Abstract

Description
본 발명은 암치료의 분야에 관한 것이다.
최근, 진행암(advanced cancer)을 대상으로 한 증식형 바이러스(replicative virus)에 의한 바이러스 요법(virotherapy)의 임상연구가 진행되고 있다. 바이러스 요법이란, HSV-1이나 아데노바이러스(adenovirus) 등의 증식형 바이러스를 종양세포에 감염시켜, 바이러스 증식에 수반되는 바이러스의 살세포 효과(cytocidal effect)에 의해 종양의 치유를 도모하는 치료법이다. HSV-1이나 아데노바이러스의 경우, 종양 치료용 증식형 바이러스는, 바이러스 게놈(viral genome)에 유전자 조작을 가한 변이 바이러스로, 종양 내에서의 복제능을 보유하면서, 인간 정상조직에서의 병원성이 최소한으로 억제되어 있다. 종양세포에 감염된 치료용 증식형 바이러스는, 세포 내에서 복제되고, 그 과정에서 감염세포는 사멸한다. 증식된 바이러스는 주위의 종양세포에 재감염되고, 항종양작용을 확산시킨다(Alemany R. et al., Replicative adenoviruses for cancer therapy. 발명의 개시
발명이 해결하고자 하는 과제
본 발명은, 암에 대한 보다 효과적인 바이러스 요법, 및 항암제의 제공을 과제로 한다. 보다 상세하게는 본 발명은, 마이너스 가닥 RNA 바이러스(minus-strand RNA virus)를 포함하는 항암제의 제조방법, 및 마이너스 가닥 RNA 바이러스를 사용한 암의 치료방법을 제공한다.
과제를 해결하기 위한 수단
본 발명자 등은, RNA 바이러스의 인 비보 투여(in vivo administration)에 의한 종양에 대한 치료효과를 검증하기 위해, 흑색종 세포주(melanoma cell line)를 복측 피하(ventral subcutaneous)로 접종한 마우스를 사용하여 실험을 행하였다. 그 결과, 치료 유전자를 갖지 않는 마이너스 가닥 RNA 바이러스를, 상기 동물의 종양 내로 주사함으로써, 종양 사이즈가 유의하게(significantly) 축소되는 것을 발견하였다. 즉, 마이너스 가닥 RNA 바이러스의 인 비보 투여는, 상기 바이러스가 치료 유전자를 갖지 않는 경우일지라도, 항종양 효과를 발휘하는 것을 알 수 있었다.
마이너스 가닥 RNA 바이러스는, 그 자체가 유효한 항암제가 되는 것으로 생각되어진다. 또한, 상기 바이러스를 직접, 종양부위(암조직)로 주입함으로써, 유효한 항종양 효과를 기대할 수 있어, 상기 바이러스는, 암의 치료에 유용하다.

발명을 실시하기
위한 최선의 형태
본 발명은, 유효성분으로서 마이너스 가닥 RNA 바이러스를 포함하는 항암제(제암제(carcinostatic agent))를 제공한다. 본 발명에 있어서 RNA 바이러스란, RNA 게놈을 갖는 바이러스를 말한다.
본 발명의 항암제의 유효성분으로 하는 RNA 바이러스는, 바람직하게는, 마이너스 가닥 RNA 바이러스이다. 마이너스 가닥 RNA 바이러스란, 마이너스 가닥(바이러스 단백직을 코드하는 센스 가닥(sense strand)에 대한 안티센스 가닥(antisense strand)의 RNA를 게놈으로서 포함하는 바이러스이다. 마이너스 가닥 RNA는 음성 가닥 RNA(negative strand RNA)라고도 불리운다. 본 발명에 있어서 사용되는 마이너스 가닥 RNA 바이러스로서는, 특히 단일 가닥 마이너스 가닥 RNA 바이러스(single-stranded minus-strand RNA virus)(비분절형(non-segmented) 마이너스 가닥 RNA 바이러스라고도 한다)를 들 수 있다. 「단일 가닥 음성 가닥 RNA 바이러스」란, 단일 가닥 음성 가닥[즉, 마이너스 가닥] RNA를 게놈에 갖는 바이러스를 말한다. 이와 같은 바이러스로서는, 파라믹소바이러스(Paramyxoviridae; Paramyxovirus, Morbillivirus, Rubulavirus, 및 Pneumovirus속 등을 포함한다), 라브도바이러스(Rhabdoviridae; Vesiculovirus, Lyssavirus, 및 Ephemerovirus속 등을 포함한다), 필로바이러스(Filoviridae), 오르토믹소바이러스(Orthomyxoviridae; Infuluenza virus A, B, C, 및 Thogoto-like viruses 등을 포함한다), 분야바이러스(Bunyaviridae; Bunyavirus, Hantavirus, Nairovirus, 및 Phlebovirus속 등을 포함한다), 아레나바이러스(Arenaviridae) 등의 과(科)에 속하는 바이러스가 포함된다.
본 발명에 있어서 적합하게 사용되는 마이너스 가닥 RNA 바이러스로서는, 예를 들면 파라믹소바이러스과(Paramyxoviridae) 바이러스의 센다이 바이러스(Sendai virus)를 들 수 있다. 그 밖의 예로서는, 뉴캐슬병 바이러스(Newcastle disease virus), 유행성 이하선염 바이러스(Mumps virus), 홍역 바이러스(Measles virus), RS 바이러스(Respiratory syncytial virus), 우역 바이러스(rinderpest virus), 디스템퍼 바이러스(distemper virus), 원숭이 파라인플루엔자 바이러스(simian parainfluenza virus)(SV5), 인간 파라인플루엔자 바이러스 1, 2, 3형(human parainfluenza virus 1, 2 and 3), 오르토믹소바이러스과(Orthomyxoviridae)의 인플루엔자 바이러스(Influenza virus), 라브도바이러스과(Rhabdoviridae)의 수포성 구내염 바이러스(Vesicular stomatitis virus), 광견병 바이러스(Rabies virus) 등을 들 수 있다.
본 발명에 있어서 사용할 수 있는 바이러스를 추가로 예시하면, 예를 들면, Sendai virus(SeV), human parainfluenza virus-1(HPIV-1), human parainfluenza virus-3(HPIV-3), phocine distemper virus(PDV), canine distemper virus(CDV), dolphin molbillivirus(DMV), peste-des-petits-ruminants virus(PDPR), measles virus(MV), rinderpest virus(RPV), Hendra virus(Hendra), Nipah virus(Nipah), human parainfluenza virus-2(HPIV-2), simian parainfluenza virus 5(SV5), human parainfluenza virus-4a(HPIV-4a), human parainfluenza virus-4b(HPIV-4b), mumps virus(Mumps), 및 Newcastle disease virus(NDV) 등이 포함된다. 보다 바람직하게는, Sendai virus(SeV), human parainfluenza virus-1(HPIV-1), human parainfluenza virus-3(HPIV-3), phocine distemper virus(PDV), canine distemper virus(CDV), dolphin molbillivirus(DMV), peste-des-petits-ruminants virus(PDPR), measles virus(MV), rinderpest virus(RPV), Hendra virus(Hendra), 및 Nipah virus(Nipah)로 이루어진 군으로부터 선택되는 바이러스를 들 수 있다.
보다 바람직하게는, 파라믹소바이러스 아과(Paramyxoviridae)(레스피로바이러스속(Respirovirus), 루블라바이러스속(Rubulavirus), 및 모빌리바이러스속(Morbillivirus)을 포함한다)에 속하는 바이러스 또는 그의 유도체이고, 보다 바람직하게는 레스피로바이러스속(genus Respirovirus)(파라믹소바이러스속(Paramyxovirus)이라고도 한다)에 속하는 바이러스 또는 그의 유도체이다. 유도체에는, 바이러스에 의한 유전자 도입능을 손상시키지 않도록, 바이러스 유전자가 개변된 바이러스, 및 화학 수식된 바이러스 등이 포함된다. 본 발명을 적용 가능한 레스피로바이러스속 바이러스로서는, 예를 들면, 인간 파라인플루엔자 바이러스 1형(HPIV-1), 인간 파라인플루엔자 바이러스 3형(HPIV-3), 소 파라인플루엔자 바이러스 3형(bovine parainfluenza virus-3)(BPIV-3), 센다이 바이러스(Sendai virus; 마우스 파라인플루엔자 바이러스 1형이라고도 불리운다), 및 원숭이 파라인플루엔자 바이러스 10형(SPIV-10) 등이 포함된다.
본 발명의 마이너스 가닥 RNA 바이러스로서는, 가장 바람직하게는 센다이 바이러스이다.
본 발명에 있어서의 마이너스 가닥 RNA 바이러스는, 천연주(natural strain), 야생주(wild-type strain), 변이주(mutant strain), 라보라토리 계대주(laboratory-passaged strain), 및 인위적으로 구축된 주(strain) 등에 유래해도 된다. 즉, 마이너스 가닥 RNA 바이러스는, 천연으로부터 단리된 마이너스 가닥 RNA 바이러스여도, 유전자 재조합에 의해 인위적으로 생성된(generate) 마이너스 가닥 RNA 바이러스여도 된다. 또한, 감염세포에 있어서 게놈 RNA를 복제하는 능력을 보유하는 한, 야생형 마이너스 가닥 RNA 바이러스가 갖는 어느 하나의 유전자에 변이나 결손이 있는 것이어도 된다. 예를 들면, 마이너스 가닥 RNA 바이러스의 엔벨로프 단백질(envelope protein) 또는 피막 단백질(coat protein)을 코드하는 적어도 하나의 유전자에 변이 또는 결손을 가지는 마이너스 가닥 RNA 바이러스를 적합하게 사용할 수 있다. 이와 같은 마이너스 가닥 RNA 바이러스는, 감염세포에 있어서는 RNA 게놈을 복제할 수는 있지만, 감염성 바이러스 입자를 형성할 수 없다. 따라서, 주위에 감염을 확대할 우려가 없기 때문에 안전성이 높다. 예를 들면, F, H, HN, 또는 G 등의 엔벨로프 단백질 또는 스파이크 단백질(spike protein)을 코드하는 적어도 하나의 유전자, 또는 그들의 조합이 포함되어 있지 않은 마이너스 가닥 RNA 바이러스를 사용할 수 있다(WO00/70055 및 WO00/70070; Li, H.-O. et al., J. Virol. 74(14) 6564-6569(2000)). 게놈 복제에 필요한 단백질(예를 들면, N, P 및 L 단백질)을 게놈 RNA에 코드하고 있는 경우, 감염세포에 있어서 게놈을 증폭시킬 수 있다. 결손형 바이러스를 제조하기 위해서는, 예를 들면, 결손되어 있는 유전자 산물 또는 그것을 상보(相補)할 수 있는 단백질을 바이러스 생산세포에 있어서 외래적으로 공급한다(WO00/70055 및 WO00/70070; Li, H.-O. et al., J. Virol. 74(14) 6564-6569(2000)). 단, 예를 들면 마이너스 가닥 RNA 바이러스에 있어서는, M 단백질 유전자를 갖는 바이러스인 경우, F 단백질이나 HN 단백질 등의 스파이크 단백질을 코드하는 유전자를 갖지 않아도 비감염성 바이러스 입자(VLP)가 방출되기 때문에, 바이러스 단백질을 상보하지 않고 VLP를 제조하는 것이 가능하다(WO00/70070). 또한, 엔벨로프 단백질 유전자를 아무것도 갖지 않아도, N, L, P 단백질, 및 게놈 RNA로 되는 RNP는 세포 내에 있어서 증폭할 수 있기 때문에, 세포 용해물(cell lysate)로부터 원심분리 등에 의해 RNP를 회수할 수 있다.
