Description
Foreliggende oppfinnelse angår en ny serie av tri-substituerte fenylderivater, fremgangsmåter for deres fremstilling, farmasøytiske preparater og blandinger som inneholder nevnte derivater.
Mange hormoner og neurotransmittere modulerer vevsfunksjonen ved å forhøye det intra-cellulære nivået av 3',5-cyklisk adenosin-monbfosfat (cAMP). Det cellulære nivået av cAMP reguleres ved mekanismer som kontrollerer både syntese og nedbrytning. Syntesen av cAMP kontrolleres av adenylcyklase som direkte kan aktiveres ved hjelp av forbindelser som forskolin, eller indirekte ved at man binder spesifikke agonister til celleoverlfate-reseptorer som blir koblet til adenylcyklase. Nedbrytningen av cAMP kontrolleres av en gruppe fosfodiesterase (PDE) isoenzymer, som også kontrollerer nedbrytningen av 3',5-cyklisk guanosin-monofosfat (cGMP). Til dags dato, er det beskrevet syv forbindelser i denne gruppen (PDE l-VII), hvis fordeling varierer fra vev til vev. Dette antyder at spesifikke inhibitorer av PDE-isoenzymer kunne frembringe forskjellige nivåer av cAMP i forskjellig vevstyper (for oversikt med hensyn til PDE-fordeling, struktur, funksjon og regulering, se Beavo & Reifsnyder (1990) TIPS, H: 150-155 og Nicholson et al. (1991) TIPS, 12:19-27).
Det er klare tegn på at en forhøyelse av cAMP i inflammatoriske leukocytter fører til en inhibering av deres aktivitet. Videre er det vist at en forhøyelse av cAMP i de glatte musklene i luftveiene fører til en spasmolytisk effekt. I disse vevene spiller PDE IV en viktig rolle med hensyn til hydrolysen av cAMP. Det kan derfor forventes at selektive inhibitorer av PDE IV vil ha terapeutiske effekter med hensyn til inflammatoriske sykdommer som f.eks. astma, ved at man oppnår både en antiinflammatorisk og en bronko-dilaterende effekt.
Utformingen av PDE IV-inhibotorer har hittil ikke gitt særlige resultater, noe som skyldes at mange av de potensielle PDE IV-inibitorene som har blitt syntetisert, har manglet styrke og/eller har vært i stand til å hemme mer enn én type PDE-isoenzymer på en ikke-selektiv måte. Mangel på en selektiv virkning har vært et spesielt problem fordi cAMP har en omfattende rolle in vivo, og det er derfor behov for sterke selektive PDE IV-inhibitorer som har hemmende virkning mot PDE IV og liten eller ingen virkning i forhold til de andre PDE-isoenzymene.
Man har nå oppdaget en ny serie tri-substituerte fenylderivater, som alle er sterke inhibitorer av PDE IV ved konsentrasjoner hvor de har liten eller ingen hemmende virkning på andre PDE-isoenzymer. Disse forbindelsene hemmer humant rekombinant PDE IV-enzym og forhøyer også cAMP i isolerte leukocytter. Forbindelsene ifølge foreliggende oppfinnelse kan derfor brukes innenfor medisinsk behandling, da spesielt ved profylakse og behandling av astma.
Foreliggende oppfinnelse angår således i ett aspekt en forbindelse med formej (I) anvendelig som en PDE IV inhibitor

hvori R<4> er et hydrogenatom eller en gruppe -(CH2)tAr eller -(CH2)tAr(Alk<4>) x (0)y (Alk<5>)zAr hvori Ar og Ar' hver representerer fenylgrupper eventuelt substituert med en substituent valgt fra d.4alkyl, CF3, N02, NH2, C^alkylkarbonylamino, Ci.4alkyl-sulfonylamino og di(Ci^alkyl)aminosulfonylamino; Alk4 og Alk<5> hver representerer en Ci_4alkylengrupe eventuelt substituert med et halogenatom eller en hydroksylgruppe;
t er null eller et heltall 1, 2 eller 3, og x, y og z hver uavhengig representerer null eller 1; og R<5> er pyridin eventuelt substituert med en fenyl-C-|.4alkylgruppe;
og de farmasøytisk akseptable salter, solvater, hydrater og N-oksyder derav.
Det er innlysende at visse forbindelser med formel (I) kan ha ett eller flere kirale sentra, avhengig av sammensetningen på gruppene L, R<1>, R<2>, R<3>, R<4>, R<5>, R6 og R<7>. Når det er tilstede ett eller flere kirale sentra, kan det eksistere enantiomerer og diastereomere former, og det er underforstått at forbindelsen innebefatter alle slike enantiomerer, diastereomerer og blandinger av disse, og heri inngår racemater.
Alkylgruppene kan være rette eller forgrenete. En C3-C8-cykloalkylgruppe kan f. eks. være cyklobutyl, cyklopentyl eller en cykloheksylgruppe.
Nærværet av visse substituenter i forbindelsene med formel (I) muliggjør dannelse av salter av forbindelsene. Egnede salter innbefatter farmasøytisk akseptable salter, f.eks. syreaddisjonssalter fremstilt ved hjelp av uorganiske eller organiske syrer, og salter avledet av uorganiske og organiske baser.
Syreaddisjonssalter innbefatter hydroklorider, hydrobromider, hydrojodider, alkylsulfonater, f.eks. metansulfonater, etansulfonater, eller isetionater, arylsulfonater, f.eks. p-toluensulfonater, benzylater eller napsylater, fosfater, sulfater, hydrogensulfater, acetater, trifluoracetater, propionanter, citrater, maleater, fumarater, malonater, suksinater, laktater, oksalater, tartrater og benzoater.
Salter avledet av uorganiske eller organiske baser innbefatter alkalimetallsalter som natrium eller kaliumsalter, alkalijordmetallsalter som magnesium eller kalsiumsalter, og organiske arhinsalter som f.eks. morfolin, piperidin, dimetylamin eller dietylaminsalter.
Forløperforbindelser av forbindelser med formel (I) innbefatter de forbindelser, f.eks. estere, alkoholer eller aminoforbindelser, som in vivo lar seg omdanne ved hjelp av metabole mekanismer, f.eks. hydrolyse, reduksjon, oksidasjon eller transforestring, til forbindelser med formel (I).
Spesielt brukbare salter av forbindelser ifølge foreliggende oppfinnelse innbefatter farmasøytiske akseptable salter, da spesielt farmasøytisk akseptable syreaddisjonssalter.
I forbindelser med formel (I) eller (2) er gruppen Z generelt fortrinnsvis en gruppe av type (A). I forbindelser av denne typen er R<5> spesielt en eventuelt substituert 4-pyridylgruppe.
Ytterligere foretrukne forbindelser er slike hvori Z er en gruppe (B) og R<4> er et hydrogenatom.
Når Ar er en fenylgruppe i de ovennevnte eksempler, så kan -(L')n-Ar-gruppen eller en eventuelt annen substituent være knyttet til ethvert tilgjengelig ringkarbonatom bortsett fra det karbonatom som knytter fenylgruppen til den gjenværende del av forbindelsen med formel (I).
Spesielt brukbare forbindelser ifølge foreliggende oppfinnelse er følgende:

Forbindelser ifølge foreliggende oppfinnelse er selektive og sterke inhibitorer av PDE IV. Forbindelsenes evne til å virke på denne måten kan enkelt bestemmes ved hjelp av prøver som er beskrevet i de etterfølgende eksempler.
Spesiell anvendelse av de foreliggende forbindelser ifølge oppfinnelsen er ved profylakse eller behandling av astma, da spesielt inflammerte lunger forbundet med astma, cystisk fibrose, eller ved behandling av inflammatoriske luftveissykdommer, kronisk bronkitt, eosinofilisk granuloma, psoriasis og andre godartede eller ondartede spredende hudlidelser, endotoksisk sjokk, septisk sjokk, ulcerativ colitis, Crohn's sykdom, reperfusjonsskader av myokardium og hjernen, inflammatorisk arthritis, kronisk glomerulonephritis, atopisk dermatitis, urticaria, voksent respiratorisk stress-syndrom, diabetes insipidus, allergisk rhinitis, allergisk conjunctivitis, vernal
conjunctivitis, arterie restenose og arterosklerose.
Forbindelser ifølge foreliggende oppfinnelse kan også undertrykke en neurogen inflammasjon gjennom en økende konsentrasjon av cAMP i de sensoriske neuroner. Forbindelsene kan derfor brukes som smertestillende middel, som hostelindrende middel og som anti-hypersmertestillende middel ved inflammatoriske sykdommer som er forbundet med irritasjon og smerte.
Forbindelser ifølge foreliggende oppfinnelse kan også forhøye cAMP i lymfocytter og derved undertrykke uønsket lymfocyttaktivering i immunbaserte sykdommer som reumatisk arthritis, ankylose spondylitis, avstøtning av transplanterte organer og sykdommer som oppstår mellom vert og implanterte organer.
Forbindelser ifølge foreliggende oppfinnelse kan også redusere mavesyre-utskillelsen og kan derfor brukes for å behandle tilstander som er forbundet med hypersekresjon.
Forbindelser ifølge foreliggende oppfinnelse kan også undertrykke cytokinsyntesen i inflammatoriske celler i forhold til en reaksjon overfor en immun eller infiserende stimulering. De kan derfor brukes ved behandling av bakterie, sopp eller virusindusert sepsis og septisk sjokk, hvor cytokiner, f.eks. en svulstnekrosefaktor (TNF), er aktive modulerende forbindelser. Videre kan forbindelsene ifølge foreliggende oppfinnelse undertrykke inflammering og pyreksi som skyldes cytokiner, og kan derfor brukes ved behandling av inflammering og cytokinstyrt kronisk vevsdegenerering som oppstår i sykdommer, f.eks. reumatisk eller osteo-arthritis.
Overproduksjon av cytokiner, f.eks. TNF i bakterie, sopp eller virusinfeksjoner eller i sykdommer som kreft, fører til kakeksi og muskelsvinn. Forbindelser ifølge foreliggende oppfinnelse kan lindre eller svekke disse symptomene, og vil følgelig øke livskvaliteten hos pasienten.
Forbindelser ifølge foreliggende oppfinnelse kan også forhøye cAMP i visse områder av hjernen og derved motvirke depresjon og hukommelsessvikt.
Forbindelser ifølge foreliggende oppfinnelse kan undertrykke celleoppformering i visse svulstceller og kan derfor brukes for å hindre svulstvekst og invadering av normalt vev.
For profylakse eller behandling av sykdommer, kan forbindelser ifølge foreliggende oppfinnelse tilføres som farmasøytiske preparater, og oppfinnelsen innbefatter således også et farmasøytisk preparat som inneholder en forbindelse med formel (I) sammen med ett eller flere farmasøytisk akseptable bærestoffer eller fortynningsmidler.
Farmasøytiske preparater ifølge foreliggende oppfinnelse kan være i form som er egnet for oral, bukkal, parenteral, nasal, topikal eller rektal bruk, eller i en form som er egnet for tilførsel ved hjelp av inhalering eller lignende.
For oral bruk, kan de farmasøytiske preparatene være i form av f.eks.
tabletter, drops eller kapsler fremstilt på vanlig kjent måte med farmasøytisk akseptable fortynnings-midler, f.eks. bindemidler (f.eks. pregelatinisert maisstivelse,
polyvinylpyrrolidon eller hydroksypropylmetylcellulose); fyllstoffer (f.eks. laktose.mikrokrystallinsk cellulose eller kalsiumhydrogenfosfat); smøremidler (f.eks.
magnesiumstearat, talkum eller silika); nedbrytningsmidler (f.eks. maisstivelse eller natriumglykollat); eller fuktemidler (f.eks. natriumlaurylsulfat). Tablettene kan belegges på velkjent måte. Flytende preparater for oral bruk kan være i form av f.eks. løsninger, siruper eller suspensjoner, eller de kan brukes som et tørrprodukt for konstituering med vann eller en annen egnet væske før bruk. Slike flytende preparater kan fremstilles på vanlig kjent måte med farmasøytisk akseptable og kjente additiver som suspenderingsmidler, emuleringsmidler, ikke-vandige væsker og konserveringsmidler. Preparatene kan også inneholde buffersalter, smaksstoffer, farge-stoffer og søtningsmidler etter behov.
Preparater for oral bruk kan egnet opparbeides slik at de gir en kontrollert
frigjøring av den aktive forbindelsen.
For bukkal bruk, kan preparatene være i form av tabletter eller drops
opparbeidet på vanlig måte.
Forbindelsene med formel (I) og (2) kan opparbeides for parenteral bruk ved injeksjon, f.eks. ved en bolusinjeksjon eller infusjon. Preparater for injeksjon kan være tilstede i enhetsdoseringsformer, f.eks. i glassampuller eller i flerdosebeholdere, f.eks. små glassflasker. Preparatene for injeksjon kan være i form av suspensjoner, løsninger eller emulsjoner i oljeholdige eller vandige væsker, og kan dessuten inneholde midler som suspenderings-, stabiliserings-, konserverings- og/eller dispergerings-midler. Alternativt kan den aktive bestanddel være i pulverform for konstituering med en egnet væske, f.eks. sterilt pyrogenfritt vann, før bruk.
I tillegg til de preparater som er beskrevet ovenfor, kan forbindelser med formel (I) og (2) også opparbeides som et depotpreparat. Slike langtvirkende preparater kan anvendes ved implantering eller ved intramuskulær injeksjon.
For nasal bruk eller ved inhalering, kan forbindelsene ifølge foreliggende oppfinnelse hensiktsmessig tilføres i form av en aerosol i en beholder under trykk eller i en anordning for tilførsel til nesehulen, ved hjelp av et egnet drivmiddel, f.eks. diklor-difluormetan, triklor-fluormetan, diklortetrafluoretan, karbondioksyd eller en annen egnet gass eller blanding av gasser.
Preparatene kan, hvis det er ønskelig, være tilstede i en pakke eller fordelings-anordning som kan inneholde én eller flere enhetsdoseringsformer som inneholder . den aktive bestanddelen. Pakken eller fordelingsanordningen kan følges av instruksjoner for bruk.
Den mengde av en forbindelse ifølge foreliggende oppfinnelse som er nødvendig for profylakse eller behandling av en spesiell inflammatorisk tilstand, vil selvsalgt være avhengig av den anvendte forbindelsen, foruten tilstanden og sykdommen hos den pasient som skal behandles. Vanligvis vil man imidlertid bruke daglige doser fra ca. 100 ng/kg til 100 mg/kg, f.eks. ca. 0,01 mg/kg til 40 mg/kg kroppsvekt for oral eller bukkal anvendelse, fra ca. 10 ng/kg til 50 mg/kg kroppsvekst for parenteral bruk, og ca. 0,05 mg til ca. 1000 mg, f.eks. fra ca. 0,5 mg til ca. 1000 mg for nasal tilførsel eller tilførsel eller bruk ved inhalering eller innånding.
Forbindelsene ifølge foreliggende oppfinnelse kan fremstilles ved hjelp av de følgende fremgangsmåter. Symbolene Z, R, Ra, R<4>, R<5>, R<6>, Ar, Ar', Alk4 og Alk5 når de brukes i formlene i det etterfølgende, angir de samme grupper som er beskrevet ovenfor i forhold til formel (I) hvis intet annet er angitt. I de reaksjoner som er beskrevet nedenfor, kan det være nødvendig å beskytte reaktive funksjonelle grupper, f.eks. hydroksy, amino, tio eller karboksygrupper, hvor disse er ønskelig i det endelige produkt, for å unngå at disse på en uventet eller uønsket måte deltar i reaksjonene. Man kan i denne sammenheng bruke vanlig kjente beskyttende grupper (se f.eks. Green, T.W. i "Protective Groups in Organic Synthesis", John Wiley and Sons, 1981).
Forbindelser med formel (I) hvor Z er en gruppe (B) kan fremstilles ved at man reagerer en forbindelse med formel (3) med en fosfonatester (R,0)(OR")P(0)CH2(R<5>) (hvor R' og R" er identiske eller forskjellige, og er en alkyl eller aralkylgruppe) i nærvær av en base i et egnet løsemiddel.
Egnede baser innbefatter organometallbaser som organolitium, f.eks. n-butyllitium, alkoksider, f.eks. alkalimetallalkoksider som natriumetoksid eller
natriummetoksid og et hydrid som kaliumhydrid eller natriumhydrid. Løsemidler innbefatter etere, f.eks. dietyleter eller cykliske etere som tetrahydrofuran og en alkohol som f.eks. metanol eller etanol.
De fosfonatderivater som brukes i denne reaksjonen er enten kjente forbindelser eller kan fremstilles ved å reagere et fosfitt P(OR')2(OR") med en forbindelse R<5>CH2Hal (hvor Hal er et halogenatom, f.eks. et bromatom) ved hjelp av kjente fremgansmåter.
Mellomprodukter med formel (3) hvor R4 er forskjellig fra et hydrogenatom, kan fremstilles ved at man reagerer den tilsvarende forbindelsen hvor R4 er et hydrogenatom, med et organometallreagens, i ét løsemiddel som f.eks. tetrahydrofuran, ved lave temperaturer, dvs. omkring I0°C, hvoretter man oksiderer den resultrende alkoholen med et oksydasjons-middel, f.eks. mangandioksid, i et løsningsmiddel, f.eks. diklormetan.
