Abstract
Det beskrives nye 3-substituerte 4-(fenyl-N-alkyl)-piperazine- og 4-(fenyl-N-alkyl)-piperidinforbindelser av Formel 1, hvor X er N, CH eller C, imidlertid slik at X kun kan være C dersom forbindelsen omfatter en dobbeltbinding ved den prikkede linje; Ri er OSO2CF3, OSO2CH3, SOR3, SO2R3, COR3, N02 og CONHR3, og idet X er CG eller C, kan Ri også være CF3, CN, F, Cl, Br eller I; R2 er en Ci-C4-alkyl, en allyl, CH2SCH3, CH2CH2OCH3, CH2CH2CH2F, CH2CF3, 3,3,3-trifluorpropyl, 4,4,4-trifluorbutyl eller (CH2)-R4; R3 er en Ci-C3-alkyl, CF3, eller N(R2)2; R4 er C3-C6-cykloalkyl, 2-tetrahydrofuran eller 3-tetrahydrofuran, og likeledes farmasøytisk akseptable salter derav. Område for oppfinnelsen
Den foreliggende oppfinnelse vedrører 3-substituert 4-(fenyl-N-alkyl)-piperazin-forbindelser, farmasøytiske materialer og farmasøytiske sammensetninger derav, samt anvendelser av forbindelsene.
Bakgrunn og teknikkens stilling
Dopamin er en neurotransmitter i hjernen. Siden dens oppdagelse, i 1950-årene, er funksjonen til dopamin i hjernen i utstrakt grad blitt undersøkt. Det er således i dag godt etablert at dopamin er essensiell i flere aspekter av hjernefunksjon, inkluderende motor, kognitive, sen-soriske, emosjonelle og autonome (f.eks. regulering av appetitt, kroppstemperatur, søvn) funksjoner. Således vil modulering av dopaminerge funksjoner være nyttige i behandling av et bredt spekter av forstyrrelser som påvirker hjernefunksjoner. Faktisk behandles både neurologiske og psykiatriske forstyrrelser med medisineringer basert på interaksjoner med dopaminsystemer og dopaminreseptorer i hj ernen.
Medikamentet som virker på, direkte eller indirekte, sentrale dopaminreseptorer anvendes vanligvis i behandlingen av neurologiske og psykiatriske forstyrrelser, f.eks. Parkinson's sykdom og schizofreni. Dagens til-gjengelige dopaminerge farmasøytika har flere bieffekter, så som ekstrapyridimale bieffekter og tardiv dyskinesi for dopaminerge antagonister anvendt som antipsykotiske midler, og dyskinesi og psykoser for dopaminerge agonister anvendt i anti-Parkinson-midler. De terapeutiske effekter er i mange sammenhenger ikke tilfredsstillende. For å forbedre effektiviteten og redusere bieffektene av dopaminerge farmasøytika, søkes det etter nye dopaminreseptorligander med selektivitet ved spesifikke dopaminreseptor subtyper eller regional selektivitet. I denne sammenheng utvikles det også partielle dopaminreseptoragonister, dvs. dopaminreseptorligander med en viss grad, men ikke full indre aktivitet ved dopaminreseptorene, for å oppnå en optimal grad av stimulering av dopaminreseptorene, og unngå utstrakt dopaminreseptorblokkade eller for høy stimulering.
Forbindelser som tilhører klasser av substituerte 4-(fenyl-N-alkyl)piperazin og substituerte 4-(fenyl-N-pipe-ridiner) er tidligere blitt rapportert. Blant disse forbindelser er noen inaktive i CNS, noen oppviser seroto-nergiske eller blandete serotonergiske/dopaminergiske farmakologiske profiler, mens andre er fullstendige eller partielle dopaminreseptorantagonister eller agonister med høy affinitet for dopaminreseptorer.
Et antall 4-fenylpiperaziner og 4-fenyl-piperidin-derivater er kjent og beskrevet, f.eks. i Costall et al., European J. Pharm., vol 31, p 94 (1975), og Mewshaw
et al., Bioorg. Med. Chem. Lett., vol 8, p 295 (1998). De rapporterte forbindelser er substituerte 4-fenyl-piperaziner, og de fleste av disse er 2-, 3- eller 4-OH-fenyl-substituert, og oppviser DA-autoreseptor-agonistegenskaper.
R. W. Fuller et al., J. Pharmacol. Exp. Therapeut., vol 218, p 636 (1981) beskriver substituerte piperaziner (f.eks. 1-(m-trifluormetylfenyl)piperazin)) som rapporteres å virke som serotonin-agonister og inhiberer serotonin-opptak. R. W. J. Boissier et al., Chem. Abstr., 61:10691c beskriver disubstituerte piperaziner. Forbindelsene rapporteres å være adrenolytiske, antihypertensive, potensiatorer av barbiturater, og depressanter i forhold til det sentrale nervesystem.
