Description
Foreliggende oppfinnelse gjelder en fremgangsmåte for å fremstille nye og hittil utilgjengelige 2-guanidino-4-(2-substituerte-amino-4-imidazolyl)tiazoler, som også er gunstig ved fremstilling av kjente forbindelser som er lavere homologe av disse.
Disse forbindelser fremstilt ifølge oppfinnelsen, er aktive som antisekretoriske midler, motvirker histamin-H^ og/ eller hemmer etanol-indusert gastrisk ulcerasjon hos rotter, og er således nyttige når det gjelder å hemme (dvs. forhindre og behandle) ulcus pepticum.
Kronisk sårdannelse i mave og tolvfingertarm, tilsammen kjent som ulcus pepticum, er en vanlig lidelse som det kan brukes en rekke forskjellige behandlingsmåter mot, inkludert diett, behandling med medisiner og operasjon, avhengig av hvor alvorlig tilstanden er. Særlig verdifulle terapeutiske midler som er nyttige ved behandling av overproduksjon av mavesyre og ulcus pepticum, er histamin-F^-receptor-antagonister, som virker ved å blokkere påvirkningen av den fysiologisk aktive forbindelse histamin på f^-receptor-posisjoner i dyrelegemet, og derved hindrer utskillelse av mavesyre. Det faktum at mange av de foreliggende forbindelser fremstilt ifølge oppfinnelsen, også hemmer etanol-indusert ulcus hos rotter, viser videre den kliniske verdi av de foreliggende forbindelser fremstilt ifølge oppfinnelsen, når det gjelder å forhindre mave.sår.
Forbindelser av den foreliggende type, som har formelen
der Ra er H, (C^-Cg)-alkyl, eller (C^-C^g)-f enylalkyl, hvor nevnte fenylgrupper er valgfritt monosubstituert med klor-, brom-, fluor-, (C-l-C^ V-alkyl eller (C1~C3)-alkoksy, er tidligere beskrevet som midler mot ulcus på grunn av deres antisekretoriske og histamin-H2-motvirkende aktivitet, se EP patentpublikasjon 50458.
Den nye fremgangsmåten i foreliggende oppfinnelse er gunstig ved fremstilling av forbindelser med formel (Ia), særlig når Ra er (C3-C6)-alkyl eller (C7~C10)-fenylalkyl. Videre tillater foreliggende fremgangsmåte fremstilling av forbindelser med beslektet struktur som ikke er tilgjengelige ved fremgangsmåter som er fremlagt tidligere i den ovenfor nevnte EP-patentpublikasjon.
En nøkkel til den foreliggende fremgangsmåte er den opp-dagelse at et primært eller sekundært amin kan reagere med 2-amino-5-acetyloksazol, med formel (IV) nedenunder, og danne forbindelsen som inngår i reaksjonen, 2-(substituert-amino)-5-acetylimidazol, med formel (II) nedenunder. Tidligere er det aldri observert noen slik reaksjon. Ringoksygenet i 2-metyloksazol-4-karboksylsyren er erstattet med NH og deretter med NCgH<- under tilsvarende betingelser med samtidig dekarboksy-lering. Cornforth og Cornforth, J. Chem. Rekken av karbonatomer i parentes refererer til det totale antall karbonatomer i gruppen som følger. Karbonkjeden kan være rett eller grenet; substituentgruppen, såsom fenyl, kan substitueres på et hvilket som helst karbon i kjeden. Farma-søytisk godtagbare syreaddisjonssalter er de med én eller to ekvivalenter syre. Egnede syrer omfatter, men er ikke begrenset til HC1, HBr, H2S04, H3P04, CH3SC>3H, p-CH3C6H4S03H, maleinsyre, fumarsyre, ravsyre og sitronsyre. Foreliggende oppfinnelse gjelder en fremgangsmåte for å fremstille forbindelser med formel
der R<4> er NHR<5> eller NR<2>R3;
<R5> er (C1-C12)-alkyl, (C3-Cg) cyk-loalkyl, pyridyl-(C^-Cg) -
alkyl eller fenyl-(C^-Cg)-alkyl; og
R 2 og R<3> begge er. uavhengig (C^-C^)alkyl, eller
R 2 og R "3 danner tilsammen med nxtrogenatomet som de er knyttet til, en pyrrolidin-, piperidin-, perhydro-lH-azepin-eller morfolin-ring. Totalreaksjonen omfatter de følgende kjemiske trinn:
Det er å foretrekke å isolere produktet (I) direkte
fra reaksjonsblandingen som monohydrogenbromidsaltet, som om ønsket kan overføres til den fri base eller til et annet alternativt farmasøytisk godtagbart salt, f.eks. dihydrogen-klorid.
