また、式(I)の化合物には、不斉炭素原子や軸不斉を有する場合があり、これに基づく光学異性体が存在しうる。本発明は、式(I)の化合物の光学異性体の分離されたもの、あるいはそれらの混合物も包含する。
式(I)の化合物及びその塩は、その基本構造あるいは置換基の種類に基づく特徴を利用し、種々の公知の合成法を適用して製造することができる。その際、官能基の種類によっては、当該官能基を原料から中間体へ至る段階で適当な保護基(容易に当該官能基に転化可能な基)に置き換えておくことが製造技術上効果的な場合がある。このような保護基としては、例えば、ウッツ(P. G. M. Wuts)及びグリーン(T. W. Greene)著、「Greene's Protective Groups in Organic Synthesis(第4版、2006年)」に記載の保護基等を挙げることができ、これらの反応条件に応じて適宜選択して用いればよい。このような方法では、当該保護基を導入して反応を行なったあと、必要に応じて保護基を除去することにより、所望の化合物を得ることができる。
また、式(I)の化合物のプロドラッグは、上記保護基と同様、原料から中間体へ至る段階で特定の基を導入、あるいは得られた式(I)の化合物を用いてさらに反応を行なうことで製造できる。反応は通常のエステル化、アミド化、脱水等、当業者に公知の方法を適用することにより行うことができる。
以下、式(I)の化合物の代表的な製造法を説明する。各製法は、当該説明に付した参考文献を参照して行うこともできる。なお、本発明の製造法は以下に示した例には限定されない。
本反応は、化合物1と脱保護試薬とを当量若しくは一方を過剰量用い、反応に不活性な溶媒中又は無溶媒下、冷却下から加熱還流下、通常0.1時間から5日間撹拌することによって行われる。ここで用いられる溶媒の例としては、特に限定はされないが、ジエチルエーテル、テトラヒドロフラン、ジオキサン、ジメトキシエタン等のエーテル類、ジクロロメタン、1,2-ジクロロエタン、クロロホルム等のハロゲン化炭化水素類等が挙げられる。脱保護試薬の例としては、特に限定はされないが、塩化水素のジオキサン溶液、塩化水素の酢酸エチル溶液、トリフルオロ酢酸等が挙げられる。
上記製造法における原料化合物は、例えば下記の方法、後述の製造例に記載の方法、公知の方法、あるいはそれらの変法を用いて製造することができる。
本工程は化合物2aとTetrahedron Letters、1993年、第34巻、第21号、p.3389-3392に記載の方法若しくはそれに準じた方法で製造した化合物2bとの縮合反応により化合物2cを得る工程である。
本工程は化合物2aと化合物2bとを当量若しくは一方を過剰量用い、これらの混合物を縮合剤の存在下、反応に不活性な溶媒中、冷却下から加熱還流下、好ましくは-20℃~60℃において、通常0.1時間~5日間攪拌することによって行われる。ここで用いられる溶媒の例としては、特に限定はされないが、ベンゼン、トルエン、キシレン等の芳香族炭化水素類、ジクロロメタン、1,2-ジクロロエタン、クロロホルム等のハロゲン化炭化水素類、ジエチルエーテル、テトラヒドロフラン(THF)、ジオキサン、ジメトキシエタン等のエーテル類、N,N-ジメチルホルムアミド(DMF)、ジメチルスルホキシド(DMSO)、酢酸エチル、アセトニトリル、水、あるいはこれらの混合物が挙げられる。縮合剤の例としては、1-(3-ジメチルアミノプロピル)-3-エチルカルボジイミド、ジシクロヘキシルカルボジイミド、1,1'-カルボニルジイミダゾール、ジフェニルリン酸アジド、オキシ塩化リン、O-(7-アザベンゾトリアゾール-1-イル)-N,N,N',N'-テトラメチルウロニウムヘキサフルオロホスフェート(HATU)等が挙げられるが、これらに限定されるものではない。1-ヒドロキシベンゾトリアゾール等の添加剤を用いることが反応の進行に好ましい場合もある。また、トリエチルアミン、N,N-ジイソプロピルエチルアミン、N-メチルモルホリン、ジメチルアミノピリジン、ピリジン等の有機塩基、又は炭酸カリウム、炭酸ナトリウム、水酸化カリウム等の無機塩基を用いることが反応の進行に好ましい場合もある。
本工程は化合物2cのエステル基部分(RcO-C(O)-)におけるRc基の脱保護により化合物2dを得る工程である。
本工程は化合物2cを金属触媒の存在下、反応に不活性な溶媒中、水素雰囲気下、室温から加熱還流下、好ましくは室温において、通常0.1時間~5日間攪拌することによって行われる。ここで用いられる溶媒の例としては、特に限定されないが、メタノール、エタノール、2-プロパノール等のアルコール類、ジエチルエーテル、テトラヒドロフラン、ジオキサン、ジメトキシエタン等のエーテル類、水、酢酸エチル、N,N-ジメチルホルムアミド、ジメチルスルホキシド及びこれらの混合物が挙げられる。金属触媒としては、パラジウム炭素、パラジウム黒、水酸化パラジウム等のパラジウム触媒、白金板、酸化白金等の白金触媒、還元ニッケル、ラネーニッケル等のニッケル触媒、ロジウム炭素、テトラキストリフェニルホスフィンクロロロジウム等のロジウム触媒、還元鉄等の鉄触媒等が好適に用いられる。反応を常圧から3気圧の水素雰囲気下で行うことが好ましい。塩酸等の無機酸を用いることが反応の進行に好ましい場合もある。
なお、本工程によりLx部分に-低級アルケニレン-を有する化合物の二重結合を還元した化合物が同時に合成できる。
本工程は化合物2dと所定のアミン化合物2eとのアミド化により化合物2fを得る工程である。
アミド化は、原料合成1の第一工程の方法を用いることもできる他、縮合剤の代わりにJournal of the Chemical Society. Chemical communications、1979年、第24巻、p.1180-1181に記載の1-クロロ-N,N,2-トリメチルプロプ-1-エン-1-アミンを用いることもできる。
本工程はR8がベンジル基、ベンジルオキシメチル基である場合に上記工程に加えてベンジル基、ベンジルオキシメチル基を脱保護し化合物2fから化合物2gを得る工程である。

所定のアミン化合物2eとトリホスゲンとを当量若しくは一方を過剰量用い、これらの混合物を塩基の存在下、反応に不活性な溶媒中、冷却下から加熱還流下、好ましくは-20℃~室温において、通常0.1時間~5日間攪拌することによって行われる。ここで用いられる溶媒の例としては、特に限定はされないが、ジクロロメタン、1,2-ジクロロエタン、クロロホルム等のハロゲン化炭化水素類が挙げられる。また塩基としてはトリエチルアミン、ピリジン等の有機塩基を用いることが反応の進行に好ましい場合もある。
次に化合物4bをイソシアネート化合物に対して当量若しくは過剰量加え、これらの混合物を塩基の存在下、反応に不活性な溶媒中、冷却下から加熱還流下、好ましくは室温において、通常0.1時間~5日間攪拌することによって行われる。ここで用いられる溶媒の例としては、特に限定はされないが、ジクロロメタン、1,2-ジクロロエタン、クロロホルム等のハロゲン化炭化水素類が挙げられる。また塩基としてはトリエチルアミン、ピリジン等の有機塩基を用いることが反応の進行に好ましい場合もある。

本工程は化合物5bと所定のアミン化合物2eとのアルキル化により化合物5cを得る工程である。
化合物5bと所定のアミン化合物2eとを当量若しくは一方を過剰量用い、これらの混合物を塩基の存在下、反応に不活性な溶媒中、冷却下から加熱還流下、好ましくは室温~100℃で、通常0.1時間~5日間攪拌することによって行われる。ここで用いられる溶媒の例としては、特に限定はされないが、ベンゼン、トルエン、キシレン等の芳香族炭化水素類、ジクロロメタン、1,2-ジクロロエタン、クロロホルム等のハロゲン化炭化水素類、ジエチルエーテル、テトラヒドロフラン(THF)、ジオキサン、ジメトキシエタン等のエーテル類、N,N-ジメチルホルムアミド(DMF)、ジメチルスルホキシド(DMSO)、酢酸エチル、アセトニトリル、あるいはこれらの混合物が挙げられる。また、トリエチルアミン、N,N-ジイソプロピルエチルアミン、N-メチルモルホリン、ジメチルアミノピリジン、ピリジン等の有機塩基、又は炭酸カリウム、炭酸ナトリウム、水酸化カリウム等の無機塩基を用いることが反応の進行に好ましい場合もある。さらに添加剤としてヨウ化カリウム等を用いることが反応の進行に好ましい場合がある。
本工程は化合物5cとTetrahedron Letters、1993年、第34巻、第21号、p.3389-3392に記載の方法若しくはそれに準じた方法で製造した化合物2bとの縮合反応により化合物5dを得る工程である。

化合物6aと所定のアミン化合物2eとを当量若しくは一方を過剰量用い、これらの混合物を、還元剤の存在下、反応に不活性な溶媒中、-45℃から加熱還流下、好ましくは0℃~室温において、通常0.1時間~5日間撹拌する。ここで用いられる溶媒の例としては、特に限定されないが、ジクロロメタン、1,2-ジクロロエタン、クロロホルム等のハロゲン化炭化水素類、メタノール、エタノール等のアルコール類、ジエチルエーテル、テトラヒドロフラン、ジオキサン、ジメトキシエタン等のエーテル類、及びこれらの混合物が挙げられる。還元剤としては、シアノ水素化ホウ素ナトリウム、トリアセトキシ水素化ホウ素ナトリウム、水素化ホウ素ナトリウム等が挙げられる。モレキュラーシーブス等の脱水剤、又は酢酸、塩酸、チタニウム(IV)イソプロポキシド錯体等の酸存在下で反応を行うことが好ましい場合がある。反応によっては、化合物6aと化合物2eとの縮合によりイミンが生成し、安定な中間体として単離できる場合がある。そのような場合には、このイミン中間体の還元反応により化合物6bを得ることができる。また、前記還元剤での処理の代わりに、メタノール、エタノール、酢酸エチル等の溶媒中、酢酸、塩酸等の酸の存在下又は非存在下で、還元触媒(例えば、パラジウム炭素、ラネーニッケル等)を用いて反応を行うこともできる。この場合、反応を常圧から50気圧の水素雰囲気下で、冷却下から加熱下で行うことが好ましい。

