Abstract
1. 청구범위에 기재된 발명이 속한 기술분야
본 발명은 백혈구 분류방법에 관한 것이다.
2. 발명이 해결하려고 하는 기술적 과제
본 발명은 값싸고 간단한 구성의 장치로 백혈구를 적어도 5분류하는 방법을 제공하는 것을 목적으로 하고 있다.
3. 발명의 해결방법의 요지
(ⅰ), (a)적혈구를 용해시킬 수 있으나 백혈구의 세포막의 일부에 손상을 주는데 충분한 양의 적어도 1종류의 이온성계면활성제와, (b)백혈구내의 양이온 하전성분과 결합해서 백혈구간에 형태적 차이를 부여하는데 충분한 양의 적어도 1종류의 음이온기를 갖는 유기화합물과, (c)비이온계면활성제와, (d)pH조정용의 완충제로된 백혈구 4분류용 시약을 전체혈액시료의 일부에 첨가하고, 세포의 크기정보와 형태정보를 측정해서 백혈구를 적어도 림프구, 단구, 및 호산구에 각각 대응하는 3집단과 호중구 및 호염기구에 대응하는 1집단과의 4집단으로 분류계수하고, (ⅱ)호염기구 측정용 시약을 전체혈액시료의 다른 일부에 첨가하여 적어도 세포의 크기정보를 측정해서 호염기구를 분류계수하고, (ⅲ)각각 얻어진 계수치에 기초해서 백혈구를 적어도 5분류하는 것을 특징으로 하는 백혈구 분류방법.
4. 발명의 중요한 용도
본 발명은 다른 시약에 의해 처리한 백혈구를 따로따로 측정하므로서 백혈구를 5분류하는 백혈구 분류방법에 관한 것이다.

Claims (9)
- (ⅰ)(a)적혈구를 용해시킬 수 있으나 백혈구의 세포막의 일부에 손상을 주는데 충분한 양의 적어도 1종류의 이온성계면활성제와, (b)백혈구내의 양이온 하전성분과 결합해서 백혈구간에 헝태적 차이를 부여하는데 충분한 양의 적어도 1종류의 음이온기를 갖는 유기화합물과, (c)비이온계면활성제와, (d)pH조정용의 완충제로된 백혈구 4분류용 시약을 전체혈액시료의 일부에 첨가하고, 세포의 크기정보와 형태정보를 측정해서 백혈구를 적어도 림프구(Lym), 단구(Mono), 및 호산구(Eo)에 각각 대응하는 3집단과 호중구 및 호염기구(GRAN)에 대응하는 1집단과의 4집단으로 분류계수하고, (ⅱ)호염기구 측정용 시약을 전체혈액시료의 다른 일부에 첨가하여 적어도 세포의 크기정보를 측정해서 호염기구(Baso)를 분류계수하고, (ⅲ)각각 얻어진 계수치에 기초해서 백혈구를 적어도 5분류하는 것을 특징으르 하는 백혈구 분류방법.
- 제1항에 있어서, 세포의 크기정보를 측정하는 공정이 저각산란광을 측정하는 것을 특징으로 하는 백혈구 분류방법.
- 제2항에 있어서, 저각산란광의 측정각도가 1∼5°인 것을 특징으로 하는 백혈구 분류방법.
- 제1항에 있어서, 형태정보를 측정하는 공정이 광각산란광을 측정하는 것을 특징으로 하는 백혈구 분류방법.
- 제4항에 있어서, 광각산란광의 측정각도가 6-20°인 것을 특징으로 하는 백혈구 분류방법.
- 제1항에 있어서, 세포의 크기정보와 형태정보를 측정하는 공정이 저각과 광각의 2가지의 산란광을 측정하는 것을 특징으로 하는 백혈구 분류방법.
- 제6항에 있어서, 측정하는 저각과 광각의 2가지의 산란광의 각도가 1∼5°및 6∼20°인 것을 특징으로 하는 백혈구 분류방법.
- 제6항 또는 제7항에 있어서, 2가지의 각도의 산란광의 검출을 1개의 포트다이오드(PD)센서로 행하는 것을 특징으로 히는 백현구 분류방법.
- 제1항에 있어서, 공정(ⅰ)의 적어도 4집단의 분류계수와 공정(ⅱ)의 호염기구(Baso)의 분류계수를 동일한 검출기를 사용해서 행하는 것을 특징으로 하는 백혈구 분류방법.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
