Description
Foreliggende oppfinnelse vedrører en analogifremgangsmåte
ved fremstilling av 5-(4-pyridyl)-6-(4-fluorfenyl)-2,3-dihydroimidazo(2,1-b)tiazol med formel I
eller et salt derav med sterk regulerende virkning på
cellebetinget immunitet og antiartritisk virkning.
De farmasøytisk anvendelig syreaddesjonssalter av forbindelsen
med formel I dannes med sterke eller middels sterke organiske eller uorganiske syrer på i og for seg kjent måte. Man ser tydelig at det er et basisk sentrum i imidazolringen og et annet i 4-pyridylsubstituenten.
F.eks. omsettes basen med en organisk eller uorganisk syre i et løsningsmiddel blandbart med vann, slik som etanol eller iso-
propanol under isolering av saltet ved at lsøningsmiddelet fjer-
nes eller i et med vann ublandbart løsningsmiddel når syren er oppløslig deri, så som etyleter eller kloroform, hvorved det øn-
skede salt utskilles direkte eller isoleres ved å fjerne løs-ningsmiddelet. Eksempler på saltene som omfattes av forbind-
elsen er maleat, fumarat, laktat, oksalat metansulfonat, etan-sulfonat, benzensulfonat, tartrat, citrat, hydroklorid, hydro-
bromid, sulfat, fosfat og nitrat. Som ovenfor nevnt har basen fysiske egenskaper som gjør dens anvendelse ekvivalent med an-vendelsen av salter derav.
Forbindelsen fremstilles ifølge foreliggende oppfinnelse ved
alkylering av 4-(4-pyridyl)-5-(4-fluorfenyl)-2-merkaptoimidazol med et etylen-1,2-halogenid som har reaktive halogenatomer så
som jod, klor eller brom, f.eks. l-brom-2-kloretan, 1,2-dibrom-
etan eller 1-jod-2-kloretan i et organisk løsningsmiddel hvor reaktantene er oppløslige. Ved anvendelse av sure løsnings-
midler skjer alkylerings-cykliseringen under dannelse av hydro-
halogenidsaltet av forbindelse med formel I. Andre polare løs-ningsmidler kan også anvendes, spesielt i nærvær av et alkali-metallreagens som er i stand til å danne alkalimetallderivatet av 2-merkaptangruppen i utgangsmaterialet så som natrium eller kaliumhydrider eller lavere alkoksyder. Løsningsmiddelet er dimetylformamid, dimetylacetamid, dimetylsulfoksyd, acetonitril, heksametylfosforamid.
Reaksjonen får løpe helt ut ved romtemperatur eller under mode-rat oppvarming slik som tilbakeløpstemperatur. Det dannede syreaddi<s>j<o>nssalt som er en blanding av isomere i 5,6 stil-ling, oppspaltes i de enkelte isomere på i og for seg kjent måte, f.eks. fraksjonert krystallisering eller kromatografi. Alternativt overføre saltet til den frie baseblanding som så adskilles til den enkelte isomere, den ønskede 5-(4-pyridyl)
-6-(4-fluorfenyl)-2,3-dihydroimidazo[2,1-b]tiazol.
En europeisk patentsøknad nr. 353 ble offentliggjort den 24. januar 1979 og beskriver en bred gruppe forbindelser beslektet med den her foreliggende. Den forbindelse hvis struktur ligger nærmest den foreliggende, og som er eksemplifisert i eksempel 6 i ovennevnte europeiske patenansøkning er blandingen av isomere 5(6)-(3-pyridyl)-6(5)-fenylimidazo[2,1-b]dihydrotiasol.
De viktigste strukturelle trekk ved forbindelsen som fremstilles ifølge foreliggende oppfinnelse, nemlig 4-fluorfenyl-gruppen, 4-pyridylgruppen og den riktige isomere struktur, er ikke beskrevet i den europeiske patentansøkningsbeskrivelse.
Den aktivitet som oppnås ved behandling av artritis bestemmes ved følgende metoder: Hemning av adjuvant indusert polyartritis hos rotter målt ved reduksjon av rottepoteødem, frembringes med forbindelsen ifølge oppfinnelsen i daglig dose fra ca. 3,125 opp til 50 mg/kg oralt med indometacin og metotreksat som kontroller. Ved denne for-søksmetode tilveiebringes adjuvant artritis hos rotter med en enkel intradermal injeksjon av 0,75 mg mykobakterium butyrikum oppslemmet i hvit parafinolje i venstre bakpotefotsåle. Den injiserte pote blir inflammert (øket volum) og når maksimal størrelse i løpet av 3 til 5 dager (primær lesjon). Dyrene har et fall i legemsvektøkning i den første perioden. Adjuvant artritis (sekundær lesjon) forekommer etter ca. 10 dager og er karakterisert ved inflammasjon av det ikke injiserte høyre bakben, fall i legemsvekt og ytterligere økning av volumet til det injiserte venstre bakben. Forsøksforbindelsen gis daglig fra og med dagen for adjuvantinjeksjon i 17 dager deretter eksklusive dagene 4,5,11 og 12. Antiartritisk aktivitet vises ved evnen til å beskytte dyrene mot utvikling av både primære og sekundære leksjoner av adjuvant artritis.