또한, 변이형 RNA 바이러스를 사용하여 본 발명의 항암제를 제조하는 것도 가능하다. 예를 들면, 엔벨로프 단백질이나 피막 단백질에 있어서 다수의 온도 감수성 변이가 알려져 있다. 이들의 온도 감수성 변이 단백질 유전자를 갖는 RNA 바이러스를 본 발명에 있어서 적합하게 사용할 수 있다. 온도 감수성 변이란, 저온(예를 들면 30℃ 내지 32℃)에 비하여, 바이러스 숙주의 통상의 온도(예를 들면 37℃ 내지 38℃)에 있어서 유의하게 활성이 저하되는 변이를 말한다. 이와 같은, 온도 감수성 변이를 갖는 단백질은, 허용 온도(저온)하에서 바이러스를 제작할 수 있기 때문에 유용하다.
예를 들면, 마이너스 가닥 RNA 바이러스의 M 유전자의 온도 감수성 변이로서는, 센다이 바이러스의 M 단백질에 있어서의 G69, T116, 및 A183으로 이루어진 군으로부터 임의로 선택되는 부위의 아미노산 치환을 들 수 있다(Inoue, M. et al., J. 아미노산 변이는, 측쇄(side chain)의 화학적 성질이 상이한 다른 아미노산으로의 치환이 바람직하고, 예를 들면 BLOSUM62 매트릭스(matrix)(Henikoff, S. and Henikoff, J. G.(1992) Proc. Natl. Acad. Sci. USA 89: 10915-10919)의 값이 3 이하, 바람직하게는 2 이하, 보다 바람직하게는 1 이하, 더욱 바람직하게는 0 이하의 아미노산으로 치환한다. 구체적으로는, 센다이 바이러스 M 단백질의 G69, T116, 및 A183 또는 다른 바이러스 M 단백질의 상동부위를, 각각 Glu(E), Ala(A), 및 Ser(S)로 치환할 수 있다. 또한, 홍역 바이러스 온도 감수성주(temperature-sensitive strain) P253-505(Morikawa, Y. et al., Kitasato Arch. Exp. Med. 1991: 64; 15-30)의 M 단백질의 변이와 상동한 변이를 이용하는 것도 가능하다. 변이의 도입은, 예를 들면 올리고뉴클레오티드(oligonucleotide) 등을 사용하여, 공지의 변이 도입방법에 따라서 실시하면 된다.
또한, HN 유전자의 온도 감수성 변이로서는, 예를 들면 센다이 바이러스의 HN 단백질의 A262, G264, 및 K461로 이루어진 군으로부터 임의로 선택되는 부위의 아미노산 치환을 들 수 있다(Inoue, M. et al., J. Virol. 2003, 77: 3238-3246). 바람직한 일례를 들면, 센다이 바이러스 HN 단백질의 A262, G264, 및 K461 또는 다른 바이러스 HN 단백질의 상동부위를, 각각 Thr(T), Arg(R), 및 Gly(G)로 치환한다. 또한, 예를 들면, 유행성 이하선염 바이러스(mumps virus)의 온도 감수성 백신주 Urabe AM9를 참고로, HN 단백질의 464 및 468번째 아미노산으로 변이 도입하는 것도 가능하다(Wright, K. E. et al., Virus Res. 2000: 67; 49-57).
또한 마이너스 가닥 RNA 바이러스는, P 유전자 또는 L 유전자에 변이를 갖고 있어도 된다. 이와 같은 변이로서는, 구체적으로는, SeV P 단백질의 86번째 Glu(E86)의 변이, SeV P 단백질의 511번째 Leu(L511)의 다른 아미노산으로의 치환, 또는 다른 마이너스 가닥 RNA 바이러스 P 단백질의 상동부위의 치환을 들 수 있다. 구체적으로는, 86번째 아미노산의 Lys로의 치환, 511번째 아미노산의 Phe로의 치환 등을 예시할 수 있다. 또한 L 단백질에 있어서는, SeV L 단백질의 1197번째 Asn(N1197) 및/또는 1795번째 Lys(K1795)의 다른 아미노산으로의 치환, 또는 다른 마이너스 가닥 RNA 바이러스 L 단백질의 상동부위의 치환을 들 수 있고, 구체적으로는 1197번째 아미노산의 Ser로의 치환, 1795번째 아미노산의 Glu로의 치환 등을 예시할 수 있다. P 유전자 및 L 유전자의 변이는, 지속 감염성(persistent infectivity), 2차 입자 방출의 억제, 또는 세포 상해성(cytotoxicity) 억제의 효과를 현저하게 높일 수 있다. 또한, 엔벨로프 단백질 유전자의 변이 및/또는 결손을 조합시킴으로써, 이들의 효과를 극적으로 상승시킬 수 있다.
또한 엔벨로프 바이러스를 사용하는 경우는, 바이러스가 본래 갖는 엔벨로프 단백질과는 상이한 단백질을 엔벨로프에 포함하는 바이러스를 사용해도 된다. 예를 들면, 바이러스 제조시에, 목적의 외래성 엔벨로프 단백질을 바이러스 생산세포로 발현시킴으로써, 이것을 포함하는 바이러스를 제조할 수 있다. 이와 같은 단백질에 특별히 제한은 없고, 포유동물 세포로의 감염능을 부여하는 목적의 단백질이 사용된다. 구체적으로는, 예를 들면 수포성 구내염 바이러스(Vesicular stomatitis virus; VSV)의 G 단백질(VSV-G)을 들 수 있다. VSV-G 단백질은, 임의의 VSV주에 유래하는 것이어도 되고, 예를 들면 Indiana 혈청형 주(J. Virology 39: 519-528(1981)) 유래의 VSV-G 단백을 사용할 수 있지만, 이것에 한정되지 않는다. 본 발명에 있어서 사용되는 마이너스 가닥 RNA 바이러스는, 다른 바이러스 유래의 엔벨로프 단백질을 임의로 조합시켜 포함할 수 있다.
또한 본 발명의 마이너스 가닥 RNA 바이러스는, 그 자체로 항암작용을 갖는 것을 특징으로 한다. 즉, 상기 마이너스 가닥 RNA 바이러스는, 외래 단백질 또는 상기 단백질을 코드하는 핵산을 가질 필요는 없다. 따라서 본 발명은, 외래 단백질을 코드하지 않는 마이너스 가닥 RNA 바이러스를 포함하는 항암제에 관한 것이다. 본 발명의 마이너스 가닥 RNA 바이러스는, 게놈 RNA 중에 외래 단백질 또는 외래 유전자를 코드해도 되고, 하지 않아도 된다. 외래 단백질을 코드하지 않는 마이너스 가닥 RNA 바이러스일지라도 항암(제암)작용을 발휘하기 때문에, 외래 유전자는 반드시 필요하지는 않다. 따라서 본 발명에는, 야생형 마이너스 가닥 RNA 바이러스나 천연으로부터 단리된 마이너스 가닥 RNA 바이러스(변이체를 포함한다) 등의 목적의 마이너스 가닥 RNA 바이러스를 이용할 수 있는 이점이 있다. 예를 들면 본 발명에 있어서 사용하는 RNA 바이러스로서는, 암 치료효과를 갖는 단백질을 코드하지 않은 RNA 바이러스를 사용할 수 있다. 이와 같은 바이러스로서는, 암 치료효과를 가지지 않는 목적의 외래 단백질을 코드하는 RNA 바이러스가 포함되고, 예를 들면 RNA 바이러스의 도입을 검출하기 위해, 녹색 형광단백질(GFP), 루시페라아제(luciferase), 각종 펩티드 태그(peptide tag) 등의 마커 단백질(marker protein)을 코드하는 RNA 바이러스 등을 사용할 수 있다. 또는, 마이너스 가닥 RNA 바이러스로 항암(제암)작용을 돕는 외래 단백질 또는 외래 유전자를 추가로 삽입함으로써, 제암(항암)작용을 보다 높이는 것도 가능하다.
외래 유전자를 갖는 재조합 마이너스 가닥 RNA 바이러스의 재구성은 공지의 방법을 이용하여 행하면 된다. 구체적인 순서는, 전형적으로는, (a) 마이너스 가닥 RNA 바이러스 게놈 RNA를 코드하는 cDNA를, 바이러스 입자형성에 필요한 바이러스 단백질을 발현하는 세포로 전사시키는 공정, (b) 생성한 바이러스를 포함하는 배양상청(culture supernatant)을 회수하는 공정에 의해 제조할 수 있다. 바이러스 단백질은, 전사시킨 바이러스 게놈 RNA로부터 발현되어도 되고, 게놈 RNA 이외로부터 트랜스(trans)에 공급되어도 된다. 게놈 RNA에 있어서, 입자형성에 필요한 바이러스 유전자가 결손되어 있는 경우는, 그 바이러스 유전자를 바이러스 생산세포로 별도 발현시켜, 입자형성을 상보한다. 바이러스 단백질이나 RNA 게놈을 세포 내에서 발현시키기 위해서는, 상기 단백질이나 게놈 RNA를 코드하는 DNA를 숙주세포에서 기능하는 적당한 프로모터(promotor)의 하류에 연결한 벡터를 숙주세포에 도입한다. 전사된 게놈 RNA는, 바이러스 단백질의 존재하에서 복제되어, 감염성 바이러스 입자가 형성된다. 엔벨로프 단백질 등의 유전자를 결손하는 결손형 바이러스를 제조하는 경우는, 결손하는 단백질 또는 그의 기능을 상보할 수 있는 다른 바이러스 단백질 등을 바이러스 생산세포에 있어서 발현시킨다.

마이너스 가닥 RNA 바이러스에 탑재시키는 외래 유전자로서는, 특별히 제한은 없지만, 천연의 단백질로서는, 예를 들면 호르몬, 사이토카인(cytokine), 증식인자, 수용체, 세포내 시그널 분자(intracellular signaling molecule), 효소, 항체(전장 항체(full-length antibody), Fab 등의 항체 단편(antibody fragment), 단일 사슬 항체(single-chain antibody) 등을 포함한다), 펩티드 등을 들 수 있다. 단백질은 분비 단백질(secretory protein), 막 단백질(membrane protein), 세포질 단백질, 핵 단밸질 등 일 수 있다. 인공적인 단백질로서는, 예를 들면, 키메라 독소(chimeric toxin) 등의 융합 단백질(fusion protein), 도미넌트 음성 단백질(dominant negative protein)(수용체의 가용성 분자 또는 막 결합형 도미넌트 음성 수용체를 포함한다), 결실형 세포 접착분자 및 세포 표면분자 등을 들 수 있다. 또한, 분비 시그널, 막 국재화 시그널(membrane-localization signal), 또는 핵 이행 시그널(nuclear translocation signal) 등을 부가한 단백질이어도 된다. 도입 유전자로서 안티센스 RNA 분자 또는 RNA 절단형 리보자임(RNA-cleaving ribozyme) 등을 발현시켜, 특정 유전자의 기능을 억제하는 것도 가능하다. 외래 유전자로서 제암작용을 나타내는 치료용 유전자를 사용하여 바이러스를 조제하면, 제암작용을 보다 높이는 것이 가능해진다.
