Abstract
본 발명은 샘플내 증폭된 핵산 피분석물을 검출하기 위한 분석에서 허위음성 결과를 제거하는 방법에 관한 것이다. 본 방법에서는 증폭시키기 전에 내부 대조군을 샘플에 첨가하는데, 이 내부 대조군은 핵산 피분석물과 동일한 증폭 시약으로 증폭될 수 있고 핵산 피분석물과 구별될 수 있는 핵산을 포함한다. 내부 대조군은 핵산 피분석물과 구별되도록 돌연변이된, 핵산 피분석물에 상응하는 핵산 서열을 포함하는 것이 바람직하다.

Claims (8)
- 샘플내 증폭된 핵산 피분석물을 검출하기 위한 분석에서 허위 음성(false negative)결과를 제거하는 방법으로서, 증폭시키기 전에, 핵산 피분석물과 동일한 증폭 시약으로 증폭될 수 있고 핵산 피분석물과 구별될 수 있는 핵산을 포함하는 내부 대조군(internal control)을 첨부하는 것을 특징으로 하는 방법.
- 제1항에 있어서, 내부 대조군이 핵산 피분석물과 구별되도록 돌연변이된, 핵산 피분석물에 상응하는 핵산 서열을 포함하는 것을 특징으로 하는 방법.
- 제1항 또는 제2항에 있어서, 내부 대조군이 핵산 피분석물보다 더 적은 효율로 증폭되는 것을 특징으로 하는 방법.
- 제1항 내지 제3항중 어느 한 항에 있어서, 내부 대조군 및 핵산 피분석물이 둘다 하나의 올리고뉴클레오티드 및 동일한 상보적 올리고뉴클레오티드와 검출 가능한 복합체를 형성할 수 있는 것을 특징으로 하는 방법.
- 제4항에 있어서, 핵산 피분석물 및 내부 대조군이 길이가 상이하며, 둘다 동일한 상보적 올리고뉴클레오티드와 검출 가능한 복합체를 형성할 수 있는 것을 특징으로 하는 방법.
- 제4항에 있어서, 상보적 올리고뉴클레오티드가 고체상상에 고정화되며, 검출 가능한 복합체가 각각 상이하게 표지한 반응물과 반응할 수 있는 것을 특징으로 하는 방법.
- 제1항 내지 제3항중 어느 한 항에 있어서, 핵산 피분석물 및 내부 대조군 핵산이 각각 상이한 상보적 올리고 뉴클레오티드와 검출가능한 복합체를 형성할 수 있는 것을 특징으로 하는 방법.
- 제7항에 있어서, 상기 올리고뉴클레오티드가 각각 고체상상의 상이한 지점에 고정화된 것을 특징으로 하는 방법.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