Et antall forskjellige substituerte piperaziner er blitt publisert som ligander ved 5-HTiA-reseptorer, f.eks. R. A. Glennon et al., J. Med. Chem., vol 31, p 1968 (1988) og B. J. van Steen, J. Med. Chem., vol 36, p 2751 (1993), M. L. Dukat, J. Med. Chem., vol 39, p 4017 (1996). R. A. Glennon beskriver, i Internasjonal Patentsøknad WO 93/00313 og WO 91/09594, forskjellige aminer, blant dem substituerte piperaziner, som sigma-reseptor-ligander. Kliniske forsøk som undersøker egenskapene til sigma-reseptorligandene i schizofrene pasienter har ikke generert bevis på antipsykotisk aktivitet, eller aktivitet i noen andre CNS-forstyrrelser. Videre beskriver WO 93/04684 og GB 2.027.703 også spesifikke substituerte piperaziner som er nyttige i behandlingen av CNS-forstyrrelser.
Sammendrag av oppfinnelsen
Formålet med foreliggende oppfinnelse er å tilveiebringe nye farmasøytisk aktive materialer, spesielt nyttige i behandling av forstyrrelser i det sentrale nervesystem, og som ikke har de ulemper som er forbundet med de ovenfor beskrevne substanser.
I det arbeid som førte frem til foreliggende oppfinnelse ble det funnet at det er ønskelig å tilveiebringe substanser med spesifikke farmakologiske egenskaper, nemlig substanser som har modulerende effekter på dopamin-neurotransmisjon. Disse egenskaper er ikke tidligere blitt beskrevet, og de er ikke mulig å oppnå med de tidligere kjente forbindelser. Forbindelsene i samsvar med foreliggende oppfinnelse har en svært overraskende og interessant dualistisk dopaminergisk virkningsprofil med antagonist-lignende effekter på hjerne-neurokjemi, og milde agonist-lignende effekter på normal adferd, men de induserer inhibering av adferd i former av hyperaktivitet.
Den foreliggende oppfinnelse vedrører således nye 3-substituerte 4-(fenyl-N-alkyl)-piperaziner og 3-substituerte-4-(fenyl-N-alkyl)-piperidiner i form av fri base eller farmasøytisk akseptable salter derav, farmasøytiske materialer inneholdende nevnte forbindelser, og anvendelse av nevnte forbindelser i terapi.
Ett formål med foreliggende oppfinnelse er å tilveiebringe nye forbindelser for terapeutisk anvendelse, og mer bestemt forbindelser for å modulere dopaminerge systemer i mammalske hjerner, inkluderende hjernen til mennesket.
Et annet formål med foreliggende oppfinnelse er å tilveiebringe forbindelser med terapeutiske effekter etter oral administrering.
Nærmere bestemt vedrører foreliggende oppfinnelse en 3-substituert 4-(fenyl-N-alkyl)-piperazin-forbindelse,
som har Formel 1:
Ri er valgt fra gruppen som består av OS02CF3, OSO2CH3, SOR3, SO2R3, COCH3, og COCH2CH3, hvor R3 er som definert nedenfor,
R2 er valgt fra gruppen som består av Ci-C4-alkyler, allyler, CH2CH2OCH3, CH2CH2CH2F, CH2CF3, 3,3,3-trif luorpropyl og 4,4,4-trifluorbutyl,
R3 er valgt fra gruppen som består av Ci-C3-alkyler, CF3 og N(CH3)2,
med det forbehold at når Ri er SO2R3 og R3 er et C1-C3 alkyl, er R2 ikke CH2CH2OCH3,
eller et farmasøytisk akseptabelt salt derav.
Det er foretrukket at Ri er valgt fra gruppen som består av OS02CF3, OS02CH3, S02CH3, S02CF3, COCH3, og S02N(CH3)2. Videre foretrekkes det at R2 er valgt fra gruppen som består av n-propyl og etyl.
Oppfinnelsen vedrører også et farmasøytisk materiale som omfatter en forbindelse i samsvar med et av kravene 1-3 og et eller flere farmasøytisk akseptable bærere eller tynnere. Et foretrukket farmasøytisk materiale er kjennetegnet ved det benyttes for behandling av en tilstand valgt fra gruppen som består av iatrogene og ikke-iatrogene Parkinsonismer, dyskinesier, dystonier og Tourette's sykdom. Videre fortrekkes det et farmasøytisk materiale som benyttes for behandling av Parkinsons sykdom, av en tilstand valgt fra gruppen som består av iatrogene og ikke-iatrogene psykoser og hallusinasjoner, av en tilstand valgt fra schizofreni og schizofreniforme forstyrrelser samt av en tilstand valgt fra gruppen som består av humør- og angstforstyrrelser. Særlig fortrukket er et farmasøytisk materiale som er kjennetegnet ved at nevnte humør- og angstforstyrrelser er valgt blant manodepressiv sykdom, depresjon og obsessiv-kompulsiv forstyrrelse.
