチン酸等の天然高分子、自己組織化能を有する両親媒性ペプチド、ポリ乳酸、ポリグリコール酸、乳酸とグルコール酸との共重合体、ポリ-ε-カプロラクトン、ε-カプロラクトンと乳酸あるいはグリコール酸との共重合体、ポリクエン酸、ポリリンゴ酸、ポリ-α-シアノアクリレート、ポリ-β-ヒドロキシ酪酸、ポリトリメチレンオキサレート、ポリテトラメチレンオキサレート、ポリプロピレンカーボネート、ポリ-γ-ベンジル-L-グルタメート、ポリ-γ-メチル-L-グルタメート、ポリ-L-アラニン等の合成高分子、ハイドロキシアパタイト、リン酸三カルシウム等の無機材料などが挙げられる。スキャフォールドの形態は、ゲル状、スポンジ状、フィルム状、メッシュ状、不織布状、編織布状などの三次元構造であればいずれの形態であってもよい。なかでも密な網目構造で構成されるゲル状またはフィルム状のスキャフォールドが好ましい。
本発明は、上記本発明の改変ラミニン、ならびにコラーゲンおよび/またはゼラチンの存在下で哺乳動物細胞を培養する培養方法を提供する。本発明の改変ラミニンが結合したコラーゲンまたはゼラチンを哺乳動物細胞の足場となる細胞外マトリックスとして用いることにより、従来フィーダー細胞を用いて培養している細胞を、フィーダー細胞を用いることなく培養することが可能となる。また、コラーゲンに親和性の低い幹細胞を、コラーゲン上で培養することが可能となり、幹細胞を分化誘導して再生医療用の三次元組織構造体を効率よく構築することが可能となる。
(1)フィーダー細胞との共培養系からヒトiPS細胞を回収する
以下の方法1または方法2のいずれかの方法で、フィーダー細胞との共培養系からヒトiPS細胞を回収する。

フィーダー細胞(例えばMEF)と共培養しているヒトiPS細胞の培養ディッシュ(3~5日目)に0.25%トリプシン/DMEM-F12を添加して(例えば1ml/60mmディッシュ)、37℃で2~3分間恒温処理し、培養ディッシュをDMEM-F12で洗浄してフィーダー細胞を除去する。培養液を加えて全体細胞を物理的に剥離した細胞懸濁液をBD Falconセルストレーナー100μm(BD Falcon #352460)に通した後、ストレーナーを洗浄することでヒトiPS細胞のコロニーのみを分離し、回収する。
フィーダー細胞(例えばMEF)と共培養しているヒトiPS細胞の培養ディッシュ(3~5日目)に細胞剥離液(例えば、ES/iPS細胞用剥離液(リプロセル RCHETP002)、1mg/ml dispase/DMEM-F12、10mg/ml collagenaseIV/DMEM-F12など)を添加して(例えば1ml/60mmディッシュ)、37℃で5分間恒温処理し、ヒトiPS細胞とMEFを剥離する。細胞を15ml遠心チューブに移し、培養液を約10ml入れて細胞を懸濁した後、5分間チューブを静置してコロニーのみを沈降させる。上清を除去し、同様の操作を2回以上繰り返して、ヒトiPS細胞のコロニーのみを沈降させ、回収する。
回収したヒトiPS細胞のコロニーを単一細胞に分散させる。単一細胞に分散させる方法は特に限定されないが、例えば、トリプシン処理する方法、P-1000ピペットマン等を用いて培養液を数回フラッシングすることによりコロニーを砕く方法等が挙げられる。単一細胞に分散させた後、適切な培養液(例えばTeSR2など)に懸濁し、例えばコラーゲンおよび改変ラミニンをコーティングした培養ディッシュに播種する。培養は、用いた培養液に適合するCO2濃度条件で行い、培養液の交換は毎日行う。
拡大エリアの不足あるいはコロニー内に死細胞の出現が目立つようになるのを目安に、継代操作を行う。本発明の培養方法では、従来の方法と同様にヒトiPS細胞を適度な大きさのコロニー形態を維持した状態で播種する方法で継代培養を行ってもよく、ヒトiPS細胞を単一細胞に分散させて播種する方法で継代培養を行ってもよい。ここで、単一細胞に分散された状態は、細胞懸濁液中の全細胞が単一細胞に分散されていることを要するものではなく、単一細胞に分散された細胞以外に数個~十数個程度が接着した状態の細胞が細胞懸濁液中に含まれている状態も単一細胞に分散された状態に含まれる。
