Abstract
Den foreliggende oppfinnelse dreier seg om anvendelse av forbindelser som har evne til å. forskyve tritiert cis-N-cykloheksyl-N-etyl-[3-(3-klor-4-cykloheksylfenyl)allyl]amin fra dets reseptorer for fremstilling av farmasøytiske forbindelser som planlegges for å motvirke celleproliferasj on.
Description
Den foreliggende oppfinnelse dreier seg om en ny anvendelse av forbindelser, for fremstilling av en farmasøytisk blanding som har en god affinitet til reseptorene som binder cis-N-cykloheksyl-N-etyl-[3-(3-klor-4-cyklo-heksylfenyl)allyl]amin, samt til farmasøytiske blandinger som har de samme egenskaper.
cis-N-cykloheksyl-N-etyl[3-(3-kloro-4-cyklo-heksyifenyl)allyl]amin, også kjent under koden CM 31747 eller SR 31747 og nedenfor her omtalt som «SR 31747» blir beskrevet i EP 376,850 som også diskuterer forbindelsens immundempende aktivitet. Det har også blitt funnet at SR 31747 hindrer kreftceller fra å proliferere og at forbindelsen således kan fremvise anti-tumoraktivitet.
Artikkelen til P.J. Brent etal, Eur. Pharmacol., 278,151-160 (1995) beskriver evnen til visse sigma-reseptoiiigander for inhibering av celleproliferasjon av cellelinjer, brystcarcinomer og kolon-carcinomer. Det indikeres også i denne artikkel at tester på celleproliferasjon varierer signifikant avhengig av sigmabindingen, av dosen som benyttes og av den aktuelle cellelinje som undersøkes. Det understrekes derfor at det ikke er funnet noen korrelasjon mellom den inhiberende effekt på cellevekst utvist ved de testede sigmaligander og deres affinitet for sigma-reseptorer.
Publikasjonen FR-A-2 445 889 foreslår aminoderivater som analgesiske og antidepressiva. I denne publikasjonen omtales ikke forbindelsen SR 31747.
Publikasjonen FR-A-2 016 169 beskriver cyklopropankarboksylsyrederivater med anti-inflammatoriske og analgesiske egenskaper.
B. Bourrié etal. Rapporterer i deres artikkel i Immunotogy, 88,389-393 (1996), at SR 317447 har en signifikant affinitet til sigma reseptorer uttrykt på lymfocytter.
Foreliggende oppfinnelse angår anvendelse av cis N-cykloheksyt-N-etyl-[3-(3-kior-4-cykloheksylfenyl)allyl] amin eller et farmasøytisk akseptabelt salt derav eller solvat derav, for fremstilling av en farmasøytisk blanding for behandling av glioblastomer, neuroblastomer, lymfomer, myelomer, leukemier, carcinomer i kolon, kolorektal-, epitelial-, hepatisk-, pulmonal-, bryst-, ovarie-, pankreatiske- eller protatacarcinomer.
Spesielt angår foreliggende oppfinnelse anvendelse som nevnt over, hvor celleproliferasjonen er hormonsensitiv eller, hormoninsensitiv.
Videre angår oppfinnelsen farmasøytiske blandinger, inneholdende cis-N-cykloheksyl-N-etyl-[3-(3-klor-4-cykloheksylfenyl)allyl] amin eller et farmasøytisk akseptabelt salt derav eller solvat derav for anvendelse som et medikament for behandling av glioblastomer, neuroblastomer, lymfomer, myelomer, leukemier, carcinomer i kolon, kolorektal-, epitelial-, hepatisk-, pulmonal-, bryst-, ovarie-, pankreatiske- eller protatacarcinomer.
Evnen som den aktuelle forbindelse har til å fjerne tritiert SR 31747 fra dets reseptorer, kan lett bestemmes biokjemisk ved å bruke <3>H-SR 31747 og binde denne til cellene.
Denne bestemmelse kan utføres med tumorceller som er utvalgt på fornuftig måte, fortrinnsvis fra cellelinjer som lett proliferer in vitro, for eksempel humant myelom, nyrekreft eller humane lungeceller eller alternativt på brystkreftcefler.
I den foreliggende oppfinnelses sammenheng ble en human brystkreft-cellelinje «MCF-7», utvalgt på tilfeldig basis for å kunne utføre bestemmelsene under standardiserte betingelser, noe som gjør det mulig å få konstante og reproduserbare resultater.