Videre omfatter foreliggende oppfinnelse en farmasøytisk sammensetning som er egnet til behandling av gruppen som består av oppmerksomhetssviktforstyrrelser, autisme og kognitive dysfunksjoner. Det foretrekkes en farmasøytisk sammensetning som er til behandling av Huntington's sykdom og søvnforstyrrelser samt til behandling av substansrelaterte forstyrrelser forbundet med alkoholmisbruk eller et vanedannende medikament. Videre forektrekkes det en farmasøytisk sammensetning som er formulert for oral administrasjon. Særlig fortrekkes det at den er formulert som en tablett. Den kan også formuleres som en kapsel. Videre foretrekkes det en farmasøytisk sammensetning som er formulert for administrasjon ved injeksjon.
Oppfinnelsen vedrører også en anvendelse av en forbindelse i samsvar med krav 1-3 for fremstilling av en farmasøytisk sammensetning for behandling av en tilstand valgt fra gruppen som består av iatrogene og ikke-iatrogene Parkinsonismer, dyskinesier, dystonier og Tourette's sykdom, for behandling av behandling av Parkinsons sykdom, samt for behandling av en tilstand valgt fra gruppen som består av iatrogene og ikke-iatrogene psykoser og hallusinasjoner. Særlig fortrekkes det en anvendelse av en forbindelse for fremstilling av en farmasøytisk sammensetning for behandling av Huntington's sykdom samt av substansrelaterte forstyrrelser forbundet med alkoholmisbruk eller et vanedannende medikament.
Forbindelsene i samsvar med foreliggende oppfinnelse oppviser dopamin-modulerende egenskaper og er nyttige i behandling av flere sentralnervesystemforstyrrelser, inkluderende både psykiatriske og neurologiske symptomer.
Sykdommer hvor forbindelsene med modulerende effekter på dopaminerge systemer kan være nyttige, er forstyrrelser relatert til aldring, for å hindre bradykinesi og depresjon og for forbedring av mentale funksjoner. De kan også anvendes for å forbedre kognitive funksjoner, og relaterte emosjonelle forstyrrelser i neurodegenerative og utviklingsforstyrrelser, og likeledes etter hjerneskade.
Forbindelsene i samsvar med foreliggende oppfinnelse
anvendes for å forbedre symptomer på psykose, inkluderende schizofreni og schizofreniforme forstyrrelser, og likeledes medikamentinduserte psykotiske forstyrrelser. Forbindelsene i samsvar med oppfinnelsen kan også anvendes i adferdsforstyrrelser som vanligvis først diagnostiseres i spedbarn, barndom, eller ungdomstid, og likeledes i impulskontroll-forstyrrelser. Også taleforstyrrelser så som stamming kan forbedres. De kan også anvendes for å behandle substans-misbruks-forstyrrelser og likeledes forstyrrelser karakterisert med misbruk av næringsstoffer.
Sinnstemnings- og angstforstyrrelser,
personalitetsforstyrrelser, og omstillingshysteri kan også behandles med forbindelsene i samsvar med oppfinnelsen.
Neurologiske indikasjoner inkluderer behandling av Huntington's sykdom og andre bevegelsesforstyrrelser, og likeledes bevegelsesforstyrrelser indusert av medikamenter. "Urolige ben" og relaterte forstyrrelser, og likeledes narkolepsi kan også behandles med forbindelsene inkludert i samsvar med oppfinnelsen. De kan også forbedre mentale og motorfunksjoner i Parkinson's sykdom, og i relaterte parkinsonske syndromer. De kan også anvendes for å lindre tremor av forskjellig opprinnelse. De kan anvendes i behandling av hodepine, og anvendes for å forbedre hjernefunksjon etter vaskulær og traumatisk hjerneskade. Videre kan de anvendes for å stille smerter i tilstander karakterisert av økt muskeltone.
Forbindelsene i samsvar med foreliggende oppfinnelse er uventet blitt funnet å virke spesifikt på dopaminerge systemer i hjernen. De har effekter på biokjemiske indisier i hjernen med de karakteristiske trekk av selektive dopa-minantagonister, f.eks. ved å produsere økninger i konsen-trasjoner av dopaminmetabolitter.