Ved denne prøve viste 5-(4-pyridyl)-6-(4-fluorfenyl)-2,3-dihy-droimidazo[2,1-b]tiazol positiv aktivitet ved 6,25, 12,5, 25
og 50 mg pr. kg oralt og gav en oral på 26,4 (15,6-54,4)
mg pr. kg pr. dag venstre benvolum på dag 3, 10,4 (5,3-19,0)
mg pr. kg dag venstre benvolum på dag 16 og 7,5 (2,7-15,6) mg pr. kg pr. dag høyre benvolum på dag 16. (ED25 refererer til den beregnede dosis som tilveiebringer en 25% reduksjon i forhold til kontrollverdiene.
Derimot hadde den omdannede isomer 6-(4-pyridyl)-5-(4-fluor-fenyl-2,3-dihydroimidazo[2,1-b]tiazol ingen aktivitet ved 50
mg pr. kg pr. dag. Det hadde heller ikke en nær beslekted ana-log 5,6-bis-(4-pyridyl)-2,3-dihydroimidazo[2,1-b]tiazol (50 mg /kg/dag).
Ved den karragen induserte rottepoteødemprøve tilveiebringes antiimflammatorisk virkning av den aktive forbindelse fremstilt ifølge oppfinnelsen i doser på 50 og 100 mg/kg oralt.
Dessuten er forbindelser som har immunoregulerende virkning anvendelige ved behandling av reumatoidartritis. Det har vist seg at den fremstilte forbindelse har evne til å regu-lere cellebetinget immunitet som påvist ved standardmetoder,
så som den oksazoloninduserte kontaktfølsomhetsprøve, ved hvilken musepote måles. Denne metode er beskrevet av Griswold m.fl., Cellular Immunology 11:198-204 (1974).
Ved oksazolonfølsomhetsprøven hadde 5-(4-pyridyl-6-(4-fluorfenyl)
-2,3-dihydroimidazo[2,1-b]tiazol signifikant virkning etter oral administrering.
Den fremstilte forbindelse hadde en oral LD^q på 214,2 mg/kg oralt hos mus over 14 dager.
I foreliggende tilfelle dreier det seg ikke om at en ren optisk isomer er bedre enn en annen, men at det er tale om to stillings-isomerer, nemlig, at den 5-(4-pyridyl)-6-(4-fluorfenyl)-substituerte forbindelse er bedre enn en annen stillings-isomer, nemlig den 6-(4-pyridyl)-5-(4-fluorfenyl)-substituerte forbindelse. Slike uventede forhold synes ikke å være tidligere kjent innenfor medisinsk kjemi.
Av de etterfølgende dato fremgår at den fremstilte forbindelse har et bredere aktivitetsspektrum enn standardfor-bindelsen på markedet, som er "Indomethocin". Det skal på-pekes at den fremstilte forbindelse har en betydelig aktivitet ved prøver som viser immuno-regulerende aktivitet, mens "Indomethocin" ikke har dette. Den fremstilte forbindelse som er angitt med SK&F nr. 86 002, har en betraktelig aktivitet i doser på 1 og 10 mg/kg, mens "Indomethocin ikke har noen virkning i doser på 2,5 og 10 mg/kg. 1 de etterfølgende tabeller er "Indomethocin" angitt med SK&F nr. 19056.
FORSØKSUTFØRELSE:
Kontaktsensitivitet fremkalles ved å påføre 0,1 ml 5% oksazolon (4-etoksymetylen-2-fenyloksazolon, Gallard-Schelessinger Chemical Manufacturing, Long Island, N.Y.) i absolutt alkohol på fastspent bakkropp av mus. Sensitiviser ing en får vare i.i 2 dager, hvoretter venstre bakben pensles med 5% oksazolon på en bomullsapplikator. Et anatomisk, referansepunkt marke-res på dyrenes baklemmer med Indiablekk for å lette målingen av leggvolumet. Den frembragte ødem måles plethysmografisk som beskrevet for Adjuvant- Induced Arthritis Test (Protocol Reference 0280), bortsett fra bruken av en mindre kvikksølv-kopp. Dataene er uttrykt i ødem (forskjell i volum mellom oksazolon-angrepet venstre bakben og det ubehandlede høyre bakben).
