Description
Foreliggende oppfinnelse angår generelt rekombinant DNA-teknologi og nærmere bestemt en fremgangsmåte for fremstilling av-et rekombinant IL-2-mutein.
Mer spesielt angår oppfinnelsen en fremgangsmåte for fremstilling av mutasjonelt endrede biologisk aktive proteiner som skiller seg fra sine stamanaloger ved substi-tusjon/utelatelse av cysteinrest.
Biologisk aktive proteiner som produseres mikrobielt via rekombinant DNA-teknologi kan inneholde cysteinrester som er uvesentlige for aktiviteten, men som er frie til å danne uønskede intermolekylaere eller intramolekylære bindinger. Et slikt protein er mikrobielt fremstilt human-e-interferon (IFN—e). Under fremstillingen av IFN-g ved rekombinante DNA-teknikker er det observert at dimerer og oligomerer av mikrobielt fremstilte IFN-p dannes i E.coli-ekstrakter inneholdende høye konsentrasjoner av IFN-e. Denne multimer-dannelse gjør rensing og separering av IFN-p meget arbeids-krevende og tidsforbrukende og nødvendiggjør flere ytterligere trinn ved rensing og isolasjon som reduksjon av proteinet under rensingen og reoksydering av dette for å gjenopprette den opprinnelige konformasjon, noe som derved øker muligheten for ukorrekt disulfidbindingsdannelse. Videre er også mikrobielt fremstilt IFN—p funnet å vise konsistent lav spesifikk aktivitet, muligens på grunn av dannelse av multimerer eller tilfeldige intramolekylære disulfidbroer. Det ville derfor være ønskelig å være i stand til å endre mikrobielt fremstilte biologisk aktive proteiner som IFN—P på en slik måte at man ikke påvirker aktiviteten ugunstig, men reduserer eller eliminerer evnen til å danne intramolekylære fornetninger eller intramolekylære bindinger som forårsaker at proteinet opptar en uønsket, tertiær struktur (det vil si en konformasjon som reduserer proteinets aktivitet).
Foreliggende oppfinnelse er rettet mot fremstilling ved direkte mutagenese-teknikker av mutasjonelt endrede biologisk aktive proteiner (slike proteiner kalles "muteiner", se "Glossary of Genetics and Cytogenetics", 4. utgave, side 381, Springer Verlag (1976)), som bibeholder aktiviteten av stamanalogene, men mangler evnen til å danne intermolekylære bindinger eller uønskede intramolekylære disulfidbindinger. I denne henseende beskriver H.M. Shepard et al. i "Nature"
(1981) 294.-563-565 et mutein av IFN—p hvori cysteinet i posisjon 141 i aminosyresekvensen (det er tre cysteiner i nativt human IFN—P ved posisjonene 17, 31 og 141, se "Gene"
(1980) 10:11-15 og "Nature" (1980) 285:542-547) er erstattet av tyrosin. Dette mutein ble fremstilt ved bakteriell eksprimering av et hybridgen konstruert fra en partiell IFN—p cDNA-klon med en G->A-transisjon ved nukleotid 485 i IFN-p-genet. Dette mutein manglet den biologiske aktivitet for nativt IFN-p og ledet forfatterne til den konklusjon at det erstattede cystein var vesentlig for aktiviteten.
Rettede mutagenese-teknikker er velkjente og er oppsummert av R.F. Lather og J.P. Lecoq i "Genetic Engineering", Academic Press, side 31-50 (1983). Oligonukleotid-rettet mutagenese er spesielt nevnt av M. Smith og S. Gillam i "Genetic Engineering: Principles and Methods", Plenum Press, (1981), 3:1-32.
I henhold til dette angår oppfinnelsen, som nevnt innled-ningsvis, en fremgangsmåte for fremstilling av et rekombinant IL-2-mutein og denne fremgangsmåte karakteriseres ved at den ikke-essensielle cysteinrest i posisjon 125 erstattes av serin, dyrking av en rekombinant E.coli-vertscelle som er transformert med en ekspresjonsvektor og som omfatter et strukturelt gen som koder det syntetiske mutein, eller avkom, av nevnte celler, og høsting av det syntetiske mutein fra kulturen.
Fortrinnsvis benytter man et ikke-glykosylert mutein.