I sammenheng med den foreliggende oppfinnelse ble <3>H-SR 31747 også valgt tilfeldig, der tritium btir bundet til vinylenbindingen, men SR 31747 kan bli merket på enhver mulig måte siden merkingen bare skal hjelpe med å følge forskyvningen av produktet vekk fra dets reseptorer.
Bestemmelse av evnen til å skyve vekk <3>H-SR 31747 fra dets reseptorer som er tilstede på cellene, spesielt celler i MCF-7 cellelinjen, ble utført ved å teste total binding og spesifikk binding.
Generelt kan ethvert produkt som når det blir utsatt for den ovenfor beskrevne preliminære undersøkelse, har evne til å fortrenge <3>H-SR 31747 fra dennes reseptorer, bli benyttet til fremstilling av farmasøytiske blandinger som virker mot celleproliferasjon. SR 31747 er for det første i stand til å forskyve <3>H-SR 31747 fra dets reseptorer og har kraftig inhibitorisk virkning på celleproliferasjon.
Ifølge biokjemiske og farmakologiske studier gir eksponering av forbindelsen som benyttes ifølge oppfinnelsen overfor normale celter, under de samme betingelser som de som gjør det mulig å fremskaffe en antiproliferativ aktivitet på kreftceller, ikke opphav til noen skadelig effekt på alle de analyserte kriterier, slik som for eksempel integritet av strukturene og cellefunksjonene eller vedlikehold av levedyktighet. Denne forbindelse og blandinger inneholdende denne fungerer således med stor spesifisitet i relasjon til tumorceller. Anti-tumoraktivitet ble etablert på en rekke humane tumorlinjer in vitro og in vivo i mus. Cellene som ble benyttet ble alle fremskaffet fra den internasjonale ATCC samling. MCF-7-celler ble benyttet for å utføre bindingsstudiene.
Membranene ble fremstillet som følger: 10<9> MCF-7-celler ble homogenisert i 10 sekunder på en Polytron® i 10 ml Hepes® buffer, pH=7,4, som inneholdt: 210 mM D-mannitol, 70 mM sukrose, 1 mM EDTA, 0,3 mM PMSF. Homogenatet ble sentrifugert ved 650 xg i 15 minutter, og supernatanten ble deretter tatt og sentrifugert i 1 time ved 100.000 xg. Bunnfallet ble resuspendert i Tris-HCI-buffer, pH=7,4, ved en konsentrasjon på 1 mg/ml og lagret ved -70°C. Den totale binding blir utført i et 5 ml reagensrør som fikk følgende ingredienser: 50 uL membransuspensjon som inneholdt 10 til 50 ug proteiner 175 ul_ 50 mM Tris-buffer, pH=7,4 25 u-L 20 nM <3>H SR 31747 i 50 mM Tris, pH=7,4 + 0,1% BSA (bovint serumalbumin).
Den ikke-spesifikke binding blir analysert ved å tilsette 25 u.L 10"<5> M SR 31747 til den ovenfor beskrevne løsning (sluttvolum 250 uL) og deretter inkubasjon i 30 minutter ved 25°C. De fri og bundne fraksjoner blir separert ved å plassere 200 ut-på en søyle som inneholder 1 ml Sephadex® LH 20, og radioaktiviteten som inneholdes i de første 2 ml som fremskaffes ved eluere søylene med 50 mM Tris-butter, pH=7,4, blir tellet.
Blant de andre celler som ble benyttet ble de følgende cellelinjer dyrket på standard måte i RPMI-medium supplementert med 10% føtalt kalveserum (FCS): humane myelomer U266 og RPMI 8226

Anti-tumoraktivitet blir målt ifølge den kolorimetriske metode som bruker MTT, 3-(4,5-dimetyltiazol-2,5-difenyltetrazolium)bromid, som beskrevet av Mosmann, T., Joum. of Imm. Methods; 1983, 65, 55.
Denne kolorimetriske analyse gjør det mulig å måle kvantitativt anti-tumoraktiviteten til en løsning som inneholder forbindelsen som benyttes ifølge oppfinnelsen.
Ifølge den prosedyren som ble benyttet, blir de suspenderte celler (slik som myelomcellene) inokulert ved 2 x 10<5> celler/ml i 1 ml brønner i definert medium. De adherente celler (slik som MCF-7-cellene) blir inokulert ved 5 x 104 celler/ml i 1 ml brønner i RPMI-medium pluss 0,5% FCS over natten. Den neste dag blir cellelaget vasket to ganger og mediet blir byttet ut med det definerte medium.
