본 발명은 신규화합물과 그 용도에 관한 것이다.
2. 발명이 해결하려고 하는 기술적과제
본 발명은 적색파장을 사용해서 망상적혈구를 정밀도가 양호하게 측정할 수 있는 신규의 화합물과 그 용도인 망상적혈구측정용의 시약 및 그 측정방법을 제공하는 것을 목적으로 한다.
3. 발명의 해결방법의 요지
식(Ⅰ)
(식중 R1은 수소원자, 또는 저급알킬기; R2및 R3은 수소원자 저급알킬기, 또는 저급알콕시기; R4는 수소원자, 아실기, 또는 저급알킬기; R5는 수소원자, 치환되어 있어도 되는 저급알킬기; Z는 유황원자, 산소원자, 또는 저급알킬기로 치환된 탄소원자; n은 1 또는 2; X-는 음이온이다)로 표시되는 화합물.
4. 발명의 중요한 용도
본 발명은 임상검사분야에 있어서의 망상적혈구 및 망상적혈구 성숙지수측정용을 위한 형광색소로서 사용가능한 신규화합물, 그 화합물을 포함하는 시약 및 망상적혈구의 측정방법에 관한 것이다.

제1도는 본 발명의 색소화합물A를 포함하는 망상적혈구측정용시약을 사용해서 혈액학적시료의 전방산란광강도 및 형광강도를 측정한 경우의 산포도,
제2도는 본 발명의 색소화합물A를 포함하는 망상적혈구측정용시약을 다시또 양이온성계면활성제를 포함하는 시약을 사용해서 혈액학적시료의 전방산란광강도 및 형광강도를 측정한 경우의 산포도,
제3도는 본 발명의 색소를 포함하는 망상적혈구측정용시약을 사용해서 혈액학적시료의 전방산란광강도 및 형광강도를 측정한 경우의 산포도,
제4도는 본 발명의 색소를 포함하는 망상적혈구측정용시약을 사용해서 혈액학적시료의 전방산란광강도 및 형광강도를 측정한 경우의 산포도,
제5도는 본 발명의 색소를 포함하는 망상적혈구측정용시약을 사용해서 본 발명의 방법 및 종래의 R-2000을 사용한 방법에서 RET를 측정한 경우의 상관도.

Claims (14)

  1. 식(I)
    (식중 R1은 수소원자, 또는 저급알킬기; R2및 R3은 수소원자 저급알킬기, 또는 저급알콕시기; R4는 수소원자, 아실기, 또는 저급알킬기; R5는 수소원자, 치환되어 있어도 되는 저급알킬기; Z는 유황원자, 산소원자, 또는 저급알킬기로 치환된 탄소원자; n은 1 또는 2; X-는 음이온이다)로 표시되는 것을 특징으로 하는 화합물.
  2. 제1항에 있어서, 식(I)의 화합물에 있어서, R2및 R3는 수소원자; R4는 수소원자, 아실기, 또는 저급알킬기; R5는 수소원자, 치환되어 있어도 되는 저급알킬기인 것을 특징으로 하는 화합물.
  3. 제1항에 있어서, 식 (I)의 화합물에 있어서, R1은 메틸기; R2및 R3은 수소원자; R4및 R5는 수소원자; n=1; Z는 유황원자, R1은 메틸기; R2및 R3은 수소원자; R4는 수소원자; R5는 히드록시메틸기; n=1; Z는 유황원자, R1은 메틸기; R2및 R3은 수소원자; R4는 COCH3기; R5는 수소원자; n=1; Z는 유황원자, R1은 메틸기; R2및 R3은 수소원자; R4및 R5는 수소원자; n=2; Z는 유황원자인 것을 특징으로 하는 화합물.
  4. 식(I)
    (식중 R1은 수소원자 또는 저급알킬기; R2및 R3은 수소원자 저급알킬기, 또는 저급알콕시기; R4는 수소원자, 아실기 또는 저급알킬기; R5는 수소원자, 치환되어 있어도 되는 저급알킬기; Z는 유황원자, 산소원자, 또는 저급알킬기로 치환된 탄소원자; n은 1 또는 2; X-는 음이온이다)로 표시되는 화합물을 포함하는 것을 특징으로 하는 망상적혈구염색용시약.
  5. 제4항에 있어서, 적혈구의 비특이염색을 억제하기 위한 다가음이온과 완충제와를 포함하는 것을 특징으로 하는 망상적혈구염색용시약.
  6. 제5항에 있어서, 다가음이온이 황산이온, 인산이온, 다가카르복시산이온으로부터 선택된 적어도 1종인 것을 특징으로 하는 망상적헐구염색용시약.
  7. 제4항 내지 제6항중의 어느 한항에 있어서, pH가 6.0∼11.0인 것을 특징으로 하는 망상적혈구염색용시약.
  8. 제4항 내지 제7항중의 어느 한항에 있어서, 염색촉진제로서 양이온성계면활성제를 포함하는 것을 특징으로 하는 망상적혈구염색용시약.
  9. 양이온성계면활성제가
    (식중 R6은 탄소수 8∼12의 알킬기, R7, R8, 및 R9는 저급알킬기, X는 할로겐으로부터 선택된 적어도 하나이다)인 것을 특징으로 하는 망상적혈구염색용시약.
  10. 제8항에 있어서, 양이온성계면활성제가 라우릴트리메틸암모늄클로라이드, 데실트리메틸암모늄브로마이드, 옥틸트리메틸암모늄브로마이드의 적어도 1종인 것을 특징으로 하는 망상적혈구염색용시약.
  11. 식(I)
    (식중 R1은 수소원자, 또는 저급알킬기; R2및 R3은 수소원자 저급알킬기, 또는 저급알콕시기; R4는 수소원자, 아실기, 또는 저급알킬기; R5는 수소원자, 치환되어 있어도 되는 저급알킬기; Z는 유황원자, 산소원자, 또는 저급알킬기로 치환된 탄소원자; n은 1 또는 2; X-는 음이온이다)로 표시되는 화합물을 포함하는 시약을 사용하는 것을 특징으로 하는 망상적혈구측정방법.
  12. 제11항에 있어서, (i) 식 (I)로 표시되는 화합물과 적혈구의 비특이염색을 억제하기 위한 다가음이온과 완충제로 된 망상적혈구염색용시약과 혈액학적시료와를 혼합하므로서 측정시료를 조제하고 (ⅱ) 상기한 측정시료를 적색파장의 광원을 갖는 플로우사이트미터의 유체계에 도입하고 (ⅲ) 시즈플로우중을 흐르는 혈액학적시료의 세포에 적색파장의 광을 조사하고 (ⅳ) 세포로부터 발하는 산란광과 형광을 측정하고 (ⅴ) 산란광강도 혹은 산란광강도와 형광강도에 의해 소혈판과 적혈구계세포와를 판별하고 (ⅵ) 형광강도 혹은 형광강도와 산란광강도에 의해 적혈구, 망상적혈구 및 백혈구를 판별하고 (ⅶ) 혈소판, 적혈구, 망상적혈구 및 백혈구를 분류계수해서 세포수, 및 세포비율을 산출하는 것을 특징으로 하는 망상적혈구측정방법.
  13. 제12항에 있어서, 적색파장의 광원이 He-Ne 레이저 또는 적색반도체레이저인 것을 특징으로 하는 망상적혈구측정방법.
  14. 제12항에 있어서, 적어도 하나의 특정범위의 형광강도를 갖는 망상적혈구의 전체망상적혈구가 점하는 비율을 구하는 것으로 이루어진 것을 특징으로 하는 망상적혈구의 측정방법.
