Description
Foreliggende oppfinnelse er rettet mot en fremgangsmåte for fremstilling av taxol, samt et nytt p-laktam for anvendelse som mellomprodukt ved fremgangsmåten.
Taxanfamilien av terpener, hvor taxol er et medlem, har tiltrukket seg betydelig interesse både innen biologi og kjemi. Taxol er et lovende kjemoterapeutisk middel mot kreft med et bredt spektrum av antileukemi- og tumorhemmende akti-vitet, og har den følgende formel:
På grunn av dets lovende virkning gjennomgår taxol for tiden kliniske forsøk både i Frankrike og USA.
Leveransen av taxol til disse kliniske forsøkene til-veiebringes for tiden ved hjelp av bark fra flere barlind-arter. Taxol finnes imidlertid bare i ubetydelige mengder i barken hos disse saktevoksende, eviggrønne planter, noe som forårsaker betydelig bekymring for at den begrensede tilgang på taxol ikke vil imøtekomme behovet. Kjemikere har følgelig i de senere år lagt vekt på å prøve å finne en mulig syntetisk vei for fremstillingen av taxoler. Så langt har resultatene ikke vært helt tilfredsstillende.
Én syntetisk vei som er blitt foreslått, er rettet mot syntesen av den tetracykliske taxankjerne fra kjemikalier som finnes i handelen. En syntese av den taxolbeslektede forbindelse taxusin er blitt rapportert av Holton et al. i JACS 110, 6558 (1988). Til tross for fremskrittet som er gjort ved denne fremgangsmåte, er det sannsynlig at den endelige total-syntese av taxol ikke desto mindre vil være en flertrinns, omstendelig og kostbar fremgangsmåte.
En alternativ fremgangsmåte for fremstillingen av taxol er blitt beskrevet av Greene et al. i JACS 110, 5917
(1988) og omfatter anvendelsen av en forbindelse som er be-slektet med taxol, 10-deacetylbaccatin III, som har den følgende formel:
10-deacetylbaccatin III er lettere tilgjengelig enn taxol, ettersom den kan fås fra bladene til Taxus baccata. Ifølge fremgangsmåten til Greene et al. omdannes 10-deacetylbaccatin III til taxol ved festing av C10-acetylgruppen og ved festing av C13-(3-amidoestersidekjeden gjennom forestringen av C-13-alkoholen med en p-amidokarboksylsyreenhet. Selv om denne fremgangsmåten krever forholdsvis få trinn, er syntesen av p-amidokarboksylsyreenheten en flertrinns fremgangsmåte som for-løper med lavt utbytte, og koblingsreaksjonen er omstendelig og forløper også med lavt utbytte. Denne koblingsreaksjonen er imidlertid et nøkkeltrinn som er påkrevd ved enhver tenkelig syntese av taxol eller biologisk aktivt derivat av taxol, ettersom det er blitt vist av Wani et al. i JACS 93, 2325
(1971) at tilstedeværelsen av p-amidoestersidekjeden i C13 er påkrevd for antitumoraktivitet.
En hovedvanskelighet som er tilbake ved syntesen av taxol og andre potensielle antitumormidler, er mangelen på en lett tilgjengelig enhet som lett kunne festes til C13-oksy-genet for å tilveiebringe p-amidoestersidekjeden. Utvikling av en slik enhet og en fremgangsmåte for dens festing i høyt utbytte ville lette syntesen av taxol samt beslektede antitumormidler med et modifisert sett av substituenter på kjernen eller en modifisert C13-sidekjede. Dette behov er blitt opp-fylt ved oppdagelsen av en ny kjemisk, lett tilgjengelig side-kjedeforløperenhet og en virkningsfull fremgangsmåte for dens festing til C13-oksygenet.
Formålene ved foreliggende oppfinnelse er derfor tilveiebringelsen av en sidekjedeforløper for syntesen av taxoler, og tilveiebringelsen av en fremgangsmåte for festingen av sidekjedeforløperen i forholdsvis høyt utbytte for å tilveiebringe et taxolprodukt.
