Abstract
Es wird die Verwendung einer Verbindung nach der Formel (I) als Kontaktbiozid beschrieben, wobei A ein Fragment, das als Kation an das freie Elektronenpaar eines Stickstoffatoms koordiniert, B ein Fragment, das als Anion mit einem Carbokation unter Bildung eines ungeladenen Produkts reagiert, R ein organischer Rest aus einer gesättigten aliphatischen Kette C6 bis C30, ein gesättigter cycloaliphatischer Rest, eine einfach oder mehrfach ungesättigte aliphatische Kette mit zumindest 6 Kohlenstoffatomen, ein Alkylspacer zu quervernetzenden Oligooxazolinen, ein Arylspacer zu quervernetzenden Oligooxazolinen, ein Phenyl, ein Alkylphenyl, ein Heteroaryl oder Copolymere davon und X ein biozid unwirksames Anion sind und wobei (n+m)>10 ist.

VERWENDUNG VON KONTAKTBIOZIDEN BASIEREND AUF POLY ( 2 - SUBSTITUIERTEN)
OXAZOLINEN
Technisches Gebiet
Um in für Einzeller charakteristische Stoffwechselvorgänge einzugreifen, sind Biozide, wie Sulfonamide, Cephalosporine, Tetracycline und dgl., kleinmolekular strukturiert und daher üblicherweise wasserlöslich. Nachteilig ist jedoch, dass auch auf andere Stoffwechselprozesse Einfluss genommen werden kann, die in einem wässrigen Milieu ablaufen. Durch Kontaktbiozide können diese Nachteile vermieden werden. Kontaktbiozide bilden in der Regel hochmolekulare, protonierbare chemische Verbindungen, die Einzeller durch bloßen Kontakt aufgrund von statischen Ladungen und der dadurch bedingten Zelllyse abtöten, ohne in wässrige Lösung überzugehen oder in den inneren Stoffwechsel einzugreifen. Allerdings konnten sich die bekannten Kontaktbiozide, beispielsweise Polymere von basischen Methacrylaten, aufgrund der systemimmanenten Estergruppen in der Praxis nicht durchsetzen, weil die Estergruppen keine ausreichende Hydrolysestabilität aufweisen und sich nur bedingt mit den marktdominanten Gruppen der Polyolefine vertragen.
Stand der Technik
Um geeignete Kontaktbiozide angeben zu können, wurden bereits Polymere, die durch kationische Polymerisation eines 2-Oxazolin erhalten werden, auf ihre antibakterielle Wirksamkeit untersucht. Dabei wurde 2-Ethyl-2-oxazolin mit
Methyltosilat als Katalysator einer ringöffnenden Polymerisation unterworfen, die zu Poly(2-ethyl-2-oxazolin) führte. Ein solches mit einem quaternären Ammoniumsalz funktionalisiertes Poly(2-ethyl-2-oxazolin) wies eine antimikrobielle Aktivität gegen S. Aureus auf, die verbessert werden konnte, wenn das quater- näre Ammoniumsalz und entsprechende funktionelle Gruppen am Polymerende in einer unimolekularen Mizelle aggregieren. Der nachfolgende gemeinsame Angriff beider Gruppen auf eine Phospholipidmembran führte zur Durchlöcherung der Membran. Allerdings ist dieses antimikrobielle Poly(2-ethyl-2-oxazolin) wasserlöslich.
Zur Herstellung von wasserunlöslichen antimikrobiellen Oberflächen wurde bereits ein Blockcopolymer aus Poly(2-Methyl-2-oxazolin) als hydrophiler Block mit quaternärem Stickstoff als Endgruppe vorgeschlagen. Außerdem ist es bekannt, ein Poly(2-Oxazolin), das einerseits über polymerisierbare Endgruppen, andererseits aber über eine quaternäre Stickstoff-Endgruppe verfügte, mit Hydroxyethylmethacrylat und Glyceroldimethacrylat zu copolymerisieren, wodurch ein Polymernetzwerk entstand. Diese bekannten Kontaktbiozide nehmen allerdings keine Rücksicht auf die Amphiphilie einer Cytoplasma- membran, die für eine verbesserte antibakterielle Wirkung eine feinfühlige Abstimmung zwischen der Hydrophilie und Hydrophobie des Kontaktbiozids erfordert.