예를 들면, 혈관 형성 또는 혈관 신생을 저해하는 유전자를 사용하면, 항암효과를 보다 높일 수 있다. 혈관 형성 또는 혈관 신생을 촉진하는 유전자로서는, 예를 들면 섬유아세포 증식인자(fibroblast growth factor) 2(FGF2)(Baffour, R. et al., J. Vasc. Surg. 16(2):181-91, 1992), 내피세포 증식인자(endothelial cell growth factor; ECGF)(Pu, L. Q. et al., J. Surg. 또한, 본 발명에 있어서 사용되는 바이러스로서, 바이러스 게놈이 유래하는 바이러스의 엔벨로프 단백질과는 상이한 단백질을 엔벨로프에 포함하는 바이러스를 제작하는 것도 가능하다. 예를 들면, 바이러스 재구성시에, 베이스가 되는 바이러스의 게놈이 코드하는 엔벨로프 단백질 이외의 엔벨로프 단백질을 세포에서 발현시킴으로써, 목적의 엔벨로프 단백질을 갖는 바이러스를 제조할 수 있다. 이와 같은 단백질에 특별히 제한은 없다. 세포로의 감염능을 부여하는 목적의 단백질이 사용된다. 예를 들면, 다른 바이러스의 엔벨로프 단백질, 예를 들면, 수포성 구내염 바이러스(Vesicular stomatitis virus; VSV)의 G 단백질(VSV-G)을 들 수 있다. VSV-G 단백질은, 임의의 VSV주에 유래하는 것이어도 된다. 예를 들면, 인디아나 혈청형 주(Indiana serotype strain)(J. K. et al., J. Virol. 70: 5701-5705(1996)). 또한, 헤르페스바이러스과(Herpesviridae)의 단백질로서는, 예를 들면, 단순 헤르페스바이러스(herpes simplex virus)의 gB, gD, gH, gp85 단백질, EB 바이러스의 gp350, gp220 단백질 등을 들 수 있다. 헤파드나바이러스과(Hepadnaviridae)의 단백질로서는, B형 간염 바이러스의 S 단백질 등을 들 수 있다. 이들의 단백질은, 세포외 도메인을 F 단백질 또는 HN 단백질의 세포내 도메인과 결합시킨 융합 단백질로서 사용해도 된다. 이와 같이 본 발명에 있어서 사용되는 바이러스에는, VSV-G 단백질 등과 같이, 게놈이 유래하는 바이러스 이외의 바이러스에 유래하는 엔벨로프 단백질을 포함하는 슈도타입 바이러스(pseudotype virus)가 포함된다. 바이러스의 게놈 RNA에는 이들의 엔벨로프 단백질을 게놈에 코드되지 않도록 설계하면, 바이러스 입자가 세포에 감염된 후에는, 바이러스로부터 이 단백질이 발현되는 경우는 없다.
또한, 본 발명에 있어서 사용되는 바이러스는, 예를 들면, 엔벨로프 표면에 특정의 세포에 접착할 수 있는 접착인자, 리간드, 수용체 등의 단백질, 항체 또는 그의 단편, 또는 이들의 단백질을 세포외 영역에 가지고, 바이러스 엔벨로프 유래의 폴리펩티드를 세포내 영역에 갖는 키메라 단백질 등을 포함하는 것이어도 된다. 이들은 바이러스 게놈에 코드되어 있어도 되고, 바이러스의 재구성시에, 바이러스 게놈 이외의 유전자(예를 들면, 다른 발현 벡터 또는 숙주 염색체상 등에 있는 유전자)의 발현에 의해 공급되어도 된다.
또한 바이러스는, 예를 들면 바이러스 단백질에 의한 면역원성(Immunogenicity)을 저하시키기 위해, 또는 RNA의 전사 효율 또는 복제 효율을 높이기 위해, 바이러스에 포함되는 임의의 바이러스 유전자가 야생형 유전자로부터 개변되어 있어도 된다. 구체적으로는, 예를 들면, 복제인자인 N, P, 및 L 유전자 중 적어도 하나를 개변하여, 전사 또는 복제의 기능을 높이는 것을 생각할 수 있다. 또한, 엔벨로프 단백질의 하나인 HN 단백질은, 적혈구 응집소인 헤마글루티닌(hemagglutinin) 활성과 뉴라미니다아제(neuraminidase) 활성의 양자의 활성을 갖지만, 예를 들면, 전자의 활성을 약하게 할 수 있다면, 혈액 중에서의 바이러스의 안정성을 향상시키는 것이 가능할 것이고, 예를 들면, 후자의 활성을 개변함으로써, 감염능을 조절하는 것도 가능하다. 또한, F 단백질을 개변함으로써 막 융합능(membrane fusion ability)을 조절하는 것도 가능하다. 또한, 예를 들면, 세포 표면의 항원 분자가 될 수 있는 F 단백질 및/또는 HN 단백질의 항원제시 에피토프(epitope) 등을 해석하고, 이것을 이용하여 이들의 단백질에 관한 항원제시능을 약하게 한 바이러스를 제작하는 것도 가능하다.
또한, 마이너스 가닥 RNA 바이러스는, 액세서리 유전자(accessory gene)가 결손된 것이어도 된다. 예를 들면, SeV의 액세서리 유전자의 하나인 V 유전자를 녹아웃(knock out)시킴으로써, 배양세포에 있어서의 유전자 발현 및 복제는 장해받지 않아, 마우스 등의 숙주에 대한 SeV의 병원성이 현저하게 감소한다(Kato, A. et al., 1997, J. Virol. 71: 7266-7272; Kato, A. et al., 1997, EMBO J. 16: 578-587; Curran, J. et al., WO01/04272, EP1067179).
마이너스 가닥 RNA 바이러스는, 예를 들면, 그 자체의 항암작용과의 상승효과가 기대되는 외래 유전자를 도입하기 위한 벡터로서의 역할을 하게 하는 것도 가능하다. 상기 벡터는, 숙주세포의 세포질에서만 전사·복제를 행하여, DNA 페이즈(DNA phase)를 갖지 않기 때문에 염색체로의 통합(integration)은 일어나지 않는다(Lamb, R. A. and Kolakofsky, D., Paramyxoviridae: The viruses and their replication. In: Fields BN, Knipe DM, Howley PM, (eds). Fields of virology. Vol. 2. Lippincott-Raven Publishers: Philadelphia, 1996, pp. 1177-1204). 이 때문에 염색체 이상에 의한 암화(癌化) 및 불멸화(immortalization) 등의 안전면에 있어서의 문제가 발생하지 않는다. 마이너스 가닥 RNA 바이러스의 이 특징은, 벡터로서 이용했을 때의 안전성에 크게 기여하고 있다. 이종 유전자 발현의 결과에서는, 예를 들면, 센다이 바이러스(SeV)를 연속 다대 계대(multiple continuous passage)해도 거의 염기의 변이가 인정되지 않고, 게놈의 안정성이 높으며, 삽입 이종 유전자를 장기간에 걸쳐 안정하게 발현하는 것이 나타내어져 있다(Yu, D. et al., Genes Cells 2, 457-466(1997)). 또한, 캡시드 구조 단백질(capsid structural protein)을 갖지 않는 것에 의한 도입 유전자의 사이즈 또는 팩키징(packaing)의 유연성(flexibility) 등 성질상의 이점이 있다. 이와 같이, 마이너스 가닥 RNA 바이러스에는, 인간의 유전자 치료를 위한 고효율 벡터로서의 기능을 부가적으로 담당하게 하는 것도 가능하다. 예를 들면, 전파능을 가지는 센다이 바이러스는, 외래 유전자를 적어도 4 kb까지 도입 가능하고, 전사 유닛(transcriptional unit)을 부가함으로써 2종류 이상의 유전자를 동시에 발현하는 것도 가능하다.
또한 센다이 바이러스는, 설치류(rodent)에 있어서는 병원성으로 폐렴을 발생시키는 것이 알려져 있지만, 인간에 대해서는 병원성이 없다. 이것은 또한, 야생형 센다이 바이러스의 경비적 투여(nasal administration)에 의해 비인간 영장류(non-human primate)에 있어서 심각한 유해작용을 나타내지 않는다고 하는 지금까지의 보고에 의해서도 지지되고 있다(Hurwitz, J.L. et al., Vaccine 15:533-540, 1997). 센다이 바이러스의 이들 특징은, 센다이 바이러스가 인간의 치료로 응용할 수 있는 것을 시사하고, 암의 유전자치료의 유망한 선택지의 하나가 되는 것을 결론짓는 것이다.
본 발명의 마이너스 가닥 RNA 바이러스는, 필수는 아니지만, 게놈 RNA 중에 외래 유전자를 코드해도 된다. 외래 유전자를 포함하는 재조합 바이러스는, 상기 바이러스의 게놈에 외래 유전자를 삽입함으로써 얻어진다. 외래 유전자로서는, 본 발명의 RNA 바이러스의 항암작용과의 상승효과 등을 기대할 수 있는 목적의 유전자를 사용할 수 있다. 외래 유전자는 천연형 단백질을 코드하는 유전자여도 되고, 또는 결실, 치환 또는 삽입에 의해 천연형 단백질을 개변한 단백질을 코드하는 유전자여도 된다. 외래 유전자의 삽입 위치는, 예를 들면, 바이러스 게놈의 단백질 비코드 영역의 목적의 부위를 선택할 수 있고, 예를 들면, 게놈 RNA의 3' 리더 영역과 3' 말단에 가장 가까운 바이러스 단백질 ORF의 사이, 각 바이러스 단백질 ORF의 사이, 및/또는 5' 말단에 가장 가까운 바이러스 단백질 ORF와 5' 트레일러 영역 사이에 삽입할 수 있다. 또한, F 또는 HN 유전자 등을 결실하는 게놈에서는, 그의 결실 영역에 외래 유전자를 코드하는 핵산을 삽입할 수 있다. 파라믹소바이러스에 외래 유전자를 도입하는 경우는, 게놈으로의 삽입 단편의 폴리뉴클레오티드의 쇄장(chain length)이 6의 배수가 되도록 삽입하는 것이 바람직하다(Journal of Virology, Vol. 67, No. 8, 4822-4830, 1993). 삽입한 외래 유전자와 바이러스 ORF의 사이에는, E-I-S 서열이 구성되도록 한다. E-I-S 서열을 매개로 하여 2 또는 그 이상의 유전자를 직렬로 배열하여 삽입할 수 있다.
마이너스 가닥 RNA 바이러스에 탑재하는 외래 유전자의 발현 레벨은, 그의 유전자의 상류(음성 가닥의 3'측)에 부가하는 전사개시서열의 종류에 따라 조절할 수 있다(WO01/18223). 또한, 게놈상의 외래 유전자의 삽입 위치에 의해 제어할 수 있어, 음성 가닥의 3' 가까이에 삽입할수록 발현 레벨이 높고, 5' 가까이에 삽입할수록 발현 레벨이 낮아진다. 이와 같이, 외래 유전자의 삽입 위치는, 상기 유전자의 목적의 발현량을 얻기 위해, 또한 전후의 바이러스 단백질을 코드하는 유전자와의 조합이 최적이 되도록 적절히 조절할 수 있다. 일반적으로, 외래 유전자의 높은 발현이 얻어지는 것이 유리하다고 생각되어지기 때문에, 외래 유전자는 효율이 높은 전사개시서열에 연결하고, 음성 가닥 게놈의 3' 말단 가까이에 삽입하는 것이 바람직하다. 구체적으로는, 3' 리더 영역과 3'에 가장 가까운 바이러스 단백질 ORF의 사이에 삽입된다. 또는, 3'에 가장 가까운 바이러스 유전자의 ORF와 2번째 유전자의 ORF 사이에 삽입해도 된다. 야생형 파라믹소바이러스에 있어서는, 게놈의 3'에 가장 가까운 바이러스 단백질 유전자는 N 유전자이고, 2번째 유전자는 P 유전자이다. 반대로, 도입 유전자의 고발현이 바람직하지 않은 경우는, 예를 들면, 바이러스에 있어서의 외래 유전자의 삽입 위치를 음성 가닥의 되도록 5'측에 설정하거나, 전사개시서열을 효율이 낮은 것으로 하는 등으로 하여, 바이러스로부터의 발현 레벨을 낮게 억제함으로써 적절한 효과가 얻어지도록 하는 것도 가능하다.
