Abstract
1. Генетически созданное антитело против клетки-мишени, узнающее антиген CD138, которое включает антигенсвязывающую область (ABR) против CD138, выделенную из антитела, отличного от человеческого, и ! дополнительную область антитела, по меньшей мере часть которой выделена из человеческого антитела, при этом генетически созданное антитело против клетки-мишени ! (а) связывается с антигеном CD138 со сродством связывания, превышающим сродство связывания указанного антитела, отличного от человеческого; и/или ! (b) обеспечивает гомогенное связывание с антигеном CD138 CD138-экспрессирующих клеток. ! 2. Генетически созданное антитело по п.1, в котором указанная дополнительная область антитела является по меньшей мере одной константной областью, включающей константную область тяжелой цепи или ее часть, выделенную из человеческого антитела, при этом указанное генетически созданное антитело относится к изотипу IgG4. ! 3. Генетически созданное антитело по п.2, которое связывается с антигеном CD138 со сродством связывания, превышающим сродство связывания указанного антитела, отличного от человеческого. ! 4. Генетически созданное антитело по п.2, которое обеспечивает гомогенное связывание с антигеном CD138 CD138-экспрессирующих клеток. ! 5. Генетически созданное антитело по п.4, которое связывается с антигеном CD138 с изменчивостью направленного воздействия менее 150%, 140%, 130%, 120%, 110%, 100%, 90%, 80%, 70%, 60% или 50%. ! 6. Генетически созданное антитело по п.1 или 2, в котором антигенсвязывающая область (ABR) включает ! (а) CDR3 вариабельной области тяжелой цепи, включающую аминокислотные остатки 99-111 SEQ ID NO: 1, и ! (b) CDR3 вариабельной области легкой цепи, включающую аминокислот�
Claims (24)
1. Генетически созданное антитело против клетки-мишени, узнающее антиген CD138, которое включает антигенсвязывающую область (ABR) против CD138, выделенную из антитела, отличного от человеческого, и
дополнительную область антитела, по меньшей мере часть которой выделена из человеческого антитела, при этом генетически созданное антитело против клетки-мишени
(а) связывается с антигеном CD138 со сродством связывания, превышающим сродство связывания указанного антитела, отличного от человеческого; и/или
(b) обеспечивает гомогенное связывание с антигеном CD138 CD138-экспрессирующих клеток.
2. Генетически созданное антитело по п.1, в котором указанная дополнительная область антитела является по меньшей мере одной константной областью, включающей константную область тяжелой цепи или ее часть, выделенную из человеческого антитела, при этом указанное генетически созданное антитело относится к изотипу IgG4.
3. Генетически созданное антитело по п.2, которое связывается с антигеном CD138 со сродством связывания, превышающим сродство связывания указанного антитела, отличного от человеческого.
4. Генетически созданное антитело по п.2, которое обеспечивает гомогенное связывание с антигеном CD138 CD138-экспрессирующих клеток.
5. Генетически созданное антитело по п.4, которое связывается с антигеном CD138 с изменчивостью направленного воздействия менее 150%, 140%, 130%, 120%, 110%, 100%, 90%, 80%, 70%, 60% или 50%.
6. Генетически созданное антитело по п.1 или 2, в котором антигенсвязывающая область (ABR) включает
(а) CDR3 вариабельной области тяжелой цепи, включающую аминокислотные остатки 99-111 SEQ ID NO: 1, и
(b) CDR3 вариабельной области легкой цепи, включающую аминокислотные остатки 89-97 SEQ ID NO: 2 соответственно.
7. Генетически созданное антитело по п.6, в котором антигенсвязывающая область (ABR) далее включает
(а) CDR1 и CDR2 вариабельной области тяжелой цепи, включающие аминокислотные остатки 31-35 и 51-68 SEQ ID NO: 1, и/или
(b) CDR1 и CDR2 вариабельной области легкой цепи, включающие аминокислотные остатки 24-34 и 50-56 SEQ ID NO: 2.