ヒトiPS細胞を培養している培養ディッシュに、TrypLE Select(商品名、Invitrogen #12563011)を添加して(例えば1ml/100mmディッシュ)、37℃で5分間恒温処理する。例えばP-1000ピペットマン等を用いて培養液を数回フラッシングすることによりヒトiPS細胞のコロニーを砕き、単一細胞に分散させる。培養液を加えて細胞を懸濁し、遠心チューブに回収する。遠心(1000×g、3分)と同培養液による洗浄操作を2度繰り返した後、新鮮な培養液に細胞を懸濁し、ヒト由来の改変ラミニンをコーティング(例えば1.0μg/cm2)した培養ディッシュに、例えば約40000細胞/cm2の播種密度で単一細胞に分散したヒトiPS細胞を播種する。培養は、用いた培養液に適合するCO2濃度条件で行い、培養液の交換は毎日行う。
ヒトiPS細胞を単一細胞に分散しない場合は、細胞の剥離にCollagenaseIV、Dispase、Accutase等の酵素を用いる。ヒトiPS細胞を培養している培養ディッシュに、10mg/ml collagenase/DMEM-F12、または2mg/ml dispase/DMEM-F12、またはAccutase(Millipore #SCR005)を添加して(例えば1ml/60mmディッシュ)、37℃で5分間恒温処理する。酵素液を除いて培養液を加え、例えばP-1000のピペットマン等を用いて培養液を数回フラッシングすることによりヒトiPS細胞が約50~約100個のコロニーを維持する程度に小さく砕く。細胞懸濁液を遠心チューブに回収し、遠心(200×g、3分)と同培養液による洗浄操作を2度繰り返した後、新鮮な培養液に細胞を懸濁し、ヒト由来の改変ラミニンをコーティング(例えば1.5μg/cm2)した培養ディッシュに2分の1~4分の1希釈量を播種する。培養は、用いた培養液に適合するCO2濃度条件で行い、培養液の交換は毎日行う。
ヒトES細胞またはヒトiPS細胞をAccutase(Millipore)で単一細胞まで分散した後、100ng/mlのアクチビンAと10ng/mlのbFGFを含む分化誘導用hESF-DIF培地(hESF-DIF培地(Cell Science & Technology Institute)に10μg/mlのヒト組換えインスリン、5μg/mlのヒトアポトランスフェリン、10μMの2-メルカプトエタノール、10μMのセレン酸ナトリウム、0.5mg/mlのウシ血清アルブミンを添加したもの)を含むマトリゲル上で2日間培養する。ここで誘導された中内胚葉細胞にFOXA2遺伝子を発現させるアデノウィルスベクターを導入し、上記のマトリゲル上で6日目まで培養して、胚体内胚葉細胞に分化させる。これにFOXA2遺伝子を発現させるアデノウィルスベクターとHNF1α遺伝子を発現させるアデノウィルスベクター導入し、30ng/mlのBMP4、20ng/mlのFGF4を添加したhepatocyte culture medium(HCM、Lonza)を含むマトリゲル上でさらに3日間培養して、肝芽細胞へ分化させる。この肝芽細胞にFOXA2遺伝子を発現させるアデノウィルスベクターとHNF1α遺伝子を発現させるアデノウィルスベクター導入し、10ng/mlのHGF、10ng/mlのFGF1、10ng/mlのFGF4、10ng/mlのFGF10を添加したHCM培地を含むマトリゲル上で3日間培養し、肝細胞方向へ分化させる。この細胞を肝細胞として成熟させるため、細胞を8.3% tryptose phosphate broth(BD)、10%FBS、10μMのhydrocortisone21-hemisuccinate、1μMインスリン、25mM NaHCO3、20ng/mlのHGF、20ng/mlのオンコスタチンMおよび10μMのデキサメタゾンを添加したL15培地(Invitrogen)中のマトリゲル上で8日間培養する。この分化誘導方法で、薬物代謝が可能な肝様細胞へ誘導できることが報告されている(参考文献:Takayama et al., J. Hepatology, 2012, 57, 628-636)。マトリゲルに代えて本発明の改変ラミニンとコラーゲンゲルとを組み合わせた三次元基質を用いることにより、ヒトに対して安全な再生医療用肝臓細胞を提供することができる。
