1.不快な味を有する薬物と第一添加剤を含有した混合物を得る工程と、
2.結合剤を造粒液調製用溶媒に溶解または懸濁させて造粒液を得る工程と、
3.前記混合物に前記造粒液を添加し、撹拌造粒装置によって造粒を行い、造粒物を得る工程と、
4.コーティング剤、滑沢剤および/または低粘度の結合剤をスプレー液調製用溶媒に溶解または懸濁させてスプレー液を得る工程と、
5.前記造粒物に前記スプレー液を噴霧し、造粒物を被覆して被覆造粒物を得る工程と、
6.前記被覆造粒物に第二添加剤およびカルボキシメチル基を有する崩壊剤を混合し、打錠する工程。
具体的には、ドネペジル塩酸塩、ゾルピデム酒石酸塩、リスペリドン、アムロジピンベシル酸塩などが挙げられる。
本発明において最も好ましい不快な味を有する薬物はドネペジル塩酸塩である。
本発明の造粒液調製時に好ましく用いられる結合剤としては、ヒドロキシプロピルセルロース、ポリビニルピロリドン、ヒプロメロースおよびポリビニルアルコールが挙げられる。
本発明のスプレー液調製時に好ましく用いられる滑沢剤としては、ステアリン酸マグネシウム、タルクおよび酸化チタンが挙げられる。
本発明において好ましく用いられる崩壊剤としては、結晶セルロースおよびトウモロコシデンプンが挙げられる。
本発明において好ましく用いられる流動化剤としては、軽質無水ケイ酸,酸化チタン,タルクが挙げられる。
本発明において好ましく用いられる着色剤としては、酸化チタン、タルク、三二酸化鉄、タール系色素が挙げられる。
本発明において好ましく用いられるコーティング剤としては、オイドラギットNE30D、オイドラギットL等のアクリルポリマー系コーティング剤である。
本発明において好ましく用いられる低粘度の結合剤としては、メチルセルロース、ヒプロメロースなどが挙げられる。
本発明において好ましく用いられるカルボキシメチル基を有する崩壊剤としてはカルメロース、クロスカルメロースナトリウム,カルメロースカルシウム、カルボキシメチルスターチナトリウムである。
また、本発明により、官能試験におけるドネペジル塩酸塩含有錠剤の服用後のドネペジル塩酸塩に起因する苦味等の不快な味の低減が可能となった。
また、本発明により、従来のドネペジル塩酸塩含有錠剤と比較してドネペジル塩酸塩含有錠剤の服用直後のドネペジル塩酸塩の溶出挙動をコントロールすること可能としたドネペジル塩酸塩含有錠剤を得ることが可能となった。
また、本発明により、従来のドネペジル塩酸塩含有錠剤と比較して摩損度の低減したドネペジル塩酸塩含有錠剤を得ることが可能となった。
また、本発明により、錠剤中の空隙率が低減したドネペジル塩酸塩含有錠剤を得ることが可能となった。
ドネペジル塩酸塩65重量部とD-マンニトール110重量部を混合して混合物を得た後、該混合物を撹拌造粒装置(VG-05、(株)パウレック製)に投入し、さらにヒプロメロース2重量部と精製水20重量部からなる造粒液を添加して造粒物を得た。該造粒物を流動層乾燥機(マルチプレックスMP-01、(株)パウレック製)にて乾燥させ、パワーミルにて粉砕・整粒した。前記乾燥後の造粒物に、オイドラギットNE30D 150重量部、タルク25重量部、酸化チタン25重量部、メチルセルロース10重量部および精製水200重量部からなるスプレー液を噴霧し、被覆造粒物を得た。調製した該被覆造粒物20重量部に軽質無水ケイ酸2重量部、カルメロース10重量部、ステアリン酸マグネシウム2重量部、トウモロコシデンプン10重量部、結晶セルロース5重量部およびD-マンニトール101重量部を添加・混合し、ロータリー打錠機(VIRGO、菊水製作所製)にて打錠を行った。
実施例1で調製した被覆造粒物20重量部に、軽質無水ケイ酸2重量部、カルメロースカルシウム10重量部、ステアリン酸マグネシウム2重量部、トウモロコシデンプン10重量部、結晶セルロース5重量部およびD-マンニトール101重量部を添加・混合し、ロータリー打錠機(VIRGO、菊水製作所製)にて打錠を行った。
実施例1で調製した被覆造粒物20重量部に、カルボキシメチルスターチナトリウム10重量部、軽質無水ケイ酸2重量部、ステアリン酸マグネシウム2重量部、トウモロコシデンプン10重量部、結晶セルロース5重量部およびD-マンニトール101重量部を添加・混合し、ロータリー打錠機(VIRGO、菊水製作所製)にて打錠を行った。