Dopaminreseptor-antagonister undertrykker karakte-ristisk adferdsaktivitet og induserer katalepsi, mens forbindelsene ifølge foreliggende oppfinnelse ikke viser noen, eller kun begrenset, inhibitoriske effekter på spontanloko-motering. I motsetning, kan de indusere en liten adferdsaktivering med påfølgende økning i småskalabevegelser, f.eks. stopp i senter av adferdsopptaksarenaen, tilsvarende det som induseres av dopaminerge agonister. Adferdsaktive-ringen er begrenset, og når ikke den uttalte økning i aktivitet som induseres av direkte eller indirekte dopaminerge atonister. På andre side reduserer de foretrukne substanser økning i aktivitet indusert av direkte- eller indirekte dopaminerge agonister, dvs. d-amfetamin og kongenerere.
Således har forbindelsene ifølge foreliggende oppfinnelse overraskende vist en interessant dopaminergisk virkningsprofil med antagonistisk-lignende effekter på hjerne-neurokjemi og milde agonist-lignende effekter på normal adferd, med inhibering av adferd i former av hyperaktivitet. Virkningsprofilen antyder modulatoriske effekter på dopaminerge funksjoner, klart forskjellig fra kjente forbindelser som tilhører disse kjemiske klasser eller effekter antatt for typiske dopaminreseptorantagonister eller -agonister fra disse eller andre kjemiske klasser.
Gitt innvirkningen av dopamin i et bredt spekter av CNS-funksjoner og mangelen på kliniske preparater som synes å virke for dopaminsystemene, vil den nye klasse av dopaminerge modulatorer som presenteres med foreliggende oppfinnelse vise seg å være overlegne dagens kjente dopaminerge forbindelser i behandling av flere forstyrrelser relatert til dysfunksjoner i CNS, i termer av effektivitet og likeledes bieffekter.
Noen forbindelser i samsvar med foreliggende oppfinnelse er funnet å ha overraskende gode farmakokinetiske egenskaper, inkluderende høy oral biotilgjengelighet. De er således stabile for fremstilling av oralt administrerte farmasøytika. Det er ingen retningslinjer i teknikkens stilling for å oppnå forbindelser med denne effekt på dopaminsystemer i hjernen.
Detaljert beskrivelse av oppfinnelsen Farmakologi
Det foreligger bevis på at neurotransmisjon i CNS er forstyrret i psykiatriske og neurologiske sykdommer. I mange tilfeller, f.eks. i schizofreni eller Parkinson's sykdom, er farmakoterapier basert på antagonisme eller agonisme ved dopaminreseptorene nyttige, men ikke optimale. I de senere år er det gjort mye for å finne nye og selektive ligander for dopaminreseptorsubtyper (Di, D2, D3, D4, D5) med det som formål å forbedre effektivitet, og å redusere bieffekter.
Den foreliggende oppfinnelse tilveiebringer et nytt prinsipp for nye terapeutika basert på interaksjoner med dopaminsystemer. Forbindelsene ifølge oppfinnelsen har effekter på hjerne-neurokjemi tilsvarende til antagonister ved dopamin D2-reseptorer. I motsetning til for tiden an-vendte dopaminreseptor-antagonister, viser forbindelsene ifølge oppfinnelsen ingen, eller begrensede inhibitoriske effekter på spontan bevegelse. De kan indusere adferdsaktivering med ledsagende økning i småskala-bevegelser, f.eks. stopp i senter av adferdsopptaksarena, tilsvarende til det som induseres av dopaminerge agonister. Adferdsaktiviteten er begrenset, og når ikke opp til den utstrakte økning i aktivitet indusert av direkte eller indirekte dopaminreseptoragonister. Overraskende viser det seg at foretrukne substanser faktisk kan redusere økningen i aktivitet indusert av direkte eller indirekte dopaminerge agonister, dvs. d-amfetamin og kongenerer.
De foretrukne strukturer er substituert i metaposisjon på den aromatiske ring. Et eksempel på en slik forbindelse er metansulfonisk syre 3-(l-propyl-piperidin-4-yl)-fenylester, som er vist i Eksempel 14 nedenfor. I rotter øker denne forbindelse 3,4-dihydroksyfenyl-eddiksyre i striatum fra 1265174 (kontroller) til 3208±236 ng/g vev 50

I rotter øker denne forbindelse 3,4-dihydroksy-fenyleddiksyre i striatum fra 914 ± 19 (kontroller) til 1703119 ng/g vev ved 50 jomol/kg s.c. Denne økning i dopamin-turnover etterfølges av en trend mot en økning i motoraktivitet fra 2030 ± 299 cm/60 min., til 28791398 cm/60 min., p=0,14. I dyr som er habituert til motilimeterboksen, øker forbindelsene beskrevet i Eksempel 6, 4-(3-metansulfonyl-fenyl)-lpropyl-piperidin, adferdsaktiviteten fra 476 ± 279 cm/60 min.