Delta-like leucine zipper kinase(DIK)陽性のマウス胎児肝臓細胞をEHS-ラミニン上で培養した細胞(hepatic progenitor cells proliferating on laminin; HPPL)を2×105細胞/ウェルで6穴プレートに播種し、コンフルエントになるまで培養する。その後、20ng/mlのオンコスタチンMを添加したDMEM/F12培地に交換し、5日間培養する。DMEM/F12培地で6倍希釈した300μlのマトリゲルに培地を交換すると、細胞層の上にゲルが形成される。さらに5日間培養すると、多糖を産生するようになり、PAS染色陽性の肝臓細胞へと分化することが報告されている(参考文献:Tanimizu et, al., J. Cell Sci., 2004, 117, 6425-6434)。この方法において、HPPL細胞に代えてヒト由来の幹細胞を用い、EHSゲルに代えて本発明の改変ラミニンとコラーゲンゲルとを組み合わせた三次元基質を用いることにより、ヒトに対して安全な再生医療用肝臓細胞を提供することができる。
上記HPPL細胞を、1cm直径のカルチャーインサートの上面に作製した20%マトリゲルを含むI型コラーゲンゲル上で2日間培養し、その後同組成のゲルをのせて37℃で2時間ゲル化させる。そこに5ng/mlのEGFとHGFを含むDMEM/F12培地500μlを、カルチャーインサートの上面と下面に添加する。2~3日間培養すると管様の構造を形成し、胆管マーカーのサイトケラチン19を発現するようになることが報告されている(参考文献:Tanimizu et al., Mol. Biol. Cell, 2009, 20, 2486-2494)。この方法はサンドウィッチ培養法と呼ばれている。このサンドウィッチ培養法において、HPPL細胞に代えてヒト由来の幹細胞を用い、マトリゲルに代えて本発明の改変ラミニンとコラーゲンゲルとを組み合わせた三次元基質を用いることにより、ヒトに対して安全な再生医療用胆管細胞を提供することができる。
マウス小腸の陰窩からLgr5陽性細胞をcrypt culture medium(10-50ng/mlのEGF、500ng/mlのR-spondin1、100ng/mlのnogginを添加したAdvanced DMEM/F12培地)中に回収し、1μMのJagged-1ペプチド(Anaspec)を含むマトリゲル5μl中に1細胞ずつ埋め込む。そのあとに10μMのY-27632を含むcrypt culture mediumを100μl添加する。成長因子は一日おきに添加し、培地全体は4日ごとに交換して1-2週間培養すると陰窩構造を再形成することが報告されている(参考文献:Sato et al., Nature, 2009, 459, 262-265)。ヒト由来細胞を用い、マトリゲルに代えて本発明の改変ラミニンとコラーゲンゲルとを組み合わせた三次元基質を用いることにより、ヒトに対して安全な再生医療用小腸細胞の三次元培養が可能となる。
ヒトフィブロネクチンコラーゲン結合ドメイン(以下「CBD」と記す)付加ラミニン511E8組換えタンパク質は、ヒトラミニンα5鎖E8の発現ベクター、ヒトラミニンβ1鎖E8の発現ベクター、ヒトラミニンγ1鎖E8の発現ベクター、N末端にCBDを付加したヒトラミニンα5鎖E8の発現ベクター、N末端にCBDを付加したヒトラミニンβ1鎖E8の発現ベクターおよびN末端にCBDを付加したヒトラミニンγ1鎖E8の発現ベクターをそれぞれ作製し、これらを組み合わせて宿主細胞にトランスフェクションし、発現させることにより作製した。
ヒトラミニンα5E8(アクセッション番号:NP_005551(表2参照)のAla2534-Ala3327)のcDNAの5’末端に6×HisタグをコードするDNAを付加した断片、ヒトラミニンβ1E8(アクセッション番号:NP_002282(表2参照)のLeu1561-Leu1786)のcDNAの5’末端にHAタグをコードするDNAを付加した断片、ヒトラミニンγ1E8(アクセッション番号:NP_002284(表2参照)のAsn1364-Pro1609)のcDNAの5’末端にFLAGタグをコードするDNAを付加した断片をそれぞれPCRで増幅した。この断片をpSecTag2Bベクター(Invitrogen)のHindIII/EcoRVサイト(α5E8)あるいはHindIII/EcoRIサイト(β1E8、γ1E8)に挿入して、pSec-LNα5E8、pSec-LNβ1E8およびpSec-LNγ1E8を作製した(Ido H. et al., J.Biol.Chem. 2007, 282, 11144-11154)。