Abstract

본 발명은 인간 EGF-수용체 분자에 특이적인 단클로항체 또는 이의 단면과 케모킨족 부류, 바람직하게는 C-X-C족, 예컨데 인터루이킨-8(IL-8)중에서 선택되는 케모킨류로 이루어지는 신규한 면역접합체를 제공한다. 본 면역접합체는 세포독성 및 주화성 활성을 유도하며 표적종양 치료에 적합하다.

Claims (14)

  1. 표피 성장 인자 수용체(EGFR)의 항원 결정기를 함유하고 있는 종양세포에 대해 지향성을 갖는 단클론성 항체 또는 이의 단편 : 및 상기 항체 또는 항체 단편에 융합된 케모킨 단백질을 포함하여 이루어지는 것을 특징으로 하는 면역접합체
  2. 제1항에 있어서, 상기한 케모킨이 C-X-C족인 것을 특징으로 하는 면역접합체
  3. 제2항에 있어서, 상기한 케모킨이 IL-8인 것을 특징으로 하는 면역접합체
  4. 제1항 내지 제3항 중의 어느 한 항에 있어서, 상기한 항체가 항체중쇄의 가변부, 불변부의 CH1 구역 및 적절한 경쇄(항체-CH1 접합체)로 구성되는 Fab 단편 및 F(ab')2 단편인 것을 특징으로 하는 면역접합체
  5. 제1항 내지 제3항 중의 어느 한 항에 있어서, 상기한 항체가 항체중쇄의 가변부, 불변부의 CH1 및 CH2 구역 및 적절한 경쇄(항체-CH2 접합체)로 구성되는 항체단편인 것을 특징으로 하는 면역접합체
  6. 제1항 내지 제3항 중의 어느 한 항에 있어서, 상기한 항체가 항체중쇄의 가변부, 불변부의 CH1, CH2 및 CH3 구역, 및 적절한 경쇄(항체-CH3 접합체)로 구성되는 항체단편인 것을 특징으로 하는 면역접합체
  7. 제1항 내지 제3항 중의 어느 한 항에 있어서, 상기한 항체가 항체중쇄의 가변부, 적절한 경쇄 및 중쇄를 연결하는 폴리펩티드 서열(항체-Fv 접합체)로 구성되는 것임을 특징으로 하는 면역접합체
  8. 제1항 내지 제6항 중 어느 한 항에 있어서, 상기한 면역접합체가 항체/항체 단편과 생물학적 활성 리간드 사이에서 완전 융합 형성물내에서 유일하게 존재하는, DNA 제한부위에 대해 연역 가능한 아미노산(서열)을 포함하여 이루어지고, 상기 제한 부위가 완전 융합 형성물내에서 유일하게 존재하는 것을 특징으로 하는 면역접합체
  9. 제1항 내지 제7항 중 어느 한 항에 있어서, 상기한 면역접합체가 항체/항체 단편과 생물학적 활성 리간드 사이의 링커 펩티드를 포함하여 이루어지는 것을 특징으로 하는 면역접합체
  10. 제1항 내지 제3항 중의 어느 한 항에 있어서, 상기한 항체 또는 항체단편이 쥐, 인간 또는 키메라성 MAb 425로부터 유도되는 것임을 특징으로 하는 면역접합체
  11. MAb 425-CH1-IL8, MAb 425-CH2-(Ncol)-IL-8, MAb 425-CH2-(Bcll)-IL-8, MAb 25-Fv-IL-8, MAb 425-CH3-IL-8 중에서 선택되는 것임을 특징으로 하는 면역접합체
  12. 제1항 내지 제10항 중의 한 항에 기재된 면역접합체를 제조하는 방법에 있어서, 목적하는 융합 DNA-서열에 상보적인 올리고 뉴클레오티드를 사용하여 항체 또는 항체단편 및 생물학적 활성 리간드 각각을 암호화하는 DNA서열을 단일가닥 DNA상에 상호 융합시키고; 생성된 형성물을 발현 벡터에 위치시킨 후, 상기 숙주세포로 도입하여 형질전환시키고; 상기 숙주세포를 배양액중에서 배양하여 융합 단백질을 발현시키는 것으로 이루어지는 면역접합체의 제조방법
  13. 제1항 내지 제10항 중의 어느 한 항에 따른 면역접합체 1종 이상과, 생리학적으로 허용되는 담체를 포함하여 이루어지는 것을 특징으로 하는 약제학적 조성물
  14. 종양 지향성 약물을 제조하기 위하여 제1항 내지 제10항 중의 어느 한 항에 기재된 면역접합체를 사용하는 방법
