Description
OPPFINNELSESOMRÅDE
Foreliggende oppfinnelse vedrører blanding bestående av en syntetisk fosfodiester-nukleotidsekvens og anvendelse derav. Blandingen kan anvendes for behandling av cancer.
BAKGRUNN FOR OPPFINNELSEN
Cancer er en abberant netto-akkumulering av atypiske celler som kan skrive seg fra overskudd av proliferasjon, utilstrekkelig celledød eller en kombinasjon av de to.
Proliferasjon er kulmineringen av en celles progresjon gjennom cellesyklusen som resulterer i deling av én celle i to celler. Cellesyklusens fem hovedfaser er G0, Gi, S, G2 og M. Under Go-fasen er cellene i hvile. De fleste celler i kroppen er til enhver tid på dette trinn. Under Gi-fasen produserer celler som respons på signaler om deling, RNA og de nødvendige proteiner for DNA-syntese. Under S-fasen (SE, tidlig S-fase; SM, middel S-fase; og SL, sen S-fase) replikerer cellene deres DNA. Under G2-fasen dannes proteiner som forberedelse for celledeling. Under den mitotiske (M) fase deler cellen seg i to datterceller. Endringer i cellesyklus-progresjonen forekommer ved alle cancere og kan være resultat av overekspresjon av gener, mutasjon av regulatoriske gener eller opphevelse av kontrollpunkter for DNA-skade (Hochhauser, D. Anti-Cancer Chemotherapeutic Agents 8:903, 1997).
Til forskjell fra cancerceller, kan de fleste normale celler ikke proliferere uendelig som følge av en prosess betegnet cellulær aldring. Cellulær aldring er en programmert celledød-respons som fører til vekststans av celler (Dimri et al., Proe. Nati. Acad. Sei. USA 92:20, 1995). DNA-skade, eksponering av kolon-, bryst- og ovariecancerceller for topoisomerase-inhibitorer og eksponering av nasofaryngeale cancerceller for cisplatin, er rapportert å forhindre proliferasjon av disse cellene ved induksjon av aldring (Wang et al., Cancer Res. 58:5019,1998; Poele et al., Br. J. Cancer 80:9, 1999).
Syntetiske oligonukleotider er polyanioniske sekvenser som er internalisert i celler (Vlassov et al., Biochim. Biophys. Acta 1197:95, 1994). Det er rapportert om syntetiske oligonukleotider som binder selektivt til nukleinsyrer (Wagner, R. Nature, 372:333, 1994), til spesifikke celleproteiner (Bates et al., J. Biol. Chem. 274:26369, 1999) og til spesifikke nukleære proteiner (Scaggiante et al., Eur. J. Biochem. 252:207, 1998) til å inhibere proliferasjon av cancerceller.
Syntetiske 27-basesekvenser som inneholder guanin (G) og ulike mengder tymin (T) (oligonukleotid GTn), hvor n er >1 eller <7 og hvor baseantallet er >20 (Scaggiante et al., Eur. J. Biochem. 252:207, 1998) er rapportert å inhibere vekst av cancercellelinjer ved sekvensspesifikk binding til et 45 kDa nukleært protein, mens GTn, hvor baseantallet er <20, er rapportert å være inaktive overfor cancercellelinjer (Morassutti et al., Nucleosides and Nucleotides 18:1711, 1999). To syntetiske GT-rike oligonukleotider på 15 og 29 baser med 3' aminoalkyl-modifikasjoner er rapportert å danne G-kvartetter som binder til nukleolin, og til å inhibere proliferasjon av cancercellelinjer (Bates et al., J. Biol. Chem. 274:26369, 1999). Den syntetiske 6-base TTAGGG-fosfortioat som har en sekvens identisk med den for pattedyrs telomer repetisjonssekvens, er rapportert å inhibere proliferasjon av Burkitts lymfomceller in vitro og in vivo (Mata et al., Toxicol. Applied Pharmacol. 144:189, 1997). Den syntetiske 6-base TTAGGG-fosfodiesteren er imidlertid rapportert å ikke ha anti-telomeraseaktivitet (US-patent 5.643.890).
Celledød bevirkes av immun-mediatorer som fremmer apoptose, og av apoptose-indusere som direkte initierer veier som fører til celledød (Muzio et al., Cell 85:817, 1996; Levine, A. Cell 88:323, 1997). Apoptose er en aktiv celledødprosess som kjennetegnes ved distinkte morfologiske endringer som inkluderer kondensasjon av nukleært kromatin, cellekrymping, kjerne-desintegrasjon, plasmamembran-blebbing og dannelse av membran-bundne apoptotiske legemer (Wyllie et al., Int. Rev. Cytol. 68:251,1980). Et molekylært kjennetegn på apoptose er nedbrytning av cellens nukleære DNA til oligonukleosomal-lange fragmenter som resultat av aktivering av endogen endonuklease (Wyllie, A. Nature 284:555, 1980).
Kaspaser (cystein-aspartyl-spesifikke proteaser) har vært trukket inn som nøkkelenzymer ved gjennomføringen av det sene apoptosetrinn. Kaspase-familien består av minst 14 beslektede cystein-aspartyl-proteaser. Alle kaspasene inneholder et konservert QACXG (hvor X er R, Q eller G) pentapeptid som aktivsete-motiv (Cohen, G. Biochim. Biophys. Acta 1366:139,1997). En rekke kaspaser syntetiseres som inaktive pro-enzymer som aktiveres etter spaltning ved kaspase-spesifikke spaltningsseter (Cohen, G. Biochim. Biophys. Acta 1366:139, 1997) eller som inaktive enzymer som fordrer assosiasjon med regulatoriske molekyler for aktivering (Stennicke et al., J. Biol. Chem. 274:8359, 1999).
