Abstract

La invencion proporciona enzimas ADN polimerasa que comprenden la secuencia aminoácido SerGlnIleXaaLeuArgXaa (SEQ ID NO: 1), en donde "Xaa" en posicion 4 de esta secuencia es cualquier residuo aminoácido pero no un residuo ácido glutámico (Glu), preferiblemente un residuo glicina y "Xaa" en posicion 7 de esta secuencia es un residuo valina (Val) o un residuo isoleucina (Ile). Las ADN polimerasas termoestables de la invencion tienen la eficiencia aumentada para incorporar nucleotidos no convencionales, tales como ribonucléotidos, en productos de ADN y son ventajosos en muchas aplicaciones de síntesis in vitro. Tales enzimas son particularmente utiles para usar en protocolos de secuenciacion de ácido nucleico y proporcionar nuevos medios para el análisis de la secuencia de ADN con ventajas de costo y eficiencia.
