Oppfinnelsens område
Foreliggende oppfinnelse angår heterocykliske tripeptider, farmasøytisk preparat omfattende slike samt anvendelse av slike for fremstilling av medikament for behandling eller forebygging av sykdom.
Bakgrunn for oppfinnelsen
Hepatitt C virus (HCV) er det etiologiske hovedagens for post-transfusjon og gruppe-tilegnet ikke-A ikke-B hepatitt verden over. Det er beregnet at over 200 millioner mennesker verden over er infisert med viruset. En høy prosentdel av bærere blir kronisk infisert og mange utvikler kronisk leversykdom, såkalt kronisk hepatitt C. Denne gruppen har igjen høy risiko for alvorlig leversykdom så som lever-cirrhose, hepatocellulært karsinom og terminal leversykdom, hvilket fører til død.
Mekanismen hvorved HCV oppnår viral persistens og forårsaker en høy grad av kronisk leversykdom er ikke grundig oppklart. Det er ikke kjent hvorledes HCV interagerer med og unngår vertens immunsystem. I tillegg må rollene for cellulære og humorale immunresponser ved beskyttelse mot HCV- infeksjon og sykdom fortsatt oppklares. Immunoglobuliner er rapportert for forebygging av transfusjon-assosiert viral hepatitt, men Center for Disease Control anbefaler for tiden ikke immunoglobulinbehandlinger for dette formål. Mangelen på en effektiv beskyttende immunrespons hemmer utviklingen av en vaksine eller tilstrekkelige post-eksponeringsforebyggingstiltak, slik at i nær fremtid er forhåpninger sterkt knyttet til antivirus-intervensjoner.
Forskjellige kliniske undersøkelser er utført med det mål å finne fram til farmasøytiske midler som er i stand til effektivt å behandle HCV-infeksjon hos pasienter som er rammet av kronisk hepatitt C. Disse undersøkelser har involvert anvendelse av interferon-alfa, alene og i kombinasjon med andre antivirusmidler. Slike undersøkelser har vist at et vesentlig antall av deltagerne ikke responderer på disse terapier, og blant dem som responderer godt, ble en stor andel funnet å få tilbakefall etter avslutning av behandlingen.
Inntil nylig var interferon (IFN) eneste tilgjengelige terapi med påviselige resultater godkjent klinisk for pasienter med kronisk hepatitt C. Imidlertid er den forlengete responsgrad lav, og interferonbehandling fremkaller også alvorlige bivirkninger (dvs. retinopati, thyreoiditt, akutt pankreatitt, depresjon) som reduserer livskvaliteten til behandlede pasienter. Nylig ble interferon i kombinasjon med ribavirin godkjent for pasienter som ikke var mottagelige for IFN alene. Imidlertid blir bivirkningene forårsaket av IFN ikke avhjulpet med denne kombinasjonsterapien. Pegylerte former av interferoner så som PEG-lntron® og Pegasys® kan tydeligvis delvis hjelpe på disse skadelige bivirkninger, men antivirusmedikamenter forblir fortsatt det aktuelle valg for oral behandling av HCV.
Derfor eksisterer et behov for utvikling av effektive antivirusmidler for behandling av HCV-infeksjon som overvinner begrensningene ved eksisterende farmasøytiske terapier.
HCV er et innkapslet positivt trådformet RNA-virus i Flaviviridae-familien. Det enkeltrådete HCV RNA-genom er omtrent 9500 nukleotider i lengde og har en enkel åpen leseramme (ORF) som koder for et enkelt stort polyprotein på ca. 3000 aminosyrer. I infiserte celler blir dette polyprotein spaltet på multiple seter med cellulære og virale proteaser for å produsere de strukturelle og ikke-strukturelle (NS) proteiner. I tilfellet av HCV blir dannelsen av ferdige ikke-strukturelle proteiner (NS2, NS3, NS4A, NS4B, NS5A og NS5B) utført med to virale proteaser. Det første hittil dårlig karakteriserte spalter på NS2-NS3 skjøten (følgelig referert til som NS2/3 protease); den andre er en serinprotease som sitter innenfor den N-terminale region av NS3 (NS3 protease) og medierer alle de påfølgende spaltninger nedstrøms for NS3, både i cis, på NS3-NS4A-spaltningssetet og i trans, for de gjenværende NS4A-NS4B, NS4B-NS5A, NS5A-NS5B seter. NS4A-proteinet synes å tjene multiple funksjoner, som virker som en kofaktor for NS3-proteasen og muligens hjelper membranlokaliseringen i NS3 og andre viral replikase- komponenter. Kompleksdannelsen av NS3-proteasen med NS4A synes nødvendig for prosesseringsforløpet og forsteker den proteolytisk effektivitet på alle setene. NS3-proteinet oppviser også nukleosid trifosfatase- og RNA helicase-aktiviteter. NS5B er en RNA-avhengig RNA polymerase som er involvert i replikasjonen av HCV.
En generell strategi for utviklingen av antivirusmidler er å inaktivere virus- kodete enzymer som er essensielle for replikasjonen av viruset.

En fordel ved foreliggende oppfinnelse er at den tilveiebringer tripeptid-forbindelser som er anvendlige for fremstilling av medikamenter som er hemmende for NS3-proteasen, et enzym som er essensielt for replikasjonen av hepatitt C virus.
En ytterligere fordel ved ett aspekt ved foreliggende oppfinnelse ligger i det faktum at forbindelsene spesifikt er anvendlige for fremstilling av medikamenter som hemmer NS3-proteasen og ikke viser nevneverdig hemmende aktivitet mot andre serinproteaser så som human leukocytt-elastase (HLE), svinepankreas-elastase (PPE) eller bovint pankreaschymotrypsin eller cysteinproteaser så som humant lever-cathepsin B (Cat B). Videre er forbindelsene aktive i cellekultur og har god farmakokinetisk profil in vivo.

En ytterligere fordel ved foreliggende oppfinnelse er at den tilveiebringer forbindelser som er anvendlige for fremstilling av medikamenter som er oralt biotilgjengelige i pattedyr.