単離、精製は、抽出、分別結晶化、各種分画クロマトグラフィー等、通常の化学操作を適用して行なわれる。
各種の異性体は、適当な原料化合物を選択することにより製造でき、あるいは異性体間の物理化学的性質の差を利用して分離することができる。例えば、光学異性体は、ラセミ体の一般的な光学分割法(例えば、光学活性な塩基又は酸とのジアステレオマー塩に導く分別結晶化や、キラルカラム等を用いたクロマトグラフィー等)により得られ、また、適当な光学活性な原料化合物から製造することもできる。
1.ヒトおよびマウスにおけるトリプシン酵素阻害活性
実験にはヒトリコンビナントトリプシン(r-h trypsin ; WAKO社、カタログ番号206-17171)及びマウストリプシン(m-trypsin ; マウス小腸内容物より自社にて精製)を使用した。
マウス小腸内容物からのマウストリプシンの抽出方法を以下に示す。
マウス10匹分の小腸内容物および消化管をPBS中でポリトロンを用いて懸濁し、15,000xgの遠心処理を数回行った。その上清を、Benzamidine Sepharose 4 Fast Flow樹脂(GE Healthcare:#17-5123-10)に4℃で16時間混和した。PBSで樹脂を洗浄した後、Glycine 溶出緩衝液(pH3.0)でm-trypsinを溶出した。精製画分は抗マウスTrypsin抗体(Santa Cruz社:sc-67388)で認識されることをWesternblot解析にて確認した。
測定法は以下に概略を示す。
化合物をジメチルスルホキシド(DMSO)で溶解し、任意の濃度に希釈した(A)。Aをbuffer(0.1M Tris(pH8.0)、0.15M NaCl、10mM CaCl2、0.05% Brij38)で100倍希釈した(B)。r-h trypsinは0.088μg/mLとなるよう、m-trypsinは上記抽出液を1/50になるようbufferで希釈した(C)。m-trypsinの希釈倍率(1/50)は、kinetics解析により0.088μg/mL r-h trypsinと同じ活性を示すように設定した。酵素反応の基質BZiPAR基質溶液(Rhodamine標識基質)は5μmol/Lになるようbufferで希釈した(D)。B;5μL、C;5μL、D;10μLを384プレートに添加し、室温にて30分インキュベーションした。蛍光シグナルはTecan Safire Fluorometerを使用してEx/Em = 497/520にて検出した。化合物は2500nMから3倍比で0.0075nMまでの計12濃度で検討し、酵素存在下で化合物未添加時(DMSOのみ)を0%阻害、酵素非存在下で化合物未添加時を100%阻害とし、各化合物の阻害率を算出した。得られた阻害率を基に非線形回帰からIC50値(nM)を算出した。対照化合物としてCamostatを用いて同様に試験を行った。
実験には6週齢のICRマウス(オス)を用い、1群5例にて実施した。マウスを15時間絶食後、対照群は0.5%メチルセルロース(MC)液を、被験薬群は化合物を0.5%MC液に溶解、又は懸濁し強制経口投与(5mg/kg)した。その直後から絶食を解除し、自由摂餌(CE-2)を開始させたのち、3時間後から9時間後までの糞を回収した。得られた全ての糞を6mLの蒸留水で溶解・懸濁し、3000rpmで10分間遠心分離した。得られた上清中の蛋白濃度をBradford法にて測定し、糞中蛋白濃度を糞全重量で徐することで糞1g中に含まれる糞中蛋白濃度として算出し、対照に対する比から薬効を検討した。複数回評価した化合物については、平均値を算出した。
実験にはBALB/cマウス(メス)を用いた。6週齢の時点でDXRを尾静脈注射し、腎症を惹起した。被検化合物はDXR投与直後から一日一回経口投与した。投与量は3, 10, 30mg/kgの3用量とし、control群にはvehicleを同用量投与した。薬剤投与開始から1、2、3、4週間後にそれぞれ採尿を行い尿中タンパク排泄量を測定し、さらに4週目の採尿終了後に採血を行ない、血漿中クレアチニン量および尿素窒素(BUN)量を測定した。
本試験の結果より、当該化合物が、尿中タンパク排泄量の増大等の腎機能低下症状を良好に抑制し、病態の悪化を抑制することを確認した。
10週齢のWistarラット(オス)に左腎摘術を行い、その1週後にDXR(5mg/kg)を尾静脈内投与することによりモデルを作製した。被検化合物はモデル作製直後0.5%メチルセルロース(MC)に懸濁し3、10、30mg/kgの用量を1日2回強制経口投与した。sham群、control群には0.5% MCを強制経口投与した。被検薬投与開始から1、2、3、4週間後にそれぞれ24時間採尿を行い、尿中タンパク排泄量を測定した。4週目の採尿終了後に採血を行い、血漿中クレアチニン濃度を測定し、腎機能に対する効果も併せて確認した。
本試験の結果より、当該化合物は尿中タンパク排泄増加抑制、血漿中クレアチニン上昇抑制作用を示し、病態進行が抑制されることを確認した。
投与は錠剤、丸剤、カプセル剤、顆粒剤、散剤、液剤等による経口投与、又は、関節内、静脈内、筋肉内等の注射剤、坐剤、点眼剤、眼軟膏、経皮用液剤、軟膏剤、経皮用貼付剤、経粘膜液剤、経粘膜貼付剤、吸入剤等による非経口投与のいずれの形態であってもよい。
経口投与のための液体組成物は、薬剤的に許容される乳濁剤、溶液剤、懸濁剤、シロップ剤又はエリキシル剤等を含み、一般的に用いられる不活性な希釈剤、例えば精製水又はエタノールを含む。当該液体組成物は不活性な希釈剤以外に可溶化剤、湿潤剤、懸濁剤のような補助剤、甘味剤、風味剤、芳香剤、防腐剤を含有していてもよい。
(示差走査熱量計分析(DSC分析)) DSC分析は、TA Instrument製 Q2000およびQ1000を用いて行った。試料およそ2mgを専用のアルミニウム製サンプルパンに充填し、窒素雰囲気下(50mL/分)において、測定範囲を室温~300℃とし、昇温速度10℃/分で試料とリファレンス(空のアルミニウム製サンプルパン)との間に発生する熱量変化を連続的に測定し記録した。なお、データ処理を含む装置の取り扱いは、各装置で指示された方法及び手順に従った。
4-(4-ヒドロキシフェニル)ブタン酸(2.00g)のN,N-ジメチルホルムアミド(28.0mL)溶液に炭酸カリウム(1.61g)及びベンジルブロミド(1.46mL)を添加し、室温で終夜攪拌した。反応懸濁液を減圧下濃縮した後、残渣に水を加え、酢酸エチルで抽出した。有機層を水、飽和塩化ナトリウム水溶液にて洗浄した後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製し、ベンジル 4-(4-ヒドロキシフェニル)ブタノアート(2.64g)を得た。
4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]安息香酸(2.96g)のジクロロメタン(20.0mL)溶液に1-[3-(ジメチルアミノ)プロピル]-3-エチルカルボジイミド 塩酸塩(1.80g)、ベンジル 4-(4-ヒドロキシフェニル)ブタノアート(2.00g)、及びN,N-ジメチル-4-アミノピリジン(286mg)を添加し、室温で終夜攪拌した。反応溶液に1M 塩酸を加えたのち、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、4-[4-(ベンジルオキシ)-4-オキソブチル]フェニル 4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾアート(3.98g)を得た。
4-[4-(ベンジルオキシ)-4-オキソブチル]フェニル 4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾアート(3.98g)のテトラヒドロフラン(40.0mL)溶液に、10% パラジウム-炭素(398mg)を加え、3気圧の水素雰囲気下、室温で3時間攪拌した。反応懸濁液をセライトろ過した後、ろ液を減圧下濃縮し、4-[4-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)フェニル]ブタン酸(3.30g)を得た。
4-[4-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)フェニル]ブタン酸(250mg)のN,N-ジメチルホルムアミド(5.00mL)溶液にtert-ブチル O-tert-ブチル-L-チロシナート 塩酸塩(152mg)、1-[3-(ジメチルアミノ)プロピル]-3-エチルカルボジイミド 塩酸塩(88.5mg)、1H-ベンゾトリアゾール-1-オール(62.4mg)、及びトリエチルアミン(0.0643mL)を添加し、室温中終夜攪拌した。反応溶液に水を加え、酢酸エチルで抽出した。有機層を水、飽和塩化ナトリウム水溶液の順に洗浄した後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、tert-ブチル N-{4-[4-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)フェニル]ブタノイル}-O-tert-ブチル-L-チロシナート(327mg)を得た。
ベンジル (2-ブロモ-4-ヒドロキシフェニル)アセタート(400mg)のジメトキシエタン(4.00mL)及び水(1.20mL)の溶液にフェニルボロン酸(182mg)、炭酸ナトリウム(396mg)、及び1,1′-ビス(ジフェニルホスフィノ)フェロセン-パラジウム(II)ジクロリド-ジクロロメタン錯体(50.9mg)を加え、80℃で終夜撹拌した。反応混合物に水を加え、酢酸エチルで抽出した。有機層を水、飽和塩化ナトリウム水溶液の順に洗浄した後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、ベンジル (5-ヒドロキシビフェニル-2-イル)アセタート(344mg)を得た。