Det definerte medium tilsvarer: RPMI + 10 ug/ml insulin + 10 u.g/ml humant transferrin.
For å utføre denne analyse blir celler vedlikeholdt i nærvær av løsningen som inneholder forbindelsen som benyttes ifølge oppfinnelsen i 5 dager, og MTT blir deretter tilsatt til kulturmediet. 5 dagers perioden som ble valgt tilsvarer det optimale tidspunkt for aktiviteten. Etter dyrkning i 5 dager blir proliferasjonen av cellene målt ved testen som er beskrevet over. Optisk tetthet ved 570 nm blir målt. En blå farve utvikles i brønnene der cellene fremdeles er i live. Intensiteten av farvereaksjonen er proporsjonal til antall levende celler.
Resultatene som ble oppnådd blir beskrevet i Tabell A nedenfor.
Det sees at konsentrasjoner så lave som 1 nM opptil 1 u.M er tilstrekkelig for å indusere 50% hemming av veksten, og at denne effekt blir oppnådd på alle tumorcellene som ble studert.
Effekten som observeres in vivo med hemming av en MCF-7-cellelinje ble studert in vivo i «nakne» mus, intraperitonealt i doser mellom 3 bg 100 mg/kg i en modell som følger Neri, C, etal., Cancer Research, 1990, 50: 5892-5897 og ifølge Berebbi, M., etal., Oncogene, 1990, 5: 505-509.
En annen studie ble også utført for å bestemme effekten av SR 31747 på in vitro og in vivo vekst av bryst- og prostata-epiteltumorceller.
Studien besto av evaluering av den potensielle anti-tumoraktivitet som
SR 31747 har på veksten av en rekke brystkreft-epitellinjer. Studien blir utført in vitro på cellekulturer, samt også in vivo etter inokulering av tumor-brystcellelinjer i «nakne» thymusløse mus.
Cellene som ble benyttet er som følger:
Den hormonsensitive MCF-7 cellelinje stammer fra brysthulevæskeeffusjon av et brystadenokarsinom. De anti-østrogenresistente MCF-7LY2-celler ble fremskaffet fra MCF-7-linjen ved selektivt press i nærvær av et høyaffinitets anti-østrogen. MCF-7LCC1- og MCF-7LCC2-linjene ble utviklet i nakne mus, etter inokulering av MCF-7-cellene. De to linjene er østrogenuavhengige. MCF-7LCC2-cellene er også anti-østrogen motstandsdyktige. MCF-7AZTD5-linjen er avledet fra MCF-7-linjen etter stabil transfeksjon med H-ras onkogenet. MCF-7AZTD5-cellene er anti-østrogenresistente in vitro. Alle disse linjer blir vedlikeholdt på DMEM/F12 (1/1, vol/vol) som inneholder 10% dekomplementert føtalt kalveserum og 16 ng/ml humant insulin. MCF-7LCC2-linjen blir dyrket i permanent nærvær av 10'<7> M hydroksytamoxifen. MDA-MB231 tumor-brystepitelcellene er østrogen- og anti-østrogen insensitive. De dyrkes i L15-medium forsterket med 10% FCS, 1% essensielle aminosyrer og 10 u.g/ml humant insulin.
InCaP, PC3 og DU145 epitellinjene er avledet fra prostatakreftsvulster. Bare LnCaP-cellene er hormonsensitive. De tre cellelinjer blir dyrket i RPMI forsterket med 10% FCS.
Alle linjene blir vedlikeholdt i 37°C i en fuktig luft/C02 (95%/5%) atmosfære, med unntak av MDA-MB231-cellene, som blir dyrket i fravær av CO2. Cellelinjene blir kontrollert regelmessig med henblikk på fravær av mykoplasmer.