I tillegg til deres rolle ved apoptose, er kaspaser involvert i aktivering og proliferasjon av B- og T-lymfocytter, i cytokin-modning under inflammasjon, ved differensiering av progenitorceller under erytropoiese og ved utvikling av linsefiber (Fadeel et al., Leukemia 14:1514, 2000). Med hensyn til B- og T-lymfocytter, bearbeides kaspase 3 under aktivering av B-lymfocytter og av CD4(+), CD8(+), CD45RA(+) og CD45RO(+) subsett av T-lymfocytter (Alam et al., J. Exp. Med. 190:1879,1999). Stimulering av T-lymfocytter med mitogenerog med interleukin-2 er dessuten assosiert med aktivering av kaspase-veien og med spaltning av PARP (Wilheim et al., Eur. J. Immunol. 28:891, 1998). Med hensyn til cytokiner, er kaspase 3-aktivitet nødvendig for frigjøringen av IL-2 av aktiverte T-lymfocytter (Posmantur et al., Exp. Cell Res. 244:302, 1998) og for bearbeidningen og modningen av det pro-inflammatoriske cytokin interleukin-16 (Zhang et al., J. Biol. Chem. 273:1144, 1998). Med hensyn til erytropoiese, er kaspaseaktivering involvert i erytropoieseregulering og har vært vist å modulere GATA-1, et nukleært regulatorisk protein som er avgjørende for modningen av erytroidforløpere (De Maria, et al., Nature 401:489, 1999).
Cytolyse er den fullstendige eller partielle nedbrytning av en celle og den medieres av immunsystemet. Aktiverte makrofager og monocytter produserer bio-aktive molekyler som innbefatter, men ikke er begrenset til, cytokiner. Cytokiner innbefatter, men er ikke begrenset til, interleukin (IL)-1, IL-1 beta, IL-6, IL-10, IL-12 og TNF-alfa. IL-1 beta reduserer benmargcellers følsomhet for cytoreduktive medikamenter, for bestråling og for in vitro tumorcelle-spyling med medikamenter ved autolog benmargtransplantasjon (Dalmau et al., Bone Marrow Transplant. 12:551, 1993). IL-6 induserer B-celledifferensiering, stimulerer IgG-sekresjon (Taga et al., J. Exp. Med. 166:967,1987), induserer cytotoksisk T-celledifferensiering (Lee et al., Vaccine 17:490, 1999), fremmer megakaryocyttmodning (Ishibashi et al., Proe. Nati. Acad. Sei. USA 86:8953, 1989) og virker både som en anti-proliferativ faktor (Mori et al., Biochem. Biophys. Res. Comm. 257:609,1999; Alexandroff et al., Biochem. Soc. Trans. 25:270, 1997; Takizawa et al., Cancer Res. 53:18, 1993; Novick et al., Cytokine 4:6, 1992) og som en pro-proliferativ faktor (Okamoto et al., Cancer Res. 57:141, 1997; Okamoto et al., Int. J. Cancer 72:149, 1997; Chiu et al., Clin. Cancer Res. 2:215, 1996; Lu et al., Clin. Cancer Res. 2:1417, 1996) for cancerceller. IL-10 øker effektiviteten av vaksiner i murine tumormodeller (Kauffman et al., J. Immunother. 22:489, 1999) og oppregulerer anti-cancer autoreaktive T-celle-responser (Alleva et al., Immunobiol. 192:155, 1995). IL-12, alene og i kombinasjon med andre cytokiner, fremmer modningen av leukocytter og induserer sekresjonen av interferon-gamma. IL-12 er rapportert å ha anti-canceraktivitet (Stine et al., Annals NY Academy of Science 795:420, 1996; Chen et al., Journal of Immunol. 159:351, 1997) som inkluderer, men ikke begrenset til, aktivering av spesifikke cytolytiske T-lymfocytter, aktivering av naturlige dreper-(NK) celler og induksjon av de anti-angiogene proteinene IP-10 og MiG.
TNF-alfa forårsaker nekrose av solide tumorer (Porter et al., Trends in Biotech. 9:158,1991), sensibiliserer cancerceller for gammastråle-indusert apoptose (Kimura et al., Cancer Res. 59:1606, 1999) og beskytter benmarg-forløperceller fra virkningene av antineoplastiske midler (Dalmau et al., Bone Marrow Transplant. 12:551, 1993).
De fleste kjente anti-cancer terapier har imidlertid enten de er rettet mot induksjon av cellesyklusarrest, inhibering av proliferasjon, induksjon av apoptose eller stimulering av immunsystemet, vist seg å være mindre enn tilstrekkelig for kliniske anvendelser. Mange av disse terapiene er ineffektive eller toksiske, har vesentlige uheldige bivirkninger, resulterer i utvikling av medikamentresistens eller immunsensibilisering og er svekkende for mottageren.
Det er derfor et stadig behov for nye blandinger og fremgangsmåter som induserer cellesyklusarrest i cancerceller, inhiberer proliferasjon av cancerceller, aktiverer kaspaser i cancerceller, induserer apoptose i cancerceller og stimulerer cytokinproduksjon i immunsystemceller.
SAMMENFATNING AV OPPFINNELSEN
Foreliggende oppfinnelse oppfyller disse behov ved å tilveiebringe en blanding kjennetegnet ved at den omfatter en isolert 3'-OH, 5'-OH syntetisk fosfodiester-nukleotidsekvens som består av SEKV. ID. NR:45 og videre omfatter en farmasøytisk akseptabel bærer. Blandingen ifølge oppfinnelsen er videre kjennetegnet ved at den ytterligere består av et kjemoterapeutisk middel.
Det kjemoterapeutiske midlet er valgt fra gruppen bestående av en antimetabolitt, et alkyleringsmiddel og en hormon-antagonist.
Foreliggende oppfinnelse vedrører videre anvendelse av en blanding ifølge et hvilket som helst av kravene 1 til 3, for fremstilling av et medikament for å indusere
en respons valgt fra gruppen bestående av induksjon av cellesyklusarrest, inhibering av proliferasjon, aktivering av kaspaser og induksjon av apoptose i cancercellene, og produksjon av cytokiner av immunsystemceller, hvor cytokinene er valgt fra gruppen bestående av IL-1-beta, IL-6, IL-10, IL-12 og TNF-alfa.
Foreliggende oppfinnelse vedrører videre anvendelse av sammensetningen ifølge et hvilket som helst av kravene 1 til 3 for fremstilling av et medikament for behandling av cancer. Canceren er valgt fra gruppen bestående av et primært carcinom, et sekundært carcinom, et primært sarcom og et sekundært sarcom. Canceren er videre valgt fra gruppen bestående av leukemi, lymfom, bryst, prostata, colorektal, ovarie og bencancer.
Disse og andre formål, trekk og fordeler ved foreliggende oppfinnelse vil bli åpenbare etter gjennomgang av den etterfølgende detaljerte beskrivelse av den beskrevne utførelsesform og de tilhørende patentkrav.