Mellomprodukter med formel (3) hvor R4 er et hydrogenatom, kan fremstilles ved å reagere et halogenid med formel (4)
hvor Hal er et halogenatom, f.eks. et brom eller kloratom, med en organometallreagens som n-butyllitium, i et løsemiddel som et amid, f.eks. dimetylformamid, ved en lav temperatur, f.eks. under -60°C.
Mellomprodukter med formel (4) kan fremstilles ved å alkylere en tilsvarende forbindelse hvor R er hydrogen, ved å bruke en forbindelse RHal (hvor Hal er et halogenatom) i nærvær av en base i et løsemiddel som dimetylformamid ved romtemperaur eller høyere, f.eks. mellom 40°C og 50°C. Slike forbindelser hvor R er hydrogen er enten kjente forbindelser eller kan fremstilles fra kjente utgangsforbindelser ved vanlig kjente fremgangsmåter, f.eks. fra det tilsvarende aldehydet ved oksidering.
I en annen fremgangsmåte ifølge foreliggende oppfinnelse, kan en forbindelse med formel (I) hvor R4 er en -(CH2)t Ar(Alk<4>)x(0)y(Alk<5>)ZAr'-gruppe fremstilles ved at man kobler en forbindelse med formel (5)
(hvor Z' er som definert som Z i formel (I), bortsett fra at R<4> er en gruppe
-(CH2)tArE hvor E er en borsyre -B(OH)2 eller en tinnreagens Sn(R<l5>)3, hvor R<15> er en alkylgruppe, f.eks. en metylgruppe), med en reagens Ar'(Alk5)z(0)y((Alk4)xL2 hvor L<2 >er en avspaltende gruppe, i nærvær av en kompleks metallkatalysator.
Spesielle avspaltende grupper L<2> innbefatter f.eks. halogenatomer som brom, jod eller kloratomer og et alkylsulfonat, f.eks. trifluormetansulfonat. Spesielle tinnreagenser innbefatter de forbindelser hvor f.eks. R<15> i formel Sn(Rl5h er en metylgruppe.
Egnede katalysatorer innbefatter tungmetallkatalysatorer, f.eks. palladium-katalysatorer som tetrakis(trifenylfosfin)palladium. Reaksjonen kan utføres i et inert løsemiddel som f.eks. et aromatisk hydrokarbon som toluen eller benzen, eller en eter som dimetoksyetan eller dioksan, hvis nødvendig, i nærvær av en base som et alkalimetall-karbonat, f.eks. natriumkarbonat, ved en forhøyet temperatur, f.eks. kokepunktet for løsemiddelet. Vanligvis vil metallkatalysatorén og reaksjonsbetingelsene velges slik, avhengig av type av forbindelsen med formel (5) og/eller forbindelsen Ar'(Alk<5>)z(0)y((Alk<4>)xL<2> fra en rekke forskjellige kjente alterantiver for reaksjoner av denne typen (se f.eks. Miyaura, N. et al., Synth. Comm.
(1981), H, 513; Thompson, W.J. og Gaudino, J., J. Org. Chem. (1984), 49, 5237 og Sharp, M.J. et al., Tetrahedron Lett. (1987), 28, 5093).
Mellomproduktene er enten kjente forbindelser eller kan fremstilles fra kjente utgangsforbindelser ved analoge fremgangsmåter til de som er brukt for fremstillingen av kjente forbindelser. Når således det er ønskelig å fremstille en forbindelse Ar'(Alk<5>)z (0)y(Alk4)xL2 hvor L<2> er et halogenatom som et brom eller kloratom, og denne forbindelsen ikke er lett tilgjengelig, så kan en slik forbindelse fremstilles ved at man diazoterer det tilsvarende aminet ved f.eks. å bruke et nitritt som natriumnitritt, i en vandig syre ved lav temperatur, fulgt av en reaksjon med et passende kobber(l)halogenid i en vandig syre.
Mellomprodukter med formel (5) kan fremstilles ved en halogen-metallutbytning med en base som n-butyl eller butyllitium, fulgt av en reaksjon med et
borat som triisopropyl-borat eller en tinnreagens (R<l5>)3SnHal, hvor R<15> er som beskrevet ovenfor og Hal er et halogenatom som f.eks. et kloratom, ved en lav temperatur, dvs. rundt -70°C, i et løsemiddel som tetrahydrofuran.
I et annet eksempel på en fremgangsmåte ifølge foreliggende oppfinnelse, kan en forbindelse med formel (I) hvor både R<4> og R<5> er en gruppe -(CH2)r-Ar(Alk4)x(0)y(Alk5)zAr', x er null og y er 1, fremstilles ved at man reagerer en forbindelse med formel (5) hvor Z' er som definert for Z i formel (I), bortsett fra at R<4 >er en gruppe -(CH2)tArOH, med en reagens Ar'(Alk<5>)zL<2>, hvor L2 er en avspaltende gruppe, som beskrevet ovenfor.
Reaksjonen kan utføres i nærvær av en base, f.eks. trietylamin eller kaliumtert-butoksid, i et løsemiddel som diklormetan eller tetrahydrofuran, ved en egnet temperatur som f.eks. romtemperatur.
Forbindelser med formel(l) kan også fremstilles ved å omdanne andre
forbindelser med formel (I)
Således kan for eksempel hydrogenerings- eller overføringsreagenser og tilstander kan brukes for å omdanne (a) forbindelser med formel (I) hvor Z er en gruppe (B), til forbindelser med formel (I) hvor Z er gruppen (A); og (b) forbindelser med formel (I) hvor en N02-gruppe er tilstede som en substituent på en Ar eller Ar'-gruppe, noe som gir forbindelser med formel (I) hvor en NH2-gruppe er tilstede som en substiutent på en Ar eller Ar'-gruppe.
Hydrogeneringen kan utføres f.eks. ved å bruke hydrogen i nærvær av en katalysator. Egnede katalysatorer innbefatter metaller som platina eller palladium eventuelt på et inert bærestoff som karbon eller kalsiumkarbonat; nikkel, f.eks. Raney-nikkel eller rhodium. Reaksjonen kan utføres i et egnet løsemiddel som f.eks. en alkohol som metanol eller etnaol, en eter som tetrahydrofuran eller dioksan, eller en ester som etylacetat, eventuelt i nærvær av en base, f.eks. en tertiær organisk base som trietylamin, noe som kan skje ved romtemperatur.
Overføringshydrogenering kan utføres ved å bruke en organisk hydrogendonor og et overføringsmiddel. Egnede hydrogendonorer innbefatter f.eks. syrer som maursyre, formater, f.eks. ammoniumformat, alkoholer som benzylalkohol eller etylenglykol, hydrazin, og cykloalkener som cykloheksen eller cykloheksadien. Som overføringsmiddel, kan man f.eks. bruke et overgangsmetall, f.eks. palladium eller platina, eventuelt plassert på et inert bærestoff som angitt ovenfor, nikkel, f.eks. Raney-nikkel, ruthenium, f.eks. tris(trifenylfosfin)rutheniumklorid eller kobber. Reaksjonen kan vanligvis uføres ved romtemperatur eller ved forhøyet temperatur, eventuelt i nærvær av et løsemiddel som en alkohol, f.eks. etanol eller en syre som eddiksyre.
I et annet eksempel på en omdannelsesprosess, kan en forbindelse med formel (I) hvor Ar eller Ar' er substituert med en amidogruppe, f.eks. acetamidogruppe, eller en alkylaminosulfonylaminogruppe, f.eks. dimetylaminosulfonylaminogruppe, fremstilles ved at man reagerer en forbindelse med formel (I) hvor Ar eller Ar' er substituert med en amino-gruppe, ved å bruke et anhydrid, f.eks. eddiksyreanhydrid, eller et sulfamoylhalogenid, f.eks. dimetylsulfamoylklorid, i et løsemiddel, f.eks. et amin som pyridin, ved romtemperatur.
I et annet eksempel på en omdannelsesprosess, kan en forbindelse med formel (I) hvor Ar eller Ar' er substituert med en alkylsulfonylaminogruppe, f.eks. en metylsulfonyl-aminogruppe, fremstilles ved å reagere en forbindelse med formel (I) hvor Ar eller Ar' er substituert med en aminogruppe, med et alkylsulfonylhalogenid, f.eks. metansulfonylklorid, i nærvær av en base, f.eks. et tertiært amin som trietylamin eller N-metylmorfolin, i et løsemiddel, f.eks. et halogenert løsemiddel som diklormetan.
N-oksider av forbindelser med formel (I) kan f.eks. fremstilles ved at man oksiderer den tilsvarende nitrogenbasen ved å bruke et oksideringsmiddel som hydrogenperoksyd i nærvær av en syre, f.eks. eddiksyre, ved en forhøyet temperatur, f.eks. mellom 70°C og 80°C, eller alternativt ved en reaksjon med en persyre som f.eks. pereddiksyre, i et løsemiddel, f.eks. diklormetan, ved romtemperatur.
Salter av forbindelser med formel (I) kan fremstilles ved at man reagerer en forbindelse med formel (I) med en passende syre eller base i et egnet løsemiddel eller en blanding av løsemidler, f.eks. et organisk løsemiddel som en eter, eksempelvis dietyleter, eller en alkohol som etanol, ved å bruke vanlige kjente fremgangsmåter.
Når det er ønskelig å fremstille en spesiell enantiomer av en forbindelse med formel (I), så kan denne fremstilles fra den tilsvarende blandingen av enantiomerer ved å bruke enhver egnet vanlig kjent fremgangsmåte for oppløsning av enantiomerer.
Således kan f.eks. diastereomere derivater, f.eks. salter, fremstilles ved at man reagerer en blanding av enantiomerer med formel (I), f.eks. et racemat, og en passende kiral forbindelse, f.eks. en kiral syre eller base. Kjente kirale syrer innbefatter f.eks. tartarsyre og andre tartrater, f.eks. dibenzoyltartrater og ditoluoyltartrater, sulfonater såsom kamfersulfonater, mandelsyre og andre mandelater og fosfater som l,r-binaftalen-2,2'-diyl-hydrogenfosfat. Diastereomerene kan så skilles på enhver hensiktsmessig måte, f.eks. ved utkrystallisering, hvoretter den forønskede enantiomer kan innvinnes, f.eks. ved behandling med en syre eller base i de tilfeller hvor diastereomeren er et salt.
I en annen oppløsningsprosess kan et racemat med formel (I) separeres ved å bruke kiral høytrykksvæskekromatografi. Alternativt kan en spesiell enantiomer fremstilles ved å bruke et passende kiralt mellomprodukt i én av de fremgangsmåter som er beskrevet ovenfor.
De følgende eksempler illustrerer oppfinnelsen. I disse eksemplene er det anvendt følgende forkortelser: DME - etylenglykoldimetyleter; THF - tetrahydrofuran;
CH2CI2 - diklormetan; Et.20 - dietyleter; EtOH - etanol; RT - romtemperatur; DMF - N,N-dimetyl-formamid; EtOAc - etylacetat; DMPU - l,3-dimetyl-3,4,5,6-tetrahydro-2(IH)pyrimidinon; BuLi - butyllitium.