Inokulering og behandling av dyrene ble utført som følger:
Dyrene som benyttes er «nakne» athymiske hunnmus, som har eller ikke har fått fjernet eggstokkene, og som ble fremskaffet ved 4 ukers alder (R. Janvier). Før inokulering med de hormonsensitive brystceller (MCF-7, MCF-7AZTD5), mottok dyrene 10 jxL av 10"<5> M østradiolløsning i etanol, som ble tilført perkutant. Behandlingen ble gjentatt tre ganger i løpet av 1 uke. En million celler suspendert i 100 uL PBS/Ca<2+> blir inokulert subkutant på oversiden av hver av hoftene. Den følgende dag mottok dyrene 200 uL injeksjonsløsning (etanol/Tween 80/fysiologisk saltvann, 1/1/18, vol/vol) som inneholdt de ulike reaktantene i de på forhånd definerte konsentrasjoner. Injeksjonene blir gjort intraperitonealt og blir gjentatt hver dag.
RESULTATER
In vitro aktivitet av SR 31747 på proliferasjon av ulike brystcancer-epitellinjer
Resultatene som dreier seg om effekten av SR 31747 på proliferasjon av
epitelbrystceller i kultur, blir fremvist i Tabell B. Aktiviteten av SR 31747 ble evaluert i nærvær av ulike konsentrasjoner av saltvann. I nærvær av reduserte konsentrasjoner av saltvann fremviste SR 31747 en inhibitorisk virkning på proliferasjon av alle tumor-epitelcellelinjene som ble analysert. Større mengder av saltvann i dyrkningsmediet motvirket delvis eller totalt den inhibitoriske aktivitet som SR 31747 hadde på cellevekst.
Ved en gitt serumkonsentrasjon varierte evnen til SR 31747 i å hemme proliferasjonen også, avhengig av linjen som ble analysert. I nærvær av 0,1% FCS, som gjør det mulig både å vedlikeholde en minimal celleproliferasjon og å observere en følgende aktivitet av SR 31747, varierte IC50 bestemt for de hormonfølsomme MCD-7-linjer (MCF-7, MCF-7AZ, MCF-7 22HTB) i områdene mellom 2 x 10<*9> M til 7 x 10"<8> M. Lignende IC5o-verdier ble også fremskaffet for MDA-MB231 og MCF-7LCC-cellene. På den annen side viste MCF-7AZTD5- og MCF-7LY2-linjene større følsomhet overfor den inhibitoriske virkning av SR 31747, siden ICso-verdiene som ble bestemt er 2 x 10"<10> til 5 x 10"<12>.
I alle tilfelle var de tilsynelatende totale hemninger av celleproliferasjon som ble observert i nærvær av 10'<6> M og noen ganger 10"<7> M SR 31747, basert på "all evidens", celletoksiske effekter.
Resultatene fremskaffet indikerer også at SR 31747 ikke har evne til å reversere den stimulatoriske effekt av østradiol på den hormonfølsomme MCF-7-cellelinje.
In vivo effekt av SR 31747 på frekvens og størrelse av tumorer utviklet etter inokulering av brystkreftepitelcellelinjer i «nakne» mus
Figurene 1 og 2 viser effekten av SR 31747 på tumorer utviklet i «nakne» mus uten eggstokk, fremkalt av MDA-MB231-celler.
Hver mus mottar 5x10<6> MDA-MB231 -celler over hver hofte (dvs. 2 punkter med celleinokulasjoner per mus). Dyrene blir deretter daglig behandlet intraperitonealt, og mottar 500 u.g SR 31747 + 0,003 ug E2 (20 mus). Kontrollgruppen mottar 0,003 ug E2 (20 mus). Mengden (som prosentdel av inokulerte tumorer) og størrelse på tumorene som utviklet seg, blir bestemt ved ulike tidspunkter i løpet av behandlingen. Ved behandlingens slutt (dag 92) ble dyrene avlivet og veiet. Tumorene ble fjernet og veid.
Figurene viser at SR 31747 har en tendens til å redusere størrelsen, vekt og frekvens av tumorer som utvikles i eggstokkfrie nakne mus fra MDA-MB231-celler. Imidlertid er forskjellene i størrelsen og vekt på tumorer mellom kontrollgruppene og SR 31747 ikke tydelig signifikante, på grunn av den store variasjon som ble observert i hver gruppe.
Således har SR 31747 in vitro anti-proliferativ aktivitet på proliferasjon av alle de brystkreftepitelcellene som ble studert. Denne aktivitet er spesielt tydelig i nærvær av reduserte serumkonsentrasjoner, og varierer som en funksjon av de cellelinjer som ble studert. Selv om IC50-verdiene målt i 0,1% FCS i de fleste tilfeller varierte mellom 2x10'<9> M og 2x10'<8> M, ble en ekstrem følsomhet overfor SR 31747 imidlertid observert i MCF-7LY2-cellene.