DETALJERT BESKRIVELSE AV OPPFINNELSEN
En blanding som omfatter en sekvens og en farmasøytisk akseptabel bærer kan administreres til et dyr, inklusivt et menneske, som har cancer, i en effektiv mengde til behandling av cancer hos dyret, inklusivt mennesket. Den uventede og overraskende evne sekvensen har til å indusere cellesyklusarrest, inhibere proliferasjon, indusere apoptose og aktivere kaspaser i cancerceller og til å stimulere cytokinproduksjon av immunsystemceller, løser et lenge følt, uoppnådd behov på det medisinske området og fører til et betydelig gode for dyr, inklusivt mennesket.
Vendingene «terapeutisk behandling» og «effektiv mengde » angir en mengde av en sekvens som er effektiv til å indusere cellesyklusarrest, inhibere proliferasjon, aktivere kaspaser og indusere apoptose i cancerceller og stimulere cytokinproduksjon av immunsystemceller.
Vendingene «suspensjon tumormodell» og «solide tumormodeller» angir primære eller sekundære karsinomer eller sarkomer.

Administrering av en effektiv mengde av en sekvens heri til et dyr, inklusivt et menneske, er en terapeutisk behandling som forhindrer, behandler eller eliminerer en sykdom så som cancer. Den terapeutiske effekt av en sekvens kan økes ved hjelp av metoder som innbefatter, men ikke er er begrenset til, kjemisk modifisering av base-, sukker- eller fosfatskjelettet, kjemisk supplering eller bioteknologisk amplifisering av sekvensene ved å benytte bakterielle plasmider som inneholder de riktige sekvenser, kompleksering av sekvensene til biologiske eller kjemiske bærere eller kobling av sekvensene til vevstype- eller celletype-rettede ligander eller antistoffer.
Blandinger som omfatter én eller flere sekvenser og en farmasøytisk akseptabel bærer, fremstilles ved jevnt og intimt å bringe sammen sekvensen og den farmasøytisk akseptable bærer. Farmasøytisk akseptable bærere innbefatter flytende bærere, faste bærere eller begge deler. Flytende bærere er vandige bærere, ikke-vandige bærere eller begge deler og innbefatter, men er ikke begrenset til, vandige suspensjoner, oljeemulsjoner, vann-i-olje-emulsjoner, vann-i-olje-i-vann-emulsjoner, stedsspesifikke emulsjoner, emulsjoner med lang oppholdstid, klebrige emulsjoner, mikroemulsjoner og nanoemulsjoner. Faste bærere er biologiske bærere, kjemiske bærere eller begge deler og innbefatter, men er ikke begrenset til, virale vektorsystemer, partikler, mikropartikler, nanopartikler, mikrokuler, nanokuler, minipumper, bakterielle celleveggekstrakter og bionedbrytbare eller ikke bionedbryt bare naturlige eller syntetiske polymerer som muliggjør forsinket frigjøring av sekvensene. Metoder benyttet til å kompleksbinde en sekvens til en fast bærer innbefatter, men er ikke begrenset til, direkte adsorpsjon til overflaten av den faste bæreren, kovalent kobling til overflaten av den faste bæreren, enten direkte eller via en sammenkoblende del; og kovalent kobling til polymeren benyttet til å fremstille den faste bæreren. Eventuelt kan en sekvens stabiliseres ved tilsetning av ikke-ioniske eller ioniske polymerer, som f.eks. polyoksyetylensorbitan-monooleater (Tweens) eller hyaluronsyre.