4-メトキシ-2-(トリフルオロメチル)フェニル酢酸(500mg)の酢酸(5.00mL)溶液に48%臭化水素酸水溶液(5.00mL)を加え145℃で終夜攪拌した。反応懸濁液を減圧下濃縮した後、残渣に水を加え、生じた固体をろ取し、4-ヒドロキシ-2-(トリフルオロメチル)フェニル酢酸(300mg)を得た。
ベンジル (2-ブロモ-4-ヒドロキシフェニル)アセタート(2.00g)のジクロロメタン(30.0mL)溶液に氷冷中、tert-ブチル (クロロ)ジメチルシラン(1.13g)及びN,N-ジメチル-4-アミノピリジン(913mg)を加え、氷冷下2時間攪拌した。反応混合物を水で希釈し酢酸エチルで抽出した。有機層を無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、ベンジル (2-ブロモ-4-{[tert-ブチル(ジメチル)シリル]オキシ}フェニル)アセタート(2.63g)を得た。
ベンジル (2-ブロモ-4-{[tert-ブチル(ジメチル)シリル]オキシ}フェニル)アセタート(250mg)のトルエン(5.00mL)溶液に水(0.250mL)、リン酸カリウム(366mg)、トリシクロヘキシルホスフィン(64.4mg)、酢酸パラジウム(II)(25.8mg)、及びシクロプロピルボロン酸(98.6mg)を加え、110℃で終夜攪拌した。反応混合物に水を加え、酢酸エチルで抽出した。有機層を水、飽和塩化ナトリウム水溶液の順に洗浄した後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製して、ベンジル (4-{[tert-ブチル(ジメチル)シリル]オキシ}-2-シクロプロピルフェニル)アセタート(210mg)を得た。
ベンジル (4-{[tert-ブチル(ジメチル)シリル]オキシ}-2-シクロプロピルフェニル)アセタート(586mg)のテトラヒドロフラン(6.89mL)溶液に、氷冷下1M フッ化テトラ-n-ブチルアンモニウムのテトラヒドロフラン溶液(2.23mL)を加えたのち、室温で終夜攪拌した。反応溶液に水を加え、酢酸エチルで抽出した。有機層を水、飽和塩化ナトリウム水溶液の順に洗浄した後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、ベンジル(2-シクロプロピル-4-ヒドロキシフェニル)アセタート(398mg)を得た。
4-[2-(ベンジルオキシ)-2-オキソエチル]-2-クロロフェニル4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾアート(1.98g)のテトラヒドロフラン(27.1mL)溶液に、10% パラジウム-炭素(196mg)を加え、常圧、水素雰囲気下、室温にて1時間攪拌した。反応懸濁液をセライトろ過した後、ろ液を減圧下濃縮し、[4-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-3-クロロフェニル]酢酸(1.78g)を得た。
1,3-ジクロロ-5-メトキシベンゼン(10.0g)、パラホルムアルデヒド(2.21g)、及び濃塩酸(100mL)の混合物に室温で濃硫酸(1.00mL)を加えた。100℃で8時間加熱還流し、室温まで放冷した後、水を加え、酢酸エチルで抽出した。有機層を水、飽和炭酸水素ナトリウム水溶液、飽和塩化ナトリウム水溶液で洗浄した後、無水硫酸マグネシウムで乾燥し、減圧下濃縮した。残渣にジオキサン(56.0mL)、1M 水酸化ナトリウム水溶液(113mL)を加えた後、100℃で3時間加熱還流した。反応溶液を放冷した後、水を加え、クロロホルムで抽出した。有機層を飽和塩化ナトリウム水溶液で洗浄し、無水硫酸マグネシウムにて乾燥した後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、(2,6-ジクロロ-4-メトキシフェニル)メタノール(4.67g)を得た。
(2,6-ジクロロ-4-メトキシフェニル)メタノール(4.66g)のテトラヒドロフラン(50.0mL)溶液に氷冷下、三臭化リン(0.846mL)を添加した後、氷冷下1時間攪拌した。反応懸濁液に飽和炭酸水素ナトリウム水溶液を加え、酢酸エチルで抽出した。有機層を水、飽和塩化ナトリウム水溶液にて洗浄した後、無水硫酸ナトリウムにて乾燥した。溶媒を減圧下濃縮し、2-(ブロモメチル)-1,3-ジクロロ-5-メトキシベンゼン(5.45g)を得た。
1,2-ジクロロ-3-メトキシベンゼン(5.00g)の酢酸(14.0mL)溶液に、パラホルムアルデヒド(1.00g)及び25%臭化水素酸の酢酸溶液(8.00mL)を添加した後、90℃で5時間撹拌した。反応混合物を室温まで放冷した後、氷水に加え、生じた固体をろ取した。得られた固体をヘキサンで再結晶し、1-(ブロモメチル)-2,3-ジクロロ-4-メトキシベンゼン(3.90g)を得た。
1-(ブロモメチル)-2,3-ジクロロ-4-メトキシベンゼン(1.00g)、水(3.00mL)、メタノール(6.00mL)、及びジメチルスルホキシド(2.00mL)の混合物に、室温でシアン化ナトリウム(246mg)を加えた後、40℃で5時間攪拌した。反応混合物を減圧下濃縮した後、水を加えた。生じた固体をろ取し、(2,3-ジクロロ-4-メトキシフェニル)アセトニトリル(776mg)を得た。
(2,3-ジクロロ-4-メトキシフェニル)アセトニトリル(770mg)を50% 酢酸水溶液(8.00mL)に溶解した後、濃硫酸(4.00mL)を加え、110℃で終夜攪拌した。反応混合物を室温まで冷却した後、氷水に加え、生じた固体をろ取した。水で洗浄し、(2,3-ジクロロ-4-メトキシフェニル)酢酸(737mg)を得た。
N-[(ベンジルオキシ)カルボニル]-4-(tert-ブトキシカルボニル)-L-フェニルアラニン(500mg)のテトラヒドロフラン(4.00mL)及びtert-ブチルアルコール(4.00mL)溶液に、二炭酸ジ-tert-ブチル(656mg)及びN,N-ジメチル-4-アミノピリジン(30.6mg)を添加した後、室温で終夜攪拌した。反応溶液に水を加え、酢酸エチルにて抽出した。有機層を水、飽和炭酸水素ナトリウム水溶液、飽和塩化ナトリウム水溶液の順に洗浄した後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮しtert-ブチル N-[(ベンジルオキシ)カルボニル]-4-(tert-ブトキシカルボニル)-L-フェニルアラニナート(641mg)を得た。
tert-ブチル N-[(ベンジルオキシ)カルボニル]-4-(tert-ブトキシカルボニル)-L-フェニルアラニナート(570mg)のエタノール(3.00mL)及びテトラヒドロフラン(3.00mL)溶液にアルゴン雰囲気下10% パラジウム-炭素(138mg)を添加した後、水素雰囲気下、室温で終夜攪拌した。反応懸濁液をセライトろ過し、ろ液を減圧下濃縮し、tert-ブチル 4-(tert-ブトキシカルボニル)-L-フェニルアラニナート(431mg)を得た。
アルゴン雰囲気下、4-(2-{[(ベンジルオキシ)カルボニル]アミノ}エチル)安息香酸(469mg)のトルエン(10.0mL)懸濁液に、80℃で1,1-ジ-tert-ブトキシ-N,N-ジメチルメタンアミン(1.50mL)を加えた後、80℃で30分間攪拌した。室温まで冷却し、反応溶液を減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、tert-ブチル 4-(2-{[(ベンジルオキシ)カルボニル]アミノ}エチル)ベンゾアート(439mg)を得た。
tert-ブチル 4-(2-{[(ベンジルオキシ)カルボニル]アミノ}エチル)ベンゾアート(515mg)のN,N-ジメチルホルムアミド(10.0mL)溶液に氷冷下、55% 水素化ナトリウム(65.0mg)を加え、氷冷下30分間攪拌した。氷冷下ブロモ酢酸tert-ブチル(0.300mL)を加え、室温で終夜攪拌した。反応混合物に氷冷下飽和塩化アンモニウム水溶液を加え、酢酸エチルで抽出した。有機層を水で洗浄後、無水硫酸マグネシウムにて乾燥した。溶媒を減圧下濃縮した後、残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、tert-ブチル 4-(2-{[(ベンジルオキシ)カルボニル](2-tert-ブトキシ-2-オキソエチル)アミノ}エチル)ベンゾアート(473mg)を得た。
ベンジル (2-ブロモ-4-ヒドロキシフェニル)アセタート(500mg)のN,N-ジメチルホルムアミド(12.5mL)溶液に塩化第一銅(1.62g)を加え、140℃で終夜攪拌した。反応混合物を放冷した後、氷水及び1M 塩酸を加え、攪拌した。酢酸エチルで抽出し、有機層を水、飽和炭酸水素ナトリウム水溶液、及び飽和塩化ナトリウム水溶液にて洗浄し、無水硫酸ナトリウムにて乾燥した。減圧下濃縮後、残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、ベンジル (2-クロロ-4-ヒドロキシフェニル)アセタート(381mg)を得た。
ベンジル N-(tert-ブトキシカルボニル)-3-フルオロ-L-フェニルアラニナート(659mg)に4M 塩化水素のジオキサン溶液(8.82mL)を加え、室温で終夜攪拌した。反応混合物を減圧下濃縮し、ベンジル 3-フルオロ-L-フェニルアラニナート 塩酸塩(505mg)を得た。
ベンジル N-{[4-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-2-クロロフェニル]アセチル}-3-フルオロ-L-フェニルアラニナート(260mg)のテトラヒドロフラン(4.00mL)溶液に、1M 塩酸を一滴加え、アルゴン雰囲気下、10% パラジウム-炭素(20.5mg)を加え、常圧、水素雰囲気下、室温にて1時間攪拌した。反応混合物をセライトろ過した後、ろ液を減圧下濃縮し、N-{[4-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-2-クロロフェニル]アセチル}-3-フルオロ-L-フェニルアラニン(187mg)を得た。