I henhold til én utførelsesform inneholder det farmasøytiske preparatet ifølge foreliggende oppfinnelse videre a-interferon (pegylert eller ikke) eller ribavirin eller ett eller flere andre anti-HCV-midler eller hvilken som helst kombinasjon av dem ovenfor.
Innenfor omfanget av foreliggende oppfinnelse ligger også anvendelse av en forbindelse med formel I som beskrevet her for fremstilling av et medikament for behandling eller forebygging av hepatitt C-virusinfeksjon.
Detaljert beskrivelse av foretrukne utførelsesformer
Definisjoner
Som anvendes her gjelder de følgende definisjoner hvis det ikke på annen måte er angitt i kravene: Med henvisning til tilfellene hvor ( R) eller ( S) blir anvendt for å betegne den absolutte konfigurasjon for et substituent- eller asymmetrisk senter i en forbindelse med formel I, blir betegnelsen anvendt i sammenheng med hele forbindelsen og ikke i sammenheng med substituenten eller det asymmetriske senter alene.
Betegnelsen "P1, P2 og P3" som anvendes her refererer til stillingen til aminosyrerestene ved å starte fra C-terminusenden til peptidanalogene og strekker seg mot N-terminus (dvs. P1 angir stilling 1 fra C-terminus, P2: andre stilling fra C-terminus, etc.) (se Berger A. & Schechter I., Transactions of the Royal Society London series B257. 249-264 (1970)).
Som anvendes heri angir betegnelsen "(1 R, 2S)-vinyl-ACCA" en forbindelse med formelen:

Betegnelsen "(Ci-6)alkyl" som anvendes her, enten alene eller i kombinasjon med en annen substituent, betyr acykliske, lineære eller forgrenete alkylsubstituenter inneholdende fra 1 til 6 karbonatomer og omfatter for eksempel metyl, etyl, propyl, butyl, 1-metyletyl, 1 -metylpropyl, 2-metylpropyl, 1,1-dimetyletyl og heksyl. Tilsvarende betyr betegnelsen "(Ci-8)alkyl" acyklisk, rettkjedet eller forgrenet alkyl inneholdende 1 til 8 karbonatomer, f.eks. oktyl.
Betegnelsen "(C3-7)cykloalkyl" som anvendes her, enten alene eller i kombinasjon med en annen substituent, betyr en cykloalkylsubstituent inneholdende fra 3 til 7 karbonatomer og omfatter cyklopropyl, cyklobutyl, cyklopentyl, cykloheksyl og cykloheptyl.

Slike midler kan velges fra: antivirusmidler, immunomodulerende midler, inhibitorer av HCV NS3-protease, inhibitorer av HCV-polymerase eller inhibitorer av et annet mål i HCV-livssyklusen.
Betegnelsen "immunomodulerende middel" som anvendes her betyr de midler (forbindelser eller biologiske) som effektivt forbedrer eller forsterker immunsystemresponsen til et pattedyr. Immunomodulerende midler omfatter for eksempel klasse I interferoner (så som a-, p-, 5- og ou-interferoner, T-interferoner, konsensus interferoner og asialo-interferoner), klasse II interferoner (så som y-interferoner) og pegylerte interferoner.
Betegnelsen "inhibitor av HCV NS3 protease" som anvendes her betyr et agens (forbindelse eller biologisk) som effektivt hemmer funksjonen av HCV NS3 protease hos et pattedyr. Betegnelsen "inhibitor av HCV-polymerase" som anvendes her betyr et agens (forbindelse eller biologisk) som effektivt hemmer funksjonen av en HCV-polymerase hos et pattedyr. Dette omfatter for eksempel inhibitorer av HCV NS5B-polymerase. Betegnelsen "inhibitor av et annet mål i HCV-livssyklusen" som anvendes her betyr et agens (forbindelse eller biologisk) som effektivt hemmer dannelsen og/eller replikasjonen av HCV hos et pattedyr på annen måte enn ved å hemme funksjonen av HCV NS3-proteasen. Dette omfatter agenser som griper inn i enten vertens eller HCV-virusmekanismer som er nødvendige for dannelsen og/eller replikasjonen av HCV hos et pattedyr. Inhibitorer av et annet mål i HCV-livssyklusen omfatter for eksempel midler som hemmer et mål valgt fra en helicase, en HCV NS2/3-protease og et indre ribosomalt inngangssete (IRES). Et spesifikt eksempel på inhibitorer med et annet mål i HCV-livssyklusen omfatter ISIS-14803 (ISIS Pharmaceuticals).
Betegnelsen "HIV-inhibitor" som anvendes her betyr et agens (forbindelse eller biologisk) som effektivt hemmer dannelsen og/eller replikasjonen av HIV hos et pattedyr. Dette omfatter agenser som griper inn i enten verts- eller virusmekanismer som er nødvendige for dannelsen og/eller replikasjonen av HIV hos et pattedyr. HIV-inhibitorer omfatter for eksempel nukleosidiske inhibitorer, ikke-nukleosidiske inhibitorer, proteaseinhibitorer, fusjonsinhibitorer og integraseinhibitorer.
Betegnelsen "HAV-inhibitor" som anvendes her betyr et agens (forbindelse eller biologisk) som effektivt hemmer dannelsen og/eller replikasjonen av HAV hos et pattedyr. Dette omfatter agenser som griper inn i enten verts- eller virusmekanismer som er nødvendige for dannelsen og/eller replikasjonen av HAV hos et pattedyr. HAV-inhibitorer omfatter Hepatitt A vaksiner, for eksempel Havrix<®>
(GlaxoSmithKline), VAQTA<®> (Merck) og Avaxim<®> (Aventis Pasteur).
Betegnelsen "HBV-inhibitor" som anvendes her betyr et agens (forbindelse eller biologisk) som effektivt hemmer dannelsen og/eller replikasjonen av HBV hos et pattedyr. Dette omfatter agenser som griper inn i enten verts- eller virusmekanismer som er nødvendige for dannelsen og/eller replikasjonen av HBV hos et pattedyr. HBV-inhibitorer omfatter for eksempel midler som hemmer HBV viral DNA-polymerase eller HBV-vaksiner. (Synergy), HepBzyme; og immunomodulatorprodukter så som: interferon alfa 2b, HE2000 (Hollis-Eden), Theradigm (Epimmune), EHT899 (Enzo Biochem), Thymosin alfa-1 (Zadaxin<®>), HBV DNA-vaksine (PowderJect), HBV DNA-vaksine (Jefferon Center), HBV-antigen (OraGen), BayHep B<®> (Bayer), Nabi-HB<®> (Nabi) og Anti-hepatitt B (Cangen); og HBV-vaksineprodukter så som de følgende: Engerix B, Recombivax HB, GenHevac B, Hepacare, Bio-Hep B, TwinRix, Comvax, Hexavac.
Betegnelsen "klasse I interferon" som anvendes her betyr et interferon valgt fra en gruppe interferoner som alle binder til reseptor type I. Dette omfatter både naturlig og syntetisk fremstilte klasse I interferoner. Eksempler på klasse I interferoner omfatter a-, ©-interferoner, x-interferoner, konsensus- interferoner, asialo-interferoner.
Betegnelsen "klasse II interferon" som anvendes her betyr et interferon valgt fra en gruppe interferoner som alle binder til reseptor type II. Eksempler på klasse II interferoner omfatter y-interferoner.
Spesifikk foretrukne eksempler på noen av disse agenser er oppført nedenfor:
■ antivirusmidler: ribavirin og amantadin; ■ immunomodulerende agenser: klasse I interferoner, klasse II interferoner og pegylerte interferoner; ■ inhibitor av et annet mål i HCV-livssyklusen som hemmer et mål valgt fra: NS3-helicase, HCV NS2/3-protease eller indre ribosom-inngangsete (IRES); ■ HIV-inhibitorer: nukleosidiske inhibitorer, ikke-nukleosidiske inhibitorer, proteaseinhibitorer, fusjonsinhibitorer og integraseinhibitorer; eller ■ HBV-inhibitorer: agenser som hemmer HBV virus DNA-polymerase eller er en HBV-vaksine.
Som beskrevet ovenfor er kombinasjonsterapi tiltenkt hvor en forbindelse med formel (1) eller et farmasøytisk akseptabelt salt derav blir samadministrert med minst ett ytterligere agens valgt fra: et antivirusagensmiddel, et immunomodulerende middel, en annen inhibitor av HCV NS3-protease, en inhibitor av HCV-polymerase, en inhibitor for et annet mål i HCV-livssyklusen, en HIV-inhibitor, en HAV-inhibitor og en HBV- inhibitor. Eksempler på slike midler er gitt i definisjonsavsnittene ovenfor. Disse ytterligere midler kan kombineres med forbindelsene ifølge foreliggende oppfinnelse for å skape en enkel farmasøytisk doseform. Alternativt kan disse ytterligere midler administreres separat til pasienten som del av en multippel doseform, for eksempel ved anvendelse av et sett. Slike ytterligere midler kan administreres til pasienten før, samtidig med eller etter administrering av en forbindelse med formel (1) eller et farmasøytisk akseptabelt salt derav.
Som det anvendes her, betyr betegnelsen "behandling" administrering av en forbindelse eller et preparat ifølge foreliggende oppfinnelse for å lindre eller fjerne symptomer på hepatitt C-sykdom og/eller å redusere virusangrep hos en pasient.
Som det anvendes her, betyr betegnelsen "forebygging" administrering av en forbindelse eller preparat ifølge foreliggende oppfinnelse etter eksponering av individet for viruset, men før symptomer på sykdommen opptrer og/eller før deteksjon av viruset i blodet.