ドネペジル塩酸塩65重量部とD-マンニトール110重量部を混合して混合物を得た後、該混合物を撹拌造粒装置(ハイスピードミキサーFS-02、(株)深江製作所製)に投入し、さらにヒドロキシプロピルセルロース2重量部と精製水20重量部からなる造粒液を添加して造粒物を得た。該造粒物を流動層乾燥機(マルチプレックスMP-01、(株)パウレック製)にて乾燥させ、パワーミルにて粉砕・整粒した。前記乾燥後の造粒物に、オイドラギットNE30D 150重量部、酸化チタン50重量部、ヒプロメロース10重量部および精製水200重量部からなるスプレー液を噴霧し、被覆造粒物を得た。調製した該被覆造粒物20重量部に軽質無水ケイ酸2重量部、カルメロース10重量部、ステアリン酸マグネシウム2重量部、トウモロコシデンプン10重量部、結晶セルロース5重量部およびD-マンニトール101重量部を添加・混合し、ロータリー打錠機(VIRGO、菊水製作所製)にて打錠を行った。
ゾルピデム酒石酸塩50重量部と乳糖水和物50重量部を混合して混合物を得た後、該混合物を撹拌造粒装置(VG-05、(株)パウレック製)に投入し、さらにメチルセルロース2重量部と精製水20重量部からなる造粒液を添加して造粒物を得た。該造粒物を流動層乾燥機(マルチプレックスMP-01、(株)パウレック製)にて乾燥させ、パワーミルにて粉砕・整粒した。前記乾燥後の造粒物に、オイドラギットL30D55 150重量部、酸化チタン50重量部、クエン酸トリエチル5重量部および精製水200重量部からなるスプレー液を噴霧し、被覆造粒物を得た。調製した該被覆造粒物30重量部にクロスカルメロースナトリウム30重量部、ステアリン酸マグネシウム2重量部およびD-マンニトール90重量部を添加・混合し、ロータリー打錠機(VIRGO、菊水製作所製)にて打錠を行った。
実施例1で調製した被覆造粒物20重量部に、クロスポビドン10重量部、軽質無水ケイ酸2重量部、ステアリン酸マグネシウム2重量部、トウモロコシデンプン10重量部、結晶セルロース5重量部およびD-マンニトール101重量部を添加・混合し、ロータリー打錠機(VIRGO、菊水製作所製)にて打錠を行った。
実施例1で調製した被覆造粒物20重量部に、結晶セルロース10重量部、ステアリン酸マグネシウム2重量部およびD-マンニトール118重量部を添加・混合し、ロータリー打錠機(VIRGO、菊水製作所製)にて打錠を行った。
実施例1で調製した被覆造粒物20重量部に、ステアリン酸マグネシウム2重量部およびエフメルトtypeC128重量部を添加・混合し、ロータリー打錠機(VIRGO、菊水製作所製)にて打錠を行った。
ドネペジル塩酸塩65重量部とD-マンニトール110重量部を混合して混合物を得た後、該混合物を流動層造粒装置(マルチプレックスMP-01、(株)パウレック製)に投入し、さらにヒドロキシプロピルセルロース3重量部と精製水50重量部からなる造粒液を添加して造粒物を得た。次に該造粒物をパワーミルにて粉砕・整粒した。粉砕・整粒後の造粒物に、オイドラギットNE30D 150重量部、タルク50重量部および精製水200重量部からなるスプレー液を噴霧し、被覆造粒物を得た。調製した該被覆造粒物20重量部に軽質無水ケイ酸2重量部、カルメロース10重量部、ステアリン酸マグネシウム2重量部、トウモロコシデンプン10重量部、結晶セルロース5重量部およびD-マンニトール101重量部を添加・混合し、ロータリー打錠機(VIRGO、菊水製作所製)にて打錠を行った。
実施例5で調製した乾燥後の造粒物20重量部に、カルメロースナトリウム30重量部、ステアリン酸マグネシウム2重量部およびD-マンニトール98重量部を添加・混合し、ロータリー打錠機(VIRGO、菊水製作所製)にて打錠を行った。
実施例4で調製した乾燥後の造粒物30重量部に、カルメロース30重量部、ステアリン酸マグネシウム5重量部およびD-マンニトール155重量部を添加・混合し、ロータリー打錠機(VIRGO、菊水製作所製)にて打錠を行った。
アリセプト(登録商標)10mgD錠(製造販売:エーザイ株式会社)を用いた。
実施例1~4および比較例7で得られたドネペジル塩酸塩含有経口製剤をそれぞれ光安定性試験室(LTB-180C型,ナガノサイエンス製)に入れ、D65ランプ下、1000ルクス/hrの照度で累積30万ルクス・hrを照射した後、各検体についてドネペジル塩酸塩の濃度が1mg/mLになるように移動層に用いる溶液に溶解した。移動相には1-デカンスルホン酸水溶液:アセトニトリル:過塩素酸が1300:700:1の割合で混合し調製された溶液を用いた。試験機器(高速液体クロマトグラフ:LC-20A、(株)島津製作所製)を用い、検出器としてUV検出器を用い、試料を入れるカラム(Mightsil ODS RP-18 GP 150-4.6)の温度を40℃に保ち、カラムへの試料注入量10μL、流速1.3mL/minの条件で測定波長271nmにてドネペジル塩酸塩のピークに対する類縁物質のピーク面積比を求め、類縁物質の定量を行った。試験結果を表1に示す。