Forbindelsene med formel (I) fremstilles lett ved bruk
av fremgangsmåten ifølge foreliggende oppfinnelse, som skissert i de kjemiske trinn ovenfor: (IV)— > (II) (I).
Det første trinn, (IV)—» (II), utføres i nærvær av et overskudd av aminreaktanten i et vandig oppløsningsmiddel.
Når reaktantene løses opp under reaksjonsbetingelsene, er det foretrukne oppløsningsmiddel vann alene. Når det er ufull-stendig oppløsning av reaktantene under reaksjonsbetingelsene, tilsettes et polart, organisk oppløsningsmiddel som er bland-bart med vann og reaksjonsinert. Brukt som her, er et "reaksjonsinert oppløsningsmiddel" ett som ikke reagerer med reagensene, de forbindelser som dannes i løpet av reaksjonen eller produktene, på noen måte som påvirker utbyttet av det ønskede produkt i gal retning. I det foreliggende tilfelle er etere, såsom tetrahydrofuran eller lavere alkoholer fortrinnsvis med forgrenet kjede, velegnet. Det foretrukne organiske oppløsningsmiddel, når det er nødvendig for å få oppløselighet, er isopropanol. Reaksjonens gang kan lett følges ved vanlige metoder i tynnsjikt-kromatografi (tic)
som er vel kjent innen faget, f.eks. ved å bruke silikagel-plater med kloroform som inneholder 1-10% metanol som eluerings-middel. Reaksjonen fortsettes gjerne inntil utgangsstoffet aminooksazol ikke lenger kan påvises ved tic, og gjør det derved lettere å isolere den rene forbindelsen som inngår videre i reaksjonen. Reaksjonen kan utføres over et vidt temperaturområde, f.eks. 50-150°C, under trykk om nødvendig. Hensiktsmessig foregår reaksjonen ved reaksjonsblandingens tilbakeløps-temperatur (50-100°C), avhengig av kokepunktet til det vandige amin eller vandig amin og organisk oppløsningsmiddel sammen. Forbindelsene (II) som skal inngå videre i reaksjonen, isoleres vanligvis ved fordampning og renses ved kolonnekromatografi.
Om ønsket kan produktet renses ytterligere ved omkrystallisering
i et organisk oppløsningsmiddel såsom acetonitril, toluen eller cykloheksan.
Det annet trinn i total-reaksjonen, (II) —>(III), er syre-katalysert bromering av. metylgruppen i (n) som aktiveres av en karbonylgruppe. Mens en rekke forskjellige standard bromerings-midler kan benyttes i denne reaksjonen, er det foretrukne reagens brom selv. Den foretrukne syre som også hensiktsmessig brukes som oppløsningsmiddel, er konsentrert hydrogenbromsyre. Forbindelsen (III), som skal inngå videre i reaksjonen, isoleres lett i form av sin fri base, som er uoppløselig i vann, ved konsentrering og nøytralisering med en vandig base.
