G I P分泌抑制剤
技術分野
[0001] 本発明は、 医薬または食品として有用な G I P分泌抑制剤に関する。
背景技術
[0002] 現代の食生活は、 1 960年当時からの摂取カロリーはほぼ横ばいながら 、 摂取される脂質の比率は 1. 1割から 2. 7割に増え、 大きく変化してい る (国民栄養調査) 。 このように高脂肪食は胃に負担をかけ、 胃もたれ等を 弓 Iき起こすとされている。
[0003] G a s t r i c i n h i b i t o r y p o l y p e p t i d e ( I P) は、 胃酸分泌抑制作用や胃運動抑制作用を有することが知られている消 化管ホルモンであり、 摂食時、 食餌中の脂質等によりその分泌が亢進される ことが知られている (非特許文献 1 ~3) 。 このため、 G I Pの分泌を少な くすることは、 消化促進や胃もたれの改善に有用であると考えられている。 そして、 これまでの研究によって、 G I Pの機能を阻害する物質として 3 _ ブロモ _5_メチル _2_フエ二ルビラゾロ [1, 5_a] ピリミジン _7 —オール (BMP P) が知られ、 G I Pの分泌を抑制する物質としてグァガ ム等が知られている (特許文献 1、 非特許文献 4〜9) 。 しかしながら、 前 者の BMP Pは、 i n V i v oにおける G I P機能阻害効果が確認されて おらず、 また後者のグァガム等は脂質摂取時の G I P分泌抑制効果が検討さ れていないという問題があり、 その上、 胃もたれ改善効果等の点で必ずしも 十分なものとはいえない。 さらに、 日々摂取しても安全性に優れたものであ ることが望まれる。
[0004] —方、 モノァシルグリセロール (以下 MAGともいう) は、 食品分野で乳 化剤等として広く用いられる安全性にすぐれた物質であり、 マーガリン、 乳 飲料、 アイスクリーム、 パン等に通常 0. 2~0. 5%程度配合されている (非特許文献 1 0、 1 1 ) 。 また、 モノァシルグリセロールは、 食後の血中
トリグリセリ ドの上昇抑制作用 (特許文献 2) を有することが知られている しかしながら、 モノァシルグリセロールと G I P分泌との関係については 知られていなかった。

発明の開示
[0005] すなわち本発明は、 下記の 1 ) 〜7 ) の発明に係るものである。
1 ) モノァシルグリセロールを有効成分とする食後 G I P分泌抑制剤。
2 ) モノァシルグリセロールを摂取又は投与することを特徴とする、 食後 G I P分泌抑制方法。
3 ) モノァシルグリセロールを摂取又は投与することを特徴とする、 消化 促進方法。
4 ) モノァシルグリセロールを摂取又は投与することを特徴とする、 胃も たれの改善方法。
5 ) モノァシルグリセロールの食後 G I P分泌抑制剤としての使用。
6 ) モノァシルグリセロールの消化促進剤としての使用。
7 ) モノァシルグリセロールの胃もたれ改善剤としての使用。
発明の詳細な説明
[0006] 本発明は、 医薬または食品として有用な G I P分泌抑制剤を提供すること に関する。
[0007] 本発明者等は、 モノァシルグリセロールが、 食後の G I P分泌を著しく抑 制し、 消化促進や胃もたれ改善に有用であることを見出した。
[0008] 本発明の G I P分泌抑制剤を用いれば、 食後の G I Pを減少させることが でき、 消化吸収を促進することができ、 胃もたれの改善等胃の状態の改善を 図ることができる。
[0009] モノァシルグリセロールとしては、 グリセリンの 1位の水酸基が脂肪酸で エステル化されたもの ( 1—モノアシルグリセロール) 、 2位の水酸基が脂 肪酸でエステル化されたもの (2—モノアシルグリセロール) 及び 3位の水 酸基が脂肪酸でエステル化されたもの (3—モノアシルグリセロール) が挙 げられるが、 1 _モノァシルグリセロールが好ましい。 脂肪酸残基の炭素数 に特に制限はないが、 8 ~ 2 4、 特に 1 6 ~ 2 2が好ましい。 脂肪酸残基と しては、 飽和のもの及び不飽和のものが挙げられ、 具体的には、 力プリル酸 、 力プリン酸、 ラウリル酸、 ミリスチン酸、 パルミチン酸、 ステアリン酸、
ォレイン酸、 リノール酸、 リノレイン酸、 エイコサペンタエン酸、 ドコサへ キサェン酸由来のァシル基、 或いはこれらの混合物や、 これらの酸を含有す る牛脂、 豚脂等の動物油、 パー厶油、 菜種油、 大豆油、 サフラワー油、 トウ モロコシ油、 シソ油、 トウハゼ油、 アマ二油、 エノ油等の植物油から誘導さ れる脂肪酸等由来のァシル基が挙げられる。 これらのモノァシルグリセロー ルは一種又は二種以上を用いることができる。
[0010] なお、 かかるモノァシルグリセロールを混合物として用いる場合、 不飽和 脂肪酸残基の量は、 全脂肪酸残基の 5 5 %以上であることが好ましく、 更に は 7 0 %以上、 特に 9 0 %以上が好ましい。 更に不飽和脂肪酸がォレイン酸 1 5〜8 5 %、 リノール酸 1 5〜 8 5 %で構成されることが好ましい。 更に 不飽和脂肪酸がォレイン酸 5 0〜 1 0 0 %で構成されることが最も好ましい 。 なお、 構成脂肪酸含量は、 構成ァシル基を脂肪酸に換算して算出する。