tert-ブチル L-フェニルアラニナート 塩酸塩(800mg)のジメチルスルホキシド(3.20mL)溶液に、炭酸カリウム(429mg)及びtert-ブチルアクリラート(0.901mL)を加えた後、マイクロウェーブにて150℃中1時間撹拌した。室温まで冷却した後、水を加え、酢酸エチルで抽出した。有機層を無水硫酸ナトリウムにて乾燥した後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、tert-ブチル N-(3-tert-ブトキシ-3-オキソプロピル)-L-フェニルアラニナート(266mg)を得た。
tert-ブチル L-フェニルアラニナート 塩酸塩(456mg)のアセトニトリル(8.00mL)溶液に炭酸カリウム(510mg)及びtert-ブチル 3-(ブロモメチル)ベンゾアート(400mg)を加え、室温で終夜攪拌した。反応懸濁液に水を加え、酢酸エチルで抽出した。有機層を無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、tert-ブチル N-[3-(tert-ブトキシカルボニル)ベンジル]-L-フェニルアラニナート(340mg)を得た。
tert-ブチル 3-(2-アミノエチル)ベンゾアート(900mg)のN,N-ジメチルホルムアミド(20.0mL)溶液に氷冷下、炭酸カリウム(600mg)及びtert-ブチル ブロモアセタート(0.600mL)を加え、室温で5時間撹拌した。反応懸濁液に水を加え、酢酸エチルで抽出した。有機層を水で洗浄し、無水硫酸マグネシウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、tert-ブチル 3-{2-[(2-tert-ブトキシ-2-オキソエチル)アミノ]エチル}ベンゾアート(738mg)を得た。
[4-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-2-クロロフェニル]酢酸(300mg)のN,N-ジメチルホルムアミド(4.00mL)溶液に氷冷下、O-(7-アザベンゾトリアゾール-1-イル)-N,N,N',N'-テトラメチルウロニウム ヘキサフルオロホスフェート(207mg)、ジイソプロピルエチルアミン(0.0950mL)及びtert-ブチル N-(2-フェニルエチル)グリシナート(128mg)を加え、室温で5時間攪拌した。反応混合物に水を加え、酢酸エチルで抽出した。有機層を水で洗浄し、無水硫酸マグネシウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、4-{2-[(2-tert-ブトキシ-2-オキソエチル)(2-フェニルエチル)アミノ]-2-オキソエチル}-3-クロロフェニル 4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾアート(267mg)を得た。
N,N-ジメチルホルムアミド(10.0mL)をアルゴンでバブリングしたのち、4-ブロモ-3-クロロフェノール(1.00g)、ベンジルアクリラート(1.00mL)、ビス(ジベンジリデンアセトン)パラジウム(0)(85.0mg)、トリス(2-メチルフェニル)ホスフィン(150mg)、及びトリエチルアミン(0.700mL)を加え、アルゴン雰囲気下、100℃で終夜撹拌した。反応懸濁液を室温まで空冷後、水を加え酢酸エチルで抽出した。有機層を飽和塩化ナトリウム水溶液で洗浄し、無水硫酸マグネシウムで乾燥した後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、ベンジル (2E)-3-(2-クロロ-4-ヒドロキシフェニル)アクリラート(1.25g)を得た。
tert-ブチル 3-(クロロメチル)ベンゾアート(25.3g)のN,N-ジメチルホルムアミド(250mL)溶液にtert-ブチルグリシナート 塩酸塩(37.4g)、炭酸カリウム(61.7g)を加え、60℃で終夜撹拌した。反応懸濁液に水を加え、酢酸エチルで抽出した。有機層を水、飽和塩化ナトリウム水溶液にて洗浄した後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、tert-ブチル 3-{[(2-tert-ブトキシ-2-オキソエチル)アミノ]メチル}ベンゾアート(23.0g)を得た。
tert-ブチル N-(3-ニトロベンジル)グリシナート(3.39g)のジクロロメタン(20.0mL)溶液にトリエチルアミン(7.99mL)を加えた。氷冷下トリフルオロ酢酸無水物(2.70mL)のジクロロメタン(13.0mL)溶液を加え、室温で4時間攪拌した。氷冷下、水で希釈し、クロロホルムにて抽出した。有機層を水、飽和塩化ナトリウム水溶液で洗浄し、無水硫酸マグネシウムで乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、tert-ブチル N-(3-ニトロベンジル)-N-(トリフルオロアセチル)グリシナート(4.30g)を得た。
tert-ブチル N-(3-ニトロベンジル)-N-(トリフルオロアセチル)グリシナート(4.30g)、鉄(13.2 g)、水(41.8mL)、及びエタノール(83.5mL)の混合物に、室温で塩化アンモニウム(2.54g)を加え、100℃で1時間加熱還流した。放冷後反応懸濁液をセライトろ過し、ろ液を減圧下濃縮した。残渣に水を加え、酢酸エチルで抽出した。有機層を無水硫酸マグネシウムで乾燥した後、減圧下濃縮して、tert-ブチル N-(3-アミノベンジル)-N-(トリフルオロアセチル)グリシナート(3.76g)を得た。
tert-ブチル N-(3-アミノベンジル)-N-(トリフルオロアセチル)グリシナート(1.95g)及びトリエチルアミン(0.981mL)のジクロロメタン(19.5mL)溶液に、氷冷下tert-ブチル (クロロスルホニル)カルバマート(1.52g)のジクロロメタン(10.0mL)溶液を加え、室温で3時間攪拌した。反応混合物に水を加え、酢酸エチルで抽出した。有機層を飽和炭酸水素ナトリウム水溶液、飽和塩化ナトリウム水溶液で洗浄した後、無水硫酸マグネシウムで乾燥し、減圧下濃縮した。残渣に酢酸エチルを加え、n-ヘキサンを加えた。析出した固体をろ取し、tert-ブチル N-(3-{[(tert-ブトキシカルボニル)スルファモイル]アミノ}ベンジル)-N-(トリフルオロアセチル)グリシナート(2.19g)を得た。
tert-ブチル N-(3-{[(tert-ブトキシカルボニル)スルファモイル]アミノ}ベンジル)-N-(トリフルオロアセチル)グリシナート(2.19g)に50%メタノール水溶液(43.9mL)、炭酸カリウム(1.19g)を加え、室温下5時間攪拌した。反応混合物を減圧下濃縮した後、残渣を水に溶解し、1M 塩酸で中和した。n-ブタノールと酢酸エチルの混合溶媒で抽出し、無水硫酸マグネシウムで乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(メタノールークロロホルム)にて精製し、tert-ブチル N-(3-{[(tert-ブトキシカルボニル)スルファモイル]アミノ}ベンジル)グリシナート(1.38g)を得た。
tert-ブチル N-(3-シアノベンジル)-N-(トリフルオロアセチル)グリシナート(1.15g)とヒドロキシルアミン 塩酸塩(703mg)をエタノール(23.1mL)に懸濁し、室温でトリエチルアミン(1.41mL)を加え、65℃で6時間攪拌した。反応懸濁液を減圧下濃縮した後、残渣に水を加え、酢酸エチルで抽出した。有機層を無水硫酸マグネシウムで乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、tert-ブチル N-[3-(N'-ヒドロキシカルバムイミドイル)ベンジル]-N-(トリフルオロアセチル)グリシナート(853mg)を得た。
tert-ブチル N-[3-(N'-ヒドロキシカルバムイミドイル)ベンジル]-N-(トリフルオロアセチル)グリシナート(853mg)の1,4-ジオキサン(17.1mL)溶液に1,1'-カルボニルジイミダゾール(479mg)を加え、100℃で2時間攪拌した。反応混合物を放冷後減圧下濃縮し、残渣に水を加え、酢酸エチルで抽出した。有機層を水、飽和塩化ナトリウム水溶液で洗浄後、無水硫酸マグネシウムで乾燥した。減圧下濃縮し、tert-ブチル N-[3-(5-オキソ-4,5-ジヒドロ-1,2,4-オキサジアゾール-3-イル)ベンジル]-N-(トリフルオロアセチル)グリシナート(1.01g)を得た。
エチル 6-ホルミルピリジン-2-カルボキシラート(1.74g)のジクロロエタン(30.0mL)溶液にtert-ブチルグリシナート 塩酸塩(4.89g)、トリエチルアミン(4.06mL)、及び酢酸(2.78mL)を加え、氷冷中トリアセトキシ水素化ホウ素ナトリウム(6.18g)を加えた。反応懸濁液を室温で終夜撹拌した。反応混合物に飽和炭酸水素ナトリウム水溶液を加えた後、塩基性になるまで炭酸水素ナトリウムを加えた。反応混合物をクロロホルムにて抽出し、有機層を無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣を塩基性シリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、エチル 6-{[(2-tert-ブトキシ-2-オキソエチル)アミノ]メチル}ピリジン-2-カルボキシラート(2.83g)を得た。
エチル 6-{[(2-tert-ブトキシ-2-オキソエチル)アミノ]メチル}ピリジン-2-カルボキシラート(2.81g)のテトラヒドロフラン(30.0mL)溶液に水(30.0mL)及び炭酸水素ナトリウム(962mg)を加えた後、氷冷下クロロギ酸ベンジル(1.47mL)を加えた。反応懸濁液を室温で終夜撹拌後、酢酸エチルにて抽出し、有機層を飽和塩化ナトリウム水溶液にて洗浄後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、エチル 6-({[(ベンジルオキシ)カルボニル](2-tert-ブトキシ-2-オキソエチル)アミノ}メチル)ピリジン-2-カルボキシラート (3.27g)を得た。