Ved hjelp av foreliggende oppfinnelse tilveiebringer man muteiner av biologisk aktive proteiner hvori cysteinrestene som ikke er vesentlige for biologisk aktivitet målrettet er fjernet eller erstattet med andre aminosyrer for å eliminere seter for intermolekylær fornetning eller ukorrekt intramolekylær disulfidbinding og mutante gener som koder slike muteiner.
Proteiner som mutasjonelt kan endres i henhold til oppfinnelsen kan identifiseres fra tilgjengelig informasjon med henblikk på cysteininnholdet av biologisk aktive proteiner og de roller som spilles av cysteinrestene med henblikk på aktivitet og tertiær struktur. For proteiner for hvilke slike informasjoner ikke er tilgjengelige i litteraturen kan denne informasjon bestemmes ved systematisk å endre hver av cysteinrestene i proteinet ved de prosedyrer som heri er beskrevet og å prøve den biologiske aktivitet av de resulterende muteiner og deres tendens til å danne uønskede intermolekylære eller intramolekylære disulfidbindinger. Mens i henhold til dette oppfinnelsen spesielt beskrives og eksemplifiseres nedenfor som angående muteiner av IL-2 skal det være klart at den følgende lære også gjelder ethvert annet biologisk aktivt protein som inneholder en funksjonelt ikke vesentlig cysteinrest som gjør proteinet tilbøyelig til å danne uønskede disulfidbindinger. Eksempler på proteiner andre enn IL—2 som er kandidater for mutasjonen endring i henhold til oppfinnelsen er lymfotoksin (tumor nekrosefaktor) og koloni-stimulerende faktor 1 samt IFN-al. Kandidat-proteinet vil vanligvis ha et ulike antall cysteinrester.
Human IL-2 har tre cysteinrester lokalisert i posisjonene 58, 105 og 125 i proteinet. IL-2 er i aggregert oligomer form når forbindelsen isoleres fra bakterielle celler og må reduseres med reduksjonsmidler for å oppnå gode utbytter fra bakterielle ekstrakter. I tillegg er det rensede reduserte IL-2-protein ustabilt og oksyderes lett ved lagring til en oligomer-inaktiv form. Nærværet av tre cysteiner betyr at proteinet ved reoksydasjon vilkårlig kan danne en av tre mulige intramolekylære disulfidbroer der kun en er den korrekte bro slik den finnes i det native molekyl. Fordi disulfidstrukturen i det native IL-2-protein ikke erkjent, er det mulig å benytte foreliggende oppfinnelse til å skape mutasjoner ved kodonene 58, 105 og 125 I IL-2-genet og å identifisere hvilke cysteinrester som er nødvendig for aktiviteten og derfor mest sannsynlig vil involveres i nativ disulfidbrodannelse. Efter de samme tanker kan cysteinresten som ikke er nødvendig for aktiviteten modifiseres for å forhindre dannelse av ukorrekte intramolekylære disulfidbroer og minimalisere sjansen for intermolekylær disulfidbrodannelse ved å fjerne eller erstatte den frie cysteinrest.
Størrelsen av oligonukleotid-primeren bestemmes av kravet for stabil hybridisering av primeren til regionen for genet hvori mutasjonen skal induseres og ved begrensninger av de idag tilgjengelige metoder for syntetisering av oligonukleotider. Faktorene som må tas med i betraktning ved konstruksjon av oligonukleotider for bruk i oligonukleotid-rettet mutagenese (for eksempel total størrelse, størrelsen på de deler som ligger ved siden av mutasjonssetet) beskrives av M. Smith og S. Gillam, supra. Generelt vil den totale lengde av oligo-nukleotidet være slik at man optimaliserer stabil, unik hybridisering på mutasjonssetet der 5'- og 3'-ekstensjonene fra mutasjonssetet er av tilstrekkelig størrelse til å unngå styring av mutasjonen av exonuklease aktiviteten for DNA-polymerasen. Oligonukleotider som benyttes for mutagenese ifølge oppfinnelsen inneholder vanligvis fra ca. 12 til ca. 24 baser, fortrinnsvis fra ca. 14 til ca. 20 baser og aller helst fra ca. 15 til 18 baser. De vil vanligvis inneholde minst ca. tre baser 3' av det endrede eller manglende kodon.