I korte trekk er derfor foreliggende oppfinnelse rettet mot en sidekjedeforløper, et p-laktam med formelen:
Foreliggende oppfinnelse er også rettet mot en fremgangsmåte for fremstilling av et taxolprodukt som omfatter å bringe en alkohol i kontakt med et p-laktam i nærvær av en tilstrekkelig mengde av et aktiveringsmiddel under betingelser som kan forårsake at p-laktamet omsettes med alkoholen til en p-amidoester, som kan anvendes som et mellomprodukt ved syntesen av taxol.
Andre trekk ved oppfinnelsen vil fremgå og fremheves nedenunder.

Taxolalkylgruppene er lavere alkyl inneholdende fra 1 til 6 kar bona tomer i hovedkjeden og opp til 10 kar bona t omer. De kan være rettkjedede eller forgrenede og omfatter metyl, etyl, propyl, isopropyl, butyl, isobutyl, tert.-butyl, aryl, heksyl o.l.
Taxolalkenylgruppene er lavere alkenyl inneholdende fra 2 til 6 karbonatomer i hovedkjeden og opp til 10 karbonatomer. De kan være rettkjedede eller forgrenede og omfatter etenyl, propenyl, isopropenyl, butenyl, isobutenyl, aryl, heksenyl o.l.
Taxolalkynylgruppene er lavere alkynyl inneholdende fra 2 til 6 karbonatomer i hovedkjeden og opp til 10 karbonatomer. De kan være rettkjedede eller forgrenede og omfatter etynyl, propynyl, butynyl, isobutynyl, aryl, heksynyl o.l.
Eksempler på alkanoyloksy omfatter acetat, propionat, butyrat, valerat, isobutyrat o.l. Det mest foretrukne alkano-
yloksy er acetat.
Foretrukne verdier for substituentene A, B, D, L, E, F, G, M, I, J, K, P, Q, S, T, U, V og W er spesifisert i tabell I.
Nedenunder er det vist eksempelvis forbindelser innenfor den generiske formel:
I overensstemmelse med fremgangsmåten ifølge foreliggende oppfinnelse omdannes p-laktamer 1 til p-amidoestere i nærvær av en alkohol og et aktiveringsmiddel, fortrinnsvis et tertiært amin, slik som trietylamin, diisopropyletylamin, pyridin, N-metylimidazol og 4-dimetylaminopyridin (DMAP). For eksempel reagerer p-laktamer 1 med forbindelser som har den tetracykliske taxankjerne og en C13-hydroksylgruppe, i nærvær av 4-dimetylaminopyridin (DMAP), hvorved man får stoffer med en p-amidoestergruppe i C13.
Alkoholene er helst 7-0-trietylsilylbaccatin III, som kan fås som beskrevet av Greene et al. i JACS 110, 5917 (1988) eller ved hjelp av andre veier. Som rapportert i Greene et al., omdannes 10-deacetylbaccatin III til 7-0-trietylsilyl-baccatin III iht. det følgende reaksjonsskjerna:
Under det som er rapportert å være nøye optimaliserte betingelser, omsettes 10-deacetylbaccatin III med 20 ekvivalenter (C2H5)3SiCl ved 23 °C under en argonatmosfære i 20 timer i nærvær av 50 ml pyridin/mmol av 10-deacetylbaccatin III, hvorved man får 7-trietylsilyl-10-deacetylbaccatin III (32a) som et reaksjonsprodukt i 84-86 % utbytte etter rensing. Reaksjonsproduktet acetyleres med 5 ekvivalenter CH3C0C1 og
25 ml pyridin/mmol av 32a ved 0 °C under argonatmosfære i
48 timer, hvorved man får 86 % utbytte av 7-0-trietylsilyl-baccatin III (32b), Greene et al. i JACS 110, 5917-5918
(1988).
