Description
Foreliggende oppfinnelse vedrører cykliske peptidanaloger av somatostatin.
Nativt somatostatin består av både en 14-aminosyreisoform (somatostatin-14) og en 18-aminosyreisoform (somatostatin-28). Heiman, et al., Neuroendocrinology, 45:429-436 (1987). Fordi det native somatostatin har kort halveringstid har en rekke somatostatinanaloger blitt utviklet, f.eks. for behandling av akromegali. Raynor, et al., Molecular Pharmacol. 43:838 (1993). Fem ulike somatostatinreseptorer har blitt identifisert og karakterisert. Hoyer, et al., Naunyn-Schmiedebergs Arch. Pharmacol., 350:441 (1994). Somatostatin produserer en rekke effekter, deriblant modulering av hormonfrisetting, f.eks. veksthormon, glukagon, insulin, amylin og neurotransmitter frisetting. Noen av disse effekter har blitt assosiert med binding til en spesifikk somatostatinreseptor. F.eks.. har hemning av veksthormon blitt koplet til somatostatin type-2 reseptoren («SSTR-2») (Raynor, et al., Molecular Pharmacol. 43:838 (1993); Lloyd, et al., Am. J. Physiol. 268:G102 (1995)) mens hemningen av insulin har blitt tilskrevet somatostatin type-5 reseptoren («SSTR-5») (Coy, et al., 197:366-371 (1993)). Oet foretrekkes at man har en analog som er selektiv for den spesielle somatostatinreseptor subtype som er ansvarlig for den ønskede biologiske respons, noe som derved reduserer interaksjon med andre reseptor subtyper som kunne føre tit uønskede bivirkninger.
US patent nr. 4 798 821 og EP 0 021 585 beskriver cykliske somatostatinanaloger som ikke har noen amid- eller aminogruppe bundet til C-terminal ende. DE 2 416 048 beskriver at amidering av somatostatinanaloger ikke reduserer biologisk aktivitet, men lærer ikke at amidering av cykliske somatostatinanaloger øker biologisk aktivitet.

Peptidene fra oppfinnelsen kan benyttes for å hemme frisetting av insulin i et individ (et pattedyr slik som en human pasient). Således er peptidene hensikts-messige for behandling av fysiologiske tilstander der hemning av frisetting av insulin er nyttig, f.eks. diabetes type II. En terapeutisk effektiv mengde av et peptid fra denne oppfinnelse og en farmasøytisk akseptabel substans (f.eks. magnesiumkarbonat, laktose eller et fosfo-lipid som sammen med den terapeutiske forbindelse kan danne en micelle) danner sammen en terapeutisk blanding (f.eks. en pille, tablett, kapsel eller væske) til tilførselen (f.eks. oralt, intravenøst, transdermalt, pulmonalt, vaginalt, subkutant, nasalt, iontoforetisk eller intratrakealt) til et individ som har behov for peptidet. Pillen, tabletten eller kapslene kan dekkes med en substans som har evne til å beskytte blandingen fra mavesyre eller tarmenzymer i individets mavesekk på et tidsrom som er tilstrekkelig til å tillate at blandingen passerer ufordøyet inn i individets tynntarm. Dosen av et peptid fra den foreliggende oppfinnelse beregnet på behandling av de ovenfor nevnte sykdommer eller forstyrrelser varierer avhengig av tilførsels-måten, alder og kroppsvekt hos individet og tilstanden hos individet som skal behandles, og vil tilslutt bli avgjort av den ansvarshavende lege eller veterinær. En slik mengde av peptidet som er bestemt av den ansvarshavende lege eller veterinær blir her referert til som en «terapeutisk effektiv mengde».
Også planlagt å være innenfor området til denne oppfinnelse er et peptid som beskrives av den ovenfor nevnte generiske formel til bruk både for å behandle sykdommer eller forstyrrelser som er assosiert med behovet for å hemme insulin-frisetting, f.eks. diabetes type II, og til bruk i å påvise somatostatinreseptorer, f.eks. radioaktiv visualisering.