Administrasjonsmåter innbefatter, men er ikke begrenset til, peroral, topisk, subkutan, transdermal, subdermal, intramuskulær, intraperitoneal, intravesikulær, intraartikulær, intraarteriell, intravenøs, intradermal, intrakranial, intralesjonal, intratumoral, intraokulær, intrapulmonal, intraspinal, intraprostatisk administrering, anbringelse i kroppshulrom, nasal inhalasjon, pulmonal inhalasjon, impresjon i hud og elektroporering.
Avhengig av administrasjonsmåte utgjør volumet per dose fortrinnsvis ca. 0,001 til 100 ml_ per dose, mer foretrukket ca. 0,01 til 50 ml_ per dose og mest foretrukket ca. 0,1 til 30 ml_ per dose. Fortrinnsvis utgjør den sekvensmengde som administreres per dose fra ca. 0,001 til 100 mg/kg, mer foretrukket fra ca. 0,01 til 10 mg/kg og mest foretrukket fra ca. 0,1 til 5 mg/kg. Blandingen omfattende sekvensen eller sekvensen pluss et terapeutisk middel, kan administreres ved en enkelt dosebehandling, ved flerdosebehandlinger eller etter en kontinuerlig infusjonsplan og over et tidsrom egnet for sykdommen som behandles, mottagerens tilstand og administrasjonsmåten. Den kan dessuten administreres før, samtidig med eller etter administrering av det terapeutiske middel.
Blandingen i kombinasjon med kjemoterapeutisk middel kan administreres til et dyr som har cancer, i en mengde som effektivt potenserer den antineoplastiske effekt av det kjemoterapeutiske middel. Fortrinnsvis utgjør den mengde terapeutisk middel som administreres per dose, fra ca. 0,001 til 1000 mg/m<2>eller fra ca. 0,01 til 1000 mg/kg, mer foretrukket fra ca. 0,01 til 500 mg/m<2>eller fra ca. 0,01 til 500 mg/kg og mest foretrukket fra ca. 0,1 til 100 mg/m<2>eller fra ca. 0,1 til 100 mg/kg. Blandingen og det eventuelt bestemte terapeutiske middel som administreres, mengden per dose, doseringsplanen og administrasjonsmåten bør bestemmes av brukeren ved å benytte metoder kjent for fagmannen, og vil avhenge av cancertype, alvoret av canceren, lokaliseringen av canceren og andre kliniske faktorer, så som mottagerens størrelse, vekt og fysiske tilstand. Det kan dessuten eventuelt benyttes in vitro tester for å bidra til identifisering av optimale områder for administrering og for administrering av blanding pluss terapeutisk middel.
Uten noe ønske om å være bundet av den følgende hypotese, antas det at blandingene ifølge foreliggende oppfinnelse danner en ny familie av strukturer og at de ikke virker som antisense RNA, antisense DNA, trippelheliks-dannende DNA, telomerase-inhibitorer, transkripsjons-aktivatorer eller transkripsjons-inhibitorer.
De etterfølgende eksempler vil tjene til ytterligere å illustrere foreliggende oppfinnelse.
Eksempel 1
Fremstilling av sekvenser
Fosfodiester- og fosfortioatsekvenser ble fremstillet av HUKABEL Scientific Ltd., (Montréal, Québec, Canada) ved å benytte EXPEDITE™ 8900 automatisert DNA-syntesesystem (PersSeptive Biosystems, Inc., Farmingham, MA) og av Sigma-Genosys (Woodlands, TX) ved å benytte Abacus Segmented Synthesis Technology. Om intet annet er angitt var anvendte sekvenser fosfodiestersekvenser. Om intet annet er angitt ble sekvensene umiddelbart før bruk dispergert i autoklavert deionisert vann eller i en autoklavert farmasøytisk akseptabel buffer, som f.eks., men ikke begrenset til, saltvann.
Eksempel 2
Celler og reagenser
Alle cellelinjer ble anskaffet fra the American Type Culture Collection (ATCC, Rockville, MD) og ble dyrket i det mediet som er anbefalt av ATCC. Tabell 1 viser cellelinjer, deres opprinnelse og deres egenskaper.
Celler ble utsådd i 6 (1 mL/brønn), 24 (0,5 mL/brønn) eller 96 (0,1 mL/brønn) flatbunnede mikroplater og ble holdt ved 37°C i en 5% C02-atmosfære. Om intet annet er angitt, ble 2,5 x 10<5>celler/ml_ inkubert med 0 |ig/ml_ (kontroll) og 100 jig/mL (behandlet) av 2- til 45-basesekvenser inneholdende A, C, G og T i 48 timer.
Eksempel 3
Måling av celleproliferasjon
Celleproliferasjon ble målt ved å benytte dimetyltiazol-difenyl-tetrazolium (MTT) reduksjon (Mosman et al., J. Immunol. Methods 65:55, 1983). MTT ble målt ved en bølgelengde på 570 nm under bruk av en multiplate spektrometerleser (ELX800, Bio-TEK Instruments, Inc., Winooski, VT).
Eksempel 4
Inhibering av Jurkat human leukemi T-celleproliferasjon
Jurkat human leukemi T-celler er en atypisk multi-medikament resistent human suspensjon tumorcellemodell. Jurkatceller ble inkubert med 27-base GT- og CT-sekvenser (Tabell 2).
Som vist i Tabell 2, ble Jurkat T-celleproliferasjon inhibert med de testede GT-sekvensene, men ikke med den testede CT-sekvens.
Jurkat T-celler ble inkubert med 3-, 6-, 9-, 12-, 14-, 15-, 18-, 21- og 24-base GT-sekvenser (Tabell 3).
Som vist i Tabell 3, inhiberte 3-, 6-, 9-, 12-, 14-, 15- og 18-basesekvenser Jurkat T-celleproliferasjon like effektivt som 21- og 24-base GT-sekvenser.
Jurkat T-celler ble inkubert med 6-base GT-, AG-, CG-, GG-, AGT- og CGT-sekvenser (Tabell 4).
Som vist i Tabell 4, inhiberte 6-base GT-sekvenser Jurkat T-celleproliferasjon. GT SEKV. ID. NR:25 inhiberte proliferasjon 60% og AGT SEKV. ID. NR:49 inhiberte proliferasjon 45%. Sammenligning av den relative styrke av GT SEKV. ID. NR:25 og AGT SEKV. ID. NR:49 ved bruk av PHARM/PCS-4 programvare (Microcomputer Specialists, Philadelphia, PA) viste at styrken av GT SEKV. ID. NR:25 var 3,4 ganger den for AGT SEKV. ID. NR:49. AGT SEKV. ID. NR:49-fosfortioat er rapportert å inhibere telomeraseaktivitet og til å indusere apoptose i Burkitts lymfomceller (Mata et al., Toxicol. Appl. Pharmacol. 144:189, 1997).
For å bestemme telomeraseaktivitet, ble ekstrakter fra 2 x 105 Jurkat T-celler analysert ved å benytte den PCR-telomere repetisjons-amplifikasjonsprotokoll
(TRAP) (Roche, Laval, Québec, Canada). Ved konsentrasjoner mellom 1 og 100 ng/mL, oppviste GT SEKV. ID. NR:25-fosfodiester mellom 0 og 10% anti-telomeraseaktivitet, mens AGT SEKV. ID. NR:49-fosfortioat oppviste mellom 30 og 75% anti-telomeraseaktivitet. Verken GT SEKV. ID. NR:25-fosfortioat eller GT SEKV.
ID. NR:49-fosfodiester oppviste noen anti-telomeraseaktivitet.
Jurkat T-celler ble inkubert med 2-, 3-, 4-, 5-, 6- og 7-base GT-sekvenser (Tabell 5).

HL-60 promyelocytiske leukemiceller er en p53 mutert human suspensjon tumormodell. HL-60-celler ble inkubert med 6-base GT-sekvenser (Tabell 6).
Som vist i Tabell 6, inhiberte 6-base GT-sekvenser HL-60 celleproliferasjon.

Inhibering av MCF-7 human brystcancer celleproliferasjon
MCF-7 humane brystcancerceller er en kaspase-3-negativ, østrogenavhengig human modell på solid tumor. MCF-7-celler (5 x 10<5>celler/mL) ble inkubert med 3-og 6-base GT-sekvenser (Tabell 7).
Som vist i Tabell 7, inhiberte 6- og 7-base GT-sekvenser MCF-7 celleproliferasjon.

Inhibering av PC-3 human prostatacancer celleproliferasjon
PC-3 prostatacancerceller er en p53 mutert, androgen-uavhengig human solid tumormodell. PC-3-celler (5 x 10<5>celler/mL) ble inkubert med 3- og 6-base GT-sekvenser (Tabell 8).
Som vist i Tabell 8, inhiberte 3- og 6-base GT-sekvenser PC-3 celleproliferasjon.

Inhibering av LNCaP human prostatacancer celleproliferasjon
LNCaP prostatacancerceller er en TGF-beta 1 reseptor-negativ, androgen-uavhengig, metastatisk human solid tumormodell. LNCaP-celler (5 x 10<5>celler/mL) ble inkubert med 6-base GT-sekvenser (Tabell 9).
Som vist i Tabell 9, inhiberte 6-base GT-sekvenser LNCaP celleproliferasjon.

THP-1 akutt monocytisk leukemiceller er en p53 null human suspensjon tumormodell. THP-1-celler (1,6 x 106 celler/mL) ble inkubert med 6-base GT-sekvenser (Tabell 10).

OVCAR-3 ovariecancerceller er en p53 mutert, p21/waf-1/Cip deletert, metastatisk human solid tumormodell. OVCAR-3-celler (5 x 105 celler/ml_) ble inkubert med 2-, 6- og 18-base GT-sekvenser og med en 6-base AGT-sekvens (Tabell 11).
Som vist i Tabell 11, inhiberte 2-, 6- og 18-base GT-sekvenser og en 6-base AGT-sekvens OVCAR-3 celleproliferasjon.