Syntese av forbindelsen ifølge foreliggende oppfinnelse utføres med fremgangsmåter som er konvensjo-nelle for syntese av beslektede kjente forbindelser. Syntese av forbindelsene i Formel 1, generelt, omfatter reak-sjon av et mellomprodukt som forsynes til alkylgrupper med et mellomprodukt av piperidin eller piperazin som forsynes til amingruppen i Formel 2:
En hensiktsmessig metode for å syntetisere de foreliggende forbindelser er ved anvendelse av et alkyljodid (f.eks. 1-propyl-jodid). Alternativt kan andre frigivelsesgrupper i tillegg til jodid anvendes på alkylgruppen, selvsagt, slik som sulfonater, spesielt metansulfonat eller toluensulfonat, brom og lignende. Katalysatoren, fortrinnsvis Pd, vil ha evnen til å danne ligandkomplekser og undergå oksidativ addisjon. Det er videre mulig å omdanne et radikal Ri i en forbindelse av Formel 1 til et annet radikal Ri, f.eks. ved å oksidere metylsulfid til metylsulfon (f.eks. med m-klorperoksybenzoisk syre), substitusjon av et triflat eller halidgruppe med en cyanogruppe (f.eks. palladium-katalysert cyanering), substituering av triflat eller halidgruppe med et keton (f.eks. palladium-katalysert Heck-reaksjon med butyl-vinyl eter), substituering av et triflat eller halidgruppe med et karboksamid (f.eks. palladium-katalysert karbonylering), eller spalting med en eter ved f.eks. å omdanne hydroksylderivat, som videre kan omdannes til korresponderende mesylat eller triflat. Illustrerende syrer er svovelsyre, nitratsyre, fosforsyre, saltsyre, sitronsyre, eddiksyre, melkesyre, tartarsyre, palmoisk syre, etan-disulfonisk syre, sulfamisk syre, sukkinisk syre, cykloheksylsulfamisk syre, fumarsyre, maleinsyre, og benzosyre. Disse salter fremstilles enkelt ved kjente fremgangsmåter.
De farmasøytiske materialer som inneholder en forbindelse i samsvar med oppfinnelsen kan også omfatte substanser anvendt for å fremme produksjon av det farmasøy-tiske preparat eller administreringen av preparatene. For å produsere farmasøytiske preparater inneholdende forbindelsen ifølge oppfinnelsen i form av doseringsenheter for oral applisering, kan de utvalgte forbindelser blandes med en faststoffeksipient, f.eks. laktose, sakkarose, sorbitol, mannitol, stivelser så som potetstivelse, kornstivelse eller amylopektin, cellulose-derivater, et binde-middel så som gelatin eller polyvinylpyrrolidin, og et smøremiddel så som magnesiumstearat, kalsiumstearat, polyetylenglykol, vokser, parafin, og lignende, og deretter kompresseres til tabletter. Dersom belagte tabletter er nødvendig, kan kjernene, fremstilt som beskrevet ovenfor, belegges med en konsentrert sukkerløsning som kan inneholde f.eks. gummi arabicum, gelatin, talk, titaniumdioksid, og lignende. Alternativt kan tabletten belegges med en polymer kjent for fagkyndige innen fagfeltet, oppløst i lett flyktige organiske løsemidler eller blandinger av organiske løsemidler. Fargestoff kan tilsettes til disse belegninger for å enkelt skille mellom tabletter som inneholder forskjellige aktive substanser eller forskjellige mengder av den aktive forbindelse.
For fremstilling av myke gelatinkapsler kan den aktive substans samblandes med f.eks. en vegetabilsk olje eller polyetylenglykol. Harde gelatinkapsler kan inneholde granuler av den aktive substans ved anvendelse enten av de angitte eksipienter for tabletter, f.eks. laktose, sakkarose, sorbitol, mannitol, stivelse (f.eks. potetstivelse, kornstivelse eller amylopektin), cellulosederivater eller gelatin. Også væsker eller semifaststoffer av medikamentet kan fylles inn i harde gelatinkapsler.
Doseringsenheter for rektal applisering kan være opp-løsninger eller suspensjoner, eller kan fremstilles i form av stikkpiller omfattende den aktive substans i blanding med en nøytral fettbase, eller gelatinrektale kapsler omfattende den aktive substans i samblanding med vegetabilsk olje eller parafinolje. Væskepreparater for oral applisering kan være i form av siruper eller suspensjoner, f.eks. løsninger inneholdende fra ca. 0,2% til ca. 20 vekt% av aktiv substans heri beskrevet, idet balansen er sukker og blandinger av etanol, vann, glyserol og propylenglykol. Valgfritt kan slike væskepreparater inneholde fargemidler, smakstilsetningsmidler, sakkarin og karboksymetylcellulose som et fortykningsmiddel, eller andre eksipienter kjent innen fagfeltet.