CBD(アクセッション番号:NP_997647(表3参照)のVal276-Thr604)のcDNAの5’末端にHindIIIサイトを付加してPCRで増幅した。ヒトラミニンα5E8のcDNAの5’末端に6×HisタグをコードするDNAを付加した断片、ヒトラミニンβ1E8のcDNAの5’末端にHAタグをコードするDNAを付加した断片、ヒトラミニンγ1E8のcDNAの5’末端にFLAGタグをコードするDNAを付加した断片をそれぞれPCRで増幅し、CBDをコードするDNA断片とそれぞれをPCRで連結し、pSec-LNα5E8のHindIII/ClaIサイト(α5E8)あるいはpSecTag2Bベクター(Invitrogen)のHindIII/EcoRIサイト(β1E8、γ1E8)に挿入して、pSec-CBD-LNα5E8、pSec-CBD-LNβ1E8およびpSec-CBD-LNγ1E8を構築した。
組換えCBD付加ラミニン511E8または組換えラミニン511E8は、FreeStyleTM293Expression System(Invitrogen)を用いて作製した。293fectin(Invitrogen)を用いて、FreeStyleTM293-F細胞に3種類の発現ベクターを組み合わせてトランスフェクションし(表1参照)、無血清FreeStyleTM293発現培地で72時間培養した。馴化培地を回収し、遠心分離により浄化した。最初は馴化培地をNi-NTAアガロースを用いたアフィニティークロマトグラフィーに供した。カラムをTBSで洗浄し、結合したタンパク質を200mMイミダゾールを含むTBSで溶出した。続いてイミダゾール溶出フラクションを抗FLAG M2アガロースカラムにアプライし、結合したタンパク質を100μg/mlのFLAGペプチドを含むTBSで溶出した。溶出タンパク質をPBSに対して透析した。透析後の精製物を0.22μmのディスクシリンジフィルター(Millipore、#SLGV033RS)でろ過滅菌し、―80℃で保存した。組換えCBD付加ラミニン511E8および組換えラミニン511E8の作製に使用した発現ベクターの組み合わせを表4に示す。
精製タンパク質の濃度は、ウシ血清アルブミン(BSA)をスタンダードに用いるBCA法で定量した。精製タンパク質の純度は非還元条件でSDS-PAGEを行い、Coomassie Brilliant Blue染色して確認した。
SDS-PAGEの結果を図1に示した。いずれの組換えラミニン511E8およびCBD付加ラミニン511E8も、非還元条件ではα5鎖E8の単量体とβ1鎖E8-γ1鎖E8二量体の2つのバンドが検出され、ヘテロ三量体タンパク質として精製できていることが確認された。
(1)結合活性の測定
I型コラーゲン(新田ゼラチンtype I-A:豚由来)またはゼラチン(Sigma G1890-100G:豚由来)を0.1M NaHCO3で10μg/mlに希釈し、96穴イムノプレート(Nunc Maxisorp)に50μl/wellずつ添加して4℃で一晩コーティングした。プレート中のコーティング液を捨て、1%BSAを含むTBSを添加し、室温で2時間インキュベートしてブロッキングを行った。その後、プレートを0.1%BSAおよび0.02%Tween-20を含むTBS(以下、「wash buffer」と記す)で2回洗浄した。続いてwash buffer中に濃度を変えて希釈した各CBD付加ラミニン511E8溶液を添加し、室温で3時間振とうさせた。プレートをwash bufferで3回洗浄した後、抗ラミニンα5抗体5D6抗血清をwash bufferで3000倍希釈したものを50μl/wellずつ添加した。室温で1時間振とうさせた後、プレートをwash bufferで3回洗浄した。HRP標識抗マウスIgG抗体をwash bufferで3000倍希釈したものを50μl/wellずつ添加し、室温で1時間振とうした。プレートをwash bufferで3回洗浄し、o-phenylenediamine溶液50μl/wellを添加して発色させた。2.5M硫酸を50μl/well添加して発色を停止し、490nmの吸光度を測定した。