Des Weiteren ist es bekannt, das durch eine ringöffnenden Polymerisation erhaltene Poly(2-ethyl-2-oxazolin) durch eine sauer katalysierte Hydrolyse zu linearem Polyethylenimin (Polyaziridin) zu überführen. Durch Copolymere können amphiphile Systeme erhalten werden, die zum Beispiel als micellare Katalysatoren, nichtionische Tenside oder Hydrogele Anwendung finden. Eine antimikrobielle Wirkung konnte jedoch wegen der nicht protonisierbaren
Amidstrukturen nicht beobachtet werden.
Darüber hinaus ist ein Verfahren bekannt (DE 1 720 437 A1 ), mit dessen Hilfe ein teilverseiftes, einen Phenylrest tragendes Poly-2-oxazolin hergestellt wer-
den kann. Diese Poly-2-oxazoline werden allerdings als Hilfsmittel in der Papier- und Textilindustrie als Antistatika und Korrosionshemmer eingesetzt. Die US 4 153466 A beschreibt ein Flammschutzmittel aus vollständig oder teilweise hydrolisierten Phosphatsalzen von Poly-2-oxazolinen. Das Polymer wird aus 2- Oxazolinen mittels kationischer Ringöffnungspolymerisation hergestellt, wobei die Ausgangspolymere als Reste an der Position 2 des Oxazolrings substituierte und unsubstituierte C-M S Alkyl- oder Alkylengruppen oder Phenyl- und Cyc- loakylreste sein können. Die US 2005238825 A1 offenbart schließlich eine vernetzte Polymerzusammensetzung von hydrolysierten Poly(2-alkyl-2- oxazolinen) für Beschichtungen von Tintenstrahldruckern.
Darstellung der Erfindung
Der Erfindung liegt somit die Aufgabe zugrunde, die an sich bekannten antibakteriellen Eigenschaften von Poly(2-oxazolinen) zu optimieren, um vorteilhafte Kontaktbiozide zu erhalten.
Die Erfindung löst die gestellte Aufgabe durch die Verwendung der Verbindung nach der Formel
A ein Fragment, das als Kation an das freie Elektronenpaar eines Stickstoffatoms koordiniert,
B ein Fragment, das als Anion mit einem Carbokation unter Bildung eines ungeladenen Produkts reagiert,
R ein organischer Rest aus einer gesättigten aliphatischen Kette C6 bis C30, ein gesättigter cycloaliphatischer Rest, eine einfach oder mehrfach ungesättigte
aliphatische Kette mit zumindest 6 Kohlenstoffatomen, ein Alkylspacer zu quervernetzenden Oligooxazolinen, ein Arylspacer zu quervernetzenden Oli- gooxazolinen, ein Phenyl, ein Alkylphenyl, Heteroaryl oder ein Copolymer davon und
X ein Anion sind
und wobei (n+m)>1 0 ist.
Durch die aufeinander abgestimmten Amid- und Aminanteile der erfindungsgemäßen Kontaktbiozide kann in vorteilhafter Weise die Amphiphilie der Cyto- plasmamembranen berücksichtigt werden, weil der Amidanteil die Lipophilie und der Aminanteil die Hydrophilie des Kontaktbiozids maßgeblich beeinflussen. Aufgrund einer entsprechenden Abstimmung der Amid- und Aminanteile wird es daher möglich, trotz einer nur geringfügigen Wasserlöslichkeit die zur Lyse der lipiddominierten Bakterienhüllen erforderliche Hydrophilie bei einer ausgeprägten Lipophilie zur Verfügung zu stellen.
Als Fragment A eignet sich vorzugsweise Wasserstoff, ein Alkyl Ci bis CQ oder eine Benzylgruppe. Als Fragment B kann vorteilhaft eine Hydroxygruppe oder eine Aminogruppe eingesetzt werden.