Løsninger for parenteral applisering ved injisering kan fremstilles i en vandig løsning av et vannløselig farmasøytisk akseptabelt salt av den aktive substans, fortrinnsvis i en konsentrasjon på fra 0,5 til ca. 10 vekt%. Disse løsninger kan også inneholde stabiliserende midler og/eller buffermedier, og kan hensiktsmessig til-veiebringes i forskjellige doseringsenhetsampuller. Anvendelse og administrering til en pasient som skal behandles klinisk vil være åpenbar for fagkyndige innen fagfeltet.
I terapeutisk behandling er en effektiv mengde eller en terapeutisk mengde av forbindelsene ifølge oppfinnelsen fra ca. 0,01 til ca. 500 mg/kg kroppsvekt daglig, fortrinnsvis 0,1-10 mg/kg kroppsvekt daglig. Forbindelsene kan administreres på enhver egnet måte, så som oralt eller parenteralt. Den daglige dosering vil fortrinnsvis administreres i individuelle doseringer 1-4 ganger daglig.
Det er kjent for fagkyndige at å erstatte et hydrogen i en ikke-substituert posisjon i den aromatiske ring med et fluoridatom, kan blokkere muligheten for enzymatisk hydroksylering som gir forbindelsen lav oral biotilgjengelighet. Denne type forandring (H til F) forandrer sjelden den farmakologiske profil. Således er det meget viktig i noen tilfeller å introdusere et fluoridatom i én ikke-substituert posisjon i den aromatiske ring av forbindelse av Formel 1 for å forbedre den orale biotilgjengelighet.
Den foreliggende oppfinnelse illustreres videre i eksemplet nedenfor, som på ingen måte er tiltenkt å begrense oppfinnelsens ramme.
Eksempel 1: 1-( 3- metansulfonyl- fenyl)- 4- propyl-piperazin
En suspensjon av 1-(3-metansulfonyl-fenyl)-piperazin (350 mg) og malt K2C03 (403 mg) ble omrørt i CH3CN (25 ml) ved romtemperatur. 1-jodpropan (712 ul) ble tilsatt. Blandingen ble reflukset over natten. Reaksjonsblandingen ble filtrert, og flyktige materialer ble evaporert in vacuo. Den olje-aktige rest ble kromatografert på en silikakolonne med MeOH:CH2Cl2 (1:30 (v/v)) som elueringsmiddel. Innsamling av fraksjonene som inneholder rent produkt, og evaporering av løsemidlet, ga ren 1-(3-metan-sulf onyl-f enyl) -4-propyl-piperazin (220 mg). Aminet ble om-dannet til HCl-saltet og rekrystallisert fra etanol/ dietyleter: Smp. 233°C MS M/Z (rel. intensitet, 70 eV) 282 (M<+>, 30), 254 (15), 253 (BP), 210 (17), 70 (21).
De følgende forbindelser i samsvar med eksemplene 2 og 3 ble fremstilt på en måte tilsvarende til det ene beskrevet i Eksempel 1.
Eksempel 2 : 1 - propyl- 4-( 3- trifluormetansulfonyl-fenyl) - piperazin
MS M/Z (rel. intensitet, 70 eV) 336 (M<+>, 16), 307 (bp), 77 (18), 70 (38), 56 (23).
Eksempel 3 : l-[ 3- ( 4- propyl- piperazin- l- yl) - fenyl]-etanon
Utgangspunktet var 1-(3-piperazin-l-yl-fenyl)-etanon og n-Pr-I: Smp. 119°C (oksalat), MS M/Z (rel. intensitet, 70 eV) 246 (M<+>, 10), 217 (33), 132 (18), 70 (bp), 56 (41): Rf=0, 23 (EtOAc) .
Eksempel 4 : Metansulfonisk syre 3-( 1- propyl-piperidin- 4- yl) - fenyl- ester
En løsning av 3-(l-propyl-piperidin-4-yl)-fenol (340 mg) og trietylamin (187 mg) i 20 ml CH2C12 ble avkjølt til 0°C. Deretter ble metansulfonylklorid (194 mg) oppløst i 10 ml CH2C12, tilsatt dråpevis. Reaksjonsblandingen ble tillatt å nå romtemperatur, og deretter omrørt i 2,5 timer ved 25°C. Reaksjonsblandingen ble til slutt stoppet med vann. Det organiske sjikt ble separert, og vasket med 10% HC1, og deretter 10% Na2C03.