エチル 6-({[(ベンジルオキシ)カルボニル](2-tert-ブトキシ-2-オキソエチル)アミノ}メチル)ピリジン-2-カルボキシラート(3.26g)のテトラヒドロフラン(10.0mL)溶液に1M 水酸化ナトリウム水溶液(8.00mL)を加え、室温で終夜撹拌した。反応混合物に1M 塩酸(8.00mL)を加え、クロロホルムにて抽出した。有機層を無水硫酸ナトリウムにて乾燥した後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、6-({[(ベンジルオキシ)カルボニル](2-tert-ブトキシ-2-オキソエチル)アミノ}メチル)ピリジン-2-カルボン酸(2.08g)を得た。
tert-ブチル 2-メチルキノリン-4-カルボキシラート(4.61g)及び四塩化炭素(50.0mL)の混合物にN-ブロモスクシンイミド(3.71g)及びアゾビスイソブチロニトリル(156mg)を加え、90℃で5時間撹拌した。反応懸濁液をろ過し、ろ液を減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、tert-ブチル 2-(ブロモメチル)キノリン-4-カルボキシラート(2.62g)を得た。
tert-ブチル 3-シアノ-5-フルオロベンゾアート(3.00g)のテトラヒドロフラン(60.0mL)-メタノール(30.0mL)の混合溶液に、氷冷下、塩化コバルト(II)(4.40g)と水(60.0mL)の混合物を加えた後、アルゴン気流下で水素化ホウ素ナトリウム(1.54g)を加え、室温で30分攪拌した。不溶物をセライトろ過した後、テトラヒドロフラン-メタノール-水(2:1:2)の混合溶媒にて洗浄した後、ろ液を減圧下濃縮した。残渣に飽和塩化ナトリウム水溶液を加えた後、クロロホルムで抽出した。有機層を硫酸マグネシウムで乾燥した後、減圧下濃縮した。残渣を塩基性シリカゲルカラムクロマトグラフィー(クロロホルム‐メタノール)にて精製し、tert-ブチル 3-(アミノメチル)-5-フルオロベンゾアート(1.46g)を得た。
tert-ブチル 3-(クロロメチル)ベンゾアート(500mg)のN,N-ジメチルホルムアミド(10.0mL)溶液にtert-ブチル 4-アミノベンゾアート(426mg)、炭酸カリウム(610mg)、及びヨウ化カリウム(36.6mg)を加え、60℃で終夜撹拌した。反応懸濁液に水を加え、酢酸エチルで抽出した。有機層を水、飽和塩化ナトリウム水溶液にて洗浄した後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製して、tert-ブチル 3-({[4-(tert-ブトキシカルボニル)フェニル]アミノ}メチル)ベンゾアート(671mg)を得た。
3-[4-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-2-クロロフェニル]プロパン酸(200mg)のジクロロメタン(3.00mL)溶液に氷冷下1-クロロ-N,N,2-トリメチルプロプ-1-エン-1-アミン(0.0700mL)を加え、室温で1時間攪拌した。氷冷下、tert-ブチル N-[3-(2-tert-ブトキシ-2-オキソエチル)フェニル]グリシナート(170mg)とピリジン(0.0500mL)のジクロロメタン(3.00mL)溶液を加え、室温にて終夜攪拌した。反応混合物に水を加え、クロロホルムで抽出した。有機層を無水硫酸マグネシウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製して、4-(3-{(2-tert-ブトキシ-2-オキソエチル)[3-(2-tert-ブトキシ-2-オキソエチル)フェニル]アミノ}-3-オキソプロピル)-3-クロロフェニル 4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾアート(210mg)を得た。
メチル 2-({[3-(2-クロロ-4-ヒドロキシフェニル)プロパノイル]アミノ}メチル)ベンゾアート(380mg)のN,N-ジメチルホルムアミド(4.00mL)溶液にクロロ tert-ブチルジメチルシラン(215mg)、及びイミダゾール(114mg)を加え、室温で4時間撹拌した。反応混合物に水、1M 塩酸を加え、酢酸エチルにて抽出した。有機層を飽和塩化ナトリウム水溶液にて洗浄後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、メチル 2-({[3-(4-{[tert-ブチル(ジメチル)シリル]オキシ}-2-クロロフェニル)プロパノイル]アミノ}メチル)ベンゾアート(500mg)を得た。
メチル 2-({[3-(4-{[tert-ブチル(ジメチル)シリル]オキシ}-2-クロロフェニル)プロパノイル]アミノ}メチル)ベンゾアート(460mg)の濃塩酸(10.0mL)溶液を50℃で4日間撹拌した。反応混合物を減圧下濃縮後、残渣にクロロホルム‐メタノールを加え、不溶物をろ別した。ろ液を減圧下濃縮し、残渣をシリカゲルカラムクロマトグラフィー(クロロホルム‐メタノール)にて精製し、2-({[3-(2-クロロ-4-ヒドロキシフェニル)プロパノイル]アミノ}メチル)安息香酸(140mg)を得た。
tert-ブチル 3-[4-(ベンジルオキシ)-2-クロロフェニル]プロパノアート(22.5g)のジクロロメタン(67.6 mL)溶液にトリフルオロ酢酸(24.8mL)を加え、室温で3時間撹拌した。反応混合物を減圧下濃縮後、残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、3-[4-(ベンジルオキシ)-2-クロロフェニル]プロパン酸(13.5g)を得た。
ベンジル [4-(ベンジルオキシ)-2-クロロフェニル]アセタート(4.39g)のテトラヒドロフラン(80.0mL)溶液に、アルゴン雰囲気下氷冷中、1M 水素化ジイソブチルアルミニウムのトルエン溶液(37.0mL)を加え、氷冷下1時間攪拌した。反応混合物に氷冷下飽和ロッシェル塩水溶液を加えた。室温で2時間攪拌後、水を加え、酢酸エチルで抽出した。有機層を無水硫酸マグネシウムにて乾燥し、溶媒を減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、2-[4-(ベンジルオキシ)-2-クロロフェニル]エタノール(3.01g)を得た。
2-[4-(ベンジルオキシ)-2-クロロフェニル]エタノール(1.50g)のジクロロメタン(30.0mL)溶液に氷冷下、1,1,1-トリアセトキシ-1,1-ジヒドロ-1,2-ベンゾヨードキソール-3(1H)-オン(2.70g)を加え、室温で1時間攪拌した。反応混合物に1M チオ硫酸ナトリウム水溶液を加え、クロロホルムにて抽出した。有機層を飽和炭酸水素ナトリウム水溶液、飽和塩化ナトリウム水溶液で洗浄後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、[4-(ベンジルオキシ)-2-クロロフェニル]アセトアルデヒド(1.01g)を得た。
(2-クロロ-4-メトキシフェニル)アセトアルデヒド(1.22g)のトルエン(30.0mL)溶液にtert-ブチル (トリフェニルホスホラニリデン)アセタート(2.75g)を加え、80℃で終夜撹拌した。反応混合物を室温まで空冷後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、tert-ブチル (2E)-4-(2-クロロ-4-メトキシフェニル)ブト-2-エノアート(1.67g)を得た。
tert-ブチル 4-(2-クロロ-4-メトキシフェニル)ブタノアート(1.73g)のジクロロメタン(10.0mL)溶液に氷冷下、1M 三臭化ホウ素のジクロロメタン溶液(12.2mL)を加え、室温で終夜攪拌した。反応混合物を減圧下濃縮後、飽和炭酸水素ナトリウム水溶液を加え、液性を塩基性にした後、1M 塩酸を液性が酸性になるまで加えた。析出した不溶物をろ取し、減圧下乾燥し4-(2-クロロ-4-ヒドロキシフェニル)ブタン酸(647mg)を得た。
メチル 5-(2-クロロ-4-ヒドロキシフェニル)ペンタノアート(2.28g)のメタノール(20.0mL)溶液に1M 水酸化ナトリウム水溶液(28.2mL)を加え、室温で1時間攪拌した。反応溶液に1M 塩酸(28.2mL)を加え、クロロホルムにて抽出した。有機層を無水硫酸ナトリウムにて乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、5-(2-クロロ-4-ヒドロキシフェニル)ペンタン酸(2.22g)を混合物として得た。
3-[4-(ベンジルオキシ)-2-クロロフェニル]プロパン酸(1.00g)のテトラヒドロフラン(20.0mL)溶液に1,1'-カルボニルジイミダゾール(1.11g)を加え、室温で2時間攪拌した。続いて水素化ホウ素ナトリウム(520mg)、及び水(4.00mL)を加え、室温で1時間攪拌した。反応混合物に1M 塩酸を加え、酢酸エチルで抽出した。有機層を飽和炭酸水素ナトリウム水溶液で洗浄後、無水硫酸ナトリウムで乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、3-[4-(ベンジルオキシ)-2-クロロフェニル]プロパン-1-オール(861mg)を得た。
4-アミノ-3-クロロフェノール 塩酸塩(3.00g)のピリジン(20.0mL)溶液に氷冷中クロロギ酸ベンジル(2.62mL)を加え、室温で終夜撹拌した。反応混合物を減圧下濃縮後、残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、ベンジル (2-クロロ-4-ヒドロキシフェニル)カルバマート(1.59g)を得た。