(IR,5S)-N-[(3R)-3-(4-benzyloksyfenyl)-3-(3-cyklopentyloksy-4-metoksyfenyl)-2-(4-pyridyl)propanoyl]-IO,IO-dimetyl-3-tia-4-azatricyklo[5.2.I.O<l5>]dekan-3,3-dioksid
En løsning av 4-brombenzyloksybenzen (22,3 g, 84,9 mmol, 2,2 ekv.) i 100 ml THF ble tilsatt magnesiumspon (2,44 g, 101,9 mmol, 2,6 ekv.) i 10 ml TH F inneholdende 2 dråper 1,2-dibrometan, og blandingen ble kokt under tilbakeløp i 30 minutter. Den avkjølte løsningen ble dråpevis ved -40°C tilsatt en løsning av mellomprodukt 6 (20,68 g, 38,6 mmol) i THF-Et20 (1:5; 600 ml), hvoretter man lot blandingen oppvarmes til -I0°C i løpet av 4 timer. Ammoniumkloridløsning (10%; 150 ml) og EtOAc (300 ml) ble tilsatt, og det organiske lag ble utskilt og blandet med ytterligere 3 x 100 ml EtOAc. Ekstraktet ble tørket over magnesiumsulfat og konsentrert i vakuum, hvoretter den gule resten ble omkrystallisert fra etanol, noe som ga 17,93 g av tittelforbindelsen som et blekgult fast stoff.

