Definitions
- the present invention relates to a novel 2-viranone derivative, a pharmacologically acceptable salt thereof, and a therapeutic agent for thrombocytopenia containing the same as an active ingredient. Furthermore, the present invention relates to their synthetic intermediates and their production methods.
- Thrombocytopenia is a disease caused by abnormalities of the immune system or metastatic tumors that damage the bone marrow, tuberculosis, leukemia, or other causes, such as the use of chemicals or radiation therapy. It is a serious disease that can lead to death and even death.
- cytoleukins such as interleukin-16, interleukin-11, and leukocyte migration inhibitory factor (LIF) have a thrombocytopenic effect (Ishibashi et al., Blood, 74: 1241-1244. 1989; Asano et al., Blood, 75: 1602-1605. 1990; J. Okada et al., Department of Hematology and Oncology, 22: 23-31, 1991).
- the production of these cytokins is regulated and controlled by various cells in vivo, and when administered from outside the body, the regulation is out of balance, resulting in severe injury such as liver injury. Causes serious side effects.
- thrombopoietin a protein called thrombopoietin (TP 0) is a factor that increases megakaryocytes and platelets (eg, Sauvage et al., Ature, 369: 533-538. 1994). No effect has been confirmed.
- derivatives such as muramyl dipeptide are known as low-molecular compounds that increase platelet count (Nakajima et al., Arzneim. -Forsch./Drug Res. 41: 60-65, 1989). These derivatives are thought to activate platelets by activating monocytes and macrophages and producing interleukin-6. Only However, it has been reported that the administration of derivatives such as muramyl dipeptide simultaneously causes other physiological activities based on macrophage activation, resulting in side effects such as fever (Journal of the Japan Society of Medical Radiology, 48 (4) : 514, 1988).
- An object of the present invention is to overcome the above-mentioned drawbacks of the prior art, and to provide a compound that is safe and has an additive action on platelets themselves.
- the present inventors have conducted intensive studies on various compounds in order to solve the above problems, and found that a novel 2-pyranone derivative has a platelet-increasing effect in mice and has low toxicity. Completed the invention.