Som vist i det følgende reaksjonsskjerna, kan 7-0-tri-etylsilylbaccatin III 32b omsettes med et p-laktam ved værel-sestemperatur, hvorved man får et taxolmellomprodukt hvor C-7-og C-2'-hydroksylgruppene er beskyttet med hhv. trietylsilyl-og etoksyetylbeskyttelsesgrupper. Disse gruppene hydrolyseres så under milde betingelser for ikke å forstyrre esterbindingen eller taxolsubstituentene.

Selv om foreliggende skjema er rettet mot syntesen av det naturlige produktet taxol, kan det anvendes med modifika-sjoner enten i p-laktamet eller den tetracykliske alkohol, som kan være avledet fra naturlige eller ikke-naturlige kilder, for å fremstille andre syntetiske taxoler.
Ifølge oppfinnelsen kan et p-laktam 1 omdannes til en p-amidoester i nærvær av et aktiveringsmiddel og en alkohol som er forskjellig fra 7-0-trietylsilylbaccatin III, hvorved man får et taxolmellomprodukt. Syntese av taxol kan så fort-sette under anvendelse av taxolmellomproduktet under et passende reaksjonsskjerna.
p-laktamalkylgruppene er lavere alkyl inneholdende fra 1 til 6 karbonatomer i hovedkjeden og opp til 15 karbonatomer. De kan være rettkjedede eller forgrenede og omfatter metyl, etyl, propyl, isopropyl, butyl, isobutyl, tert.-butyl,
aryl, heksyl o.l.
p-laktamalkenylgruppene er lavere alkenyl som inne-holder fra 2 til 6 karbonatomer i hovedkjeden og opp til 15 karbonatomer. De kan være rettkjedede eller forgrenede og omfatter etenyl, propenyl, isopropenyl, butenyl, isobutenyl, aryl, heksenyl o.l.
p-laktamalkynylgruppene er lavere alkynyl inneholdende fra 2 til 6 karbonatomer i hovedkjeden og opp til 15 karbonatomer. De kan være rettkjedede eller forgrenede og omfatter etynyl, propynyl, butynyl, isobutynyl, aryl, heksynyl o.l.
Eksempler på p-laktamalkanoyloksy omfatter acetat, propionat, butyrat, valerat, isobutyrat o.l. Det mest foretrukne alkanoyloksy er acetat.
R2 i p-laktam 1 kan være alkyl, acyl, etoksyetyl, 2,2,2-trikloretoksymetyl eller annen hydroksylbeskyttelsesgruppe, slik som acetaler og etere, dvs. metoksymetyl og benzyloksymetyl; estere, slik som acetater; karbonater, slik som metylkarbonater; o.l. Flere forskjellige beskyttelsesgrup-per for hydroksylgruppen og syntesen derav kan finnes i "Protective Groups in Organic Synthesis" av T.W. Greene, John Wiley and Sons, 1981. Den valgte hydroksylbeskyttelsesgruppe bør fjernes lettvint under betingelser som er tilstrekkelig milde, slik at esterbindingen eller andre substituenter i taxolmellomproduktet ikke forstyrres. R2 er imidlertid fortrinnsvis etoksyetyl eller 2,2,2-trikloretoksymetyl, og helst etoksyetyl.
Eksempelvise forbindelser innenfor den generiske formel er vist nedenunder:
Ettersom p-laktamet 1 har flere asymmetriske karbonatomer, er det kjent for fagfolk innen teknikken at forbindel-sene som har asymmetriske karbonatomer, kan foreligge i dia-stereomere, racemiske eller optisk aktive former. Alle disse formene er tenkt på innenfor omfanget av denne formel. Nærmere bestemt omfatter den enantiomerer, diastereomerer, racemiske blandinger og andre blandinger derav.
p-laktamet 1 kan fremstilles fra lett tilgjengelige materialer, slik som vist for p-laktam 2 i skjemaet nedenunder :
Reagenser: (a) trietylamin, CH2C12, 25 °C, 18 timer; (b)
4 ekvivalenter serumammoniumnitrat, CH3CN, -10 °C, 10 min; (c) KOH, THF, H20, 0 °C, 30 min; (d) etylvinyleter, THF, toluen-sulfonsyre (kat.), 0 °C, 1,5 time; (e) CH3Li, eter, -78 °C,
10 min; benzoylklorid, -78 °C, 1 time.
