本发明的目的是为了克服现有技术中的不足, 提供一种从沙棘中制备含 有天然维生素 P的混合物的制备方法。
本发明的另一目的在于提供一种含有天然维生素 P的混合物。
本发明的一种从沙棘中制备含有天然维生素 P的混合物的制备方法,它包 括如下步骤: .
( 1 )取汁: 沙棘果冲洗后以 3500 4500转 /分钟的转速离心 5 ~ 10分钟制 得沙棘汁;
( 2 )脱气:将上述所得沙棘汁在真空度为 5.13 ~ 5.33Pa,温度为 41 ~ 43 °C 的条件下脱气至搅拌状态下无明显气泡;
( 3 )酶解: 在脱完气的沙棘汁中加 95%的氢氧化钠溶液调 pH至 3 ~ 4, 在 膜分离式酶解反应器中按 2 ~ 4克 /升的量加果胶酶酶解 2 ~ 6小时;
( 4 )脱脂: 酶解后的沙棘汁通入碟片式脱脂机脱脂, 其脂肪含量控制在 0.2 ~ 0.5%;
(5)均质: 脱脂后的沙棘汁在温度为 26~40°C的条件下, 用高压均质机: 分两次均质, 第一次在 14~ 16.4MPa压力下均质 3 ~ 5小时, 第二次在 9~.' 11.8MPa压力下均质 0.5~3小时;
(6) 降酸: 再按沙棘汁重量的 1~ 5%加碳酸钙通过离子交换树脂后, 再 加 10 ~ 25ppm浓度的乳酸菌将酸度降到 7 ~ 10克 /升;
(7)脱糖: 将沙棘汁升温至 20~30°C时, 按 0.005 -0.015%加葡萄糖氧 化酶、 以每小时 0.2 ~ 0.5%的量加 5 - 10%双氧水反应时间 1 ~ 3小时脱糖;
(8)微孔过滤: 在操作压力 3-14 MPa下釆用乙酸纤维膜过滤;
(9)冷冻干燥: 待真空度达到 22~26Pa后继续抽真空 1~2小时, 温度调 至 -40 ~ -45 °C时将沙棘汁冷冻干燥 15 ~ 26小时;
( 10)粉碎: 将冷冻干燥所得的含有维生素 P的混合物通过低温粉碎机在 -20~-30°C时粉碎 2~8分钟, 粉碎细度至 38~58um。
其中, 所述脱气优选为将沙棘汁在真空度为 5.2~5.3Pa, 温度为 42°C的 条件下脱气。

( 1 )取汁: 将 100公斤新鲜沙棘果冲洗后以 3500转 /分钟的转速离心 10分 钟制得沙棘汁;
( 2 )脱气: 取上述所得沙棘汁 100kg在真空度为 5.13Pa, 温度为 43°C的 条件下脱气至搅拌状态下无明显气泡;
( 3 ) 酶解: 在脱完气的沙棘汁中加 95%的氢氧化钠溶液调 pH至 3 , 在膜 分离式酶解反应器中按 2克 /升的量加果胶酶酶解 6小时;
( 4 )脱脂: 酶解后的沙棘汁通入碟片式脱脂机脱脂, 其脂肪含量控制在 0.2%;
(5)均质: 脱脂后的沙棘汁在温度为 26°C的条件下, 用高压均质机分两 次均质第一次在 14MPa压力下均质 3小时, 第二次在 11.8MPa压力下均质 3小 时;

(9)冷冻干燥: 待真空度达到 26Pa后继续抽真空 1小时, 温度调至 -45°C 时将沙棘汁冷冻干燥 15小时;
( 10)粉碎: 将冷冻干燥所得的含有维生素 P的混合物通过低温粉碎机在 -30°C时粉碎 2分钟, 粉碎细度至 45um;

( 6 ) 降酸: 再按沙棘汁重量的 3%加碳酸钙通过离子交换树脂 (D-92大 孔弱碱性苯乙烯系阴离子交换树脂、 D*70A均孔均粒凝胶型苯乙烯系强碱树 脂、 D-66酒类精制专用大孔强酸树脂按 1:1 :1比例比例混合树脂)后, 再加 18ppm浓度的足球菌将酸度降到 8克 /升;
( 7 )脱糖: 将沙棘汁升温至 25°C , 按 0.01%加葡萄糖氧化酶、 以每小时 0.35%的量加 7.5%双氧水反应时间 2小时脱糖;
( 8 )微孔过滤: 在操作压力 8MPa下釆用乙酸纤维膜(0.5um )使沙棘汁 中的蛋白质大分子、 杂质及细菌被截留, 低分子可溶性物质则可随滤液通过;
( 9 )冷冻干燥: 待真空度达到 23.5 Pa后继续抽真空 1.4小时, 温度调至 -43 °C时将沙棘汁冷冻干燥 20小时;
( 10 )粉碎: 将冷冻干燥所得的含有维生素 P的混合物通过低温粉碎机在 -25°C时粉碎 6分钟, 粉碎细度至 45um;
( 11 )成品: 釆用铝箔真空包装制得含有 10%维生素 P (其中总黄酮 65%、 花青素 15%、 儿茶素 20% ), 20%维生素 C和 70%果粉的混合物 538g。 工业实用性
本发明提供一种从沙棘中制备含有天然维生素 P的混合物的制备方法。其 经取汁、 脱气、 酶解、 脱脂、 均质、 降酸、 脱糖、 微孔过滤、 冷冻干燥和粉 碎, 制得含有 0.5 ~ 20%维生素?和2 ~ 30%维生素 C的混合物, 该方法简单, 釆 用低温生物物理相结合的方法解决了沙棘中高活性物质因高温氧化而褐变的 问题, 工艺中不需任何溶剂, 无残留, 成本低, 生物活性物质得率高, 产品 中维生素 P与维生素 C共存, 促进人体吸收, 能够减少血管脆性, 降低血管通 透性, 增强维生素 C的活性, 增强免疫、 抗氧化, 预防脑溢血、 视网膜出血、 紫癜等疾病的物质, 也将为高档天然药品和保健品提供优质的原料。