Der organische Rest R soll eine Wasserlöslichkeit des Amidanteils verhindern und bildet daher vorzugsweise längere Ketten, wobei die erforderliche Kettenlänge von der Beweglichkeit der Kettenmoleküle abhängt. So benötigen gesättigte aliphatische Ketten zu diesem Zweck wenigstens 6 Kohlenstoffatome, wobei die angestrebte Wirkung durch eine Substitution mit einer Mercapto-, einer Hydroxy- oder einer Aminogruppe unterstützt werden kann. Bevorzugt werden Ketten mit 9 bis 1 8 Kohlenstoffatomen.
Wird ein gesättigter cycloaliphatischer Rest R eingesetzt, so kann die Anzahl der Kohlenstoffatome wegen der geringeren Molekülbeweglichkeit verringert sein. Es kann daher ein gegebenenfalls mit einer Mercapto- oder einer Hydroxygruppe substituiertes Cyclopentyl, Cyclohexyl, Cycloheptyl oder Cyclooctyl
zum Einsatz kommen. Noch geringere Molekülbeweglichkeit wird durch ein- oder mehrfach ungesättigte aliphatische Ketten erreicht. Ausgangsstoffe für derartige Zwischenprodukte sind als ungesättigte Fettsäuren aus biologischem Ursprung vorteilhaft verfügbar.
Die zumindest weitgehende Unlöslichkeit des Amidanteils in Wasser kann auch durch einen Alkylspacer zu quervernetzenden Oligooxazolinen oder einen Arylspacer zu quervernetzenden Oligooxazolinen erreicht werden.
Mit Phenyl als Rest R, das gegebenenfalls mit einer Hydroxy-, Amino-, Nitro-, Halogen-, Alkyl-, und/oder Trifluormethylgruppe substituiert ist, kann auf die Basizität der Amidgruppe und damit ebenfalls auf die Wasserlöslichkeit Einfluss genommen werden. Gleiches gilt für den Einsatz eines Heteroaryls als Rest R, wobei vorzugsweise ein Pyridyl, ein Thienyl, ein Thiazolyl, ein Triazolyl, ein Pyrimidinyl oder ein Thiadiazolyl Verwendung finden.
Das an das Stickstoffkation der Amingruppe gebundene Anion kann unterschiedlich gewählt werden. Als bevorzugte Beispiele können ein Chlorid-, ein Hydrogensulfat-, ein Sulfat-, ein Dihydrogenphosphat-, ein Hydrogenphosphat-, ein Phosphat-, ein Nitrat-, ein Hydrogencarbonat-, ein Carbonat- und ein Alka- noat-Anion angegeben werden.
Natürlich ist die Erfindung nicht auf Polymere mit Chlorid als Gegenion beschränkt. Anstelle einer Hydrolyse mit Chlorwasserstoff kann ein anderes Hydrolysemedium eingesetzt werden, beispielsweise Schwefelsäure. Es kann aber auch eine nachträgliche Protonierung eines Nitrats, Phosphats, Carbonats oder dgl. vorgenommen werden. Außerdem kann eine Hydrolyse mittels organischer Mono-, Di- oder Polycarbonsäuren als Eonat vorgenommen werden.