Eksempel 12
Inhibering av celleproliferasjon med fosfodiester- og fosfortioatsekvenser
Modifikasjon av naturlige fosfodiestersekvenser ved å benytte et svovelatom i stedet for et ikke-brodannende oksygenatom på én eller flere av fosfatgruppene har vært rapportert å øke stabiliteten av oligonukleotidsekvenser til endonukleaser i biologiske væsker og celler (Crooke et al. Anticancer Drugs 6:609, 1991).
Jurkat humane leukemi T-celler (Tabell 13), LNCaP humane prostatacancerceller (5 x 10<5>celler/mL) (Tabell 14) og MCF-7 humane brystcancerceller (5 x 10<5>celler/mL) (Tabell 15) ble inkubert med 6-base GT-sekvenser som enten hadde oksygen (fosfodiester) eller svovel (fosfortioat) som et ikke-brodannende atom på fosfatgruppen.
Som vist i Tabell 13, 14 og 15, inhiberte 6-base GT-fosfodiestersekvenser Jurkat T, LNCaP og MCF-7 celleproliferasjon mer effektivt enn 6-base GT-fosfortioatsekvenser.
Eksempel 13
Inhibering av celleproliferasjon med blandede fosfodiester- og fosfortioatsekvenser
Jurkat human leukemi T-celler (Tabell 16) og MCF-7 humane brystcancerceller (5 x 105 celler/mL) (Tabell 17) ble inkubert med 6-base GT SEKV. ID. NR:25, hvor enten oksygen (fosfodiester) eller svovel (fosfortioat) var det ikke-brodannende atom på fosfatgruppen.
Som vist i Tabell 16 og 17 resulterte anvendelse av et svovelatom i stedet for et ikke-brodannende oksygenatom på én eller flere fosfatgrupper av 6-base GT SEKV. ID. NR:25, i en signifikant minsket inhibering av Jurkat T og MCF-7 celleproliferasjon.
Eksempel 14
Inhibering av murin cancercelleproliferasjon
EL-4 murine lymfom T-celler er en suspensjon tumormodell. EL-4 murine lymfom T-celler ble inkubert med 6-, 18-, 27- og 33-base GT-sekvenser og med en 15-base ACG-sekvens (Tabell 18).
Som vist i Tabell 18, inhiberte ikke 6-, 18-, 27- og 33-base GT-sekvenser og en 15-base ACG-sekvens EL-4 murin celleproliferasjon.
A20 murin leukemi B-celler er en suspensjon tumormodell. A20 murine leukemi B-celler ble inkubert med 6-base GT-sekvenser (Tabell 19).
Som vist i Tabell 19, inhiberte 6-base GT-sekvenser proliferasjon av A20 murine B-leukemiceller.
Eksempel 15
Inhibering av hunde-cancercelleproliferasjon
D-17 hunde-osteosarkomceller og CF-51 hunde-brystkjertel-cancerceller er modeller på solide tumorer. D-17 hunde-osteosarkomceller (5 x 10<5>celler/ml_)
(Tabell 20) og CF-51 hunde-brystkjertel-cancerceller (5 x 10<5>celler/mL (Tabell 21) ble inkubert med 6-, 9-, 17-, 27- og 33-base GT-sekvenser og med en 15-base ACG-sekvens.
Som vist i Tabell 20 og 21, inhiberte 6-, 9-, 17-, 27- og 33-base GT-sekvenser og en 15-base ACG-sekvens både D-17 og CF-51 hunde-celleproliferasjon.

Inhibering av cancercelleproliferasjon

10,0 ng/ml_ 5-fluoruracil eller Cisplatin (Tabell 24). 5-fluoruracil er en anti-metabolitt som interfererer med DNA- og RNA-syntese. Cisplatin er et alkyleringsmiddel som krysskobler DNA og inhiberer DNA-forløpere.
Som vist i Tabell 24, potenserte 6-base GT SEKV. ID. NR:25 og 27-base GT SEKV. ID. NR:1 den antineoplastiske effekt av 0,1 og 1,0 jig/mL 5-fluoruracil på Jurkat T-celleproliferasjon og GT SEKV. ID. NR:25 potenserte den antineoplastiske effekt av 0,1 og 1,0 ng/ml_ Cisplatin på Jurkat T-celleproliferasjon.
MCF-7 humane brystcancerceller (5 x 10<5>celler/ml_) ble inkubert med
1,0 ng/mL av 6-base GT SEKV. ID. NR:25 og av 27-base GT SEKV. ID. NR:1 i nærvær av 0, 0,1,1,0 eller 10,0 ng/ml_ 5-fluoruracil eller tamoxifen (Tabell 25). Tamoxifen er en østrogen-antagonist.
Som vist i Tabell 25, potenserte 6-base SEKV. ID. NR:25 den antineoplastiske effekt av 0,1 ng/mL 5-fluoruracil og av 0,1 ng/mL tamoxifen på MCF-7 celleproliferasjon. 27-base SEKV. ID. NR:1 potenserte ikke den antineoplastiske aktivitet av 5-fluoruracil eller av tamoxifen på MCF-7 celleproliferasjon.

Inhibering av proliferasjon av Bacillus Calmette- Guerin (BCG) avledede sekvenser BCG-avledede sekvenser er rapportert å inhibere tumorvekst in vivo (Kataoka et al., Jpn. J. Cancer Res. 83:244, 1992). Dessuten er A-2 (SEKV. ID. NR:72) og BCG A-4 (SEKV. ID. NR:74) når de ble forblandet med IMC-celler og injisert i CDF-1 mus, rapportert å inhibere IMC-tumorvekst med henholdsvis 88% og 37%.
Jurkat human leukemi T-celler ble inkubert med 45-basesekvenser avledet fra BCG (Tabell 27).

Eksempel 21
Induksjon av cellesyklusarrest
Cellesyklustilstand ble bestemt ved å benytte et kommersielt sett CYCLETEST™ PLUS DNA kit (Becton Dickinson). I korte trekk ble cellekjerner opp-nådd ved å løse cellemembranen i et ikke-ionisk overflateaktivt middel, eliminere cellens cytoskjelett og nukleære proteiner med trypsin, oppslutte den cellulære RNA med RNase og stabilisere det nukleære kromatin med spermin. Propidiumjodid ble tilsatt til cellekjernene, og deres fluorescens ble analysert i et strømningscytometer forsynt med mulighet for elektronisk dublettdiskriminering (FACSCalibur, Becton Dickinson, San Jose, CA). Akkumulering av celler i G0/G1, tidlig S (SE), midt S (SM), sen S (SL) eller G2/M-fasene av cellesyklusen ble analysert ved å benytte MODFIT LT programvare (Verity Software House, Inc, Topsham, MA).
Eksponensielt voksende Jurkat human leukemi T-celler (Tabell 28) og MCF-7 humane brystcancerceller (5 x 10<5>celler/mL) (Tabell 29) ble inkubert i 24 timer med 2-, 6-, 15-, 18-, 27- og 45-basesekvenser inneholdende A, C, G og T. Cellene ble oppsamlet og sentrifugert og cellesyklustilstanden bestemt.

