Abstract
Nye metoder for nukleinsyreamplifisering ved hjelp av kontinuerlig, hovedsakelig isotermal selvunderholdende sekvensreplikasjon ("3SR") under anvendelse av RNA-avhengig DNA polymeraseaktivitet, DNA-avhengig DNA polymeraseaktivitet, RNAse H aktivitet og DNA-avhengig RNA polymeraseaktivitet. Ved en av metodene, før enzymatiske aktiviteter kan tilveiebringes ved hjelp av bare to enzymer, anvendes en revers transkriptase og en DNA-avhengig RNA polymerase. Metoden kan anvende to eller tre enzymer til å gi de nødvendige enzyma- tiske aktiviteter. I enkelte av metodene anvendes således en eksogen kilde for RNAse H, som E. coli RNAse H, i kombinasjon med en revers transkriptase og en DNA-avhengig RNA polymerase. I andre metodene i samsvar med den foreliggende oppfinnelse anvendes reaksjonsmedia hvori den inherente RNAse H aktivitet av retrovirale reverse transkriptaser er effektiv til å tilveiebringe høye amplifiseringsnivåer, slik at bare to enzymer, revers transkriptase og DNA-avhengig RNA polymerase er nødvendig. Nye blandinger for gjennomføring av metodene i samsvar med oppfinnelsen er også anført.