4-アミノ-3-クロロフェノール 4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾアート(600mg)のエタノール(6.00mL)-テトラヒドロフラン(6.00mL)溶液に、1H-1,2,3-ベンゾトリアゾール-1-イルメタノール(195mg)を加え、室温で終夜撹拌した。反応混合物を減圧下濃縮後、残渣にN,N-ジメチルホルムアミド(6.00mL)及びテトラヒドロフラン(6.00mL)を加え、氷冷下水素化ホウ素ナトリウム(89.9mg)を加え、室温で3時間撹拌した。反応混合物に水を加え、酢酸エチルで抽出した。有機層を水、飽和塩化ナトリウム水溶液で洗浄後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、3-クロロ-4-(メチルアミノ)フェニル 4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾアート(362mg)を得た。
3-クロロ-4-(メチルアミノ)フェニル 4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾアート(200mg)のジクロロメタン(4.00mL)溶液に氷冷下ピリジン(0.0341mL)、及びトリホスゲン(40.0mg)のジクロロメタン(2.00mL)溶液を加え、同温にて30分間攪拌した。さらにピリジン(0.0155mL)、及びトリホスゲン(22.9mg)のジクロロメタン(2.00mL)溶液を加え、同温にて30分間攪拌した。反応溶液に水を加え、クロロホルムで抽出した。有機層を無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣にジクロロメタン(4.00mL)を加え、氷冷下ピリジン(33.5mg)、及びtert-ブチル 3-{[(2-tert-ブトキシ-2-オキソエチル)アミノ]メチル}ベンゾアート(124mg)のジクロロメタン(4.00mL)溶液を加え、室温で終夜撹拌した。反応混合物に水を加え、酢酸エチルで抽出した。有機層を水、飽和塩化ナトリウム水溶液で洗浄後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、tert-ブチル 3-{[{[4-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-2-クロロフェニル](メチル)カルバモイル}(2-tert-ブトキシ-2-オキソエチル)アミノ]メチル}ベンゾアート(315mg)を得た。
tert-ブチル [4-(ベンジルオキシ)-2-クロロフェニル]カルバマート(3.00g)のN,N-ジメチルホルムアミド(45.0mL)溶液に窒素気流下、氷冷中、55% 水素化ナトリウム(471mg)を加え、氷冷下30分間撹拌した。氷冷下ヨードメタン(0.673mL)を加え、30分間撹拌した。反応混合物に水を加え、酢酸エチルで抽出した。有機層を無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、tert-ブチル [4-(ベンジルオキシ)-2-クロロフェニル]メチルカルバマート(1.42g)を得た。
ベンジル (2-クロロ-4-ヒドロキシフェニル)カルバマート(9.26g)のジクロロメタン(100mL)溶液に、氷冷下ヨウ化テトラn-ブチルアンモニウム(12.3g)、ジイソプロピルエチルアミン(11.4mL)、及び[2-(クロロメトキシ)エチル](トリメチル)シラン(11.8mL)を加え、室温で2時間撹拌した。反応混合物に水を加え、酢酸エチルで抽出した。有機層を無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、ベンジル (2-クロロ-4-{[2-(トリメチルシリル)エトキシ]メトキシ}フェニル)カルバマート(10.3g)を得た。
2-クロロ-N-メチル-4-{[2-(トリメチルシリル)エトキシ]メトキシ}アニリン(1.00g)のジクロロメタン(20.0mL)溶液に、氷冷下クロロアセチルクロリド(0.415mL)及びトリエチルアミン(0.872mL)を加え、氷冷下1時間撹拌した。反応混合物に水を加え、酢酸エチルで抽出した。有機層を飽和塩化ナトリウム水溶液で洗浄後、無水硫酸ナトリウムで乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、2-クロロ-N-(2-クロロ-4-{[2-(トリメチルシリル)エトキシ]メトキシ}フェニル)-N-メチルアセトアミド(1.08g)を得た。
2-クロロ-N-(2-クロロ-4-{[2-(トリメチルシリル)エトキシ]メトキシ}フェニル)-N-メチルアセトアミド(500mg)のアセトニトリル(8.95mL)溶液にtert-ブチル 3-{[(2-tert-ブトキシ-2-オキソエチル)アミノ]メチル}ベンゾアート(441mg)、炭酸カリウム(285mg)、及びヨウ化カリウム(22.8mg)を加え、60℃で終夜撹拌した。反応懸濁液に水を加え、酢酸エチルで抽出した。有機層を水、飽和塩化ナトリウム水溶液で洗浄し、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、tert-ブチル 3-{[(2-tert-ブトキシ-2-オキソエチル){2-[(2-クロロ-4-{[2-(トリメチルシリル)エトキシ]メトキシ}フェニル)(メチル)アミノ]-2-オキソエチル}アミノ]メチル}ベンゾアート(809mg)を得た。
tert-ブチル 3-{[(2-tert-ブトキシ-2-オキソエチル){2-[(2-クロロ-4-{[2-(トリメチルシリル)エトキシ]メトキシ}フェニル)(メチル)アミノ]-2-オキソエチル}アミノ]メチル}ベンゾアート(805mg)のテトラヒドロフラン(16.1mL)溶液に1M フッ化テトラ-n-ブチルアンモニウムのテトラヒドロフラン溶液(3.72mL)を加え、50℃で4日間攪拌した。反応溶液に水を加え、酢酸エチルで抽出した。有機層を水、飽和塩化ナトリウム水溶液で洗浄後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、tert-ブチル 3-{[(2-tert-ブトキシ-2-オキソエチル){2-[(2-クロロ-4-ヒドロキシフェニル)(メチル)アミノ]-2-オキソエチル}アミノ]メチル}ベンゾアート(583mg)を得た。
tert-ブチルN-[(ベンジルオキシ)カルボニル]-L-チロシナート(1.00g)のN,N-ジメチルホルムアミド(10.0mL)溶液に炭酸カリウム(2.71g)及びtert-ブチル 2-ブロモ-2-メチルプロパノアート(2.01mL)を加え、90℃で終夜撹拌した。反応懸濁液に水を加え、酢酸エチルで抽出した。有機層を無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、tert-ブチル N-[(ベンジルオキシ)カルボニル]-O-(1-tert-ブトキシ-2-メチル-1-オキソプロパン-2-イル)-L-チロシナート(1.21g)を得た。
6-ヒドロキシ-1H-インドール-2-カルボン酸(2.66g)のN,N-ジメチルホルムアミド(25.0mL)溶液に、炭酸リチウム(1.22g)を加えて、室温で10分間攪拌した。ベンジルブロミド(2.14mL)を加え、100℃で2時間攪拌した。反応混合物を60℃まで降温し、1M 塩酸(40.0mL)を加えた後、放冷し、酢酸エチルにて抽出した。有機層を飽和炭酸水素ナトリウム水溶液、飽和塩化ナトリウム水溶液で洗浄し、無水硫酸ナトリウムにて乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、粗精製物を冷エタノール(5.00mL)で洗浄することにより、ベンジル 6-ヒドロキシ-1H-インドール-2-カルボキシラート(1.95g)を得た。
6-ヒドロキシ-1H-インドール-3-カルボン酸(1.05g)のN,N-ジメチルホルムアミド(11.0mL)溶液に炭酸ナトリウム(722mg)、ベンジルブロミド(0.775mL)を加え、室温で終夜攪拌した。反応懸濁液に水を加え、酢酸エチルで抽出した。有機層を水、飽和塩化ナトリウム水溶液にて洗浄した後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(クロロホルム‐メタノール)にて精製して、ベンジル 6-ヒドロキシ-1H-インドール-3-カルボキシラート(1.04g)を得た。
tert-ブチル 4-(ブロモメチル)チオフェン-2-カルボキシラート(2.00g)のN,N-ジメチルホルムアミド(19.8mL)溶液にtert-ブチル L-アラニナート 塩酸塩(2.62g)及びトリエチルアミン(4.02mL)を加えたのち、60℃で12時間加熱した。反応懸濁液に水を加え、酢酸エチルで抽出した。有機層を飽和塩化ナトリウム水溶液にて洗浄したのち、無水硫酸マグネシウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、tert-ブチル 4-({[(2S)-1-tert-ブトキシ-1-オキソプロパン-2-イル]アミノ}メチル)チオフェン-2-カルボキシラート(1.31g)を得た。
tert-ブチル 3-(2,6-ジフルオロ-4-メトキシフェニル)プロパナート(11.7g)を48% 臭化水素水(200mL)に懸濁し、120℃にて16時間攪拌した。反応混合物を減圧下濃縮したのち、残渣に水を加え、酢酸エチルにて抽出した。有機層を水、飽和塩化ナトリウム水溶液にて洗浄したのち、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製して、3-(2,6-ジフルオロ-4-ヒドロキシフェニル)プロパン酸(3.17g)を得た。