Zur Herstellung erfindungsgemäßer Kontaktbiozide kann in einfacher Weise von einem 2-Oxazolin ausgegangen werden, das zunächst kationisch ringöffnend polymerisiert wird, bevor das gewonnene Polymer einer partiellen Hydro-
lyse unterworfen wird. Obwohl sich für die Hydrolyse ein Chlorwasserstoff anbietet, ist die Erfindung selbstverständlich nicht auf dieses Hydrolysemedium beschränkt. Durch die Teilhydrolyse des Poly(2-oxazolins) geht sein Amidanteil zum Teil in einen Aminanteil über, der über den Hydrolysegrad eingestellt werden kann, sodass sich eine einfache Möglichkeit zur gegenseitigen Abstimmung der Hydrophobie und der Hydrophilie eröffnet. Wegen der durch den erfindungsgemäßen Rest R bedingten längeren Ketten ergibt sich ein zeitlicher Hydrolyseverlauf, der die Einhaltung eines vorgegebenen Hydrolysegrads in einfacher weise erlaubt. Die Reaktionsgeschwindigkeit ist ja lediglich von der jeweiligen Konzentration des Amidanteils abhängig. Der Hydrolysegrad ist proportional der Wasserlöslichkeit des Kontaktbiozids, dessen Wasserlöslichkeit somit ebenfalls über den Hydrolysegrad eingestellt werden kann. Bei einem Hydrolysegrad von etwa 10 % zeigt sich eine antibakterielle Wirksamkeit, die sich mit zunehmender Hydrolyse steigert, der bevorzugte Hydrolysegrad liegt bei 20 bis 75 %, weil bei einem solchen Hydrolysegrad die Wasserlöslichkeit bei geeignetem Rest R des Amidanteils im Allgemeinen noch keine Rolle spielt.
Der Aminanteil des erfindungsgemäßen Kontaktbiozids kann zur Erhöhung der Wasserunlöslichkeit in die Polymermatrix eingebunden werden. Zu diesem Zweck Können die Stickstoffatome mittels polymerer oder copolymerer Carbonsäuren, beispielsweise Poly[propylen-co-methacryl- oder -maleinsäure], ionisch gebunden werden. Eine andere Möglichkeit besteht in einer Teiladditionsreaktion der Stickstoffatome an polymeren Epoxidstrukturen, wie Poly(bis- phenolglycidylether) oder Polyolefin-co-glyzidylmethacrylate. Es können aber auch Quervernetzungen angestrebt werden, und zwar mit Hilfe kleiner Anteile (0,5 bis 10 Gew.%) an mehrfachfunktionellen 2-Oxazolinen während der Ringöffnungspolymerisation. Als Arylspacer können in diesem Zusammenhang vorteilhaft Phenylen-1 ,3-bis-2-oxazolin und Phenyl-1 ,3,5-tris-2-oxazolin sowie als Alkylspacer Tetramethylen-1 ,4-bis-2-oxazolin und Oktamethylen-1 ,8-bis-2 oxazolin eingesetzt werden. Außerdem können Substituenten in die Polymermatrix eingebunden werden, die am organischen Rest des Amidanteils zur
Raktion zur Verfügung stehen. Hiefür eignen sich Ester-. Amid-, Urethan-, Thio- click-bindungen oder Oxiranadditionen.
Weg zur Ausführung der Erfindung
Beispiel:
Zur Synthese von Poly(2-nonyl-2-oxazolin) wurde eine Stammlösung aus 2- Nonyl-2-oxazolin (15,06 g; 76,1 mmol), aus Methyltosylat (0,236 g; 1 ,27 mmol) und aus Acetonitril (38 ml) unter Schutzgas hergestellt. Es wurden jeweils 15 ml der Stammlösung in ein Mikrowellengefäß überführt, dieses verschlossen und bei 150 °C für 16 Minuten polymerisiert. Anschließend wurde das Polymer abfiltriert, mit Acetonitril nachgewaschen und im Vakuum getrocknet.
Ausbeute: 94% weißer Feststoff
Das so gewonnene Poly(2-nonyl-2-oxazolin) (0,500 g; 2,53 mmol) wurde in ein Mikrowellenvial eingewogen, mit 6 M HCl (2 ml; 0,01 mol) versetzt und während unterschiedlicher Zeitspannen in der Mikrowelle bei 160 °C hydrolysiert. Der entstandene hellgelbe Feststoff, Poly(2-nonyl-2-oxazolin-co- (Aziridiniumchlorid), wurde mit 5 ml Aceton gewaschen, das Produkt abfiltriert und im Vakuum getrocknet. Der Hydrolysegrad kann in Abhängigkeit von der Reaktionszeit der nachstehenden Tabelle entnommen werden.
