Abstract
Description
〔1〕α-ヒドロキシ酸アンモニウム塩水溶液に塩基性金属類を接触させてα-ヒドロキシ酸金属塩を製造する工程であって、溶液中に残存するα-ヒドロキシ酸アミドを500[重量ppm/α-ヒドロキシ酸]以下にする工程(1)と、前記α-ヒドロキシ酸金属塩を脱塩してα-ヒドロキシ酸を製造する工程(2)と、を含むα-ヒドロキシ酸の製造方法;
〔2〕前記工程(1)において、溶液中に残存するアンモニア濃度を3[重量%/α-ヒドロキシ酸]以下にする、上記〔1〕に記載のα-ヒドロキシ酸の製造方法;
〔3〕前記工程(1)において、発生したアンモニアを気相部に回収する、上記〔1〕または〔2〕に記載のα-ヒドロキシ酸の製造方法;
〔4〕前記アンモニアを気相部に回収するときの温度を、60℃以上とする、上記〔3〕に記載のα-ヒドロキシ酸の製造方法;
〔5〕前記α-ヒドロキシ酸アンモニウム塩水溶液がα-ヒドロキシニトリルの加水分解反応によって得られたものである、上記〔1〕から〔4〕のいずれか1項に記載のα-ヒドロキシ酸の製造方法;
〔6〕前記α-ヒドロキシニトリルの加水分解を、ニトリラーゼ、及び/又はニトリルヒドラターゼとアミダーゼの組み合わせによって酵素触媒的に行う、上記〔5〕に記載のα-ヒドロキシ酸の製造方法;
〔7〕前記α-ヒドロキシニトリルの加水分解が、ニトリラーゼによって行われる、上記〔5〕に記載のα-ヒドロキシ酸の製造方法;
〔8〕前記ニトリラーゼが、Acinetobacter属由来である、上記〔6〕または〔7〕に記載のα-ヒドロキシ酸の製造方法;
〔9〕前記ニトリラーゼが、Acinetobacter sp.AK226由来である、上記〔8〕に記載のα-ヒドロキシ酸の製造方法;
〔10〕前記塩基性金属類が、アルカリ金属類、ベリリウムまたはマグネシウムの水酸化物、酸化物及び炭酸塩からなる群から選択される一種以上であって、前記工程(2)において、前記α-ヒドロキシ酸金属塩をイオン交換法によって脱塩する、上記〔1〕から〔9〕のいずれか1項に記載のα-ヒドロキシ酸の製造方法;
〔11〕前記塩基性金属類が、アルカリ金属類、ベリリウム、またはマグネシウムの水酸化物、酸化物及び炭酸塩からなる群から選択される一種以上であって、且つ、前記工程(2)において、前記α-ヒドロキシ酸金属塩を電気透析法によって脱塩する、上記〔1〕から〔9〕のいずれか1項に記載のα-ヒドロキシ酸の製造方法;
〔12〕前記塩基性金属類が、カルシウム、ストロンチウム、バリウムまたはラジウムの水酸化物、酸化物及び炭酸塩からなる群から選択される一種以上であって、前記工程(2)において、前記α-ヒドロキシ酸金属塩を、硫酸を添加することによって脱塩する、上記〔1〕から〔9〕のいずれか1項に記載のα-ヒドロキシ酸の製造方法;
〔13〕前記工程(1)の後、固液分離により固体のα-ヒドロキシ酸金属塩を回収、洗浄し、前記工程(2)において、該固体のα-ヒドロキシ酸金属塩又はこれに水を加えたα-ヒドロキシ酸金属塩スラリーに硫酸を添加する、上記〔12〕に記載のα-ヒドロキシ酸の製造方法;
〔14〕前記塩基性金属類が、水酸化カルシウム、酸化カルシウム及び炭酸カルシウムからなる群から選択される一種以上である、上記〔12〕又は〔13〕に記載のα-ヒドロキシ酸の製造方法;
〔15〕前記工程(2)の後、アニオン交換樹脂によって不純物アニオンを除去する工程と、カチオン交換樹脂によって不純物カチオンを除去する工程と、をさらに含む、上記〔1〕から〔14〕のいずれか1項に記載のα-ヒドロキシ酸の製造方法;
〔16〕前記不純物カチオンが、副生α-アミノ酸またはイミノジアルキル酸を含む、上記〔15〕に記載のα-ヒドロキシ酸の製造方法;
〔17〕前記α-ヒドロキシ酸が、乳酸またはグリコール酸である、上記〔1〕から〔16〕のいずれか1項に記載のα-ヒドロキシ酸の製造方法;