Etter tørking (MgS04) ble løsemidlet fjernet under redusert trykk. Resten ble kromatografert på en silikakolonne ved anvendelse av MeHO:CH2Cl2 (1:9 (v/v)) som elueringsmiddel. Fraksjonene som inneholder ren metansulfonisk syre 3-(l-propyl-piperidin-4-yl)-fenylester ble innsamlet, og løsemidlet ble fjernet in vacuo, noe som ga 206 mg av tittelforbindelsen. MS M/Z (rel. intensitet, 70 eV) 297 (M<+>, 3), 268 (bp), 189 (24), 131 (13), 79 (16). Den følgende forbindelse i eksempel 5 ble fremstilt på tilsvarende måte som det ene beskrevet i Eksempel 4.
Eksempel 5: Metansulfonisk syre 3-( 4- propyl-piperazin- l- yl) - fenyletylester
Utgangspunktet var 3-(4-propyl-piperazin-l-yl)-fenol og metansulfonyl-klorid: Smp. 143-144°C (fumarat), MS M/Z (rel. intensitet, 70 eV), 298 (M<+>, 35), 269 (95), 121 (25), 84 (30) , 70 (bp) .
Fremstilling 1: 1- ( 3- metansulfonyl- fenyl) - piperazin
En blanding av l-brom-3-metansulfonyl-benzen (0,8 g), piperazin (1 g), natrium-tert-butoksid (0,5 g), BINAP
(42 mg) og [Pd2(dba)3] (38 mg) i toluen (7 ml) ble oppvarmet under argon ved 80°C i 24 timer. Etter avkjøling til romtemperatur, ble løsemidlet evaporert til tørrhet. Rå-materialet ble renset med flashkromatografi på silikagel ved anvendelse av EtOAc: Utbytte ga 0,48 g: MS M/Z (rel. intensitet, 70 eV), 240 (M<+>, 17), 199 (12), 198 (bp), 119 (9), 56 (7) .
Fremstilling 2 : 1-( 3- trifluormetansulfonyl- fenyl)-piperazin
Med utgangspunkt i 3-brom-trifluormetansulfonyl-benzen og piperazin ble tittelforbindelsen DMP fremstilt med prosedyren beskrevet i Fremstilling 1: MS M/Z (rel. intensitet, 70 eV), 294 (M<+>, 22), 252 (bp), 119 (32), 104 (10), 56 (15). (45).
Fremstilling 3 : 1-( 3- piperazin- l- yl- fenyl)- etanon
Med utgangspunkt i 3-brom-acetofenon og piperazin ble det angitte cmp fremstilt med prosedyren beskrevet i Fremstilling 1: MS M/Z (rel. intensitet, 70 eV), 204 (M<+>, 5), 162 (35), 77 (30), 57 (35), 56 (bp).
In vivo test: Adferd
For adferdstesten ble dyrene plassert i separate bevegelighetsmålebokser (50x50x50 cm) utstyrt med et arrangement av 16x16 fotoceller (Digiscan activity monitor, RXYZM (16) TAO, Omnitech Electronics, USA)), tilkoblet til Omnitech Digiscan-analyserer og en Apple Macintosh datamaskin utstyrt med et digitalt grensesnittkort (NB DIO-24, National Instruments, USA). Adferdsdata fra hver bevegelithetsmåleboks, representerende posisjonen
(gravitasjonssenter) av dyret ved hvert tidspunkt, ble opptatt med en samplingsfrekvens på 25 Hz og innsamlet ved anvendelse av en konvensjonell LABView™-applikasjon. Dataene fra hver opptakssesjon ble analysert med hensyn til distanse tilbakelagt, og små-skala bevegelser, f.eks. stopp i senter av adferdsopptaksareaen, under opptakssesjonen. For å bestemme stopp i senteret beregnes hastighet ved hvert tidspunkt som avstanden tilbakelagt siden foregående prøve, delt med tiden som er gått siden foregående prøve. Antallet stopp beregnes deretter som antallet ganger hastigheten forandres fra ikke-null verdi til null. Antallet stopp i senter av adferds-opptaksarenaen beregnes som antallet stopp som foregår ved en posisjon minst 10 cm fra kantene av opptaksarenaen. For adferds-testing av habituerte rotter, ble dyrene plassert i bevegelsesmålebokser 30 min. for administrering av testforbindelsen. Hver adferds-opptakssesjon varte 60 eller 30 min., og startet umiddelbart etter injisering av testforbindelsene. Tilsvarende adferds-opptaksprosedyrer ble anvendt for ikke-habituerte rotter, habituerte rotter og medikament-behandlede rotter. Rotter pre-behandlet med d-amfetamin gis dosen 1,5 mg/kg s.c. 5 minutter før adferdsels-sesjonen i motilimeteret. Rotter forbehandlet md dizolcipin (Mk-801) gis doseringen 0,7 mg/kg i.p. 90 min. før adferdssesjonen i bevegelsesmåleren.