마이너스 가닥 바이러스를 제조하기 위해서는, 포유동물세포에 있어서, 바이러스의 성분인 RNP의 재구성에 필요한 바이러스 단백질, 즉, N, P, 및 L 단백질의 존재하, 바이러스의 게놈 RNA를 코드하는 cDNA를 전사시킨다. 전사에 의해 음성 가닥 게놈(즉 바이러스 게놈과 동일한 안티센스 가닥)을 생성시켜도 되고, 또는 양성 가닥(positive strand)(바이러스 단백질을 코드하는 센스 가닥)을 생성시켜도, 바이러스 RNP를 재구성할 수 있다. 마이너스 가닥 RNA 바이러스의 재구성 효율을 높이기 위해서는, 바람직하게는 양성 가닥을 생성시킨다. RNA 말단은, 천연의 바이러스 게놈과 동일하게 3' 리더 서열과 5' 트레일러 서열의 말단을 되도록 정확하게 반영시키는 것이 바람직하다. 전사 산물의 5' 말단을 정확하게 제어하기 위해서는, 예를 들면, 전사개시부위로서 T7 RNA 폴리머라아제 인식서열을 이용하여, 상기 RNA 폴리머라아제를 세포 내에서 발현시키면 된다. 전사 산물의 3' 말단을 제어하기 위해서는, 예를 들면, 전사 산물의 3' 말단에 자기 절단형 리보자임을 코드시켜 두고, 이 리보자임에 의해 정확하게 3' 말단이 잘려내어지도록 할 수 있다(Hasan, M. K. et al., J. Gen. Virol. 78: 2813-2820, 1997, Kato, A. et al., 1997, EMBO J. 16: 578-587 및 Yu, D. et al., 1997, Genes Cells 2: 457-466).
예를 들면, 외래 유전자를 갖는 재조합 센다이 바이러스는, Hasan, M. K. et al., J. Gen. Virol. 78: 2813-2820, 1997, Kato, A. et al., 1997, EMBO J. 16: 578-587 및 Yu, D. et al., 1997, Genes Cells 2: 457-466의 기재 등에 준하여, 다음과 같이 하여 구축할 수 있다.
먼저, 목적의 외래 유전자의 cDNA 염기서열을 포함하는 DNA 시료를 준비한다. DNA 시료는, 25 ng/micro-L 이상의 농도로 전기영동적으로 단일한 플라스미드라고 확인할 수 있는 것이 바람직하다. 이하, NotI 부위를 이용하여 바이러스 게놈 RNA를 코드하는 DNA에 외래 유전자를 삽입하는 경우를 예로서 설명한다. 목적으로 하는 cDNA 염기서열 중에 NotI 인식부위가 포함되는 경우는, 부위 특이적 변이 도입법 등을 사용하여, 코드하는 아미노산서열을 변화시키지 않도록 염기서열을 개변하고, NotI 부위를 미리 제거해 두는 것이 바람직하다. 이 시료로부터 목적의 유전자 단편을 PCR에 의해 증폭하여 회수한다. 2개의 프라이머(primer)의 5' 부분에 NotI 부위를 부가해 둠으로써, 증폭된 단편의 양쪽 말단을 NotI 부위로 한다. 바이러스 게놈상에 삽입된 후의 외래 유전자의 ORF와 그의 양쪽 바이러스 유전자의 ORF 사이에 E-I-S 서열이 배치되도록, 프라이머 중에 E-I-S 서열을 포함할 수 있도록 설계한다.
예를 들면, 포워드측(forward side) 합성 DNA 서열은, NotI에 의한 절단을 보증하기 위해 5'측에 임의의 2 이상의 뉴클레오티드(바람직하게는 GCG 및 GCC 등의 NotI 인식부위 유래의 서열이 포함되지 않는 4 염기, 더욱 바람직하게는 ACTT)를 선택하여, 그의 3'측에 NotI 인식부위 gcggccgc를 부가하고, 추가로 그의 3'측에 스페이서 서열(spacer sequence)로서 임의의 9 염기 또는 9에 6의 배수를 더한 수의 염기를 부가하고, 추가로 그의 3'측에 목적의 cDNA의 개시 코돈 ATG(initiation codon ATG)로부터 이것을 포함하여 ORF의 약 25 염기 상당의 서열을 부가한 형태로 한다. 마지막 염기는 G 또는 C가 되도록 상기 목적의 cDNA로부터 약 25 염기를 선택하여 포워드측 합성 올리고 DNA의 3'의 말단으로 하는 것이 바람직하다.
리버스측(reverse side) 합성 DNA 서열은 5'측으로부터 임의의 2 이상의 뉴클레오티드(바람직하게는 GCG 및 GCC 등의 NotI 인식부위 유래의 서열이 포함되지 않는 4 염기, 더욱 바람직하게는 ACTT)를 선택하여, 그의 3'측에 NotI 인식부위 gcggccgc를 부가하고, 추가로 그의 3'측에 길이를 조절하기 위한 삽입 단편 올리고 DNA를 부가한다. 이 올리고 DNA의 길이는, 최종적인 PCR 증폭 산물의 NotI 단편의 쇄장이 6의 배수가 되도록 염기수를 설계한다(이른바 「6의 룰(rule of six)」; Kolakofski, D. et al., J. PCR은, Taq 폴리머라아제 또는 그 밖의 DNA 폴리머라아제를 사용하는 통상의 방법을 사용할 수 있다. 증폭한 목적 단편은 NotI로 소화한 후, pBluescript 등의 플라스미드 벡터의 NotI 부위에 삽입한다. 얻어진 PCR 산물의 염기서열을 시퀀서(sequencer)로 확인하여, 올바른 서열의 플라스미드를 선택한다. 이 플라스미드로부터 삽입 단편을 NotI로 잘라내고, 게놈 cDNA를 포함하는 플라스미드의 NotI 부위에 클로닝한다. 또한 플라스미드 벡터를 매개로 하지 않고 게놈 cDNA의 NotI 부위에 직접 삽입하여, 재조합 센다이 바이러스 cDNA를 얻는 것도 가능하다.

Virol. 78: 2813-2820, 1997). 예를 들면, NotI 제한부위를 갖는 18 bp의 스페이서 서열(5'-(G)-CGGCCGCAGATCTTCACG-3')(서열번호: 3)을, 클로닝된 센다이 바이러스 게놈 cDNA(pSeV(+))의 리더 서열과 N 단백질의 ORF의 사이에 삽입하고, 델타 간염 바이러스(delta hepatitis virus)의 안티게놈 가닥(antigenomic strand) 유래의 자기개열 리보자임 부위(auto-cleavage ribozyme site)를 포함하는 플라스미드 pSeV18+b(+)를 얻는다(Hasan, M. K. et al., 1997, J. General Virology 78: 2813-2820). pSeV18+b(+)의 NotI 부위에 외래 유전자 단편을 삽입하여, 목적의 외래 유전자가 삽입된 재조합 센다이 바이러스 cDNA를 얻을 수 있다.
이와 같이 하여 제작한 재조합 바이러스의 게놈 RNA를 코드하는 DNA를, 상기의 바이러스 단백질(L, P, 및 N) 존재하에 있어서 세포 내에서 전사시킴으로써, 바이러스를 재구성할 수 있다.
또한, 재조합 바이러스의 재구성은 공지의 방법을 이용해도 행할 수 있다(WO97/16539; WO97/16538; Durbin, A. P. et al., 1997, Virology 235: 323-332; Whelan, S. P. et al., 1995, Proc. Natl. Acad. Sci. USA 92: 8388-8392; Schnell. M. J. et al., 1994, EMBO J. 13: 4195-4203; Radecke, F. et al., 1995, EMBO J. 14: 5773-5784; Lawson, N. D. et al., Proc. Natl. Acad. Sci. USA 92: 4477-4481; Garcin, D. et al., 1995, EMBO J. 14: 6087-6094; Kato, A. et al., 1996, Genes Cells 1: 569-579; Baron, M. D. and Barrett, T., 1997, J. Virol. 71: 1265-1271; Bridgen, A. and Elliott, R. M., 1996, Proc. Natl. Acad. Sci. USA 93: 15400-15404). 이들의 방법에 의해, 파라인플루엔자, 수포성 구내염 바이러스, 광견병 바이러스, 홍역 바이러스, 우역 바이러스, 센다이 바이러스 등을 포함하는 마이너스 가닥 RNA 바이러스를 DNA로부터 재구성시킬 수 있다. 이들의 방법에 준하여, 본 발명의 바이러스를 재구성시킬 수 있다. 바이러스 DNA에 있어서, F 유전자, HN 유전자, 및/또는 M 유전자를 결실시킨 경우에는, 그대로는 감염성 바이러스 입자를 형성하지 않지만, 숙주세포에 이들을 결실시킨 유전자 및/또는 다른 바이러스의 엔벨로프 단백질을 코드하는 유전자 등을 별도로 세포에 도입하여 발현시킴으로써, 감염성 바이러스 입자를 형성시키는 것이 가능하다.
구체적인 순서는, (a) 마이너스 가닥 RNA 바이러스 게놈 RNA(음성 가닥 RNA) 또는 그의 상보가닥(양성 가닥)을 코드하는 cDNA를, N, P, 및 L 단백질을 발현하는 세포에서 전사시키는 공정, (b) 생성한 마이너스 가닥 RNA 바이러스를 포함하는 배양상청을 회수하는 공정에 의해 제조할 수 있다. 전사를 위해, 게놈 RNA를 코드하는 DNA는 적당한 프로모터의 하류에 연결된다. 전사된 게놈 RNA는 N, L, 및 P 단백질의 존재하에서 복제되어 RNP 복합체를 형성한다. 그리고 M, HN, 및 F 단백질의 존재하에서 엔벨로프에 싸여진 바이러스 입자가 형성된다. 게놈 RNA를 코드하는 DNA는, 예를 들면, T7 프로모터의 하류에 연결시켜, T7 RNA 폴리머라아제에 의해 RNA에 전사시킨다. 프로모터로서는, T7 폴리머라아제의 인식서열을 포함하는 것 이외에도 목적의 프로모터를 이용할 수 있다. 또는, 인 비트로에서 전사시킨 RNA를 세포에 트랜스펙트해도 된다.
DNA로부터의 게놈 RNA의 최초의 전사에 필요한 T7 RNA 폴리머라아제 등의 효소는, 이것을 발현하는 플라스미드 또는 바이러스 벡터의 도입에 의해 공급할 수 있고, 또는, 예를 들면, 세포의 염색체에 RNA 폴리머라아제 유전자를, 발현을 유도할 수 있도록 삽입해 두고, 바이러스 재구성시에 발현을 유도함으로써 공급하는 것도 가능하다. 또한 게놈 RNA, 및 바이러스 재구성에 필요한 바이러스 단백질은, 예를 들면, 이들을 발현하는 플라스미드의 도입에 의해 공급된다. 이들의 바이러스 단백질의 공급에 있어서, 야생형 또는 어떤 종의 변이 마이너스 가닥 RNA 바이러스 등의 헬퍼 바이러스(helper virus)를 사용하는 것도 가능하다.
게놈 RNA를 발현하는 DNA를 세포 내에 도입하는 방법으로는, 예를 들면, 다음과 같은 방법, (ⅰ) 목적의 세포를 내재시킬 수 있는 DNA 침전물을 만드는 방법, (ⅱ) 목적의 세포에 의한 내재화(internalization)에 적합하고, 또한 세포독성이 적은 양전하 특성을 가지는 DNA를 포함하는 복합체를 만드는 방법, (ⅲ) 목적의 세포막에, DNA 분자가 빠져나갈 수 있을 만큼 충분한 구멍을 전기 펄스(electric pulse)에 의해 순간적으로 뚫는 방법 등이 있다.