Andre egenskaper og fordeler med den foreliggende oppfinnelse vil bli klare fra den detaljerte beskrivelse og fra patentkravene.
Syntese og bruk av somatostatinanaloger ifølge denne oppfinnelse er klart innenfor evnen til en person med normal kompetanse i faget. Dersom det ikke defineres på annen måte har alle tekniske og vitenskapelige ord og begreper som benyttes her den samme betydning som vanligvis forstås av en med normal kompetanse i det fag som oppfinnelsen tilhører.
Det antas at en som er øvet i faget kan, basert på de herværende beskrivelser, benytte den foreliggende oppfinnelse fullt ut. De følgende spesifikke utførelser illustrerer foreliggende oppfinnelse.
Syntese av somatostatinanaloger
Syntese av korte peptider har blitt grundig studert i peptidfaget. Se f.eks. Stewart, et al., Solid Phase Peptide Synthesis (Pierce Chemical Co., 2. utg., 1984). Det følgende er syntesen av Analog I. Andre peptider fra denne oppfinnelse kan fremstilles på en analog måte av en person med normal kompetanse i faget.
Benzylhydrolaminpolystyrenresin (Advanced ChemTech, Inc., Lousville, KY)
(1,1 g, 0,5 mmol) i kloridionformen ble plassert i reaksjonskaret i en Advanced ChemTech ACT 200 peptidsyntesemaskin programmert for å levere de følgende reagenser/løsningsmidler; (a) metylenklorid; (b) 33% trifluoreddiksyre i metylenklorid
(2 ganger, 1 og 25 min hver); (c) metylenklorid; (d) etanol; (e) metylenklorid; og (f) 10% trietylamin i kloroform.
Det nøytraliserte resin ble omrørt med Boc-S-4-metylbenzyl-Cys og diisopropylkarbodiimid (hver 1,5 mmol) i metylenklorid i 1 time, og det resulterende aminosyreresin ble deretter ført gjennom trinnene (a) til (f) i det ovenfor beskrevne vaskeprogram. De følgende aminosyrer (1,5 mmol) ble deretter koplet etter hverandre ved hjelp av den samme prosedyre: Boc-Phe, Boc-O-benzyl-Thr, Boc-N-benzyloksykarbonyl-Lys, Boc-D-Trp, Boc-Phe, Boc-Phe og Boc-S-metylbenzyl-Cys. Etter vasking og tørking veide det avsluttede resin 1,6 g.
Resinet (1,6 g, 0,5 mmol) ble deretter blandet med anisol (5 ml), ditiotreitol (100 mg) og anhydrøst hydrogenfluorid (35 ml) ved 0°C og omrørt i 45 min. Overskudd hydrogenfluorid ble raskt fordampet under en strøm av tørr nitrogen, og det frie peptid ble felt ut og vasket med eter. Det urene peptid ble deretter oppløst i 500 ml 90% eddiksyre som fikk tilført en konsentrert løsning av l2/MeOH til en permanent brun farve ble observert. Overskudd av l2 ble fjernet av tilsetning av askorbinsyre, og løsningen ble fordampet ned til et lite volum som ble applisert på en søyle (2,5 x 90 cm) av SEPHADEX™ G-25 som ble eluert med 50% eddiksyre. Fraksjoner som inneholdt en stor komponent, vurdert med ultrafiolett (UV) absorpsjon og tynnsjiktskromatografi ble deretter slått sammen, fordampet til et lite volum og applisert til en søyle (1,5 x 70 cm) av VYDAC™ oktadecylsilansilika (10-15 nm). Denne søyle ble eluert med en lineær gradient fra 80% A og 20% B til 40% A og 60% B, der A er 0,1% trifluoreddiksyre (TFA) i vann og B er 79,9% acetonitril, 20% vann og 0,1% TFA. Fraksjonene ble analysert ved hjelp av tynnsjiktskromatografi (tic) og analytisk høytrykksvæskekromatografi (hplc) og slått sammen for å gi maksimal renhet. Gjentatte inntørkninger av løsningen fra vann ga 95 mg av produkt som et hvitt, lett pulver.