Oligonukleotid-rettet mutagenese kan benyttes for å frem-stille et mutant IL-2-gen som koder et mutein med IL-2-aktivitet, men har cys-125 forandret til serin-125. Den foretrukne oligonukleotid-primer som benyttes ved fremstilling av dette mutante IL-2-gen når fagene bærer sense-tråden av genet, er GATGATGCTTCTG AGAAAAGGTAATC. Dette oligonukleotid har en C-*G-f orandring ved middelbasen på tripletten som er parret med kodonet 125 i IL-2-genet.
Den resulterende mutasjonelle heterodupleks benyttes så for å transformere en kompetent vertsorganisme eller celle. Replikering fra denne heterodupleks av verten gir avkom fra begge tråder. Efter replikeringen kan mutantgenet isoleres fra avkommet av mutanttråden, innføres i en egnet ekpri-meringsvektor og vektoren benyttes for å transformere en egnet vertsorganisme eller celle. Foretrukne vektorer er plasmidene pBR322, pCRl og varianter derav, syntetiske vektorer og lignende.
Egnede vertsorganismer er E. coli, Pseudomonas, Bacillus subtilis, Bacillus thuringiensis, forskjellige gjærstammer, Bacillus thermophilus, animalske celler slik som ovarieceller fra mus, rotter eller kinesisk hamster (CHO)-celler, planteceller, animalske og planteverter og lignende. Det skal påpekes at når en valgt vert transformeres med vektoren, blir egnede promoter-operatorsekvenser også innført for å uttrykke muteinet. Verter kan være prokaryote eller eukaryote (prosesser for innføring av DNA i eukaryote celler er beskrevet i PCT-søknad US 81/00239 og US 81/00240, publisert 3. september 1981). E. coli og CHO-celler er de foretrukne verter. Mutelnene som oppnås ved fremgangsmåten Ifølge oppfinnelsen kan være glykosylerte eller ikke-glykosylerte, avhengig av glykosyleringen som opptrer i det native stamprotein og vertsorganismen som benyttes for å produsere muteinet. Hvis ønskelig kan eventuelt ikke-glykosylert mutein oppnådd når E.coli eller en Bacillus er vertsorganisme glykosyleres In vitro ved kjemisk, enzymatisk eller en annen kjent modifisering.
De følgende eksempler 1-5 skal gis for å bidra til en bedre forståelse av oppfinnelsen og er kun Illustrerende for fremstilling av et mutein av IL-2.

Nukleotidsekvensen for en cDNA-klon som koder for human IL—2, prosedyrer for fremstilling av IL-2 cDNA-bibliotek og screening av disse for IL-2 er beskrevet av T. Taniguchi et al. i "Nature", 1983, vol. 24, side 305 og følgende.
cDNA-bibliotek anriket på potensielle IL-2 cDNA-kloner ble fremstilt fra på IL-2-anrikede mRNA-fraksjoner oppnådd fra induserte perifere blodlymfocytter (PBL) og Jurkat-celler ved konvensjonelle prosedyrer. Anrikningen av mRNA på IL-2-signal ble oppnådd ved fraksjonering av mRNA og identifisering av fraksjonen med IL-2 mRNA-aktivitet ved injisering av fraksjonene i Xenopus laevis-oocytter og prøving av oocytt-lysatene på IL-2-aktivitet på HT-2-celler (J. Watson, "J. Exp. Med." (1979) 150:1570-1519 og S. Gillis et al., "J. Immun." (1978) 120:2027-2032).
Eksempel 2
Screening og identifisering av IL- 2- cDNA- kloner
IL-2 cDNA-bibliotek ble screenet ved bruk av kolonihybridi-seringsprosedyren. Hver mikrotiterplate ble replikert på duplikate nitrocellulosefilterpapir (S & S type BA-85) og kolonier ble tillatt å vokse ved 37°C i 14-16 timer på L-agar inneholdende 50 jjg/ml ampicillin. Koloniene ble lysert og DNA fiksert til filteret ved sekvensiell behandling i 5 minutter med 500 mM NaOH, 1,5 M NaCl, vasket to ganger, 5 minutter hver gang med 5 x SSC. Filtrene ble lufttørket og oppvarmet til 80°C i 2 timer. DuplikatfUtrene ble prehybridisert ved 42°C i 6-8 timer med 10 ml/filter DNA-hybridiseringsbuffer (5056 formamid, 5* SSC, pH 7,0, 5 x Denhardts's oppløsning (polyvinylpyrrolidin pluss ficoll og storfeserum albumin; lx = 0,2* av hver) 50 mM natriumfosfatbuf f er ved pH 7,0, 0,2* SDS, 20 jjg/ml Poly U og 50 jjg/ml denaturert laksesperm DNA.