I型コラーゲンに対する結合活性の結果を図2に示した。CBDを付加していないラミニン511E8(図中LN511-E8)はほとんどI型コラーゲンに結合しないのに対し、CBDをβ1鎖またはγ1鎖に1つだけ付加したCBD-E8(β)とCBD-E8(γ)は顕著にI型コラーゲンに結合していた。また、CBDをβ1鎖とγ1鎖の2つに付加したCBD-E8(βγ)と、CBDをαβγの3鎖すべてに付加したCBD-E8(αβγ)は、CBDが一つだけ付加されたものにくらべてさらに低濃度でI型コラーゲンと結合し、10nMで飽和に達した。この結果から、CBDの付加数が1つ(1価)よりも2つ(2価)以上の方がI型コラーゲンに対する結合活性が、概ね1桁高いことがわかった。ただし、2価と3価の間には違いは認められなかった。なお、示していないが、CBDをα5鎖とβ1鎖の2つに付加したCBD-E8(αβ)、CBDをα5鎖とγ1鎖の2つに付加したCBD-E8(αγ)も、CBD-E8(βγ)と同等の結合活性を有していた。
(1)ヒトiPS細胞
ヒトiPS細胞は独立行政法人医薬基盤研究所生物資源バンクより購入した株(クローン名:tic(JCRB1331))を使用した。tic細胞は、独立行政法人医薬基盤研究所生物資源バンクが推奨する方法に従い、マウスフィーダー細胞との共培養で維持した。共培養ディッシュに、1U/mlディスパーゼ/DMEM-F12を添加し、tic細胞のコロニーをスクレーパーでかき集めた。このtic細胞コロニーとマウスフィーダー細胞を含む溶液をBD Falconセルストレーナー100μmに通し、ストレーナーを洗浄することでtic細胞コロニーを分離した。残留したコロニーをmTeSR1(商品名、STEMCELL TECHNOLOGIES社)培地で回収し、P-1000ピペットマンで細かく砕いた後、mTeSR1培地に再懸濁し、マトリゲルをコートした培養器に播種した。37℃、5%CO2条件下、毎日培養液交換を行い、4~5日まで拡大培養を行った。このように拡大培養された細胞を実験に供した。
I型コラーゲン(新田ゼラチンtype I-C:豚由来)をPBSで200μg/mlに希釈したもの、あるいは0.1%ゼラチン(Sigma)を1ml/wellずつ12穴プレートに添加し、37℃で1時間インキュベートした。その後、コラーゲンコート条件ではコラーゲン溶液をアスピレートして0.1%ゼラチンを1ml/wellずつ添加し、37℃で2時間インキュベートすることによりブロッキングを行った。その後ゼラチン液を除き、プレートをPBSで2回洗浄した。各ラミニンE8を8nMになるようPBSで希釈して1ml/wellずつ添加し、4℃で一晩インキュベートした。その後、プレートをPBSで3回洗浄した。
マトリゲルコートディッシュ(10cm)上で培養していた上記iPS細胞の培地をアスピレートし、4.8mM EDTA入りPBSを添加して室温で3分間インキュベートした。EDTA液を除いてTrypLE Express(Gibco)1mlを添加して37℃で1分間インキュベートし、iPS細胞を剥がした。細胞をサプリメント添加mTeSR1培地(STEMCELL TECHNOLOGIES社)で懸濁し、15mlチューブに移した後、1000rpmで3分間遠心分離した。上清をアスピレートした後、細胞をサプリメント添加mTeSR1培地で7.6×104個/mlになるように懸濁した。プレート中のPBSをアスピレートして吸引除去した後、iPS細胞を1ml/wellずつ播種した。プレートは37℃5%CO2インキュベーター中で培養した。培地交換を毎日行い、3日間培養した。
I型コラーゲンコート条件における培養3日目のiPS細胞の写真を図4に示した。図4から明らかなように、I型コラーゲンコートのみ(図中Col I only)ではほとんどiPS細胞の増殖が認められなかった。CBDを付加していないLN511-E8(図中+511E8)を添加した場合も同様に、ほとんどiPS細胞の増殖が認められなかった。一方、CBDが1価のLN511-E8(図中+CBD-E8(β)および+CBD-E8(γ))を添加した条件では、わずかにiPS細胞の増殖が認められた。CBDが2価のLN511-E8(図中+CBD-E8(βγ))を添加した条件では、CBDが1価のLN511-E8よりiPS細胞の数が多くなっている様子が観察された。