Omfordeling av plasmamembran-fosfatidylserin og frigjøring av nukleært matriksprotein (NuMA) er karakteristisk for celler som gjennomgår apoptose (Martin et al., J. Exp. Med., 182:1545, 1995; Miller et al., Biotechniques, 15:1042, 1993).
Omfordelingen av fosfatidylserin i plasmamembranen under apoptose ble målt ved strømningscytometri under bruk av FITC-konjugert annexin V (BD Pharmingen, San Diego, CA). NuMA-frigjøring i supernatanten ble bestemt ved å benytte et kommersielt ELISA-sett (Oncogen/Calbiochem, Cambridge, MA).
Jurkat human leukemi T-celler ble inkubert med 50 \ iM av 3-, 4-, 5-, 6- og 7-GT basesekvenser, en 5-base ACGT-sekvens, 6-base AG-, GG-, AGT- og CGT-sekvenser og en 7-base GG-sekvens. Tabell 31 viser % celler i apoptose (positive for fosfatidylserin/annexin V farving (PS/A-V)) og % NuMA-frigjort fra cellene.
Som vist i Tabell 31, induserte 3-, 4-, 5- og 6-base GT-, AG-, CG- og AGT-sekvenser apoptose av Jurkat T-celler. 5-base ACGT- og 6-base CGT- og GG-sekvenser induserte ikke apoptose av Jurkat T-celler.

Økning av intracellulært kalsium (Ca<2+>)i
Økninger i intracellulært kalsium (Ca<2+>)ier rapportert assosiert med apoptose-induksjon (Lam et al., Mol. Endocrinol. 7:686, 1993). (Ca<2+>)ible fulgt ved å benytte fluorescens-proben Fluo-3-AM (Cell Permaant, Molecular Probes, Inc., Eugene, OR). En økning i Fluo-3-AM fluorescens tyder på en økning i (Ca<2+>)i.
Jurkat human leukemi T-celler ble inkubert i 24 timer med 6-base GT SEKV. ID. NR:25. Celler ble oppsamlet ved sentrifugering, suspendert i PBS inneholdende 1% FBS og ladet med 10 \ iM Fluo-3-AM i 1 time ved 37<0>C. Cellefluorescens ble målt ved 488 nm eksitasjon og 530 nm emisjon (FL1 detektor). Data ble analysert på en FACSCALIBUR ved å benytte programmet CellQUEST (Becton Dickinson).
Som vist i Figur 1, forårsaket inkubering av Jurkat T-celler med 6-base GT SEKV. ID. NR:25 en 88% økning i cellefluorescens, hvilket tyder på en økning i (Ca<2+>)i.

Induksjon av apoptose
Apoptose kan initieres av ligander som binder til celleoverflatereseptorer, inklusivt, men ikke begrenset til, Fas (CD95) og tumornekrosefaktor (TNF). Fas-binding til Fas-ligand og TNF-binding til TNF-reseptor 1 initierer intracellulær signalisering som resulterer i aktiveringen av cystein-aspartyl-proteaser (kaspaser). Kaspaser initierer den letale proteolytiske kaskade av apoptose-eksekvering assosiert med nukleær DNA-fragmentering, frigjøring av nukleære matriksproteiner (NuMA) og tap av cellesubstratkontakt.
Jurkat human leukemi T-celler (1 x 10<6>/ml_) ble inkubert med 6- og 27-GT basesekvenser (Tabell 32). NuMA ble bestemt som i Eksempel 23.

Som vist i Tabell 34, inhiberte SEKV. ID. NR:81, 82 og 83 som var rike på GT, proliferasjon av, induserte cellesyklusarrest i, og induserte apoptose av Jurkat T-celler, mens SEKV. ID. NR:15, rik på AC, ikke inhiberte proliferasjon av, induserte ikke cellesyklusarrest i, eller induserte ikke apoptose i Jurkat T-celler.

Modulering av kaspase-aktivering med GT-sekvenser
Kaspaser gjenkjenner tre hovedpeptid-substratsekvenser: 1) Tyr-Val-Ala-Asp (YVAD, kaspase-1, -4 og -5) (SEKV. ID. NR:84); 2) Asp-Glu-Val-Asp (DEVD, kaspase-2, -3 og -7) (SEKV. ID. NR:85); og 3) lle-(Leu)-Glu-X-Asp (l(L)EXD; kaspase-8 og -10) (SEKV. ID. NR:86) (Thornberry et al., J. Biol. Chem. 272:17907, 1997). Sekvens-gjenkjenning i et proteinmål resulterer i en begrenset og spesifikk proteolyse av målet som, i et første eksempel, moduleringen av kaspase 7 aktivering med kaspase 3 eller, som i et annet eksempel, nedbrytningen av strukturelle proteinmål inklusivt, men ikke begrenset til, laminer, eller som i et tredje eksempel, aktiveringen av enzymer inklusivt, men ikke begrenset til, PARP.
NH2-XXXD-COO-GT-sekvens-konstruksjoner utvikles ved kjemisk konjugasjon av en kjemisk beskyttet GT-sekvens eller av en kjemisk beskyttet AC-sekvens til et kjemisk beskyttet peptid valgt fra gruppen bestående av NH2-YVAD-COOH (SEKV. ID. NR:84), NH2-DEVD-COOH (SEKV. ID. NR:85) og NH2-IEGD-COOH (SEKV. ID. NR:87) ved å benytte et oligonukleotid syntetisert med en 5'-C2-amid-avstandsarm og standard amid-karboksyl vannløselige karboheksimid-konjugasjonsteknikker (Guy et al., J. Chromatography, B. Biomed. Sei. Appl. 706:149, 1998). Reaktive karboksylat- og reaktive amingrupper avbeskyttes etter konjugasjon.
Peptid-GT (heretter PGT) sekvenskonstruksjoner inklusivt, men ikke begrenset til, NH2-YVAD-COO-GT; NH2-DEVD-COO-GT og NH2-l(L)EXD-COO-GT, spaltes ved karboksylatfunksjonen mellom D- og GT-sekvensen av enzymer som innbefatter, men ikke er begrenset til, kaspaser, cancermetastase-assosierte enzymer, kollagenaser og metalloproteinaser. Slik spaltning resulterer i frigjøringen av den kaspase-aktiverende GT-sekvens fra PGT. Den resulterende økning i intracellulær kaspase-aktivitet kan for eksempel forsterke den terapeutiske effekt av kjemoterapeutiske midler i multi-medikamentresistente cancerceller eller immun-responsen på svakt antigene stimuli.
For å bestemme kaspase-aktivering blir kontroll og behandlede celler vasket, fiksert, permeabilisert og inkubert med et FITC-konjugert antistoff som gjenkjenner den aktive form av kaspasen (Pharmingen, San Diego, CA) ved å benytte de betingelser som er anbefalt av produsenten. Fluorescens assosiert med aktiv kaspase 3 analyseres ved strømningscytometri på en FACSCALIBUR ved å benytte programmet CellQUEST (Becton Dickinson). Alternativt bestemmes kaspase-aktiveringen kolorimetrisk ved å benytte en test basert på spaltningen av et kaspase-spesifikt peptid konjugert til farverapportør-molekylet p-nitroanilid, som kan kvantifiseres spektrofotometrisk ved en bølgelengde på 405 nm.