(ⅱ)로서는, 여러 가지의 트랜스펙션 시약을 이용할 수 있다. 예를 들면, DOTMA(Roche), Superfect(QIAGEN #301305), DOTAP, DOPE, DOSPER(Roche #1811169) 등을 들 수 있다. (ⅰ)로서는 예를 들면 인산칼슘을 사용한 트랜스펙션법을 들 수 있고, 이 방법에 의해 세포 내에 든 DNA는 식포(phagosome)에 내재되지만, 핵 내에도 충분한 양의 DNA가 들어가는 것이 알려져 있다(Graham, F. L. and Van Der Eb, J., 1973, Virology 52: 456; Wigler, M. and Silverstein, S., 1977, Cell 11: 223). Chen 및 Okayama는 트랜스퍼 기술의 최적화를 검토하여, 1) 세포와 공침전물의 인큐베이션 조건을 2~4% CO2, 35℃, 15~24시간, 2) DNA는 직쇄상(linear)보다 환상(circular)의 것이 활성이 높고, 3) 침전 혼액 중의 DNA 농도가 20~30 micro-g/㎖일 때 최적의 침전이 얻어진다고 보고하고 있다(Chen, C. and Okayama, H., 1987, Mol. Cell. Biol. 7: 2745). (ⅱ)의 방법은, 일과적인 트랜스펙션에 적합하다. 예전부터 DEAE-덱스트란(Sigma #D-9885 M. W. 5×105) 혼액을 목적의 DNA 농도비로 조제하고, 트랜스펙션을 행하는 방법이 알려져 있다. 복합체의 대부분은 엔도솜(endosome) 중에서 분해되어 버리기 때문에, 효과를 높이기 위해 클로로퀸(chloroquine)을 첨가하는 것도 가능하다(Calos, M. P., 1983, Proc. Natl. Acad. Sci. USA 80: 3015). (ⅲ)의 방법은 전기천공법(electroporation method)으로 불리우는 방법으로, 세포 선택성이 없다고 하는 점에서 (ⅰ) 또는 (ⅱ)의 방법에 비하여 범용성이 높다. 효율은 펄스 전류의 지속시간, 펄스의 형태, 전계(전극간의 갭, 전압)의 세기, 버퍼(buffer)의 도전율, DNA 농도, 세포밀도의 최적 조건하에서 좋다고 여겨지고 있다.
이상, 3개의 카테고리 중에서 (ⅱ)의 방법은 조작이 간편하고 다량의 세포를 사용하여 다수의 검체를 검토할 수 있기 때문에, 바이러스 재구성을 위한 DNA의 세포로의 도입에는, 트랜스펙션 시약이 적합하다. 적합하게는 Superfect Transfection Reagent(QIAGEN. Cat No. 301305), 또는 DOSPER Liposomal Transfection Reagent(Roche, Cat No. 1811169)가 이용되지만, 이들에 제한되지 않는다.
cDNA로부터의 바이러스의 재구성은 구체적으로는 예를 들면 이하와 같이 하여 행할 수 있다.
24웰(well)에서 6웰 정도의 플라스틱 플레이트 또는 100 ㎜ 페트리 접시(petri dish) 등에서, 10% 소 태아 혈청(FCS) 및 항생물질(100 units/㎖ 페니실린 G 및 100 micro-g/㎖ 스트렙토마이신(streptomycin))을 포함하는 최소 필수배지(MEM)를 사용하여 원숭이 신장 유래 세포주 LLC-MK2(ATCC CCL-7)를 거의 100% 컨플루언트(confluent)가 될 때까지 배양하고, 예를 들면, 1 micro-g/㎖ 소랄렌(psoralen) 존재하, 자외선(UV) 조사처리를 20분 처리로 불활성화시킨, T7 RNA 폴리머라아제를 발현하는 재조합 백시니아 바이러스(vaccinia virus) vTF7-3(Fuerst, T. R. et al., Proc. Natl. Acad. Sci. USA 83: 8122-8126, 1986, Kato, A. et al., Genes Cells 1: 569-579, 1996)를 2 PFU/세포로 감염시킨다. 소랄렌의 첨가량 및 UV 조사시간은 적절히 조정할 수 있다. 감염 1시간 후, 2~60 micro-g, 보다 바람직하게는 3~20 micro-g의 재조합 센다이 바이러스의 게놈 RNA를 코드하는 DNA를, 바이러스 RNP의 생성에 필수인 트랜스에 작용하는 바이러스 단백질을 발현하는 플라스미드(0.5~24 micro-g의 pGEM-N, 0.25~12 micro-g의 pGEM-P, 및 0.5~24 micro-g의 pGEM-L)(Kato, A. et al., Genes Cells 1: 569-579, 1996)와 함께 Superfect(QIAGEN사)를 사용한 리포펙션법(lipofection method) 등에 의해 트랜스펙션한다. N, P, 및 L을 코드하는 발현 벡터의 양비는 예를 들면, 2:1:2로 하는 것이 바람직하고, 플라스미드량은, 예를 들면 1~4 micro-g의 pGEM-N, 0.5~2 micro-g의 pGEM-P, 및 1~4 micro-g의 pGEM-L 정도로 적절히 조정한다.
트랜스펙션을 행한 세포는, 목적에 따라 100 micro-g/㎖의 리팜피신(rifampicin)(Sigma) 및 시토신 아라비노시드(cytosine arabinoside)(Arac), 보다 바람직하게는 40 micro-g/㎖의 시토신 아라비노시드(Arac)(Sigma)만을 포함하는 혈청이 포함되지 않은 MEM에서 배양하고, 백시니아 바이러스에 의한 세포독성을 최소로 억제하여, 바이러스의 회수율을 최대로 하도록 약제의 최적 농도를 설정한다(Kato, A. et al., 1996, Genes Cells 1: 569-579). 트랜스펙션으로부터 48~72시간 정도 배양 후, 세포를 회수하고, 동결 융해를 3회 반복하여 세포를 파쇄한 후, RNP를 포함하는 파쇄물을 LLC-MK2 세포에 재차 트랜스펙션하여 배양한다. 또는, 배양상청을 회수하고, LLC-MK2 세포의 배양액에 첨가하여 감염시켜 배양한다. 트랜스펙션은, 예를 들면, 리포펙트아민(lipofectamine) 또는 폴리카티오닉 리포솜(polycationic liposome) 등과 함께 복합체를 형성시켜 세포에 도입하는 것이 가능하다. 구체적으로는, 여러 가지의 트랜스펙션 시약을 이용할 수 있다. 예를 들면, DOTMA(Roche), Superfect(QIAGEN #301305), DOTAP, DOPE, DOSPER(Roche #1811169) 등을 들 수 있다. 엔도솜 중에서의 분해를 막기 위해, 클로로퀸을 첨가하는 것도 가능하다(Calos, M, P., 1983, Proc. Natl. Acad. Sci. USA 80: 3015). RNP가 도입된 세포에서는, RNP로부터의 바이러스 유전자의 발현 및 RNP 복제의 과정이 진행되어 벡터가 증폭된다. 얻어진 바이러스 용액을 희석(예를 들면, 106배)하여 재증폭을 반복함으로써, 백시니아 바이러스 vTF7-3는 완전히 제거할 수 있다. 재증폭은, 예를 들면, 3회 이상 반복한다. 얻어진 바이러스는 -80℃에서 보존할 수 있다. 엔벨로프 단백질을 코드하는 유전자를 결손한 전파능을 가지지 않는 바이러스를 재구성시키기 위해서는, 엔벨로프 단백질을 발현하는 LLC-MK2 세포를 트랜스펙션에 사용하거나, 또는 엔벨로프 발현 플라스미드를 함께 트랜스펙션하면 된다. 또한, 트랜스펙션을 행한 세포에 엔벨로프 단백질을 발현하는 LLC-MK2 세포를 중층하여 배양함으로써 결손형 바이러스를 증폭하는 것도 가능하다(국제공개번호 WO00/70055 및 WO00/70070 참조).
회수된 바이러스의 역가(titer)는, 예를 들면 CIU(Cell-Infected Unit) 측정 또는 적혈구 응집 활성(hemagglutination activity)(HA)을 측정함으로써 결정할 수 있다(WO00/70070; Kato, A. et al., 1996, Genes Cells 1: 569-579; Yonemitsu, Y. & Kaneda, Y., Hemagglutinating virus of Japan-liposome-mediated gene delivery to vascular cells. Ed. by Baker AH. Molecular Biology of Vascular Diseases. Method in Molecular Medicine: Humana Press: pp. 295-306, 1999). 또한, GFP(녹색 형광 단백질(green fluorescent protein)) 등의 마커 유전자를 탑재한 바이러스에 대해서는, 마커를 지표로 직접적으로 감염 세포를 카운트함으로써 역가를 정량할 수 있다(예를 들면, GFP-CIU로서). 이와 같이 하여 측정한 역가는, CIU와 동일하게 취급할 수 있다(WO00/70070).
바이러스가 재구성되는 한, 재구성에 사용하는 숙주세포는 특별히 제한되지 않는다. 예를 들면, 센다이 바이러스 등의 재구성에 있어서는, 원숭이 신장 유래의 LLC-MK2 세포 및 CV-1 세포, 햄스터 신장 유래의 BHK 세포 등의 배양세포, 인간 유래 세포 등을 사용할 수 있다. 이들의 세포에 적당한 엔벨로프 단백질을 발현시킴으로써, 그 단백질을 엔벨로프에 갖는 감염성 바이러스 입자를 얻는 것도 가능하다. 또한, 대량으로 센다이 바이러스를 얻기 위해, 상기의 숙주로부터 얻어진 바이러스를 발육 계란에 감염시켜, 상기 바이러스를 증폭시킬 수 있다. 계란을 사용한 바이러스의 제조방법은 이미 개발되어 있다(나카니시 등 편, (1993), 「신경과학연구의 첨단기술 프로토콜 Ⅲ, 분자 신경세포 생리학」, 고세이샤, 오사카, pp. 153-172). 구체적으로는, 예를 들면, 수정란을 배양기에 넣고 9~12일간 37~38℃에서 배양하여, 배(embryo)를 성장시킨다. 바이러스를 요막강(allantoic cavity)으로 접종하고, 수일간(예를 들면, 3일간) 알을 배양하여 바이러스를 증식시킨다. 배양기간 등의 조건은, 사용하는 재조합 센다이 바이러스에 의해 변할 수 있다. 그 다음, 바이러스를 포함한 요액(allantoic fluid)을 회수한다. 요액으로부터의 센다이 바이러스의 분리·정제는 통상적인 방법에 따라서 행할 수 있다(다시로 마비토, 「바이러스 실험 프로토콜」, 나가이, 이시하마 감수, 메티컬 뷰사, pp. 68-73, (1995)).
예를 들면, F 유전자를 결실한 센다이 바이러스의 구축과 조제는, 이하와 같이 행할 수 있다(WO00/70055 및 WO00/70070 참조).