〔18〕前記α-ヒドロキシ酸が、グリコール酸である、上記〔17〕に記載のα-ヒドロキシ酸の製造方法;
〔19〕上記〔1〕から〔18〕のいずれか1項に記載の方法で得られたα-ヒドロキシ酸の水溶液を原料としてα-ヒドロキシ酸オリゴマーを合成する工程と、前記α-ヒドロキシ酸オリゴマーを解重合させてその環状二量体エステルを得る工程と、を含む、環状二量体エステルの製造方法;及び
〔20〕上記〔19〕に記載の方法で得られた環状二量体エステルを原料として開環重合反応でポリα-ヒドロキシ酸を得る工程を含む、ポリα-ヒドロキシ酸の製造方法、に関する。

菌体懸濁液中の乾燥菌体触媒重量の測定は、以下のごとく実施した。まず、適当な濃度の菌体触媒懸濁液を適量取り、-80℃まで冷却した後、凍結乾燥機を用いて完全に乾燥し、その重量値から前記菌体触媒懸濁液の濃度を算出した。既知濃度となった菌体触媒懸濁液を適当な複数の濃度に希釈し、分光光度計を用いて室温において透過光度(600nm)を測定し、該分光光度計での該菌体触媒の検量線を作成した。以後、該分光光度計の指示値から任意の該菌体触媒懸濁液の乾燥菌体触媒濃度を算出した。
反応液及び処理液の分析は、以下のごとく実施した。基質であるグリコロニトリル及び生成物であるグリコール酸(アンモニウム)と乳酸(アンモニウム)及び副生成物であるグリコロアミドと乳酸アミドは、高速液体クロマトグラフィーで測定した。カラムはイオン排除カラム(島津Shim-pack SCR-101H)、カラム温度は40℃、移動相はリン酸水溶液(pH=2.3)、流速は0.7cc/min、検出器はUV(島津SPD-10AV vp、210nm)及びRI(島津RID-6A)で実施した。
塩化ナトリウム0.1重量%、リン酸二水素カリウム0.1重量%、硫酸マグネシウム七水和物0.05重量%、硫酸第一鉄七水和物0.005重量%、硫酸アンモニウム0.1重量%、硝酸カリウム0.1重量%硫酸マンガン五水和物0.005重量%を含む培養液250mlを三角フラスコに仕込み、pH7になるように水酸化ナトリウムで調整し、121℃で20分間滅菌した後、アセトニトリル0.5重量%を添加した。これにAcinetobacter sp.AK226を接種して30℃で振とう培養した(前培養)。ミーストパウダー0.3重量%、グルタミン酸ナトリウム0.5重量%、硫酸アンモニウム0.5重量%、リン酸水素二カリウム0.2重量%、リン酸ニ水素カリウム0.15重量%、塩化ナトリウム0.1重量%、硫酸マグネシウム七水和物0.18重量%、塩化マンガン4水和物0.02重量%、塩化カルシウム二水和物0.01重量%、硫酸鉄7水和物0.003重量%、硫酸亜鉛7水和物0.002重量%、硫酸銅5水和物0.002重量%、大豆油2重量%を含む培養液3Lを5Lジャーファーメンターに仕込み、121℃で20分間滅菌した後、前記の前培養液を接種して30℃で通気攪拌を行った。培養開始10時間後から大豆油のフィードを開始した。pH7になるようにリン酸及びアンモニア水でコントロールし、最終的に約5重量%のAcinetobacter sp.AK226懸濁液を得た。更に0.06Mリン酸バッファーを用いて2回洗浄を行い、最終的にリン酸バッファーに懸濁されたAcinetobacter sp.AK226懸濁液(乾燥菌体濃度5重量%)を得た。