I henhold til en annen alternativ utførelsesform kan det farmasøytiske preparatet ifølge foreliggende oppfinnelse i tillegg omfatte en annen inhibitor av HCV NS3-protease.
I henhold til en annen alternativ utførelsesform kan det farmasøytiske preparatet ifølge foreliggende oppfinnelse i tillegg omfatte en inhibitor av HCV-polymerase.
I henhold til enda en annen alternativ utførelsesform kan det farmasøytiske preparatet ifølge foreliggende oppfinnelse i tillegg omfatte en inhibitor for andre mål i HCV-livssyklusen, omfattende helicase, NS2/3-protease eller indre ribosominngangssete (IRES).
Det farmasøytiske preparatet ifølge foreliggende oppfinnelse kan administreres oralt, parenteralt eller via et implantert reservoir. Oral administrering eller administrering ved injeksjon er foretrukket. Det farmasøytiske preparatet ifølge foreliggende oppfinnelse kan inneholde hvilke som helst konvensjonelle ikke-toksiske farmasøytisk akseptable bærere, adjuvanser eller konstituenter. I noen tilfeller kan pH i preparatet reguleres med farmasøytisk akseptable syrer, baser eller buffere for å forbedre stabiliteten til den formulerte forbindelsen eller dens leveringsform. Betegnelsen parenteral som anvendes her omfatter subkutan, intrakutan, intravenøs, intramuskulær, intra-artikulær, intrasynovial, intrasternal, intratekal og intralesjonal injeksjon eller infusjonsteknikker.
Det farmasøytiske preparatet kan foreligge i form av et sterilt injiserbart preparat, for eksempel som en steril injiserbar vandig eller oljeaktig suspensjon. Denne suspensjonen kan formuleres i henhold til teknikker som er kjente på området ved anvendelse av egnete dispergerings- eller fuktemidler (så som for eksempel Tween 80) og oppslemmingsmidler.
Det farmasøytiske preparatet ifølge foreliggende oppfinnelse kan administreres oralt i hvilken som helst oralt akseptabel doseform omfattende kapsler, tabletter og vandige suspensjoner og løsninger. I tilfellet av tabletter for oral anvendelse, omfatter bærere som blir vanlig anvendt laktose og maisstivelse. Smøremidler, så som magnesiumstearat, blir også typisk tilsatt. For oral administrering i en kapsel-form omfatter anvendelige fortynningsmidler laktose og tørket maisstivelse. Når vandige suspensjoner blir administrert oralt, er den aktive bestanddel kombinert med emulgerings- og oppslemmingsmidler. Om ønsket kan visse søtnings-og/eller smaks- og/eller fargemidler tilsettes.
Andre egnete konstituenter eller bærere for de ovenfor nevnte formuleringer og preparater kan finnes i farmasøytiske standardverker, f.eks. i "Remington's Pharmaceutical Sciences", The Science and Practice of Pharmasy, 19<nde> utg. Mack Publishing Company, Easton, Penn., (1995).
Dosenivåer på mellom ca. 0,01 og ca. 100 mg/kg kroppsvekt pr. dag, fortrinnsvis mellom ca. 0,1 og ca. 50 mg/kg kroppsvekt pr. dag av
proteaseninhibitorforbindelse beskrevet heri er anvendelige i en monoterapi for fremstilling av medikament for forebygging og behandling av HCV-betinget sykdom. Typisk vil det farmasøytiske preparatet ifølge foreliggende oppfinnelse administreres fra ca. 1 til ca. 5 ganger pr. dag, eller alternativt som en kontinuerlig infusjon. Slik administrering kan anvendes som en vedvarende eller akutt terapi. Mengden av aktiv bestanddel som kan kombineres med bærermaterialene for å produsere en enkel doseform, vil variere avhengig med verten som blir behandlet og den spesielle administreringsmetode. Et typisk preparat vil inneholde fra ca. 5 vekt-% til ca. 95 vekt-% aktiv forbindelse. Fortrinnsvis inneholder slike preparater fra ca. 20% til ca. 80% aktiv forbindelse.
Som fagfolk vil forstå, kan lavere eller høyere doser enn de som er angitt ovenfor være nødvendige. Spesifikk dose og behandlingsregimer for hvilken som helst spesiell pasient vil avhenge av en rekke faktorer, omfattende aktiviteten til den spesifikke forbindelse som anvendes, alderen, kroppsvekten, den generelle helsestatus, kjønn, diett, tid for administrering, utskillingshastighet, medikamentkombinasjon, alvorlighetsgraden og forløpet av infeksjonen, pasientens disposisjon for infeksjonen og bedømmelsen til den behandlende lege. Generelt blir behandling igangsatt med små doser som hovedsakelig er mindre enn den optimale dose av peptidet. Deretter blir dosen øket med små inkrementer inntil den optimale effekt under omstendighetene blir oppnådd. Generelt blir forbindelsen mest ønskelig administrert i et konsentrasjonsnivå som generelt vil gi antiviralt effektive resultater uten å forårsake noen ugunstige eller skadelige bivirkninger.
Når preparatet ifølge foreliggende oppfinnelse omfatter en kombinasjon av en forbindelse med formel I og ett eller flere ytterligere terapeutiske eller profylaktiske midler, bør både forbindelsen og det ytterligere middel foreligge i dosenivåer på mellom ca. 10 til 100% og mer foretrukket mellom ca. 10 og 80% av dosen som normalt blir administrert i et monoterapi-opplegg.
Når disse forbindelser eller deres farmasøytisk akseptable salter blir formulert sammen med en farmasøytisk akseptabel bærer, kan det resulterende preparat administreres in vivo til pattedyr, så som mennesker, for å hemme HCV NS3-protease eller for fremstilling av medikament for å behandle eller forhindre HCV-virusinfeksjon. Slik behandling kan også oppnås ved anvendelse av en forbindelse i kombinasjon med midler som omfatter: a-, |3-, 5-, co-, x- eller y-interferon, ribavirin, amantadin; andre inhibitorer av HCV NS3-protease; inhibitorer av HCV-polymerase, inhibitorer av andre mål i HCV-livssyklusen, som omfatter, helicase, NS2/3-protease eller indre ribosominnganssete (IRES); eller kombinasjoner derav. De ytterligere midler kan kombineres med forbindelser ifølge foreliggende oppfinnelse for å danne en enkel doseform. Alternativt kan disse ytterligere midler administreres separat til et pattedyr som en del av en multippel doseform.
Følgelig tilveiebringer en annen utførelsesform av oppfinnelsen anvendelse av en forbindelse med formel I for fremstilling av medikament for å hemme HCV NS3-proteaseaktivitet hos et pattedyr.
I en foretrukket utførelsesform anvendes dette for å redusere NS3-proteaseaktiviteten til hepatitt C viruset som infiserer et pattedyr.
Hvis det farmasøytiske preparatet omfatter bare en forbindelse ifølge foreliggende oppfinnelse som den aktive komponent, kan en slik anvendelse i tillegg omfatte trinnet administrering til nevnte pattedyr av et middel valgt fra et immunomodulerende middel, et antivirusmiddel, en HCV NS3- proteaseinhibitor, en inhibitor for HCV-polymerase eller en inhibitor for andre mål i HCV-livssyklusen så som helicase, NS2/3-protease eller IRES. Slike ytterligere midler kan administreres til pattedyret før, samtidig med eller etter administrering av preparatet ifølge foreliggende oppfinnelse.
En forbindelse med formel 1 angitt her kan også anvendes som et laboratoriereagens. En forbindelse ifølge foreliggende oppfinnelse kan også anvendes for å behandle eller forhindre viral forurensning av materialer og derfor redusere risikoen for virusinfeksjon av laboratorie- eller medisinsk personale eller pasienter som kommer i kontakt med slike materialer (f.eks. blod, vev, kirurgiske instrumenter og klesplagg, laboratorieinstrumenter og klesplagg og blodsamlingsapparater og materialer).
En forbindelse med formel 1 angitt her kan også anvendes som et forskningsreagens. En forbindelse med formelen 1 kan også anvendes som positiv kontroll for å bedømme surrogat-celle-baserte målinger eller in vitro virusreplikasjonsforsøk.

