Description
Foreliggende oppfinnelse vedrører fumaratsaltet av (R)-5-(metylaminosulfonylmetyl) -3- (N-metylpyrrolidin-2-ylmetyl) - lH-indol med strukturen:-
Forbindelse (I) i sin fri baseform er beskrevet i eksempel
5A i WO-A-92/06973. Fumaratsaltet av (I) har ikke tidligere blitt beskrevet, skjønt fumaratsalter er nevnt i generelle vendinger bare i en liste av egnede akseptable farmasøy-
tiske syreaddisjonssalter i WO-A-92/06973.
Det er funnet at fumaratsaltet av (I) uventet har forbedret stabilitet mot oksidativ degradering. Også, og igjen uven-
tet, har det utmerket løslighet og faststoffstabilitet og er ikke-hygroskopisk.
I overenstemmelse med dette, foreskriver den foreliggende oppfinnelsen fumaratsaltet av (R) -5- (metylaminosulfonylmetyl) -3- (N-metylpyrrolidin-2-ylmetyl) -3- (N-metylpyrrolidin-2-ylmetyl)-lH-indol, farmasøytiske sammensetninger inneholdende denne, og dens bruk i behandling av migrene.
Saltet kan fremstilles ved reaksjonen av forbindelse (I)
med fumarsyre, typisk ca. 1 ekvivalent, i et egnet organisk løsemiddel eller blanding av løsemidler som illustrert i det følgende eksemplet.
Det kan formuleres og administreres til mennesker for å be-handle migrene og andre indikasjoner som beskrevet i WO-A-92/06973, som herved er innlemmet ved referanse.
Eksempel
( R) - 5- ( Metylaminosulfonylmetyl) - 3- ( N- metylpyrrolidin- 2-ylmetyl)- lH- indol
Til en suspensjon av (R)-5-(metylaminosulfonylmetyl)-3-(N-metylpyrrolidin-2-ylmetyl)-lH-indol (16,25 g, 0,0506 mol) i metanol (81,25 ml) ble fumarsyre (5,87 g, 0,0506 mol) tilsatt i én porsjon ved romtemperatur, dette gav en fin suspensjon som ble filtrert og vasket med metanol (16 ml) . De rørte væskene ble varmet til refluks og fortynnet med acetonitril (50 ml). Løsemiddel ble fjernet ved destillasjon under atmosfæretrykk og erstattet med acetonitril opp til en damptemperatur på 80EC. Under destillasjonen ble løs-ningen podet med krystallisasjonskim av (R)-5-(metylaminosulfonylmetyl) -3- (N-metylpyrrolidin-2-ylmetyl) - lH-indol fumarat og en dispersjon ble fremstilt. Suspensjonen ble avkjølt til romtemperatur og så granulert ved 0EC i1time.Filtrering ga produktet (21,45 g, 97%) som off-white krys-taller, smp. 159EC (med DSC). Rf 0,2 (kiselgel, dietyleter/
etylacetat/ DEA, metanol, 10:10:1:1); [a] D +13,7E (c=l,
H20) .

Mettet løslighet av den fri basen og fumaratsaltet av forbindelse ( I) .
For både den fri basen og fumaratsaltet, ble ca. 50 mg av fast bulkmaterialet nøyaktig innveid i et 1,5 ml plast "Eppendorf"-rør (Sigma). 0,3 ml vann (MilliQ) ble så tilsatt rørene. Rørene ble så vortex-blandet ved 1300omdr./min ("LKB") i 16 timer ved romtemperatur. Supernatan-ten ble separert fra uløst materiale ved sentrifugering ved 13000 omdr./min i 20 minutter ("Heraeus Biofuge 13"), og deretter fortynnet og analysert med hensyn på forbindelse (I) med HPLC. Analysen benyttet en mobilfase av acetonitril (20%), vann (80%) og trifluroeddiksyre (0,1%) og en 150 x 4,2 mm "Zorbax SB CN" kolonne ved 40EC med U.V. deteksjon ved 220 nm. Resultatene er fremlagt under.