健康な成人男子(年齢27歳、身長175cm、体重65kg)の口腔内のだ液で錠剤が完全に溶解するまでの味を評価した。試験は2回行われ、結果は2回の総合評価を表2に表す。
-:不快な味または苦味を全く感じない
±:不快な味または苦味を僅かに感じる
+:不快な味または苦味を感じる
++:不快な味または苦味をかなり感じる
実施例1~3および比較例1~3にて調製した錠剤3個を、注射筒正倒立法にて医薬成分の溶出率(%)を比較した。なお、溶出率は試験開始30秒後の測定値とし、結果を表3に示した。
先端に開孔径0.22μm~ 0.45μmのサイズ25mmのフィルター(ポール社製 エキクロディスク) を装着し、液漏れ防止のためにフィルター先端の液出口を熱可塑性フィルム(PARAFILM(登録商標)M) で巻いて塞いだプラスチック製の10mL注射筒(ニプロ社製)を用意する。このガラス10mL注射筒にドネペジル塩酸塩15mg相当量の錠剤を入れ、37±1℃に保った水10mLを入れる。それと同時にピストンを装着し、3秒に一回の割合で30秒間、計10回注射筒を正倒立させてパラフィルムを外し、フィルターによりろ過、そのはじめのろ液を5mL捨てた後、残りのろ液を回収し、ドネペジル塩酸塩量を測定する。
回収したろ液を試験用液とする。別途、105℃2時間減圧乾燥させたドネペジル塩酸塩標準品を精密に約150mg量りとり、本試験の溶出に用いた水を加え、超音波照射して溶かし、更に水を加え正確に100mLとして標準溶液とする。この標準溶液および試験溶液を検出器として紫外可視吸光光度計(U-3300、日立製作所(株))を用い、試料を10mm×10mmの石英セルに4mL入れ、測定波長271nmにより吸光度測定を行った。検出したドネペジル塩酸塩の数値を用いて以下に示す(1)式により簡易溶出試験(注射筒正倒立法)による溶出量を求める。得られた試験結果を表3に示す。

実施例1,4,5および比較例7で得られたドネペジル塩酸塩含有経口製剤について、錠剤の摩損度試験法(日本薬局方15改正)に準拠して試験を実施し、摩損度(%)を比較し、結果を表4に示した。
実施例4および比較例4にてそれぞれ得られた造粒物および被覆造粒物の拡大図を以下の図1~図4に示す。
Claims (8)
- 下記の1~6の工程によって得られる不快な味を有する薬物を含有する経口剤
1.不快な味を有する薬物と第一添加剤を含有した混合物を得る工程と、
2.結合剤を造粒液調製用溶媒に溶解または懸濁させて造粒液を得る工程と、
3.前記混合物に前記造粒液を添加し、撹拌造粒装置によって造粒を行い、造粒物を得る工程と、
4.コーティング剤、滑沢剤および/または低粘度の結合剤をスプレー液調製用溶媒に溶解または懸濁させてスプレー液を得る工程と、
5.前記造粒物に前記スプレー液を噴霧し、造粒物を被覆して被覆造粒物を得る工程と、
6.前記被覆造粒物に第二添加剤およびカルボキシメチル基を有する崩壊剤を混合し、打錠する工程。 - 不快な味を有する薬物が、ドネペジル塩酸塩、ゾルピデム酒石酸塩、リスペリドン、アムロジピンベシル酸塩から選ばれる1種の薬物を含有する請求項1記載の経口剤。
- 不快な味を有する薬物が、ドネペジル塩酸塩である請求項1記載の経口剤。
- 第一添加剤が、賦形剤である請求項1記載の経口剤。
- コーティング剤がアクリルポリマー系コーティング剤である請求項1記載の経口剤。
- コーティング剤がオイドラギットNEである請求項1記載の経口剤。
- カルボキシメチル基を有する崩壊剤が、カルメロース、カルメロースナトリウム、カルメロースカルシウム、クロスカルメロースナトリウム、カルボキシメチルスターチナトリウム、カルボキシメチルエチルセルロースである請求項1記載の経口剤。
- 下記の1~6の工程によって示される不快な味を有する薬物を含有する経口剤の製造方法
1.不快な味を有する薬物と第一添加剤を含有した混合物を得る工程と、
2.結合剤を造粒液調製用溶媒に溶解または懸濁させて造粒液を得る工程と、
3.前記混合物に前記造粒液を添加し、撹拌造粒装置によって造粒を行い、造粒物を得る工程と、
4.コーティング剤、滑沢剤および/または低粘度の結合剤をスプレー液調製用溶媒に溶解または懸濁させてスプレー液を得る工程と、
5.前記造粒物に前記スプレー液を噴霧し、造粒物を被覆して被覆造粒物を得る工程と、
6.前記被覆造粒物に第二添加剤およびカルボキシメチル基を有する崩壊剤を混合し、打錠する工程。