En 3<Jp_meri£e-t 20-mer oligonukleotidsonde ble preparert basert på IL-2-gensekvensen som angitt av T. Taniguchi et al. ovenfor. Nukleotidsekvensen for proben var
GTGGCCTTCTTGGGCATGTA.
Prøvene ble hybridisert ved 42°C i 24-36 timer med 5 ml/filter DNA-hybridiseringsbuffer inneholdende <3>^p cDNA-proben. Filtrene ble vasket to ganger 30 minutter hver gang ved 50"C med 2 x SSC, 0,1* SDS, derefter vasket to ganger med 1 x SSC og 0,1* SDS ved 50° C i 90 minutter, lufttørket og autoradiografert ved -70°C i 2 til 3 dager. Positive kloner ble identifisert og screenet om igjen med proben. Fullengde-kloner ble identifisert og bekreftet ved restriksjonsenzym-kartleggelse og sammenligning med sekvensen av IL-2 cDNA som var klonet som angitt av T. Taniguchi et al. ovenfor.
Eksempel 3
Kloning av IL- 2- genet inn i M13- vektor
IL-2-genet ble klonet inn i M13mp9 ved bruk av plasmid pLWl (figur 1) inneholdende IL-2-genet under kontroll av E.coli trp-promoteren. En prøve av pLWl er deponert i "American Type Culture Collection", 12301 Parklawn Drive, Rockville, Maryland 20852, USA, den 4. august 1983 og er gitt ATCC-nummer 39 405. Restriksjonskartet for en klon (betegnet M13-IL2) inneholdende IL-2-innskuddet er vist i figur 2.
Enkelttrådet fag DNA ble fremstilt fra klon M13 IL-2 for å tjene som templat for oligonukleotid-rettet mutagenese.
Eksempel 4
Ollgonukleotld- rettet mutagenese
Som antydet tidligere inneholder IL-2 cysteinrester i aminosyreposisjonene 58, 105 og 125. Basert på nukleotid-sekvensene for andelene av IL-2-genet som inneholder kodonene for disse tre cysteinrester, ble tre oligonukleotid-primere konstruert og syntetisert for mutasjon av kodonene for disse rester til kodoner for serin. Disse oligonukleotider har de følgende sekvenser: CTTCTAGAGACTGCAGATGTTTC (DM27) for å forandre cys 58, CATCAGCATACTCAGACATGAATG (DM28) for å forandre cys 105 og GATGATGCTCTGAGAAAAGGTAATC (DM29) for å forandre cys 125. 40 picomol av hvert nukleotid ble kinasebehandlet separat i nærvær av 0,9 mM ATP, 50 mM Tris-HCl, pH 8,0, 10 mM MgCl2, 5 mM DTT og 9 enheter T^kinase i 50 jjI ved 37"C i 1 time. Hver av dekinasebehandlede primere (10 pmol) ble hybridisert til 2,6 jjg ss M13-IL2 DNA i 15 pl av en blanding inneholdende 100 mM NaCl, 20 mM Tris-HCL, pH 7,9, 20 mM MgCl2 og 20 mM p-merkaptoetanol, ved oppvarming til 67"C i 5 minutter og 42°C i 25 minutter. Den oppvarmede blanding ble avkjølt på is og derefter justert il et sluttvolum på 25 pl av en reaksjons-blanding inneholdende 0,5 mM av hver dNTP, 17 mM Tris-HCl, pH 7,9, 17 mM MgCl2, 83 mM NaCl, 17 mM p<->merkaptoetanol, 5 enheter DNA-polymerase i Klenow-fragment, 0,5 mM ATP og 2 enheter DNA-ligase, inkubert ved 37°C i 5 timer. Reaksjonen ble avsluttet ved oppvarming til 80°C og reaksjonsblandingen benyttet for å transformere kompetente JM103-celler, platert på agarplater og inkubert over natt, for å oppnå fag-plaquer.