実施例1で作製した各鎖E8発現ベクター以外に、α5鎖以外のラミニンα鎖に由来するヒトラミニンE8フラグメントの発現ベクター(α1鎖E8、α2鎖E8、α3鎖E8、α4鎖E8)、ラミニンβ2鎖E8およびCBD付加ラミニンβ2鎖E8の発現ベクターをそれぞれ作製した。γ鎖については、実施例1で作製したラミニンγ1鎖E8およびCBD付加ラミニンγ1鎖E8の発現ベクターを使用した。これらの発現ベクターを組み合わせて宿主細胞にトランスフェクションすることにより、ラミニン511以外のラミニンアイソフォームに由来する組換えラミニンE8およびCBD付加ラミニンE8を作製した。
(1-1)ヒトラミニンα1鎖E8フラグメント発現ベクターの作製
クローニング用プラスミドpBluescript KS(+)(Stratagene社)を鋳型として、以下のプライマーセット(i)を用いてPCRを行い、プラスミドのマルチクローニング部位内のEcoRVの5’側に制限酵素AscI認識配列と6×HisタグをコードするDNAが挿入されたpBluescript KS(+)を作製した。
(i) 6×Hisタグ・AscIサイト導入用プライマー

次に、ヒトラミニンα1鎖のcDNA配列を含むプラスミド(Ido et al., J. Biol. Chem., 279, 10946-10954, 2004)を鋳型としてPCRを行い、α1鎖(アクセッション番号:NP_005550(表2参照)のPhe1878-Gln2700)に相当する領域を増幅した。なお、リバース(reverse)プライマーの5’側にはBamHI認識配列が付加されている。
増幅したcDNAを、AscI認識配列と6×Hisタグ配列を付加したpBluescript KS(+)のマルチクローニング部位のEcoRVとBamHI部位に挿入した。この後、5’側の6×Hisタグとα1鎖E8フラグメントをコードする配列を含むcDNAを制限酵素AscIとBamHIとで切り出し、哺乳細胞用発現ベクターpSecTag2A(Invitrogen)の当該部位に挿入し、ヒトα1鎖E8フラグメント(N末端側に6×Hisタグを含む)の発現ベクターpSec-LNα1E8を作製した。
ヒトラミニンα2鎖のcDNA配列を含むプラスミド(Ido et al., J. Biol. Chem., 283, 28149-28157, 2008)を鋳型としてPCRを行い、α2鎖(アクセッション番号:NP_000417(表2参照)のLeu1900-Ala2722)に相当する領域を増幅した。なお、リバース(reverse)プライマーの5’側にはBamHI認識配列(GGATCC)が付加されている。
増幅したcDNAを、AscI認識配列と6×Hisタグ配列を付加したpBluescript KS(+)のマルチクローニング部位のEcoRVとBamHI部位に挿入した。この後、5’側の6×Hisタグとα1鎖E8フラグメントをコードする配列を含むcDNAを制限酵素AscIとBamHIとで切り出し、哺乳細胞用発現ベクターpSecTag2A(Invitrogen)の当該部位に挿入し、ヒトα2鎖E8フラグメント(N末端側に6×Hisタグを含む)の発現ベクターpSec-LNα2E8を作製した。
ヒトラミニンα3鎖(ラミニン球状ドメインの4番目と5番目を除く)のcDNA配列を含むプラスミド(Ido et al., J. Biol. Chem., 282, 11144-11154, 2007)を鋳型としてPCRを行い、α3鎖(アクセッション番号:NP_000218(表2参照)のAla579-Ala1364)に相当する領域を増幅した。なお、リバース(reverse)プライマーの5’側にはXbaI認識配列が付加されている。
増幅したcDNAを、AscI認識配列と6×Hisタグ配列を付加したpBluescript KS(+)のマルチクローニング部位のEcoRVとXbaI部位に挿入した。この後、5’側の6×Hisタグとα3鎖E8フラグメントをコードする配列を含むcDNAを制限酵素AscIとNotIとで切り出し、哺乳細胞用発現ベクターpSecTag2A(Invitrogen)の当該部位に挿入し、ヒトα3鎖E8フラグメント(N末端側に6×Hisタグを含む)の発現ベクターpSec-LNα3E8を作製した。
5’側から、マウスIg-κ鎖V-J2-Cシグナルペプチド・6×Hisタグ・α4鎖E8フラグメントを順にコードするcDNA断片を獲得するために、マウスIg-κ鎖V-J2-Cシグナルペプチド・6×HisタグをコードするcDNA断片とα4鎖E8をコードするcDNA断片をそれぞれ取得し、エクステンションPCRによってそれら2種類の断片を連結・増幅した。
