Abstract
본 발명은 표지에 대한 수용체에 의하여 비혼성화된 유전자 프로브의 신호를 변환시킴을 기초로 하는 동종 유전자 프로브 테스트에 관한 것이다.

본 내용은 요부공개 건이므로 전문내용을 수록하지 않았음
제1도는 선행 기술에 따른 유전자 프로브 분석의 일반원리를 나타낸다,
제2도는 아크리디늄 아실설폰아미드를 나타낸다,
제3도는 적합한 유전자 프로브의 구조를 나타낸다,
제4도는 본 발명에 따른 바람직한 변형을 도식적으로 나타내고 있다,
제56도는 일본쇄 유전자 프로브의 표지를 2회 제거하여 측정하는 방법을 나타내고 있다.
Claims (7)
- (a) 표적 서열과 혼성화되지 않은 임의의 유전자 프로브 잉여부분의 표지와 결합하는 수용체를 첨가하여 표지로 인한 신호를 정성적 및/또는 정량적으로 변환시키는 단계 및 (b) 상기 목적에 적합한 방법을 사용하여 표지로 인한 신호를 정성적으로 또는 정량적으로 탐지하는 단계를 추가로 포함하는, 임의로 표적 서열을 포함하는 샘플을, 표적 서열과 유전자 프로브가 서로 혼성화되도록 상기 표적 서열의 결정에 적합한 유전자 프로브와 접촉시키는 표적서열의 결정 방법.
- 제1항에 있어서, 추가의 단계에서, 임의의 비혼성화된 잉여의 유전자 프로브의 표지를 선택 시약과 함께 반응시킴으로서 부분적으로 불활성화시키는 방법.
- 제2항에 있어서, 처음에 선택 시약과 반응시키고 이어서 수용체를 첨가하는 방법.
- 제1항 내지 제3항 중 어느 한 항에 있어서, 화학적 변형후에도 항원 결합 능력이 어느 정도 유지되는 경우에, 수용체가 모노클로날 또는 폴리클로날 항체, 항체의 단편, 화학적으로 변형된 항체 또는 화학적으로 변형된 항체의 단편인 방법.
- 제1항 내지 제4항 중 어느 한 항에 있어서, 표지가 형광, 인광, 화학발광, 생체발광, 생체발광 또는 전자발광할 수 있는 그룹인 방법.
- 제5항에 있어서, 표지가 아크리디늄 에스테르, 아크리디늄 아실설폰아미드, 루미놀, 이소루미놀 또는 이의 유도체, 디옥스에탄, 루시페린, 옥살산 에스테르 또는 옥스아미드인 방법.
- 제1항 내지 제4항 중 어느 한 항에 있어서, 표지가 효소인 방법.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