Syntese av forbindelser med formel (I):
Generelt blir forbindelsene med formel 1 og mellomprodukter for disse fremstilt ved kjente metoder ved anvendelse av reaksjonsbetingelser som er kjent for å være egnet for reaktantene. Mange slike metoder er beskrevet i WO 00/09543, WO 00/09558 og US Pat. No. 6,323,180, inntatt her som referanse.
Fremstilling av tiourinstoffer
Fremstilling av Tiourinstoff 2a
Tiourinstoff (5,0 gm, 66 mmol) ble oppløst i toluen (50 ml) og tert-butylacetylklorid (8,88 gm, 66 mmol) ble tilsatt. Blandingen ble oppvarmet ved tilbakeløp i 14 timer, hvilket ga en gul løsning. Blandingen ble konsentrert til tørrhet og residuet fordelt mellom EtOAc og mettet NaHC03. Den gule organiske fasen ble tørket over MgS04, filtrert og konsentrert, hvilket ga et gult, fast stoff. Det faste stoffet ble oppløst i en minimums- mengde av EtOAc og gnidd ut med heksan, hvilket ga 2a som et hvitt, fast stoff (8,52 g; 75%). M.S. (elektrospray): 173 (M-H)-175 (M+H)+. Revers- fase HPLC homogenitet (0,06 % TFA ; CH3CN : H20): 99 %.
Fremstilling av Tiourinstoff 2b
Anvendelse av metoden beskrevet ovenfor og ved anvendelse av kommersielt tilgjengelig cyklopentylacetylklorid istedenfor tert-butylacetylklorid ga tiourinstoff 2b.
Syntese av mellomprodukter 3
Fremstilling av karbamat 3a
Tetrahydrofuran (350 ml) ble satt til en kolbe inneholdende karbonsyre-cyklopentylester-2,5-diokso-pyrrolidin-1-ylester (9,00 g; 39,6 mmol) og fert-leucin (6,24 g; 47,5 mmol), hvilket resulterte i en suspensjon. Destillert vann (100 ml) ble tilsatt under kraftig røring. En liten mengde av fast stoff forble uoppløst. Trietylamin (16,6 ml; 119 mmol) ble deretter tilsatt, hvilket resulterte i en homogen løsning som ble rørt ved R.T. Etter 2,5 timer ble THF inndampet, og det vandige residuet fortynnet med vann (100 ml) og reaksjonsblandingen gjort basisk ved tilsetning av 1 N NaOH (25 ml - endelig pH >10). Løsningen ble vasket med EtOAc (2 x 200 ml) og den vandige fasen deretter surgjort med 1 N HCI (ca. 70 ml - endelig pH <2). Den uklare løsningen ble ekstrahert med EtOAc (200 + 150 ml). Ekstraktet ble tørket (MgS04) og inndampet, hvilket ga forbindelsen 3a som et hvitt, fast stoff (8,68 g).
Fremstilling av karbamater 3b, 3c og 3d
Ved anvendelse av metoden beskrevet ovenfor og ved anvendelse av de riktige kombinasjoner av ferf-butyl-glycin eller cykloheksyl-glycin og karbonsyre-cyklobutyl-, -cyklopentyl- eller-cykloheksylester-2,5-diokso-pyrrolidin-1-ylester ble de følgende karbamater fremstilt:
Fremstilling av mellomprodukt 5
Fremstilling av mellomprodukt 5a:
a-Bromketon 4 (3,61 g ; 5,71 mmol) ble kombinert med tiourinstoff 2a (1,09 g ; 6,28 mmol) i isopropanol (140 ml) og den gule løsningen ble plassert i et forvarmet oljebad på 70°C i 1,5 timer. Løsningen ble avkjølt til R.T. og inndampet til tørrhet. Residuet ble oppløst i EtOAc. EtOAc-løsningen ble vasket med mettet NaHC03 (2x), vann (2x) og saltvann (1x), tørket (MgS04) og inndampet, hvilket ga produktet som et oransjebrunt skum. Flash- kolonnekromatografi i 7:3 heksan : EtOAc fjernet de mindre polare urenheter, og 6:4 heksan : EtOAc ga rent produkt som et lysegult, fast stoff (3,05 g ; 76%). M.S.(elektrospray): 706,3 (M-H)- 708,4 (M+H)+. Reversfase-HPLC homogenitet (0,06 % TFA; CH3CN : H20): 99 %.