En generell tendens til SR 31747 i å redusere på den samme tid størrelse, vekt og hyppighet av tumorer utviklet fra MDA-MB231 -celler ble observert. Det ble benyttet in vivo fremstilling av svulster som ble benyttet som mål, for forbindelsen som benyttes ifølge oppfinnelsen, radioaktivt merket for eksempel med et 123l, 1<8>F, <11>C eller <13>N, reseptorene som binder <3>H SR 31747. Grunnen til dette er at den høye tetthet av disse reseptorer i tumorceller har antydet at de med fordel kan benyttes for å binde radioligandene ved bruk av PET (positron emisjonstomografi) og SPECT (enkel foton emisjonstomografi) teknikker.
Det ble brukt forbindelser radioaktivt merket med isotoper, som er velkjent for de som er øvet i faget, som bildefremkallende midler for radioaktiv merking.
Videre kan andre bildefremskapende teknikker som benytter et kompleks med antigenradioaktivt merket antistoff, bli benyttet.
Slike fremgangsmåter inkluderer teknikken kalt SPECT eller teknikken kjent som PET.
Bruk av forbindelsen omfatter også preparering av medisinale produkter beregnet på behandling som dreier seg om målretting av radioaktivt merkede produkter som strålingsfrigjørende midler nær ved, eller inne i, tumoren selv, der de nevnte forbindelser brukes for å målrette radioaktivt merkede produkter mot tumoren eller i dens umiddelbare omgivelser, for å utsette tumorecellene for stråling som sendes ut av de nevnte radioaktivt merkede produkter.
Bruk av forbindelsen kan også omfatte et radioterapitrinn, der cellene i kontakt med forbindelsen blir utsatt for effekten av ioniserende stråling, der i blant for eksempel gammastråling, betastråling, røntgenstråling, eller alfapartikler som kan leveres fra en ytre kilde som for røntgenstrålene eller gammastrålene, eller ved hjelp av radionukleider som blir tilført direkte til pasienten, som beskrevet for eksempel i Principles of Radiation Therapy in Cancer, Principles and Practice of Oncology, Devita, V.T., etal., red. 4. utg. J.B. Lippincort Co. Philadelphia, 1993,15,248-275.
Forbindelsen kan også tilføres i kombinasjon med andre anti-canceraktive midler som er vel kjent for de som er øvet i faget.
Ifølge de biokjemiske og farmakologiske studier som er beskrevet over, ble aktiviteten bestemt for følgende forbindelse:
cis-N-cykloheksyl-N-etyl-t3-(3-klor-4-cykloheksylfenyl)allyl]amin,
eller ett av dens farmasøytisk akseptable salter eller solvater.
Forbindelsen som har evne til å konkurrere vekk <3>H SR 31747 fra dets reseptorer og kan benyttes i terapi i enhver patologisk prosess som inkluderer proliferasjon av tumorceller. Denne celleproliferasjon kan være enten hormonsensitiv eller hormoninsensrtiv.
Mer presist kan kliniske applikasjoner der bruk av denne forbindelsen kan tenkes, omfatte sykdommer som resulterer fra celleproliferasjon, spesielt glioblastomer, nevroblastomer, lymfomer, myelomer, leukemi og colon, colorektale, epiteliale, lever, lunge, bryst, eggstokk, pankreas eller prostata karsinomer.
For disse formål kan forbindelsen som har evne til å konkurrere vekk
<3>H-SR 31747 fra dets reseptorer, spesielt forbindelsen cis N-cykloheksyl-N-etyl-[3-(3-klor-4-cykloheksylfenyl)allyl] amin og dets farmasøytisk akseptable salter, benyttes
for fremstilling av orale, parenterale, sublingvale, transdermale eller lokale farmasøytiske blandinger. Disse farmasøytiske blandinger inneholder den ovenfor nevnte forbindelse, i kombinasjon med et farmasøytisk uvirksomt bæremiddel.
Mer spesifikt dreier den foreliggende oppfinnelse seg ifølge et annet av dets aspekter, om farmasøytiske blandinger som inneholder som aktivt prinsipp forbindelsen cis N-cykloheksyl-N-etyl-[3-(3-klor-4-cykloheksylfenyl)allyl] amin eller et av dets farmasøytisk akseptable salter.