Eksempel 5
Screening og identifisering av muterte fag- plaquer
Plater inneholdende muterte M13-IL2-plaque så vel som 2 plater inneholdende ikke muterte M12-IL2-fagplaque ble avkjølt til 4°C og fag-plaque fra hver plate overført til to nitrocellulosefiltersirkler ved å legge et tørrfilter på agarplaten i 5 minutter for det første filter og 15 minutter for det andre filter. Filtrene ble derefter anbragt på tykke filterpapirer dynket i 0,2N NaOH, 1,5 M NaCl og 0,2* Triton i 5 minutter og nøytralisert ved å legge dem på filterpapiret dynket med 0,5 M Tris-HCl, pH 7,5 og 1,5 M NaCl i ytterligere 5 minutter. Filtrene ble vasket på en tilsvarende måte to ganger på filteret dynket i 2 x SSC, tørket og derefter oppvarmet i en vakuumovn til 80"C i 2 timer. DuplikatfUtrene var forhybridiserte ved 42°C i 4 timer med 10 ml pr. filter DNA-hybridiseringsbuffer (5 x SSC, pH 7,0, 4 x Denhardts-oppløsning (polyvinylpyrrolidon, ficoll og storfeserum albumin, lx = 0,02* av hver), 0,1* SDS 50 mM natriumfosfat-buffer, pH 7,0 og 100 pg/ml denaturert laksesperma DNA. <32>P-merket sonder ble preparert ved kinasebehandling av oligonukleotid-primerene med merket ATP. Filtrene ble hybridiserte til 0,1 x IO<5> cpm/ml <32>P-merket primer i 5 ml/filter DNA hybridiseringsbuffer ved 42°C i 8 timer. Filtrene ble vasket to ganger ved 50° C 30 minutter hver gang i vaskebuffere inneholdende 0,1* SDS og 0,2* SSC, og to ganger ved 50°C i 30 minutter hver med 0,1* SDS og 0,2 x SSC. Filtrene ble lufttørket og autoradiografert ved -70°C i 2-3 dager.
Fordi oligonukleotid-primerene DM28 og DM29 ble konstruert for å skape et nytt Ddel-restriksjonssete i de muterte kloner (figur 3), ble RF-DNA fra et antall av klonene som hybridiserte med hver av disse kinasebehandlede primere spaltet med restriksjonsenzymet Ddel. En av de muterte M13-IL2-plaque som hybridiserte med primeren DM28 og hadde et nytt Ddel-restriksjonssete (M13-LW44) ble plukket ut og inokulert i en kultur av JM103, ssDNA ble preparert fra kultur-supernatanten og dsRF-DNA ble preparert fra cellepelleten. På samme måte ble en plaque som hybridiserte med primeren DM29 plukket ut (M13-LW46) og ssDNA og RF-DNA fremstilt derfra. Oligonukleotid-primeren DM27 ble konstruert til å skape et nytt Pstl-restriksjonssete i stedet for et Ddel-sete. Derfor ble plaque som hybridiserte til denne primer screenet med henblikk på nærvær av et nytt PstI-sete. En slik. fagplaque ble identifisert (M13-LW42) og ssDNA og RF-DNA fremstilt derfra. DNA fra alle tre av disse kloner ble sekvensert for å bekrefte at de ønskede TGT-kodoner for cystein var omdannet til en TCT-kodon for serIn.

Subklonlng av mutert IL- 2- gen for eksprimerlng 1 E. coli RF-DNA fra M13-LW42, M13-LW44 og M13-LW46 ble spaltet med restriksjonsenzymene HindiII og BanII og innskuddsfragmentene renset fra en 1% agarosegel. På samme måte ble plasmidet pTrp3 spaltet med Hindlll og BanII der det store plasmid-fragment inneholdende trp-promoteren ble renset på en agarosegel og derefter ligert med hver av innskuddsfragmentene isolert fra M13-LW42, M13-LW44 og M13-LW46. De ligerte plasmider ble transformert til kompetent E.coli K12 stamme MM294. Plasmid DNA fra disse transformanter ble analysert ved restriksjonsenzyms kartlegging for å bekrefte nærværet av plasmidene pLW42, pLW44 og pLW46. Figur 3 er et restriksjonskart av pL¥46. Når hver av disse individuelle kloner ble dyrket i fravær av tryptofan for å indusere trp-promoter, og cellefrie ekstrakter analysert på SDS-polyakryl-amidgeler, viste alle de tre kloner pLW42, pLW44 og pLW46 seg å syntetisere et 14,5 kd protein tilsvarende det som finnes i den positive kontroll, pLW21, som ble vist å syntetisere et 14,4 kd IL-2-protein. Når disse samme ekstrakter ble underkastet prøving på IL-2-aktivitet på muse HT-2-celler, hadde kun klonene pLW21 (positiv kontroll) og pLW46 signifi-kant mengde IL-2-aktivitet (tabell I nedenfor), noe som antyder at cys 58 og cys 105 er nødvendige for biologisk aktivitet og forandring av disse til seriner (pLW42 henholdsvis pLW44) resulterte i tapet av biologisk aktivitet. Cys 125 må på den annen side ikke være nødvendig for biologisk aktivitet fordi forandring av den til ser 125 (pL¥46) ikke påvirket den biologiske aktivitet.