En blanding av forbindelsen fra eksempel 3 (9,35 g, 19,5 mmol), cykloheksadien (25 ml, 195 mmol) og 10% Pd/C (1,0 g) i 300 ml EtOH ble kokt under tilbakeløp i 2 timer. Reaksjonsblandingen ble så filtrert gjennom celitt, og filtratet og vaskeløsningene konsentrert i vakuum, noe som ga 7,6 g av tittelforbindelsen som et hvitt skum.

4-Metyl-3-pyridinkarboksaldehyd
n-BuLi (1,6 M i heksaner) (2,2 mmol, 1,37 ml) ble dråpevis tilsatt en løsning av N,N,N'-trimetyletylendiamin (2,4 mmol, 0,245 g) i THF ved -78°C. En løsning av 3-pyridinkarboksaldehyd (2 mmol, 0,214 g) i THF ble tilsatt, og blandingen ble rørt i 15 minutter før den ble tilsatt ytterligere n-BuLi (1,6 M i heksaner) (4 mmol, 2,5 ml). Etter røring i 2 timer ved -70°C, ble jodmetan tilsatt, og blandingen ble rørt ved RT i 30 minutter, og så helt over i en vandig NaCI-løsning, ekstrahert to ganger med etyleter, tørket over magnesiumsulfat, filtrert og konsentrert i vakuum. Resten ble kromatografert (Si02; Et20), noe som ga tittelforbindelsen som en olje.
Mellomprodukt 10
(4-Metyl-3-fenyletyenyl)pyridin
Benzyltrifenylfosfoniumbromid (blanding med natriumamid) (99 mmol, 4,96 g) ble oppløst i THF ved RT og rørt i 30 minutter. Man fikk en rød løsning. Mellomprodukt 9 (8,3 mmol, I g) i løsning i THF ble tilsatt, og reaksjonsblandingen ble rørt i 16 timer og så helt over i en vandig NaHC03-løsning, ekstrahert to ganger med CH2CI2, tørket over magnesiumsulfat, filtrert og konsentrert i vakuum. Resten ble kromatografert (Si02; Et20), noe som ga 1,5 g av tittelforbindelsen som en oljeaktig forbindelse.
Mellomprodukt II
(4-Metyl-3-fenyletyl)pyridin
En rørt løsning av 1,8 g av mellomrpodukt 10 i etanol ble hydrogenert ved å
bruke H2-Pd/C i 16 timer. Tittelforbindelsen ble fremstilt som en olje.

3-Cyklopentyloksy-4-metoksybenzaldehyd
Tittelforbindelsen ble fremstilt som beskrevet i internasjonal patentspesifikasjon nr. WO 94/20455.
Mellomprodukt 13
2-Brom-4-metylpyridin
En vandig løsning av 48% HBr (56,55 ml, 520,64 mmol) ble langsomt tilsatt 2-amino-4-metylpyridin (10 g, 92,47 mmol) under -5°C. Brom (14 ml, 274 mmol) ble så dråpevis tilsatt mens man holdt temperaturen under 0°C, hvoretter man langsomt og forsiktig tilsatte en løsning av natriumnitritt (17,5 g, 254 mmol) i 20 ml vann mens man holdt temperaturen under 5°C. Etter røring i I time, tilsatte man en løsning av NaOH (37 g, 940 mmol) i 50 ml vann ved en temperatur under 25°C. Blandingen ble så ekstrahert med 3 x 75 ml etyleter, de samlede lag ble vasket med saltløsning, tørket over magnesiumsulfat og fordampet til tørrhet, noe som ga en uren olje. Kromatografi (Si02; heksan/Et20, 50:50), ga 10,2 g av tittelforbindelsen som en gul olje.