<1> F 유전자 결실형 센다이 바이러스 게놈 cDNA 및 F 발현 플라스미드의 구축
센다이 바이러스(SeV) 전장 게놈 cDNA, pSeV18+ b(+)(Hasan, M. K. et al., 1997, J. General Virology 78: 2813-2820)(「pSeV18+ b(+)」는 「pSeV18+」라고도 한다)의 cDNA를 SphI/KpnI로 소화하여 프래그먼트(14673 bp)를 회수하고, pUC18에 클로닝하여 플라스미드 pUC18/KS로 한다. F 유전자 결손부위의 구축은 이 pUC18/KS상에서 행한다. F 유전자의 결손은, PCR-라이게이션 방법의 조합으로 행하고, 결과로서 F 유전자의 ORF(ATG-TGA=1698 bp)를 제거하고, 예를 들면, atgcatgccggcagatga(서열번호: 4)에서 연결하여, F 유전자 결실형 SeV 게놈 cDNA(pSeV18+/△F)를 구축한다. PCR은, F의 상류에는 [forward: 5'-gttgagtactgcaagagc/서열번호: 5, reverse: 5'-tttgccggcatgcatgtttcccaaggggagagttttgcaacc/서열번호: 6], F 유전자의 하류에는 [forward: 5'-atgcatgccggcagatga/서열번호: 7, reverse: 5'-tgggtgaatgagagaatcagc/서열번호: 8]의 프라이머쌍(pair of primer)을 사용한 PCR의 산물을 EcoT22I로 연결한다. 이와 같이 얻어진 플라스미드를 SacI와 SalI로 소화하고, F 유전자 결손부위를 포함하는 영역의 단편(4931 bp)을 회수해서 pUC18에 클로닝하여, pUC18/dFSS로 한다. 이 pUC18/dFSS를 DraⅢ로 소화하고, 단편을 회수하여 pSeV18+의 F 유전자를 포함하는 영역의 DraⅢ 단편과 치환하며, 라이게이션하여 플라스미드 pSeV18+/△F를 얻는다.
외래 유전자는, 예를 들면, pUC18/dFSS의 F 유전자 결실부위에 있는 제한효소 NsiI 및 NgoMⅣ 부위에 삽입한다. 이를 위해서는, 예를 들면, 외래 유전자 단편을, NsiI-tailed 프라이머 및 NgoMIV-tailed 프라이머로 증폭하면 된다.
<2> SeV-F 단백을 유도 발현하는 헬퍼 세포의 제작
센다이 바이러스의 F 유전자(SeV-F)를 발현하는 Cre/loxP 유도형 발현 플라스미드의 구축은 SeV-F 유전자를 PCR로 증폭하고, Cre DNA 리콤비나아제(recombinase)에 의해 유전자 산물이 유도 발현되도록 설계된 플라스미드 pCALNdlw(Arai, T. et al., J. Virology 72, 1998, p1115-1121)의 유니크 사이트 SwaI 부위에 삽입하여, 플라스미드 pCALNdLw/F를 구축한다.
F 유전자 결손 게놈으로부터 감염 바이러스 입자를 회수하기 위해, SeV-F 단백을 발현하는 헬퍼 세포주를 수립한다. 세포는, 예를 들면, SeV의 증식에 자주 사용되고 있는 원숭이 신장 유래 세포주 LLC-MK2 세포를 사용할 수 있다. LLC-MK2 세포는, 10%의 열처리한 불활성화 소 태아 혈청(FBS), 페니실린 G 나트륨 50 단위/㎖, 및 스트렙토마이신(streptomycin) 50 micro-g/㎖를 첨가한 MEM으로 37℃, 5% CO2에서 배양한다. SeV-F 유전자 산물은 세포상해성을 가지기 때문에, Cre DNA 리콤비나아제에 의해 F 유전자 산물이 유도 발현되도록 설계된 상기 플라스미드 pCALNdLw/F를, 인산칼슘법(mammalian transfection kit(Stratagene))에 의해, 주지의 프로토콜에 따라서 LLC-MK2 세포에 도입한다.
10 ㎝ 플레이트를 사용하여, 40% 컨플루언트까지 생육한 LLC-MK2 세포에 10 micro-g의 플라스미드 pCALNdLw/F를 도입 후, 10 ㎖의 10% FBS를 포함하는 MEM 배지에, 37℃의 5% CO2 인큐베이터 중에서 24시간 배양한다. 24시간 후에 세포를 벗겨, 10 ㎖ 배지에 현탁 후, 10 ㎝ 샬레 5장을 사용하여, 5 ㎖ 1장, 2 ㎖ 2장, 0.2 ㎖ 2장에 뿌리고, G418(GIBCO-BRL)을 1200 micro-g/㎖를 포함하는 10 ㎖의 10% FBS를 포함하는 MEM 배지에 배양을 행하여, 2일마다 배지를 교환하면서, 14일간 배양하고, 유전자의 안정 도입주의 선택을 행한다. 상기 배지에 의해 생육되어온 G418에 내성을 나타내는 세포는 클로닝 링(cloning ring)을 사용하여 회수한다. 회수한 각 클론은 10 ㎝ 플레이트에서 컨플루언트가 될 때까지 확대 배양을 계속한다.
F 단백질의 발현 유도는, 세포를 6 ㎝ 샬레에 컨플루언트까지 생육시킨 후, 아데노바이러스 AxCANCre를 사이토 등의 방법(Saito et al., Nucl. Acids Res. 23: 3816-3821(1995); Arai, T. et al., J. Virol 72, 1115-1121(1998))에 의해, 예를 들면, moi=3으로 감염시켜 행할 수 있다.
<3> F 유전자 결실 SeV 바이러스의 재구축 및 증폭
상기 pSeV18+/△F의 외래 유전자가 삽입된 플라스미드를 이하와 같이 하여 LLC-MK2 세포에 트랜스펙션한다. LLC-MK2 세포를 5×106 cells/dish로 100 ㎜의 샬레에 뿌린다. T7 RNA 폴리머라아제에 의해 게놈 RNA의 전사를 행하게 하는 경우에는, 세포 배양 24시간 후, 소랄렌(psoralen)과 장파장 자외선(365 ㎚)으로 20분간 처리한 T7 RNA 폴리머라아제를 발현하는 재조합 백시니아 바이러스(recombinant vaccinia virus)(PLWUV-VacT7: Fuerst, T. R. et al., Proc. Natl. Acad. Sci. USA 83, 8122-8126(1986))를 MOI 2 정도로 실온에서 1시간 감염시킨다. 백시니아 바이러스로의 자외선 조사에는, 예를 들면, 15 와트 벌브(watt bulb) 5개가 장비된 UV Stratalinker 2400(카탈로그 번호 400676(100 V), 스트라타진사, La Jolla, CA, USA)을 사용할 수 있다. 세포를 무혈청의 MEM으로 세정한 후, 게놈 RNA를 발현하는 플라스미드, 및 마이너스 가닥 RNA 바이러스의 각각 N, P, L, F 및 HN 단백질을 발현하는 발현 플라스미드를, 적당한 리포펙션 시약을 사용하여 이 세포에 트랜스펙트한다. 플라스미드의 양비는, 이것에 한정되지 않지만, 적합하게는 순서대로 6:2:1:2:2:2로 할 수 있다. 예를 들면, 게놈 RNA를 발현하는 플라스미드, 및 N, P, L, F 및 HN 단백질을 발현하는 발현 플라스미드(pGEM/NP, pGEM/P, pGEM/L 및 pGEM/F-HN; WO00/70070, Kato, A. et al., Genes Cells 1, 569-579(1996))를, 각각 12 micro-g, 4 micro-g, 2 micro-g, 4 micro-g 및 4 micro-g/dish의 양비로 트랜스펙트한다. 수시간 배양 후, 혈청을 포함하지 않는 MEM으로 세포를 2회 세정하여, 40 micro-g/mL의 Cytosine β-D-arabinofuranoside(AraC: Sigma, St. Louis, MO) 및 7.5 micro-g/mL의 Trypsin(Gibro-BRL, Rockville, MD)을 포함하는 MEM에서 배양한다. 이들의 세포를 회수하여, 펠렛(pellet)을 OptiMEM에 현탁한다(107 cells/㎖). 동결 융해를 3회 반복하여 리포펙션시약(lipofection reagent) DOSPER(Boehringer mannheim)와 혼합하고(106 cells/25 micro-L DOSPER) 실온에서 15분 방치한 후, 상기에서 클로닝한 F 발현 헬퍼 세포에 트랜스펙션(106 cells/well 12-well-plate)하여, 혈청을 포함하지 않는 MEM(40 micro-g/㎖ AraC, 7.5 micro-g/㎖ 트립신을 포함한다)에서 배양하고, 상청을 회수한다. F 이외의 유전자, 예를 들면, HN 또는 M 유전자를 결손한 바이러스도 이것과 동일한 방법으로 조제할 수 있다.
또한, 본 발명의 마이너스 가닥 RNA 바이러스는, 재조합 바이러스가 아닌, 천연 바이러스일지라도 사용하는 것이 가능하고, 각 RNA 바이러스의 정제방법, 증식방법, 단리주의 취득방법에 대해서는, 바이러스학 실험학 각론, 개정 2판(국립예방위생연구소 학우회 편, 마루젠, 1982)을 참조할 수 있다. 예를 들면, 파라믹소바이러스과(Paramyxoviridae)의 센다이 바이러스 등 파라인플루엔자 바이러스는, 각 형태 모두 원숭이 신장(MK2), 인간 태아 폐(human embryonic lung), 신장, 양막(amnion)의 초대 배양세포, 및 트립신 첨가 Vero 세포에서 잘 증식하여(상기와 동일, p334; Itoh H. et al., Jap. J. Med. Sci. Biol. 23, 227 (1970)) 회수할 수 있다. 정제는, 수크로오스 밀도 구배원심법(sucrose density gradient centrifugation method), 평형원심법(equilibrium centrifugation method) 등으로 정제할 수 있다(p336). 홍역 바이러스는, 원숭이 유래의 각종 세포에서 잘 증식할 수 있어(Matsumoto M, Bact. Rev. 30, 152(1966)) Vero 세포가 가장 범용되고 있지만, CV1, FL, KB, HeLa HEp2 등을 사용하여 증식 가능하다(p351). 광견병 바이러스 등의 라브도바이러스과(Rhabdoviridae)에서는 조직배양법을 사용하여, BHK, CE 및 Vero 세포 등이 사용되고 있다. 정제법으로서는, 감염 3~4일의 배양액을 pH 7.4 이상으로 보정하고, 저속 원심으로 세포편(cell debris)을 제거하고 농축한다(p376). 라사 바이러스(lassa virus) 등의 아레나바이러스과(Arenaviridae)는, 인 비트로로 계대되어 있는 거의 모든 배양세포에서 잘 증식하지만, HK-21/13S 세포에 감염시킨 후, 한천(agar) 중에 현탁한 형태로 배양함으로써 증식이 가능하다(Sedwik W. D., J. Virol. 1, 1224 (1967))(p240). 인플루엔자 바이러스 등의 오르토믹소바이러스과(Orthomyxoviridae)는, 발육 계란이나 MDCK 세포에 있어서 증식할 수 있다(p295). 정제는 원심분획법, 적혈구로의 흡착·유출(release)에 의한 정제(Laver W. G.., Fundamental Techniques in Virology, 82(1969)) 등으로 정제할 수 있다(p317).
본 발명의 하나의 태양에 있어서, 마이너스 가닥 RNA 바이러스에 의해 도입하는 외래 유전자로서는, 특별히 제한은 없지만, 천연 단백질로서는, 예를 들면, 호르몬, 사이토카인, 증식인자, 수용체, 세포내 시그널 분자, 효소, 펩티드 등을 들 수 있다. 단백질은 분비 단백질, 막 단백질, 세포질 단백질, 핵 단백질 등일 수 있다. 인공적인 단백질로서는, 예를 들면, 키메라 독소 등의 융합 단백질, 도미넌트 음성 단백질(dominant negative protein)(수용체의 가용성 분자 또는 막 결합형 도미넌트 음성 수용체를 포함한다), 결실형 세포 접착분자 및 세포 표면분자 등을 들 수 있다. 또한, 분비 시그널, 막 국재화 시그널(membrane-localization signal), 또는 핵 이행 시그널 등을 부가한 단백질이어도 된다. 도입 유전자로서 안티센스 RNA 분자 또는 RNA 절단형 리보자임 등을 발현시켜, 특정 유전자의 기능을 억제하는 것도 가능하다. 외래 유전자로서 질환의 치료용 유전자를 사용하여 본 발명의 바이러스를 조제하면, 이 바이러스를 투여하여 유전자 치료를 행하는 것이 가능해진다.