Syrer, både organiske og anorganiske, kan benyttes for å danne addisjonssalter med syrer av aminet cis N-cykloheksyl-N-etyl-[3-(3-klor-4-cykloheksylfenyl)allyl] amin som er ikke-toksiske og farmasøytisk akseptable, spesielt svovelsyre, salpetersyre, fosforsyre, saltsyre, sitronsyre, eddiksyre, melkesyre, tartarsyre, pamoinsyre, etandisulfonsyre, metansulfonsyre, succinsyre, cykloheksylsulfonsyre, fumarsyre, eplesyre og benzosyre.
Hva angår oral eller sublingval tilførsel, blir enkle eller sukkerdekkede
tabletter, gelatinkapsler, granula som kan ha en forsinket frigjøringsvirkning, dråper eller liposomer spesielt benyttet. Hva angår intravenøs, subkutan eller intramuskulær tilførsel, blir sterile eller steriliserbare løsninger benyttet, spesielt for venøs infusjon, mens vanlige plastre kan produseres for transdermal tilførsel. For lokal bruk kan kremer eller væsker som skal smøres ut på huden bli benyttet.
De farmasøytiske blandingene ifølge den foreliggende oppfinnelse kan fremstilles ifølge vanlige fremgangsmåter, som er vel kjent i det tekniske farmasøytiske felt.
Det aktive prinsipp kan inkorporeres inn i bindemidlene som vanligvis benyttes i disse farmasøytiske blandinger, slik som talkum, gummi arabicum, laktose, stivelse, magnesiumstearat, vandige eller ikke-vandige bæremidler, fettsubstanser fra dyr eller planter, parafinderivater, glykoler, ulike våtgjørende, fordelende og emulsifiserende midler, konserveringsmidler, og så videre.
Oppfinnelsens farmasøytiske blandinger kan med fordel inneholde forbindelsen cis N-cykloheksyl-N-etyl-[3-(3-klor-4-cykloheksylfenyl)allyl] amin eller et av dets farmasøytisk akseptable addisjonssalter i kombinasjon med ett eller flere medisinske produkter som er kjent og vanligvis benyttes for de samme terapeutiske indikasjoner.
Mengden av aktivt prinsipp som skal tilføres per dag ifølge den foreliggende oppfinnelses fremgangsmåte, avhenger av den spesifikke terapeutiske indikasjon, alvoret i plagene som skal behandles og pasientens kroppsvekt, samt tilførselsvei. For systemisk tilførsel varierer den totale dose hos mennesket vanligvis mellom 1 og 100 mg per dag, for eksempel fra 2 til 50 mg og mer passende fra 3 til 40 mg per dag.
Enhetsdoseformer for systemisk tilførsel vil vanligvis omfatte fra 3 til 50 mg (nemlig 3, 5,10,20,30,40 og 50 mg produkt). Disse enhetsdoser vil vanligvis bli tilført én eller flere ganger per dag, fortrinnsvis én til tre ganger per dag.
For lokal tilførsel vil de farmasøytiske blandinger generelt inneholde fra 0,0001 til 10% aktivt prinsipp og fortrinnsvis fra 0,01 til 5%.

1. Anvendelse av cis N-cykloheksyl-N-etyl-[3-(3-klor-4-cykloheksylfenyl)allyl] amin eller et farmasøytisk akseptabelt salt derav eller solvat derav, for fremstilling av en farmasøytisk blanding for behandling av glioblastomer, neuroblastomer, iymfomer, myelomer, leukemier, carcinomer i kolon, kolorektal-, epitelial-, hepatisk-, pulmonal-, bryst-, ovarie-, pankreatiske- eller protatacarcinomer.
2. Anvendelse ifølge krav 1, hvor celleproliferasjonen er hormonsensitiv.
3. Anvendelse ifølge krav 1, hvor celleproliferasjonen er hormon-insensitiv.
4. Farmasøytisk blanding, karakterisert ved at den omfatter cis-N-cykloheksyl-N-etyl-[3-(3-klor-4-cykloheksylfenyl)allyl] amin eller et farmasøytisk akseptabelt salt derav eller solvat derav for anvendelse som et medikament for behandling av glioblastomer, neuroblastomer, Iymfomer, myelomer, leukemier, carcinomer i kolon, kolorektal-, epitelial-, hepatisk-, pulmonal-, bryst-, ovarie-, pankreatiske- eller protatacarcinomer.