Mellomprodukt 14
2-cylklopentyloksy-4-metylpyridin
Til en kald (0°C) suspensjon av natriumhydrid (2 g, 50 mmol) i 20 ml DMF ble dråpevis tilsatt cyklopentanol (4,99 ml, 55 mmol), og løsningen ble oppvarmet til RT og rørt i I time. Mellomprodukt 13 (8,6 g, 49,67 mmol) ble dråpevis tilsatt, og
reaksjons-blandingen ble holdt på 80°C i 2 timer. Etter avkjøling, tilsatte man 100 ml • vann, og blandingen ble ekstrahert over i 3 x 100 ml etyleter, vasket med saltløsning, tørket over magnesiumsulfat og så fordampet til tørrhet, noe som ga en oransje olje. Kromatografi (Si02; heksan/etyleter, 50:50), ga 7,5 g av tittelforbindelsen som en oransje olje. M/Z 179 (M + I). 5H (CDCI3) 1,5-2,0 (8H, m, (CH2)4), 2,25 (3H, s, CH3), 5,35 (IH, m, OCH), 6,45 (IH, s, pyridin H2), 6,65 (IH, d, pyridin H5) og 7,96 (IH, d, pyridin Hg).
Eksempel 1
a) (E) °9 (Z)-isomerer av 4-{2-(3-cyklopentyloksy-4-metoksyfenyl)-2-[4-(4-trifluormetylfenyl)fenyl]etenyl}pyridin
En blanding av mellomprodukt 5 (500 mg, I,I mmol) og tetrakis(trifenylfosfin)-palladium i 20 ml DME ble rørt ved romtemperatur i 30 minutter. Man tilsatte så en natriumkarbonatløsning (2 M; 1,5 ml, 3,0 mmol) og 4-trifluormetylfenylborsyre (I90 mg, l,0 mmol), og blandingen ble kokt under tilbakeløp i I8 timer. Den ble så avkjølt, delt mellom I0 ml vann og 20 ml etylacetat, hvoretter det organiske laget ble utskilt og blandet med ytterligere 2 x I0 ml etylacetat. Ekstraktet ble vasket med 15 ml saltløsning, tørket over magnesiumsulfat og konsentrert i vakuum, noe som ga en blekbrun olje. Denne ble så kromatografert (Si02; etylacetat-heksan, l:l), noe som ga 4I0 mg av tittelforbindelsen som en fargeløs olje.
6H (CDCI3) 1,5-2,0 (8H, br m, (CH2)4), 3,86 og 3,89 (3H, s, Orne), 4,57 og 4,70 (IH, br m, OCH), 6,6-7,8 (I4H, m, 2 x C6H4 + C6H3 + pyridin H3, H5 + C=CH) og 8,3-8,45 (2H, m, pyridin hb, H6).
Den følgende forbindelsen ble fremstilt på samme måte som beskrevet i eksempel la).
b) (E) og (Z)-isomerer av 4-[2-(3-cyklopentyloksy-4-metoksyfenyl)-2-(4'-metyl-4-bifenyl)etenyl]pyridin
Fra mellomprodukt 5 (0,40 g, 0,89 mmol), tetrakis(trifenylfosfin)palladium (0,051 g, 0,045 mmol) i 20 ml DME, natriumkarbonatløsning (2 M, 1,49 ml, 2,7 mmol) og 4-metylbenzenborsyre (0,1151 g, 0,846 mmol). Kromatografi (Si02; etylacetat-heksan, 1:1) ga 0,311 g av tittelforbindelsen.

Eksempel 2
a) (+)-4-{2-(3-Cyklopentyloksy-4-metoksyfenyl)-2-[4-(4-trifluormetylfenyl)-fenyletyl]}pyridiniumformat
En blanding av (380 mg) av forbindelsen fra eksempel I og 50 mg 10% Pd/C i 5 ml maursyre og 35 ml etanol ble kokt under tilbåkeløp i 3 timer. Reaksjonsblandingen ble så filtrert gjennom celitt og konsentrert i vakuum, noe som ga en gul olje som så ble kromatografert (Si02; etylacetat-heksan, 1:1), noe som ga 190 mg av tittelforbindelsen som et blekgultfast stoff.

Den følgende forbindelsen ble fremstilt som beskrevet i eksempel 2a).
b) (+)-4-[2-(3-Cyklopentyloksy-4-metoksyfenyl)-2-(4'-metyl-4-bifenyl)etyl]-pyridin
Fra forbindelsen fra eksempel Ib) (0,311 g, 0,673 mmol) i 10% maursyre i 35 ml etanol og 10% Pd/C. Rensing ved HPLC (60-80% acetonitril-vann) ga 0,23 g av tittelforbindelsen.

Eksempel 3
(R)-4-[2-(4-Benzyloksyfenyl)-2-(3-cyklopentyloksy-4-metoksyfenyl)etyl]pyridin
n-BuLi (1,6 M løsning i heksan) (27 ml, 43,0 mmol, 1,5 ekv.) ble dråpevis tilsatt en løsning av etantiol (10,6 ml, 14,3 mmol, 5 ekv.) i 300 ml THF ved ca. -I0°C. Etter ca. 0,5 timer, tilsatte man dråpevis en løsning av mellomprodukt 7 (20,6 g, 28,7 mmol) i 200 ml THF, og reaksjonsblandingen ble rørt ved RT i 12,5 timer. 10 ml vann ble tilsatt, og løsemiddelet fjernet i vakuum. Det gjenværende skummet ble oppløst i I00 ml etanol og 200 ml av en 2 M løsning av vandig natriumhydroksid, hvoretter blandingen ble kokt under tilbakeløp i I time. Reaksjonsblandingen ble så avkjølt til ca. 50°C og behandlet med 37 ml konsentrert saltsyre til pH 5 og så kokt under tilbakeløp i 0,75 timer. De organiske løse-midler ble fjernet i vakuum, og resten ble delt mellom vandig NaOH-løsning (l,0 M; 400 ml) og 400 ml etyleter. Det organiske laget ble utskilt, blandet med ytterligere 3 x I00 ml etyleter, og ekstraktet ble så vasket med vandig NaOH-løsning (1,0 Mj 2 x 100 ml), så tørket over magnesiumsulfat og konsentrert i vakuum. Man fikk en gul gummi som så ble kromatografert (Si02; heksan-Et20, 7:3 til 100% Et20), noe som ga 9,35 g av tittel-forbindelsen som et hvitt fast stoff (funnet: C, 79,76; H, 6,98; N, 2,62. C23H33NO3 krever C, 79,63; H, 7,11; N, 3,00%).

Eksempel 4
(R)-4-{2-(3-Cyklopentyloksy-4-metoksyfenyl)-2-[4-(4-nitrofenyloksy)fenyl]etyl}-pyridin
Kalium tert-butoksid (258 mg, 2,19 mmol) ble tilsatt en løsning av mellomprodukt 8 (710 mg, 1,83 mmol) i 25 ml DMF og 10 ml DMPU, og blandingen ble
rørt ved rom-temperatur. Etter 0,5 timer, tilsatte man 4-bromnitrobenzen (553 mg, 2,74 mmol), og løsningen ble rørt over natten ved RT. 100 ml vann og 20 ml av en 2
M vandig natrium-hydroksydløsning ble tilsatt, og blandingen ble ekstrahert med 3 x I50 ml etyleter. Ekstraktet ble vasket med 2 x 30 ml av en I M vandig natriumhydroksidløsning, 2 x I00 ml vann og I00 ml saltløsning, så tørket over magnesiumsulfat og konsentrert i vakuum. Den
gule resten ble så kromatografert (Si02; Et_0), noe som ga 729 mg av tittelforbindelsen som et blekgult fast stoff.