8. Генетически созданное антитело по п.6, в котором указанная дополнительная область антитела включает
(а) аминокислотные остатки 123-448 SEQ ID NO: 1 и/или
(b) аминокислотные остатки 108-214 SEQ ID NO: 2
и их мутации, которые
(i) сохраняют или снижают антителозависимую цитотоксичность и/или комплементзависимую цитотоксичность генетически созданного антитела против клетки-мишени, и/или
(ii) стабилизируют генетически созданное антитело против клетки-мишени.
9. Генетически созданное антитело по п.1 или 2, в котором указанная антигенсвязывающая область указанного антитела, отличного от человеческого, выделена из мышиного антитела.
10. Генетически созданное антитело по п.1 или 2, являющееся химерным антителом, в котором указанное антитело, отличное от человеческого, является антителом В-В4.
11. Генетически созданное антитело по п.3, включающее по меньшей мере одну тяжелую цепь и одну легкую цепь, в котором
(а) последовательность указанной тяжелой цепи по меньшей мере примерно на 70% идентична SEQ ID NO: 1; и/или
(b) последовательность указанной легкой цепи по меньшей мере примерно на 70% идентична SEQ ID NO: 2.
12. Генетически созданное антитело по п.8, в котором
(а) последовательность указанной тяжелой цепи по меньшей мере примерно на 90% идентична SEQ ID NO: 1; и/или
(b) последовательность указанной легкой цепи по меньшей мере примерно на 90% идентична SEQ ID NO: 2.
13. Генетически созданное антитело по п.1 или 2, в котором антигенсвязывающая область (ABR) состоит в основном из антигенсвязывающей области антитела, отличного от человеческого.
14. Фармацевтическая композиция, включающая антитело по п.1 или 2 и фармацевтически приемлемый носитель.
15. Гибридома, которая продуцирует антитело по п.1 или 2.
16. Анализ на основе антитела, включающий антитело по п.1 или 2.
17. Выделенный полипептид, включающий аминокислотную последовательность тяжелой цепи иммуноглобулина или ее части, при этом последовательность указанной тяжелой цепи иммуноглобулина или ее части по меньшей мере на 70%, по меньшей мере на 80%, по меньшей мере на 90%, по меньшей мере на 95% или по меньшей мере на 98% идентична SEQ ID NO: 1, и агент против клетки-мишени, включающий указанную тяжелую цепь иммуноглобулина или ее часть, направленно воздействует на CD138.
18. Выделенный полипептид по п.17, в котором константная область указанной тяжелой цепи иммуноглобулина или ее части является константной областью изотипа IgG4.
19. Выделенный полипептид по п.17, в котором указанный агент против клетки-мишени является химерным антителом ”мышь-человек”.
20. Выделенный полипептид по п.17, который дополнительно включает аминокислотную последовательность легкой цепи иммуноглобулина или ее части, при этом последовательность указанной легкой цепи иммуноглобулина или ее части по меньшей мере на 70%, по меньшей мере на 80%, по меньшей мере на 90%, по меньшей мере на 95% или по меньшей мере на 98% идентичная SEQ ID NO: 2.
21. Выделенный полипептид по п.17, в котором указанная тяжелая цепь иммуноглобулина идентична последовательности SEQ ID NO: 1.
22. Выделенный полинуклеотид по п.20 или 21, в котором указанная легкая цепь иммуноглобулина идентична последовательности SEQ ID NO: 2.
23. Способ гомогенного связывания с CD138, который включает
получение генетически созданного антитела против клетки-мишени по п.1 или 2 и введение указанного генетически созданного антитела против клетки-мишени в CD138-экспрессирующие клетки, при этом указанное генетически созданное антитело против клетки-мишени гомогенно связывается с CD138, экспрессированным на указанных CD138-экспрессирующих клетках.
24. Способ по п.23, в котором указанное генетически созданное антитело против клетки-мишени связывается с CD138 с изменчивостью направленного воздействия менее 150%, 140%, 130%, 120%, 110%, 100%, 90%, 80%, 70%, 60% или 50%.