tert-ブチル 4-{[(2-tert-ブトキシ-2-オキソエチル){2-[(4-{[tert-ブチル(ジメチル)シリル]オキシ}-2-クロロフェニル)アミノ]-2-オキソエチル}アミノ]メチル}チオフェン-2-カルボキシラート(375mg)のテトラヒドロフラン(7.50 mL)溶液に氷冷中、55% 水素化ナトリウム(25.2 mg)を加え、室温中30分間撹拌した。室温中ヨードメタン(0.187mL)を加え、室温中2時間撹拌した。反応混合物に酢酸エチルを加え、氷冷下、水及び飽和塩化アンモニウム水溶液を加えた。有機層を抽出したのち、無水硫酸マグネシウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製して、tert-ブチル 4-{[(2-tert-ブトキシ-2-オキソエチル){2-[(4-{[tert-ブチル(ジメチル)シリル]オキシ}-2-クロロフェニル)(メチル)アミノ]-2-オキソエチル}アミノ]メチル}チオフェン-2-カルボキシラート(209mg)を得た。
4-アミノ-3-フルオロ安息香酸(500mg)のテトラヒドロフラン(10.0mL)溶液に、N,N'-ビス-tert-ブトキシカルボニル-1-グアニルピラゾール(1.20g)を加え、室温中3日間攪拌した。反応懸濁液を減圧下濃縮したのち、残渣をシリカゲルカラムクロマトグラフィー(クロロホルム‐メタノール)にて精製して、4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]-3-フルオロ安息香酸(669mg)を得た。
tert-ブチル (3-ホルミルフェノキシ)アセタート(1.50g)のジクロロエタン(20.0mL)溶液にtert-ブチルグリシナート(874mg)及び酢酸(1.09mL)を加えたのち、氷冷中トリアセトキシ水素化ホウ素ナトリウム(2.69g)を加えた。反応懸濁液を室温中16時間撹拌した。反応混合物に飽和炭酸水素ナトリウム水溶液を加えたのち、酢酸エチルにて抽出した。有機層を飽和塩化ナトリウム水溶液にて洗浄し、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(クロロホルム‐メタノール)にて精製して、tert-ブチル N-[3-(2-tert-ブトキシ-2-オキソエトキシ)ベンジル]グリシナート(1.18g)を得た。
tert-ブチル [2-(クロロメチル)フェニル]アセタート(2.31g)のアセトニトリル (23.1mL)溶液にtert-ブチル グリシナート 塩酸塩(2.41g)及びトリエチルアミン(4.01mL)を加えたのち、60℃で3時間加熱した。反応懸濁液を減圧下濃縮したのち、残渣に水、酢酸エチルを加え、有機層を抽出した。有機層を無水硫酸マグネシウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、tert-ブチル N-[2-(2-tert-ブトキシ-2-オキソエチル)ベンジル]グリシナート(2.54g)を得た。
5-メチル-1-ベンゾチオフェン-2-カルボン酸(1.50g)のN,N-ジメチルホルムアミド(10.5mL)溶液に1,1'-カルボニルジイミダゾール(1.27g)を加えたのち、室温中2.5時間攪拌した。続いてtert-ブチルアルコール(1.44mL)及び1,8-ジアザビシクロ[5.4.0]ウンデカ-7-エン(1.17mL)を加えたのち、50℃で24時間加熱した。反応混合物に酢酸エチルを加え、有機層を1M 塩酸、飽和塩化ナトリウム水溶液の順に洗浄したのち、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、tert-ブチル 5-メチル-1-ベンゾチオフェン-2-カルボキシラート(1.78g)を得た。
tert-ブチル 3-{[{2-[4-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-2-クロロフェニル]プロパノイル}(2-tert-ブトキシ-2-オキソエチル)アミノ]メチル}ベンゾアート(300mg)をUVトリガーによる超臨界クロマトグラフィー法(CO2-メタノール)にて分取精製後、減圧下濃縮し、tert-ブチル 3-{[{2-[4-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-2-クロロフェニル]プロパノイル}(2-tert-ブトキシ-2-オキソエチル)アミノ]メチル}ベンゾアートの両エナンチオマー体としてPEx.69-1(135mg、RT:5.90min)及びPEx.69-2(134mg、RT:7.35min)をそれぞれ得た。なお、RTを求める為に行った超臨界クロマトグラフィー法の条件を以下に示す。

tert-ブチル [4-(アミノメチル)フェニル]アセタート(1.00g)のアセトニトリル(15.0mL)溶液にtert-ブチル ブロモアセタート(0.668mL)及びトリエチルアミン(0.693mL)を加えたのち、室温中4時間撹拌した。反応懸濁液を減圧下濃縮したのち、残渣に酢酸エチルを加えた。有機層を0.1M 塩酸、飽和炭酸水素ナトリウム水溶液、飽和塩化ナトリウム水溶液の順に洗浄したのち、無水硫酸マグネシウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、tert-ブチル N-[4-(2-tert-ブトキシ-2-オキソエチル)ベンジル]グリシナート(747mg)を得た。
1-[4-(ベンジルオキシ)-2-クロロフェニル]エタノン(500mg)とトルエン(15.0mL)の混合物にp-トルエンスルホン酸(110 mg)及びtert-ブチル グリシナート(0.800mL)を加えたのち、120℃で18時間加熱した。終了後、反応混合物を放冷したのち、減圧下濃縮した。残渣にメタノール(10.0mL)を加えたのち、氷冷中水素化ホウ素ナトリウム(90.0mg)を添加し、氷冷中2時間撹拌した。反応混合物に飽和塩化アンモニウム水溶液、酢酸エチルを加え、有機層を抽出した。有機層を無水硫酸マグネシウムにて乾燥し、減圧下濃縮した。残渣を残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製して、tert-ブチル N-{1-[4-(ベンジルオキシ)-2-クロロフェニル]エチル}グリシナート(560mg)を得た。
2-ベンジル 1-tert-ブチル (2S,4R)-4-ヒドロキシピロリジン-1,2-ジカルボキシラート(565mg)のテトラヒドロフラン(10.0mL)溶液にベンジル 4-ヒドロキシベンゾアート(405mg)及び1,1'-(アゾジカルボニル)ジピペリジン(555 mg)を加えたのち、氷冷下、トリn-ブチルホスフィン(0.540mL)を加え、室温中終夜撹拌した。反応混合物を減圧下濃縮したのち、残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、2-ベンジル 1-tert-ブチル (2S,4S)-4-{4-[(ベンジルオキシ)カルボニル]フェノキシ}ピロリジン-1,2-ジカルボキシラート(273mg)を得た。
2-クロロ-4-ヒドロキシベンズアルデヒド(400mg)のジクロロメタン(4.00mL)溶液に無水硫酸マグネシウム(615mg)及びtert-ブチル グリシナート 塩酸塩(679mg)を加えたのち、室温中終夜撹拌した。反応混合物を濾過したのち、濾液を減圧下濃縮した。残渣をメタノール(12.0 mL)に溶解させたのち、水素化ホウ素ナトリウム(193 mg)を加え、室温中2時間撹拌した。反応混合物に水、酢酸エチルを加え、有機層を抽出した。有機層を飽和塩化ナトリウム水溶液にて洗浄したのち、無水硫酸マグネシウムにて乾燥し、減圧下濃縮して、tert-ブチル N-(2-クロロ-4-ヒドロキシベンジル)グリシナート(613mg)を得た。
ジメチル (ベンジルオキシカルボニル)メチル ホスホナート(2.20g)のテトラヒドロフラン(32.0mL)溶液に氷冷中、55% 水素化ナトリウム(406mg)を加え、室温中30分間撹拌した。続いてtert-ブチル 3-ホルミルベンゾアート(1.60g)を加え、室温中16時間撹拌した。反応混合物に水、酢酸エチルを加え、有機層を抽出した。有機層を飽和炭酸水素ナトリウム水溶液、飽和塩化ナトリウム水溶液にて洗浄したのち、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製して、tert-ブチル 3-[(1E)-3-(ベンジルオキシ)-3-オキソプロパ-1-エン-1-イル]ベンゾアート(1.60g)を得た。
ジ-tert-ブチル 5-(ブロモメチル)イソフタル酸(1.00g)のテトラヒドロフラン(10.0mL)溶液に70%エチルアミン水溶液(2.21mL)を加えたのち、室温で終夜撹拌した。反応混合物に水、酢酸エチルを加え、有機層を抽出した。有機層を水にて洗浄し、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル、続いてクロロホルム‐メタノール)にて精製して、ジ-tert-ブチル 5-[エチルアミノ(メチル)]イソフタル酸(558mg)を得た。
エチル 3-(2-クロロ-4-{[2-(トリメチルシリル)エトキシ]メトキシ}フェニル)ブタ-2-エノアート(525mg)のテトラヒドロフラン(10.0mL)溶液に、5%のロジウム炭素(200mg)を加えたのち、水素雰囲気下で、室温中18時間攪拌した。反応懸濁液をセライト濾過し、濾液を減圧下濃縮したのち、残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製して、エチル 3-(2-クロロ-4-{[2-(トリメチルシリル)エトキシ]メトキシ}フェニル)ブタノアート(282mg)を得た。