Kaspase 3 aktivering ble også bestemt kolorimetrisk som i Eksempel 28. Som vist i Figur 2B var kaspase 3 aktivitet i 33-base GT SEKV. ID. NR:66 og 11-base GT SEKV. ID. NR:81, 82 og 83 behandlede celler 105%, 77%, 100% og 60% høyere enn den i kontrollceller, mens i 15-base ACG SEKV. ID. Aktivering av kaspase-7 og PARP-spaltning med GT SEKV. ID. NR:25
Jurkat T-celle leukemiceller ble inkubert i 72 timer med 6-base GT SEKV. ID. NR:25. Cellene ble vasket 3X med PBS, lysert med 10 mM HEPES, pH 7,5, inneholdende 5 mM MgCb, 1 mM ditiotreitol, 1,5 nM aprotinin, 10 mM leupeptin og 2,5 nm Na-ortovanadat, og proteininnholdet av lysatet ble bestemt (Bradford, J. Anal. Biochem. 72:248, 1976).
Aktivert kaspase 7 og PARP-spaltning ble påvist ved western blotting. Lysatet ble blandet med Laemmli-buffer (Laemmli, U. Nature 15:680, 1970), ristet og oppvarmet til 100°C i 4 minutter. 50 jxg protein ble tilsatt til hver bane, og proteiner ble separert ved elektroforese i en 10% (kaspase) eller en 17% (PARP) natrium-dodekylsulfat-polyakrylamidgel (SDS-PAGE) ved en konstant spenning på 100V i ca. 1,5 timer. De separerte proteiner ble elektroblottet over på en PVDF-membran. Lik protein-lading ble overvåket ved Ponceau-rødt farving av membranen.
Membranen ble blokkert over natten ved 4°C med en buffer inneholdende 1% Tris-buffret saltvann (2 mM Tris-HCI, 13,7 mM NaCI, pH 7,6, og 0,1% polyetylen-sorbitan-monolaurat (TWEEN 20) (TBST) + 5% fettfri tørrmelk. Membranen ble vasket og inkubert i 1 time ved RT med et monoklonalt muse IgG anti-kaspase 7 antistoff (Pharmingen) (fortynnet 1:1000 i TBST + 1% BSA) eller med et monoklonalt muse IgG anti-PARP antistoff (fortynnet 1:1000 i TBST + 1% BSA) (Pharmingen). IgG bundet til kaspase 7 eller til PARP ble påvist med sau anti-mus IgG konjugert til pepperrotperoksydase (fortynnet 1:1000 i TBST + 5% fettfri tørrmelk) (Pharmingen). Blott ble fremkalt ved å benytte et forbedret kjemiluminescens påvisningssystem (ECL, Amersham, Corp., Amersham, UK).
Som vist i Figur 3B, indusert 6-base GT SEKV. ID. NR:25 prosessering av inaktiv pro-kaspase 7 (30 kDa) til aktiv kaspase 7 (19-20 kDa) og aktivt PARP til dets inaktive 85 kDa nedbrytningsprodukt.

NH2-YVAD-CO-GT, NH2-DEVD-CO-GT og NH2-IEGD-COO-GT induserte hver prosessering av inaktiv kaspase 3 til aktiv kaspase 3 og av inaktiv kaspase 7 til aktiv kaspase 7, mens NH2-YVAD-CO-ACG, NH2-DVED-CO-ACG og NH2-IEGD-CO-ACG ikke induserte prosessering av inaktiv pro-kaspase 3 til aktiv kaspase 3 eller av inaktiv kaspase 7 til aktiv kaspase 7.
Uten noe ønske om å bindes av den følgende hypotese, antas det at basal kaspase-aktivitet i kaspase-holdige celler, medierer frigjøringen av kaspase-aktiverende GT-sekvenser fra NH2-XXXD-COO-GT-konstruksjoner ved proteolyse/hydrolyse. Dette resulterer i positiv amplifikasjon av kaspase-aktivitet (økte nivåer av kaspase 3 og kaspase 7) i cellene, av de frigjorte kaspase-aktiverende GT-sekvensene.
Eksempel 33
Induksjon av cytokinproduksjon
Om intet annet er angitt, ble 1 x 10<6>celler inkubert med 100 ng/mL av hver av sekvensene testet i 48 timer ved 37°C i 5% CO2. Produksjon av cytokin IL-6, IL-10, IL-12, IL-1 beta og TNF-alfa ble bestemt i pg/mL i 100 jiL kultursuspernatant ved å benytte den relevante kommersielle ELISA (BioSource, Camarillo, CA). IL-12 ELISA måler både IL-12 p70 kompleks og fri p40-subenhet. Resultater uttrykkes som «ganger» (x) økning i cytokinproduksjon i behandlede celler sammenlignet med kontrollceller.
THP-1 humane akutt monocytisk leukemiceller ble inkubert med 2-, 3-, 6-, 9-, 12-, 14-, 15- og 18-base GT-sekvenser, og produksjon av cytokinet IL-6 ble bestemt.