Claims

权 利 要 求 书
1、 一种从沙棘中制备含有天然维生素 P的混合物的方法, 其特征在于, 包括如下步骤:
(1)取汁: 沙棘果冲洗后以 3500~4500转 /分钟的转速离心 5~ 10分钟制 得沙棘汁;
( 2 )脱气:将上述所得沙棘汁在真空度为 5.13 ~ 5.33Pa,温度为 41 ~ 43 °C 的条件下脱气至搅拌状态下无明显气泡;
(3)酶解: 在脱完气的沙棘汁中加 95%的氢氧化纳溶液调 pH至 3 -4, 在 膜分离式酶解反应器中按 2 ~ 4克 /升的量加果胶酶酶解 2 ~ 6小时;
(4)脱脂: 酶解后的沙棘汁通入碟片式脱脂机脱脂, 其脂肪含量控制在 0.2 ~ 0.5%;
(5)均质: 脱脂后的沙棘汁在温度为 26~40°C的条件下, 用高压均质机 分两次均质, 第一次在 14~ 16.4MPa压力下均质 3 ~ 5小时, 第二次在 9~ 11.8MPa压力下均质 0.5 ~ 3小时;
(6) 降酸: 再按沙棘汁重量的 1 ~ 5%加碳酸钙通过离子交换树脂后, 再 加 10~25ppm浓度的乳酸菌将酸度降到 7~10克 /升;
(7)脱糖: 将沙棘汁升温至 20~30°C时, 按 0.005 ~ 0.015%加葡萄糖氧 化酶、 以每小时 0.2 -0.5%的量加 5 ~ 10%双氧水反应时间 1~ 3小时脱糖;
(8)微孔过滤: 在操作压力 3 ~ 14 MPa下釆用乙酸纤维膜过滤;
(9)冷冻干燥: 待真空度达到 22~26Pa后继续抽真空 1 2小时, 温度调 至 _40 ~ -45 °C时将沙棘汁冷冻干燥 15 ~ 26小时;
( 10)粉碎: 将冷冻干燥所得的含有维生素 p的混合物通过低温粉碎机在 -20~-30°C时粉碎 2~8分钟, 粉碎细度至 38~58um。
2、 如权利要求 1所述的方法, 其特征在于, 所述脱气为将沙棘汁在真空 度为 5.2~5.3Pa, 温度为 42'C的条件下脱气。
3、 如权利要求 1或 2所述的方法, 其特征在于, 所述离子交换树脂为阴离 子交换树脂。 4、 如权利要求 1-3任意一项所述的方法, 其特征在于, 所述乙酸纤维膜 孔径为 0.3 ~ 0.7um 。
5、 如权利要求 1-4任意一项所述的方法, 其特征在于, 所述微孔过滤的 操作压力为 8 ~ 10MPa。
6、 如权利要求 1-5任意一项所述的方法, 其特征在于, 所述冷冻干燥是 在真空度达为 24 ~ 25 Pa后继续抽真空 1.5小时, 温度调至 -42〜- 43Ό时将沙棘 汁冷冻干燥 18 ~ 20小时。
7、 如权利要求 1-6任意一项所述的方法, 其特征在于, 所述粉碎为将冷 冻干燥所得的含有维生素 P的混合物通过低温粉碎机在 -25 ~ -28Ό时粉碎 5分 钟, 粉碎细度至 45um。
8、 一种由权利要求 1-7任意一项所述方法制备的含有天然维生素 P的混 合物, 其特征在于, 其包含维生素 P、 维生素 C和果粉。
9、 根据权利要求 8所述含有天然维生素 P的混合物, 其特征在于, 其组分 含量为: 0.5 ~ 20%维生素?、 2 - 30%维生素 C, 其余为果粉。
10、 根据权利要求 8或 9所述含有天然维生素 P的混合物, 其特征在于, 其 中, 所述维生素 P包含总黄酮 60 ~ 80%、 花青素 12 ~ 30%和儿茶素 3 ~ 20%。