[001 1 ] 本発明で使用するモノアシルグリセロールは、 不飽和ァシル基を含有する アマ二油、 エゴマ油、 シソ油、 大豆油、 ナタネ油等の加水分解反応、 これら 各種油脂とグリセリンとのエステル交換反応、 かかる油脂由来の脂肪酸とグ リセリンとのエステル化反応等任意の方法により得ることができる。 反応方 法は、 アルカリ触媒等を用いた化学反応法、 リパーゼ等の酵素を用いた生化 学反応法のいずれでもよい。 得られた反応生成物を分画して所期のモノァシ ルグリセ口一ルを単離することができる。
[0012] 後記実施例に示すように、 モノァシルグリセロールは、 トリァシルグリセ ロール (以下 T A Gともいう) と共に摂取した場合に、 グルコース及びトリ ァシルグリセロールの摂取による G I P分泌量の増加を有意に抑制する作用 を有する。 従って、 モノァシルグリセロールは、 食後 G I P分泌抑制剤とし て、 ヒト若しくは動物用の食品若しくは医薬品又はこれらの素材となり得る なお、 「食後の G I P分泌抑制」 とは、 脂質及び糖質を含む食事、 特に脂 質を多く含む食事、 そのなかでもトリアシルグリセロールを多く含む食事を 摂取することにより消化管からの G I P分泌量が増えるのを抑制することを
いう。
[0013] 本発明の食後 G I P分泌抑制剤は、 本発明のモノァシルグリセロールを単 体でヒト及び動物に投与できる他、 各種飲食品、 医薬品、 ぺットフード等に 配合して摂取することができる。 食品又は飲料としては、 モノァシルグリセ ロールを含有し、 消化促進または胃もたれの改善効果を有することを特徴と し、 消化促進または胃もたれの改善のために用いられるものである旨の表示 を付した飲料及び食品として使用することができる。 また、 胃酸分泌促進、 消化促進、 胃もたれ改善等胃の状態の改善のために用いられるものである旨 の表示を付した美容食品、 病者用食品、 特定保健用食品等の食品又は飲料に 応用できる。 医薬品として使用する場合は、 例えば、 錠剤、 顆粒剤等の経口 用固形製剤や、 内服液剤、 シロップ剤等の経口用液体製剤とすることができ る。
[0014] なお、 経口用固形製剤を調製する場合には、 本発明のモノァシルグリセ口 一ルに賦形剤、 必要に応じて結合剤、 崩壊剤、 滑沢剤、 着色剤、 矯味剤、 矯 臭剤等を加えた後、 常法により錠剤、 被覆錠剤、 顆粒剤、 散剤、 カプセル剤 等を製造することができる。 また、 経口用液体製剤を調製する場合は、 矯味 剤、 緩衝剤、 安定化剤、 矯味剤等を加えて常法により内服液剤、 シロップ剤 、 エリキシル剤等を製造することができる。
上記各剤中のモノァシルグリセロールの配合量は、 全組成物中通常 0 . 1 質量0 /0以上、 好ましくは 1質量0 /0以上、 さらに好ましくは 5重量%以上であ る。
[0015] 上記各剤中の有効投与 (摂取) 量は、 モノァシルグリセロールとして、 1 日当たり 0 . 0 1〜 1 0 gとするのが好ましい。 また、 本発明の食後 G I P 分泌抑制剤は、 食前■食中■食後に用いると効果的である。
[001 6] 実施例 1 モノアシルグリセ口ールの G I P分泌抑制作用
トリァシルグリセロール (T A G ) としてトリオレイン、 モノァシルグリ セロール (M A G ) として 1 _モノォレインを用い、 下記の実験を行った。
マウス (C57 B LZ6 J雄、 8週令) を 1群 1 2または 1 4匹とし、 グ ルコース 2m gZg体重のみ、 及びさらにトリオレイン 2mgZg体重を 0 . 02mgZg体重卵黄レシチンにより乳化させたもの (それぞれダルコ一 ス, TAG 1 ) 、 または TAG 1の乳化物にそれぞれ 0. 08, 0. 2, 0 . 4mgZg体重の 1 _モノォレイン (それぞれ MAG 1, MAG 2, MA G3) を添加したものを、 ゾンデにより経口投与した。 乳化物の組成を表 1 に示す。 1 0分後、 腹部大静脈より採血し、 血中 G I Pを、 E L I SA法 ( (j a s t r i c I n h i b i t o r y P e p t i d e t I A k i t , P h o e n i x P h a r ma c e u t i c a l I n c. ) により測定 した。 乳化剤を投与しなかったマウスの血中 G I P値を初期値と仮定して、 1 0分後のマウス血中の G I P增加量を表 2に示す。

量を統計学的に有意に濃度依存的に抑制することができ、 G I P分泌抑制効 果を示すことがわかった。 具体的には、 例えば、 TAGに対して 5分の 1の MAGを摂取したマウスでは、 MAG無添加の場合に比べて血中 G I Pの増 加量が低く、 血中への G I P分泌抑制効果が認められることがわかる。