Hygroskopisitet
Ca. 10 mg av hver av den fri basen og fumaratsaltene av forbindelse (I) ble eksponert for 8 forskjellige relative fuktigheter (RHs=relative humidities) mellom 0 og 94% ved 30EC i "Surface Measurement Systems Ltd" fuktighetsmikro-vekt. Prøvene fikk lov til å nå likevekt ved hver av RHs'ene og endringen i vekt fra den initielle verdien, når prøven først ble plassert i vekten, ble beregnet. Dataene ble benyttet til å konstruere fuktighets-sorpsjonsisoter-mene for materialene. Materialenes hygroskopisitet ble sammenlignet ved å beregne fuktighets-opptaket ved 90% RH, som var som følgende:-
Økt løslighet av fumaratsaltet i vann forenkler formulering av vandig løsning og hjelper oppløsning av faste doserings-former. Den økte vandige løsligheten av fumaratsaltet ble ikke fulgt av økt hygroskopisitet av bulkmaterialet [som potensielt kunne lede til redusert bulkstabilitet].
Oksidativ stabilitet av forbindelse ( I)
Myke gelatinkapsel-formuleringer er attraktive leverings-systemer for forbindelse (I) siden de forbedrer in vivo utløsnings- og innholds-ensartethet ved tillaging. Forbin- deise (I) kan formuleres som en væskefylt myk gelatinkapsel. Men, oksidativ degradering begrenser ofte holdbarheten på slike formuleringer. PEG 400 er reperesentativ for et typisk tilsetningsstoff til væskefyll brukt i en slik hen-sikt. Det følgende studiet ble designet for å bestemme hvorvidt oksidativ degradering oppstår med forbindelse (I) i en slik formulering, og hvorvidt fumaratsaltet utøver beskyttelse mot oksidativ degradering.1mg/ml løsninger av forbindelse (I) som fri base, forbindelse (I) som hydrokloridsalt og forbindelse (I) som f urna - ratsalt ble laget i 90% PEG 400 (BDH) og 10% vann (MilliQ) . 10% vann ble tilsatt til formuleringen for å mimikere inntrengning av vann fra det myke gelatinkapselskall. Den frie basen var ikke særlig løslig i formuleringen noe som forhindret videre undersøkelse av denne bulkformen. Men for å lette løsningens tillaging ble hydrokloridsaltet benyttet i steden.
Hydrogenperoksidet (BDH) ble tilsatt til en endelig konsen-trasjon på 0,3 vekt% i formuleringene for å forårsake en oksidativ belastning. En kontrollformulering uten hydrogen-peroksid ble også brukt i studiet. 1 ml av hver formulering ble forseglet i 2 ml HPLC flasker ("Cromacol") og plassert i en termos tåtert ovn ved 40EC. Prøvene ble fjernet etter1,5 og 3 dager og lagret frosset (-20EC) inntil analyse. Prøvene ble fortynnet og analysert ved å benytte den stabilitetsindikerende HPLC metoden som beskrevet over. Degradering ble uttrykket som % gjenværende forbindelse (I) . Stabilitet av forbindelse (I) i 90% PEG 400 ved 40eC med og uten oksidativ belastning (gjenomsnittsverdi"standardav-vik, n=3)
Stabilitetsstudiet demonstrerte at begge saltformene av forbindelse (I) var relativt stabile i PEG 400 formulering i fravær av oksidativ belastning. Men, når en oksidativ belastning ble påført formuleringen ved tilsetting av hyd-rogenperoksid ble det observert en signifikant økning i degradering. Dette demonstrerer at oksidativ degradering kan forekomme i mykgelformuleringer av forbindelse (I) med en resulterende innvirkning på holdbarheten. Slik vil bulk-former med forbedret stabilitet i et oksidativt miljø på-hjelpe formulering i myke gelatinkapsler.
