Abstract

Det er tilveiebragt avvikende familie B DNA polymeraser uten 3'-> 5' eksonukleaseaktivitet. Disse avvikende polymeraser har anvendbarhet som DNA sekvenseringspolymeraser. Fremgangsmåter for DNA sekvensering med familie B DNA polymeraser og kjedetermineringsnukleotider som ikke tidligere er anvendt for sekvensering er blitt utviklet. Disse fremgangsmåter omfatter bruk av familie B DNA polymeraser som inntil nå ikke har vært kjent å kunne anvendes i DNA sekvensering, slik som avvikende eller villtype former av fag T4 DNA polymerase eller Escherichia coli DNA polymerase II, med nye kombinasjoner av deoksynukleotider og kjedetermineringsnukleotider.