Utgangsmaterialene er lett tilgjengelige, a-acyloksy-acetylklorid fremstilles fra glykolsyre, og i nærvær av et tertiært amin ringkondenseres det med iminer fremstilt fra aldehyder og p-metoksyanilin, hvorved man får 1-p-metoksyfenyl-3-acyloksy-4-arylazetidin-2-oner.
p-metoksyfenylgruppen kan lett fjernes gjennom oksi-dasjon ved serumammoniumnitrat, og acyloksygruppen kan hydrolyseres under standardbetingelser, som er velkjente for de som har erfaring innenfor teknikken, for å tilveiebringe 3-hyd-
roksy-4-arylazetidin-2-oner.
3-hydroksylgruppen kan beskyttes med flere forskjellige standardbeskyttelsesgrupper, slik som 1-etoksyetylgrup-pen. Fortrinnsvis oppløses det racemiske 3-hydroksy-4-aryl-azetidin-2-on i de rene enantiomerene før beskyttelse ved rekrystallisering av tilsvarende 2-metoksy-2-(trifluormetyl)-fenyleddiksyreestere, og bare den dekstrorotatoriske enantio-mer anvendes ved fremstillingen av taxol. I alle tilfeller kan 3-(1-etoksyetoksy)-4-fenylazetidin-2-on omdannes til p-laktam 2 ved behandling med en base, fortrinnsvis n-butyllitium, og et aroylklorid ved -78 °C eller lavere.
De følgende eksempler illustrerer oppfinnelsen.
Eksempel 1
Fremstilling av cis- l- benzoyl- 3-( 1- etoksyetoksy)- 4- fenyl-azetidinon 2
Cis- l- p- metoksyfenyl- 3- acetoksy- 4- fenylazetidin- 2- on
Til en oppløsning av 962 mg (4,56 mmol) av iminet, avledet fra benzaldehyd og p-metoksyanilin, og 0,85 ml (6,07 mmol) trietylamin i 15 ml CH2C12 ved -20 °C ble det dråpevis tilsatt en oppløsning av 413 mg (3,04 mmol) a-acetoksyacetylklorid i 15 ml CH2C12. Reaksjonsblandingen fikk varmes opp til 25 °C i løpet av en 18-timers periode. Reaksjonsblandingen ble så fortynnet med 100 ml CH2C12 og oppløs-ningen ekstrahert med 30 ml 10 % vandig HC1. Det organiske sjikt ble vasket med 30 ml vann og 30 ml mettet, vandig natriumbikarbonat, tørket over natriumsulfat og oppkonsentrert, hvorved man fikk en fast masse. Det faste stoff ble triturert med 50 ml heksan og blandingen filtrert. Det gjen-værende faste stoff ble rekrystallisert fra etylacetat/heksan, hvorved man fikk 645 mg (68 %) cis-l-p-metoksyfenyl-3-acetoksy-4-fenylazetidin-2-on som hvite krystaller, sm.p. 163 °C.
Cis- 3- acetoksy- 4- fenylazetidin- 2- on
Til en oppløsning av 20,2 g cis-l-p-metoksyfenyl-3-acetoksy-4-fenylazetidin-2-on i 700 ml acetonitril ved -10 °C ble det sakte tilsatt en oppløsning av serumammoniumnitrat i 450 ml vann i løpet av et tidsrom på 1 time. Blandingen ble omrørt i 30 min ved -10 °C og fortynnet med 500 ml eter. Vann-sjiktet ble ekstrahert med to 100 ml porsjoner eter, og det kombinerte organiske sjikt ble vasket med to 100 ml porsjoner vann, to 100 ml porsjoner mettet, vandig natriumbisulfitt, to 100 ml porsjoner mettet, vandig natriumbikarbonat og oppkonsentrert, hvorved man fikk 18,5 g av et fast stoff. Rekrystallisering av det faste stoff fra aceton/heksan gav cis-3-acetoksy-4-fenylazetidin-2-on som hvite krystaller, sm.p. 152-154 °C.