Degraderingshastigheten av fumaratsalter var signifikant lavere (analyse av variansen p < 0,001) enn for hydrokloridet ved begge tidspunktene. Antioksidantegenskapene av fumaratsaltet gir signifikant beskyttelse mot degradering i formuleringen og hjelper formuleringen i mykgelhjelpemid-delet.
Såvidt vites finnes det ingen tidligere rapporter i litte-raturen vedrørende overlegen oksidativ stabilitet for fumaratsalter.
Faststoff- stabilitet
Ca.1mg av hver av den fri basen, hydrokloridet, hydrobro-midet og fumaratsaltet av forbindelse (I) ble nøyaktig innveid i små glassflasker. Disse ble lagret i 9 uker ved hver av 4EC/omgivelsenes fuktighet, 40EC/omgivelsenes fuktighet, 40EC/75% RH og 50EC/omgivelsenes fuktighet. Prøvene ble så analysert ved å bruke den følgende stabilitetsindikerende HPLC-metoden. En mobilfase bestående av 0,05 M kaliumdi-hydrogenortofosfat justert til pH 2 med fosforsyre (90%) og acetonitril (fjern DV) (10%), ble pumpet gjennom en "Zorbax SB-CNn 150 x 4,6 mm kolonne (40EC) med en hastighet på1ml/minutt med UV deteksjon ved 225nm.
Prøvene med unntak av den fri basen ble laget ved å løse dem i mobilfasen i en 25 ml volumetrisk flaske. Den fri basen ble løst i noen dråper metanol før fortynning med den mobile fasen.
Stabiliten av prøvene ble kvantifisert ved å undersøke forekomst av nye topper i kromatogrammet av lagrede prøver
og økningen i de foreliggende toppene sammenlignet med kon-trollprøvene lagret ved 4EC. Degraderingen uttrykt på denne måten for den fri basen og fumaratsaltet lagret ved 50EC er vist i tabellen under. Lignende trender ble observert ved
lagringsbetingelser med lavere temperatur. Det kan sees fra dataene at fumaratsaltet ikke hadde degradert i det hele tatt etter 9 uker ved 50EC, mens den fri basen viste målbar degradering med forekomst av 4 nye droge-relaterte topper. Derfor er fumaratsaltet det mest stabile i fast tilstand og har en egnet holdbarhet i bulkform for farmasøytisk utvik-ling.
Dthevet og understreket indikerer at en ny topp har oppstått.
Hydroklorid- og hydrobromidsaltene referert til over ble fremstilt på vanlig måte, for eksempel, for å lage hydrokloridsaltet ble en løsning av forbindelse (I) i etanol behandlet ved 65EC med ca. 1 ekvivalent av konsentrert HCl og deretter avkjølt. Fjerning av løsemiddel in vacuo og re-krystallisering av residuet fra absolutt etanol ga hydrokloridsaltet.

Claims (6)
1. Fumaratsaltet av (R)-5-(metylaminosulfonylmetyl)-3 (N-metylpyrrolidin-2-ylmetyl) -lH-indol.
2. En farmasøytisk sammensetning inneholdende fumaratsaltet som krevet i krav 1 og et farmasøytisk akseptabelt tilsettingsstoff eller bærer.
3.Fumaratsaltet som krevet i krav 1 for bruk som et medikament.
4. Anvendelse av fumaratsaltet som krevet i krav l for fremstilling av et medikament for behandling av migrene.
5. Fremgangsmåte for å fremstille fumaratsaltet av (R)-5-(metylaminosulf onylmetyl) -3 (N-metylpyrrolidin-2-ylmetyl)-lH-indol
karakterisert vedå reagere (R)-5-(metylaminosulfonylmetyl) -3 (N-metylpyrrolidin-2-ylmetyl)-lH-indol med fumarsyre.
6. Fremgangsmåte som krevet i krav 6, hvori ca. 1 ekvivalent med fumarsyre blir brukt.