Det tredje trinn i total-reaksjonen, (III)—> (I) , omfatter kondensering av forbindelsen med formel (III) med omtrent én molekvivalent amidinotiourinstoff. Reaksjonen foregår i et reaksjonsinert organisk oppløsningsmiddel såsom tetrahydrofuran, en lavere alkohol, såsom metanol, etanol eller isopropanol,
eller et lavere keton såsom aceton eller metyletylketon. Aceton er et foretrukket oppløsningsmiddel ettersom produktet bunnfelles rent som monohydrogenbromidsaltet fra dette oppiøsningsmidlet og kan derfor greit samles opp med stort utbytte ved enkel filtrering. Hydrogenbromidsaltet overføres i^tt til fri base ved vanlige nøytraliserings/ekstraherings-metoder. For å fremstille andre farmasøytisk godtagbare salter, løses den fri base opp i et organisk oppløsningsmiddel og enten én eller minst to ekvivalenter av syren som tilsvarer det ønskede salt, tilsettes. Saltet samles opp-ved filtrering, konsentrering eller tilsetning av et ikke-oppløsende reagens, eller ved en kombinasjon av disse trinn. Det foretrukne salt er dihydrogenkloridet, som lett felles ut fra en metanol-oppløsning av den fri base ved diffusjon av oppløsningen med et overskudd av vannfritt hydrogenklorid.
5-acetyl-2-amino-oksazol som trengs til den foreliggende fremstillingsmåte, er en kjent forbindelse. Kochetikov et al., Chemical Abstracts 54 : 14230h (1960). Det er gitt eksempel på en foretrukket fremstillingsmåte nedenfor. Amidinotiourinstoff og aminene som også trengs til foreliggende fremgangsmåte, er kommersielt tilgjengelige eller tilgjengelige ifølge litteratur-metoder.
For eksempel, 2-aminoheptan er tilgjengelig ut fra 2-heptanon ved reduksjon av oksimet, reduktiv aminering eller Leukart-reaksjonen, Rohrmann og Schonle, J. Am. Chem. Soc. 70: 2811 (1948); 2-metyl-l-aminocykloheksan er tilgjengelig ved
Hoffmann-degradering av 2-metylcykloheksan-l-karboksamid,
Gutt. Ber. 40 : 2061 (1907); N-etyl-2-feny1-2-propylamin er tilgjengelig ved reduktiv aminering, Woodruff et al., J. Am. Chem. Soc. 62 : 922 (1940); og 2-(3-pyridyl)-2-propylamin fremstilles ved Leukart-reaksjonen, Burger og Walters, ibid, 72: 1988 (1950).
Nytten av forbindelsene med formel ( x) når det gjelder å forhindre ulcus hos pattedyr, innbefattet menneske, avspeiles i deres antisekretoriske, histamin-H2-motvirkende effekt og/eller ved at de forhindrer etanol-indusert ulcus hos rotter, som vist i detalj i eksemplene nedenunder. For å hemme (forhindre eller behandle) mavesår hos et pattedyr, administreres forbindelsene fremstilt ifølge foreliggende oppfinnelse på en rekke vanlige administreringsmåter, inkludert oral og parenteral. Fortrinnsvis administreres forbindelsene oralt. Generelt administreres disse forbindelser oralt i doser mellom 0,1 og 20 mg/kg kroppsvekt av pasienten som skal behandles, pr. dag, i enkle eller delte doser. Hvis det er ønskelig med parenteral administrering, kan disse forbindelser gis i daglige totaldoser mellom 0,1 og 1,0 mg/kg kroppsvekt av pasienten som skal behandles. Imidlertid, etter den behandlende leges skjønn, vil noen variasjon i doseringen nødvendigvis forekomme, avhengig av tilstanden til pasienten som behandles og den spesielle forbindelse som benyttes.
Foreliggende oppfinnelse er illustrert ved de følgende eksempler. Imidlertid må det være forstått at oppfinnelsen ikke er begrenset til de spesifikke detaljer i disse eksempler.