Produktet ble funnet å være homogen som vurdert ved hplc og tic. Aminosyreanalyse av et surt hydrolysat og matriks-assistert laser desorpsjons masse-spektrometri (MALDI MS) bekreftet sammensetningen av et cykliske oktapeptid (MW kalkulert, 1077; MW funnet, 1080).
Det følgende er syntesen av Analog V. Benzylhydrylamin-polystyrenresin (Advanced Chemtech), Inc.) (0,7 g, 0,25 mmol) i kloridioneformen ble plassert i reaksjonskaret i en Advanced ChemTech ACT 200 peptidsyntesemaskin programmert for å gi de følgende reagenser/løsningsmidler; (a) metylenklorid; (b) 33% trifluroeddiksyre i metylenklorid (2 ganger, 1 og 25 min hver); (c) metylenklorid; (d) etanol; (e) metylenklorid; og (f) 10% trietylamin i kloroform.
Det nøytraliserte resin ble omrørt med Boc-S-metylbenzyl-Cys og diisopropylkarbodiimid (hver 1,5 mmol) i metylenklorid i 1 time, og det resulterende aminosyreresin ble deretter ført gjennom trinnene (a) til (g) i det ovenfor beskrevne vaskeprogram. De følgende aminosyrer (1,5 mmol) ble deretter koplet etter hverandre ved hjelp av den samme prosedyre: Boc-Phe, Boc-O-benzyl-Thr, Boc-N-benzyloksy-karbonyl-Lys, Boc-D-Trp, Boc-Phe, Boc-Phe, Boc-Asn og Boc-S-metylbenzyl-Cys. Etter vasking og tørking veide det avsluttede resin 1,2 g. Peptidresinet ble deretter utsatt for kløving i HF og cyklisering med b som beskrevet over. Søylerensning som beskrevet over ga 21 mg av det cyklilske nonapeptid som ble funnet å være homogen ved hplc og tic. Aminosyreanalyse av et surt hydrolysat og MALDI MS bekreftet sammensetningen av det cykliske nonapeptid (MW kalkulert, 1192; funnet, 1192).
Syntesen av jodinerte somatostatinanaloger på tyrosinresten (f.eks. kloramin-T metoden) er godt beskrevet og er innenfor evnen til en person med vanlig kompetanse i faget. Se f.eks. Czernick, et al., J. Biol. Chem. 258:5525 (1993) og europeisk patent nr. 389,180 B1.
SOMATOSTATINRESEPTOR BINDINGSANALYSE
(1) Humant SSTR-2 bindingsanalyse:
CHO-K1 (ovarie, kinesisk hamster) celler ble fremskaffet fra American Type Culture Collection (ATCC) (Rockville, MD) (ATCC nr. CCL61) og ble transfektert med det humane SSTR-2 cDNA, beskrevet i Yamada, et al., Proe. Nati. Acad. Sei. USA, 89:251-255 (1992) og tilgjengelig fra ATCC (ATCC nr. 79046), ved hjelp av standard teknikker som er kjent i molekylærbiologisk fagkunnskap. Se f.eks. Patel, et al., Biochem. Biophys. Res. Commun. 198:605 (1994). Urene membranpreparater ble preparert ved å homogenisere CHO-K1 celler transfektert med humant SSTR-2 i 20 ml med iskald 50 mM Tris-HCI (buffer A) med en POLYTRON™ homogenisator (Brinkmann Instruments, Westbury, NY) satt på merke 6 i 15 sek. Ytterligere buffer A ble tilsatt for å oppnå et sluttvolum på 40 ml, og homogenatet ble sentrifugert i en SORVAL™ SS-34 rotor (DuPont, Newtown, CT) ved 39.000 g i 10 min ved 0-4°C. Den resulterende supernatant ble skilt fra og kastet. Bunnfallet ble rehomogenisert i iskald buffer A, fortynnet og sentrifugert som beskrevet over. Sluttbunnfallet ble resuspendert i 10 mM Tris HCI og oppbevart på is for reseptor bindingsanalysen.