(E) og (Z)-isomerer av 4-[2-(3-cyklopentyloksy-4-metoksyfenyl)etenyl]-3-(fenyletyl)-pyridin
En katalytisk mengde av tosinsyre ble tilsatt en løsning av forbindelsen fra eksempel 10a) (1,8 g, 4,32 mmol) i 150 ml toluen, og blandingen ble kokt under tilbakeløp i et Dean Stark utstyr i 8 timer. Reaksjonsblandingen ble avkjølt, helt over i en vandig NaHC03-løsning og ekstrahert to ganger med metylenklorid. Den samlede organiske fase ble så tørket over magnesiumsulfat, filtrert og så konsentrert i vakuum. Resten ble underkastet kromatografi, noe som ga 1,3 g av tittelforbindelsen som et gult fast stoff. Smp. 78-80°C. (Funnet C, 81,44; H, 7,35; N, 3,62. C27H29N02 krever C, 81,17; H, 7,32; N, 3,51%).

Forbindelsen fra eksempel 5 (800 mg, 2 mmol) ble hydrogenert ved å bruke reagensene og betingelsene som beskrevet for fremstilling av mellomprodukt II fra mellom-produkt 10. Rensing ved kromatografi (Si20; Et20) ga 605 mg av tittel-forbindelsen som en fargeløs gummi. (Funnet C, 80,82; H, 7,77, N, 3,54. C27H3|N02 krever C, 80,76; H, 7,78; N.3,49%).

Eksempel 7
4-{2-(S)-[4-(4-Aminofenyloksy)fenyl]-2-(3-cyklopentyloksy-4-metoksyfenyl)etyl}-pyridindihydroklorid
En rørt løsning av forbindelsen fra eksempel 4 (800 mg, 1,46 mmol) i 100 ml etanol ble hydrogenert med 100 ml 10% Pd/C under en hydrogenatomosfære i 24 timer ved RT. Reaksjonsblandingen ble filtrert gjennom celitt, og filtratet konsentrert i vakuum, noe som ga et gulbrunt glass som ble delt mellom I M NaOH og etylacetat. Den smalede organiske fase ble tørket over natriumsulfat og så konsentrert i vakuum til et oljeaktig fast stoff. Kromatografi (Si02; EtOAc-Et20,1:1) ga 384,9 mg av tittelforbindelsens frie base som et nesten hvitt fast stoff.

Det resulterende oljeaktige bunnfallet ble oppløst ved å tilsette etanol, hvoretter løsemiddelet ble fjernet i vakuum. Resten ble suspendert i etyleter, hvoretter løsemiddelet ble fjernet i vakuum, noe som ga et meget blekt, beige fast stoff. Omkrystallisering (THF-EtOH) ga tittelforbindelsen som et meget blekt, beige, sterkt hygroskopisk fast stoff.

Eksempel 8
a) 4-{2-(S)-[4-(4-Acetamldofenyloksy]fenyl]-2-(3-cyklopentyloksy-4-metoksy-fenyl)etyl}pyridinhydroklorid
En løsning av den frie basen av forbindelsen fra eksempel 7 (322 mg, 0,67 mmol) i 10 ml pyridin ble oppvarmet sammen med 126 jai eddiksyreanhydrid, og blandingen ble rørt ved romtemperatur i 24 timer. Løsemiddelet ble fjernet i vakuum, resten ble så azeotropisk destillert to ganger med toluen og delt mellom vandig NaOH (pH 13) og etylacetat. Det vandige laget ble ytterligere ekstrahert med etylacetat, og det samlede organiske lag ble tørket over natriumsulfat og konsentrert i vakuum, noe som ga et hvitt glass. Kromatografi (Si02; EtOAc) ga 303,6 mg av tittelforbindelsen fri base som et hvitt glass.
Behandling av den frie base (303,6 mg) i EtOH-ET20 (30 ml; 1:2) med 0,62 ml av en 1,0 M HCI-løsning i etyleter som beskrevet i eksempel 7, fulgt av en omkrystallisering (THF-Et20), ga tittelforbindelsen som et hvitt, amorft, sterkt hygroskopisk fast stoff.

Fra forbindelsen fra eksempel 7 (250 mg, 0,52 mmol) i 15 ml pyridin og dimetylsulfamoylklorid (112 ul, 1,04 mmol). Kromatografi (Si02; EtOAc-Et20,1:2) ga 188,1 mg av tittelforbindelsen fri base som et hvitt glass.

Eksempel 9
4-{2-(S)-(3-Cyklopentyloksy-4-metoksyfenyl)-2-[4-(4-metylsulfonylaminofenyl)-fenyloksy]etyl}pyridinhydroklorid
En løsning av den frie basen av forbindelsen fra eksempel 7 (0,3667 g, 0,6 mmol) og N-metylmorfolin (264 0,24 mmol) i 15 ml vannfri metylenklorid ble oppvarmet sammen med metansulfonylklorid (93 [il, 1,2 mmol), og blandingen ble rørt ved romtemperatur i 24 timer. Blandingen ble delt mellom vann og etylacetat, og det vandige laget ytterligere ekstrahert med etylacetat. De samlede organiske lag ble vasket med vandig 0,5 M NaOH og saltløsning, tørket over natriumsulfat og konsentrert i vakuum, noe som ga et blekbrunt glass. Kromatografi (Si02; EtOAc-Et20, 2:1) ga tittelforbindelsen fri base (230 mg) som et hvitt, glassaktig fast stoff.