Eksempel 1
5- acetyl- 2- aminooksazol ( IV)
En blanding av 132,3 g (0,80 m) 2-brom-l-hydroksy-3-okso-l-buten, 120,1 g (2,0 m) urinstoff og 1,85 1 aceton ble varmet med tilbakeløp og røring ovenfra i 1 time. Blandingen ble konsentrert og olje-residuet ble tatt opp i 600 ml vann, og deretter gjort basisk med konsentrert ammoniumhydroksyd. Etter henstand ved romtemperatur i 0,5 time, ble det dannet et bunnfall. Dette ble samlet og tørret i vakuum og det ga 61,1 g råprodukt. Filtratet ble konsentrert igjen og olje-residuet ble tatt opp
i 50 ml. vann og igjen gjort basisk med konsentrert ammoniumhydroksyd. Etter henstand over natten, ble en ny <p>orsjon rå-
produkt, 17,6 g, isolert. Begge porsjoner ble samlet og omkrystallisert fra metanol, og det ga 50,3 g (50%) 5-acetyl-2-aminooksazol, smp. 214-215°.
Eksempel 2
Generell fremgangsmåte for å fremstille 2- substituerte- amino- 5- acetylimidazoler ( II)
En blanding av 2,0 g (16 mmol) 5-acetyl-2-aminooksazol,
20 ml av det passende amin og 30 ml vann ble varmet opp med tilbakeløp i 3 til 96 timer, som indikert under (når det gjaldt noen lipofile aminer, ble isopropanol tilsatt som en nødvendig-het for å oppnå en homogen reaksjonsblanding). Blandingen ble konsentrert (destillert om nødvendig, for å fjerne alle spor av aminet) og residuet ble deretter kromatografert på 60 g silikagel, med bruk av 4:1 etylacetat/heksan som eluant. Da alt det mindre polare materialet (pyrimidin biprodukt) var eluert, ble kolonnen eluert med 19:1 kloroform/metanol for å få det mer polare imidazol-produkt. Analytisk rent imidazol ble fremstilt ved omkrystallisering fra de rette oppløsningsmidler som vist i detalj nedenfor. På denne måte ble de følgende 2-substituerte amino-5-acetylimidazoler fremstilt:
Eksempel 3
Generell fremgangsmåte for å fremstille 2- substituerte- amino- 5- ( 2- bromacetyl) - Imidazoler ( HI)
En oppløsning av 1,0 g av det rette keton i 25 ml konsentrert hydrogenbromidsyre ble rørt ved romtemperatur og 5% molart overskudd av brom ble tilsatt dråpevis i løpet av 2 minutter. Blandingen ble deretter varmet ved 80° (eksternt) i 1 time,
og i løpet av den tiden forsvant brom-farven. Blandingen ble avkjølt, og deretter konsentrert. Residuet ble triturert med mettet natriumbikarbonatoppløsning og bunnfallet som ble dannet fra dette basismediet ble samlet, vasket med vann, og deretter tørret i vakuum, og det ga de følgende 2-amino-5-(2-bromacetyl)-imidazoler som faste stoff som bie karakterisert ved nmr-spektroskooi i DMSO-dg, hvis ikke annet er angitt:
Eksempel 4
Generell fremgangsmåte for å fremstille 2-guanidin-4-(2-substituert-amino-4-imidazolyl)-tiazol-hydrogen-bromid ( I. HBR)
En blanding av 1 g bromacetylimidazol fra det foregående eksempel, en ekvimolar mengde amidinotiourinstoff og 50 ml aceton, ble varmet med tilbakeløp i 1 time, og i løpet av den tiden ble produktet felt ut som sitt monohydrogenbromidsalt. Dette faste stoff ble samlet, vasket med aceton, og deretter tørret i vakuum. Det ble deretter overført til sitt c<f>ihydrogen-kloridsalt som skissert i eksempel 5.