Deler av membranfraksjonen ble inkubert i 90 min ved 25°C med 0,05 nM [<125>l-Tyr]MK-678 (2000 Ci/mmol; c[N-metyl-Ala-Tyr(l<125>)-D-Trp-Lys-Va-Phe] i 50 mM HEPES (pH 7,4) som inneholdt et peptid til testing i ulike konsentrasjoner (f.eks. 10" <11> til 10"6 M), 10 mg/ml bovint serumalbumin (fraksjon V, Sigman Chemical Co., St. Louis, MO), MgCI2 (5 mM), Trasylol (200 KlU/ml), bacitracin (0,02 mg/ml), og fenylmetylsulfonylfluorid (0,02 mg/ml). Det avsluttende analysevolum var 0,3 ml. Inkubasjonene ble avsluttet med rask filtrering gjennom GF/c-filtere (på forhånd fuktet i 0,3% polyetylenimin i 30 min) ved hjelp av en filtrasjonsmanifold (Brandel, Gaithersburg, MD). Hvert rør og hvert filter ble deretter vasket tre ganger med 5 ml volumer av iskald buffer A. Spesifikk binding ble definert som den totale [<125>l-Tyr]MK-678 minus den binding som var tilstede i nærvær av 200 nM somatostatin-14.
De følgende testpeptider ble analysert; somatostatin-14, somatostatin-28, Analog I, Analog II, Analog III, Analog IV og Analog V. Strukturene av analogene I-V er vist over. Verdiene for Kj (inhibisjonskonstantene) for testpeptidene ble kalkulert ved hjelp av den følgende formel: Ki = IC5o/[1 +(LC/LEC)] der IC50 er konsentrasjonen av testpeptidet som er nødvendig for å hemme 50% av den spesifikke binding av den radioaktive merkede ligand [125l-Tyr]MK-678, LC er konsentrasjonen av den radioaktive merkede ligand (0,05 nM), og LEC er ekvilibriumdissosiasjonskonstanten til den radioaktive merkede ligand (0,155 nM). (2) Humant SSTR-5 bindingsanalyse
CHO-K1 celler ble transfektert med det humane SSTR-5 cDNA, beskrevet i Yamada, et al., Biochem Biophys. Res. Commun., 195-844 (1993) ved hjelp av standard teknikker som er kjent i det molekylærbiologiske fag. Se f.eks. Patel, et al., Biochem. Biophys. Res. Commun. 198:605 (1994). Råe membranpreparater ble laget ved homogenisering av CHO-K1 cellene transfektert med det humane SSTR-5 i 20 ml med iskald 50 mM Tris-HCI med en POLYTRON™ homogenisator (hastighet 6,15 sek). Buffer ble tilsatt for å oppnå et sluttvolum på 40 ml, og homogenatet ble sentrifugert i en SORVAL™ SS-34 rotor ved 39.000 g i 10 min ved 0-4°C. Den resulterende supernatant ble skilt fra og kastet. Bunnfallet ble rehomogenisert i iskald buffer, fortynnet og sentrifugert som før. Det avsluttende bunnfallet ble resuspendert 1 10 mM Tris HCI og oppbevart på is for analyse av reseptorbinding.
Deler av membranpreparasjonen ble inkubert i 30 min ved 30°C med 0,05 nM [<125>l-Tyr<11>]somatostatin-14 (2000 Ci/mmol; Amersham Corp., Arlington Heights, IL) i 50 mM HEPES (pH 7,4) som inneholdt et testpeptid i ulike konsentrasjoner (f.eks. 10" <11> til 10"<6> M), 10 mg/ml bovint serumalbumin (fraksjon V), MgCI2 (5 mM), Trasylol (200 KlU/ml), bacitracin (0,02 mg/ml), og fenylmetylsulfonylfluorid (0,02 mg/ml). Det avsluttende analysevolum var 0,3 ml. Inkubasjonene ble avsluttet med rask filtrering gjennom GF/c-filtere (på forhånd fuktet i 0,3% polyetylenimin i 30 min) ved hjelp av Brandel filtrasjonsmanifold.