前記のように得られたAcinetobacter sp.AK226懸濁液(5.1重量%)1.8gと蒸留水225gを1L四ツ口フラスコに仕込み懸濁させた。該フラスコにpH計と温度計を設置し反応液のpHと温度をモニタリングできるようにして、50℃恒温水槽に入れてスターラー攪拌を実施し、内温が50℃になるまでしばらく保持した。次に原料の55重量%グリコロニトリル水溶液(東京化成製)を、液体クロマトグラフィー用ポンプを用いて0.33g/minでフィードした。原料グリコロニトリル中に安定剤として含まれる硫酸を中和するため、チューブポンプで1.5重量%アンモニア水をフィードした。尚、アンモニア水フィードポンプはpH計による制御で内液pHが6.9±0.1になるようにセットした。反応中は定期的にサンプリングを行い、高速液体クロマトグラフィーでグリコロニトリルとグリコール酸アンモニウム濃度を測定し、定常グリコロニトリル濃度が2重量%以下になるように原料の添加量を調節した。最終的なグリコール酸アンモニウム蓄積濃度は52重量%、基質のグリコロニトリルは検出されなかった。次に得られたグリコール酸アンモニウム塩水溶液を、遠心分離機(クボタ製:高速遠心機7700)を用いて、回転数:10000pm、処理時間:20min、処理温度:4℃で処理し、上澄みを回収後、MF(旭化成ケミカルズ製:PSP-003)を用いて、流速2ml/min、処理温度:30℃で処理して52重量%のグリコール酸アンモニウム塩水溶液を1065g得た。本液は若干の着色を生じていたため、0.83gの市販活性炭(白鷺A:日本エンバイロケミカルズ製)を加えて、室温で45分攪拌した後、デカンテーションで処理液を回収し、着色成分を除去した。グリコール酸アンモニウム濃度52重量%、副生グリコロアミド濃度0.33重量%であった。
本実施例で使用した乳酸アンモニウム塩水溶液は、市販の40%乳酸アンモニウム(和光純薬製)に市販の97%乳酸アミド(和光純薬製)を添加して調製した。乳酸アンモニウム濃度40重量%、乳酸アミド濃度は0.35重量%であった。
52重量%グリコール酸アンモニウム塩水溶液400gを1L四ツ口フラスコに仕込み、側管に温度計と還流器とN2バブリング用キャピラリーを取り付け、中央にスリーワンモーターの撹拌羽根を取り付け、恒温水槽に全体を浸した。N2バブリングを行いながら、内液を70℃に昇温させ、40重量%水酸化ナトリウム水溶液225gを徐々に滴下した。20分間常圧での操作を行った後、徐々に真空ポンプで減圧操作を行い、最終的に140mmHgまで減圧した。その間、N2バブリングは継続し、内温は63℃となった。
実施例1と同様にグリコール酸アンモニウムの脱アンモニア操作を行い、アンモニア濃度0.225[重量%/グリコール酸]、グリコロアミドが検出限界(1重量ppm)以下の37.2重量%粗グリコール酸ナトリウム水溶液583gを得た。
52重量%グリコール酸アンモニウム塩水溶液50gを100mL四ツ口フラスコに仕込み、側管に温度計と還流器を取り付け、中央にスリーワンモーターの撹拌羽根を取り付け、恒温水槽に全体を入れ、内液を40℃、60℃、80℃または100℃まで上昇させ、30重量%の水酸化カルシウムスラリー37.9gを15minかけて滴下した。これにより、グリコール酸アンモニウム塩1molに対して0.55molの水酸化カルシウムを用いたことになる。
比較例として、α-ヒドロキシ酸アンモニウム塩の加熱加水分解により、アンモニアを除去した。
40重量%乳酸アンモニウム塩水溶液50gを100mL四ツ口フラスコに仕込み、側管に温度計と還流器を取り付け、中央にスリーワンモーターの撹拌羽根を取り付け、恒温水槽に全体を浸して、内液を80℃まで昇温させ、30重量%の水酸化カルシウムスラリー25.1gを15minかけて滴下した。これにより、乳酸アンモニウム塩1molに対して0.55molの水酸化カルシウムを用いたことになる。
Claims (20)
- α-ヒドロキシ酸アンモニウム塩水溶液に塩基性金属類を接触させてα-ヒドロキシ酸金属塩を製造する工程であって、溶液中に残存するα-ヒドロキシ酸アミドを500[重量ppm/α-ヒドロキシ酸]以下にする工程(1)と、
前記α-ヒドロキシ酸金属塩を脱塩してα-ヒドロキシ酸を製造する工程(2)と、