본 명세서에 기재한 바이러스 제조방법에 따르면, 본 발명의 바이러스는, 예를 들면, 1×105 CIU/mL 이상, 바람직하게는 1×106 CIU/mL 이상, 보다 바람직하게는 5×106 CIU/mL 이상, 보다 바람직하게는 1×107 CIU/mL 이상, 보다 바람직하게는 5×107 CIU/mL 이상, 보다 바람직하게는 1×108 CIU/mL 이상, 보다 바람직하게는 5×108 CIU/mL 이상의 역가로 바이러스 생산세포의 세포외액 중에 방출시키는 것이 가능하다. 바이러스의 역가는, 본 명세서 및 그 외에 기재된 방법에 의해 측정할 수 있다(Kiyotani, K. et al., Virology 177(1), 65-74(1990); WO00/70070).
회수한 바이러스는 실질적으로 순수하게 되도록 정제할 수 있다. 정제방법은 여과(filtration), 원심분리, 흡착, 및 칼럼 정제 등을 포함하는 공지의 정제·분리방법 또는 그의 임의의 조합에 의해 행할 수 있다. 「실질적으로 순수」란, 바이러스를 포함하는 용액 중에서 바이러스의 성분이 주요한 비율을 차지하는 것을 말한다. 예를 들면, 실질적으로 순수한 바이러스 조성물은, 용액 중에 포함되는 전체 단백질(단, 캐리어(carrier)나 안정제로서 첨가한 단백질은 제외한다) 중, 바이러스의 성분으로서 포함되는 단백질의 비율이 10%(중량/중량) 이상, 바람직하게는 20% 이상, 보다 바람직하게는 50% 이상, 바람직하게는 70% 이상, 보다 바람직하게는 80% 이상, 더욱 바람직하게는 90% 이상을 차지함으로써 확인할 수 있다. 예를 들면, 파라믹소바이러스인 경우, 구체적인 정제방법으로서는, 셀룰로오스 황산 에스테르 또는 가교 폴리사카라이드 황산 에스테르를 사용하는 방법(일본국 특허공고 제(소)62-30752호 공보, 일본국 특허공고 제(소)62-33879호 공보, 및 일본국 특허공고 제(소)62-30753호 공보), 및 푸코오스 황산 함유 다당 및/또는 그의 분해물에 흡착시키는 방법(WO97/32010) 등을 예시할 수 있지만, 이들에 제한되지 않는다.
본 발명에 있어서의 마이너스 가닥 RNA 바이러스는, 예를 들면, 감염성 바이러스 입자여도 비감염성 바이러스 입자여도 된다. 또한, 본 발명의 RNA 바이러스는, 게놈 RNA-단백 복합체(ribonucleoprotein complex; RNP)여도 된다. 이들의 입자 또는 복합체를 이용하는 경우에는, 이들 입자 또는 복합체를, 리포펙션 시약과 혼합시켜 인 비보 투여하는 것이 바람직하다. 예를 들면 리포펙트아민이나 폴리카티오닉 리포솜 등과 입자 또는 복합체를 혼합시켜, 그의 혼합물을 인 비보 투여할 수 있다(WO00/70055). 이 방법에 있어서는, 여러 종류의 트랜스펙션 시약을 이용할 수 있다. 예를 들면, DOTMA(Boehringer), Superfect(QIAGEN #301305), DOTAP, DOPE, DOSPER(Boehringer #1811169) 등을 들 수 있다. 엔도솜 중에서의 분해를 막기 위해, 클로로퀸을 첨가하는 것도 가능하다(Calos, M. P., 1983, Proc. Natl. Acad. Sci. USA 80: 3015).
본 발명에 있어서의 마이너스 가닥 RNA 바이러스를 포함하는 항암제의 제조에 있어서는, 마이너스 가닥 RNA 바이러스는 필요에 따라서 약리학적으로 허용되는 목적의 담체 또는 매체와 혼합(조합)할 수 있다.
즉 본 발명은, 마이너스 가닥 RNA 바이러스와, 약학적으로 허용되는 담체 또는 매체를 혼합하는 공정을 포함하는, 항암제의 제조방법을 제공한다.
「약학적으로 허용되는 담체 또는 매체」란, 마이너스 가닥 RNA 바이러스와 함께 투여하는 것이 가능하고, 마이너스 가닥 RNA 바이러스의 감염을 유의하게 저해하지 않는 재료이다. 이와 같은 담체 또는 매체로서는, 예를 들면, 탈이온수, 멸균수, 염화나트륨 용액, 덱스트로오스 용액(dextrose solution), 덱스트로오스 및 염화나트륨, 유산(lactic acid) 함유 링거 용액, 배양액, 혈청, 인산 완충 생리식염수(PBS) 등을 들 수 있고, 이들과 마이너스 가닥 RNA 바이러스를 적절히 조합시켜 제제화하는 것을 생각할 수 있다. 또한, 필요에 따라서 원심 등에 의해 농축되어, 배양액 또는 생리식염수 등의 생리적 용액 중에 재현탁되어도 된다. 또한, 리포솜의 막 안정화제(예를 들면, 콜레스테롤 등의 스테롤류)를 포함하고 있어도 된다. 또한, 항산화제(예를 들면, 토코페롤 또는 비타민 E 등)를 포함하고 있어도 된다. 추가로, 그 밖에도 식물유, 현탁제, 계면활성제, 안정제, 살생물제 등이 함유되어 있어도 된다. 또한 보존제나 그 밖의 첨가제를 첨가할 수 있다. 본 발명의 조성물은, 수용액, 캡슐, 현탁액, 시럽 등의 형태일 수 있다. 또한 본 발명의 바이러스 조성물은 용액, 동결 건조물, 또는 에어졸(aerosol) 형태의 조성물이어도 된다. 동결 건조물의 경우는 안정화제로서 소르비톨(sorbitol), 수크로오스(sucrose), 아미노산 및 각종 단백질 등을 포함하고 있어도 된다.
본 발명의 마이너스 가닥 RNA 바이러스는 항암 활성(anticancer activity)을 갖고 있어, 종양(암조직)에 투여함으로써 항암(제암)작용을 발휘할 수 있다. 즉 본 발명은, 마이너스 가닥 RNA 바이러스를 암조직으로 투여(인 비보 투여)하는 공정을 포함하는, 암의 억제방법(암세포 증식의 억제방법, 또는 암의 치료방법)에 관한 것이다.
예를 들면, 암환자에 대하여 암치료를 행할 수 있다. 이 방법은, 마이너스 가닥 RNA 바이러스(본 발명의 항암제)를 투여하는 공정을 포함하는 방법이다. 구체적으로는, 마이너스 가닥 RNA 바이러스의 치료상 유효량을, 환자에게 투여하는 공정을 포함하는 방법이다. 본 방법에 의해, 본 발명의 마이너스 가닥 RNA 바이러스를 투여하지 않는 경우에 비하여, 암세포의 증식을 억제하는 것을 기대할 수 있다. 마이너스 가닥 RNA 바이러스는, 외래 유전자를 갖지 않는 것이어도 되고, 또는 암 항원, 면역자극성 사이토카인, 혈관 신생을 저해하는 단백질 등의 하나 또는 복수를 코드하는 유전자(외래 유전자)를 갖는 것이어도 된다.
본 발명은, 목적의 고형암(solid cancer)에 적용할 수 있고, 예를 들면 설암(tongue cancer), 치은암(gum cancer), 악성 림프종, 악성 흑색종(멜라노마), 상악암(upper jaw cancer), 비암(nose cancer), 비강암(nasal cavity cancer), 후두암(larynx cancer), 인두암(pharyngeal cancer), 신경교종(glioma), 수막종(meningioma), 신경교종, 폐암, 유방암, 췌장암, 소화기암(식도암, 위암, 십이지장암, 대장암), 편평상피암(squamous cancer), 선암(adenocarcinoma), 폐포상피암(alveolar cell cancer), 고환 종양(testis tumor), 전립선암, 갑상선암, 간암, 난소암, 횡문근육종(rhabdomyosarcoma), 섬유육종(fibrosarcoma), 골육종(osteosarcoma), 연골육종(chondrosarcoma) 등을 들 수 있다. 대상이 되는 암은, 바람직하게는 상피암이고, 보다 바람직하게는, 피부 편평상피암, 피부 기저세포암(basal cell cancer of the skin), 피부 보웬병(Bowen's disease of the skin), 피부 파제트병(Paget's disease of the skin), 피부 악성 흑색종 등을 포함하는 피부암이다.
본 발명의 마이너스 가닥 RNA 바이러스(본 발명의 항암제)의 인 비보에서의 투여량은, 질환, 환자의 체중, 연령, 성별, 증상, 투여목적, 투여 조성물의 형태, 투여방법, 도입 유전자 등에 따라 다르지만, 당업자라면 적절히 결정하는 것이 가능하다. 투여경로는 적절히 선택할 수 있지만, 예를 들면, 경피적(percutaneous), 비강내적(intranasal), 경기관지적(transbronchial), 근육내적(intramuscular), 복강내(intraperitoneal), 정맥내(intravenous), 관절내(intraarticular), 또는 피하 등에 행하여질 수 있다. 투여는 국소 또는 전신이어도 된다. 투여되는 바이러스는 바람직하게는 약 105 CIU/㎖~약 1011 CIU/㎖, 보다 바람직하게는 약 107 CIU/㎖~약 109 CIU/㎖, 가장 바람직하게는 약 1×108 CIU/㎖~약 5×108 CIU/㎖ 범위 내의 양을 약학상 용인 가능한 담체 중에서 투여하는 것이 바람직하다. 인간에 있어서는 1회당 투여량은 2×105 CIU~2×1011 CIU가 바람직하고, 투여 횟수는 1회 또는 임상상 용인 가능한 부작용의 범위에서 복수회 가능하며, 1일의 투여 횟수에 대해서도 동일하다. 인간 이외의 동물에 대해서도, 예를 들면, 목적의 동물과 인간과의 체중비 또는 투여 표적부위의 용적비(예를 들면 평균값)로 상기의 투여량을 환산한 양을 투여할 수 있다. 또한, 전파성의 마이너스 가닥 RNA 바이러스를 개체 또는 세포에 투여 후, 치료가 완료되는 등의 바이러스의 증식을 억지(抑止)할 필요가 발생했을 때에는, RNA 의존성 RNA 폴리머라아제 저해제를 투여하면, 숙주에 장해를 부여하지 않고, 바이러스의 증식만을 특이적으로 억지하는 것도 가능하다.
본 발명의 마이너스 가닥 RNA 바이러스를 투여하는 경우는, 바람직하게는, 환자의 암 병소(cancerous lesion)에 투여한다. 암 병소란, 암조직 또는 그의 주위(예를 들면 암으로부터 5 ㎜ 이내, 바람직하게는 3 ㎜ 이내)의 영역을 말한다. 또한 투여량은, 암의 타입이나 스테이지, 도입 유전자의 유무 등에 의해 적절히 조절되어도 된다. 마이너스 가닥 RNA 바이러스는 외래 유전자를 탑재하고 있지 않아도 항종양 효과가 기대되지만, 보다 상승적인 효과를 기대하여, 예를 들면, IFN-beta 유전자 또는 가용성 FGF 수용체 유전자 등을 RNA 바이러스에 탑재시키는 것도 가능하다.