まず、ヒトラミニンα5鎖E8発現ベクター(Ido et al., J. Biol. Chem., 282, 11144-11154, 2007)を鋳型として、以下のプライマーセット(ii)を用いてPCRを行い、マウスIg-κ鎖V-J2-Cシグナルペプチド・6×Hisタグに相当する領域を増幅した。なお、リバース(reverse)プライマーの5’側にはエクステンションPCRに使用する配列が付加されている。
(ii) シグナルペプチド配列・6×Hisタグ配列増幅用プライマー

得られた2種類のcDNA断片を、エクステンションPCRにより連結・増幅させ、マウスIg-κ鎖V-J2-Cシグナルペプチド・6×Hisタグ・α4鎖E8をコードするcDNA断片を得た。増幅したcDNAを、制限酵素HindIIIとEcoRIで消化し、哺乳細胞用発現ベクターpSecTag2B(Invitrogen)の当該部位に挿入し、ヒトα4鎖E8フラグメント(N末端側に6×Hisタグを含む)の発現ベクターpSec-LNα4E8を作製した。
ヒトラミニンβ2鎖のcDNA配列を含むプラスミド(Ido et al., J. Biol. Chem., 283, 28149-28157, 2008)を鋳型としてPCRを行い、ヒトラミニンβ2E8(アクセッション番号:NP_002283(表2参照)のLeu1573-Gln1798)に相当する領域を増幅した。なお、リバース(reverse)プライマーの5’側にはEcoRI認識配列が付加されている。cDNAの5’末端にHAタグをコードするDNAを付加した断片をそれぞれPCRで増幅した。
増幅したcDNAを、HAタグ配列を付加したpBluescript KS(+)のマルチクローニング部位のEcoRVとEcoRI部位に挿入した。この後、5’側のHAタグとβ2鎖E8フラグメントをコードする配列を含むcDNAを制限酵素KpnIとEcoRIとで切り出し、哺乳細胞用発現ベクターpSecTag2B(Invitrogen)の当該部位に挿入し、ヒトβ2鎖E8フラグメント(N末端側にHAタグを含む)の発現ベクターpSec-LNβ2E8を作製した(Taniguchi Y. et al., J.Biol.Chem. 2009, 284, 7820-7831)。
CBD(アクセッション番号:NP_997647(表3参照)のVal276-Thr604)のcDNAの5’末端にHindIIIサイトを付加して断片をPCRで増幅した。ヒトラミニンβ2E8のcDNAの5’末端にHAタグをコードするDNAを付加した断片をPCRで増幅し、CBDをコードするDNA断片とPCRで連結し、pSecTag2Bベクター(Invitrogen)のHindIII/EcoRIサイトに挿入して、pSec-CBD-LNβ2E8を作製した。
pSec-CBD-LNβ2E8により発現されるタンパク質(CBD-LNβ2E8)のアミノ酸配列を配列番号11に、当該タンパク質をコードするDNA(Sec-CBD-LNβ2E8に含まれる)の塩基配列を配列番号12に示した。
組換えラミニンE8およびCBD付加ラミニンE8は、実施例1に記載の方法に従い、FreeStyleTM293Expression System(Invitrogen)を用いて作製した。培地中に分泌された組換えタンパク質は、実施例1に記載の方法に従い、馴化培地からNi-NTAアガロースと抗FLAG M2アガロースを用いる2段階のアフィニティークロマトグラフィーにより精製した。精製した組換えタンパク質はPBSに対して透析した後、22μmのディスクディスクシリンジフィルター(Millipore, #SLGV033RS)でろ過滅菌し、―80℃で保存した。組換えCBD付加ラミニンE8および組換えラミニンE8の作製に使用した発現ベクターの組み合わせを表5に示す。
精製タンパク質の濃度は、ウシ血清アルブミン(BSA)をスタンダードに用いるBCA法で定量した。精製タンパク質の純度は非還元条件でSDS-PAGEを行い、Coomassie Brilliant Blue染色して確認した。
各アイソフォームの組換えラミニンE8およびCBD付加ラミニンE8SDS-PAGEの結果を図6~14に示した。いずれの組換えラミニンE8およびCBD付加ラミニンE8も、非還元条件ではα鎖E8の単量体とβ鎖E8-γ1鎖E8二量体の2つのバンドが検出され、ラミニン511E8およびCBD付加ラミニン511E8と同様のヘテロ三量体タンパク質として精製できていることが確認された。