DEFINISJON AV SIGNIFIKANT AKTIVITET;
En statistisk signifikant aktivitet bestemmes ved å bruke studentenes "t" test. Resultatene er sammenfattet i den følgende tabell.
OKSAZOLON- INDUSERT KONTAKTSENSITIVITET
Forsøksresultater:
Foruten å være anvendelig ved reumatoid artritis kan immuno-regulerende midler anvendes ved andre sykdommer hvor cellebetinget immunitet bringes i fare. Eksempler på slike sykdommer er systemisk lupus erytematosus og autoimmun tyroiditis. Også sykdommer så som atopisk dermatitis, periodisk aptus ul-cerasjon, periodisk infeksjon av de øvre åndedrett organer hos barn og influensa, lunge og brystkreft, forbigående granulocylo-penia og allergiske hudreaksjoner er med godt resultat blitt behandlet med levamisol, som er et middel, som gjenoppretter skadet cellebetinget immunreaksjon.
De farmasøytisk virksomme forbindelser fremstilt ifølge oppfinnelsen gis
i vanlig enhetsdoseringsformer som fremstilles ved blande forbindelsen med formel I i en tilstrekkelig mengde til å gi antiartrisk virkning eller regulering av cellebetinget immunitet med standard farmasøytiske bærere på vanlige måter.
Fortrinnsvis inneholder hver enhetsdose den aktive bestanddel
i en mengde fra ca. 25 mg til ca. 100 mg.

EKSEMPEL 1
En vandig oppløsning (75 ml) av natriumcyanid (19,6 g, 400 mm)
og bensyltrietylammoniumklorid (3,0 g, 13 mm) ble tilsatt isonikotinaldehyd (10 g, 93 mm) i 100 ml metylenklorid ved 0°c. Man rørte kraftig i 15 minutter og tilsatte langsomt en metylen-kloridoppløsning av benzoylklorid (14,0 g, 100 mm, 50 ml). Kjøling ble avbrutt etter en halv times reaksjon og blandingen fikk nå stuetemperatur. Det organiske skjiktet ble skilt fra og vasket med 5 % natriumkarbonat og saltvann, tørket og inndampet til en olje, som ble grundig ekstrahert med eter (4 liter). Eteroppløsningen ble konsentrert til ca. 500 ml og fikk krys-tallisere hvilket ga 5,0 g isonikotinaldehyd-O-benzoylcyanhydrin Cyanhydrinet (3,0 g, 12 mm) ble rørt i 50 ml tertiær butylalko-hol med p-fluorbenzaldehyd (12 mm) og natriumhydrid (12 mm) ble tilsatt. Omrøring ble fortsatt i 1 1/2 time ved romtemperatur. En mineraloljesuspensjon av kaliumhydrid (24 % suspensjon, 4
ml, 23 mmol) ble tilsatt forsiktig. En tynnskjiktkromatografisk undersøkelse av en del av reaksjonsblandingen etter 1 1/2 time viste dannelse av et enkelt produkt. Dette ble opparbeidet ved at suspensjonen ble helt i 200 ml is/vann blanding og deretter ekstrahert med kloroform. Kloroformekstraktene ble inndampet til en krystallinsk rest som ytterligere ble krystallisert ved tilsetning av eter. Man fikk et krystallinsk produkt som eventuelt kunne renses ved krystallisering fra blandinger av metylenklorid og eter. Dette var ikke nødvendig for å oppnå til-fredsstillende utbytter.
9,0 g (37 mmol) av hydroksyketonene ble oppvarmet under til-bakeløpskoking i 14 0 ml dimetylformamid med 5,3 g (70 mmol) tiourea. Utgangsmaterialet var ikke lenger påviselig etter 4 timers reaksjon. Oppløsningsmiddelet ble konsentrert til halvparten av det opprinnelige volum for å starte utfelling av det krystallinske 2-merkaptoimidazol. Krystalliseringen ble fullført natten over i kjoleskap. Den erholdte forbindelse ble renset ved krystallisering fraetanol.
4-(4-pyridyl)-5-(4-fluorfenyl)-2-merkaptoimidazol erholdtes i et samlet utbytte på 40 % råprodukt beregnet på benzoylcyanhy-drinkondensasjon, smeltepunkt 386 - 388°C.