を含むα-ヒドロキシ酸の製造方法。 - 前記工程(1)において、溶液中に残存するアンモニア濃度を3[重量%/α-ヒドロキシ酸]以下にする、請求項1に記載のα-ヒドロキシ酸の製造方法。
- 前記工程(1)において、発生したアンモニアを気相部に回収する、請求項1または2に記載のα-ヒドロキシ酸の製造方法。
- 前記アンモニアを気相部に回収するときの温度を、60℃以上とする、請求項3に記載のα-ヒドロキシ酸の製造方法。
- 前記α-ヒドロキシ酸アンモニウム塩水溶液がα-ヒドロキシニトリルの加水分解反応によって得られたものである、請求項1から4のいずれか1項に記載のα-ヒドロキシ酸の製造方法。
- 前記α-ヒドロキシニトリルの加水分解を、ニトリラーゼ、及び/又はニトリルヒドラターゼとアミダーゼの組み合わせによって酵素触媒的に行う、請求項5に記載のα-ヒドロキシ酸の製造方法。
- 前記α-ヒドロキシニトリルの加水分解が、ニトリラーゼによって行われる、請求項5に記載のα-ヒドロキシ酸の製造方法。
- 前記ニトリラーゼが、Acinetobacter属由来である、請求項6または7に記載のα-ヒドロキシ酸の製造方法。
- 前記ニトリラーゼが、Acinetobacter sp.AK226由来である、請求項8に記載のα-ヒドロキシ酸の製造方法。
- 前記塩基性金属類が、アルカリ金属類、ベリリウムまたはマグネシウムの水酸化物、酸化物及び炭酸塩からなる群から選択される一種以上であって、前記工程(2)において、前記α-ヒドロキシ酸金属塩をイオン交換法によって脱塩する、請求項1から9のいずれか1項に記載のα-ヒドロキシ酸の製造方法。
- 前記塩基性金属類が、アルカリ金属類、ベリリウム、またはマグネシウムの水酸化物、酸化物及び炭酸塩からなる群から選択される一種以上であって、且つ、前記工程(2)において、前記α-ヒドロキシ酸金属塩を電気透析法によって脱塩する、請求項1から9のいずれか1項に記載のα-ヒドロキシ酸の製造方法。
- 前記塩基性金属類が、カルシウム、ストロンチウム、バリウムまたはラジウムの水酸化物、酸化物及び炭酸塩からなる群から選択される一種以上であって、前記工程(2)において、前記α-ヒドロキシ酸金属塩を、硫酸を添加することによって脱塩する、請求項1から9のいずれか1項に記載のα-ヒドロキシ酸の製造方法。
- 前記工程(1)の後、固液分離により固体のα-ヒドロキシ酸金属塩を回収、洗浄し、前記工程(2)において、該固体のα-ヒドロキシ酸金属塩又はこれに水を加えたα-ヒドロキシ酸金属塩スラリーに硫酸を添加する、請求項12に記載のα-ヒドロキシ酸の製造方法。
- 前記塩基性金属類が、水酸化カルシウム、酸化カルシウム及び炭酸カルシウムからなる群から選択される一種以上である、請求項12又は13に記載のα-ヒドロキシ酸の製造方法。
- 前記工程(2)の後、アニオン交換樹脂によって不純物アニオンを除去する工程と、カチオン交換樹脂によって不純物カチオンを除去する工程と、をさらに含む、請求項1から14のいずれか1項に記載のα-ヒドロキシ酸の製造方法。
- 前記不純物カチオンが、副生α-アミノ酸またはイミノジアルキル酸を含む、請求項15に記載のα-ヒドロキシ酸の製造方法。
- 前記α-ヒドロキシ酸が、乳酸またはグリコール酸である、請求項1から16のいずれか1項に記載のα-ヒドロキシ酸の製造方法。
- 前記α-ヒドロキシ酸が、グリコール酸である、請求項17に記載のα-ヒドロキシ酸の製造方法。
- 請求項1から18のいずれか1項に記載の方法で得られたα-ヒドロキシ酸の水溶液を原料としてα-ヒドロキシ酸オリゴマーを合成する工程と、前記α-ヒドロキシ酸オリゴマーを解重合させてその環状二量体エステルを得る工程と、を含む、環状二量体エステルの製造方法。
- 請求項19に記載の方法で得られた環状二量体エステルを原料として開環重合反応でポリα-ヒドロキシ酸を得る工程を含む、ポリα-ヒドロキシ酸の製造方法。