(1)結合活性の測定
実施例2(1)の「抗ラミニンα5抗体5D6抗血清をwash bufferで3000倍希釈したもの」に代えて「抗FLAG抗体M2(Sigma)をwash bufferで2000倍希釈したもの」を用いた以外は、実施例2(1)の方法と同じ方法でコラーゲン結合活性を測定した。
Claims (13)
- ラミニン、または、ヘテロ3量体を形成しているラミニンフラグメントのα鎖のN末端、β鎖のN末端およびγ鎖のN末端の少なくとも1か所にコラーゲン結合性分子が結合していることを特徴とする改変ラミニン。
- ラミニン、または、ヘテロ3量体を形成しているラミニンフラグメントのα鎖のN末端、β鎖のN末端およびγ鎖のN末端の2か所以上にコラーゲン結合性分子が結合していることを特徴とする請求項1に記載の改変ラミニン。
- ラミニンフラグメントが、インテグリン結合活性を有していることを特徴とする請求項1または2に記載の改変ラミニン。
- ラミニンフラグメントが、ラミニンE8フラグメントである請求項3に記載の改変ラミニン。
- ラミニン、または、ヘテロ3量体を形成しているラミニンフラグメントが、α1~α5から選択される1種のα鎖もしくはそのフラグメント、β1~β3から選択される1種のβ鎖もしくはそのフラグメント、およびγ1~γ3から選択される1種のγ鎖もしくはそのフラグメントからなることを特徴とする請求項1~4のいずれかに記載の改変ラミニン。
- ラミニン、または、ヘテロ3量体を形成しているラミニンフラグメントが、ラミニンα5β1γ1もしくはそのフラグメント、ラミニンα3β3γ2もしくはそのフラグメント、ラミニンα1β1γ1もしくはそのフラグメント、ラミニンα1β2γ1もしくはそのフラグメント、ラミニンα2β1γ1もしくはそのフラグメント、ラミニンα2β2γ1もしくはそのフラグメント、ラミニンα3β1γ1もしくはそのフラグメント、ラミニンα3β2γ1もしくはそのフラグメント、ラミニンα4β1γ1もしくはそのフラグメント、ラミニンα4β2γ1もしくはそのフラグメントまたはラミニンα5β2γ1もしくはそのフラグメントである請求項5に記載の改変ラミニン。
- コラーゲン結合性分子が、以下の(a)~(r)から選択される少なくとも1種以上であることを特徴とする請求項1~6のいずれかに記載の改変ヒトラミニン。
(a)フィブロネクチンまたはそのコラーゲン結合活性部位を含むフラグメント
(b)コラゲナーゼまたはそのコラーゲン結合活性部位を含むフラグメント
(c)インテグリンα1鎖またはそのコラーゲン結合ドメインを含むフラグメント
(d)インテグリンα2鎖またはそのコラーゲン結合ドメインを含むフラグメント
(e)インテグリンα10鎖またはそのコラーゲン結合ドメインを含むフラグメント
(f)インテグリンα11鎖またはそのコラーゲン結合ドメインを含むフラグメント
(g)血小板グリコプロテインVIまたはそのコラーゲン結合部位を含むフラグメント
(h)ディスコイディンドメイン受容体1またはそのコラーゲン結合部位を含むフラグメント
(i)ディスコイディンドメイン受容体2またはそのコラーゲン結合部位を含むフラグメント
(j)マンノース受容体またはそのコラーゲン結合部位を含むフラグメント
(k)ホスホリパーゼA2受容体またはそのコラーゲン結合部位を含むフラグメント
(l)DEC205またはそのコラーゲン結合部位を含むフラグメント

- 請求項1~8のいずれかに記載の改変ラミニン、ならびにコラーゲンおよび/またはゼラチンを含むことを特徴とする細胞外マトリックス材料。
- 請求項1~8のいずれかに記載の改変ラミニン、ならびにコラーゲンおよび/またはゼラチンがコーティングされていることを特徴とする培養基材。
- 請求項1~8のいずれかに記載の改変ラミニン、ならびにコラーゲンおよび/またはゼラチンを含むことを特徴とするスキャフォールド。
- 哺乳動物細胞の培養方法であって、請求項1~8のいずれかに記載の改変ラミニン、ならびにコラーゲンおよび/またはゼラチンの存在下で培養することを特徴とする培養方法。
- 哺乳動物細胞が、ES細胞、iPS細胞または体性幹細胞である請求項12に記載の培養方法。