Figur 4 viser nukleotidsekvensen av kodingstråden av klon pLW46. Sammenlignet med kodingstråden av den native human IL-2-genklon pLW46 hadde en enkeltbaseforandring g-+C ved nukleotid 374. Figur 4 viser den tilsvarende aminosyresekvens for IL-2-mutein kodet av pLW46. Dette mutein kalles IL—2 inc* Sammenlignet med nativt IL-2 har muteinet serin i stedet for cystein i posisjon 125.
En prøve av E. coli K12 stamme MM294, transformert med pLW46, er deponert i "American Type Culture Collection", 12301 Parklawn Drive, Rockville, Maryland 20852, USA, den 26. september 1983 under nr. ATCC 39 452.
Muteiner av IL-2 hvori cysteinet i posisjon 125 er utelatt eller erstattet med en annen aminosyre slik som mutein IL-2 40e beholder sin IL-2-aktivitet. De kan derfor

formuleres og benyttes på samme måte som nativt IL-2.
I henhold til dette er slike IL-2-muteiner brukbare ved diagnose og behandling av bakterielle, virale, parasittiske, protozo- og fungalinfeksjoner; ved manifestering av lymfokin eller immunoeffekter; for rekonstituering av normal immuno-funksjon hos eldre mennesker og dyr; ved utvikling av diagnostiske prøver som de som benytter enzymforsterkning, radiomerking, radiobilder og andre metoder som er kjent for å overvåke IL-2-nivåer i den syke tilstand; for fremming av T-cellevekst in vitro for terapeutiske og diagnostiske formål for å blokkere reseptorseter for lymfoklner; og forskjellige andre terapeutiske, diagnostiske og forskningsanvendelser. De forskjellige terapeutiske og diagnostiske anvendelser av human IL-2 er undersøkt og angitt av S.A. Rosenberg, E.A. Grim et al., A. Mazumder et al. samt E.A. Grim og S.A. Rosenberg. IL-2-muteiner kan benyttes som sådanne eller i kombinasjon med andre immunologiske relevante B- eller T-celler eller andre terapeutiske midler. For terapeutiske eller diagnostiske anvendelser kan de formuleres i ikke-toksiske, ikke-allergene, fysiologisk kompatible bærere som destillert vann, Ringers-oppløsning, Hanks-oppløsning, fysiologisk saltoppløsning og lignende. Inngivelse av IL-2-muteiner til mennesker eller dyr kan skje oralt, intra-peritonealt, intramuskulært eller subkutant alt efter hva som er mest hensiktsmessig. Eksempler på relevante celler er B-eller T-celler, naturlige utrydderceller og lignende og eksempler på terapeutiske reagenser som kan benyttes i kombinasjon med polypeptidene ifølge oppfinnelsen er de forskjellige interferoner, spesielt T-interferon, B-celle-vekstfaktor, IL-1 og lignende.
Claims (2)
1.
Fremgangsmåte for fremstilling av et rekombinant IL-2-mutein, karakterisert ved at den ikke-essensielle cysteinrest I posisjon 125 erstattes av serin, dyrking av en rekombinant E.coli-vertscelle som er transformert med en ekspresjonsvektor og som omfatter et strukturelt gen som koder det syntetiske mutein, eller avkom, av nevnte celler, og høsting av det syntetiske mutein fra kulturen.

Fremgangsmåte ifølge krav 1, karakterisert ved at muteinet er ikke-glykosylert.