Cis- 3- hydroksy- 4- fenylazetidin- 2- on
Til en blanding av 200 ml THF og 280 ml 1 M vandig kaliumhydroksidoppløsning ved 0 °C ble det tilsatt en opp-løsning av 4,59 g (22,4 mmol) cis-3-acetoksy-4-fenylazetidin-2-on i 265 ml THF via en dråpetrakt i løpet av et tidsrom på 40 min. Cis- 3- ( 1- etoksyetoksy)- 4- fenylazetidin- 2- on
Til en oppløsning av 3,41 g (20,9 mmol) cis-3-hydroksy-4-fenylazetidin-2-on i 15 ml THF ved 0 °C ble det tilsatt 5 ml etylvinyleter og 20 mg (0,2 mmol) metansulfonsyre. Blandingen ble omrørt ved 0 °C i 20 min, fortynnet med 20 ml mettet, vandig natriumbikarbonat og ekstrahert med tre 40 ml porsjoner etylacetat. De kombinerte etylacetatsjikt ble tørket over natriumsulfat og oppkonsentrert, hvorved man fikk 4,87 g (99 %) cis-3-(1-etoksyetoksy)-4-fenylazetidin-2-on som en fargeløs olje.
Cis- l- benzoyl- 3-( 1- etoksyetoksy)- 4- fenylazetidin- 2- on
Til en oppløsning av 2,35 g (10 mmol) cis-3-(1-etoksyetoksy)-4-fenylazetidin-2-on i 40 ml THF ved -78 °C ble det tilsatt 6,1 ml (10,07 mmol) av en 1,65 M oppløsning av n-butyllitium i heksan. Blandingen ble omrørt i 20 min ved
-78 °C og en oppløsning av 1,42 g (10,1 mmol) benzoylklorid i 10 ml THF ble tilsatt. Blandingen ble omrørt ved -78 °C i 1 time og fortynnet med 70 ml mettet, vandig natriumbikarbonat og ekstrahert med tre 50 ml porsjoner etylacetat. De kombinerte etylacetatekstrakter ble tørket over natriumsulfat og oppkonsentrert, hvorved man fikk 3,45 g av en olje. Kromato-grafi av oljen på silikagel eluert med etylacetat/heksan gav 3,22 g (95 %) cis-l-benzoyl-3-(1-etoksyetoksy)-4-fenylazetidin-2-on (2) som en fargeløs olje.
Eksempel 2
Fremstilling av B- amidoestere fra cis- l- benzoyl- 3-( 1- etoksy-etoksy )- 4- fenylazetidin- 2- on- 2
Benzyl- 3- benzamido- 3- fenyl- 2- hydroksypropionat
Til en oppløsning av 88 mg (0,26 mmol) cis-l-benzoyl-3- (1-etoksyetoksy)-4-fenylazetidin-2-on i 0,3 ml THF ble det tilsatt 28 mg (0,26 mmol) benzylalkohol og 32 mg (0,26 mmol) 4- dimetylaminopyridin (DMAP). Etter 5 timer ved 25 °C ble blandingen fortynnet med 10 ml mettet, vandig natriumbikarbo-natoppløsning og ekstrahert med tre 20 ml porsjoner etylacetat. De kombinerte etylacetatsjikt ble ekstrahert med 10 ml 5 % vandig HC1 og 10 ml mettet natriumbikarbonat, tørket over natriumsulfat og oppkonsentrert, hvorved man fikk 112 mg (100 %) benzylester som en olje som var mer enn 97 % ren ved NMR-analyse. Til en oppløsning av denne olje i 4 ml THF ble det tilsatt 1 ml 10 % vandig HCl-oppløsning. Blandingen ble omrørt ved 25 °C i 30 min, fortynnet med 20 ml mettet, vandig natriumbikarbonatoppløsning og ekstrahert med fire 30 ml porsjoner etylacetat. De kombinerte etylacetatekstrakter ble tørket over natriumsulfat og oppkonsentrert, hvorved man fikk et fast stoff. Rekrystallisering av det faste stoff fra kloro-form gav 92 mg (9S %) benzyl-3-benzamido-3-fenyl-2-hydroksypropionat som hvite krystaller, sm.p. 129-131 °C.