Das mit einem Hydrolysegrad von 75 % gewonnene erfindungsgemäße Kon- taktbiozid wurde in einer Menge von 1 Gew.% in ein Polypropylen mit einem Schmelzflussindex MFR von 0,42 bei einer Temperatur zwischen 170 und 180 °C eingeknetet und aus der homogenen Polymermasse Probeplättchen von 60 x 60 x 2 mm gespritzt. Diese Probeplättchen wurden gemäß JIS Z 2801 :2000 gegenüber E.coli, S.Aureus und Listeria Monocytogenes getestet. Gegenüber
allen erwähnten grampositiven und gramnegetiven Keimen wurde eine Hemmung von über 99,9% erreicht. Dieser Wert blieb auch nach einer einwöchigen Lagerung der Probeplattchen in destilliertem Wasser gleich.
Claims
1 . Verwendung einer Verbindung nach der Formel
A ein Fragment, das als Kation an das freie Elektronenpaar eines Stickstoffatoms koordiniert,
B ein Fragment, das als Anion mit einem Carbokation unter Bildung eines ungeladenen Produkts reagiert,
R ein organischer Rest aus einer gesättigten aliphatischen Kette CQ bis C30, ein gesättigter cycloaliphatischer Rest, eine einfach oder mehrfach ungesättigte aliphatische Kette mit zumindest 6 Kohlenstoffatomen, ein Alkylspacer zu quervernetzenden Oligooxazolinen, ein Arylspacer zu quervernetzenden Oligooxazolinen, ein Phenyl, ein Alkylphenyl, ein Heteroaryl oder Copolymere davon und
X ein Anion sind
und wobei (n+m)>10 ist.
2. Verwendung nach Anspruch 1 , dadurch gekennzeichnet, dass das Fragment A Wasserstoff, ein Alkyl C1 bis C6 oder eine Benzylgruppe ist.
3. Verwendung nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass das Fragment B eine Hydroxygruppe oder eine Aminogruppe ist.
4. Verwendung nach einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass der organischer Rest R eine gesättigte aliphatische Kette Cg bis Ci8 ist.
5. Verwendung nach einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass der organischen Rests R eine einfach oder mehrfach ungesättigte aliphatische Kette von C8 bis C25 ist
6. Verwendung nach einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass der cycloaliphatische Ring des organischen Rests R ein Cyclopentyl, Cyclohexyl, Cycloheptyl oder Cyclooctyl ist.
7. Verwendung nach einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass der organischen Rests R als Alkylphenyl ein Benzyl aufweist.
8. Verwendung nach einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass der organische Rest R als Heteroaryl ein Pyridyl, ein Thienyl, ein Thiazolyl, ein Triazolyl, ein Pyrimidinyl oder ein Thiadiazolyl ist.
9. Verwendung nach einem der Ansprüche 4 bis 8, dadurch gekennzeichnet, dass der organische Rest R durch Hydroxy-, Mercapto- oder Aminogrup- pen substituiert ist.
10. Verwendung nach einem der Ansprüche 7 bis 9, dadurch gekennzeichnet, dass beim Einsatz von Phenyl, Alkylphenyl oder Heteroaryl als organischer Rest R dieser durch eine Halogen-, Alkyl-, Alkoxy-, Nitro- und/oder Trifluorme- thylgruppe substituiert ist.

Verwendung nach einem der Ansprüche 1 bis 10, dadurch gekennzeichnet, dass das biozid unwirksame Anion X ein Chlorid-, ein Hydrogensulfat- , ein Sulfat-, ein Dihydrogenphosphat-, ein Hydrogenphosphat-, ein Phosphat-, ein Nitrat-, ein Hydrogencarbonat-, ein Carbonat- oder ein Alkanoat-Anion ist.
12. Verwendung nach einem der Ansprüche 1 bis 1 1 , dadurch gekennzeichnet, dass zunächst ein 2-Oxazolin oder wenigstens zwei unterschiedliche 2-Oxazoline kationisch ringöffnend polymerisiert wird bzw. copolymerisiert werden und anschließend das gewonnene Polymer einer partiellen Hydrolyse unterworfen wird.