EKSEMPEL 1
Syntese av Forbindelse 100
Trinn 1: Fremstilling av mellomprodukt 6a
Boc-dipeptid 5a (3,05 g ; 4,31 mmol) ble oppløst i 4N HCI/dioksan (22 ml). Etter røring ved R.T. i 30 min., falt HCI-saltet ut. MeOH (2 ml) ble tilsatt for å oppløse fellingen. Etter 2 timer ble reaksjonsblandingen inndampet til tørrhet. Det resulterende HCI-salt ble oppløst i DCM (22 ml) og DIEA (3,0 ml; 17,24 mmol); karbamat 3a (1,15 g; 4,47 mmol), og HATU (1,72 g ; 4,516 mmol) ble tilsatt. Løsningen ble rørt ved romtemperatur i 6 timer. Blandingen ble deretter fortynnet med EtOAc og løsningen vasket med mettet NaHC03 (2x), vann (2x) og saltvann (1x), tørket (MgS04), filtrert og inndampet for å oppnå forbindelsen 6a som et gult, fast stoff. Flash-kolonnekromatografi under eluering først med heksan : EtOAc 7:3 og deretter 6:4 ga ren Me-ester 6a som et hvitt skum (3,25 g ; 90%). M.S.

Trinn 2: Hydrolyse av ester
Metylester 6a (3,24 mg; 3,89 mmol) ble oppløst i TH F (40 ml) og MeOH (20 ml) og en vandig løsning av LiOH (1,63 mg; 38,9 mmol i 25 ml) ble tilsatt. Den gule reaksjonsblanding ble rørt i 5,5 timer og deretter konsentrert for å oppnå en gråhvit suspensjon. Suspensjonen ble oppløst i EtOAc og saltvann, fremstilt med ionefritt vann. pH i den resulterende løsning ble regulert til 6 ved tilsetning av 1N HCI. Sjiktene ble separert og det vandige sjiktet videre ekstrahert med EtOAc (2x). De samlede EtOAc-ekstrakter ble vasket med ionefritt vann (2x), saltvann fremstilt fra ionefritt vann (1x), tørket (MgS04), filtrert og inndampet for å oppnå forbindelse 100 som et blekgul-hvitt fast stoff (3,02 g; 95% utbytte).
Omdannelse til Na-salt:
Den nøytrale forbindelsen 100 (1,22 g; 1,49 mmol) ble oppløst i MeOH (30 ml) og 1 ekvivalent 0,01 N NaOH (14,85 ml) ble tilsatt - ingen produkt- utfelling. Den klare gule løsning ble konsentrert, fortynnet med ionefritt vann, frosset og frysetørket for å oppnå produktet (Na-salt) som et gulhvitt amorft fast stoff (1,24 g; 99 % utbytte). Forbindelse 6b (70 mg, 0,095 mmol) ble oppløst i 2 ml DCM og suksessivt behandlet med DIEA (0,045 ml, 0,26 mmol) og pivaloylklorid (0,015 ml, 0,12 mmol). Etter røring i 1 time ved 40° ble ytterligere pivaloylklorid (0,015 ml, 0,12 mmol) tilsatt og røring fortsatte i ytterligere 2 timer. Etter at løsningen var konsentrert, ble residuet oppløst i EtOAc. Løsningen ble vasket med en mettet løsning av NaHC03 og saltvann, tørket (MgS04) og konsentrert, hvilket ga 82 mg av råproduktet 6c som ble anvendt uten rensning.
Metylesterderivatet 6c ble hydrolyser! som i trinn 2 i eksempel 1 og renset ved preparativ HPLC ved anvendelse av en YMC Combi-Prep. ODS-AQ- kolonne, 50 x 20mm. ID, S - 5mikron, 120 A og et lineært gradientprogram fra 2 til 100% AcCN /vann (0,06% TFA).
Fraksjoner ble analysert ved analytisk HPLC, og de rene fraksjoner ble samlet, konsentrert, frosset og frysetørket, hvilket ga forbindelse 101 som trifluoracetatsaltet:

Forbindelse 100 fra eksempel 1 (29,8 mg, 0,036 mmol) ble kombinert med HATU (1,2 ekv., 19,7 mg, 0,052 mmol) og oppløst i vannfritt DMF (4 ml). Løsningen ble rørt ved R.T. og DIPEA (5 ekv., 31,4uL, 0,18 mmol) ble tilsatt dråpevis over ca. 1 min. Blandingen ble rørt i 20min. ved romtemperatur og analysert ved analytisk HPLC for dannelse av den aktiverte ester. En løsning av metansulfonamid (5,8ekv., 19,7 mg, 0,207 mmol), DMAP (5,4ekv., 23,5 mg, 0,193 mmol) og DBU (4,8ekv., 25,8uL, 0,172 mmol) ble tilsatt i DMF (1 ml). Reaksjonsblandingen ble rørt 16 timer ved romtemperatur før den ble konsentrert / vakuum. Residuet ble rekonstituert i DMSO og renset ved preparativ HPLC. Lyofilisering ga sluttproduktet (23 mg, 71,3%) som et grå-hvitt amorft fast stoff.