Taxol
Til et lite reaksjonskar ble det tilsatt 109 mg (0,320 mmol) (+)-cis-l-benzoyl-3-(l-etoksyetoksy-4-fenylazeti-
din-2-on, 45 mg (0,064 mmol) 7-0-trietylsilylbaccatin III,
7,8 mg (0,064 mmol) 4-dimetylaminopyridin (DMAP) og 0,032 ml pyridin. Blandingen ble omrørt ved 25 "C i 12 timer og fortynnet med 100 ml etylacetat. Etylacetatoppløsningen ble ekstrahert med 20 ml 10 % vandig kobbersulfatoppløsning, tørket over natriumsulfat og oppkonsentrert. Resten ble filtrert gjennom en plugg av silikagel eluert med etylacetat. Hurtigkromato-grafi på silikagel eluert med etylacetat/heksan, etterfulgt av rekrystallisering fra etylacetat/heksan, gav 61 mg (92 %) 2'-(1-etoksyetoksy)-7-0-trietylsilyl-taxol som en 2:l-blanding av diastereomerer.
En 5 mg prøve av 2'-(1-etoksyetoksy)-7-0-trietylsilyl-taxol ble oppløst i 2 ml etanol, og 0,5 ml 0,5 % vandig HC1-oppløsning ble tilsatt. Blandingen ble omrørt ved 0 °C i 30 timer og fortynnet med 50 ml etylacetat. Oppløsningen ble ekstrahert med 20 ml mettet, vandig natriumbikarbonatoppløs-ning, tørket over natriumsulfat og oppkonsentrert. Resten ble renset ved hjelp av kolonnekromatografi på silikagel eluert med etylacetat/heksan, hvorved man fikk 4,5 mg (ca. 90 %) taxol som var identisk med en autentisk prøve i alle hen-seender .

1. Fremgangsmåte for fremstilling av en taxol med formelen:

E og F er uavhengig av hverandre hydrogen eller lavere alkanoyloksy, alkenoyloksy, alkynoyloksy eller benzoyloksy, eller
E og F danner til sammen okso,
G er hydrogen eller hydroksy eller lavere alkanoyloksy, alkenoyloksy, alkynoyloksy eller benzoyloksy, eller
G og M danner til sammen okso eller metylen, eller G og M danner til sammen oksiran, eller M og F danner til sammen oksetan,
J er hydrogen, hydroksy eller lavere alkanoyloksy, alkenoyloksy, alkynoyloksy eller benzoyloksy, eller
I er hydrogen, hydroksy eller lavere alkanoyloksy, alkenoyloksy, alkynoyloksy eller benzoyloksy, eller

R2 er en hydroksylbeskyttelsesgruppe, og
R3 er fenyl, naftyl eller fenyl eller naftyl som er substituert med lavere alkoksy, eller er lavere alkyl, alkenyl eller alkynyl,
bringes i kontakt med en alkohol med formelen:
hvor A, B, D, E, F, G, I, J, K, L, M, P og Q er som definert ovenfor, idet (3-laktamet og alkoholen bringes i kontakt med hverandre i nærvær av en tilstrekkelig mengde av et aktiveringsmiddel under betingelser som er effektive når det gjelder å få p-laktamet til å reagere med alkoholen, slik at det dannes en p-amidoester som er egnet for anvendelse som et mellomprodukt i syntesen av taxol, og at mellomproduktet omdannes til taxol.

5. Fremgangsmåte ifølge krav 2, karakterisert ved at det anvendes en alkohol hvor R4 er etoksyetyl, trimetylsilyl eller trietylsilyl.
6. Fremgangsmåte ifølge krav 1, karakterisert ved at det som aktiveringsmiddel anvendes trietylamin, diisopropyletylamin, pyridin, N-metylimidazol eller 4-dimetylaminopyridin.