Eksempel 5
Generell fremgangsmåte for å fremstille 2-guanidin-4-(2-substituert-amino-4-imidazoly1)-tiazol-dihydrogen-klorid[T.(HCl) 2]
Hydrogenbromidsaltet ble rørt i 50 ml mettet NaKCO-^-oppløsning i 15-30 minutter. Det faste stoff, nå som fri base, ble samlet, vasket med vann og deretter tørret i vakuum. Dette faste stoff ble tatt opp i en minimal mengde metanol. Metanol-oppløsningen ble mettet med HCl-gass og deretter sakte for-tynnet med eter. Det resulterende bunnfall ble samlet, vasket med eter, deretter tørret i vakuum, og det ga dihydrogenklorid-saltet som ble karakterisert ved forbrenningsanalyse og/eller spektraldata som følger:
SAMMENLIGNINGSFORSØK
Fremgangsmåten ifølge europeisk patentpublikasjon 50458 ble sammenlignet med fremgangsmåten ifølge foreliggende oppfinnelse. Det ble funnet at den kjente fremgangsmåte ikke gav noen utbytter når R<4> (NHR<5>) var NH(C7H15) eller en høyere homolog og forholdsvis lave utbytter for lavere homologer. Dette fremgår av de følgende resultater som ble oppnådd ved forsøkene:
Antisekretorisk aktivitet mot mavesyre
Den antisekretoriske aktivitet mot mavesyre til disse forbindelsene fremstilt ifølge foreliggende oppfinnelse, ble bestemt på bevisste "Heidenhain pouch dogs" som hadde fastet over natten. Pentagastrin (Pentavolon-Ayerst) ble brukt for å stimulere utskillelsen av syre ved kontinuerlig infusjon inn i en overfladisk leggvene, i doser som tidligere var bestemt til å stimulere bortimot maksimal syreutskillelse fra mavesekken. Mavesaft ble samlet opp med 30 minutters mellomrom etter starten av pentagastrin-infusjonen, og ble målt til nærmeste 0,1 ml. 10 oppsamlinger ble tatt av hver hund i løpet av et eksperiment. Syrekonsentrasjonen ble bestemt ved titrering av 1,0 ml mavesaft til pH 7,4 med 0,IN natriumhydroksyd, med hjelp av en Autobyrette og et glass-elektrode-pH-meter (Radiometer). Medikament eller hjelpestoff ble gitt intravenøst 90 minutter etter start av pentagastrin-infusjonen, i en dose på 1 mg/kg eller mindre. Den antisekretoriske virkning på mave-saften ble beregnet ved å sammenligne den laveste syreproduksjon etter administrering av medikamentet med den midlere syreproduksjon umiddelbart før medikamentet.
Produktene a til h, n, r og cc i eksempel 5, i en dose på
1 mg/kg, hemmet utskillelsen av mavesyre minst 21%. De foretrukne produktene f til h og w, hemmet utskillelse av mavesyre minst 97% ved samme eller lavere dose. I en dose på 3 mikrog/kg,
ga forbindelse n 42% hemning. Ved 0,1 mg/kg, ga forbindelse cc 72% hemning.
Histamin- H0- antagonistisk aktivitet
Den histamin-H2~antagonistiske aktivitet til forbindelsene fremstilt ifølge foreliggende oppfinnelse, ble bestemt ved følgende fremgangsmåte:
Marsvin drepes raskt med et slag i hodet, hjertet fjernes
og de høyre forkammere dissekeres fri. Hjertekammerene suspenderes, isometrisk, i et temperatur-kontrollert (32° + 2°) vevsbad (10 ml) som inneholder oksygenert '(95% 0,,; 5% CO.,) Krebs-Henseleit-buffer (pH 7,4), og får stabiliseres ca. 1 time,
og i løpet av den tiden gjennomstrømmes vevsbadet flere ganger. Individuelle forkammer-kontraksjoner følges med en kraft-forskyvnings-transduser forbundet med et kardiotakometer og Grass polygraph recorder. Etter å ha fått en dose-respons-kurve overfor histamin, gjennomspyles hvert hjertekammer flere ganger med ny buffer, og hjertekammerene ekvilibreres påny til utgangsverdiene. Etter tilbakegang til utgangsverdiene, tilsettes testforbindelsene i utvalgte endelige konsentrasjoner, og dose-respons for histamin bestemmes påny i nærvær av antagonist. Resultatene uttrykkes som dose-forhold, forholdet mellom histamin-konsentrasjonen som trengs for å gi halvparten av maksimal stimulering i nærvær og fravær av antagonist, og den tilsynelatende dissosiasjonskonstanten til H2_reseptor-antagonisten, pA2 bestemmes.