Celle Basert HCV RNA Replikasjonsmåling
Cellekultur
Huh7 celler som stabilt opprettholdt et subgenomisk HCV-replikon ble opprettet
som tidligere beskrevet (Lohman et al., 1999. Science 285:110-113) og betegnet som S22.3 celle-linje. S22.3 celler blir holdt i Dulbecco's Modifisert Earle Medium (DMEM) supplert med 10% FBS og 1 mg/ml neomycin (Standard Medium). Under målingen ble DMEM-medium supplert med 10% FBS, inneholdende 0,5% DMSO, og som mangler neomycin, anvendt (Målemedium). 16 timer før forbindelsens tilsetning blir S22.3 celler trypsinisert og fortynnet til 50 000 celler/ml i Standard Medium. 200ul (10 000 celler) blir fordelt i hver brønn i en 96-brønners plate. Platen ble deretter inkubert ved 37° med 5% C02 til neste dag.

Fremstilling av testforbindelse
10ul testforbindelse (i 100% DMSO) ble satt til 2 ml Målemedium til en sluttkonsentrasjon i DMSO på 0,5%, og løsningen ble ultralydbehandlet i 15 min og filtrert gjennom en 0,22 uM Millipore filterenhet. 900ul ble overført til rekke A i
en Polypropylen dyp-brønn titerplate. Rekkene B til H inneholder 400ul alikvoter av Målemedium (inneholdende 0,5% DMSO) og blir anvendt for å fremstille serie-fortynninger (1/2) ved overføring av 400 ul fra rekke til rekke (ingen forbindelse forlå i rekke H).
Applikasjon av testforbindelse til celler
Celledyrkningsmedium ble fordampet fra 96-brønners platen inneholdende S22.3 cellene. 175ul målemedium med den riktige fortynning av testforbindelse ble overført fra hver brønn i forbindelsesplaten til den tilsvarende brønn i cellekulturplaten (rekke H ble anvendt som "Ikke- hemningskontroll"). Cellekulturplaten ble inkubert ved 37° med 5% C02 i 72 timer.
Ekstraksjon av totalt cellulært RNA
Etter 72-timers inkuberingsperioden ble det totale cellulære RNA ekstrahert fra S22.3 cellene i 96-brønners platen ved anvendelse av RNeasy 96 settet (Qiagen®, RNeasy Handbook. 1999.). I korthet ble målemediet fullstendig fjernet fra celler og 100 ul RLT buffer (Qiagen®) inneholdende 143 mM p-merkaptoetanol ble satt til hver brønn i 96-brønners celle-kulturplaten. Mikroplaten ble forsiktig ristet i 20 sekunder. 100 pl 70% etanol ble deretter satt til hver mikroplatebrønn og blandet ved pipettering. Lysatet ble fjernet og påført brønnene i en RNeasy 96 (Qiagen®) plate som ble plassen" på toppen av en Qiagen® Square-Well Block. RNeasy 96 platen ble forseglet med tape og Square-Well Block med RNeasy 96 platen ble satt i holderen og plassert i en rotorbøtte i en 4K15C sentrifuge. Prøven ble sentrifugert ved 6000 opm (-5600 x g) i 4 min ved romtemperatur. Tapen ble fjernet fra platen og 0,8 ml Buffer RW1 (Qiagen® RNeasy 96 sett) ble satt til hver brønn i RNeasy 96 platen. RNeasy 96 platen ble forseglet med et nytt stykke tape og sentrifugert ved 6000 opm i 4 min ved romtemperatur. RNeasy 96 platen ble plassert på toppen av en annen ren Square-Well Block, tapen fjernet og 0,8 ml Buffer RPE (Qiagen® RNeasy 96 sett) ble satt til hver brønn i RNeasy 96 platen. RNeasy 96 platen ble forseglet med et nytt stykke tape og sentrifugert ved 6000 opm i 4 min ved romtemperatur. Tapen ble fjernet og ytterligere 0,8 ml Buffer RPE (Qiagen® RNeasy 96 sett) ble satt til hver brønn i RNeasy 96 platen. RNeasy 96 platen ble forseglet med et nytt stykke tape og sentrifugert ved 6000 opm i 10 min ved romtemperatur. Tapen ble fjernet, RNeasy 96 platen ble plassert på toppen av en rack inneholdende 1,2-ml samlemikrorør. RNA'et ble eluert ved tilsetning av 50 ul RNase-fritt vann til hver brønn, platen lukket med et nytt stykke tape og inkubert i 1 min ved romtemperatur. Platen ble deretter sentrifugert ved 6000 opm i 4 min ved romtemperatur. Elueringstrinnet ble gjentatt med en nytt volum av 50 pl RNase-fritt vann. Mikrorørene med totalt cellulært RNA blir lagret ved -70°.
Kvantifisering av totalt cellulært RNA
RNA ble kvantifisert på the STORM® system (Molecular Dynamics®) ved anvendelse av RiboGreen® RNA Quantification Kit (Molecular Probes®). I korthet ble RiboGreen reagens fortynnet 200 ganger i TE (10mM Tris-HCI pH =7,5,1mM EDTA). Generelt ble 50ul reagens fortynnet i 10 ml TE. En standardkurve av ribosomalt RNA ble fortynnet i TE til 2 Mg/m I og forut bestemte mengder (100, 50, 40, 20,10, 5, 2 og 0 pl) av den ribosomale RNA løsning blir deretter overført i en ny 96-brønners plate (COSTAR # 3997) og volumet ble fylt opp til 100ul med TE. Generelt ble kolonne 1 i 96-brønners platen anvendt for standardkurven, og de andre brønner blir anvendt for RNA prøvene som skal kvantifiseres. 10ul av hver RNA-prøve som skulle kvantifiseres, ble overført til den tilsvarende brønn i 96-brønners platen og 90pl av TE ble tilsatt. Et volum (100ul) av fortynnet RiboGreen reagens ble satt til hver brønn i 96-brønners platen og inkubert i 2 til 5 minutter ved romtemperatur beskyttet mot lys (en a 10 uJ RNA-prøve i en 200 uJ slutt-volum genererer en 20 X fortynning). Fluorescensintensiteten til hver brønn ble målt i STORM® systemet (Molecular Dynamics®). En standardkurve ble dannet på basis av de kjente mengder av det ribosomale RNA og de resulterende fluorescensintensiteter. RNA-konsentrasjonen i de eksperimentelle prøver ble bestemt fra standardkurven og korrigert for 20X fortynningen.