55%水素化ナトリウム(128mg)のテトラヒドロフラン(15.0mL)溶液に氷冷中ベンジル 2-(ジメトキシホスホリル)プロパノアート(796mg)を加えたのち、氷冷中30分撹拌した。続いて4-{[tert-ブチル(ジメチル)シリル]オキシ}-2-クロロベンズアルデヒド(720mg)のテトラヒドロフラン(3.00mL)溶液を加えたのち、室温中4時間撹拌した。反応混合物に飽和塩化アンモニウム水溶液、水、酢酸エチルを加え、有機層を抽出した。有機層を無水硫酸マグネシウムにて乾燥し、減圧下濃縮した。残渣のテトラヒドロフラン(10.0mL)溶液に氷冷中、1M フッ化テトラ-n-ブチルアンモニウムのテトラヒドロフラン溶液(2.70mL)を加え、氷冷中30分撹拌した。終了後、反応混合物に飽和塩化アンモニウム水溶液、水、酢酸エチルを加え、有機層を抽出した。有機層を無水硫酸マグネシウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製して、ベンジル (2E)-3-(2-クロロ-4-ヒドロキシフェニル)-2-メチルアクリラート(587mg)を得た。
6-ヒドロキシキノリン-2-カルボン酸 (2.00 g)のN,N-ジメチルホルムアミド(20.0 mL)溶液に炭酸カリウム(3.07g)、ベンジルブロミド(3.02 mL)を添加し、50℃で、6時間攪拌した。反応混合物を放冷したのち、1M 塩酸、酢酸エチル、テトラヒドロフランを加え、有機層を抽出した。有機層を水、飽和塩化ナトリウム水溶液にて洗浄したのち、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をヘキサン‐酢酸エチルにて洗浄して、ベンジル 6-(ベンジルオキシ)キノリン-2-カルボキシラート(1.80g)を得た。
ベンジル 6-(ベンジルオキシ)キノリン-2-カルボキシラート(650mg)のテトラヒドロフラン(13.0mL)溶液に、-78℃中、1M 水素化ジイソブチルアルミニウムのトルエン溶液(2.64mL)を加えたのち、-78℃中1時間攪拌した。反応混合物に硫酸ナトリウム10水和物(680mg)を加えたのち、室温中終夜攪拌した。不溶物を濾取し、濾液を減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製して、6-(ベンジルオキシ)キノリン-2-カルバルデヒド(170mg)を得た。
2-ブロモ-1,3,5-トリメチルベンゼン(1.22g)のテトラヒドロフラン(20.0mL)溶液に-78℃中、1.59M n-ブチルリチウムのn-ヘキサン溶液(3.86mL)を加えたのち、-78℃中30分撹拌した。続いてtert-ブチル(4-ブロモ-2-チエニル)アセタート(1.55g)のテトラヒドロフラン(15.0 mL)溶液を加えたのち、-78℃中30分撹拌した。続いて-78℃中、1.59M n-ブチルリチウムのn-ヘキサン溶液(3.51mL)を加えたのち、-78℃中30分撹拌した。さらにN,N-ジメチルホルムアミド(0.451mL)を加えたのち、-78℃中1時間撹拌した。反応混合物に飽和塩化アンモニウム水溶液、酢酸エチルを加え、有機層を抽出した。有機層を無水硫酸マグネシウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製して、tert-ブチル (4-ホルミル-2-チエニル)アセタート(355mg)を得た。
tert-ブチル N-{[4-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-2-クロロフェニル]アセチル}-O-tert-ブチル-L-チロシナート (210mg)に4M 塩化水素のジオキサン溶液(6.38mL)を加え、終夜攪拌した。反応混合物を減圧下濃縮し、N-(4-{4-[(4-カルバムイミドアミドベンゾイル)オキシ]-2-クロロフェニル}アセチル)-L-チロシン 塩酸塩(120mg)を得た。
tert-ブチル N-{[4-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-2-クロロフェニル]アセチル}-1-トリチル-L-ヒスチジナート(265mg)のジクロロメタン(2.70mL)溶液に氷冷下、トリフルオロ酢酸(0.619mL)を加え、室温で終夜撹拌した。反応混合物を減圧下濃縮後、水で洗浄し、ろ液を減圧下濃縮した。残渣をODSシリカゲルカラムクロマトグラフィー(0.01M 塩酸-アセトニトリル)にて精製し、精製物を減圧下乾燥して、N-({4-[(4-カルバムイミドアミドベンゾイル)オキシ]-2-クロロフェニル}アセチル)-L-ヒスチジン 2塩酸塩(54.1mg)を得た。
3-{[(3-{4-[(4-カルバムイミドアミドベンゾイル)オキシ]-2-クロロフェニル}プロパノイル)(カルボキシメチル)アミノ]メチル}安息香酸 塩酸塩(727mg)を50%アセトニトリル水溶液(24.0mL)に溶解し、室温中1M 水酸化ナトリウム水溶液(1.20mL)を加えた。室温にて5時間撹拌した後、生じた固体をろ取し、50%アセトニトリル水溶液にて洗浄した。得られた固体に50%アセトニトリル水溶液(50.0mL)を加え、100℃で30分間撹拌した後、徐々に室温まで冷却し、室温にて終夜撹拌した。生じた固体をろ取し、50%アセトニトリル水溶液にて洗浄し、減圧下、40℃にて乾燥した。得られた固体(601mg)を50%アセトン水溶液(16.0mL)に懸濁させ、室温にてメタンスルホン酸(0.160mL)を加えた。室温にて終夜撹拌し、溶媒を減圧留去した。次に、アセトン(25.0mL)を加え、50℃で2時間撹拌した。室温まで冷却し、析出した結晶をろ取した後、40℃にて減圧下乾燥することで、3-{[(3-{4-[(4-カルバムイミドアミドベンゾイル)オキシ]-2-クロロフェニル}プロパノイル)(カルボキシメチル)アミノ]メチル}安息香酸 メシル酸塩(679mg)を得た。
tert-ブチル 3-{[{[6-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)キノリン-2-イル]カルボニル}(2-tert-ブトキシ-2-オキソエチル)アミノ]メチル}ベンゾアート(659mg)のジクロロメタン(5.00mL)溶液にトリフルオロ酢酸(2.40mL)を加えたのち、室温中終夜撹拌した。反応溶液を減圧下濃縮したのち、残渣にアセトニトリル(20.0mL)を加え、再び減圧下濃縮した。この操作を2回繰り返したのち、アセトニトリル(20.0mL)を加え、室温中1時間撹拌、さらに100℃中2時間加熱還流した。反応溶液を氷冷したのち、1M 水酸化ナトリウム(1.54mL)を加えた。続いて飽和炭酸水素ナトリウム水溶液を、pH5付近になるまで加えた。氷冷中1時間撹拌したのち、不溶物を濾取し、濾取物を50%アセトニトリル水溶液にて洗浄したのち、50℃中減圧下乾燥した。乾燥物(354mg)を33%アセトニトリル水溶液(9.00mL)に懸濁させたのち、メタンスルホン酸 (0.100mL)を添加した。60℃で30分撹拌したのち、水(6.00mL)を加え、室温中終夜撹拌した。析出した結晶を濾取し、10%アセトニトリル水溶液にて洗浄したのち、減圧乾燥して3-{[({6-[(4-カルバムイミドアミドベンゾイル)オキシ]キノリン-2-イル}カルボニル)(カルボキシメチル)アミノ]メチル}安息香酸(236mg)を得た。
tert-ブチル 3-{[{2-[4-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-2-クロロフェニル]プロパノイル}(2-tert-ブトキシ-2-オキソエチル)アミノ]メチル}ベンゾアート(180mg)のジクロロメタン(1.92mL)溶液にトリフルオロ酢酸(0.646mL)を加えたのち、室温中終夜撹拌した。終了後、反応溶液を減圧下濃縮したのち、残渣にアセトニトリル(20.0mL)を加え、再び減圧下濃縮した。この操作を2回繰り返したのち、残渣に50%アセトニトリル水溶液(5.00mL)を加え、氷冷中1M 水酸化ナトリウム水溶液(0.208mL)を加えた。氷冷中1時間撹拌したのち、析出した結晶を濾取し、濾取物を10%アセトニトリル水溶液にて洗浄したのち、減圧下乾燥して3-{[(2-{4-[(4-カルバムイミドアミドベンゾイル)オキシ]-2-クロロフェニル}プロパノイル)(カルボキシメチル)アミノ]メチル}安息香酸(92mg)を得た。
L- アスパラギン酸 ジ‐tert-ブチルエステル 塩酸塩(40μmoL)に、3-(4-ヒドロキシフェニル)プロピオン酸(299mg)及び4-ジメチルアミノピリジン(220mg)のN,N-ジメチルホルムアミド(30mL)溶液を0.5mL、並びにN-(3-ジメチルアミノプロピル)-N'-エチルカルボジイミド 塩酸塩(518mg)のN,N-ジメチルホルムアミド(30mL)溶液を0.5mL加え、室温にて終夜攪拌した。減圧下濃縮後、4-[N',N''-ビス(tert-ブトキシカルボニル)カルバミンイミドアミド]安息香酸(911mg)のジクロロメタン(60mL)溶液を1.0mL及びN-(3-ジメチルアミノプロピル)-N'-エチルカルボジイミド 塩酸塩(518mg)のジクロロメタン(30mL)溶液を0.5mL加え、室温で終夜攪拌した。トリフルオロ酢酸(0.5mL)を加え、室温でさらに終夜攪拌した。減圧下濃縮後MSトリガーによるLC(ギ酸水溶液/メタノール)にて分取精製後減圧下濃縮しN-(3-{4-[(4-カルバミンイミドアミドベンゾイル)オキシ]フェニル}プロパノイル)-L-アスパラギン酸(4.7mg)を得た。

環Aは下記式(a)、(b)又は(c)であり、
R2はH、(ハロゲン、-CO2H、-OH、-O-低級アルキル、置換されていてもよいシクロアルキル、置換されていてもよいアリール、置換されていてもよい芳香族へテロ環及び置換されていてもよい非芳香族へテロ環からなる群より選択される1以上の置換基で置換されていてもよい低級アルキル)、置換されていてもよいシクロアルキル、置換されていてもよいアリール、置換されていてもよい芳香族へテロ環、置換されていてもよい非芳香族へテロ環又は-C(O)-低級アルキレン-置換されていてもよいアリールであり、
L1は-Y1-低級アルキレン-Y2-又は-C(O)-N(R6)-であり、環Aが式(b)又は式(c)のとき、L1はさらに-C(O)-であってもよく、