마이너스 가닥 RNA 바이러스의 투여 횟수는, 1회 또는 임상상 용인 가능한 부작용의 범위에서 복수회 가능하며, 1일의 투여 횟수에 대해서도 동일하다. 투여대상으로서는 특별히 제한은 없지만, 예를 들면, 닭, 메추라기, 마우스, 랫트, 개, 돼지, 고양이, 소, 토끼, 양, 염소, 원숭이, 및 인간 등을 포함하는 조류, 포유동물(인간 및 비인간 포유동물)을 들 수 있다. 인간 이외의 동물에 투여하는 경우는, 예를 들면, 목적의 동물과 인간과의 체중비로 상기의 투여량을 환산한 양을 투여할 수 있다.
또한 본 발명은, 마이너스 가닥 RNA 바이러스의, 암치료에 있어서의 사용에 관한 것이다. 또한, 본 발명은, 마이너스 가닥 RNA 바이러스의, 항암제(또는 제암제, 암 증식억제제 등)의 제조에 있어서의 사용에 관한 것이다.
또한 본 발명은, 마이너스 가닥 RNA 바이러스를 포함하는 패키지로서, 상기 마이너스 가닥 RNA 바이러스를 암 억제에(항암제로서) 사용하는 것의 기재를 포함하는 패키지에 관한 것이다. 마이너스 가닥 RNA 바이러스는, 배양액 또는 생리식염수 등의 용액 중에 현탁되어 있어도 된다. 암 억제에 사용하는 것이란, 예를 들면 마이너스 가닥 RNA 바이러스, 또는 그것을 포함하는 조성물이, 종양 증식억제, 암의 축퇴(縮退), 암 치료, 암환자의 처치·치료, 암환자의 연명, 또는 항암제로서 사용되는 것이다. 상기 기재는, 패키지에 직접 기재되어 있어도 되고, 상기 기재를 포함하는 종이나 스티커를 패키지에 포함하고 있어도 된다. 패키지는, 마이너스 가닥 RNA 바이러스를 포함하는 용기여도 되고, 그 경우, 용기는, 예를 들면 병(bottle), 튜브(tube), 비닐 봉지(plastic bag), 바이알(vial), 시린지(syringe) 등이어도 된다. 또한 본 발명의 패키지는, 상기 용기를 수납하는 백 또는 바깥상자(outer box) 등을 포함해도 된다. 패키지에는, 마이너스 가닥 RNA 바이러스의 투여방법이 기재된 설명서를 포함해도 되고, 마이너스 가닥 RNA 바이러스 투여를 위한 시린지, 카테터(catheter), 및/또는 주사바늘 등을 추가로 포함해도 된다.
추가로 본 발명은, 마이너스 가닥 RNA 바이러스를 적어도 구성성분으로서 함유하는, 암 치료용 또는 항암제 제조용 키트(kit)에 관한 것이다. 본 발명의 키트는, 마이너스 가닥 RNA 바이러스 이외에, 예를 들면, 담체, 생리식염수, 완충액, 병, 튜브, 비닐 봉지, 바이알, 시린지 등을, 적절히 포함하고 있어도 된다. 또한, 사용서를 패키징하는 것도 가능하다.
또한, 본 명세서 중에 인용된 문헌은, 모두 본 명세서의 일부로서 포함된다.
이하, 실시예에 의해 본 발명을 더욱 상세하게 설명하지만, 본 발명은 이들 실시예에 제한되는 것은 아니다.
[실시예 1] 치료 유전자를 갖지 않는 센다이 바이러스에 의한 항종양 효과
흑색종 세포주 B16F1(ATCC CRL-6323) 1×105개를, C57BL/6 마우스(6~8주령, 암컷)의 복측 피하로 접종하였다(n=4). 접종 후 5일(day 5) 및 12일(day 12)에, 특별한 치료 유전자를 갖지 않는 센다이 바이러스(GFP 발현 SeV; SeV-GFP), 인간 가용형 FGF 수용체를 발현하는 센다이 바이러스(SeV-sFGFR), 또는 인간 PDGFRα 가용형을 발현하는 센다이 바이러스(SeV-hsPDGFRα), 각 1×108 PFU를 종양내 주사하고, 이후, 종양 사이즈를 경시적으로 측정하였다.
그 결과, 어느 SeV 투여군에 있어서도, SeV 비투여군에 비하여 종양 사이즈가 유의하게 축소되었다(도 1). 이와 같이, SeV의 인 비보 투여는, 치료 유전자를 갖지 않는 것일지라도 항종양 효과를 발휘할 수 있었다. 가용형 FGF 수용체를 발현하는 SeV를 투여한 경우는, SeV-GFP 투여군 보다도 강한 종양 증식억제 효과가 확인되고, 가용형 PDGFRα를 발현하는 SeV를 투여한 경우의 항종양 효과는 가장 현저하여, 종양 사이즈는 거의 증가하지 않았다.
이상으로부터, 치료 유전자 등의 외래 유전자를 갖지 않는 SeV일지라도, 충분한 항종양 효과를 발휘하는 것이 명백해졌다.
[실시예 2] 치료 유전자(IFNβ)를 갖는 센다이 바이러스에 의한 항종양 효과
본 실시예는, RNA 바이러스의 인 비보 및 엑스 비보(ex vivo) 투여에 의한 종양의 치료방법의 일례를 나타낸다.
종양 모델로서, MHC Class Ⅰ을 매우 낮은 레벨로밖에 발현하지 못하고 면역원성이 부족한 B16 흑생종의 이식 모델을 채용하였다. 종양 모델 마우스에게는, C57BL/6 마우스(6~8주령, 암컷)(일본 찰스 리버)를 사용하고, 수상세포(dendritic cell)는 C57BL/6 마우스(8주령, 암컷)(일본 찰스 리버)로부터 채취하였다. 수상세포는, C57BL/6 마우스의 대퇴골로부터 골수를 채취하여, SpinSepTM, murine hematopoetic progenitor enrichmentcocktail(항CD5 항체, 항CD45R 항체, 항CD11b 항체, 항Gr-1 항체, 항TER119 항체, 항7/4 항체, StemCell technology)을 사용하고, T세포를 제거한 후, IL-4 및 GM-CSF를 첨가하여 1주간 배양하여 얻었다. 1×105/100 micro-L의 B16 흑색종 세포를 day 0에 마우스의 복부 피하(s. c.) 접종하였다. Day 10, 17, 및 24에 활성화 자극을 가하지 않는 수상세포, LPS로 활성화시킨 수상세포(LPS DC), 또는 SeV-GFP 또는 마우스 인터페론(mouse interferon) β를 발현하는 SeV-IFN β를 도입하여 활성화시킨 수상세포(각각 SeV GFP DC 또는 SeV IFN β DC)를 종양 주위에 투여하였다. 이 때, 수상세포에 종양항원(B16의 냉동과 해동(freeze and thaw)에 의한 종양 용해물)을 펄스한 후 투여하는 실험도 행하였다. 이들과는 별도로, 종양 접종 후 10일째(day 10)에 SeV-IFN β를 직접, 종양내 주입(intratumoral injection)하여 항종양 효과를 조사하는 실험도 행하였다.
수상세포로의 SeV의 도입에서는, 상기와 같이 1주간 배양한 수상세포에 SeV- IFN β를 MOI 40에서 감염시켜 8시간 배양하였다. 종양항원을 펄스하는 경우는, 상기와 같이 1주간 배양한 수상세포를 회수하여, 종양항원이 되는 종양 용해물(tumor lysate)을 펄스(DC: tumor lysate=1:3)하고, 18시간 배양한 후, SeV-IFN β를 MOI 40에서 감염시켜 8시간 배양하였다. 그 다음, 이들의 수상세포를 회수하여, 5×105~10×105 세포수를 마우스의 종양 주위에 투여하였다.
도 2에 나타내는 바와 같이, SeV-IFN β의 직접 종양내 주입 및 수상세포를 매개로 한 엑스 비보 투여는, 모두 종양 증식을 억제하였다. 특히 DC/SeV-IFN β로 처리한 마우스에서는, 매우 강한 종양 억제효과가 관찰되었다.
상기의 각 치료군에 있어서의 항종양 효과를 보다 상세하게 검토하였다. 내츄럴 킬러(natural killer)(NK) 세포 활성을 어세이하기 위해, 상기의 각 치료군에 대해서, 3회의 DC 요법 종료 7일 후의 마우스로부터 비장을 적출하여 이펙터 세포(effector cell)를 작성하였다. 타겟으로서 Yac-1을 사용하여, 51Cr 방출 어세이(release assay)를 행하였다. 또한, T 림프구의 세포상해성을 어세이하기 위해, 상기의 NK세포 활성 어세이에 사용한 비장세포의 나머지를 사용하고, B16의 종양항원인 TRP-2 펩티드와 함께, 5일간 배양한 세포를 이펙터 세포로서 사용해서, mTRP-2 펩티드를 펄스한 EL-4 타겟 세포와 공배양하여, 51Cr 방출 어세이를 행하였다. 특이적 51Cr 방출의 비율은 이하와 같이 계산하였다.

여기에서, 최대 방출은 1% triton X와 함께 인큐베이트한 타겟 세포, 자발 방출은 배양액만으로 인큐베이트한 타겟 세포를 사용하였다.
내츄럴 킬러(NK) 세포의 활성화는, 직접 SeV를 주입한 마우스에만 보이고, 수상세포 투여군에서는 검출되지 않았다(도 3). 이것에 대하여, 세포상해성 T 림프구(cytotoxic T-lymphocytes, CTL)의 활성화는, DC/LPS 처리군 및 DC/SeV-IFN β로 처리한 마우스에서 가장 강하고, DC/SeV-GFP 처리군에서는 그것보다도 약간 약하며, SeV-IFN β의 직접 주입군에서는 검출되지 않았다(도 4). 종양 용해물의 펄스는, 종양 증식에도 CTL 응답에도 유의하게 영항을 끼치지 않았다. 이와 같이, SeV-IFN β의 직접투여는 NK세포를 강하게 활성화시키는 것이 판명되었다.
따라서, SeV를 투여한 경우의 종양 억제효과는, NK세포의 활성화를 메커니즘(mechanism)로 하는 것임이 시사되었다. 더욱이, 치료 유전자를 갖지 않는 SeV를 투여하는 것에 추가하여, 치료효과가 기대되는 유전자를 갖는 SeV를 투여하는 것에 의해서도, 암에 대하여, 보다 유효한 치료효과가 발휘되는 것이 나타내어졌다.
본 발명에 의해, 마이너스 가닥 RNA 바이러스를 유효성분으로 하는 항암제가 제공되었다. 본 발명의 RNA 바이러스는, 치료효과를 갖는 외래 유전자를 갖지 않는 상태에서, 효과적인 암 치료효과가 발휘되기 때문에, 외래 유전자의 도입을 위한 비용의 삭감, 및 바이러스의 조제를 위한 조작시간의 삭감에 크게 기여할 것으로 생각되어진다. 본 발명은, 마이너스 가닥 RNA 바이러스에 의한 새로운 바이러스 요법을 가능하게 한다.

- 제1항 내지 제3항 중 어느 한 항에 있어서, RNA 바이러스가 게놈 RNA-단백 복합체인 항암제.
- 제1항 내지 제4항 중 어느 한 항에 있어서, 마이너스 가닥 RNA 바이러스가 파라믹소바이러스인 항암제.
- 제5항에 있어서, 파라믹소바이러스가 센다이 바이러스인 항암제.
- 마이너스 가닥 RNA 바이러스와, 약학적으로 허용되는 담체 또는 매체를 혼합하는 공정을 포함하는 항암제의 제조방법.