Sanntid RT-PCR
Sanntid RT-PCR ble utført på ABI Prism 7700 Sequence Detection System ved anvendelse av TaqMan EZ RT-PCR Kit fra (Perkin-Elmer Applied Biosystems®). RT-PCR ble optimalisert for kvantifisering av 5' IRES i HCV RNA ved anvendelse av Taqman teknologi (Roche Molecular Diagnostic Systems) lignende teknikken tidligere beskrevet (Mariell et al., 1999. J. Clin. Microbiol. 37: 327-332). Systemet utnytter den 5'-3' nukleolytiske aktiviteten til AmpliTaq DNA polymerase. I korthet anvender metoden en dobbelt-merket fluorogen hybridiserings-probe (PUTR Probe) som spesifikt kobler til templatet mellom PCR-primerne (primere 8125 og 7028). 5'-enden av proben inneholder en fluorescerende rapportør (6-karboksyfluorescein [FAM]) og 3'-enden inneholder en fluorescerende stopper (6-karboksytetrametylrhodamin [TAMRA]). FAM rapportørens emisjonspektrum ble undertrykket med stopperen på den intakte hybridiseringsprobe. Nukleasenedbrytning av hybridiseringsproben frigjør rapportøren, hvilket resulterer i en økning i fluorescensemisjon. ABI Prisme 7700 sekvensdetektoren måler økningen i fluorescensemisjon kontinuerlig under PCR-amplifiseringen, slik at det amplifiserte produkt var direkte proporsjonalt med signalet. Amplifiserings- plotten ble analysert tidlig under reaksjonen på et punkt som representerer den logaritmiske fase av produktakkumulering. Et punkt som representerer en definert deteksjonsterskel for økningen i fluorescenssignalet forbundet med den eksponentielle vekst av PCR-produktet for sekvensdetektoren ble definert som syklusterskelen (Cr). Crverdier er omvendt proporsjonale med mengden av innført HCV RNA; slik at under identiske PCR-betingelser, var startkonsentrasjon av HCV RNA større jo lavere Cr var. En standard kurve ble dannet automatisk med ABI Prisme 7700 deteksjonssystemet ved plotting av Cr mot hver standardfortynning av kjent HCV RNA-konsentrasjon.
Referanseprøver for standardkurven inngår på hver RT-PCR-plate. HCV Replicon RNA ble syntetisert (ved T7 transkripsjon) in vitro, renset og kvantifisert ved OD26o-I betraktning av at 1ug av dette RNA = 2,15 X 10<11> RNA kopier, blir fortynninger gjort for å få 10<8>,10<7>,10<6>,10<5>,10<4>,10<3> eller 10<2> genomiske RNA-kopier/5ul. Totalt cellulært Huh-7 RNA ble også inntatt med hver fortynning (50ng / 5ul). 5ul av hver referansestandard (HCV Replicon + Huh-7 RNA) ble kombinert med 45ul Reagens Mix og anvendt i sanntid RT-PCR-reaksjonen.
Sanntid RT-PCR-reaksjonen ble satt opp for de eksperimentelle prøver som ble renset på RNeasy 96-brønners plater ved å kombinere 5ul av hver total cellulær RNA-prøve med 45pl Reagens Mix.

Termiske sirkuleringsbetingelser:
Etter avslutningen av RT-PCR-reaksjonen krever dataanalysene fastsetting av terskelfluorescenssignal for PCR-platen, og en standardkurve ble konstruert ved å plotte Ct-verdien mot RNA-kopitall anvendt i hver referanse- reaksjon. Ct-verdiene oppnådd for måleprøvene blir anvendt til å interpolere et RNA-kopitall basert på standardkurven.
Til slutt ble RNA-kopitallet normalisert (basert på RiboGreen RNA kvantifisering av det totale RNA ekstrahert fra cellekulturbrønnen) og uttrykt som genom ekvivalenter / ug av totalt RNA [ge/ug].
RNA-kopitallet [g.e./ug] fra hver brønn i cellenkulturplaten var et mål for mengden av replikerende HCV RNA i nærvær av forskjellige konsentrasjoner av inhibitor. % hemning ble beregnet med den følgende formel:
En ikke-lineær kurvetilpasning med Hill-modellen ble anvendt på hemnings-konsentrasjonsdataene, og den 50% effektive konsentrasjon (EC5o) ble beregnet ved anvendelse av SAS programvare (Statistical Software System; SAS Institute, Inc. Cary, N.C.).
Når forbindelsene ifølge foreliggende oppfinnelse ble evaluert i den forutgående enzymatiske og cellebaserte måling, ble forbindelsene funnet å være meget aktive. Mer spesifikt hadde forbindelsene IC5o'er under 0,1 uM i NS3-NS4A proteasemåling og EC5o'er under 0,5 uM i den cellebaserte HCV RNA-replikasjonsmåling.