Produktene a til 1, n, o og q til bb i eksempel 5, ga pA2~ verdier på minst 5,9. De foretrukne produkter f til i, n, s, t og w ga pA2 på 7,0 eller større. Den høyeste verdi (8,8) ble oppnådd med produkt n.
Hemning av etanol- indusert ulcus- dannelse hos rotter Anti-ulcus-aktiviteten til produktene fremstilt ifølge denne oppfinnelse, ble også bestemt ved en analyse av etanol-indusert ulcus hos rotter. I denne test ble han-rotter som hadde fastet over natten gitt medikament (5 mg/kg) eller vann oralt 15 minutter før en oralt administrert dose absolutt etanol (1,0 ml). 1 time etter påkjenningen med etanol, drepes dyrene (8/gruppe) og mavesekkene undersøkes med henblikk på skader. Etter avliving åpnes abdomen, og en låsende hemostat plasseres ved pyrolus. 6 ml av en 4°5 oppløsning av formaldehyd ble injisert i maven med et rør til å fylle mavesekken, og en annen låsende hemostat ble brukt for å forsegle spiserøret. Mavesekken ble fjernet, åpnet langs den store kurvatur og undersøkt med henblikk på ulcus-dannelse.

Ulcus- bedømmelsestabell
For hver gruppe dyr beregnes en ulcus-indeks som følger: Ulcus-dannelses-indeks = (summen av poengene i gruppen) x (summen av antall ulcera i gruppen) x (andelen av gruppen som har noe som helst tegn på ulcus-dannelse).
Prosent hemning av ulcus-dannelse beregnes som følger:
% hemning = 100 x [ (ulcus-indeks kontroll) - (ulcus-indeks medikament-behandlet)] r (ulcus-indeks kontroll).
Ved en oral dose på 30 mg/kg, viste produktene a til k, m,
r til u, w, y, z, bb og cc i eksempel 5 minst 19% hemning av etanol-indusert ulcus-dannelse. Ved den samme dosering, viste forbindelsene o, v, x og aa ingen signifikant aktivitet, forbindelsene n, p og q viste 7-13% hemning, forbindelsene u og cc viste 21-51% hemning, og foretrukne forbindelser f til i, s, t og w viste 86-100% hemning.
Claims (1)
- Fremgangsmåte for å fremstille en forbindelse med formelder R<4> er NHR<5> eller NR<2>R<3>;R<5> er (C1-C12)-alkyl, (C3-C8)-cykloalkyl, pyridyl-(C^-Cg)-alkyl eller fenyl-(C1-C6)-alkyl; ogR<2> og R<3> er begge uavhengig (C^-Cg)-alkyl, ellerR<2> og R<3> danner sammen med det nitrogen som de er bundet til,en pyrrolidin-, piperidin-, perhydro-lH-azepin- eller morfolin-ring; karakterisert ved trinnene: (a) omsetning av oksazolet med formelmed et overskudd av et amin med formelH2NR5 eller HNR<2>R<3 >der R<2>, R<3> og R<5> er som definert ovenfor, i et vandig opp-løsningsmiddel, under dannelse av et 5-acetylimidazol med formelder R<4> er som definert ovenfor; (b) bromering av imidazolet med formel (II) i surt miljø og dannelse av et bromacetylimidazol med formelder R<4> er som definert ovenfor; og (c) kobling av bromacetylimidazolet med formel (III) med en forbindelse med formelog dannelse av forbindelsen med formel (I)•