In yivo- test: Neurokjemi
Etter adferdsaktivitets-sesjonene, ble rottene de-kapitert, og deres hjerner hurtig tatt ut og lagt på iskald petriskål. Den limbiske forhjerne, striatum, frontal cortex og den resterende hemisfæriske del av hver rotte ble dissikert og frosset. Hver hjernedel ble deretter analysert med hensyn til dets innhold av monoaminer og deres metabolitter. Monoaminerge indisier som ble analysert var dopamin (DA), 3,4-dihydroksy-fenyl-eddiksyre (DOPAC), homovanillisk syre (HVA), 3-metoksytyramin (3-MT), serotonin (5-HT), 5-hydroksyindol-eddiksyre (5-HIAA) og noradrenalin (NA). Alle monoaminergiske indisier i det dissikerte vev ble analysert ved hjelp av HPLC med elektrokjemisk deteksjon som beskrevet av K. Svensson et al., 1986, Naunyn-Schmiedeberg's Arch. Pharmacol., vol 334, p 234-245 og inkorporert med referanse heri.
In vivo- test: Farmakokinetikk i rotten
For å bestemme oral tilgjengelighet (F) og plasma-halveringstid (tl/2) for testforbindelsene ifølge oppfinnelsen, ble eksperimenter utført i rotten. På dag 1 ble rottene implantert med ett kateter i den jugulare vene og ett kateter i den karotide arterie under ketamin-anestesi. På dag 3 injiseres testforbindelsen eller oralt eller i det jugulare venekateter. Blodprøver innsamles i løpet av 8 timer fra det arterielle kateter. Blodprøvene ble hepa-rinisert og sentrifugert. Plasma opptas fra de sentri-fugerte prøver, og fryses. Nivåene av testforbindelser ble deretter bestemt i hver prøve ved hjelp av et gass-kromatografi-massespektrometer (Hewlett-Packard 597MSD). Plasmaprøvene, tatt fra rottene på Sprague-Dawley-stammen, 0,5 ml, ble fortynnet med vann (0,5 ml), og 30 pmol
(50 (il) ( (-)-S-3-(3-etylsulfonylfenyl)-N-n-propylpiperidin som indre standard ble tilsatt. pH ble justert til 11,0 ved tilsetning av 25 ul mettet Na2CC>3. Etter blanding ble prøvene ekstrahert med 4 ml diklormetan ved risting i 20 min. Det organiske sjikt ble, etter sentrifugering, over-ført til et mindre rør og evaporert til tørrhet under en strøm av nitrogen og deretter oppløst på nytt i 4 0 ul toluen for GC-MS-analyse. En stahdardkurve over området 1-500 pmol ble fremstilt ved å tilsette egnede mengder testforbindelse til blanke plasmaprøver. GC ble utført på en HP-Ultra 2-kapillærkolonne (12 mm x 0,2 mm ID), og 2 ul
ble injisert i splitless-modus. GC-temperaturen ble holdt ved 90°C i 1 minutt etter injisering, og deretter øket ved 30°C/min. til den finale temperatur på 290°C. Hver prøve ble kjørt i duplikat. Den laveste detekterbare konsentrasjon av testforbindelsen var generelt funnet å være 1 pmol/ml.
Claims (28)
1. 3-substituert 4-(fenyl-N-alkyl)-piperazin-forbindelse, karakterisert ved at forbindelsen har Formel 1:
hvor: Ri er valgt fra gruppen som består av OS02CF3, OSO2CH3, SOR3, SO2R3, COCH3, og COCH2CH3, hvor R3 er som definert nedenfor, R2 er valgt fra gruppen som består av Ci-C4-alkyler, allyler, CH2CH2OCH3, CH2CH2CH2F, CH2CF3, 3,3,3-trifluorpropyl og 4,4,4-trifluorbutyl, R3 er valgt fra gruppen som består av Ci~C3-alkyler, CF3 og N(CH3)2, med det forbehold at når Ri er SO2R3 og R3 er et C1-C3 alkyl, er R2 ikke CH2CH2OCH3, eller et farmasøytisk akseptabelt salt derav.
2. Forbindelse i samsvar med krav 1, karakterisert ved at Ri er valgt fra gruppen som består av OS02CF3, OS02CH3, S02CH3, S02CF3, COCH3, og S02N(CH3)2.
3. Forbindelse i samsvar med et av kravene 1-2, karakterisert ved at R2 er valgt fra gruppen som består av n-propyl og etyl.