Fra en løsning av den frie basen (224,8 mg, 0,4024 mmol) i Et20-EtOH (40 ml, 3:1) og vandig 1,0 M HCI i Et20 (0,44 ml). Kromatografi (Si02; CH2CH2 -5% MeOH) ga et fargeløst glass som ble omkrystallisert (THF-Et20), noe som ga tittelforbindelsen som et meget hygroskopisk hvitt fast stoff, m/z (El<+>) 550 (M<+>).
Eksempel 10
a) (+)-4-[2-(3-Cyklopentyloksy-4-metoksyfenyl)-2-hydroksyetyl]-3-fenyletyl-pyridin
n-BuLi (1,6 M i heksaner) (8,03 mmol, 5,02 ml) ble dråpevis tilsatt en løsning av mellomprodukt II (1,45 g, 7,3 mmol) i THF, og blandingen ble rørt i 30 minutter før man tilsatte en løsning av mellomprodukt 12 (1,77 g, 8,03 mmol) i THF. Blandingen ble rørt i 30 minutter ved -78°C, hensatt for oppvarming til romtemperatur, helt over i en vandig NaHC03-løsning, ekstrahert to ganger med metylenklorid, tørket over magnesiumsulfat, filtrert og konsentrert i vakuum. Resten ble kromatografert (Si02; EtOAc), noe som ga 2,3 g av tittelforbindelsen som et hvitt fast stoff.
Den følgende forbindelsen ble fremstilt på samme måte som beskrevet i eksempel 10a).
b) [2-(3-Cyklopentyloksypyrid-4-yl)-l-pyrid-4-yl]etanol
Fra mellomprodukt 14 (I g, 5,61 mmol) i 10 ml THF under N2, n-BuLi (1,6 M) (3,5
ml) pyridin-4-karboksaldehyd (534 (il, 5,61 mmol). Kromatografi (Si02; Et20) ga 550 mg av tittelforbindelsen som en gul olje.
8H (CDCI3) 1,5-20 ((H, m, (CH2)4), 2,9 (2H, m, Chb-CHOH), 4,93 (IH, t, CHOH), 5,37 (IH.m, OCH), 6,51 (IH, s, pyridin H'3), 6,65 (IH, d, pyridin H'5), 7,24 (2H, d, pyridin H3, Hs), 8,2 (IH, d, pyridin H'5) og 8,51 (2H, d, pyridin H2, H6).
Aktivitet og selektivitet for forbindelsene ifølge foreliggende oppfinnelse, ble demonstrert i følgnde prøver. I disse prøver betyr forkortelsen FMLP peptidet N-formyl-met-leu-phe.
I. Isolert enzym
Styrke og selektivitet for forbindelsene ifølge foreliggende oppfinnelse, ble bestemt ved å bruke distinkte PDE-isoenzymer som følger:
i. PDE I, kaninhjerte
ii. PDE II, kaninhjerte
iii. PDE III, kaninhjerte, Jurka-celler
iv. PDE IV, HL60-celler, kaninhjerne, kaninnyre og humant rekombinant PDE IV
v. PDE V, kaninlunge, marsvinlunge
Et gen som koder for humant PDE IV var blitt klonet fra humane monocytter (Livi et al., 1990, Molecular and Cellular Biology, 10, 2678). Ved å bruke en lignende fremgangsmåte, har man klonet humane PDE IV-gener fra en rekke forskjellige kilder som innbefatter eosinofiler, neutrofiler, lymfocytter, monocytter, hjerne og nervevev. Disse gener var blitt overført til gjær ved å bruke en induserbar vektor, hvoretter forskjellige rekombinante proteiner ble uttrykt med biokjemiske egenskaper som tilsvarer de man har i PDE IV (Beavo and Reifsnyder, 1990, TIPS, il 150). Disse rekombinante enzymer, da spesielt det humane eosinofile rekombinante PDE IV, ble så brukt som basis for en utsortering for sterke, selektive PDE IV-inhibitorer.
Enzymene ble renset til isoenzymhomogenitet ved å bruke standardkromatografisk teknikk.
Fosfodiesteraseaktiviteten ble undersøkt som følger. Reaksjonen ble utført i en 150 ul standardblanding inneholdende (sluttkonsentrasjoner): 50 mM 2-[[tris(hydroksymetyl)-metyl]amino]-l-etansulfonsyre (TES) -NaOH-buffer (pH 7,5), 10 mM MgCI2, 0,1 (iM [<3>H]-cAMP og en bærevæske og forskjellige konsentrasjoner av prøveforbindelsene. Reaksjonen ble startet ved å tilsette enzymet, og reaksjonen ble utført ved 30°C imellom 5 til 30 minutter. Reaksjonen ble stoppet ved å tilsette 50 p.1 2% trifluoreddiksyre inneholdende [<l4>C]-5'-AMP for å bestemme innvinning av produktet. En porsjon av prøven ble så påsatt en kolonne av nøytral aluminiumoksyd og [<3>H]-cAMP ble eluert med 10 ml 0,1 TES-NaOH-buffer (pH 8). [<3>H]-5'-AMP-produktet ble eluert med 2 ml 2 M NaOH over i en skintillasjonsampulle inneholdende 10 ml av en skintillasjonsblanding. Innvinning av [<3>H]-5'-AMP ble bestemt ved å bruke [<l4>C]-5'-AMP og alle prøvene ble utført i et lineært område av reaksjonen.
Forbindelser ifølge foreliggende forbindelse, f.eks. de som er nevnt i de foregående eksempler, gir en konsentrasjonsavhengig inhibering av rekombinant PDE IV i en konsentrasjon fra 0,1-1000 nM med liten eller ingen aktivitet i forhold til PDE I, II, III eller V ved konsentrasjoner opp til 100 ^M.
2. Forhøyelse av cAMP i leukocytter
Effekten av forbindelsene ifølge foreliggende oppfinnelse på intracellulært cAMP, ble undersøkt ved å bruke humane neutrofiler eller marsvin-eosinofiler. Humane neutrofiler ble utskilt fra perifert blod, inkubert med dihydrocytokalasin B og prøveforbindelsen i 10 minutter og så stimulert med FMLP. Marsvin-eosinofilene ble innhøstet ved hjelp av en peritoneal slange som ble innsatt i dyr som på forhånd var behandlet med intraperitoneale injeksjoner av humant serum. Eosinofilene ble utskilt fra det peritoneale eksudat og inkubert med isoprenalin og prøveforbindelsen. Med begge celletyper, ble suspensjonene sentrifugert ved slutten av inkuberingen, og cellepellets ble resuspendert i buffer og kokt i 10 minutter før måling av cAMP ved en spesifikk radioimmunoprøve (DuPont).
De sterkeste forbindelsene ifølge eksemplene induserte en konsentrasjonavhengig forhøyelse av cAMP i neutrofiler og/eller eosinofiler ved konsentrasjoner fra 0,1 nM til I |iM.
3. Undertrykkelse av leukocyttfunksion
Forbindelser ifølge forliggende oppfinnelse ble undersøkt med hensyn til effekter på superoksidutvikling, kjemotaksis og tilfestning til neutrofiler og eosinofiler. Isolerte leuko-cytter ble inkubert med dihydrocytokalasin B for superoksidutvikling, og prøveforbindelsen før stimulering med FMLP. De mest virksomme forbindelser som angitt i eksemplene, ga en konsentrasjonsavhengig inhibering av
superoksidgenerering, chemotaksis og tilfesting ved konsentrasjoner fra 0,1 nM til I
HM.
Lipopolysakkarid (LPS)-indusert syntese av svulstnekrosefaktor (TNF) ved hjelp av humane perifere blodmonocytter (PBM), ble hemmet av forbindelsene fra eksemplene i konsentrasjoner fra 0,01 nM til 10 \ iM.
4. Skadelige effekter
Generelt har man i våre prøver funnet at forbindelsene ifølge forliggende oppfinnelse ikke har noen toksiske effekter når de blir tilført dyr i farmakologisk effektive doser.

1. Forbindelse med formel (I) anvendelig som en PDE IV inhibitor
