Abstract

Винахід належить до синтетичних лідерних пептидних послідовностей для секреції поліпептидів в дріжджах. ДНК експресуюча касета, причому включає послідовність 5'-P-SP-LS-PS-“ген”-(T)i-3', де З означає промоторну послідовність; SP означає ДНК послідовність, що кодує сигнальний пептид; LS означає ДНК послідовність, що кодує лідерний пептид загальної формули I: GlnProIle(Asp/Glu) (Asp/Glu)X1(Glu/Asp)X2Asnz(Thr/Ser)X3 , (I), де X1 означає пептидний зв'язок або кодовану амінокислоту, X2 означає пептидний зв'язок або кодовану амінокислоту, або послідовність до 4 однакових або різних кодованих амінокислот; Ζ означає кодовану амінокислоту окрім Pro, і X3 означає послідовність від 4 до 30 однакових або різних кодованих амінокислот; PS означає ДНК послідовність, що кодує процесинговий сайт; “ген” означає ДНК послідовність, що кодує поліпептид; Т означає термінаторну послідовність, і і дорівнює 0 або 1.