Farmakokinetiske egenskaper
Foreliggende forbindelser viser også gode farmakokinetiske egenskaper så som detekterbare plasmanivåer i rotte ved 1 time og 2 timer etter en oral dose på 4 eller 5 mg/kg.
Mer eksplisitt ble den følgende måling, en in vivo oral absorpsjonscreen, anvendt for å bestemme plasmanivåer for testforbindelser i rotte etter oral administrering:
Materialer og metoder:
1. Metode anvendt til sammenslåing av forbindelser ("kassett-
seleksjon"): Seleksjonen av forbindelser som skal samles i en "kassett" var basert på deres strukturelle likhet og fysiskalsk-kjemiske egenskaper. En fastfase-ekstraksjonsmetode som var anvendbar for alle de valgte forbindelser ble oppsatt. Basert på de innledende tester hvor hver forbindelse ble ført inn i rotteplasma og kjørt gjennom HPLC eller HPLC/MS i en konsentrasjon på 0,5 uM, ble retensjonstiden, ionisk masse og mulig separering av forbindelser ved HPLC og/eller HPLC/MS anvendt som grunnlag for samling av 3-4 forbindelser i én "kasser.
2. Oral konstituent- og forbi ndelsespreparat:
Hver "kassett" inneholder 3-4 forbindelser ved 5 eller 4 mg/kg for hver forbindelse. Kassettene ble fremstilt som en oral suspensjon i 0,5% vandig metylcellulose og 0,3% polyoksyetylen(20)sorbitonmonooleat (Tween-80). Doseringsvolumet varl 0 ml/kg gjennom oral gavage.
3. Dosering og plasmaprøvetaking:
Sprague Dawley hannrotter ble fastet natten over i individuelle bur med tilgang til vandig 10% dekstrose. To rotter ble dosert med hver "kassett". Plasmaprøver (~1 ml) ble oppsamlet ved 1 og 2 timer etter dosering fra de 2 rotter og samlet for ekstraksjon og analyse.
4. Forbindelsesekstraksjon og analyse:
Fra hver kassett blir plasmaprøver ved 1 og 2 timer, blindplasma, blind- plasma forsynt med alle forbindelsene ved 0,5 uM av hver, ekstrahert ved fastfase-ekstraksjonsmetoden. Prøver ble analysert med HPLC og HPLC/MS for sammenligningsformål. Plasmakonsentrasjoner blir beregnet basert på den ene konsentrasjon av 0,5 uM standard.
Resultater
Ved måling i den forutgående screen ble forbindelsene fra eksempler 1 til 8 ifølge foreliggende oppfinnelse funnet å være tilstede i signifikante nivåer i plasmaet ved 1 times og 2 timers intervaller etter oral administrering, henholdsvis gjennomsnittlige blodplasmanivåer på 0,83 uM og 0,75 uM.
Denne demonstrasjon av in vivo oral absorpsjon for tripeptidforbindelsene ifølge foreliggende oppfinnelse er bemrkelsesverdig i betraktning av den dårlige orale absorbsjon som generelt tilskrives denne klassen av peptider. Den raske orale absorpsjon gjør forbindelsene anvendelige for fremstilling av medikament for behandling av HCV-infeksjon.
Claims (24)
1. Forbindelse med formel (I):
hvor R<1> er hydroksy eller NHS02R<1A> hvor R<1A> er (Ci-8)alkyl, (C3.7)cykloalkyl eller {(Ci-6)alkyl-(C3-7)cykloalkyl}, som alle eventuelt er substituert fra 1 til 3 ganger med halogen, cyano, nitro, 0-(Ci-e)alkyl, amido, amino eller fenyl eller R<1A> er C6 eller C10 aryl, som eventuelt er substituert fra 1 til 3 ganger med halogen, cyano, nitro, (Ci-e)alkyl, 0-(Ci-6)alkyl, amido, amino eller fenyl; R2 er t-butyl, -CH2-C(CH3)3 eller -CH2-cyklopentyl; R<3> er t-butyl eller cykloheksyl og R4 er cyklobutyl, cyklopentyl eller cykloheksyl; eller et farmasøytisk akseptabelt salt derav.
2. Forbindelse med formel I ifølge krav 1, hvor R<1> er hydroksy, NHS02Me, NHS02-cyklopropyl eller NHS02Ph.
3. Forbindelse med formel I ifølge krav 2, hvor R<1> er NHS02Me eller hydroksy.
4. Forbindelse med formel I ifølge krav 3, hvor R<1> er hydroksy.
5. Forbindelse med formel I ifølge hvilket som helst av kravene 1 til 4, hvor R<2> er t-butyl eller CH2-C(CH3)3.
6. Forbindelse med formel I ifølge krav 5, hvor R2 er CH2-C(CH3)3.
7. Forbindelse med formel I ifølge hvilket som helst av kravene 1 til 6, hvor R<3> er t-butyl.
8. Forbindelse med formel I ifølge hvilket som helst av kravene 1 til 7, hvor R<4> er cyklopentyl eller cykloheksyl.
9. Forbindelse med formel I ifølge krav 8, hvor R<4> er cyklopentyl.
10. Forbindelse med formel I som definert i krav 1, hvor R<1> er hydroksy, R2 er CH2-C(CH3)3, R3 er t-butyl og R<4> er cyklopentyl.
11. Forbindelse med formel I som definert i krav 1, hvor R<1> er hydroksy, R2 og R<3> hver er t-butyl og R<4> er cyklopentyl.
12. Forbindelse med formel I som definert i krav 1, hvor R<1> er hydroksy, R2 er CH2-C(CH3)3, R3 er cykloheksyl og R<4> er cyklopentyl.
13. Forbindelse med formel I som definert i krav 1, hvor R<1> er hydroksy, R2 er CH2-C(CH3)3 og R3 og R4 hver er cykloheksyl.
14. Forbindelse med formel I som definert i krav 1, hvor R<1> er hydroksy, R2 er cyklopentylmetyl, R3 er t-butyl og R<4> er cyklobutyl.
15. Forbindelse med formel I som definert i krav 1, hvor R<1> er hydroksy, R2 er CH2-C(CH3)3, R3 er t-butyl og R<4> er cyklobutyl.
16. Forbindelse'med formel I som definert i krav 1, hvor R<1> er NHS02Me, R<2> er CH2-C(CH3)3 R3 er t-butyl og R4 er cyklopentyl.
17. Forbindelse med formel I som definert i krav 1, hvor R<1> er NHS02Ph, R<2> er CH2-C(CH3)3, R3 er t-butyl og R<4> er cyklopentyl.
18. Farmasøytisk preparat omfattende en anti-hepatitt C viralt effektiv mengde av en forbindelse med formel I ifølge hvilket som helst av kravene 1 til 17 eller et farmasøytisk akseptabelt salt derav i blanding med en farmasøytisk akseptabelt bærermedium eller hjelpemiddel.
19. Farmasøytisk preparat ifølge krav 18, videre omfattende en terapeutisk effektiv mengde av én eller flere andre anti-HCV-midler.
20. Farmasøytisk preparat ifølge krav 19, hvor nevnte andre anti-HCV-middel er valgt
fra: a-interferon eller pegylert a-interferon.

21. Farmasøytisk preparat ifølge krav 19 eller 20, hvor nevnte andre anti-HCV- middel er ribavirin.
22. Farmasøytisk preparat ifølge krav 19, 20 eller 21, hvor nevnte andre anti-HCV middel er valgt fra inhibitorer av: helicase, NS2/3 protease og indre ribosom-inngangsete (IRES).
23. Anvendelse av en forbindelse med formel I ifølge hvilket som helst av kravene 1 til c 17 for fremstilling av et medikament for behandling eller forebygging av hepatitt C virusinfeksjon.
24. Farmasøytisk preparatet ifølge krav 19, 20 eller 21, hvor nevnte andre anti-HCV-middel er en inhibitor for HCV-polymerase.