Stimulering av cytokinsyntese i humane perifere mononukleære blodceller
Perifere mononukleære blodceller (heretter «PBMC») ble isolert fra 7 friske personer ved Ficoll-Hypaque (Amersham Pharmacia Biotech. Baie d'Urfée, Québec, Canada) ved tetthets gradientsentrifugering av helblod. PBMC ble inkubert med 6-base GT-, AGT-, CGT-, AG-, CG- og GG-sekvenser, og produksjon av cytokinene IL-1beta, IL-6, IL-10 og IL-12 ble bestemt.
Som vist i Tabell 41, økte 6-base GT-, AGT-, CTG-, AG-, CG- og GG-sekvenser human PBMC celleproduksjon av cytokinene IL-1 beta, IL-6, IL-10 og IL-12.

Cytokinsyntese med sjimpanse perifere mononukleære blodceller
PBMC ble isolert fra 4 friske sjimpanser som i Eksempel 35. Sjimpanse PBMC ble inkubert med 6-base GT-, AGT-, CGT-, AG-, CG- og GG-sekvenser, og produksjon av cytokinene IL-10, IL-12 og TNF-alfa ble bestemt (Tabell 41).
Som vist i Tabell 41, økte 6-base GT-, AGT-, CGT-, AG-, CG- og GG-sekvenser sjimpanse PBMC celleproduksjon av cytokinene IL-10 og IL-12 og TNF-alfa.

MCF-7 humane brystcancerceller implanteres subkutant som xenografter i nakne BALB/c hunnmus. Musene deles i 6 grupper på 10 mus. På dag 0 får gruppe 1 mus saltvann, gruppe 2 mus får 6-base GT SEKV. ID. NR:25, gruppe 3 mus får 5-fluoruracil, gruppe 4 mus får tamoxifen, gruppe 5 mus får 6-base GT SEKV. ID. NR:25 + 5-fluoruracil og gruppe 6 mus får 6-base GT SEKV. ID.
NR:25 + tamoxifen. Etter 4 ukers behandling avlives musene og tumormassen bestemmes. Gruppe 1 mus har den største tumormasse, gruppe 2, 3 og 4 mus har mindre tumormasse enn gruppe 1 mus og gruppe 5 og 6 mus har mindre tumormasse enn gruppe 1, 2, 3 og 4 mus.
Eksempel 39
Effekt av 3- og 6-base GT-sekvenser og 45-base BCG A-3-sekvens på LNCaP humane prostatacancertumorer
LNCaP humane prostatacancerceller implanteres subkutant, som xenografter, i nakne nu/nu hannmus. Musene fordeles i 5 grupper på 10 mus. På dag 0 får gruppe 1 mus saltvann, gruppe 2 mus får 3-base SEKV. ID. NR:8, gruppe 3 mus får 6-base GT SEKV. ID. NR:25, gruppe 4 mus får 6-base AG SEKV. ID. NR:45 og gruppe 5 mus får 45-base BCG A-3 SEKV. ID. NR:69. Etter 4 ukers behandling avlives musene og tumormassen bestemmes. Gruppe 1 mus har den største tumormasse, gruppe 5 mus har mindre tumormasse enn gruppe 1 mus og gruppe 2, 3 og 4 mus har mindre tumormasse enn gruppe 1 og 5 mus.
Eksempel 41
Effekt av 3-, 6-, 8- og 27-basesekvenser på EL-4 murine T-lymfomer
EL-4 murine T-lymfomceller implanteres i C57/BL6 mus. Musene fordeles på 6 grupper å 10 mus. På dag 0 får gruppe 1 mus saltvann, gruppe 2 mus får 3-base GT SEKV. ID. NR:8, gruppe 3 mus får 6-base SEKV. ID. NR:25, gruppe 4 mus får 6-base AG SEKV. ID. NR:45, gruppe 5 mus får 18-base GT SEKV. ID. NR:18 og gruppe 6 mus får 27-base GT SEKV. ID. NR:1. Etter 4 ukers behandling avlives musene og tumormassen bestemmes. Gruppe 1 mus har den største tumormasse, gruppe 2, 3, 4, 5 og 6 mus har mindre tumormasse enn gruppe 1 mus.
Eksempel 42
Humane koloncancercellelinjer opprettholdes som adherente cellekulturer.
Celler i den eksponensielle vekstfase behandles med 2-20-base GT-, GA-, GC- eller GG-sekvenser i doseringsområdet 0 jig/mL til 100 ng/mL i 24-72 timer. Inhibering av celleproliferasjon måles ved MTT-reduksjon, cellesyklusarrest ved strømningscytometri og apoptose ved annexin-V-binding eller NuMA-frigjøring. GT-, GA-, GC- eller GG-sekvenser inhiberer proliferasjon, induserer cellesyklusarrest og induserer apoptose i koloncancercellelinjene.
SCID-mus som bærer subkutane humane kolorektale cancercellelinjer behandles med saltvann eller med 2-20-base GT-, GA-, GC- eller GG-sekvenser som har anti-canceraktivitet mot humane kolorektale cancercellelinjer in vitro. Mus behandlet med 2-20-base GT-, GA-, GC- eller GG-sekvenser, som har anti-canceraktivitet mot humane kolorektale cancercellelinjer in vitro, oppviser en signifikant reduksjon i tumormasse sammenlignet med mus behandlet med saltvann.
Claims (7)
1. Blanding,karakterisert vedat den omfatter en isolert 3'-OH, 5'-OH syntetisk fosfodiester-nukleotidsekvens som består av SEKV. ID. NR:45 og videre omfatter en farmasøytisk akseptabel bærer.
2. Blanding ifølge krav 1,karakterisert vedat den ytterligere består av et kjemoterapeutisk middel.
3. Blanding ifølge krav 1 eller 2,karakterisert vedat det kjemoterapeutiske middel er valgt fra gruppen bestående av en antimetabolitt, et alkyleringsmiddel og en hormon-antagonist.
4. Anvendelse av en blanding ifølge et hvilket som helst av kravene 1 til 3, for fremstilling av et medikament for å indusere en respons valgt fra gruppen bestående av induksjon av cellesyklusarrest, inhibering av proliferasjon, aktivering av kaspaser og induksjon av apoptose i cancercellene, og produksjon av cytokiner av immunsystemceller, hvor cytokinene er valgt fra gruppen bestående av IL-1-beta, IL-6, IL-10, IL-12 og TNF-alfa.
5. Anvendelse av sammensetningen ifølge et hvilket som helst av kravene 1 til 3 for fremstilling av et medikament for behandling av cancer.
6. Anvendelse ifølge et hvilket som helst av kravene 4 til 5, hvor canceren er valgt fra gruppen bestående av et primært carcinom, et sekundært carcinom, et primært sarcom og et sekundært sarcom.
7. Anvendelse ifølge krav 6, hvor canceren er valgt fra gruppen bestående av leukemi, lymfom, bryst, prostata, colorektal, ovarie og bencancer.