2-merkaptoimidazolet (12,5 mm) ble oppslemmet i 100 ml dimetylformamid og natriumhydrid (13,0 mm) ble tilsatt- Saltdannelsen fikk forgå ved stuetemperatur i 1/2 time, hvorpå man tilsatte l-brom-2-kloretan fra en hypodermisk sprøyte og oppløsningen ble rørt natten over under argonatmosfære.
Nesten vannfritt kaliumkarbonat (20,0 mm) ble satt til reaksjonsblandingen og tilbakeløpskoking ble utført i 2 1/2 time. Fortynning av dimetylformamidoppløsningen med en blanding av
is og vann til 300 ml forårsaket utfelling av et oljeaktig produkt.
Den oljeaktige rest ble kromatografert over kiselgel under anvendelse av en tørr søyleteknikk med 1:1 etylacetat/eter. Dette skilte den isomere blanding fra alle forurensninger. Isomerene ble så separert ved middeltrykks væskekromatografi på kiselgel med isopropanol/eter (1:2). Den hurtiggående flekk (Rf:0,55 kiselgel)isopropanol) etter eluering ble ytterligere renset ved krystallisering fra isopropanol, smeltepunkt 165 - 167°C,utbytte 0,75 g 5-(4-fluorfenyl)-6-(4-pyridyl)-isomeren.

Den langsomt gående flekk, den foretrukne 5-(4-pyridyl)-6-(4-fluorfenyl)isomer (R^: 0,25 kiselgel/isopropanol) etter eluering ble omkrystallisert fra isopropanol, smeltepunkt 186 til 189°C,0,62 g.

1,9 g 6-(4-fluorfenyl)-5-(4-pyridyl)-2,3-dihydroimidazo=[2,1-b] tiazol i 25 ml metylenklorid ved isbadtemperatur ble tilsatt 1,36 g m-klorperbenzosyre. Omrøring ble fortsatt i 1 time, og deretter ble oppløsningen fortynnet med metylenklorid og vasket med 1:1 blanding av 5 % natriumkarbonat og saltvann. Behandling med magnesiumsulfat, filtrering og avdestillering ga 2,1 g 6-4-fluorfenyl)-5-(4-pyridyl)-2,3-dihydroimidazo[2,1-b]tiazol-1 -oksyd som ble revet med eter, filtrert og tørket under dannelse av 1,8 g produkt, smeltepunkt 229 til 231°C.

Behandling av utgangsmaterialet med 2,72 g m-klorperbenzosyre gir 1-dioksydderivatet.
EKSEMPEL 3
50 ml iseddik ble tilsatt 1,0 g 4-(4-pyridyl)-5-(4-fluorfenyl) -2-merkaptoimidazol og 0,7 g dibrometan. Den melkeaktige opp-løsning ble klar etter tilbakeløpskoking i 1 time. På dette tidspunkt ble ytterligere 2,1 g dibrometan tilsatt og tilbake-løpskokingen opprettholdt i ytterligere 3 timer. Eddiksyren ble avdestillert i våkum. Resten ble fordelt mellom metylenklorid og 5N natriumkarbonatoppløsning. Det organiske eks-traktet ble vasket med 5N natriumkarbonatoppløsning og saltvann og deretter behandlet med magnesiumsulfat. Avdamping av løsningsmiddelet levnet en blanding av 6-(4-fluorfenyl)-5-(4-pyridyl)-2,3-dihydroimidazo[2,1-b]tiazol og 5-(4-fluorfenyl) -6-(4-pydidyl)-2,3-dihydro-imidazo[2,1-b]tiazol som ble separert som ovenfor beskrevet.
Claims (1)
- Analogifremgangsmåte ved fremstilling av 5-(4-pyridyl)-6- (4-£luorfenyl)-2,3-dihydroimidazo(2,1-b)tiazol med formel eller et salt derav med sterk regulerende virkning på cellebetinget immunitet og antiartritisk virkning,karakterisert ved at man omsetter 4-(4-pyridyl)-5-(4-fluorfenyl)-2-merkaptoimidazol med 1,2-di-halogenetan hvor halogen er klor, brom eller jod, hvilket gir en blanding av 5-(4-pyridyl)-6-(4-fluorfenyl)-2,3-dihydroimidazo[2,1-b]tiazol og 5-(4-fluorfenyl)-6-(4-pyridyl)-2,3-dihydroimidazo[2,1-b]tiazol, fra hvilken blanding den ønskede forbindelse isoleres ved fraksjonert krystallisering eller kromatografi, eventuelt etterfulgt av overføring i et syreaddisjonssalt.
